THOUGH HIGHLY EFFECTIVE HIV PREVENTION LIKE PRE-EXPOSURE PROPHYLAXIS (PREP) IS CENTRAL TO THE GLOBAL STRATEGY FOR HIV EPIDEMIC CONTROL, AWARENESS OF PREP AMONG AGYW IS LOW, WITH UPTAKE AND CONTINUATION EVEN LOWER—HEALTH SYSTEMS ARE OFTEN ILL EQUIPPED TO OVERCOME THE NUMEROUS ACCESS BARRIERS FACED BY AGYW, INCLUDING STIGMA, HARMFUL NORMS, AND THE INCONVENIENCES OF CLINIC-BASED HEALTH SERVICES. THERE IS GROWING RECOGNITION THAT COMMUNITY-BASED PHARMACIES HAVE THE POTENTIAL TO BRIDGE GAPS IN HIV PREVENTION AND MITIGATE ACCESS BARRIERS AMONG AGYW. THUS, THE GOAL OF THIS STUDY IS TO EVALUATE THE MALKIA KLABU (“QUEEN CLUB”) IMPLEMENTATION STRATEGY IN TANZANIA, A LOYALTY PROGRAM DESIGNED FOR AND BY AGYW THAT CREATES YOUTH-FRIENDLY PHARMACIES WHERE AGYW CAN ACCESS HIV PREVENTION AND SEXUAL AND REPRODUCTIVE HEALTH (SRH) SERVICES, WITH STRONG LINKAGES TO FACILITY-BASED CARE. BASED ON PROMISING DATA FROM OUR PILOT STUDY, WE HYPOTHESIZE THAT THE CREATION OF A WELCOMING AND SAFE PHARMACY ENVIRONMENT THROUGH THE MALKIA KLABU IMPLEMENTATION MODEL WILL ATTRACT AGYW AND BOLSTER UPTAKE OF PREP, HIV SELF-TESTING (HIVST), AND OTHER SRH SERVICES, THEREBY EMPOWERING AGYW TO AVOID HIV AND UNINTENDED PREGNANCY. WE HAVE DESIGNED AN INNOVATIVE, 5-YEAR IMPLEMENTATION SCIENCE STUDY TO SCALE AND EVALUATE SEVERAL MODELS OF MALKIA KLABU’S PHARMACY-BASED APPROACH FOR PREP AND HIVST DELIVERY IN FIVE RESEARCH SITES IN LAKE ZONE, TANZANIA USING THE RE-AIM FRAMEWORK. THIS RESEARCH WILL BE ENHANCED BY IMPLEMENTATION SCIENCE TRAINING OF LOCAL RESEARCHERS AND POLICYMAKERS TO OPTIMIZE MALKIA KLABU AND BENEFIT FUTURE HIV PREVENTION STRATEGIES. WE WILL FIRST CONDUCT A TYPE 3 HYBRID IMPLEMENTATION-EFFECTIVENESS STUDY OF FOUR PHARMACY-BASED DIFFERENTIATED PREP DELIVERY MODELS OF MALKIA KLABU IN 50 PHARMACIES, EXAMINING IMPLEMENTATION OUTCOMES BEFORE AND AFTER THE ADDITION OF USER COST-SHARING WHICH IS ESSENTIAL FOR REAL-WORLD SUSTAINABILITY (AIM 1). WE WILL EXAMINE THE SUCCESS OF THE MALKIA KLABU DELIVERY MODELS ON EFFECTIVE USE OF PREP AMONG A COHORT OF 575 AGYW, REACH OF THE PROGRAM IN A CROSS-SECTIONAL HOUSEHOLD SURVEY WITH 750 AGYW IN STUDY AREAS; PHARMACY EXIT SURVEYS WITH ~200 AGYW WILL HELP DESCRIBE THE SUCCESS OF PHARMACY- BASED PREP SERVICE OFFERINGS (AIM 2). A CERTIFICATE IN IMPLEMENTATION SCIENCE COUPLED WITH SEED GRANTS FOR GRADUATES WILL ENSURE THE PROJECT HAS LASTING IMPACT (AIM 3). AT THE STUDY’S CONCLUSION, WE WILL DETERMINE THE STRONGEST AND MOST SUSTAINABLE VERSION OF THE MALKIA KLABU IMPLEMENTATION STRATEGY FOR PHARMACY-BASED PREP AND HIVST DELIVERY TO AGYW, AND LAUNCHED AN IMPLEMENTATION SCIENCE TRAINING PROGRAM TO ENABLE LOCALLY-LED SCALING AND ADAPTATION TO REACH VULNERABLE AGYW ACROSS TANZANIA AND THE REGION. NEARLY ALL ADULTS IN THE UNITED STATES ARE CONSIDERED AT RISK OF DEVELOPING TOOTH DECAY, AND FOR CHILDREN IT IS A COMMON CHRONIC DISEASE. THOUGH WIDELY AND ROUTINELY USED, THERE ARE ALWAYS CONCERNS ABOUT PATIENT EXPOSURE TO X-RAYS DURING RADIOGRAPHIC IMAGING – A CONCERN ESPECIALLY HEIGHTENED WITH CHILDREN. RADIATION MONITORING DEVICES (RMD) PROPOSES TO DEVELOP AN IMPROVED DIGITAL X-RAY DETECTOR FOR DENTAL RADIOGRAPHY THAT WILL RETAIN ALL THE BENEFITS OF A CURRENT DIGITAL TECHNOLOGY, WHILE IMPROVING SENSITIVITY AND REDUCING PATIENT DOSE. THE DEVICE WILL INVOLVE THE INTEGRATION OF TWO TECHNOLOGIES - SILICON BASED IMAGING ARRAYS AND SEMICONDUCTOR X-RAY CONVERSION LAYERS. THE SILICON CAPABILITIES ARE WELL UNDERSTOOD, WITH CCD AND CMOS DEVICES WELL ESTABLISHED IN DENTAL IMAGING. THE APPLICATION OF A SEMICONDUCTOR CONVERSION LAYER IS THE MORE INNOVATIVE AND CHALLENGING ASPECT OF THE CONCEPT. RMD'S INNOVATION IS TO REPLACE THE COMMONLY USED CSI OR AMORPHOUS SELENIUM X-RAY RECEPTORS WITH THE SEMICONDUCTOR THALLIUM BROMIDE (TLBR). TLBR WILL PROVIDE A MUCH GREATER DOSE EFFICIENCY AND HIGHER IMAGE CONTRAST. THE ADVANTAGE TO DENTAL IMAGING IS THAT DENTISTS WILL GET THE HIGH RESOLUTION IMAGES THAT THEY ARE ACCUSTOMED TO, BUT NOW WITH A MUCH LOWER DOSE TO THE PATIENT. DURING PHASE I, RMD WILL DEVELOP THE PROCESSES TO FABRICATE HIGH QUALITY FILMS OF TLBR AND COMPARE THE TECHNOLOGY TO EXISTING DETECTOR FILMS TO DEMONSTRATE THE SUPERIORITY OF TLBR. THE WORK PLAN WILL ENCOMPASS FILM DEPOSITION, INVESTIGATION OF COMPATIBLE ELECTRODE LAYERS TO WORK IN CONJUNCTION WITH THE TLBR, IDENTIFICATION AND MITIGATION STRATEGIES OF TECHNICAL CHALLENGES THAT ARISE, AND COMPLETION OF A WIDE RANGE OF ELECTRICAL AND PHYSICAL TESTS TO GAUGE X-RAY SENSITIVITY AND IMAGE QUALITY. IN PHASE II RMD WILL OPTIMIZE THE PROCESSES, FABRICATE X-RAY IMAGING DETECTORS ON KA IMAGING CMOS ROICS AND FULLY CHARACTERIZE A COMPLETE DETECTOR SYSTEM. BUT LSO, AFTER YEARS OF DEVELOPMENT, HAS REACHED ITS PERFORMANCE LIMIT, ESPECIALLY FOR THE SCANNERS WITH A LONG AXIAL FIELD OF VIEW (AFOV) THAT ARE CURRENTLY BEING DEVELOPED. THE GOAL OF THESE SCANNERS IS TO INCREASE THE GEOMETRICAL COVERAGE AND SIGNIFICANTLY INCREASE DETECTION SENSITIVITY (BY A FACTOR OF 30-40), THEREBY REDUCING THE SCANNING TIMES (30-40 TIMES FASTER) OR THE PATIENT’S RADIATION EXPOSURE. HOWEVER, LONG AFOV SCANNERS FACE TWO MAIN CHALLENGES: GREATER DEPTH-OF-INTERACTION (DOI) EFFECTS, WHICH INCREASE BLURRING AND NOISE; AND AN INCREASE IN THE VOLUME REQUIRED FOR THE CONSTITUENT CRYSTALS, WHICH MAKE UP SOME 50% OF THE COST OF THE ENTIRE SCANNER. THE USE OF SHORTER CRYSTALS CAN COUNTERACT BOTH THE DOI EFFECTS AND THE INCREASED CRYSTAL VOLUME (HENCE COST), BUT WITH A MAJOR LOSS OF DETECTION EFFICIENCY, DEFEATING THE ORIGINAL PURPOSE. ANOTHER APPROACH FOR REDUCING DOI EFFECTS IS A DOUBLE-ENDED READ-OUT BUT THIS INCREASES BOTH COST AND SYSTEM COMPLEXITY. THEREFORE, TO ACHIEVE VIABLE AND AFFORDABLE LONG AFOV SCANNERS, A NEW SCINTILLATION MATERIAL IS REQUIRED THAT WOULD PROVIDE HIGHER STOPPING POWER THAN LSO, WITH SIMILAR OR BETTER TIMING PROPERTIES, AND AT A LOWER COST. THESE REQUIREMENTS CAN BE MET BY A SCINTILLATOR BASED ON TLCL. THIS HOST HAS A COMPARABLE DENSITY (7.0 G/CM3 VS. 7.4 G/CM3 FOR LSO) AND A MUCH HIGHER EFFECTIVE Z OF 77 (VS. 65 TO LSO.) WHEN DOUBLE DOPED WITH BE AND I, IT EXHIBITS A FAST DONOR-ACCEPTOR TYPE LUMINESCENCE WITH DECAY TIME <10 NS ALONG WITH ULTRA-FAST CHERENKOV COMPONENT (VS. 40 NS OF LSO.) WHILE THE MATERIAL’S LIGHT YIELD IS LOW, ITS TIMING PROPERTIES ARE EXCELLENT WITH BETTER THAN 300 PS RESOLUTION FWHM WHEN PAIRED WITH LYSO, WITHOUT ANY SIGNIFICANT OPTIMIZATION. THE ONLY PROPERTY WHERE THE MATERIAL IS DEFICIENT IS ITS ENERGY RESOLUTION (>15% AT 511 KEV, DUE TO ITS LOW LIGHT YIELD). THE KEY METRIC FOR TLCL IS ALSO ITS VERY LOW COST, IN TERMS OF RAW MATERIALS AS WELL AS PRODUCTION – ESTIMATED 50% LOWER THAN LSO. IN THIS PROJECT, WE PLAN TO OPTIMIZE THE DOPING CONTENT OF TLCL TO MAXIMIZE ITS SCINTILLATION PROPERTIES AND ACHIEVE ENERGY RESOLUTION OF ABOUT 10% AT 511 KEV AND TIMING RESOLUTION OF 200 PS. IN PHASE II WE WILL INCREASE THE VOLUMES OF PRODUCED MATERIAL, DEVELOP COST REDUCTION SCHEMES, AND PRODUCE AND EVALUATE PET DETECTION MODULES WITH THE SAME PERFORMANCE GOALS. | $1.5M | FY2021 | Jul 2021 – May 2024 |

BRAIN PETS WITH A SPATIAL RESOLUTION APPROACHING 1 MM AT THE CENTER OF THE FIELD-OF-VIEW (FOV) ARE UNDER DEVELOPMENT USING LYSO OR LSO SCINTILLATORS, WHICH ARE THE CURRENTLY AVAILABLE SCINTILLATORS THAT CAN PROVIDE THE BEST OVERALL PERFORMANCE. HOWEVER, TO OBTAIN A HIGH SENSITIVITY, WHICH STRONGLY AFFECTS THE QUANTITATIVE PRECISION OF PET IMAGING, L(Y)SO WITH A THICKNESS OF AROUND 20 MM IS WIDELY USED. THE FIRST AND THE MAJOR DRAWBACK OF USING THE THICK L(Y)SO CRYSTAL IS THE PARALLEL ERROR, WHICH DECREASES THE SPATIAL RESOLUTION OFF THE CENTER OF THE FOV. DEPTH-OF-INTERACTION (DOI) INFORMATION CAN BE USED TO IMPROVE SPATIAL RESOLUTION OFF THE CENTER OF THE FOV, HOWEVER, THE BEST DOI RESOLUTION FROM 20 MM THICK L(Y)SO IS ~2.0 MM OBTAINED USING THE DUAL-ENDED READOUT, WHICH IS NOT GOOD ENOUGH. THE SECOND DRAWBACK OF THE L(Y)SO IS THE HIGH COST. FOR MOST SCANNERS, THE L(Y)SO COST IS HALF OF THE TOTAL SCANNER COST. THE PERFORMANCE OF BRAIN PET SCANNERS CAN BE IMPROVED USING A NEW SCINTILLATOR BASED ON LU2O3 (LO), WHICH HAS A HIGHER DENSITY (9.4 VS. 7.4 G/CM3 FOR LSO) AND HIGHER EFFECTIVE Z (68 VS. 65 FOR LSO). WHEN DOPED WITH LA3+ AND YB3+, IT EXHIBITS AN ULTRA-FAST RISE TIME (<0.6NS) COMBINED WITH PROMPT CHARGE TRANSFER LUMINESCENCE WITH DECAY TIME OF 1.7 NS, GIVING TIMING RESOLUTION (TR) OF 180 PS FWHM. THE ENERGY RESOLUTION AT 511 KEV IS BETTER THAN 10% (>10% FOR LYSO), DUE TO THE GOOD LIGHT YIELD FROM SLOWER COMPONENTS (<20,000 PH/MEV) AND GOOD PROPORTIONALITY (BETTER THAN FOR LYSO). HENCE, LO:LA,YB CAN BE AN EXCELLENT CHOICE FOR A BRAIN PET SCANNER REQUIRING HIGH SENSITIVITY AND HIGH SPATIAL RESOLUTION. THE LO IS PRODUCED USING CERAMIC METHODS WHICH MAY ALSO HAVE AN EDGE OVER THE CRYSTAL GROWTH METHODS IN TERMS OF COST OF MANUFACTURING. THEREFORE, FOR THIS PROJECT, WE PROPOSE TO 1) OPTIMIZE THE LO:LA,YB AND ITS MANUFACTURING FOR HIGH-RESOLUTION AND HIGH-SENSITIVITY BRAIN PET APPLICATIONS, 2) OPTIMIZE LO:LA,YB BASED PET DETECTOR FOR BRAIN PET THROUGH EXPERIMENTAL EVALUATION, AND 3) TO ESTIMATE THE PERFORMANCE OF A FULL BRAIN PET SYSTEM BASED ON THESE RESULTS THROUGH GATE MONTE CARLO SIMULATIONS AND BENCHMARK IT AGAINST LYSO SYSTEM. | $1.2M | FY2023 | Aug 2023 – Sep 2025 |

CAPACITY: BUILD LOCAL CAPACITY OF TRAINED AND CERTIFIED INDIVIDUALS AND FIRMS TO ADDRESS LEAD HAZARDS SAFELY AND EFFECTIVELY DURING LEAD HAZARD CONTROL, RENOVATION, REMODELING, AND MAINTENANCE ACTIVITIES. ANOTHER CORE ELEMENT FOR CAPACITY INCLUDES THE DEVELOPMENT OF COMPREHENSIVE, COMMUNITY-BASED APPROACHES TO INTEGRATING THIS GRANT PROGRAM WITHIN OTHER LOCAL INITIATIVES THROUGH PUBLIC AND PRIVATE PARTNERSHIPS THAT ADDRESS HOUSING RELATED HEALTH AND SAFETY HAZARDS AND/OR SERVE LOW-INCOME FAMILIES WITH CHILDREN UNDER THE AGE OF SIX (6). THIS PROPOSAL AIMS TO VALIDATE AND LEVERAGE A NOVEL MOBILE ELECTROENCEPHALOGRAPHY (EEG) PLATFORM TO PROVIDE ACCESSIBLE, COST-EFFECTIVE COGNITIVE ASSESSMENTS FOR INDIVIDUALS SUFFERING FROM NEUROLOGICAL MANIFESTATIONS OF PASC (NPASC). OUR PRELIMINARY DATA SUGGESTS THAT EEG/EVENT-RELATED POTENTIAL (ERP) BIOSIGNATURES, INCLUDING DELAYED NEURAL RESPONSES DURING AUDITORY AND MEMORY TASKS, ARE SENSITIVE TO COGNITIVE IMPAIRMENTS IN NPASC. HOWEVER, THE TRAJECTORY AND SPECIFICITY OF THESE DEFICITS REMAIN UNCLEAR. THIS STUDY WILL VALIDATE THESE PRELIMINARY FINDINGS IN A LARGER COHORT (N=52) AND LONGITUDINALLY MONITOR CHANGES OVER ONE YEAR TO MAP RECOVERY OR DETECT EARLY SIGNS OF PERSISTENT COGNITIVE DECLINE. MALAYSIA STANDS AS A PIVOTAL HUB IN THE GLOBAL TRADE OF TFT, SERVING AS A SOURCE, TRANSIT POINT, AND CONSUMER MARKET FOR BOTH LEGALLY AND ILLEGALLY TRADED SPECIES, INCLUDING NATIVE AND NON-NATIVE SPECIES MANY OF WHICH ARE HIGHLY THREATENED. THERE ARE 18 SPECIES OF TFT NATIVE TO MALAYSIA, 13 OF WHICH ARE CONSIDERED PRIORITY SPECIES DUE TO THEIR HIGH EXTINCTION RISK (AS LISTED IN TABLE 1. OF THE NOFO). THE TFT PET TRADE POSES A SIGNIFICANT RISK TO THE SURVIVAL OF NATIVE AND NON-NATIVE SPECIES. DRAWING ON SOCIAL SCIENCE AND CRIMINOLOGICAL THEORY, THIS PROJECT EXAMINES THE ILLEGAL TFT TRADE FOR PETS IN MALAYSIA. THE GOAL OF THE PROJECT IS TO CONSIDER THE HUMAN DIMENSION AND MAP POTENTIAL GUARDIANS ALONG THE TRADE CHAIN AND WITHIN THE REGULATORY LANDSCAPE THAT CAN EITHER PASSIVELY OR ACTIVELY PREVENT ILLEGAL TRADE THROUGH THEIR PRESENCE OR ACTIONS. WITH THE ADVENT OF MICROFLUIDICS TECHNOLOGY, NEW METHODS HAVE BEEN INTRODUCED TO GROW TISSUES IN 3D INSIDE PERFUSED CHAMBERS AND PRECISELY CONTROL BIOLOGICAL FACTORS, SUCH AS CELLS, NUTRIENTS AND OXYGEN, AT A SPATIAL AND TEMPORAL LEVEL. THESE MODELS CAN INCORPORATE 3D EXTRACELLULAR MATRICES (ECM) AND PERFUSABLE NEOVASCULATURE, BOTH KEY COMPONENTS OF SOLID TUMORS. BEING OPTICALLY TRANSPARENT, THEY PERMIT EXCELLENT VISUALIZATION OF LIVE CELLS THROUGH ADVANCED OPTICAL MICROSCOPY TECHNIQUES. RADIOLUMINESCENCE MICROSCOPY (RLM) IS A METHOD THAT WAS DEVELOPED TO IMAGE CLINICAL RADIOTRACERS IN LIVE CELLS WITH HIGH SPATIAL RESOLUTION. HOWEVER, THIS METHOD IN ITS CURRENT FORM CANNOT BE USED TO ADEQUATELY IMAGE 3D CELL CULTURES DUE TO THE LOSS OF SPATIAL RESOLUTION AND LACK OF TOMOGRAPHIC CAPABILITIES FOR IMAGING THICK SAMPLES. THE GOAL OF THIS PROJECT IS TO DEVELOP A NOVEL LAYERED SCINTILLATOR DESIGN FOR LIMITED-ANGLE TOMOGRAPHIC IMAGING OF 3D CELL CULTURES AND OTHER IN VITRO TISSUES SUCH AS ORGANOIDS AND TUMOR-CHIPS. THE DUAL-LAYER SCINTILLATOR WILL PROVIDE ANGULAR INFORMATION THAT CAN BE USED FOR 3D RECONSTRUCTION OF RADIOTRACER DISTRIBUTION IN THESE THICK SAMPLES. THUS, SUCH A TECHNOLOGICAL ADVANCE HAS THE POTENTIAL FOR WIDESPREAD USE IN RESEARCH AND MEDICINE USING THE ARSENAL OF EXISTING DIAGNOSTIC AND THERAPEUTIC RADIOISOTOPES. IT COULD BE USED TO BRIDGE THE GAP BETWEEN THESE EMERGENT TUMOR MODELS AND CLINICAL TRIALS, WHICH USE PET BIOMARKERS AS DISEASE ENDPOINTS. IN ADDITION, THE TECHNOLOGY COULD BE USED TO CHARACTERIZE HOW PROPERTIES SPECIFIC TO THE 3D MICROENVIRONMENT SURROUNDING MICROTUMORS COULD AFFECT THE UPTAKE AND RETENTION OF RADIOTRACERS. HIGHER SPATIAL RESOLUTION WILL ALLOW CELLS TO BE PROBED IN SITU, IN DENSE TISSUE SECTIONS. THESE NEW CAPABILITIES WILL BE CRITICAL TO HELP RESEARCHERS DEVELOP PATIENT-DERIVED TUMOR MODELS THAT RECAPITULATE THE MOST SALIENT FEATURES OF SOLID TUMORS AND CAN BE IMAGED USING CLINICALLY RELEVANT PET TRACERS. MAKING WOMEN'S OPTIONS FOR HIV PREVENTION IN TANZANIA ACCESSIBLE AND JOINING IMPLEMENTATION SCIENCE CAPACITY BUILDING (MWOTAJI) - SUMMARY ADOLESCENT GIRLS AND YOUNG WOMEN (AGYW; AGES 15-24) IN SUB-SAHARAN AFRICA FACE A DISPROPORTIONATE BURDEN OF HIV INFECTION. THOUGH HIGHLY EFFECTIVE HIV PREVENTION LIKE PRE-EXPOSURE PROPHYLAXIS (PREP) IS CENTRAL TO THE GLOBAL STRATEGY FOR HIV EPIDEMIC CONTROL, AWARENESS OF PREP AMONG AGYW IS LOW, WITH UPTAKE AND CONTINUATION EVEN LOWER—HEALTH SYSTEMS ARE OFTEN ILL EQUIPPED TO OVERCOME THE NUMEROUS ACCESS BARRIERS FACED BY AGYW, INCLUDING STIGMA, HARMFUL NORMS, AND THE INCONVENIENCES OF CLINIC-BASED HEALTH SERVICES. THERE IS GROWING RECOGNITION THAT COMMUNITY-BASED PHARMACIES HAVE THE POTENTIAL TO BRIDGE GAPS IN HIV PREVENTION AND MITIGATE ACCESS BARRIERS AMONG AGYW. THUS, THE GOAL OF THIS STUDY IS TO EVALUATE THE MALKIA KLABU (“QUEEN CLUB”) IMPLEMENTATION STRATEGY IN TANZANIA, A LOYALTY PROGRAM DESIGNED FOR AND BY AGYW THAT CREATES YOUTH-FRIENDLY PHARMACIES WHERE AGYW CAN ACCESS HIV PREVENTION AND SEXUAL AND REPRODUCTIVE HEALTH (SRH) SERVICES, WITH STRONG LINKAGES TO FACILITY-BASED CARE. BASED ON PROMISING DATA FROM OUR PILOT STUDY, WE HYPOTHESIZE THAT THE CREATION OF A WELCOMING AND SAFE PHARMACY ENVIRONMENT THROUGH THE MALKIA KLABU IMPLEMENTATION MODEL WILL ATTRACT AGYW AND BOLSTER UPTAKE OF PREP, HIV SELF-TESTING (HIVST), AND OTHER SRH SERVICES, THEREBY EMPOWERING AGYW TO AVOID HIV AND UNINTENDED PREGNANCY. WE HAVE DESIGNED AN INNOVATIVE, 5-YEAR IMPLEMENTATION SCIENCE STUDY TO SCALE AND EVALUATE SEVERAL MODELS OF MALKIA KLABU’S PHARMACY-BASED APPROACH FOR PREP AND HIVST DELIVERY IN FIVE RESEARCH SITES IN LAKE ZONE, TANZANIA USING THE RE-AIM FRAMEWORK. THIS RESEARCH WILL BE ENHANCED BY IMPLEMENTATION SCIENCE TRAINING OF LOCAL RESEARCHERS AND POLICYMAKERS TO OPTIMIZE MALKIA KLABU AND BENEFIT FUTURE HIV PREVENTION STRATEGIES. WE WILL FIRST CONDUCT A TYPE 3 HYBRID IMPLEMENTATION-EFFECTIVENESS STUDY OF FOUR PHARMACY-BASED DIFFERENTIATED PREP DELIVERY MODELS OF MALKIA KLABU IN 50 PHARMACIES, EXAMINING IMPLEMENTATION OUTCOMES BEFORE AND AFTER THE ADDITION OF USER COST-SHARING WHICH IS ESSENTIAL FOR REAL-WORLD SUSTAINABILITY (AIM 1). WE WILL EXAMINE THE SUCCESS OF THE MALKIA KLABU DELIVERY MODELS ON EFFECTIVE USE OF PREP AMONG A COHORT OF 575 AGYW, REACH OF THE PROGRAM IN A CROSS-SECTIONAL HOUSEHOLD SURVEY WITH 750 AGYW IN STUDY AREAS; PHARMACY EXIT SURVEYS WITH ~200 AGYW WILL HELP DESCRIBE THE SUCCESS OF PHARMACY- BASED PREP SERVICE OFFERINGS (AIM 2). A CERTIFICATE IN IMPLEMENTATION SCIENCE COUPLED WITH SEED GRANTS FOR GRADUATES WILL ENSURE THE PROJECT HAS LASTING IMPACT (AIM 3). AT THE STUDY’S CONCLUSION, WE WILL DETERMINE THE STRONGEST AND MOST SUSTAINABLE VERSION OF THE MALKIA KLABU IMPLEMENTATION STRATEGY FOR PHARMACY-BASED PREP AND HIVST DELIVERY TO AGYW, AND LAUNCHED AN IMPLEMENTATION SCIENCE TRAINING PROGRAM TO ENABLE LOCALLY-LED SCALING AND ADAPTATION TO REACH VULNERABLE AGYW ACROSS TANZANIA AND THE REGION. THE PROJECT IS A COLLABORATION BETWEEN MZUMBE UNIVERSITY, HEALTH FOR A PROSPEROUS NATION, THE TANZANIA MINISTRY OF HEALTH, AND UNIVERSITY OF CALIFORNIA (SAN FRANCISCO & BERKELEY).

SEMICONDUCTOR DETECTOR FOR LOW-DOSE, HIGH-RESOLUTION DENTAL DR IMAGING - RESEARCH & RELATED OTHER PROJECT INFORMATION – PROJECT SUMMARY/ABSTRACT DENTAL CARIES IS A PERSISTENT PUBLIC HEALTH PROBLEM THAT IMPACTS A LARGE FRACTION OF THE US POPULATION. NEARLY ALL ADULTS IN THE UNITED STATES ARE CONSIDERED AT RISK OF DEVELOPING TOOTH DECAY, AND FOR CHILDREN IT IS A COMMON CHRONIC DISEASE. THOUGH WIDELY AND ROUTINELY USED, THERE ARE ALWAYS CONCERNS ABOUT PATIENT EXPOSURE TO X-RAYS DURING RADIOGRAPHIC IMAGING – A CONCERN ESPECIALLY HEIGHTENED WITH CHILDREN. RADIATION MONITORING DEVICES (RMD) PROPOSES TO DEVELOP AN IMPROVED DIGITAL X-RAY DETECTOR FOR DENTAL RADIOGRAPHY THAT WILL RETAIN ALL THE BENEFITS OF A CURRENT DIGITAL TECHNOLOGY, WHILE IMPROVING SENSITIVITY AND REDUCING PATIENT DOSE. THE DEVICE WILL INVOLVE THE INTEGRATION OF TWO TECHNOLOGIES - SILICON BASED IMAGING ARRAYS AND SEMICONDUCTOR X-RAY CONVERSION LAYERS. THE SILICON CAPABILITIES ARE WELL UNDERSTOOD, WITH CCD AND CMOS DEVICES WELL ESTABLISHED IN DENTAL IMAGING. THE APPLICATION OF A SEMICONDUCTOR CONVERSION LAYER IS THE MORE INNOVATIVE AND CHALLENGING ASPECT OF THE CONCEPT. RMD'S INNOVATION IS TO REPLACE THE COMMONLY USED CSI OR AMORPHOUS SELENIUM X-RAY RECEPTORS WITH THE SEMICONDUCTOR THALLIUM BROMIDE (TLBR). TLBR WILL PROVIDE A MUCH GREATER DOSE EFFICIENCY AND HIGHER IMAGE CONTRAST. THE ADVANTAGE TO DENTAL IMAGING IS THAT DENTISTS WILL GET THE HIGH RESOLUTION IMAGES THAT THEY ARE ACCUSTOMED TO, BUT NOW WITH A MUCH LOWER DOSE TO THE PATIENT. DURING PHASE I, RMD WILL DEVELOP THE PROCESSES TO FABRICATE HIGH QUALITY FILMS OF TLBR AND COMPARE THE TECHNOLOGY TO EXISTING DETECTOR FILMS TO DEMONSTRATE THE SUPERIORITY OF TLBR. THE WORK PLAN WILL ENCOMPASS FILM DEPOSITION, INVESTIGATION OF COMPATIBLE ELECTRODE LAYERS TO WORK IN CONJUNCTION WITH THE TLBR, IDENTIFICATION AND MITIGATION STRATEGIES OF TECHNICAL CHALLENGES THAT ARISE, AND COMPLETION OF A WIDE RANGE OF ELECTRICAL AND PHYSICAL TESTS TO GAUGE X-RAY SENSITIVITY AND IMAGE QUALITY. IN PHASE II RMD WILL OPTIMIZE THE PROCESSES, FABRICATE X-RAY IMAGING DETECTORS ON KA IMAGING CMOS ROICS AND FULLY CHARACTERIZE A COMPLETE DETECTOR SYSTEM. THE RESULT OF THIS RESEARCH WILL BE A NEW DIGITAL RADIOGRAPHY DETECTOR WITH COMPARABLE OR BETTER IMAGE QUALITY AND SIGNIFICANTLY LOWER PATIENT DOSE THAN CURRENTLY USED DETECTORS.

AT PRESENT, MOST PET SYSTEMS USE CRYSTALS OF LSO (LU2SIO5:CE) OR ITS ANALOG LYSO, WHICH SATISFY MANY OF THE LISTED REQUIREMENTS. BUT LSO, AFTER YEARS OF DEVELOPMENT, HAS REACHED ITS PERFORMANCE LIMIT, ESPECIALLY FOR THE SCANNERS WITH A LONG AXIAL FIELD OF VIEW (AFOV) THAT ARE CURRENTLY BEING DEVELOPED. THE GOAL OF THESE SCANNERS IS TO INCREASE THE GEOMETRICAL COVERAGE AND SIGNIFICANTLY INCREASE DETECTION SENSITIVITY (BY A FACTOR OF 30-40), THEREBY REDUCING THE SCANNING TIMES (30-40 TIMES FASTER) OR THE PATIENT’S RADIATION EXPOSURE. HOWEVER, LONG AFOV SCANNERS FACE TWO MAIN CHALLENGES: GREATER DEPTH-OF-INTERACTION (DOI) EFFECTS, WHICH INCREASE BLURRING AND NOISE; AND AN INCREASE IN THE VOLUME REQUIRED FOR THE CONSTITUENT CRYSTALS, WHICH MAKE UP SOME 50% OF THE COST OF THE ENTIRE SCANNER. THE USE OF SHORTER CRYSTALS CAN COUNTERACT BOTH THE DOI EFFECTS AND THE INCREASED CRYSTAL VOLUME (HENCE COST), BUT WITH A MAJOR LOSS OF DETECTION EFFICIENCY, DEFEATING THE ORIGINAL PURPOSE. ANOTHER APPROACH FOR REDUCING DOI EFFECTS IS A DOUBLE-ENDED READ-OUT BUT THIS INCREASES BOTH COST AND SYSTEM COMPLEXITY. THEREFORE, TO ACHIEVE VIABLE AND AFFORDABLE LONG AFOV SCANNERS, A NEW SCINTILLATION MATERIAL IS REQUIRED THAT WOULD PROVIDE HIGHER STOPPING POWER THAN LSO, WITH SIMILAR OR BETTER TIMING PROPERTIES, AND AT A LOWER COST. IN PHASE II WE WILL INCREASE THE VOLUMES OF PRODUCED MATERIAL, DEVELOP COST REDUCTION SCHEMES, AND PRODUCE AND EVALUATE PET DETECTION MODULES WITH THE SAME PERFORMANCE GOALS.

FOR HUMAN BRAIN STUDIES, PET OFFERS A SENSITIVE AND NONINVASIVE WINDOW INTO BRAIN METABOLISM AND NEUROCHEMISTRY, PROVIDING INFORMATION FROM LIVING HUMAN SUBJECTS THAT CANNOT BE OBTAINED USING ANY OTHER TECHNIQUE. THE KEY CHARACTERISTICS OF BRAIN PET ARE SPATIAL RESOLUTION AND SENSITIVITY. BRAIN PETS WITH A SPATIAL RESOLUTION APPROACHING 1 MM AT THE CENTER OF THE FIELD-OF-VIEW (FOV) ARE UNDER DEVELOPMENT USING LYSO OR LSO SCINTILLATORS, WHICH ARE THE CURRENTLY AVAILABLE SCINTILLATORS THAT CAN PROVIDE THE BEST OVERALL PERFORMANCE. HOWEVER, TO OBTAIN A HIGH SENSITIVITY, WHICH STRONGLY AFFECTS THE QUANTITATIVE PRECISION OF PET IMAGING, L(Y)SO WITH A THICKNESS OF AROUND 20 MM IS WIDELY USED. THE FIRST AND THE MAJOR DRAWBACK OF USING THE THICK L(Y)SO CRYSTAL IS THE PARALLEL ERROR, WHICH DECREASES THE SPATIAL RESOLUTION OFF THE CENTER OF THE FOV. DEPTH-OF-INTERACTION (DOI) INFORMATION CAN BE USED TO IMPROVE SPATIAL RESOLUTION OFF THE CENTER OF THE FOV, HOWEVER, THE BEST DOI RESOLUTION FROM 20 MM THICK L(Y)SO IS ~2.0 MM OBTAINED USING THE DUAL-ENDED READOUT, WHICH IS NOT GOOD ENOUGH. THE SECOND DRAWBACK OF THE L(Y)SO IS THE HIGH COST. FOR MOST SCANNERS, THE L(Y)SO COST IS HALF OF THE TOTAL SCANNER COST. THE PERFORMANCE OF BRAIN PET SCANNERS CAN BE IMPROVED USING A NEW SCINTILLATOR BASED ON LU2O3 (LO), WHICH HAS A HIGHER DENSITY (9.4 VS. 7.4 G/CM3 FOR LSO) AND HIGHER EFFECTIVE Z (68 VS. 65 FOR LSO). WHEN DOPED WITH LA3+ AND YB3+, IT EXHIBITS AN ULTRA-FAST RISE TIME (<0.6NS) COMBINED WITH PROMPT CHARGE TRANSFER LUMINESCENCE WITH DECAY TIME OF 1.7 NS, GIVING TIMING RESOLUTION (TR) OF 180 PS FWHM. THE ENERGY RESOLUTION AT 511 KEV IS BETTER THAN 10% (>10% FOR LYSO), DUE TO THE GOOD LIGHT YIELD FROM SLOWER COMPONENTS (<20,000 PH/MEV) AND GOOD PROPORTIONALITY (BETTER THAN FOR LYSO). HENCE, LO:LA,YB CAN BE AN EXCELLENT CHOICE FOR A BRAIN PET SCANNER REQUIRING HIGH SENSITIVITY AND HIGH SPATIAL RESOLUTION. THE LO IS PRODUCED USING CERAMIC METHODS WHICH MAY ALSO HAVE AN EDGE OVER THE CRYSTAL GROWTH METHODS IN TERMS OF COST OF MANUFACTURING. THEREFORE, FOR THIS PROJECT, WE PROPOSE TO 1) OPTIMIZE THE LO:LA,YB AND ITS MANUFACTURING FOR HIGH-RESOLUTION AND HIGH-SENSITIVITY BRAIN PET APPLICATIONS, 2) OPTIMIZE LO:LA,YB BASED PET DETECTOR FOR BRAIN PET THROUGH EXPERIMENTAL EVALUATION, AND 3) TO ESTIMATE THE PERFORMANCE OF A FULL BRAIN PET SYSTEM BASED ON THESE RESULTS THROUGH GATE MONTE CARLO SIMULATIONS AND BENCHMARK IT AGAINST LYSO SYSTEM.

IN ADDITION, THERE IS HEALTHY HOMES SUPPLEMENTAL FUNDING AVAILABLE THAT IS INTENDED TO ENHANCE THE LEAD-BASED PAINT HAZARD CONTROL ACTIVITIES BY COMPREHENSIVELY IDENTIFYING AND ADDRESSING OTHER HOUSING HAZARDS THAT AFFECT OCCUPANT HEALTH. INFORMATION ABOUT WHERE THE SUPPLEMENTAL FUNDING CAN BE USED CAN BE FOUND AT. TARGETED UNITS: TARGET LEAD HAZARD CONTROL EFFORTS IN HOUSING UNITS WHERE CHILDREN LESS THAN 6 YEARS OF AGE ARE AT GREATEST RISK OF LEAD POISONING (PRE-1960, AND, ESPECIALLY, PRE-1940 CONSTRUCTION), WHICH HAS HISTORICALLY INCLUDED CHILDREN IN LOW-INCOME AND MINORITY NEIGHBORHOODS, TO REDUCE THE LIKELIHOOD OF ELEVATED BLOOD LEAD LEVELS IN THESE CHILDREN. B. COST EFFECTIVENESS: UTILIZE COST-EFFECTIVE LEAD HAZARD CONTROL METHODS AND APPROACHES THAT ENSURE THE LONG-TERM SAFETY OF THE BUILDING OCCUPANTS. C. CAPACITY: BUILD LOCAL CAPACITY OF TRAINED AND CERTIFIED INDIVIDUALS AND FIRMS TO ADDRESS LEAD HAZARDS SAFELY AND EFFECTIVELY DURING LEAD HAZARD CONTROL, RENOVATION, REMODELING, AND MAINTENANCE ACTIVITIES. ANOTHER CORE ELEMENT FOR CAPACITY INCLUDES THE DEVELOPMENT OF COMPREHENSIVE, COMMUNITY-BASED APPROACHES TO INTEGRATING THIS GRANT PROGRAM WITHIN OTHER LOCAL INITIATIVES THROUGH PUBLIC AND PRIVATE PARTNERSHIPS THAT ADDRESS HOUSING RELATED HEALTH AND SAFETY HAZARDS AND/OR SERVE LOW-INCOME FAMILIES WITH CHILDREN UNDER THE AGE OF SIX (6). D. AFFIRMATIVE MARKETING: ESTABLISH AND IMPLEMENT A DETAILED PROCESS OF MONITORING AND ENSURING THAT UNITS MADE LEAD-SAFE ARE AFFIRMATIVELY MARKETED, AND PRIORITY GIVEN, TO FAMILIES WITH CHILDREN UNDER AGE 6 YEARS FOR NOT LESS THAN THREE YEARS. E. DATA COLLECTION: GATHER PRE- AND POST-TREATMENT DATA THAT SUPPORTS AND VALIDATES LEAD HAZARD CONTROL INVESTMENTS. PROGRAM DATA COLLECTED SHOULD SUPPORT THE EVALUATION OF GRANT PROGRAM ACTIVITIES AND OUTCOMES. F. TARGETED OUTREACH AND EDUCATION: CONDUCTING TARGETED OUTREACH, AFFIRMATIVE MARKETING, EDUCATION OR OUTREACH PROGRAMS ON LEAD HAZARD CONTROL AND LEAD POISONING PREVENTION DESIGNED TO INCREASE THE ABILITY OF THE APPLICANT TO DELIVER THE SPECIFIED LEAD HAZARD CONTROL SERVICES THROUGH THIS PROGRAM; INCLUDING EDUCATING OWNERS OF ELIGIBLE RENTAL PROPERTIES, TENANTS, AND OTHERS ON THE BENEFITS AND EXPECTATIONS OF PARTICIPATING IN THIS PROGRAM PROVIDED BY "TITLE X" OF THE RESIDENTIAL LEAD-BASED PAINT HAZARD REDUCTION ACT OF 1992.; EXPECTED OUTCOMES: TO IDENTIFY AND CLEAN UP DANGEROUS LEAD IN LOW-INCOME FAMILIES’ HOMES WHERE LOW-INCOME FAMILIES WERE CHILDREN 6 AND UNDER RESIDE. THESE INVESTMENTS WILL PROTECT FAMILIES AND CHILDREN BY TARGETING SIGNIFICANT LEAD AND HEALTH HAZARDS IN OVER 3,700 LOW-INCOME HOMES FOR WHICH OTHER RESOURCES ARE NOT AVAILABLE.; INTENDED BENEFICIARIES: TO ASSIST STATES, CITIES, COUNTIES/PARISHES, NATIVE AMERICAN TRIBES OR OTHER UNITS OF LOCAL GOVERNMENT IN UNDERTAKING COMPREHENSIVE PROGRAMS TO IDENTIFY AND CONTROL LEAD-BASED PAINT HAZARDS IN ELIGIBLE PRIVATELY-OWNED RENTAL OR OWNER-OCCUPIED HOUSING POPULATIONS WERE CHILDREN UNDER 6 RESIDE.; SUBRECIPIENT ACTIVITIES: THE SUBRECIPIENT ACTIVITIES ARE UNKNOWN AT THE TIME OF AWARD.

THIS PROJECT AIMS TO ADDRESS ISSUES CONNECTED TO ILLEGAL AND UNSUSTAINABLE SONGBIRD TRADE THAT CAN HAVE DEVASTATING IMPACTS ON WILD SONGBIRD POPULATIONS. THE GOAL OF THE PROJECT IS TO ENSURE THAT INTERNATIONAL TRADE IN SONGBIRDS FOR USE IN SINGING COMPETITIONS IN CANADA AND THE NETHERLANDS IS NOT A THREAT TO WILD SONGBIRDS IN THE GUIANA SHIELD AND THE CARIBBEAN. THE PROJECT HAS FOUR INTERCONNECTED OBJECTIVES: 1) UNDERSTAND THE RELEVANT REGULATORY LANDSCAPE AND INTERNATIONAL TRADE CHAIN FOR SONGBIRD SPECIES NATIVE TO THE GUIANA SHIELD AND CARIBBEAN USED IN SINGING COMPETITIONS IN CANADA 2) UNDERSTAND DEMAND FOR SONGBIRD SPECIES NATIVE TO THE GUIANA SHIELD AND THE CARIBBEAN USED IN SINGING COMPETITIONS IN CANADA AND 3) UNDERSTAND THE REGULATORY AND DEMAND FOR SONGBIRD SPECIES IN THE NETHERLANDS BY SUPPORTING RESEARCH WITH THE INTERNATIONAL FUND FOR ANIMAL WELFARE. TO ACCOMPLISH THESE OBJECTIVES THE PROJECT INCORPORATES RESEARCH ON THE HUMAN DIMENSIONS ASSOCIATED WITH COMMUNITIES THAT INTERSECT WITH THE SONGBIRD TRADE TO BETTER UNDERSTAND THE DRIVERS AND MECHANISMS OF INTERNATIONAL TRADE AND PLACE THEM WITHIN THE APPROPRIATE CULTURAL FRAMEWORKS. THE PROJECT TAKES AN INCLUSIVE CONSERVATION APPROACH THAT WILL WORK WITH COMMUNITIES TO BE CONSCIOUS AND RESPECTFUL OF DIVERSE CULTURES LANGUAGES AND MARGINALIZED GROUPS THROUGHOUT THE RESEARCH PROCESS.

MIGRANTS AND RETURNEES WHO MOVE BACK TO EL SALVADOR OFTEN STRUGGLE TO REINTEGRATE INTO THE COUNTRY'S LIFE. THEY FREQUENTLY ARRIVE OR RETURN WITH NO MONEY OR JOB PROSPECTS, OFTEN HAVING EXPERIENCED VIOLENCE AND DISCRIMINATION. THE INTER-AMERICAN FOUNDATION (IAF) SUPPORTS COMMUNITY-LED SOLUTIONS THAT AIM TO PROMOTE SOCIAL AND ECONOMIC INCLUSION FOR MARGINALIZED GROUPS. IAF GRANTEE GRUPO DE MONITOREO INDEPENDIENTE DE EL SALVADOR (GMIES) IS A NON-PROFIT ORGANIZATION THAT ADVOCATES FOR FAIR AND EQUAL ACCESS TO LIVELIHOODS FOR MARGINALIZED AND VULNERABLE POPULATIONS IN EL SALVADOR, INCLUDING MIGRANTS AND RETURNED SALVADORANS. GMIES IS WORKING CLOSELY WITH VARIOUS STAKEHOLDERS FROM THE PRIVATE AND LOCAL DEVELOPMENT SECTORS TO CREATE OPPORTUNITIES THAT CAN LEAD TO SUSTAINABLE OUTCOMES FOR MIGRANTS AND RETURNEES IN CENTRAL CHALATENANGO, NORTH CHALATENANGO, AND MASAHUAT IN SANTA ANA. THE ORGANIZATION PROVIDES ASSISTANCE FOR EMPLOYMENT, LEADERSHIP TRAINING, AND ACCESS TO LEGAL AND PSYCHOSOCIAL SUPPORT SERVICES TO IMPROVE ECONOMIC OPPORTUNITIES. CHARACTERIZING NEUROCOGNITIVE DEFICITS IN POST-ACUTE SEQUELAE OF COVID-19 - ABSTRACT POST-ACUTE SEQUELAE OF COVID-19 (PASC), OR LONG COVID, IS AN EMERGING GLOBAL HEALTH CRISIS WITH AN URGENT NEED FOR OBJECTIVE ASSESSMENT TOOLS TO QUANTIFY NEUROCOGNITIVE DEFICITS. THIS PROPOSAL AIMS TO VALIDATE AND LEVERAGE A NOVEL MOBILE ELECTROENCEPHALOGRAPHY (EEG) PLATFORM TO PROVIDE ACCESSIBLE, COST-EFFECTIVE COGNITIVE ASSESSMENTS FOR INDIVIDUALS SUFFERING FROM NEUROLOGICAL MANIFESTATIONS OF PASC (NPASC). OUR PRELIMINARY DATA SUGGESTS THAT EEG/EVENT-RELATED POTENTIAL (ERP) BIOSIGNATURES, INCLUDING DELAYED NEURAL RESPONSES DURING AUDITORY AND MEMORY TASKS, ARE SENSITIVE TO COGNITIVE IMPAIRMENTS IN NPASC. HOWEVER, THE TRAJECTORY AND SPECIFICITY OF THESE DEFICITS REMAIN UNCLEAR. THIS STUDY WILL VALIDATE THESE PRELIMINARY FINDINGS IN A LARGER COHORT (N=52) AND LONGITUDINALLY MONITOR CHANGES OVER ONE YEAR TO MAP RECOVERY OR DETECT EARLY SIGNS OF PERSISTENT COGNITIVE DECLINE. THE INNOVATIVE APPROACH INTEGRATES RESTING-STATE EEG WITH MULTIPLE TASK-BASED ERP PROTOCOLS, ENABLING COMPREHENSIVE PROFILING OF NEURAL DYSFUNCTION ASSOCIATED WITH NPASC. LEVERAGING ADVANCED BRAIN MONITORING'S FDA-CLEARED MOBILE EEG PLATFORM AND VALIDATED ANALYSIS PIPELINES WILL PROVIDE A SCALABLE SOLUTION FOR WIDESPREAD CLINICAL USE AND REMOTE ASSESSMENTS. IF SUCCESSFUL, THIS STUDY WILL DELIVER A QUANTITATIVE EEG/ERP BIOMARKER PROFILE SPECIFIC TO NEURO-PASC, ENABLING OBJECTIVE EVALUATION OF COGNITIVE DEFICITS. LONGITUDINAL MONITORING WILL ELUCIDATE THE NATURAL TRAJECTORY, IDENTIFY POTENTIAL RISK FACTORS FOR PERSISTENT IMPAIRMENT, AND PROVIDE OUTCOME MEASURES FOR FUTURE THERAPEUTIC INTERVENTIONS. THIS INNOVATIVE DIAGNOSTIC TOOL WILL EMPOWER PATIENTS AND CLINICIANS BY PROVIDING OBJECTIVE EVIDENCE OF NPASC SEVERITY AND TREATMENT EFFICACY.

OUR PROPOSED INITIATIVE PROVIDES A WELL-INFORMED, SUSTAINABLE, FEASIBLE AND IMPACTFUL APPROACH TO REDUCING ILLEGAL TRADE IN TORTOISES AND FRESHWATER TURTLES (TFT) IN MALAYSIA. MALAYSIA STANDS AS A PIVOTAL HUB IN THE GLOBAL TRADE OF TFT, SERVING AS A SOURCE, TRANSIT POINT, AND CONSUMER MARKET FOR BOTH LEGALLY AND ILLEGALLY TRADED SPECIES, INCLUDING NATIVE AND NON-NATIVE SPECIES MANY OF WHICH ARE HIGHLY THREATENED. THERE ARE 18 SPECIES OF TFT NATIVE TO MALAYSIA, 13 OF WHICH ARE CONSIDERED PRIORITY SPECIES DUE TO THEIR HIGH EXTINCTION RISK (AS LISTED IN TABLE 1. OF THE NOFO). THE TFT PET TRADE POSES A SIGNIFICANT RISK TO THE SURVIVAL OF NATIVE AND NON-NATIVE SPECIES. DRAWING ON SOCIAL SCIENCE AND CRIMINOLOGICAL THEORY, THIS PROJECT EXAMINES THE ILLEGAL TFT TRADE FOR PETS IN MALAYSIA. THE GOAL OF THE PROJECT IS TO CONSIDER THE HUMAN DIMENSION AND MAP POTENTIAL GUARDIANS ALONG THE TRADE CHAIN AND WITHIN THE REGULATORY LANDSCAPE THAT CAN EITHER PASSIVELY OR ACTIVELY PREVENT ILLEGAL TRADE THROUGH THEIR PRESENCE OR ACTIONS. BY MAPPING THE TFT SUPPLY CHAIN AND IDENTIFYING POTENTIAL GUARDIANS, INCLUDING FORMAL GUARDIANS SUCH AS LAW ENFORCEMENT AND INFORMAL GUARDIANS SUCH AS COMMUNITY MEMBERS, THIS PROJECT AIMS TO DEVELOP STRATEGIES TO ENHANCE THEIR CAPABILITIES IN COMBATTING ILLEGAL WILDLIFE TRADE. THE PROJECT WILL PRIORITIZE CAPACITYBUILDING ACTIVITIES WITH POTENTIAL GUARDIANS BASED ON URGENCY, FEASIBILITY, AND POTENTIAL IMPACT, ULTIMATELY AIMING TO CREATE A REPLICABLE TEMPLATE FOR MAPPING GUARDIANSHIP ALONG WILDLIFE TRADE CHAINS, WITH THE GOAL OF MAXIMIZING RESOURCE ALLOCATION FOR CRIME PREVENTION EFFORTS. BY EMPHASIZING THE ROLE OF GUARDIANSHIP IN CRIME PREVENTION, THIS PROJECT AIMS TO MOVE THINKING ABOUT BEHAVIOR CHANGE BEYOND CONSUMERS AND OFFENDERS. BASED ON A HOLISTIC CONSERVATION APPROACH THAT INTEGRATES INSIGHTS FROM MULTIPLE DISCIPLES, IT WILL IDENTIFY POINTS OF INTERVENTION WHERE RESOURCES COULD BE MOST EFFECTIVELY UTILIZED TO SUPPORT GUARDIANSHIP CAPABILITY ACROSS STAKEHOLDERS AND INTEREST GROUPS AND REDUCE OPPORTUNITIES FOR ILLEGAL TRADE. THIS PROJECT WILL MAKE AN IMPORTANT LONG-TERM AND SUSTAINABLE IMPACT ON THE ILLEGAL TFT TRADE IN MALAYSIA, AND GLOBALLY.

DIRECTIONAL SENSOR FOR RADIOLUMINESCENCE MICROSCOPY OF NEXT-GENERATION TUMOR MODELS - MICROPHYSIOLOGICAL TUMOR MODELS (ΜPTM) ARE INCREASINGLY USED FOR PRECLINICAL RESEARCH DUE TO THEIR ABILITY TO CLOSELY SIMULATE, IN VITRO, THE PHYSIOLOGY OF SOLID TUMORS. WITH THE ADVENT OF MICROFLUIDICS TECHNOLOGY, NEW METHODS HAVE BEEN INTRODUCED TO GROW TISSUES IN 3D INSIDE PERFUSED CHAMBERS AND PRECISELY CONTROL BIOLOGICAL FACTORS, SUCH AS CELLS, NUTRIENTS AND OXYGEN, AT A SPATIAL AND TEMPORAL LEVEL. THESE MODELS CAN INCORPORATE 3D EXTRACELLULAR MATRICES (ECM) AND PERFUSABLE NEOVASCULATURE, BOTH KEY COMPONENTS OF SOLID TUMORS. BEING OPTICALLY TRANSPARENT, THEY PERMIT EXCELLENT VISUALIZATION OF LIVE CELLS THROUGH ADVANCED OPTICAL MICROSCOPY TECHNIQUES. RADIOLUMINESCENCE MICROSCOPY (RLM) IS A METHOD THAT WAS DEVELOPED TO IMAGE CLINICAL RADIOTRACERS IN LIVE CELLS WITH HIGH SPATIAL RESOLUTION. HOWEVER, THIS METHOD IN ITS CURRENT FORM CANNOT BE USED TO ADEQUATELY IMAGE 3D CELL CULTURES DUE TO THE LOSS OF SPATIAL RESOLUTION AND LACK OF TOMOGRAPHIC CAPABILITIES FOR IMAGING THICK SAMPLES. THE GOAL OF THIS PROJECT IS TO DEVELOP A NOVEL LAYERED SCINTILLATOR DESIGN FOR LIMITED-ANGLE TOMOGRAPHIC IMAGING OF 3D CELL CULTURES AND OTHER IN VITRO TISSUES SUCH AS ORGANOIDS AND TUMOR-CHIPS. THE DUAL-LAYER SCINTILLATOR WILL PROVIDE ANGULAR INFORMATION THAT CAN BE USED FOR 3D RECONSTRUCTION OF RADIOTRACER DISTRIBUTION IN THESE THICK SAMPLES. THUS, SUCH A TECHNOLOGICAL ADVANCE HAS THE POTENTIAL FOR WIDESPREAD USE IN RESEARCH AND MEDICINE USING THE ARSENAL OF EXISTING DIAGNOSTIC AND THERAPEUTIC RADIOISOTOPES. IT COULD BE USED TO BRIDGE THE GAP BETWEEN THESE EMERGENT TUMOR MODELS AND CLINICAL TRIALS, WHICH USE PET BIOMARKERS AS DISEASE ENDPOINTS. IN ADDITION, THE TECHNOLOGY COULD BE USED TO CHARACTERIZE HOW PROPERTIES SPECIFIC TO THE 3D MICROENVIRONMENT SURROUNDING MICROTUMORS COULD AFFECT THE UPTAKE AND RETENTION OF RADIOTRACERS. HIGHER SPATIAL RESOLUTION WILL ALLOW CELLS TO BE PROBED IN SITU, IN DENSE TISSUE SECTIONS. THESE NEW CAPABILITIES WILL BE CRITICAL TO HELP RESEARCHERS DEVELOP PATIENT-DERIVED TUMOR MODELS THAT RECAPITULATE THE MOST SALIENT FEATURES OF SOLID TUMORS AND CAN BE IMAGED USING CLINICALLY RELEVANT PET TRACERS. THE OBJECTIVE OF THIS PHASE I PROJECT IS TO DEMONSTRATE THE FEASIBILITY OF SUCCESSFULLY FABRICATING THIN LAYERS OF A HIGHLY DENSE TRANSPARENT SCINTILLATOR, SEPARATED BY A LAYER OF NON-SCINTILLATING TRANSPARENT MATERIAL. THIS NOVEL DESIGN ENABLES VISUALIZATION OF TWO SCINTILLATION SPOTS SO THAT THE ANGLE OF INCIDENCE CAN BE ESTIMATED TO PROVIDE LIMITED-ANGLE TOMOGRAPHIC PROJECTIONS. THIS INNOVATIVE DESIGN WILL PROVIDE THE SPATIAL RESOLUTION REQUIRED FOR VISUALIZATION OF RADIOTRACER UPTAKE IN 3D CELL CULTURES, MICROTUMORS, AND OTHER THICK SPECIMENS.
