NOVEL APPROACHES FOR TAU PROTAC DRUG DISCOVERY - NOVEL APPROACHES FOR TAU PROTAC DRUG DISCOVERY ALZHEIMER’S DISEASE (AD), FRONTOTEMPORAL DEMENTIA (FTD), PROGRESSIVE SUPRANUCLEAR PALSY (PSP) ARE A GROUP OF NEURODEGENERATIVE DISEASES CHARACTERIZED BY THE PATHOLOGICAL ACCUMULATION OF HYPER-PHOSPHORYLATED TAU (P-TAU) PROTEIN, IN THE FORM OF INTRACELLULAR PAIRED HELICAL FILAMENTS (PHFS) OR NEUROFIBRILLARY TANGLES (NFTS), OR AGGREGATES, WITHIN NEURONS AND GLIA OF AFFECTED BRAIN REGIONS, LEADING TO CELL DEATH. SEVERAL APPROACHES TO REMOVE AGGREGATES OF PATHOGENIC TAU HAVE FAILED, INCLUDING ANTIBODY THERAPY. NOVEL APPROACHES TO REMOVE AGGREGATED PROTEINS ARE DESIRED. TARGETED PROTEIN DEGRADATION BY PROTEOLYSIS TARGETING CHIMERAS (PROTACS) HAS EMERGED AS NOVEL THERAPEUTIC MODALITY. PROTACS ARE HETEROBIFUNCTIONAL SMALL MOLECULES THAT SIMULTANEOUSLY BIND TO A TARGET PROTEIN AND A UBIQUITIN E3 LIGASE, THEREBY LEADING TO UBIQUITINATION AND SUBSEQUENT DEGRADATION OF THE TARGET. THEY PRESENT A NEW OPPORTUNITY TO MODULATE PROTEINS IN A MANNER INDEPENDENT OF ENZYMATIC OR SIGNALING ACTIVITY. PROTAC DRUGS OFFER MANY ADVANTAGES, SUCH AS: 1) TARGET CAN BE SELECTIVELY DEGRADED WITH CATALYTIC SPECIFICITY; 2) WEAK BINDERS CAN BE CONVERTED INTO SELECTIVE PROTAC DRUGS; 3) OVEREXPRESSED OR MUTANT TARGETS CAN BE DEGRADED; AND 4) LIMITED TARGET ENGAGEMENT CAN LEAD TO MAXIMAL DEGRADATION. ALTHOUGH MOST MISFOLDED AND AGGREGATED PROTEINS IN THE HUMAN PROTEOME CAN BE DEGRADED BY PROTEASOMAL SYSTEM, SOME NATIVE AND MUTANT PROTEINS PRONE TO AGGREGATION, SUCH AS TAU OLIGOMERS ARE RESISTANT TO ALL KNOWN PROTEOLYTIC PATHWAYS THUS SUBJECT TO AUTOPHAGIC OR LYSOSOMAL PATHWAY. DEVELOPMENT OF PROTACS THAT CAN EFFICIENTLY DEGRADE PATHOGENIC TAU AGGREGATES IS AN ATTRACTIVE AND BREAKTHROUGH THERAPY FOR AD AND FTD. NO ONE, INDUSTRY OR ACADEMIA HAS SHOWN CREDIBLE DATA FOR TAU PROTACS TO SUCCEED IN THE CLINIC. TRADITIONAL METHODS FOR PROTAC ANALYSIS, WESTERN BLOT OR REPORTER GENE ANALYSIS ARE TIME CONSUMING, CUMBERSOME, AND PRONE TO ARTIFACTS. IN THIS PROPOSAL WE DESCRIBE A HIGH THROUGHPUT METHOD FOR RAPID DEVELOPMENT OF TAU PROTACS. DEVELOPMENT OF PROTACS THAT PROMOTE TAU DEGRADATION BY LYSOSOMAL/AUTOPHAGIC MECHANISM IS CHALLENGING. THIS GRANT PROPOSES STRATEGIES TO SCREENING DIFFERENT E3 UBIQUITIN LIGASES FOR TAU PROTAC THAT WILL REMOVE PATHOGENIC (MONOMERIC) AS WELL AS AGGREGATES THROUGH AUTOPHAGIC MECHANISM. GOAL OF THE PROJECT IS TO ESTABLISH RULES THAT GOVERN PATHOGENIC TAU DEGRADATIONS, SUCH AS POLY-UBIQUITIN SIGNATURES CONJUGATED ON TAU AGGREGATES TO DIFFERENTIATE PROTEASOMAL VS LYSOSOMAL DEGRADATION. DEVELOPMENT OF THE TECHNOLOGY THAT DEGRADES TAU AGGREGATES CAN BE DIRECTLY APPLIED TO PARKINSON’S DISEASE, HUNTINGDON DISEASE AND AMYOTROPHIC LATERAL SCLEROSES.

SBIR FAST TRACK: DEVELOPMENT OF A HIGH-THROUGHPUT MAGNETIC CYTOMETER FOR SINGLE CELL SORTING - PROJECT SUMMARY THIS SMALL BUSINESS INNOVATION RESEARCH PROJECT AIMS TO DEVELOP A HIGH-THROUGHPUT EASY-TO-USE CELL SORTER MAGICYTE™ TO RAPIDLY AND PRECISELY SORT INTACT AND VIABLE RARE CELLS AT SINGLE-CELL RESOLUTION. SINGLE-CELL SORTING FROM A HETEROGENEOUS CELL POPULATION HAS BEEN ESSENTIAL TO LIFE SCIENCE APPLICATIONS SUCH AS MONOCLONAL ANTIBODY PRODUCTION AND CELL LINE DEVELOPMENT. RECENTLY, WITH THE RAPID GROWTH OF PRECISION MEDICINE, THERE HAS ALSO BEEN A STRONG DEMAND FOR SINGLE-CELL SORTING TECHNOLOGIES FOR RARE CELLS, SUCH AS CIRCULATING TUMOR CELLS (CTCS), CIRCULATING FETAL CELLS, STEM CELLS AND ANTIGEN-SPECIFIC T- OR B-CELLS. HOWEVER, COLLECTING SUCH RARE CELLS AT SINGLE-CELL RESOLUTION TO MEET THE REQUIREMENTS OF SINGLE-CELL ANALYSIS IS NOT FEASIBLE FOR CONVENTIONAL SINGLE-CELL SORTING TECHNIQUES. MICROMANIPULATION AND SERIAL DILUTION ARE TIME CONSUMING AND LOW-THROUGHPUT, WHILE FLUORESCENT-ACTIVATED CELL SORTING (FACS) SUFFERS FROM REMARKABLE CELL LOSS AND IS INEFFICIENT IN ISOLATING RARE CELLS. IN ADDITION, MANY SUCH TECHNIQUES REQUIRE CELL FIXATION AND/OR FLUORESCENT STAINING, WHICH DECREASES CELL VIABILITY AND THUS PROHIBITS MANY DOWNSTREAM MOLECULAR ANALYSES, AS WELL AS AND CELL CULTURE. AS SUCH, THERE IS AN UNADDRESSED NEED FOR HIGH THROUGHPUT SINGLE-CELL SORTING OF RARE CELLS. WE AIM TO DEVELOP A HIGH-THROUGHPUT CHIP-BASED CYTOMETER MAGICYTE™ TO RAPIDLY ISOLATE, IDENTIFY AND SORT SINGLE INTACT AND VIABLE TARGET CELLS FROM HETEROGENEOUS CELL POPULATION. THE CORE COMPONENT OF MAGICYTE IS A SEMICONDUCTOR CHIP MAGICHIP™ WHICH IS USED FOR SINGLE CELL IDENTIFICATION AND SINGLE CELL SORTING. IN THIS FAST- TRACK PROJECT, WE WILL FIRST DEVELOP THE CONTROL SOFTWARE AND DEMONSTRATE AUTOMATED SINGLE-CELL SORTING; IN PHASE II, WE WILL DEVELOP THE NEXT-GENERATION MAGICHIP, BUILD THE MAGICYTE BETA PRODUCT AND VALIDATE THE SYSTEM ON CLINICAL SAMPLES WITH OUR COLLABORATORS.

A DRUG DISCOVERY PLATFORM TO PRIORITIZE DEVELOPMENT OF NATURAL PRODUCT NEUROMODULATORS FOR TREATING COGNITIVE IMPAIRMENT - PROJECT SUMMARY MILD COGNITIVE IMPAIRMENT (MCI) IS ONE OF THE EARLIEST AND MOST NOTICEABLE SYMPTOMS ASSOCIATED WITH ALZHEIMER’S DISEASE (AD) AND OTHER AD RELATED DEMENTIAS (ADRD). THERE IS NO CURE FOR ADRD, AND CONVENTIONAL TREATMENTS ONLY TARGET MORE DISEASE STAGES. NOTABLY, MCI ASSOCIATED WITH AD TYPICALLY OCCURS AFTER AD BIOMARKERS (RELATED TO Β-AMYLOID AND TAU PATHOLOGIES) ARE DETECTABLE IN PATIENTS, CREATING A NARROW WINDOW PRIOR TO MCI-AD ONSET AND DIAGNOSIS, TERMED PRECLINICAL AD, WHERE INTERVENTIONS SLOWING MCI ONSET HAVE THE POTENTIAL TO GREATLY IMPROVE PATIENT QUALITY OF LIFE AND DRAMATICALLY REDUCE COSTS OF AD CARE AT LARGE. HOWEVER, DEVELOPING PHARMACEUTICAL TREATMENTS FOR PRECLINICAL AD MUST MEET HIGH SAFETY MARGINS GIVEN THE DURATION OF TREATMENT, AND PAST ATTEMPTS HAVE FAILED TO SHOW EFFICACY. RECENTLY, APPROVALS OF ANTIDEPRESSANTS LIKE KETAMINE AND BREXANOLONE (BOTH 2019) AND THE RESURGENCE OF PSYCHEDELIC RESEARCH HAS HIGHLIGHTED THE POWER OF NEUROPLASTICITY MODIFIERS – COMPOUNDS DRIVING THE CREATION OR MODULATION OF SYNAPTIC CONNECTIONS WITHIN THE BRAIN – FOR MODIFYING NEUROLOGICAL FUNCTION TO IMPACT MENTAL HEALTH. ENGAGING NEUROPLASTICITY MECHANISMS THROUGH PHARMACOLOGICAL MEANS OR HOLISTIC APPROACHES (I.E. EXERCISE & DIETARY CHANGES) HAS BEEN SHOWN TO POSITIVELY ENHANCE COGNITION AND MEMORY. NOTABLY, SOME OF THE MOST POTENT NEUROPLASTICITY MODULATORS ARE NATURAL PRODUCTS (NPS) DERIVED FROM MEDICINAL PLANTS AND HERBS USED BY HUMANS FOR CENTURIES. AS SUCH, NPS HAVE BEEN THE SUBJECT OF NUMEROUS STUDIES INVESTIGATING THEIR POTENTIAL TO IMPROVE COGNITION AND PROGRESSION TO MCI IN AD. HOWEVER, INCOMPLETE DESCRIPTIONS OF NP MOLECULAR TARGETS, MECHANISMS OF ACTION, STABILITY, PURITY, POTENCY AND BIOAVAILABILITY HAVE LARGELY PREVENTED THEIR DEVELOPMENT AS CLINICAL DRUG CANDIDATES. WHAT’S MORE, THE QUANTITY OF KNOWN NPS FOUND IN BOTANICAL PRODUCTS NUMBERS IN THE TENS OF THOUSANDS. TO IDENTIFY AND DEVELOP PSYCHOACTIVE NATURAL PRODUCTS THAT IMPACT NEUROPLASTICITY AND CAN SLOW MCI ONSET, SENSORIUM THERAPEUTICS IS EXPANDING THE CAPACITY OF THEIR BIODYNAMIC DISCOVERY PLATFORMTM (BDP). OUR NOVEL BDPTM LEVERAGES AI-ASSISTED ANALYSIS OF ETHNOBOTANICAL DATA AND MEDICINAL CHEMISTRY COUPLED WITH IN VITRO HIGH-THROUGHPUT SCREENING USING HUMAN IPSCS TO PROVIDE AN EFFECTIVE DRUG DEVELOPMENT PIPELINE FOR IDENTIFYING NOVEL NPS AS MCI-AD TREATMENTS. OUR PLATFORM HAS ALREADY LED TO THE IDENTIFICATION OF A LEADING NATURAL PRODUCT DRUG CANDIDATE FOR DEPRESSION AND ANXIETY. ADDITIONALLY, WE HAVE IDENTIFIED A CLASS OF BOTANICALLY-DERIVED ALKALOID COMPOUNDS THAT HAVE FAVORABLE CNS PHARMACOLOGICAL PROFILES, EXCELLENT CHEMICAL AND IN VITRO SAFETY, AND ARE LINKED TO IMPROVING COGNITIVE FUNCTION IN HUMANS. THE OVERARCHING GOAL OF THIS FAST TRACK PROPOSAL IS TO EXPAND OUR BDPTM TO INCLUDE SCALABLE CELLULAR/NEURONAL READOUTS OF NEUROPLASTICITY WHILE INCORPORATING MCI-AD IN VITRO MODELS (PHASE I) AND IDENTIFY AND VALIDATE NP CANDIDATE EFFICACY FOR MCI-AD TREATMENT (PHASE II). OUR RESEARCH TEAM HAS PREVIOUSLY PILOTED HEAT ISLAND MEASUREMENT PROJECTS FOR CITIZEN SCIENCE RESEARCH INVOLVING COLLEGE STUDENTS AND AS STEM LEARNING EXPERIENCES AT THE HIGH SCHOOL LEVEL. THROUGH THE PILOT PROJECTS, OUR TEAM HAS IDENTIFIED KEY USABILITY HURDLES THAT LIMIT THE WIDE-SPREAD SCALABILITY AND APPLICABILITY OF THESE ACTIVITIES. THIS SBIR FAST-TRACK PHASE I/II PROPOSAL (CALLED HEAT VIZ) AIMS TO BUILD AND TEST AN INTERACTIVE DIGITAL MEDIA PLATFORM TO ENABLE HEAT ISLAND PROJECTS TO BE IMPLEMENTABLE BY HIGH SCHOOLS ACROSS THE COUNTRY. THE GOAL OF THE PROJECT IS FOCUSED ON IMPROVEMENTS OF FOUR STUDENT OUTCOMES 1. CONTENT KNOWLEDGE OF RELEVANT DISCIPLINARY CORE IDEAS AROUND WEATHER, CLIMATE, BIOGEOLOGY, AND HUMAN IMPACTS ON THE EARTH 2. ULTRAFAST AND PRECISE EXTERNAL BEAM MONITOR FOR FLASH AND OTHER ADVANCED RADIATION THERAPY MODALITIES - SUMMARY/ABSTRACT FLASH RADIOTHERAPY (FLASH-RT) IS A NOVEL FORM OF RADIATION THERAPY THAT PROMISES LARGE SPARING OF NORMAL- TISSUES IN CANCER TREATMENT WHILE SHOWING NO TUMOR SPARING. IN FLASH-RT, THE RADIATION DOSE IS DELIVERED TO THE TUMOR AND NORMAL TISSUES IN MILLISECONDS RATHER THAN MINUTES. FLASH-RT IS ONLY EFFECTIVE WHEN GIVEN WITH LARGE DOSES PER FRACTION DELIVERED IN 1-3 TREATMENT SESSIONS. IT WOULD SHORTEN A STANDARD 30-DAY TREATMENT TO 1- 3 DAYS, THUS GREATLY REDUCING SIDE-EFFECTS AND RADIATION THERAPY COSTS TO BOTH THE PATIENT AND SOCIETY. THE FIRST HUMAN PATIENT WAS SUCCESSFULLY TREATED IN 2018. THE FIRST CLINICAL TRIAL WITH TEN (10) PATIENTS STARTED IN THE U.S. IN NOVEMBER 2020. ADDITIONAL CLINICAL TRIALS ARE PLANNED FOR 2021 IN THE U.S. AND IN EUROPE WITH AT LEAST 100 PATIENTS. A MAJOR LIMITATION OF FLASH-RT, PREVENTING A FAST TRANSLATION TO CLINICAL USE, IS THE LACK OF DETECTORS CAPABLE OF MEETING THE REQUIREMENTS NEEDED TO MONITOR AND TERMINATE THE FLASH-RT TREATMENT IN REAL TIME. WE PROPOSE TO DEVELOP AND DEMONSTRATE A LARGE AREA, ULTRAFAST AND PRECISE EXTERNAL BEAM MONITOR FOR FLASH-RT, UNIVERSALLY SUITABLE FOR ELECTRONS, PROTONS, PHOTONS, AND IONS, THAT CAN TERMINATE THE BEAM IN =1 MS WHILE THE PATIENT IS BEING TREATED. FOR THIS PROPOSAL, WE WILL PRIMARILY FOCUS ON DEVELOPING AND DEMONSTRATING THE SYSTEM FOR ELECTRON FLASH-RT WITH LINACS AND PROTON FLASH-RT WITH EXISTING CYCLOTRONS, BUT WILL ALSO DEMONSTRATE PERFORMANCE USING X-RAYS. UNLIKE STRIP OR WIRE IONIZATION CHAMBERS, THE PROPOSED SYSTEM IS BASED ON OUR PATENTED (JAN 2020 AND NOV 2020) IONIZING-RADIATION BEAM MONITORING SYSTEM TECHNOLOGY, WHICH CAN PROVIDE ULTRAFAST READOUT WITH CONCURRENT ANALYSIS OF THE RADIATION BEAM POSITION, PROFILE, AND FLUENCE/DOSIMETRY IN REAL TIME AT A RATE OF =10 KHZ (I.E., BEAM ANALYSIS =100 ΜS). THE PROPOSED SYSTEM PROVIDES REAL-TIME DOSIMETRY, BEAM CONTROL, AND VERIFICATION FOR FLASH-RT. IT PROVIDES AN ACCURATE 2D POSITION AND BEAM PROFILE OF RAPIDLY SCANNED BEAMS WITH A SPATIAL RESOLUTION OF A FEW MICRONS OVER AN ACTIVE BEAM MONITORING AREA OF 26 CM X 30 CM. THE BEAM MONITOR RESPONSE IS LINEAR, WITHOUT SATURATION, FOR ALL FLASH-RT BEAM LUMINOSITIES. PROTON BEAM TESTING WILL BE PRIMARILY AT THE UNIVERSITY OF MICHIGAN ION BEAM LABORATORY, AND ELECTRON BEAM TESTING AT THE NOTRE DAME RADIATION LABORATORY. THE PROPOSED PROGRAM IS FOR 3-YEARS AND WILL EVOLVE FROM FABRICATION AND TESTING OF A QUARTER-SCALE BEAM MONITOR IN YEAR 1 TO A FULL-SIZE SYSTEM WITH SELF-CALIBRATION CAPABILITY IN YEAR 3. OUR PRINCIPAL COLLABORATORS ON THIS PROGRAM INCLUDE THE UNIVERSITY OF MICHIGAN, PHYSICS DEPARTMENT, AND LOMA LINDA UNIVERSITY, SCHOOL OF MEDICINE. THE PROPOSED BEAM MONITOR CONSTITUTES A CRITICAL ENABLING TECHNOLOGY FOR ALL TYPES OF FLASH-RT. IT WILL ENSURE THE SAFETY, QUALITY, AND EFFICIENCY OF FLASH RADIATION THERAPY, ALLOWING CANCER PATIENTS TO BE SUCCESSFULLY TREATED WITH MUCH HIGHER DOSES, FEWER SIDE-EFFECTS, AND EXCELLENT TUMOR CONTROL. IT IS ALSO SUITABLE FOR SPATIALLY FRACTIONATED RADIATION THERAPY TECHNIQUES SUCH AS GRID, LATTICE, MICROBEAM RT (MRT), AND PROTON-MINIBEAM RT (PMBRT). PROTEASOMAL DEGRADATION OF TARGET PROTEINS IS A WELL-ESTABLISHED MEANS WHEREBY THE UB PROTEASOME SYSTEM (UPS) CONTROLS PROTEIN CONTENT. ENZYMES CALLED UB E3 LIGASES CONJUGATE UB TO TARGET PROTEINS BY FORMING AN ISOPEPTIDE BOND BETWEEN THE E- AMINO GROUP OF THE TARGET PROTEIN LYSINE AND THE CARBOXYLATE OF THE C-TERMINAL GLYCINE OF UB. CONJUGATION OF MULTIPLE UBS FORMS POLY-UB CHAINS AT ANY OF ITS SEVEN LYSINES (K), AND K6, K11, K27, K29, K33, K48, AND K63 UB CHAINS HAVING VARIOUS ROLES ARE PRESENT IN ALL TISSUES. RECENTLY, SEVERAL GROUPS DESIGNED SMALL MOLECULES THAT BIND TO AN E3 LIGASE AT ONE END AND A TARGET PROTEIN AT THE OTHER, PHYSICALLY FACILITATING UBIQUITYLATION OF THE TARGET PROTEIN, WHICH IS THEN DEGRADED. THIS HIJACKING OF A LIGASE TO UBIQUITYLATE A DESIRED PROTEIN HAS LAUNCHED A NEW CLASS OF DRUG CALLED PROTACS (PROTEOLYSIS TARGETING CHIMERAS). PROTAC-BASED APPROACHES FOR THERAPEUTICS OFFER SEVERAL ADVANTAGES: 1) SELECTIVE, CATALYTIC DEGRADATION OF THE TARGET; 2) CONVERSION OF WEAK BINDERS INTO SELECTIVE PROTAC DRUGS; 3) DEGRADATION OF OVEREXPRESSED OR MUTANT TARGETS; AND 4) MAXIMAL DEGRADATION FROM LIMITED TARGET ENGAGEMENT. TO DATE, CEREBLON AND VHL LIGASE BINDERS HAVE BEEN MOST COMMONLY USED AS VEHICLES TO UBIQUITYLATE TARGET PROTEINS SUCH AS NUCLEAR RECEPTORS, KINASES, TRANSCRIPTION FACTORS, AND NEURONAL PROTEINS TAU AND A-SYNUCLEIN. A MAJOR PROBLEM HAS HINDERED DEVELOPMENT OF NEW PROTAC DRUGS, HOWEVER. CHEMICAL OPTIMIZATION OF PROTAC MOLECULES DEPENDS ON RAPID EVALUATION OF SYNTHESIZED COMPOUNDS TO GUIDE THE SYNTHETIC STRATEGY FOR PRODUCING DRUG CANDIDATE MOLECULES. ASSAYS CURRENTLY AVAILABLE ARE LABOR INTENSIVE AND DO NOT PROVIDE RESULTS TO THE MEDICINAL CHEMISTS FAST ENOUGH – OFTEN, A WEEK IS REQUIRED. IN PHASE I, A FACILE IN VITRO METHOD EMPLOYING UB LIGASES CEREBLON AND HDM2 WAS DEVELOPED TO SCREEN FOR POTENTIAL PROTAC DRUGS; PROTAC-MEDIATED UBIQUITYLATION OF SELECTED PROTEINS WAS RECAPITULATED IN VITRO IN A WAY THAT MIMICS OBSERVED PROTAC-DEPENDENT UBIQUITINATION AND DEGRADATION OF THESE PROTEINS IN VIVO, ACHIEVING THE AIMS OF PHASE I. IN PHASE II, THE UTILITY OF THIS METHOD WILL BE EXPANDED TO INCLUDE REPRESENTATIVE MEMBERS OF ALL UB LIGASE FAMILIES (CULLIN FAMILIES, RING FINGER LIGASES, HECT FAMILY LIGASES, AND SUMO LIGASE), INCREASING THE BIOCHEMICAL AND CHEMICAL SPACE FOR PROTAC DRUG DISCOVERY. TO SCALE UP PROTAC SCREENS, A MICROTITER PLATE-BASED, HIGH THROUGHPUT METHOD WILL BE ESTABLISHED TO MONITOR IN VITRO PROTAC DRUG DISCOVERY, AND BIOCHEMICAL AND UB MASS SPEC PROTEOMICS WILL BE EMPLOYED TO DEMONSTRATE THAT TARGET PROTEIN LYSINES UBIQUITYLATED IN VITRO ARE CORRELATED WITH IN VIVO PROTAC MEDIATED DEGRADATION OF TARGET PROTEINS. CITIZEN-SCIENCE TECHNOLOGY FOR HEALTHY LIVING AT HOME - ABSTRACT HUMAN EXPOSURE TO AIR POLLUTION IS THE LEADING ENVIRONMENTAL RISK FACTOR FOR PREMATURE MORBIDITY AND MORTALITY WORLDWIDE AND, IN THE UNITED STATES (U.S.), PEOPLE OF COLOR SUFFER FROM MANY DISEASES ASSOCIATED WITH EXPOSURE TO AIR POLLUTION AT HIGHER RATES THAN WHITE PEOPLE. OUR UNDERSTANDING OF HUMAN EXPOSURE TO AIR POLLUTION IS BASED LARGELY ON MEASUREMENTS OF OUTDOOR POLLUTANT CONCENTRATIONS AND A MULTITUDE OF STUDIES HAVE FOUND THAT, FOLLOWING DECADES OF DISCRIMINATORY HOUSING AND LAND-USE POLICIES, PEOPLE OF COLOR ARE MORE LIKELY TO LIVE IN U.S. NEIGHBORHOODS WITH HIGHER CONCENTRATIONS OF OUTDOOR AIR POLLUTION; HOWEVER, OUTDOOR AIR POLLUTION DATA DO NOT CAPTURE THE FULL PICTURE OF HUMAN EXPOSURE. PEOPLE LIVING IN THE U.S. SPEND MOST OF THEIR TIME INDOORS AT HOME AND PRIOR STUDIES HAVE FOUND THAT IN-HOME AIR POLLUTANT CONCENTRATIONS—WHICH, BEING A COMBINATION OF POLLUTION FROM OUTDOOR AND INDOOR SOURCES, ARE ALSO LIKELY TO BE IMPACTED BY THE AFOREMENTIONED DISCRIMINATORY POLICIES— TEND TO BE BETTER-CORRELATED WITH PERSONAL EXPOSURES THAN OUTDOOR CONCENTRATIONS. IN-HOME AIR POLLUTION IS RARELY CHARACTERIZED DUE TO THE COST AND COMPLEXITY OF SETTING UP LARGE, NOISY, FINICKY MONITORS IN PRIVATE RESIDENCES. THE GOAL OF THIS PHASE II SBIR PROJECT IS TO REDUCE THE AMOUNT OF AIR POLLUTION U.S. RESIDENTS ARE EXPOSED TO AT HOME BY MAKING CHARACTERIZATION OF IN-HOME AIR QUALITY EASY; AFFORDABLE; USEFUL TO HOME OCCUPANTS; AND WIDESPREAD ENOUGH TO INFLUENCE LOCAL-, STATE-, AND NATIONAL-LEVEL POLICIES. THE FIRST AIM OF THIS PROJECT IS TO DEVELOP A SMALL, QUIET, EASY-TO-USE “HOME HEALTH BOX” (HHB) THAT MEASURES INDOOR CONCENTRATIONS OF PARTICULATE MATTER, VOLATILE ORGANIC COMPOUNDS, CARBON DIOXIDE, AND NITROGEN DIOXIDE USING A COMBINATION OF LOW-COST SENSORS AND HIGH-FIDELITY INTEGRATED SAMPLERS. THE HHB IS DESIGNED TO ELIMINATE THE NEED FOR AN AIR QUALITY PROFESSIONAL TO GO TO THE HOME; ONCE THE HHB IS DELIVERED, AN OCCUPANT SIMPLY NEEDS TO SET IT IN A CENTRAL LOCATION, PLUG IT IN, AND SWITCH IT ON. DATA COLLECTED BY THE HHB WILL UNDERGO AUTOMATED ANALYSES, BE REVIEWED BY AN AIR QUALITY PROFESSIONAL, AND THEN BE SYNTHESIZED INTO A REPORT THAT WILL (A) COMPARE MEASURED POLLUTANT LEVELS TO GUIDELINES, (B) DESCRIBE POTENTIAL POLLUTION SOURCES, AND (C) SUGGEST AFFORDABLE ACTIONS THAT OCCUPANTS COULD TAKE TO IMPROVE THEIR IN-HOME AIR QUALITY. THE SECOND AIM OF THIS PROJECT IS TO EVALUATE WHETHER HHB SAMPLING AND SUBSEQUENT RECEIPT OF A PERSONALIZED “HOME HEALTH REPORT” (HHR) HELPS HOUSEHOLDS IMPROVE THEIR IN-HOME AIR QUALITY OVER TIME. A RACIALLY- AND ETHNICALLY-DIVERSE GROUP OF HOUSEHOLDS WILL BE RECRUITED IN PARTNERSHIP WITH HEALTHCARE PROVIDERS AND COMMUNITY ORGANIZATIONS IN DENVER AND CHICAGO. HOMES WILL BE RANDOMIZED INTO INTERVENTION AND CONTROL GROUPS. HOMES IN BOTH GROUPS WILL RECEIVE THREE, ONE-WEEK HHB DEPLOYMENTS SPACED SIX WEEKS APART. AFTER EACH OF THE FIRST TWO DEPLOYMENTS, HOMES IN THE INTERVENTION GROUP WILL RECEIVE A HHR. WE WILL (A) INVESTIGATE WHETHER AND HOW AIR QUALITY CHANGES OVER TIME IN CONTROL AND INTERVENTION HOMES AND (B) SURVEY INTERVENTION HOUSEHOLDS ON THE UTILITY OF THE HOME HEALTH REPORTS. THE EDA-FUNDED EARTHEN BERM WILL PROVIDE CRITICAL PROTECTION TO THE FACILITY DURING FUTURE FLOOD EVENTS, AND WILL ENSURE CRITICAL SERVICES REMAIN ONLINE AND RESILIENT IN THE FACE OF FUTURE ECONOMIC SHOCKS. THE PROJECT BEING FUNDED IS THE BOBBY BENSON CENTER'S FACILITY RENOVATION PROJECT. THE BOBBY BENSON CENTER IS A NON-PROFIT RESIDENTIAL TREATMENT FACILITY FOR YOUTH AGES 13-17 YEARS OLD WITH SUBSTANCE USE AND MENTAL HEALTH DISORDERS. THE FACILITY CONSISTS OF FOUR CABINS (YOUTH LIVING QUARTERS AND THERAPIST OFFICES) WITH A COMBINED 8,000 SQUARE FEET AND A 3,600 SQUARE FOOT ADMINISTRATION BUILDING WHERE YOUTH PARTICIPATE IN THE INTAKE PROCESS, MEET WITH THE NURSE/OPERATIONS MANAGER/CLINICAL DIRECTOR/INTAKE COORDINATOR, AND ALSO HOUSES OUR ACCOUNTING, HUMAN RESOURCE, QUALITY ASSURANCE, AND IT DEPARTMENTS. OUR UPMOST PRIORITY AT THE BOBBY BENSON CENTER IS THE HEALTH (BOTH PHYSICAL AND MENTAL) & SAFETY OF THE YOUTH, EMPLOYEE, VISITORS AND OTHER CUSTOMERS. BOBBY BENSON CENTER HAS BEEN IN OPERATIONS FOR 33 YEARS AND IN THAT TIME THE BUILDINGS HAVE BEEN SUBJECT TO AGING, WEAR AND TEAR AND ENVIRONMENTAL ELEMENTS THAT HAVE MADE IT NECESSARY RENOVATE THE FACILITY. THIS FUNDING WILL HELP SUPPORT THE CENTER IN MAKING THE NECESSARY RENOVATIONS OF THE FACILITY TO ENSURE THAT ALL WHO ENTER MAY ENJOY A CLEAN AND MODERN FACILITY AND MAKE AN ENVIRONMENT CONDUCIVE TO HEALING FOR THE YOUTH.

PURPOSE: THE PUBLIC HOUSING OPERATING FUND (PH OPFUND) PROVIDES OPERATING SUBSIDIES TO HOUSING AUTHORITIES (HAS) TO ASSIST IN FUNDING THE OPERATING AND MAINTENANCE EXPENSES OF THEIR DWELLINGS, IN ACCORDANCE WITH SECTION 9 OF THE U.S. HOUSING ACT OF 1937, AS AMENDED. THE SUBSIDIES ARE REQUIRED TO HELP MAINTAIN SERVICES AND PROVIDE MINIMUM OPERATING RESERVES. THE PH OPFUND IS A $5 BILLION DOLLAR PROGRAM PROVIDING FUNDING TO APPROXIMATELY 6,000 HAS SERVING 1,590,321 PUBLIC HOUSING RESIDENTS IN 902,436 HOUSEHOLDS (44% ARE ELDERLY AND 35% OF RESIDENTS HAVE CHILDREN). SUCH ACTIVITIES INCLUDE MANAGEMENT AND OPERATIONS, ROUTINE AND PREVENTATIVE MAINTENANCE, ANTI-CRIME, ANTI-DRUG AND SECURITY ACTIVITIES, OPERATING COSTS FOR PRIVATELY OWNED PUBLIC HOUSING UNITS WITHIN MIXED-FINANCE PROJECTS, ENERGY COSTS, RESIDENT SUPPORTIVE SERVICES, INSURANCE, DEBT SERVICE AND COSTS ASSOCIATED WITH ADMINISTRATION AND PROGRAM IMPLEMENTATION. TURNKEY III PROJECTS ARE FUNDED FOR UNITS UNDER THE FINAL LEASE PURCHASE AGREEMENT FOR CLOSING OUT THE PROGRAM. TO SUPPORT THESE ACTIVITIES, THERE IS CONTINUED MODERNIZATION OF THE INFORMATION TECHNOLOGY PLATFORMS. PHAS HAVE ACCESS TO WEB-BASED PLATFORMS THAT UTILIZE REAL-TIME DATA TO PROVIDE INSIGHT INTO THEIR PORTFOLIOS. PHAS CAN OBTAIN METRICS ON THEIR FUNDING LEVELS, OCCUPANCY RATES, AND THE NUMBER OF FAMILIES SERVED THROUGH RENTAL ASSISTANCE.; EXPECTED OUTCOMES: AS A RESULT OF THE ACTIVITIES PERFORMED, THIS PROGRAM IS EXPECTED TO ASSIST IN FUNDING THE OPERATING AND MAINTENANCE EXPENSES. THIS MAY INCLUDE INCREASED OCCUPANCY IN PUBLIC HOUSING, DECREASED ENERGY COSTS THROUGH REGULAR MAINTENANCE AND ENERGY PERFORMANCE CONTRACTING AND LEVERAGE FEDERAL RESOURCES. SBIR PHASE II: HIGH RESOLUTION TERAHERTZ VIDEO CAMERA FOR MEDICAL IMAGING -THIS SMALL BUSINESS INNOVATION RESEARCH PHASE II PROJECT WILL DEVELOP AND LAUNCH A NEW, COMMERCIALLY VIABLE TERAHERTZ (THZ) IMAGING TECHNOLOGY INITIALLY TARGETED AT MEDICAL IMAGING APPLICATIONS. THE PROTOTYPE INSTRUMENT USES THZ REFLECTIVE IMAGING PHENOMENOLOGY, A TECHNIQUE THAT HAS BEEN SHOWN TO EFFICIENTLY DISCRIMINATE BETWEEN HEALTHY AND DISEASED TISSUES. WITH A RESOLUTION OF MORE THAN 300X300 PIXELS, OPERATION AT BETWEEN 10-30 FRAMES PER SECOND, AND A COMPACT FORM FACTOR, THE SYSTEM IS APPROPRIATE FOR MEDICAL ENVIRONMENTS SUCH AS CANCER SURGERIES, INCLUDING GLIOBLASTOMA RESECTION. THE THZ CAMERA WILL BE A NEW ADDITION TO THE MEDICAL IMAGING MARKET THAT HAS MATURED WITH X-RAY, MRI, VISIBLE AND INFRARED TECHNOLOGIES TO REACH $45 BILLION. THIS NEW INSTRUMENT IS ENABLED BY A LARGE FORMAT SENSOR ARRAY THAT USES A LOW-COST MANUFACTURING PROCESS ALREADY IMPLEMENTED IN A HIGH-VOLUME FOUNDRY. IN ADDITION TO THE MEDICAL APPLICATION, SIMILAR IMAGING SYSTEMS CAN BE USED IN SECURITY SCREENING APPLICATIONS ($2 BILLION MARKET/YEAR) SUCH AS PASSENGER SCREENING AT AIRPORTS. MORE BROADLY, THE UNDERLYING SENSOR TECHNOLOGY CAN BE ADAPTED FOR USE ACROSS THE EXTREMELY WIDE THZ SPECTRUM, WHICH HAS IMPLICATIONS FOR MANY INDUSTRIES. THE INTELLECTUAL MERIT OF THIS PROJECT IS PRIMARILY ITS INTEGRATION OF BOTH HIGH RESOLUTION AND HIGH-SPEED ASPECTS OF THZ IMAGING, AS THESE CAPABILITIES HAVE NOT YET BEEN DEMONSTRATED IN A COMMERCIALLY VIABLE FORM. PREVIOUS DEMONSTRATIONS HAVE EITHER INVOLVED SLOW SCANNING SYSTEMS THAT PRECLUDE REAL TIME APPLICATIONS OR CAMERAS THAT TARGET MUCH HIGHER BANDS OF THE THZ SPECTRUM. THE LATTER RESTRICTION STEMS FROM AN INABILITY TO MANUFACTURE ARRAYS OF LARGE PIXELS TO IMAGE AT THE LONGER THZ WAVELENGTHS. GOING BEYOND ONE OF A KIND LABORATORY PROOF OF CONCEPT EXPERIMENTS, THE THZ CAMERA SEEKS TO PROVIDE RESOLUTION AND FRAME UPDATE RATES THAT ARE RELEVANT TO ACTUAL APPLICATIONS. THE COMMERCIAL MANUFACTURING VIABILITY OF THE SENSING ELEMENT MAY ENSURE THAT APPLICATIONS BEYOND THE INITIAL MEDICAL MARKET WILL BE PRACTICAL. THE CAMERA WILL FEATURE NOT ONLY THE HIGH-RESOLUTION THZ IMAGING NEAR THE 250-350 GHZ SPECTRUM BUT ALSO INCORPORATE HIGH-RESOLUTION VISIBLE AND NEAR-INFRARED CAMERAS THAT CAN COMPLEMENT THE THZ VIDEO, WITH TRIALS BEING PLANNED FOR HOSPITAL USAGE. IN ADDITION TO CLINICAL USAGE SCENARIOS, THE IMAGING SYSTEM CAN BE ADAPTED FOR USE IN AIRPORT SECURITY AND OTHER SCREENING APPLICATIONS, AS PORTABILITY AND EASE OF USE WILL GUIDE THE DEMONSTRATION SYSTEM DESIGN. OPTIMIZING A MINIATURIZED AND INTEGRATED CONTINUOUS GLUCOSE MONITORING PLATFORM - PROJECT SUMMARY THE LONG-TERM GOAL OF THIS PROJECT IS TO DEVELOP A SCALABLE, USER-FRIENDLY, CONTINUOUS GLUCOSE MONITORING (CGM) SYSTEM FOR BETTER DIABETES MANAGEMENT. IMS HAS DEVELOPED THE WORLD'S SMALLEST ELECTROCHEMICAL ANALYTE SENSING PLATFORM BASED ON AN APPLICATION-SPECIFIC INTEGRATED CIRCUIT (ASIC) USING COMPLEMENTARY METAL-OXIDE SEMICONDUCTOR (CMOS) TECHNOLOGY. THIS DESIGN OFFERS SOME UNIQUE ADVANTAGES DUE TO ITS ACTIVE DIGITAL SENSOR (COMPARED TO PASSIVE ANALOG SENSORS IN CURRENT LEADING CGMS) INCLUDING EXTREME MINIATURIZATION, ON-CHIP INTEGRATION OF MULTIPLE SENSORS AND PROCESSING ELECTRONICS, IMPROVEMENT IN SIGNAL-TO-NOISE (SNR) RATIO (THUS IMPROVING HYPOGLYCEMIA ACCURACY), AND ON-CHIP TEMPERATURE CALIBRATION (IMPROVE ACCURACY, RELIABILITY BY CONFIRMING THE SENSOR IS IN THE TISSUE, AND PERFORMING ON-SITE TEMPERATURE CALIBRATION DURING SICKNESS, EXERCISE, AND SLEEP). RECENTLY, WE VERIFIED THE FEASIBILITY OF SYSTEM OPERATION FOR 14 DAYS IN A SECOND HUMAN STUDY (ONGOING). OUR OBJECTIVE IN THIS CRP GRANT IS TO OPTIMIZE THE MANUFACTURING AND THE QUALITY FRAMEWORK TO ENABLE LARGER HUMAN STUDIES, PAVING THE WAY TOWARDS ENABLING AN FDA IDE FOR APPROVAL TRIALS. THIS WORK IS SIGNIFICANT AS IT WILL CATALYZE THE COMMERCIALIZATION OF THE FIRST CGM+ SYSTEM CAPABLE OF THE SMALLEST NEEDLE INSERTION (28-GAUGE NEEDLE) THAT CAN WORK FOR A LONG TIME (>14 DAYS) WITH FACTORY CALIBRATION (NO FINGER STICKS). AFTER THE FIRST APPROVAL, THIS SYSTEM CAN BE EXTENDED TO INCLUDE OTHER ANALYTES (E.G., KETONES, INSULIN, GLUCAGON) WITHOUT INCREASING ITS SIZE, OWING TO THE ON-CHIP INTEGRATION OF MULTIPLE SENSORS. PURPOSE: THE PUBLIC HOUSING OPERATING FUND (PH OPFUND) PROVIDES OPERATING SUBSIDIES TO HOUSING AUTHORITIES (HAS) TO ASSIST IN FUNDING THE OPERATING AND MAINTENANCE EXPENSES OF THEIR DWELLINGS, IN ACCORDANCE WITH SECTION 9 OF THE U.S. HOUSING ACT OF 1937, AS AMENDED. THE SUBSIDIES ARE REQUIRED TO HELP MAINTAIN SERVICES AND PROVIDE MINIMUM OPERATING RESERVES. THE PH OPFUND IS A $5 BILLION DOLLAR PROGRAM PROVIDING FUNDING TO APPROXIMATELY 6,000 HAS SERVING 1,590,321 PUBLIC HOUSING RESIDENTS IN 902,436 HOUSEHOLDS (44% ARE ELDERLY AND 35% OF RESIDENTS HAVE CHILDREN). INFORMATION ON THE CURRENT OPERATING FUND GRANT PROCESSING CAN BE FOUND AT HTTPS://WWW.HUD.GOV/PROGRAM_OFFICES/PUBLIC_INDIAN_HOUSING/PROGRAMS/PH/AM/FUNDING.; ACTIVITIES TO BE PERFORMED: OPERATING FUNDS ARE USED TO FUND DAY-TO-DAY OPERATIONAL EXPENSES ASSOCIATED WITH PUBLIC HOUSING AS WELL AS THE ADMINISTRATIVE AND PROGRAM IMPLEMENTATION EXPENSES THAT PUBLIC HOUSING AGENCIES (PHAS) ARE REQUIRED TO UNDERTAKE UNDER THE 1937 HOUSING ACT AND PROGRAM REGULATIONS. SUCH ACTIVITIES INCLUDE MANAGEMENT AND OPERATIONS, ROUTINE AND PREVENTATIVE MAINTENANCE, ANTI-CRIME, ANTI-DRUG AND SECURITY ACTIVITIES, OPERATING COSTS FOR PRIVATELY OWNED PUBLIC HOUSING UNITS WITHIN MIXED-FINANCE PROJECTS, ENERGY COSTS, RESIDENT SUPPORTIVE SERVICES, INSURANCE, DEBT SERVICE AND COSTS ASSOCIATED WITH ADMINISTRATION AND PROGRAM IMPLEMENTATION. TURNKEY III PROJECTS ARE FUNDED FOR UNITS UNDER THE FINAL LEASE PURCHASE AGREEMENT FOR CLOSING OUT THE PROGRAM. TO SUPPORT THESE ACTIVITIES, THERE IS CONTINUED MODERNIZATION OF THE INFORMATION TECHNOLOGY PLATFORMS. PHAS HAVE ACCESS TO WEB-BASED PLATFORMS THAT UTILIZE REAL-TIME DATA TO PROVIDE INSIGHT INTO THEIR PORTFOLIOS. PHAS CAN OBTAIN METRICS ON THEIR FUNDING LEVELS, OCCUPANCY RATES, AND THE NUMBER OF FAMILIES SERVED THROUGH RENTAL ASSISTANCE.; EXPECTED OUTCOMES: AS A RESULT OF THE ACTIVITIES PERFORMED, THIS PROGRAM IS EXPECTED TO ASSIST IN FUNDING THE OPERATING AND MAINTENANCE EXPENSES. THIS MAY INCLUDE INCREASED OCCUPANCY IN PUBLIC HOUSING, DECREASED ENERGY COSTS THROUGH REGULAR MAINTENANCE AND ENERGY PERFORMANCE CONTRACTING AND LEVERAGE FEDERAL RESOURCES. IN ADDITION TO ADDRESSING THE DEPARTMENT’S STRATEGIC GOALS OF: • ADDRESSING THE NEED FOR QUALITY AFFORDABLE RENTAL HOMES BY MAINTAINING OR IMPROVING UPON THE 96% OCCUPANCY RATE OF HABITABLE UNITS; • PROMOTING HOUSING AS A PLATFORM TO IMPROVE QUALITY OF LIFE THROUGH SUPPORTIVE SERVICES, CRIME PREVENTION EFFORTS AND RESIDENT ENGAGEMENT ACTIVITIES; AND • HELPING TO BUILD INCLUSIVE COMMUNITIES FREE FROM DISCRIMINATION BY FACILITATING THE IMPLEMENTATION OF AFFIRMATIVELY FURTHERING FAIR HOUSING MEASURES.; INTENDED BENEFICIARIES: THE OPERATING FUND PROVIDES FOR THE OPERATION AND MAINTENANCE OF LOW-INCOME HOUSING PROJECTS TO PHAS/PROJECTS. IT WAS CREATED TO ASSIST HOUSING AUTHORITIES IN PROVIDING DECENT AND SAFE RENTAL HOUSING FOR ELIGIBLE LOW-INCOME FAMILIES OR INDIVIDUALS, THE ELDERLY, AND PERSONS WITH DISABILITIES. A HA DETERMINES ELIGIBILITY BASED ON 1) ANNUAL GROSS INCOME; 2) A PERSON ON WHO IS ELDERLY, A PERSON WITH A DISABILITY, OR AS A FAMILY; AND 3) U.S. CITIZENSHIP OR ELIGIBLE IMMIGRATION STATUS.; SUBRECIPIENT ACTIVITIES: THE SUBRECIPIENT ACTIVITIES ARE UNKNOWN AT THE TIME OF AWARD.

HIGH FREQUENCY RADAR (HFR) PROVIDES ONE OF THE 30 ESSENTIAL OCEAN VARIABLES OUTLINED IN THE U.S. INTEGRATED OCEAN OBSERVING SYSTEM (IOOS) MISSION, AND IS THE ONLY INSTRUMENT SYSTEM OPERATIONALLY PROVIDING BOTH HIGH TEMPORAL AND SPATIAL RESOLUTION OBSERVATIONS OVER THE CONTINENTAL SHELF. APPROXIMATELY 85% OF THE IOOS HFRS ARE CODAR SEASONDES, WITH MEASUREMENTS CONSISTING OF RANGE, BEARING, AND VELOCITY OF A SEA SURFACE PATCH. THE SEASONDES UTILIZE ALGORITHMS ALONG WITH A COMPACT ANTENNA SYSTEM TO DETERMINE THE BEARING. RANGE AND VELOCITY ARE HIGHLY ACCURATE, WHILE THE BEARING DETERMINATION IS THE GREATEST SOURCE OF ERROR. A FURTHER ISSUE IS THAT THESE MEASUREMENTS PRESENTLY PROVIDE NO INDICATION OF MEASUREMENT UNCERTAINTY, WHICH HAS HINDERED UTILIZATION OF HFR OBSERVATIONS BY MODELERS AND FORECASTERS, AND PRESENTS A CHALLENGE FOR HFR OPERATORS PERFORMING DATA QUALITY CONTROL. THIS PHASE II WORK IS BROKEN DOWN INTO THREE MAIN CATEGORIES: HARDWARE AND INTEGRATION (~30%), DATA COLLECTION (~25%), AND ALGORITHM DEVELOPMENT AND DATA ANALYSIS (~45%). MUCH OF THIS WORK WILL BE HAPPENING IN PARALLEL AS EXISTING SYSTEMS ARE UPDATED AND USES AS NEXT STAGE SYSTEMS ARE UNDER DEVELOPMENT. NWB SENSORS, INC. IMPLEMENTS AN ITERATIVE SCIENTIFIC / ENGINEERING APPROACH ON ALL OUR PROJECTS. THIS APPROACH FOLLOWS THE STEPS OF 1. MODEL THE PROBLEM OR DESIGN THE SYSTEM, 2. VALIDATE THE MODEL OR SYSTEM DATA BY COLLECTING REAL WORLD DATA (TESTING), 3. REFINE THE MODEL OR SYSTEM IF DESIRED PERFORMANCE WAS NOT ACHIEVED. THROUGHOUT THE PROJECT ENGINEERING PROJECT MANAGEMENT METHODS WILL BE IMPLEMENTED. THESE METHODS INCLUDE INDUSTRY STANDARD SOFTWARE TOOLS FOR: COLLABORATIVE PROJECT MANAGEMENT; DOCUMENTATION; SOFTWARE VERSION CONTROL, AND BACKUP OF PROJECT DOCUMENTS.DEVELOPMENT ON THE IMPROVED CAMERA CONTROLLER WILL START IN FALL 2019 AND CONTINUE INTO EARLY SPRING 2020. AT THIS TIMETHE CAMERA CONTROLLER WILL BE UPDATED TO IMPLEMENT IMPROVED MOVEMENT DETECTION AND CAMERA CONTROL. DURING DEVELOPMENT THESE MOTION DETECTION AND CAMERA CONTROL ROUTINES WILL BE TESTED USING AUTOMOBILES IN SIMULATED FIELD TESTS. WORK DURING THE EARLY SPRING WILL VALIDATE THESE IMPROVEMENTS IN FARM TESTS. INITIAL WORK ON THE NVIDIA JETSON EMBEDDED MACHINE VISION PLATFORM WILL ALSO START AT THIS TIME WITH THE GOAL OF PROTOTYPE FIELD TEST DURING THE ON-FARM EXPERIMENTS IN SUMMER 2020. BASED ON THE RESULTS OF THESE TESTS THE PROTOTYPE SYSTEM WILL BE IMPROVED UPON DURING FALL AND WINTER 2020 TO BE READY FOR FIELD TESTS IN SUMMER 2021.DEVELOPMENT TO IMPROVE THE DATA PROCESSING TOOLCHAIN AND TO IMPLEMENT THE END USER DATA APPLICATION WILL HAPPEN IN PARALLEL AND WILL TAKE PLACE THROUGHOUT THE PROJECT STARTING IN FALL 2019. THE FIRST TASK OF THIS EFFORT WILL BE TO IMPLEMENT OBJECT TRACKING IN THE DATA PROCESSING TOOL CHAIN. THE ILLUMINATION CORRECTION AND IMPROVED ANOMALY DETECTION WILL UTILIZETHE OBJECT TRACKING ROUTINES. AS NEW IMAGERY IS ADDED DURING THE 2020 AND 2021 SUMMER THESE ROUTINES WILL BE IMPROVED AS NECESSARY. TESTING OF THE METHODS TO COMMUNICATE IN FIELD DATA WILL BE DEVELOPED WITH A PROTOTYPE READY FOR SUMMER 2020.CRITICAL TO THE SUCCESS OF THIS PROJECT IS THE CREATION OF TOOLS TO ALLOW FOR SYSTEM OF LEARNING FOR NEW ENVIRONMENTS. DEVELOPMENT ON THE METHODS FOR ASSISTED TRAINING USING AUTO-SORTED CLASSES CAN START IN FALL 2019. THIS WORK CAN USE OBJECT IDENTIFICATION GRAPHS TRAINED ON GRAIN CROPS BUT BEING ADAPTED TO PULSE CROPS. VALIDATION OF THESE TECHNIQUES WILL USE COMPARISONS WITH EXISTING HUMAN BUILT GRAPHS FOR PULSE CROP OBJECT IDENTIFICATION. AFTER THE MOTION TRACKING HAS BEEN INTEGRATED INTO THE TOOL CHAIN FURTHER DEVELOPMENT ON AUTOMATED NEW CROP AND OBJECT DETECTION CAN TAKE PLACE.WORK TO VALIDATE DATA PROCESSING AND TRAINING TOOL CHAINS WILL TAKE PLACE THROUGHOUT THE PROJECT USING BOTH NEWLY COLLECTED AND EXISTING DATA. VALIDATION WILL REQUIRE CLOSE WORK WITH OUR CO-OPERATING GROWERS AND LANDOWNERS TO ENSURE THE MAPPING DATA IS ACCURATE AND IN FORMATS USABLE IN THEIR OPERATIONS.

STEVENSON CARSON SCHOOL DISTRICT IS WORKING WITH DAVID CORR, A SECURITY CONSULTANT TO BRING THE SCHOOL DISTRICT UP TO A MORE SECURE STATE THAN THE CURRENT SITUATION. THIS UPDATING BEGAN LAST SUMMER WITH SMALL, EASY UPGRADES WHILE THE BIGGER PICTURE, LARGER PROJECTS WERE DETERMINED. THERE WILL CONTINUE TO BE MORE PROJECTS AFTER THE GRANT PERIOD BUT A LARGE CHUNK OF COSTS ARE PROPOSED WITHIN THIS GRANT APPLICATION. THE APPLICATION CONTAINS REQUESTS FOR: MANY MORE CAMERAS; A BULLET PROOF LAMINATE/FILM TO APPLY TO WINDOWS; PA SYSTEM UPGRADES THAT WILL ASSIST WITH EMERGENCY SITUATIONS; THE CONSULTANT; HARDENING OF WALLS/PLATING; ADDING A LAYER TO SEVERAL SCHOOLS TO REQUIRE PERMISSIVE ACCESS AND CARD READER DOOR ACCESS POINTS; AND A CHAIN LINK FENCE AROUND A DIFFICULT PART OF THE HIGH SCHOOL TO SECURE. SOME OF THIS WILL BE PAID FOR BY OUR ENTITY AND THE REST BY THE GRANT. THIS LAST MONTH HAS BEEN VERY CHALLENGING TO OUR SCHOOL DISTRICT. WE HAVE HAD LOCK DOWNS, LOCK OUTS, WE HAD TO CANCEL ONE SCHOOL DAY BECAUSE OF THE EXHAUSTIVE ISSUES. WE EVEN MADE THE NEWS. THIS IS A SMALL SCHOOL DISTRICT (CURRENTLY 780 STUDENTS IN 4 SCHOOLS) WITH AN EVER INCREASING ARRAY OF CHALLENGES. THESE CHANGES WILL HELP THE SCHOOL DISTRICT MEET THE SECURITY GOALS DETERMINED BY THE CONSULTANT, COMMUNITY AND SCHOOL BOARD. THE 4 SCHOOLS ARE DIVIDED INTO 2 COMMUNITIES. STEVENSON IS THE CITY PROPER AND IT HOLDS THE YOUNGEST ELEMENTARY SCHOOL TTK-2ND GRADE AND THE HIGH SCHOOL 9TH-12TH GRADE. CARSON IS ON THE OUTSKIRTS OF STEVENSON WITH THE OLDER ELEMENTARY 3RD-5TH GRADE AND THE MIDDLE SCHOOL 6TH-8TH GRADE. THE PROJECTS ARE VARIED PER SCHOOL BASED ON NEEDS. THE OUTCOMES: SCHOOL BUSINESS CAN BE CONDUCTED WITHOUT ACCESS INTO SCHOOL; MORE CAMERA FOOTAGE TO DETERMINE ISSUES; INCREASED PROTECTION FROM POSSIBLE ATTACKS WITH PROTECTIVE LAYERS; LIMITED ACCESS USING CARD READERS VERSUS KEYS THAT ARE NOT RETURNED; QUICKER/CONSISTENT RESPONSE WITH UPGRADED PA SYSTEMS. THE CONSULTANT AND THE SHERIFF ARE WORKING WITH THE SCHOOL DISTRICT ON THE EMERGENCY OPERATING PROCEDURES AND UPDATING AS BOTH PLANNED DRILLS AND THE UNFORTUNATE NON-DRILLS CONTINUE TO TAKE PLACE IN THE DISTRICT.

IMS HAS DEVELOPED THE WORLD'S SMALLEST WIRELESS ELECTROCHEMICAL CGM USING A NOVEL SEMICONDUCTOR (CMOS) PLATFORM. THIS DESIGN OFFERS SOME UNIQUE ADVANTAGES DUE TO ITS SMART SENSOR (COMPARED TO PASSIVE SENSORS IN COMPETITOR DESIGNS) INCLUDING EXTREME MINIATURIZATION (SYSTEM IS SMALLER THAN HALF A SESAME SEED), ON-CHIP INTEGRATION OF MULTIPLE SENSORS, IMPROVEMENT IN SIGNAL-TO-NOISE RATIO (THUS IMPROVING HYPOGLYCEMIA ACCURACY), DECREASE IN SENSOR CAPACITANCE AND MEMBRANE THICKNESS (THUS IMPROVING RESPONSE SPEED), AND ON-CHIP TEMPERATURE CALIBRATION. THIS SYSTEM CAN ENABLE AUTOMATIC CLOSED-LOOP GLUCOSE CONTROL WITHOUT MEAL ANNOUNCEMENTS AND CARBOHYDRATE COUNTING AND WITHOUT THE FEAR OF MISSED HYPOGLYCEMIC EVENTS; GREATLY SIMPLIFYING PATIENT EXPERIENCE AND IMPROVING SAFETY. WE HAVE DEMONSTRATED THE FUNCTION OF OUR CGM PLATFORM IN THE LAB, IN ANIMALS, AND RECENTLY IN HUMANS. THE ADDITION OF CKM CAN SIGNIFICANTLY IMPROVE THE VALUE OF THE SYSTEM TO DETECT IMPENDING DKA THAT GOES UNNOTICED WITH THE CURRENT CGM SYSTEMS, CAUSING SIGNIFICANT HARM TO THE AT-RISK PATIENTS. THEREFORE, OUR OBJECTIVE IN THE ORIGINAL PROPOSAL WAS TO DEVELOP A MULTI-ANALYTE SENSING PLATFORM THAT CAN SENSE BOTH GLUCOSE AND KETONE IN THE INTERSTITIAL FLUID (ISF) USING A SINGLE DEVICE. THE SENSING PLATFORM WOULD BE TESTED IN THE LAB. THIS RESEARCH IS SIGNIFICANT AS IT WILL ENABLE THE FIRST CDM SYSTEM WITH A SMALL, PAIN-FREE NEEDLE- INSERTION (26-GAUGE NEEDLE) THAT CAN WORK FOR A LONG TIME (>30 DAYS). ONCE PROVEN, THIS SYSTEM CAN BE EXTENDED TO INCLUDE OTHER IMPORTANT ANALYTES (E.G., INSULIN, GLUCAGON) WITHOUT INCREASING ITS SIZE OR COMPLEXITY, OWING TO ON-CHIP INTEGRATION OF MULTIPLE SENSORS ON A SINGLE MICROSCALE DEVICE. DURING OUR WORK, WE DISCOVERED AN UNFORESEEN ISSUE WITH THE KETONES SENSING CHEMISTRY THAT PREVENTS LONG-TERM STABILITY. WE HAVE DISCOVERED A POTENTIAL SOLUTION TO THAT BY USING A CONJUGATED ENZYME. A MINIATURIZED AND INTEGRATED CONTINUOUS METABOLIC MONITORING PLATFORM - PROJECT SUMMARY THE LONG-TERM GOAL OF THIS PROJECT IS TO DEVELOP A SCALABLE, USER-FRIENDLY, CONTINUOUS METABOLIC MONITORING (CMM) SYSTEM TO IMPROVE THE PERFORMANCE AND RELIABILITY OF HORMONE REPLACEMENT THERAPY FOR MILLIONS OF DIABETES PATIENTS. IMS HAS DEVELOPED THE WORLD'S SMALLEST WIRELESS ELECTROCHEMICAL ANALYTE SENSING PLATFORM BASED ON AN APPLICATION-SPECIFIC INTEGRATED CIRCUIT (ASIC) USING COMPLEMENTARY METAL-OXIDE SEMICONDUCTOR (CMOS) TECHNOLOGY. THIS DESIGN OFFERS SOME UNIQUE ADVANTAGES DUE TO ITS ACTIVE DIGITAL SENSOR (COMPARED TO PASSIVE ANALOG SENSORS IN COMPETITOR DESIGNS) INCLUDING EXTREME MINIATURIZATION, ON-CHIP INTEGRATION OF MULTIPLE SENSORS, IMPROVEMENT IN SIGNAL-TO- NOISE (SNR) RATIO (THUS IMPROVING HYPOGLYCEMIA ACCURACY), DECREASE IN SENSOR CAPACITANCE AND MEMBRANE THICKNESS (THUS IMPROVING RESPONSE SPEED), UNIVERSAL INTERFERENCE REJECTION (USING A BACKGROUND REFERENCE SENSOR AND SUBTRACTING ITS SIGNAL FROM ALL SENSORS), AND ON-CHIP TEMPERATURE CALIBRATION (IMPROVE ACCURACY, RELIABILITY BY CONFIRMING THE SENSOR IS IN THE TISSUE AND MEASURING THE CORRECT TEMPERATURE). THE PROPOSED CMM WILL ENABLE USERS TO BENEFIT FROM USEFUL DRUGS (E.G., METFORMIN, SGLT2 INHIBITORS), EXERCISE, AND AUTOMATED INSULIN DELIVERY SYSTEMS WITHOUT MEAL ANNOUNCEMENTS, CARBOHYDRATE COUNTING, AND THE RISK OF ADVERSE SIDE EFFECTS. HENCE, THE DESIGN IS HIGHLY INNOVATIVE. WE HAVE DEMONSTRATED THE FUNCTION OF THE FIRST-GENERATION PLATFORM FOR CONTINUOUS GLUCOSE MONITORING (CGM) IN HUMANS. CONTINUOUS LACTATE MONITORING (CLM) CAN SIGNIFICANTLY IMPROVE THE SYSTEM'S VALUE TO DETECT IMPENDING LACTIC ACIDOSIS THAT GOES UNNOTICED WITH THE CURRENT CGM SYSTEMS. WE HAVE DEMONSTRATED THE CLM IN ANIMALS AND A MULTIPLEXED CGM-CLM AT THE PROPOSED SIZE SCALE IN THE LAB. OUR OBJECTIVE IN THIS PROPOSAL IS TO DEVELOP A CMM THAT CAN SENSE GLUCOSE, LACTATE, AND TEMPERATURE IN THE INTERSTITIAL FLUID (ISF) USING A SINGLE WIRELESS SENSOR SMALLER THAN HALF A SESAME SEED. THE SMALL AND FLEXIBLE (20UM THICK WITH POLYMER COATINGS) SENSOR MINIMIZES THE FOREIGN BODY RESPONSE (FBR). THIS RESEARCH IS SIGNIFICANT AS IT WILL ENABLE THE FIRST CMM SYSTEM CAPABLE OF A SMALL, PAIN-FREE NEEDLE INSERTION (26-GAUGE NEEDLE) THAT CAN WORK FOR A LONG TIME (>30 DAYS) WITH FACTORY CALIBRATION (NO FINGER STICKS). ONCE PROVEN, THIS SYSTEM CAN BE EXTENDED TO INCLUDE OTHER IMPORTANT ANALYTES (E.G., KETONES, INSULIN, GLUCAGON) WITHOUT INCREASING ITS SIZE, OWING TO THE ON-CHIP INTEGRATION OF MULTIPLE SENSORS. PURPOSE: THE PUBLIC HOUSING OPERATING FUND (PH OPFUND) PROVIDES OPERATING SUBSIDIES TO HOUSING AUTHORITIES (HAS) TO ASSIST IN FUNDING THE OPERATING AND MAINTENANCE EXPENSES OF THEIR DWELLINGS, IN ACCORDANCE WITH SECTION 9 OF THE U.S. HOUSING ACT OF 1937, AS AMENDED. THE SUBSIDIES ARE REQUIRED TO HELP MAINTAIN SERVICES AND PROVIDE MINIMUM OPERATING RESERVES. THE PH OPFUND IS A $5 BILLION DOLLAR PROGRAM PROVIDING FUNDING TO APPROXIMATELY 6,000 HAS SERVING 1,590,321 PUBLIC HOUSING RESIDENTS IN 902,436 HOUSEHOLDS (44% ARE ELDERLY AND 35% OF RESIDENTS HAVE CHILDREN). INFORMATION ON THE CURRENT OPERATING FUND GRANT PROCESSING CAN BE FOUND AT HTTPS://WWW.HUD.GOV/PROGRAM_OFFICES/PUBLIC_INDIAN_HOUSING/PROGRAMS/PH/AM/FUNDING.; ACTIVITIES TO BE PERFORMED: OPERATING FUNDS ARE USED TO FUND DAY-TO-DAY OPERATIONAL EXPENSES ASSOCIATED WITH PUBLIC HOUSING AS WELL AS THE ADMINISTRATIVE AND PROGRAM IMPLEMENTATION EXPENSES THAT PUBLIC HOUSING AGENCIES (PHAS) ARE REQUIRED TO UNDERTAKE UNDER THE 1937 HOUSING ACT AND PROGRAM REGULATIONS. SUCH ACTIVITIES INCLUDE MANAGEMENT AND OPERATIONS, ROUTINE AND PREVENTATIVE MAINTENANCE, ANTI-CRIME, ANTI-DRUG AND SECURITY ACTIVITIES, OPERATING COSTS FOR PRIVATELY OWNED PUBLIC HOUSING UNITS WITHIN MIXED-FINANCE PROJECTS, ENERGY COSTS, RESIDENT SUPPORTIVE SERVICES, INSURANCE, DEBT SERVICE AND COSTS ASSOCIATED WITH ADMINISTRATION AND PROGRAM IMPLEMENTATION. TURNKEY III PROJECTS ARE FUNDED FOR UNITS UNDER THE FINAL LEASE PURCHASE AGREEMENT FOR CLOSING OUT THE PROGRAM. TO SUPPORT THESE ACTIVITIES, THERE IS CONTINUED MODERNIZATION OF THE INFORMATION TECHNOLOGY PLATFORMS. PHAS HAVE ACCESS TO WEB-BASED PLATFORMS THAT UTILIZE REAL-TIME DATA TO PROVIDE INSIGHT INTO THEIR PORTFOLIOS. PHAS CAN OBTAIN METRICS ON THEIR FUNDING LEVELS, OCCUPANCY RATES, AND THE NUMBER OF FAMILIES SERVED THROUGH RENTAL ASSISTANCE.; EXPECTED OUTCOMES: AS A RESULT OF THE ACTIVITIES PERFORMED, THIS PROGRAM IS EXPECTED TO ASSIST IN FUNDING THE OPERATING AND MAINTENANCE EXPENSES. THIS MAY INCLUDE INCREASED OCCUPANCY IN PUBLIC HOUSING, DECREASED ENERGY COSTS THROUGH REGULAR MAINTENANCE AND ENERGY PERFORMANCE CONTRACTING AND LEVERAGE FEDERAL RESOURCES. IN ADDITION TO ADDRESSING THE DEPARTMENT’S STRATEGIC GOALS OF: • ADDRESSING THE NEED FOR QUALITY AFFORDABLE RENTAL HOMES BY MAINTAINING OR IMPROVING UPON THE 96% OCCUPANCY RATE OF HABITABLE UNITS; • PROMOTING HOUSING AS A PLATFORM TO IMPROVE QUALITY OF LIFE THROUGH SUPPORTIVE SERVICES, CRIME PREVENTION EFFORTS AND RESIDENT ENGAGEMENT ACTIVITIES; AND • HELPING TO BUILD INCLUSIVE COMMUNITIES FREE FROM DISCRIMINATION BY FACILITATING THE IMPLEMENTATION OF AFFIRMATIVELY FURTHERING FAIR HOUSING MEASURES.; INTENDED BENEFICIARIES: THE OPERATING FUND PROVIDES FOR THE OPERATION AND MAINTENANCE OF LOW-INCOME HOUSING PROJECTS TO PHAS/PROJECTS. IT WAS CREATED TO ASSIST HOUSING AUTHORITIES IN PROVIDING DECENT AND SAFE RENTAL HOUSING FOR ELIGIBLE LOW-INCOME FAMILIES OR INDIVIDUALS, THE ELDERLY, AND PERSONS WITH DISABILITIES.
