EXPANDING BIOMEDICAL RESEARCH CAPACITY AT BRYANT UNIVERSITY - BRYANT UNIVERSITY REQUESTS NIH FUNDING TO BUILD THREE SHARED RESEARCH SPACES THAT WILL SIGNIFICANTLY EXPAND THE INSTITUTION’S BIOMEDICAL RESEARCH CAPACITY. THE PROPOSED PROJECT WILL INCLUDE (1) THE EXPANSION AND MODERNIZATION OF AN ANALYTICAL LABORATORY DESIGNED TO SUPPORT FACULTY RESEARCH AND UNDERGRADUATE TRAINING ACROSS A VARIETY OF SCIENTIFIC DISCIPLINES; (2) THE CONSTRUCTION OF A TISSUE CULTURE LAB THAT WILL HOUSE A LAMINAR FLOW HOOD, A BIOSAFETY LEVEL II CABINET, AND INCUBATORS TO SUPPORT LEVEL II CELL CULTURE WORK; AND (3) THE CONSTRUCTION OF A MICROSCOPY ROOM THAT WILL HOUSE THE FIRST CONFOCAL MICROSCOPE IN RHODE ISLAND WITH RAMAN CAPABILITIES TO IMAGE LIVE CELLS. CREATING THESE SHARED RESEARCH SPACES WILL REVOLUTIONIZE THE BIOMEDICAL RESEARCH THAT BRYANT CAN CONDUCT. THE CURRENT ANALYTICAL LABORATORY IS A 761-SQUARE- FOOT FACILITY THAT IS OUTDATED AND UNDER-OUTFITTED TO SUPPORT CUTTING-EDGE RESEARCH. THIS PROJECT WILL INCREASE THE SIZE OF THE SPACE BY 45%, TO 1,500 SQUARE FEET, AND WILL MODERNIZE THIS SHARED RESEARCH SPACE WITH EQUIPMENT NECESSARY FOR THE INSTITUTION TO BECOME A LEADER IN BIOMEDICAL RESEARCH. CURRENTLY, BRYANT DOES NOT HAVE A MICROSCOPY ROOM OR ANY SPACE ON CAMPUS DEDICATED TO TISSUE CULTURE WORK. THIS MEANS THAT THE FACULTY CAN ONLY CONDUCT RUDIMENTARY SAMPLING OR CANNOT CONDUCT RESEARCH FOR RISK OF BIOHAZARD. LASTLY, A 250-SQUARE-FOOT MICROSCOPY ROOM WILL BE BUILT TO HOUSE A CONFOCAL MICROSCOPE WITH RAMAN CAPABILITIES. THE RESEARCH THESE SPACES WILL MAKE POSSIBLE WILL ENHANCE CRITICAL WORK ON WOMEN’S HEALTH DISPARITIES, PARTICULARLY IN THE AREAS OF ANTIBIOTIC RESISTANCE AND ADDICTION, BOTH CONDITIONS THAT DISPROPORTIONATELY AFFECT WOMEN. WITH THE PROPOSED CONSTRUCTION, BRYANT WILL REALIZE THE FOLLOWING FOUR OBJECTIVES: (1) TRANSFORM AND EXPAND CRITICAL RESEARCH SPACE AVAILABLE TO BRYANT FACULTY AND STUDENTS (WITH A 10% INCREASE IN STUDENT RESEARCHERS ANNUALLY OVER FIVE YEARS); (2) MAKE POSSIBLE NEW RESEARCH ON WOMEN’S HEALTH, PARTICULARLY REGARDING ANTIBIOTIC RESISTANCE AND ADDICTION (WITH A 50% INCREASE IN PUBLICATIONS OVER FIVE YEARS); (3) ENABLE INTERDISCIPLINARY COLLABORATIONS AND FOSTER INNOVATION IN BIOMEDICAL RESEARCH IN RHODE ISLAND (WITH A 100% INCREASE IN OUTSIDE RESEARCHERS WHO USE BRYANT FACILITIES OVER FIVE YEARS); AND (4) PREPARE STUDENTS FOR CAREERS IN THE BIOMEDICAL SCIENCES THROUGH HANDS-ON RESEARCH AND TRAINING EXPERIENCES (WITH 100% OF ALL SHBS STUDENTS, EXCLUDING PSYCHOLOGY, TRAINED ON NEW LAB INSTRUMENTS AND TECHNIQUES).

SPECIFICALLY, FUNDS WILL SUPPORT BRYANT UNIVERSITY’S DOCTORAL PROGRAM IN CLINICAL PSYCHOLOGY, PROVIDING A STATE-OF-THE-ART PSYCHOTHERAPY TRAINING SUITE, CHILD DEVELOPMENT TEACHING AND RESEARCH LAB, AS WELL AS DEDICATED INSTRUCTIONAL AND RESEARCH SPACE FOR PSYCHOPHYSIOLOGY, COGNITIVE, AND SOCIAL PSYCHOLOGY. ADVANCEMENT OF THE PSYCHOLOGY UNDERGRADUATE PROGRAM AND DEVELOPMENT OF THE DOCTORAL PROGRAM IN CLINICAL PSYCHOLOGY ARE CRITICAL PIECES OF BRYANT UNIVERSITY’S COMMITMENT TO THE EXPANSION OF ITS SCHOOL OF HEALTH AND BEHAVIORAL SCIENCES, WHICH IS DEDICATED TO TRAINING THE NEXT GENERATION OF HEALTHCARE LEADERS AND ENHANCING AND DIVERSIFYING THE HEALTHCARE WORKFORCE. THIS PROJECT WILL EXPAND THE CURRENT PSYCHOLOGY LABS FROM APPROXIMATELY 900 SQUARE FEET TO JUST OVER 4000 SQUARE FEET, PROVIDING AMPLE SPACE TO SUPPORT ITS UNDERGRADUATE AND NEW GRADUATE PROGRAM. NEURAL CIRCUITRY MECHANISMS UNDERLYING MALADAPTIVE REWARD MEMORIES IN DROSOPHILA - PROJECT SUMMARY: ALCOHOL ADDICTION CONSTITUTES ONE OF THE MOST SERIOUS PUBLIC HEALTH PROBLEMS WORLDWIDE. ACCORDING TO THE CDC, THE TOTAL ECONOMIC COST OF EXCESSIVE ALCOHOL USE IS ESTIMATED TO BE $249 BILLION. DESPITE ITS DEVASTATING IMPACT, THERE ARE FEW EFFECTIVE TREATMENTS. NEURAL CIRCUIT BASED APPROACHES TO TREAT ADDICTION PROVIDE A POWERFUL OPPORTUNITY TO DEVELOP MORE SPECIFIC AND EFFECTIVE TREATMENTS. HOWEVER, CIRCUIT COMPLEXITY IS A SIGNIFICANT OBSTACLE TO A COMPREHENSIVE UNDERSTANDING OF THE MECHANISMS BY WHICH MEMORY CIRCUITS ARE ALTERED TO CREATE ENDURING PREFERENCES FOR ALCOHOL ASSOCIATED CUES. THE CURRENT PROPOSAL WILL TAKE PLACE AT BRYANT UNIVERSITY, A PRIMARILY UNDERGRADUATE INSTITUTION, AND WILL CAPITALIZE ON THE GENETIC TRACTABILITY OF DROSOPHILA MELANOGASTER AND PRECISE NEUROGENETIC TOOLS AVAILABLE TO INTERROGATE HOW IDENTIFIED MEMORY CIRCUITS ARE ALTERED BY ALCOHOL AND WHY ALCOHOL ASSOCIATED MEMORIES ARE SO RESISTANT TO CHANGE. ALTHOUGH ONLY 100,000 NEURONS COMPRISE THE CENTRAL NERVOUS SYSTEM OF DROSOPHILA, THE NEURAL CIRCUITRY UNDERLYING REWARD AND ADDICTION IS REMARKABLY COMPLEX AND SIMILAR TO MAMMALS. LIKE MAMMALS, DROSOPHILA SHOW ROBUST PREFERENCE FOR CUES ASSOCIATED WITH ALCOHOL INTOXICATION THAT PERSIST IN THE FACE OF AVERSIVE CONSEQUENCES. STUDIES PROPOSED HERE WILL CAPITALIZE ON ESTABLISHED AND NEWLY DEVELOPED NEUROGENETIC TOOLS THAT ALLOW FOR THE PRECISE MANIPULATION OF INDIVIDUAL NEURONS AND VISUALIZATION OF CIRCUIT ACTIVITY. DOING SO WILL ENABLE US TO TEST THE HYPOTHESIS THAT ALCOHOL DISRUPTS MEMORY CIRCUIT ACTIVITY AND DRIVES MALADAPTIVE REWARD SEEKING BY ENGAGING SIGNIFICANTLY MORE DOPAMINE NEURONS DURING ACQUISITION. SPECIFICALLY, WE WILL USE A COMBINATION OF OPTOGENETICS, THERMOGENETICS, AND TWO-PHOTON MICROSCOPY TO CAPTURE THE PRECISE CHANGES IN DOPAMINE NEURAL DYNAMICS WHILE DROSOPHILA LEARN TO ASSOCIATE ODOR CUES WITH ALCOHOL INTOXICATION. ADDITIONALLY, WE WILL DETERMINE IF ALCOHOL-INDEPENDENT ACTIVATION OF DOPAMINE NEURONS IS SUFFICIENT TO INDUCE MALADAPTIVE MEMORIES FOR NATURAL REWARDS. FINALLY, WE WILL INVESTIGATE THE ROLE OF RECENTLY IDENTIFIED DOWNSTREAM REGIONS OF THE FAN-SHAPED BODY IN MODULATING ALCOHOL REWARD MEMORIES TO UNDERSTAND THE STABILITY AND PERSISTENCE OF THESE MEMORIES. WE EXPECT THAT EXTRACELLULAR DOPAMINE LEVELS AND NUMBER OF RECRUITED DOPAMINE NEURONS WILL SIGNIFICANTLY INCREASE ACROSS ALCOHOL EXPOSURES AND THIS INCREASE IS SUFFICIENT TO DRIVE MALADAPTIVE REWARD SEEKING BEHAVIOR. FURTHER, WE PREDICT THAT DOWNSTREAM CONVERGENT REGIONS PLAY A CRITICAL ROLE IN BEHAVIORAL FLEXIBILITY AND DISRUPTION TO THESE FUNCTIONAL CONNECTIONS RESULTS IN MEMORIES THAT LACK FLEXIBILITY AND DRIVE MALADAPTIVE REWARD SEEKING. ULTIMATELY, UNDERSTANDING GENERAL CIRCUITRY PRINCIPLES DESCRIBED FIRST IN DROSOPHILA WILL PROVIDE INSIGHT TO HOW ALCOHOL CO-OPTS MAMMALIAN CIRCUITS TO CREATE ENDURING PREFERENCE FOR ALCOHOL AND DRIVE MALADAPTIVE CHOICE. AN AREA GRANT IS ESSENTIAL TO CONTINUE OUR EFFORTS IN EXPOSING STUDENTS TO HANDS-ON MERITORIOUS RESEARCH, PROVIDING SUPERIOR RESEARCH EXPERIENCES, AND STRENGTHENING THE RESEARCH ENVIRONMENT AT BRYANT UNIVERSITY, THEREBY ENABLING MORE STUDENTS TO PARTICIPATE.

IRES: ECOLOGICAL RESILIENCE OF AFROMONTANE FORESTS -THIS IRES PROJECT ENGAGES U.S. UNDERGRADUATE AND GRADUATE STUDENTS IN RESEARCH ON THE LONG-TERM CLIMATE AND ENVIRONMENTAL HISTORY AND RESILIENCE OF THE AFROMONTANE FORESTS OF THE USAMBARA MOUNTAINS IN NORTHEASTERN TANZANIA, AN AREA WIDELY KNOWN AS A GLOBAL BIODIVERSITY HOTSPOT, RICH WITH PLANT AND ANIMAL SPECIES FOUND NOWHERE ELSE ON EARTH. HOWEVER, CHANGING RAINFALL PATTERNS, URBAN DEVELOPMENT, AND EXPANDING AGRICULTURE ARE ENDANGERING THE MOUNTAIN ECOSYSTEMS. THIS IRES PROJECT GIVES PARTICIPATING STUDENTS THE OPPORTUNITY TO EXPLORE HOW THESE FORESTS HAVE RESPONDED TO PAST ENVIRONMENTAL CHANGES TO BETTER UNDERSTAND FUTURE IMPACTS. THROUGH RESEARCH EXPERIENCES WITH SCIENTISTS IN THE UNITED STATES, TANZANIA, AND GERMANY, PARTICIPATING STUDENTS ARE HELPING TO RECONSTRUCT THE REGION?S LONG-TERM ECOLOGICAL HISTORY AND DEVELOP CONSERVATION PLANS FOR THE FORESTS. UNDER THE LEADERSHIP OF BRYANT UNIVERSITY, IN COLLABORATION WITH TANZANIAN AND GERMAN RESEARCHERS, THIS IRES PROJECT INVESTIGATES THE ECOLOGICAL HISTORY OF THE USAMBARA MOUNTAINS TO REVEAL PAST FOREST RESPONSES AND INFORM POTENTIAL FUTURE RESILIENCE. THROUGH THE STUDENT RESEARCH ACTIVITIES, U.S. STUDENTS RECEIVE PREPARATORY TRAINING AT BRYANT, CONDUCT FIELDWORK AND SEDIMENT CORE SAMPLING IN TANZANIA IN COLLABORATION WITH THE UNIVERSITY OF DAR ES SALAAM, AND LABORATORY ANALYSIS IN GERMANY AT THE MAX PLANCK INSTITUTE OF GEOANTHROPOLOGY. THIS PROJECT IS THE FIRST EFFORT TO RETRIEVE EARLY-TO-MID HOLOCENE, AND POSSIBLY EVEN OLDER, SEDIMENTS FROM THE USAMBARAS. BY ANALYZING MULTIPLE PALEOENVIRONMENTAL INDICATORS, THIS RESEARCH PRODUCES A NEW ECOLOGICAL RESILIENCE MODEL FOR EASTERN AFRICA AND CONTRIBUTES FOUNDATIONAL DATA TO THE STUDY OF HUMAN-ENVIRONMENT INTERACTIONS. THE PROJECT EMPHASIZES TRAINING AND MENTORSHIP, INTERNATIONAL COLLABORATION, AND ETHICAL COMMUNITY ENGAGEMENT, ENSURING THAT BOTH LOCAL PARTNERS AND STUDENTS BENEFIT FROM THE RESEARCH AND ITS LONG-TERM IMPACTS. RUI: EXPANDING THE CHEMICAL BIOLOGY TOOLBOX FOR STUDYING PEPTIDOGLYCAN METABOLISM -WITH THE SUPPORT OF THE CHEMISTRY OF LIFE PROCESSES (CLP) PROGRAM IN THE DIVISION OF CHEMISTRY, DR. CHRISTOPHER REID AND HIS STUDENTS AT BRYANT UNIVERSITY ARE STUDYING THE DEVELOPMENT OF SMALL MOLECULE PROBES TO STUDY BACTERIAL CELL WALL METABOLISM. MOST BACTERIA ARE SURROUNDED BY A CARBOHYDRATE-BASED POLYMER CALLED PEPTIDOGLYCAN. THIS POLYMER DEFINES CELL SHAPE AND WITHSTANDS THE OUTWARD FACING OSMOTIC PRESSURE. THE METABOLISM OF PEPTIDOGLYCAN IS A HIGHLY COMPLEX AND COORDINATED PROCESS BETWEEN SYNTHETIC AND DEGRADATIVE PROCESSES. WHILE UNDERSTANDING OF PEPTIDOGLYCAN BIOSYNTHESIS IS RELATIVELY ROBUST, THERE ARE SIGNIFICANT KNOWLEDGE GAPS REGARDING THE DEGRADATIVE STEPS AND HOW THEY ARE COORDINATED WITH BIOSYNTHESIS. TO ADDRESS THE LACK OF CHEMICAL BIOLOGY METHODS TO STUDY PEPTIDOGLYCAN DEGRADATION, THIS PROPOSAL LOOKS TO EXPAND ON THE SUCCESS OF THE N-ACETYLGLUCOSAMINIDASE INHIBITOR MASARIMYCIN IN THE GRAM-POSITIVE ORGANISM BACILLUS SUBTILIS TO DEVELOP AN ARRAY OF MASARIMYCIN-BASED CHEMICAL BIOLOGY REAGENTS FOR THE MICROBIAL GLYCOSCIENCE COMMUNITY. THIS WORK COULD PROVIDE NEW INSIGHTS INTO THE ROLE DEGRADATIVE ENZYMES SUCH AS N-ACETYLGLUCOSAMINIDASES PLAY IN CELL GROWTH AND DIVISION. THIS RESEARCH WILL BE CONDUCTED WITH A SIGNIFICANT NUMBER OF UNDERGRADUATE RESEARCHERS. AS SUCH, THE REID LABORATORY WILL CONTINUESTO CONTRIBUTE TO THE TRAINING AND DEVELOPMENT OF THE NEXT GENERATION OF SCIENTISTS. THIS SUPPORT WILL ALLOW FOR THE REID LABORATORY TO CONTRIBUTE TO THE GROWTH AND DEVELOPMENT OF THE CHEMICAL LIFE SCIENCE ECOSYSTEM IN RHODE ISLAND. MOST BACTERIA ARE SURROUNDED BY A PEPTIDOGLYCAN CELL WALL THAT DEFINES THEIR SHAPE AND WITHSTANDS THE OUTWARD FACING OSMOTIC PRESSURE. PEPTIDOGLYCAN METABOLISM IS A HIGHLY COMPLEX AND COORDINATED PROCESS BETWEEN SYNTHETIC AND DEGRADATIVE ENZYMES. WHILE UNDERSTANDING OF THE BIOSYNTHETIC STEPS IS RELATIVELY ROBUST, SIGNIFICANT KNOWLEDGE GAPS EXIST REGARDING THE DEGRADATIVE STEPS AND HOW THEY ARE COORDINATED WITH BIOSYNTHESIS. TO ADDRESS THE LACK OF TOOLS TO STUDY PEPTIDOGLYCAN METABOLISM AND DEGRADATIVE ENZYMES IN PARTICULAR, THE REID LAB AT BRYANT UNIVERSITY HAS DEVELOPED THE SMALL MOLECULE PROBE, MASARIMYCIN (MAS), TO INHIBIT THE MAJOR ACTIVE N-ACETYLGLUCOSAMINIDASE LYTG FROM BACILLUS SUBTILIS. THROUGH THIS PROPOSAL, THE REID LAB SEEKS TO EXPAND THIS WORK BY DEVELOPING NEW GLYCOSYL TRIAZOLE AND DIAMIDE INHIBITORS OF N-ACETYLGLUCOSAMINIDASES AGAINST REPRESENTATIVE GRAM-NEGATIVE, POSITIVE, AND MYCOLICIBACTERIA. MASARIMYCIN PROVIDES A VEHICLE THROUGH WHICH TO STUDY THE DIFFERENCES IN PEPTIDOGLYCAN METABOLISM AND REGULATION IN ROD- AND COCCOID-SHAPED BACTERIA. TO ACHIEVE THESE GOALS, THE REID LAB WILL PURSUE THREE OBJECTIVES IN PARALLEL. FOR THE FIRST OBJECTIVE, STUDIES WILL BUILD ON THEIR PRELIMINARY IDENTIFICATION OF GLCNACASE DIAMIDE INHIBITORS IN B. SUBTILIS AND S. PNEUMONIAE TO CREATE A SECOND-GENERATION MASARIMYCIN INHIBITOR WITH POTENTIALLY IMPROVED POTENCY AND PHYSICAL-CHEMICAL PROPERTIES. FOR THE SECOND OBJECTIVE, PRELIMINARY GLYCOSYL-TRIAZOLE AND DIAMIDE LEADS AGAINST ESCHERICHIA COLI AND MYCOLICIBACTERIUM SMEGMATIS WILL BE EVALUATED TO DETERMINE IF THESE NEW INHIBITORS DISRUPT CELL WALL METABOLISM IN GRAM-NEGATIVE AND ACID-FAST BACTERIA IN THE SAME MANNER AS IN GRAM-POSITIVE ORGANISMS. FINALLY, A COMPARATIVE STUDY BETWEEN B. SUBTILIS AND S. PNEUMONIAE WILL BE PERFORMED, USING MASARIMYCIN AS A PROBE, TO INVESTIGATE THE DIFFERENCES IN PG METABOLISM BETWEEN ROD- AND COCCOID-SHAPED BACTERIA. THE PROPOSED WORK COULD OPEN THE DOOR FOR MASARIMYCIN, AND DIAMIDE INHIBITORS IN GENERAL, TO SERVE AS VALUABLE CHEMICAL BIOLOGY REAGENTS FOR STUDYING PG METABOLISM ACROSS KEY BACTERIAL GENERA.
