THE PROJECT TEAM WILL COLLABORATE WITH THE SAN MATEO COUNTY FLOOD AND SEA-LEVEL RISE RESILIENCY DISTRICT ('ONESHORELINE') TO INTEGRATE THE PARK DESIGN WITH THEIR EMERGING REGIONAL SOLUTION FOR FLOOD PROTECTION ALONG THE COUNTY SHORELINE. | National Science Foundation | STTR PHASE I: A NOVEL TISSUE-ENGINEERED PLATFORM FOR CANCER MODELING AND DRUG SCREENING -THE BROADER IMPACT OF THIS SMALL BUSINESS TECHNOLOGY TRANSFER PHASE I PROJECT LIES PRIMARILY IN PROVIDING MORE EFFECTIVE DRUG SCREENING MODELS TO ELIMINATE INEFFECTIVE DRUG CANDIDATES AT AN EARLY STAGE. THIS MAY EXPEDITE THE ANTI-CANCER DRUG DEVELOPMENT PROCESS AND REDUCE EXCESSIVE EXPENSES DURING CLINICAL TRIALS, WHICH MAY INCREASE THE R&D RETURNS FOR PHARMACEUTICAL COMPANIES AND REDUCE HEALTHCARE COSTS FOR THE CUSTOMERS. FURTHERMORE, THE SOONER THAT AN EFFICACIOUS ANTI-CANCER DRUG CAN BE BROUGHT TO THE MARKET, THE SOONER THAT PATIENTS CAN DIRECTLY EXPERIENCE IMPROVED HEALTH OUTCOMES. CANCER HAS BEEN ONE OF THE LEADING CAUSES OF DEATH IN THE UNITED STATES AND WORLDWIDE. DEVELOPMENT OF SAFER AND MORE EFFECTIVE ANTICANCER DRUGS CAN PROVIDE PATIENTS THE OPPORTUNITY FOR BOTH A LONGER AND POTENTIALLY BETTER QUALITY OF LIFE. THE PROPOSED TECHNOLOGY MAY ALSO HAVE IMMENSE VALUE FOR PHARMACEUTICAL COMPANIES. IN DRUG DEVELOPMENT, RESEARCHERS SCREEN NUMEROUS COMPOUNDS TO UNDERSTAND DRUG EFFICACY AND DISCARD INEFFECTIVE ONES. 90% OF DRUG CANDIDATES IN THE PIPELINE WILL FAIL, IN PART DUE TO POOR CLINICAL TRANSLATION. THE PRODUCT AIMS TO HELP COMPANIES ELIMINATE INEFFICACIOUS DRUGS EARLIER BEFORE MILLIONS OF DOLLARS ARE SPENT ON THEIR DEVELOPMENT. THE PROPOSED PROJECT WILL DEVELOP A MORE PHYSIOLOGICALLY RELEVANT, CONSISTENT, AND VERSATILE HIGH-THROUGHPUT SCREENING (HTS) MODEL TO IMPROVE THE TRANSLATION RATE BETWEEN PRECLINICAL AND CLINICAL TESTING. CURRENT DRUG SCREENING APPROACHES, WHICH RELY ON TWO-DIMENSIONAL (2D) CELL CULTURES AND THREE-DIMENSIONAL (3D) CELL AGGREGATES, FAIL TO PROVIDE THE NECESSARY MICROENVIRONMENTAL CUES FOR ACCURATELY REPLICATING HUMAN PATIENT DRUG RESPONSES. THEREFORE, THE PROPOSED TECHNOLOGY HAS BEEN DEVELOPED, AIMING TO STRIKE A BALANCE BETWEEN THROUGHPUT, WHICH INCLUDES SCALABILITY AND UNIFORMITY, AND PHYSIOLOGICAL RELEVANCE, SUCH AS THE ABILITY TO MODULATE KEY ATTRIBUTES OF THE TUMOR MICROENVIRONMENT. UTILIZING A TISSUE ENGINEERING TOOLSET, THE 3D HYDROGEL SCAFFOLD OFFERS A MORE PHYSIOLOGICALLY PERTINENT MICROENVIRONMENT FOR CELL GROWTH AND DRUG RESPONSE. MEANWHILE, A PATENTED MICROFLUIDIC CELL ENCAPSULATION PLATFORM ALLOWS FOR THE RAPID AND CONSISTENT PRODUCTION OF THE PRODUCTS, WHICH ARE ESSENTIAL FOR HTS APPLICATIONS. THIS PROJECT AIMS TO 1) TEST ADDITIONAL CANCER CELL TYPES TO DEMONSTRATE THE PLATFORM'S BROAD APPLICABILITY, 2) EXPLORE CRYOPRESERVATION TO REDUCE RESPONSE TIME, AND 3) TRANSITION TO COMMERCIALLY AVAILABLE GMP-GRADE HYDROGEL PRECURSORS TO IMPROVE REPRODUCIBILITY AND SCALABILITY. THE OVERALL PROJECT, LOCATED ON RECLAIMED LAND ON THE EDGE OF SAN FRANCISCO BAY, WILL RESTORE TIDAL WETLANDS AND TRANSITION-ZONE HABITATS ABSENT FROM THE BURLINGAME SHORELINE, PROVIDE EDUCATIONAL AND RECREATIONAL AMENITIES, IMPROVE THE BAY TRAIL, AND DEMONSTRATE HOW GREEN INFRASTRUCTURE CAN PROTECT THE BAYFRONT. THE PROJECT TEAM WILL COLLABORATE WITH THE SAN MATEO COUNTY FLOOD AND SEA-LEVEL RISE RESILIENCY DISTRICT ('ONESHORELINE') TO INTEGRATE THE PARK DESIGN WITH THEIR EMERGING REGIONAL SOLUTION FOR FLOOD PROTECTION ALONG THE COUNTY SHORELINE. PEROSPHERE TECHNOLOGIES INTENDS TO COMMERCIALIZE THE DEVICE THROUGH A MARKETING, SALES AND DISTRIBUTION PARTNERSHIP WITH A GLOBAL MEDICAL DEVICE COMPANY. THE DEVICE IS SIMILARLY SUITED FOR USE IN EMERGENCY SITUATIONS (E.G. EMERGENCY ROOM, EMERGENCY VEHICLE) AND FOR ROUTINE CARE IN THE INPATIENT AND OUTPATIENT SETTINGS (HOSPITALS, CLINICS, SURGICAL SUITES, DOCTOR’S OFFICES, PHARMACIES) BY HEALTHCARE PROFESSIONALS WHO CAN PERFORM OR ORDER FRESH VENOUS WHOLE BLOOD DRAWS. THE DEVICE PROMISES TO ADDRESS SEVERAL UNMET MEDICAL NEEDS, INCLUDING POINT-OF- CARE MONITORING OF DOACS, AND HAS THE POTENTIAL TO DEFINE NEW STANDARDS OF CARE. TO HELP THE AMERICAN ECONOMY PREPARE FURTHER IN ADVANCE FOR IMPACTFUL WEATHER AND CLIMATE EVENTS, THIS PROJECT WILL DEVELOP A FORECAST SYSTEM THAT CAN TRANSLATE CLIMATE FORECASTS INTO LIKELIHOODS OF IMPACTFUL WEATHER EVENTS. THIS INFORMATION WILL BE USED TO INFORM THE ENERGY AND INSURANCE MARKETS OF FINANCIAL RISK, PARTICULARLY WITHIN THE RENEWABLE ENERGY SECTOR WHERE WEATHER AND CLIMATE CONTROL THE AMOUNT OF ENERGY PRODUCED. RENEWABLE ENERGY IS PRODUCED LOCALLY WITHIN THE UNITED STATES, WHICH MEANS THIS PROJECT WILL IMPROVE THE NATION?S ENERGY SECURITY BY INFORMING WHEN AND WHERE RENEWABLE ENERGY WILL BE MOST AVAILABLE. CONSERVATIVE ESTIMATES OF THE TECHNOLOGY?S POTENTIAL INCLUDE A $2.5 MILLION PER YEAR BENEFIT TO THE RENEWABLE ENERGY INDUSTRY, WITH A SIMILAR MULTI-MILLION DOLLAR IMPACT TO THE MORE GENERAL PARAMETRIC INSURANCE MARKET. THIS PROJECT WILL ENHANCE A CLIMATE FORECAST SYSTEM DEVELOPED DURING THE COMPANY'S NSF SBIR PHASE I AWARD THAT ISSUES CLIMATE FORECASTS UP TO A DECADE INTO THE FUTURE. THE ENHANCEMENTS INCLUDE INCREASED FORECAST ACCURACY THROUGH AUTOMATED MACHINE LEARNING MODEL PARAMETER TUNING, FORECAST POST-PROCESSING, AND THE TRANSLATION OF THE CLIMATE FORECASTS INTO REALISTIC WEATHER PATTERNS. THE TECHNOLOGY USES MACHINE LEARNING TO IDENTIFY PATTERNS IN THE LAND, ATMOSPHERE, AND OCEAN THAT HELP DETERMINE HOW THE CLIMATE SYSTEM WILL EVOLVE ON TIMESCALES OF ONE MONTH TO ONE DECADE. THESE CLIMATE FORECASTS WILL THEN BE TRANSLATED INTO REALISTIC POSSIBILITIES OF FUTURE WEATHER PATTERNS ON DAILY TIMESCALES. THESE DAILY WEATHER PATTERNS CAN THEN BE USED TO INFORM RENEWABLE ENERGY POWER PRODUCTION FORECASTS AND EXTREME WEATHER EVENT RISK FOR NUMEROUS INDUSTRIES, INCLUDING THE ENERGY SECTOR AND THE GENERAL INSURANCE INDUSTRY. A KEY TECHNICAL BENEFIT TO THE PROPOSED FORECAST SYSTEM IS ITS HIGH COMPUTATIONAL SCALABILITY, WHICH ENABLES THE RAPID CREATION OF CLIMATE FORECASTS THAT ARE TYPICALLY PRODUCED USING SUPERCOMPUTERS. VIVOSPHERES: TISSUE-ENGINEERED SPHEROIDAL MODELS FOR HIGH-THROUGHPUT SCREENING - ABSTRACT THE DEVELOPMENT OF THREE-DIMENSIONAL (3D) DISEASE MODELS HAS PAVED THE WAY FOR ACCELERATED DESIGN, DISCOVERY, AND VALIDATION OF POTENTIAL TARGETS AND DRUG COMPOUNDS, THEREBY SIGNIFICANTLY REDUCING THE TIME, COST, AND LABOR INVOLVED IN THE DRUG DISCOVERY PIPELINE. 3D SPHEROIDAL CELL AGGREGATE MODELS HAVE BEEN ESTABLISHED AS MORE ACCURATE AND PREDICTIVE THAN TWO-DIMENSIONAL (2D) MODELS FOR DRUG SCREENING. HOWEVER, CURRENT COMMERCIAL TECHNIQUES FOR SPHEROIDAL TISSUE FORMATION RELY HEAVILY ON THE SELF-AGGREGATION OF SINGLE CELLS AND HAVE SUBSTANTIAL LIMITATIONS CONCERNING PRODUCTION CONSISTENCY, CONTROL, AND FLEXIBILITY. THAT MAKES VIVOSPHERE A BREAKTHROUGH PRODUCT. VIVOSPHERES ARE CELL-LADEN HYDROGEL MICROSPHERES THAT ARE PRODUCED RAPIDLY USING OUR MICROFLUIDIC PLATFORM. COMPARED TO SELF-AGGREGATION SYSTEMS, INCORPORATION OF HYDROGEL MATRICES IN VIVOSPHERES CAN IMPROVE THE CONSISTENCY OF TISSUES IN SIZE AND SHAPE, WHICH IS CRITICAL IN GETTING HIGH-QUALITY RESULTS IN HIGH-THROUGHPUT SCREENING (HTS), AND PROVIDE THE ABILITY TO MANIPULATE THE CELLULAR MICROENVIRONMENT, WHICH IS AN IMPORTANT DRIVER OF APPROPRIATE CELL PHENOTYPE AND FUNCTION. THE CELL TYPES WILL NO LONGER BE LIMITED TO SELF-AGGREGATING CELLS. INSTEAD, THERE WILL BE GREAT FLEXIBILITY IN SELECTING EITHER AGGREGATING OR NON-AGGREGATING CELLS, AND EVEN MULTIPLE CELL TYPES FOR CO-ENCAPSULATION. THIS PROJECT WILL USE CANCER CELLS TO DEMONSTRATE THE HIGH CONSISTENCY OF VIVOSPHERES AS MORE SUITABLE FOR HTS COMPARED TO SELF-AGGREGATION SYSTEMS, ESTABLISH THE ABILITY TO FORM VIVOSPHERES USING NON-AGGREGATING CANCER CELLS, AND OPTIMIZE KEY ASSAYS USED IN HTS. THIS MAY EXPEDITE THE ANTI-CANCER DRUG DEVELOPMENT PROCESS AND REDUCE EXCESSIVE EXPENSES DURING CLINICAL TRIALS, WHICH MAY INCREASE THE R&D RETURNS FOR PHARMACEUTICAL COMPANIES AND REDUCE HEALTHCARE COSTS FOR THE CUSTOMERS. FURTHERMORE, THE SOONER THAT AN EFFICACIOUS ANTI-CANCER DRUG CAN BE BROUGHT TO THE MARKET, THE SOONER THAT PATIENTS CAN DIRECTLY EXPERIENCE IMPROVED HEALTH OUTCOMES. CANCER HAS BEEN ONE OF THE LEADING CAUSES OF DEATH IN THE UNITED STATES AND WORLDWIDE. DEVELOPMENT OF SAFER AND MORE EFFECTIVE ANTICANCER DRUGS CAN PROVIDE PATIENTS THE OPPORTUNITY FOR BOTH A LONGER AND POTENTIALLY BETTER QUALITY OF LIFE. THE PROPOSED TECHNOLOGY MAY ALSO HAVE IMMENSE VALUE FOR PHARMACEUTICAL COMPANIES. IN DRUG DEVELOPMENT, RESEARCHERS SCREEN NUMEROUS COMPOUNDS TO UNDERSTAND DRUG EFFICACY AND DISCARD INEFFECTIVE ONES. 90% OF DRUG CANDIDATES IN THE PIPELINE WILL FAIL, IN PART DUE TO POOR CLINICAL TRANSLATION. THE PRODUCT AIMS TO HELP COMPANIES ELIMINATE INEFFICACIOUS DRUGS EARLIER BEFORE MILLIONS OF DOLLARS ARE SPENT ON THEIR DEVELOPMENT. THE PROPOSED PROJECT WILL DEVELOP A MORE PHYSIOLOGICALLY RELEVANT, CONSISTENT, AND VERSATILE HIGH-THROUGHPUT SCREENING (HTS) MODEL TO IMPROVE THE TRANSLATION RATE BETWEEN PRECLINICAL AND CLINICAL TESTING. CURRENT DRUG SCREENING APPROACHES, WHICH RELY ON TWO-DIMENSIONAL (2D) CELL CULTURES AND THREE-DIMENSIONAL (3D) CELL AGGREGATES, FAIL TO PROVIDE THE NECESSARY MICROENVIRONMENTAL CUES FOR ACCURATELY REPLICATING HUMAN PATIENT DRUG RESPONSES. THEREFORE, THE PROPOSED TECHNOLOGY HAS BEEN DEVELOPED, AIMING TO STRIKE A BALANCE BETWEEN THROUGHPUT, WHICH INCLUDES SCALABILITY AND UNIFORMITY, AND PHYSIOLOGICAL RELEVANCE, SUCH AS THE ABILITY TO MODULATE KEY ATTRIBUTES OF THE TUMOR MICROENVIRONMENT. UTILIZING A TISSUE ENGINEERING TOOLSET, THE 3D HYDROGEL SCAFFOLD OFFERS A MORE PHYSIOLOGICALLY PERTINENT MICROENVIRONMENT FOR CELL GROWTH AND DRUG RESPONSE. MEANWHILE, A PATENTED MICROFLUIDIC CELL ENCAPSULATION PLATFORM ALLOWS FOR THE RAPID AND CONSISTENT PRODUCTION OF THE PRODUCTS, WHICH ARE ESSENTIAL FOR HTS APPLICATIONS.
