HOWEVER, MANY EDUCATORS AND CLASSROOMS ARE NOT ADEQUATELY SUPPORTED WITH THE FUNDING, MATERIALS, OR TRAINING NECESSARY TO CONDUCT AND ASSESS EFFECTIVE LEARNING ACTIVITIES IN THESE AREAS. FURTHERMORE, EVEN WITH SUCH SUPPORT, MANY CONCEPTS ARE DIFFICULT TO GRASP. HOWEVER, THE USE OF INTERACTIVE DIGITAL MEDIA (IDM), SPECIFICALLY AUGMENTED REALITY (AR) HOLDS GREAT PROMISE AND SHOWS RESULTS FOR ADDRESSING THIS GAP. THIS PROJECT’S LONG-TERM OBJECTIVE IS TO UTILIZE AR TO PROVIDE INTERACTIVE, COLLABORATIVE GAME-BASED ACTIVITIES THAT ARE FEASIBLE FOR DELIVERY ACROSS EDUCATIONAL SETTINGS AND IN PARTICULAR FOR LOW RESOURCE COMMUNITIES. FURTHERMORE, WITH EACH CONTENT AREA STUDENTS AND TEACHERS WILL HAVE THE OPPORTUNITY TO APPRECIATE THE NATURE AND BEHAVIOR OF SCIENTIFIC SYSTEMS WHILE ALSO SOLVING PROBLEMS THROUGH DESIGN-THINKING FOR REAL-LIFE APPLICATIONS. THE PROJECT ALIGNS WITH THE NIH MISSION BY DEVELOPING THE SCIENTIFIC HUMAN RESOURCES AT THE CRITICAL ELEMENTARY AGE WITH THE KNOWLEDGE AND SKILL SETS NECESSARY TO STUDY, UNDERSTAND, AND IMPACT SYSTEMS FOR BETTER HEALTH OUTCOMES. THIS SBIR PHASE II PROJECT WILL EXPAND THE IMPACT OF THE FRAMEWORK AND INFRASTRUCTURE FROM THE MOBILE APPLICATION DEVELOPED IN PHASE I FOR SIMULATION-BASED LEARNING; DESIGN AND CRITICAL-THINKING; AND INCREASING DIVERSITY AND INCLUSION IN STEM. THE FIRST SPECIFIC AIM FOCUSES ON THE FEEDBACK CAPABILITIES. EACH INTERACTION PROVIDES EVIDENCE TO TRIGGER FORMATIVE FEEDBACK FOR THE STUDENT AND REALTIME SUPPORT BASED ON STUDENTS' AR ACTIONS AND REQUESTS. THE SECOND SPECIFIC AIM DEVELOPS THE VISUALIZATION DASHBOARD TO PROVIDE ACTIONABLE INSIGHTS FOR EDUCATORS. THESE FIRST TWO AIMS PROPOSE TO RESEARCH, DEVELOP, AND REFINE THE MACHINE LEARNING OR ARTIFICIAL INTELLIGENCE ALGORITHMS AND USER INTERFACES FOR CRITICAL STUDENT FEEDBACK AND EDUCATOR DASHBOARDS FOR CONTENT SPECIFIC KNOWLEDGE AND DESIGN AND PROBLEM-SOLVING PROCESSES, TARGETING TROUBLESHOOTING AND PROTOTYPING. THE THIRD SPECIFIC AIM IS TO DEVELOP THE SOFTWARE AND HARDWARE SYSTEMS NECESSARY TO INCREASE THE IMPACT OF THE TECHNOLOGY THROUGH BOTH CURRICULAR EXPANSION AND HARDWARE COMPATIBILITY. ADDITIONALLY, EDUCATIONAL INTERVENTIONS THAT HELP CLOSE THE PARTICIPATION AND ACHIEVEMENT GAP OF UNDERREPRESENTED DEMOGRAPHIC GROUPS IN STEM ARE SORELY NEEDED. IN THIS NIGMS PHASE II SBIR PROJECT, D’VINCI INTERACTIVE, IN COLLABORATION WITH THE JACKSON LABORATORY, PROPOSES TO DEVELOP AND EVALUATE A FULL-FUNCTION PROTOTYPE OF THE “BLUEPRINT OF LIFE” ONLINE EDUCATIONAL SOLUTION, WHICH IS AIMED AT EMBEDDING GENOMICS INTO MIDDLE SCHOOL AND HIGH SCHOOL CLASSROOMS. THE SOLUTION WILL FEATURE AN EXPANDED, NGSS-ALIGNED SUPPLEMENTAL CURRICULUM THAT TEACHES KEY CONCEPTS AROUND MOLECULAR GENETICS OF PERSONALIZED MEDICINE, USE OF BIOINFORMATICS TOOLS, THE VIRAL GENOME (PARTICULARLY WITH REGARD TO THE COVID-19 PANDEMIC), CAREERS IN GENETICS SCIENCE, AND THE ETHICS OF GENETICS RESEARCH. THE CURRICULUM WILL BE DELIVERED VIA AN ONLINE STUDENT LEARNING PORTAL DESIGNED TO PROVIDE LEARNING EXPERIENCES THAT ARE ENGAGING, INSPIRATIONAL, ACCESSIBLE, INCLUSIVE, AND RELEVANT TO STUDENTS’ LIVES, WITH FEATURES SUCH AS AN IMMERSIVE 3D/VR GENOME EXPLORATION MODULE AND AN IMMERSIVE VIRTUAL LABORATORY ENVIRONMENT (WHICH TOGETHER REPRESENT THE CORE OF THE PROJECT’S TECHNICAL INNOVATION), CREATIVE CONTENT THAT APPEALS TO UNDERREPRESENTED DEMOGRAPHIC GROUPS, A SPANISH-LANGUAGE CURRICULUM VERSION, AND ADAPTIVE LEARNING TECHNOLOGY. EDUCATORS WILL HAVE ACCESS TO A LEARNING MANAGEMENT SYSTEM, ONLINE CLASSROOM TOOLS THAT ARE EASY TO IMPLEMENT BOTH VIRTUALLY AND IN-CLASS, AND AN ADMINISTRATIVE DASHBOARD FOR REAL-TIME SCHOOL AND SCHOOL-DISTRICT LEVEL REPORTING ON STUDENT LEARNING PROGRESS. THE AIMS FOR THIS PHASE II PROJECT ARE TO SHOW THAT THE SOLUTION DELIVERS THE FOLLOWING LEARNER BENEFITS: (1) EFFECTIVE MASTERY OF THE FUNDAMENTALS OF GENOMICS/GENETICS AND PERSONALIZED MEDICINE, (2) EFFECTIVE UNDERSTANDING OF THE ETHICAL AND HEALTHCARE IMPLICATIONS OF GENETICS/GENOMICS, (3) MOTIVATIONAL IMPACT ON STUDENTS, BY PROMOTING INTEREST IN GENOMICS/STEM EDUCATION AND CAREERS, (4) STRONG LEARNING AND MOTIVATIONAL IMPACT ON UNDERREPRESENTED DEMOGRAPHIC GROUPS. WESTED, WILL CONDUCT TO VERIFY THE ATTAINMENT OF THESE AIMS, THE EXTERNAL RESEARCH AND EVALUATION PARTNER, A RANDOMIZED CONTROLLED TRIAL (WITH 750 9TH GRADE STUDENTS FROM CLASSROOMS WITH HIGH POPULATIONS OF UNDERSERVED STUDENTS) ON THE FULLY DEVELOPED EDUCATIONAL SOLUTION. AIMS 1-3 ARE SUPPORTED IF STUDENTS IN THE TREATMENT GROUP SHOW A PRE- VERSUS POST-ASSESSMENT PERFORMANCE IMPROVEMENT (MEASURED IN TERMS OF INFORMATION RETENTION (AIM 1), ATTITUDES TOWARDS THE PRACTICAL USE OF SCIENCE (AIM 2), AND INTEREST IN PURSUING ADDITIONAL GENOMICS/STEM EDUCATION AND A GENOMICS/STEM CAREER (AIM 3)) THAT IS STATISTICALLY LARGER THAN THE IMPROVEMENTS OBSERVED IN THE CONTROL GROUP. AIM 4 IS SUPPORTED IF PERFORMANCE IN THE TREATMENT GROUP IS UNCORRELATED WITH DEMOGRAPHIC FACTORS (I.E. MALES VS. FEMALES AND MINORITY VS. NON-MINORITY STUDENTS). OVER THE LAST FIVE YEARS AS PART OF OUR PREVIOUS SEPA AWARD, WE HAVE DEVELOPED AN AWARD-WINNING APPROACH TO INFORMAL BIOLOGY EDUCATION, BIOTINKERING, THAT AUTHENTICALLY ENGAGES YOUNG PEOPLE WITH SCIENCE AS A PERSONALLY RELEVANT AND CREATIVE PROCESS BY SUPPORTING LEARNER AGENCY AND CHOICE. OUR PROJECT, ADAPTING AND SCALING THE BIOTINKERING APPROACH THROUGH A COMMUNITY OF PRACTICE (COP) MODEL, AIMS TO EMPOWER OTHER INFORMAL SCIENCE INSTITUTIONS TO PARTICIPATE IN AND CO-AUTHOR THE FUTURE OF BIOTINKERING WITH US. TO BE THOUGHTFUL ABOUT ENSURING THAT OUR BURGEONING APPROACH CAN MEANINGFULLY CONTRIBUTE TO BRINGING MORE DIVERSE PERSPECTIVES INTO STEM FIELDS, OUR PROPOSED COP CENTERS COMMUNITY AND CO-DESIGN. COMMUNITY RELATIONSHIPS AT EACH PARTICIPATING SITE WILL BE INTEGRATED THROUGHOUT THE PROJECT TO HELP BUILD A FOUNDATION FOR BIOTINKERING THAT IS MORE INCLUSIVE AND CULTURALLY RELEVANT, ESPECIALLY FOR MARGINALIZED COMMUNITIES. TO REACH OUR DESIRED IMPACT, WE HAVE THE FOLLOWING 3 AIMS. FIRST, WE WILL ESTABLISH A THRIVING BIOTINKERING COMMUNITY OF PRACTICE TO SUPPORT DIVERSE PRACTITIONERS IN GAINING CONFIDENCE WITH DOING BIOTINKERING IN THEIR OWN CONTEXTS. BY USING A COP MODEL TO DO INSTITUTIONAL CAPACITY BUILDING, WE CAN CREATE LASTING IMPACT ON THE FIELD BY BUILDING A DISPERSED NETWORK OF BIOTINKERING HUBS EQUIPPED TO PROVIDE ONGOING SUPPORT AND MENTORSHIP TO OTHER PRACTITIONERS. SECOND, ALL FOUR FOUNDING MEMBERS OF OUR BIOTINKERING COP WILL DEVELOP CULTURALLY RELEVANT BIOTINKERING ACTIVITIES, CO-DESIGNED WITH LOCAL COMMUNITIES, TO GENERATE A MORE DIVERSE REPERTOIRE OF EXPERIENCES FOR VARIED AUDIENCES AND ENVIRONMENTS. THIS WILL RESULT IN HANDS-ON BIOTINKERING EXPERIENCES BEING IMPLEMENTED AROUND THE NATION, REACHING TENS OF THOUSANDS OF YOUNG PEOPLE AND PROVIDING LEARNERS WITH PATHWAYS TO DEVELOP STRONGER STEM IDENTITIES. AND THIRD, WE WILL IDENTIFY STRATEGIES AND PROMISING PRACTICES THAT CAN BE USED BY PRACTITIONERS TO ADOPT BIOTINKERING AS AN INCLUSIVE APPROACH THAT CENTERS COMMUNITY VOICES. OUR NEW COMMUNITY AND INCLUSION-FOCUSED RESOURCES, FORGED FROM DIVERSE COMMUNITY PERSPECTIVES AND CO-DESIGN LEARNINGS, WILL BETTER SUPPORT THE ADAPTATION OF BIOTINKERING TO DIVERSE SITUATIONS AND CONTEXTS, MAKING IT EASIER TO FURTHER SCALE THIS WORK TO ADDITIONAL INFORMAL LEARNING SITES AND NEW COMMUNITIES. THIS APPROACH CAN GENERATE THE TYPES OF INFORMAL LEARNING EXPERIENCES SOCIETY NEEDS TO SERVE AS INSPIRATIONAL ENTRY POINTS FOR A MORE DIVERSE NEXT GENERATION OF BIOTECH AND BIOMEDICAL INNOVATORS. DESPITE THE RAPIDLY GROWING AGING POPULATION AND THE HIGH COST AND CONSEQUENCES OF AD AND MILD COGNITIVE IMPAIRMENT (MCI), AD AND MCI CLINICAL TRIALS HAVE BEEN LARGELY UNSUCCESSFUL, COSTLY, AND HAVE NOT BEEN ABLE TO DEMONSTRATE CLINICALLY MEANINGFUL EFFECTS SUCH AS DELAYING THE ONSET OR PROGRESSION OF CLINICAL SYMPTOMS. ONE SIGNIFICANT BARRIER TO SUCCESSFUL DEMENTIA PREVENTION IS THAT CURRENTLY AVAILABLE OUTCOME MEASURES USED IN MCI CLINICAL TRIALS ARE UNABLE TO DETECT MEANINGFUL CHANGES IN COGNITION AND FUNCTION THAT OCCUR IN MCI, RESULTING IN SMALL EFFECT SIZES THAT REQUIRE LARGE SAMPLE SIZES FOR CLINICAL TRIALS. DEVELOPMENT OF NEW ASSESSMENT TOOLS THAT ARE SENSITIVE TO THE EARLIEST CHANGES IN CLINICALLY MEANINGFUL COGNITION AND FUNCTION ARE DESPERATELY NEEDED TO TRANSFORM MCI CLINICAL TRIALS AND LEAD TO SUCCESSFUL DEMENTIA PREVENTION. TO ADDRESS THE CRITICAL NEED FOR IMPROVED OUTCOME MEASURES FOR MCI CLINICAL TRIALS THAT INCORPORATE FOOD AND DRUG ADMINISTRATION (FDA) RECOMMENDATIONS AND NATIONAL INSTITUTE ON AGING (NIA) STRATEGIC PRIORITIES, WE WILL DESIGN, DEVELOP, AND TEST A NOVEL COMPUTERIZED ASSESSMENT TOOL THAT COMBINES CLINICALLY MEANINGFUL COGNITION AND FUNCTION TO IMPROVE OUTCOME MEASURES IN MCI CLINICAL TRIALS. THE “COGNITION AND FUNCTION IN AN IADL TASK” (C-FIT) ASSESSMENT TOOL WILL BE EASY TO SELF-ADMINISTER VIA SPEECH OR TOUCHSCREEN INPUT; RELIABLE AND REPEATABLE; AND EFFECTIVE IN DETECTING EARLY AND MEANINGFUL COGNITIVE AND FUNCTIONAL CHANGES IN OLDER ADULTS WITH MCI. THE GOAL OF THE C-FIT ASSESSMENT TOOL IS TO IMPROVE EARLY DETECTION OF MEANINGFUL CHANGE IN COGNITION AND FUNCTION IN MCI CLINICAL TRIALS. IN THIS SBIR PHASE I PROJECT WE WILL EXECUTE THREE AIMS: AIM 1: DESIGN AND DEVELOP THE C-FIT ASSESSMENT TOOL. AIM 2: INITIAL TEST OF THE C-FIT ASSESSMENT TOOL. AIM 3: DEMONSTRATE INITIAL RELIABILITY AND VALIDITY OF THE C-FIT ASSESSMENT TOOL. TO ACHIEVE THESE AIMS, WE WILL DESIGN A NOVEL COMPUTERIZED ASSESSMENT THAT EVALUATES KEY COGNITIVE AND FUNCTIONAL DOMAINS THAT ARE MOST SENSITIVE TO MCI. WE WILL TEST THE C-FIT ASSESSMENT TOOL WITH 10 ADULTS AGES 65 OR OLDER: 5 WHO ARE COGNITIVELY HEALTHY AND 5 WHO HAVE A CLINICAL DIAGNOSIS OF MCI. WE WILL UTILIZE A USER-CENTERED DESIGN PROCESS AND ITERATIVELY IMPROVE THE C-FIT ASSESSMENT USING PARTICIPANT FEEDBACK. WE WILL CONDUCT PSYCHOMETRIC TESTING WITH A NEW SAMPLE OF 30 ADULTS AGES 65 OR OLDER: 15 WHO ARE COGNITIVELY HEALTHY AND 15 WHO HAVE A CLINICAL DIAGNOSIS OF MCI. TO ASSESS TEST-RETEST RELIABILITY, PARTICIPANTS WILL COMPLETE THE C-FIT ASSESSMENT TWICE, TWO WEEKS APART. SCORES AT EACH TIMEPOINT WILL BE COMPARED USING INTRACLASS CORRELATION COEFFICIENTS. NO EFFECTIVE THERAPEUTICS EXIST TO COUNTER THE ADDICTION; CURRENT OUD THERAPEUTICS SUCH AS METHADONE, BUPRENORPHINE, AND NALTREXONE ARE OPIOID MIMETICS, EITHER EXERTING WEAKER POTENCY OR BLOCKING THE OPIOIDS FROM BINDING WITH THEIR COGNATE RECEPTORS. EPIGENETIC MODIFICATIONS ON HISTONE MARKS PLAY KEY CAUSATIVE ROLES IN MEDIATING OPIOID ADDICTION. SUCH EPIGENETIC MODIFICATIONS GENERATE “PERSISTING IMPACTS” ON THE REWARD CIRCUITRY OF THE BRAIN BY ALTERING CHROMATIN STRUCTURE AND SYNAPTIC REWIRING. UNFORTUNATELY, CURRENT EPIGENETIC DRUGS ARE NOT SUITABLE FOR DRUG ADDICTION THERAPEUTICS. THE FUNDAMENTAL PROBLEM IS THAT EPIGENETIC REGULATORS PLAY NUMEROUS ROLES IN DIFFERENT TISSUES AND UNDER DIVERSE CELLULAR CONTEXTS; THUS, ENZYME-INHIBITORY EPIGENETIC DRUGS TEND TO CAUSE VERY HIGH OFF-TARGET TOXICITIES THAT ARE UNACCEPTABLE OUTSIDE OF ONCOLOGICAL INDICATIONS. RECENTLY, WE DEVELOPED A NOVEL APPROACH THAT IDENTIFIED HIGH-PRECISION SMALL-MOLECULES BY TARGETING PROTEIN-PROTEIN INTERACTIONS (PPI) OF AN EPIGENETIC PROTEIN COMPLEX INVOLVING HISTONE DEACETYLASE (HDAC). IN PARTICULAR, WE DEMONSTRATED A SIN3-BINDING MOLECULE THAT DISRUPTS THE HDAC SIN3 COMPLEX RECRUITMENT TO CHROMOSOMAL LOCI. THIS MOLECULE COULD BLOCK LSD-INDUCED HALLUCINATION AS EFFECTIVELY AS A GENETIC HDAC2 KNOCKOUT IN THE MOUSE PREFRONTAL CORTEX, WHILE ONLY MINIMALLY INDUCING OFF-TARGET GENE EXPRESSION. BY ADAPTING THIS APPROACH, WE SEEK TO DEVELOP HIGH-PRECISION PPI-TARGETING MOLECULES FOR OUD. THE NEURONAL G9A COMPLEX IS AN ATTRACTIVE TARGET REGULATING ΔFOSB EXPRESSION UNDER CHRONIC OPIOID EXPOSURE. WE WILL APPLY A MAMMALIAN CELL-BASED COMPOUND SCREENING PLATFORM TO IDENTIFY FUNCTIONAL MODULATORS OF THE G9A COMPLEX AND BIOPHYSICALLY CHARACTERIZE THEIR DRUG-PROTEIN INTERACTIONS. BY COMBINING INTERACTIVE GAMEPLAY, MNEMONIC CHARACTERS REPRESENTING MEDICAL CONCEPTS, AND AI-ENABLED NON- PLAYABLE CHARACTERS (NPCS) FOR PERSONALIZED DIALOGUES, THIS APPROACH TARGETS THE EFFECTIVE LEARNING AND RETENTION OF COMPLEX MEDICAL KNOWLEDGE. OUR PRELIMINARY STUDIES INDICATE THE PLATFORM’S EFFICACY ACROSS EDUCATIONAL LEVELS. A SECTION OF THE ENDOCRINE MODULE BOOSTED HIGH SCHOOL STUDENTS' UNDERSTANDING OF PANCREATIC FUNCTIONS BY OVER 200%. FURTHERMORE, MEDICAL STUDENTS EXPOSED TO A SECTION OF THE IMMUNE MODULE OUTPERFORMED THEIR PEERS ON RELATED EXAM QUESTIONS. WE EXPECT THAT LONG-TERM OUTCOMES OF OUR PLATFORM WILL INCLUDE A DECREASE IN DISEASE BURDEN AS HEALTH LITERACY IS THE BEST FORM OF PREVENTATIVE MEDICINE. WE HYPOTHESIZE THAT OUR NOVEL HEALTH SCIENCES EDUCATION PLATFORM, WHICH INTEGRATES GAME-BASED LEARNING, MNEMONICS, AND AI STRATEGIES WILL PROMOTE STUDENT LEARNING AND ENGAGEMENT WITH MEDICAL KNOWLEDGE. THIS WILL BE ASSESSED THROUGH THE FOLLOWING SPECIFIC AIMS. AIM 1: DEVELOP A PILOT PROTOTYPE PLATFORM TO SUPPORT HEALTH EDUCATION. THIS AIM DEVELOPS A PROTOTYPE PLATFORM WITH THREE MODULES CENTERED AROUND THE ENDOCRINE, IMMUNE, AND CIRCULATORY SYSTEMS, INCORPORATING MULTIPLE MNEMONIC-BASED CHARACTERS AND ITEMS. AIM 2: INCORPORATE AI-POWERED MEDIMASTERS INTO THE PROTOTYPE PLATFORM. THIS AIM SEEKS TO INCORPORATE FINE- TUNED GENERATIVE AI TECHNOLOGIES TO PERSONALIZE THE LEARNING EXPERIENCE, CREATING NPC MEDIMASTERS THAT ADAPT CONTENT AND CHALLENGES BASED ON THE INDIVIDUAL LEARNER. AIM 3: ASSESS PLATFORM PROTOTYPE FEASIBILITY TO ENHANCE STUDENT ENGAGEMENT AND SATISFACTION WITH HEALTH EDUCATION MATERIAL. IN ADDITION, TRANSPORTATION MAY NOT BE FEASIBLE DEPENDING ON TRAVEL DISTANCE, 33,34 AND THE TYPES OF INTERVENTIONS DO NOT ADDRESS THE MOTIVATIONAL FACTORS FOR AAS THAT INTERFERE WITH ADHERENCE TO EVIDENCE-BASED RECOMMENDATIONS. FIT FAMILIES, THE INTERVENTION ON WHICH OUR PROPOSAL IS BASED, ADDRESSES ACCESS ISSUES THROUGH AN AT-HOME EVIDENCE-BASED INTERVENTION. THIS PROPOSAL AIMS TO EXPAND THE FIT FAMILIES INTERVENTION THROUGH OUR INNOVATION THE DIGITAL WELLNESS NURSE (DWN)-FIT THAT UTILIZES MOBILE TECHNOLOGY AND AUTOMATION TO INCREASE ACCESS, ADHERENCE AND ENGAGEMENT IN HEALTHY BEHAVIORS USING HUMAN-IN-THE-LOOP AUTOMATION, WEB, AND MOBILE TECHNOLOGY TO DELIVER EVIDENCE-BASED INTERVENTIONS. AS DEFINED BY THE 2021 CENSUS, WINDHAM COUNTY (WC) IS 70.94% RURAL. LOCATED IN SOUTH-EASTERN VERMONT, THE POPULATION OF WC IS 45,966. DUE TO ITS RURAL NATURE, WC IS DEFINED AS AN UNDERSERVED COMMUNITY. DATA PROVIDED DETAILS HIGHER PREVALENCE OF BOTH CANNABIS AND PRESCRIPTION DRUG USE AMONG RURAL WC YOUTH AND YOUNG ADULTS, LOW SES AND THE LGB POPULATION. WCPP WILL UTILIZE A COMPREHENSIVE APPROACH INCLUDING INFORMATION DISSEMINATION, EDUCATION, ALTERNATIVES, POLICY CHANGE, ID AND REFERRAL AND COMMUNITY-BASED PROCESSES. DECREASE UNDERAGED ALCOHOL USE THROUGH A MULTI-STRATEGY APPROACH BASED ON THE STRATEGIC PREVENTION FRAMEWORK. THE POPULATION OF FOCUS INCLUDES YOUTH 12-20 LIVING IN THE 7 TOWNS THAT MAKE UP THE WEST RIVER VALLEY, AS WELL AS ADULTS, PRIMARILY PARENTS AND CAREGIVERS, IN THESE TOWNS WITH WHOM THE COMMUNITY NORMS AROUND DRINKING AND YOUTH ACCESS TO ALCOHOL CAN BE ADDRESSED AND CHANGED. THE PROJECTED NUMBER OF PERSONS SERVED ANNUALLY INCLUDES: 500 PARENTS, 300 YOUTH 12 - 20 AND 2,000 COMMUNITY MEMBERS. WE ESTIMATE TOTALS REACHED DURING THE LIFE OF THE PROJECT TO BE: 2,000 PARENTS, 1,200 YOUTH AND 8,000 COMMUNITY MEMBERS. OBJECTIVES INCLUDE: INCREASE PARENT AND CARING ADULT INVOLVEMENT IN THE COALITION AND CAPACITY TO REDUCE UNDERAGE DRINKING; INCREASE PERCEPTION OF HARM AND LOWER EASE OF ACCESS TO DECREASE PAST 30-DAY USE; INCREASE ADULT AWARENESS OF RISKS AND CONSEQUENCES ASSOCIATED WITH, AND BEST PRACTICES TO PREVENT, YOUTH ACCESS TO ALCOHOL; ADDRESS ENVIRONMENTAL FACTORS AND COMMUNITY NORMS THAT CONTRIBUTE TO PERCEIVED ACCEPTANCE OF UNDERAGE DRINKING. AUGMENTED REALITY PLATFORM FOR FEEDBACK AND ASSESSMENT IN STEM ELEMENTARY EDUCATION - DELIVERING MEANINGFUL, PROJECT-BASED LEARNING FOR SCIENCE AND ENGINEERING IN ELEMENTARY CLASSROOMS IS INCREASINGLY RECOGNIZED AS ESSENTIAL FOR TRAINING THE WORKFORCE REQUIRED TO SOLVE OUR MOST PRESSING HEALTH AND ENVIRONMENTAL PROBLEMS. HOWEVER, MANY EDUCATORS AND CLASSROOMS ARE NOT ADEQUATELY SUPPORTED WITH THE FUNDING, MATERIALS, OR TRAINING NECESSARY TO CONDUCT AND ASSESS EFFECTIVE LEARNING ACTIVITIES IN THESE AREAS. FURTHERMORE, EVEN WITH SUCH SUPPORT, MANY CONCEPTS ARE DIFFICULT TO GRASP. HOWEVER, THE USE OF INTERACTIVE DIGITAL MEDIA (IDM), SPECIFICALLY AUGMENTED REALITY (AR) HOLDS GREAT PROMISE AND SHOWS RESULTS FOR ADDRESSING THIS GAP. THIS PROJECT’S LONG-TERM OBJECTIVE IS TO UTILIZE AR TO PROVIDE INTERACTIVE, COLLABORATIVE GAME-BASED ACTIVITIES THAT ARE FEASIBLE FOR DELIVERY ACROSS EDUCATIONAL SETTINGS AND IN PARTICULAR FOR LOW RESOURCE COMMUNITIES. FURTHERMORE, WITH EACH CONTENT AREA STUDENTS AND TEACHERS WILL HAVE THE OPPORTUNITY TO APPRECIATE THE NATURE AND BEHAVIOR OF SCIENTIFIC SYSTEMS WHILE ALSO SOLVING PROBLEMS THROUGH DESIGN-THINKING FOR REAL-LIFE APPLICATIONS. THE PROJECT ALIGNS WITH THE NIH MISSION BY DEVELOPING THE SCIENTIFIC HUMAN RESOURCES AT THE CRITICAL ELEMENTARY AGE WITH THE KNOWLEDGE AND SKILL SETS NECESSARY TO STUDY, UNDERSTAND, AND IMPACT SYSTEMS FOR BETTER HEALTH OUTCOMES. THIS SBIR PHASE II PROJECT WILL EXPAND THE IMPACT OF THE FRAMEWORK AND INFRASTRUCTURE FROM THE MOBILE APPLICATION DEVELOPED IN PHASE I FOR SIMULATION-BASED LEARNING; DESIGN AND CRITICAL-THINKING; AND INCREASING DIVERSITY AND INCLUSION IN STEM. THE FIRST SPECIFIC AIM FOCUSES ON THE FEEDBACK CAPABILITIES. EACH INTERACTION PROVIDES EVIDENCE TO TRIGGER FORMATIVE FEEDBACK FOR THE STUDENT AND REALTIME SUPPORT BASED ON STUDENTS' AR ACTIONS AND REQUESTS. THE SECOND SPECIFIC AIM DEVELOPS THE VISUALIZATION DASHBOARD TO PROVIDE ACTIONABLE INSIGHTS FOR EDUCATORS. THESE FIRST TWO AIMS PROPOSE TO RESEARCH, DEVELOP, AND REFINE THE MACHINE LEARNING OR ARTIFICIAL INTELLIGENCE ALGORITHMS AND USER INTERFACES FOR CRITICAL STUDENT FEEDBACK AND EDUCATOR DASHBOARDS FOR CONTENT SPECIFIC KNOWLEDGE AND DESIGN AND PROBLEM-SOLVING PROCESSES, TARGETING TROUBLESHOOTING AND PROTOTYPING. THE THIRD SPECIFIC AIM IS TO DEVELOP THE SOFTWARE AND HARDWARE SYSTEMS NECESSARY TO INCREASE THE IMPACT OF THE TECHNOLOGY THROUGH BOTH CURRICULAR EXPANSION AND HARDWARE COMPATIBILITY. THE CURRICULAR AREAS OF FOCUS WOULD INCLUDE MAGNETISM, SOUND (AUDITORY) AND LIGHT WAVES (OPTICAL) AS WELL AS BIOENGINEERING. THE SOLUTION WILL FEATURE AN EXPANDED, NGSS-ALIGNED SUPPLEMENTAL CURRICULUM THAT TEACHES KEY CONCEPTS AROUND MOLECULAR GENETICS OF PERSONALIZED MEDICINE, USE OF BIOINFORMATICS TOOLS, THE VIRAL GENOME (PARTICULARLY WITH REGARD TO THE COVID-19 PANDEMIC), CAREERS IN GENETICS SCIENCE, AND THE ETHICS OF GENETICS RESEARCH. THE CURRICULUM WILL BE DELIVERED VIA AN ONLINE STUDENT LEARNING PORTAL DESIGNED TO PROVIDE LEARNING EXPERIENCES THAT ARE ENGAGING, INSPIRATIONAL, ACCESSIBLE, INCLUSIVE, AND RELEVANT TO STUDENTS’ LIVES, WITH FEATURES SUCH AS AN IMMERSIVE 3D/VR GENOME EXPLORATION MODULE AND AN IMMERSIVE VIRTUAL LABORATORY ENVIRONMENT (WHICH TOGETHER REPRESENT THE CORE OF THE PROJECT’S TECHNICAL INNOVATION), CREATIVE CONTENT THAT APPEALS TO UNDERREPRESENTED DEMOGRAPHIC GROUPS, A SPANISH-LANGUAGE CURRICULUM VERSION, AND ADAPTIVE LEARNING TECHNOLOGY. EDUCATORS WILL HAVE ACCESS TO A LEARNING MANAGEMENT SYSTEM, ONLINE CLASSROOM TOOLS THAT ARE EASY TO IMPLEMENT BOTH VIRTUALLY AND IN-CLASS, AND AN ADMINISTRATIVE DASHBOARD FOR REAL-TIME SCHOOL AND SCHOOL-DISTRICT LEVEL REPORTING ON STUDENT LEARNING PROGRESS. THE AIMS FOR THIS PHASE II PROJECT ARE TO SHOW THAT THE SOLUTION DELIVERS THE FOLLOWING LEARNER BENEFITS: (1) EFFECTIVE MASTERY OF THE FUNDAMENTALS OF GENOMICS/GENETICS AND PERSONALIZED MEDICINE, (2) EFFECTIVE UNDERSTANDING OF THE ETHICAL AND HEALTHCARE IMPLICATIONS OF GENETICS/GENOMICS, (3) MOTIVATIONAL IMPACT ON STUDENTS, BY PROMOTING INTEREST IN GENOMICS/STEM EDUCATION AND CAREERS, (4) STRONG LEARNING AND MOTIVATIONAL IMPACT ON UNDERREPRESENTED DEMOGRAPHIC GROUPS. WESTED, WILL CONDUCT TO VERIFY THE ATTAINMENT OF THESE AIMS, THE EXTERNAL RESEARCH AND EVALUATION PARTNER, A RANDOMIZED CONTROLLED TRIAL (WITH 750 9TH GRADE STUDENTS FROM CLASSROOMS WITH HIGH POPULATIONS OF UNDERSERVED STUDENTS) ON THE FULLY DEVELOPED EDUCATIONAL SOLUTION. AIMS 1-3 ARE SUPPORTED IF STUDENTS IN THE TREATMENT GROUP SHOW A PRE- VERSUS POST-ASSESSMENT PERFORMANCE IMPROVEMENT (MEASURED IN TERMS OF INFORMATION RETENTION (AIM 1), ATTITUDES TOWARDS THE PRACTICAL USE OF SCIENCE (AIM 2), AND INTEREST IN PURSUING ADDITIONAL GENOMICS/STEM EDUCATION AND A GENOMICS/STEM CAREER (AIM 3)) THAT IS STATISTICALLY LARGER THAN THE IMPROVEMENTS OBSERVED IN THE CONTROL GROUP. AIM 4 IS SUPPORTED IF PERFORMANCE IN THE TREATMENT GROUP IS UNCORRELATED WITH DEMOGRAPHIC FACTORS (I.E. MALES VS. FEMALES AND MINORITY VS. NON-MINORITY STUDENTS). OVERALL, THE ENGAGING AND INSPIRATIONAL LEARNING SOLUTION PROPOSED IN THIS PROJECT WILL HELP BETTER PREPARE TOMORROW’S HEALTHCARE CONSUMERS AND SCIENTISTS AND PROMOTE GREATER DIVERSITY IN STEM EDUCATION AND PRACTICE.

ADAPTING AND SCALING THE BIOTINKERING APPROACH THROUGH A COP MODEL - PROJECT SUMMARY AND ABSTRACT DESPITE THE RAPIDLY ADVANCING LANDSCAPE OF MODERN BIOLOGY, THE MAJORITY OF EXPERIENCES THAT K-12 LEARNERS HAVE WITH BIOLOGY ARE STILL ROOTED IN TRADITIONAL PROCEDURAL AND PRESCRIPTIVE APPROACHES. THERE IS A CRITICAL NEED TO ENGAGE STUDENTS IN A MORE 21ST CENTURY APPROACH TO BIOLOGY – ONE THAT EMPHASIZES OPEN-ENDED EXPLORATION AND CREATIVITY AS KEY ELEMENTS OF SCIENTIFIC PRACTICE. OVER THE LAST FIVE YEARS AS PART OF OUR PREVIOUS SEPA AWARD, WE HAVE DEVELOPED AN AWARD-WINNING APPROACH TO INFORMAL BIOLOGY EDUCATION, BIOTINKERING, THAT AUTHENTICALLY ENGAGES YOUNG PEOPLE WITH SCIENCE AS A PERSONALLY RELEVANT AND CREATIVE PROCESS BY SUPPORTING LEARNER AGENCY AND CHOICE. OUR PROJECT, ADAPTING AND SCALING THE BIOTINKERING APPROACH THROUGH A COMMUNITY OF PRACTICE (COP) MODEL, AIMS TO EMPOWER OTHER INFORMAL SCIENCE INSTITUTIONS TO PARTICIPATE IN AND CO-AUTHOR THE FUTURE OF BIOTINKERING WITH US. TO BE THOUGHTFUL ABOUT ENSURING THAT OUR BURGEONING APPROACH CAN MEANINGFULLY CONTRIBUTE TO BRINGING MORE DIVERSE PERSPECTIVES INTO STEM FIELDS, OUR PROPOSED COP CENTERS COMMUNITY AND CO-DESIGN. COMMUNITY RELATIONSHIPS AT EACH PARTICIPATING SITE WILL BE INTEGRATED THROUGHOUT THE PROJECT TO HELP BUILD A FOUNDATION FOR BIOTINKERING THAT IS MORE INCLUSIVE AND CULTURALLY RELEVANT, ESPECIALLY FOR MARGINALIZED COMMUNITIES. TO REACH OUR DESIRED IMPACT, WE HAVE THE FOLLOWING 3 AIMS. FIRST, WE WILL ESTABLISH A THRIVING BIOTINKERING COMMUNITY OF PRACTICE TO SUPPORT DIVERSE PRACTITIONERS IN GAINING CONFIDENCE WITH DOING BIOTINKERING IN THEIR OWN CONTEXTS. BY USING A COP MODEL TO DO INSTITUTIONAL CAPACITY BUILDING, WE CAN CREATE LASTING IMPACT ON THE FIELD BY BUILDING A DISPERSED NETWORK OF BIOTINKERING HUBS EQUIPPED TO PROVIDE ONGOING SUPPORT AND MENTORSHIP TO OTHER PRACTITIONERS. SECOND, ALL FOUR FOUNDING MEMBERS OF OUR BIOTINKERING COP WILL DEVELOP CULTURALLY RELEVANT BIOTINKERING ACTIVITIES, CO-DESIGNED WITH LOCAL COMMUNITIES, TO GENERATE A MORE DIVERSE REPERTOIRE OF EXPERIENCES FOR VARIED AUDIENCES AND ENVIRONMENTS. THIS WILL RESULT IN HANDS-ON BIOTINKERING EXPERIENCES BEING IMPLEMENTED AROUND THE NATION, REACHING TENS OF THOUSANDS OF YOUNG PEOPLE AND PROVIDING LEARNERS WITH PATHWAYS TO DEVELOP STRONGER STEM IDENTITIES. AND THIRD, WE WILL IDENTIFY STRATEGIES AND PROMISING PRACTICES THAT CAN BE USED BY PRACTITIONERS TO ADOPT BIOTINKERING AS AN INCLUSIVE APPROACH THAT CENTERS COMMUNITY VOICES. OUR NEW COMMUNITY AND INCLUSION-FOCUSED RESOURCES, FORGED FROM DIVERSE COMMUNITY PERSPECTIVES AND CO-DESIGN LEARNINGS, WILL BETTER SUPPORT THE ADAPTATION OF BIOTINKERING TO DIVERSE SITUATIONS AND CONTEXTS, MAKING IT EASIER TO FURTHER SCALE THIS WORK TO ADDITIONAL INFORMAL LEARNING SITES AND NEW COMMUNITIES. THIS APPROACH CAN GENERATE THE TYPES OF INFORMAL LEARNING EXPERIENCES SOCIETY NEEDS TO SERVE AS INSPIRATIONAL ENTRY POINTS FOR A MORE DIVERSE NEXT GENERATION OF BIOTECH AND BIOMEDICAL INNOVATORS. LESSONS LEARNED FROM THE SUCCESSES AND CHALLENGES OF THIS PROJECT CAN ALSO OFFER VALUABLE INSIGHTS ABOUT USING COPS AS A MODEL FOR SELF-SUSTAINING SCALING AND KNOWLEDGE TRANSFER WITHIN THE INFORMAL SCIENCE LEARNING FIELD.

INTERACTIVE SOFTWARE FOR ASSESSING MEANINGFUL COGNITIVE FUNCTION - PROJECT SUMMARY ALZHEIMER’S DISEASE (AD) AND AD-RELATED DEMENTIAS (ADRD) ARE ASSOCIATED WITH SIGNIFICANT DISABILITY AND LOSS OF INDEPENDENCE, BURDEN, AND COSTS TO AFFECTED INDIVIDUALS, FAMILY CAREGIVERS, AND HEALTH CARE SYSTEMS. DESPITE THE RAPIDLY GROWING AGING POPULATION AND THE HIGH COST AND CONSEQUENCES OF AD AND MILD COGNITIVE IMPAIRMENT (MCI), AD AND MCI CLINICAL TRIALS HAVE BEEN LARGELY UNSUCCESSFUL, COSTLY, AND HAVE NOT BEEN ABLE TO DEMONSTRATE CLINICALLY MEANINGFUL EFFECTS SUCH AS DELAYING THE ONSET OR PROGRESSION OF CLINICAL SYMPTOMS. ONE SIGNIFICANT BARRIER TO SUCCESSFUL DEMENTIA PREVENTION IS THAT CURRENTLY AVAILABLE OUTCOME MEASURES USED IN MCI CLINICAL TRIALS ARE UNABLE TO DETECT MEANINGFUL CHANGES IN COGNITION AND FUNCTION THAT OCCUR IN MCI, RESULTING IN SMALL EFFECT SIZES THAT REQUIRE LARGE SAMPLE SIZES FOR CLINICAL TRIALS. DEVELOPMENT OF NEW ASSESSMENT TOOLS THAT ARE SENSITIVE TO THE EARLIEST CHANGES IN CLINICALLY MEANINGFUL COGNITION AND FUNCTION ARE DESPERATELY NEEDED TO TRANSFORM MCI CLINICAL TRIALS AND LEAD TO SUCCESSFUL DEMENTIA PREVENTION. TO ADDRESS THE CRITICAL NEED FOR IMPROVED OUTCOME MEASURES FOR MCI CLINICAL TRIALS THAT INCORPORATE FOOD AND DRUG ADMINISTRATION (FDA) RECOMMENDATIONS AND NATIONAL INSTITUTE ON AGING (NIA) STRATEGIC PRIORITIES, WE WILL DESIGN, DEVELOP, AND TEST A NOVEL COMPUTERIZED ASSESSMENT TOOL THAT COMBINES CLINICALLY MEANINGFUL COGNITION AND FUNCTION TO IMPROVE OUTCOME MEASURES IN MCI CLINICAL TRIALS. THE “COGNITION AND FUNCTION IN AN IADL TASK” (C-FIT) ASSESSMENT TOOL WILL BE EASY TO SELF-ADMINISTER VIA SPEECH OR TOUCHSCREEN INPUT; RELIABLE AND REPEATABLE; AND EFFECTIVE IN DETECTING EARLY AND MEANINGFUL COGNITIVE AND FUNCTIONAL CHANGES IN OLDER ADULTS WITH MCI. THE GOAL OF THE C-FIT ASSESSMENT TOOL IS TO IMPROVE EARLY DETECTION OF MEANINGFUL CHANGE IN COGNITION AND FUNCTION IN MCI CLINICAL TRIALS. IN THIS SBIR PHASE I PROJECT WE WILL EXECUTE THREE AIMS: AIM 1: DESIGN AND DEVELOP THE C-FIT ASSESSMENT TOOL. AIM 2: INITIAL TEST OF THE C-FIT ASSESSMENT TOOL. AIM 3: DEMONSTRATE INITIAL RELIABILITY AND VALIDITY OF THE C-FIT ASSESSMENT TOOL. TO ACHIEVE THESE AIMS, WE WILL DESIGN A NOVEL COMPUTERIZED ASSESSMENT THAT EVALUATES KEY COGNITIVE AND FUNCTIONAL DOMAINS THAT ARE MOST SENSITIVE TO MCI. WE WILL TEST THE C-FIT ASSESSMENT TOOL WITH 10 ADULTS AGES 65 OR OLDER: 5 WHO ARE COGNITIVELY HEALTHY AND 5 WHO HAVE A CLINICAL DIAGNOSIS OF MCI. WE WILL UTILIZE A USER-CENTERED DESIGN PROCESS AND ITERATIVELY IMPROVE THE C-FIT ASSESSMENT USING PARTICIPANT FEEDBACK. WE WILL CONDUCT PSYCHOMETRIC TESTING WITH A NEW SAMPLE OF 30 ADULTS AGES 65 OR OLDER: 15 WHO ARE COGNITIVELY HEALTHY AND 15 WHO HAVE A CLINICAL DIAGNOSIS OF MCI. TO ASSESS TEST-RETEST RELIABILITY, PARTICIPANTS WILL COMPLETE THE C-FIT ASSESSMENT TWICE, TWO WEEKS APART. SCORES AT EACH TIMEPOINT WILL BE COMPARED USING INTRACLASS CORRELATION COEFFICIENTS. TO ASSESS CONCURRENT VALIDITY, PARTICIPANTS WILL COMPLETE VALIDATED COGNITIVE AND FUNCTIONAL TESTS USED IN MCI CLINICAL TRIALS. SCORES WILL BE COMPARED USING PEARSON CORRELATION COEFFICIENTS.

DEVELOPING HIGH-PRECISION EPIGENETIC THERAPEUTICS FOR OPIOID USE DISORDER - ABSTRACT OPIOID ABUSE DISORDER (OUD) IS A GROWING EPIDEMIC THAT DISRUPTS NUMEROUS LIVES AND LOCAL COMMUNITIES IN THE UNITED STATES. NO EFFECTIVE THERAPEUTICS EXIST TO COUNTER THE ADDICTION; CURRENT OUD THERAPEUTICS SUCH AS METHADONE, BUPRENORPHINE, AND NALTREXONE ARE OPIOID MIMETICS, EITHER EXERTING WEAKER POTENCY OR BLOCKING THE OPIOIDS FROM BINDING WITH THEIR COGNATE RECEPTORS. EPIGENETIC MODIFICATIONS ON HISTONE MARKS PLAY KEY CAUSATIVE ROLES IN MEDIATING OPIOID ADDICTION. SUCH EPIGENETIC MODIFICATIONS GENERATE “PERSISTING IMPACTS” ON THE REWARD CIRCUITRY OF THE BRAIN BY ALTERING CHROMATIN STRUCTURE AND SYNAPTIC REWIRING. UNFORTUNATELY, CURRENT EPIGENETIC DRUGS ARE NOT SUITABLE FOR DRUG ADDICTION THERAPEUTICS. THE FUNDAMENTAL PROBLEM IS THAT EPIGENETIC REGULATORS PLAY NUMEROUS ROLES IN DIFFERENT TISSUES AND UNDER DIVERSE CELLULAR CONTEXTS; THUS, ENZYME-INHIBITORY EPIGENETIC DRUGS TEND TO CAUSE VERY HIGH OFF-TARGET TOXICITIES THAT ARE UNACCEPTABLE OUTSIDE OF ONCOLOGICAL INDICATIONS. RECENTLY, WE DEVELOPED A NOVEL APPROACH THAT IDENTIFIED HIGH-PRECISION SMALL-MOLECULES BY TARGETING PROTEIN-PROTEIN INTERACTIONS (PPI) OF AN EPIGENETIC PROTEIN COMPLEX INVOLVING HISTONE DEACETYLASE (HDAC). IN PARTICULAR, WE DEMONSTRATED A SIN3-BINDING MOLECULE THAT DISRUPTS THE HDAC SIN3 COMPLEX RECRUITMENT TO CHROMOSOMAL LOCI. THIS MOLECULE COULD BLOCK LSD-INDUCED HALLUCINATION AS EFFECTIVELY AS A GENETIC HDAC2 KNOCKOUT IN THE MOUSE PREFRONTAL CORTEX, WHILE ONLY MINIMALLY INDUCING OFF-TARGET GENE EXPRESSION. BY ADAPTING THIS APPROACH, WE SEEK TO DEVELOP HIGH-PRECISION PPI-TARGETING MOLECULES FOR OUD. THE NEURONAL G9A COMPLEX IS AN ATTRACTIVE TARGET REGULATING ΔFOSB EXPRESSION UNDER CHRONIC OPIOID EXPOSURE. THE EFFICACY AND SPECIFICITY OF THE FINAL HITS WILL BE TESTED IN OUD ANIMAL MODELS WHERE BEHAVIORAL RESPONSES AND BIOMARKERS IN THE BRAIN WILL BE EXAMINED. THESE METHODS INTRODUCE INTERACTIVE LEARNING, IMPROVE MEMORY RECALL, AND OFFER PERSONALIZED EDUCATIONAL EXPERIENCES, ADDRESSING THE CHALLENGES OF CONVEYING COMPLEX MEDICAL KNOWLEDGE EFFECTIVELY. OUR PROJECT INTEGRATES THESE THREE PEDAGOGICAL TECHNIQUES INTO A PLATFORM FOR HEALTH SCIENCES EDUCATION. BY COMBINING INTERACTIVE GAMEPLAY, MNEMONIC CHARACTERS REPRESENTING MEDICAL CONCEPTS, AND AI-ENABLED NON- PLAYABLE CHARACTERS (NPCS) FOR PERSONALIZED DIALOGUES, THIS APPROACH TARGETS THE EFFECTIVE LEARNING AND RETENTION OF COMPLEX MEDICAL KNOWLEDGE. OUR PRELIMINARY STUDIES INDICATE THE PLATFORM’S EFFICACY ACROSS EDUCATIONAL LEVELS. A SECTION OF THE ENDOCRINE MODULE BOOSTED HIGH SCHOOL STUDENTS' UNDERSTANDING OF PANCREATIC FUNCTIONS BY OVER 200%. FURTHERMORE, MEDICAL STUDENTS EXPOSED TO A SECTION OF THE IMMUNE MODULE OUTPERFORMED THEIR PEERS ON RELATED EXAM QUESTIONS. WE EXPECT THAT LONG-TERM OUTCOMES OF OUR PLATFORM WILL INCLUDE A DECREASE IN DISEASE BURDEN AS HEALTH LITERACY IS THE BEST FORM OF PREVENTATIVE MEDICINE. WE HYPOTHESIZE THAT OUR NOVEL HEALTH SCIENCES EDUCATION PLATFORM, WHICH INTEGRATES GAME-BASED LEARNING, MNEMONICS, AND AI STRATEGIES WILL PROMOTE STUDENT LEARNING AND ENGAGEMENT WITH MEDICAL KNOWLEDGE. THIS WILL BE ASSESSED THROUGH THE FOLLOWING SPECIFIC AIMS. AIM 1: DEVELOP A PILOT PROTOTYPE PLATFORM TO SUPPORT HEALTH EDUCATION. THIS AIM DEVELOPS A PROTOTYPE PLATFORM WITH THREE MODULES CENTERED AROUND THE ENDOCRINE, IMMUNE, AND CIRCULATORY SYSTEMS, INCORPORATING MULTIPLE MNEMONIC-BASED CHARACTERS AND ITEMS. AIM 2: INCORPORATE AI-POWERED MEDIMASTERS INTO THE PROTOTYPE PLATFORM. THIS AIM SEEKS TO INCORPORATE FINE- TUNED GENERATIVE AI TECHNOLOGIES TO PERSONALIZE THE LEARNING EXPERIENCE, CREATING NPC MEDIMASTERS THAT ADAPT CONTENT AND CHALLENGES BASED ON THE INDIVIDUAL LEARNER. AIM 3: ASSESS PLATFORM PROTOTYPE FEASIBILITY TO ENHANCE STUDENT ENGAGEMENT AND SATISFACTION WITH HEALTH EDUCATION MATERIAL. THE FINAL AIM WILL ASSESS THE PLATFORM'S EFFECTIVENESS IN IMPROVING STUDENT ENGAGEMENT AND SATISFACTION WITH HEALTH EDUCATION MATERIAL THROUGH PRE/POSTTEST, SURVEYS, FOCUS GROUP ANALYSIS, AND ANALYSIS OF STUDENT-NPC INTERACTIONS. FURTHER IMPACTING THESE DISPARITIES AND THEIR CORRESPONDING HEALTH RISKS, ADHERENCE TO PHYSICAL ACTIVITY GUIDELINES IS SUBOPTIMAL, ESPECIALLY AMONG AA FAMILIES WITH ADOLESCENTS3,4. DESPITE THE PREVALENCE OF OBESITY AMONG AA ADOLESCENTS, ACCESS TO INTENSIVE BEHAVIORAL INTERVENTIONS IS A CHALLENGE, ESPECIALLY FOR THOSE LIVING IN RURAL AREAS WHO FACE LIMITED HEALTHCARE OPTIONS32 SPECIALIZING IN THESE INTERVENTIONS 33. IN ADDITION, TRANSPORTATION MAY NOT BE FEASIBLE DEPENDING ON TRAVEL DISTANCE, 33,34 AND THE TYPES OF INTERVENTIONS DO NOT ADDRESS THE MOTIVATIONAL FACTORS FOR AAS THAT INTERFERE WITH ADHERENCE TO EVIDENCE-BASED RECOMMENDATIONS. FIT FAMILIES, THE INTERVENTION ON WHICH OUR PROPOSAL IS BASED, ADDRESSES ACCESS ISSUES THROUGH AN AT-HOME EVIDENCE-BASED INTERVENTION. THIS PROPOSAL AIMS TO EXPAND THE FIT FAMILIES INTERVENTION THROUGH OUR INNOVATION THE DIGITAL WELLNESS NURSE (DWN)-FIT THAT UTILIZES MOBILE TECHNOLOGY AND AUTOMATION TO INCREASE ACCESS, ADHERENCE AND ENGAGEMENT IN HEALTHY BEHAVIORS USING HUMAN-IN-THE-LOOP AUTOMATION, WEB, AND MOBILE TECHNOLOGY TO DELIVER EVIDENCE-BASED INTERVENTIONS. SPECIFICALLY, THIS PROPOSAL AIMS TO DEVELOP AND VALIDATE A PROTOTYPE OF THE DWN-FIT PLATFORM; EVALUATE THE DWN-FIT INTERVENTION (DELIVERED BY 4 COMMUNITY HEALTHCARE WORKERS) WITH 16 OBESE AA ADOLESCENTS AND THEIR FAMILIES; DETERMINE THE DEGREE TO WHICH SOCIAL SUPPORT, AS AN ADDITIONAL ELEMENT OF DWN-FIT, ENHANCES ENGAGEMENT AND RETENTION OF FAMILIES IN THE TREATMENT; CONDUCTS USER CENTERED EVALUATIONS THAT DETERMINE IF DWN-FIT MET THE NEEDS OF THE FAMILIES AND COMMUNITY HEALTHCARE WORKERS DURING THE TREATMENT IN PHASE 2 WE WILL EXPAND OUR RANDOMIZED TRIAL BY INCREASING THE NUMBER OF PARTICIPANTS AND LENGTH OF THE INTERVENTION, AND EVALUATE THE DIFFERENCES BETWEEN THE DWN-FIT AND A STANDARD TREATMENT BASED ON ACCESS, ADHERENCE, WEIGHT-LOSS OUTCOMES, AND COST BENEFITS. THE PRIMARY GOALS ARE: 1) INCREASE THE PREVENTION INFRASTRUCTURE IN WINDHAM COUNTY THROUGH CAPACITY BUILDING TO BETTER SERVE RURAL AND LOW SES COMMUNITIES; 2) REDUCE WC YOUTH & YOUNG ADULT CANNABIS USE BY IMPLEMENTING EVIDENCE-BASED STRATEGIES WITHIN THE COMMUNITY AND SCHOOL DISTRICTS THAT ADDRESS BEHAVIORS THAT MAY LEAD TO THE INITIATION AND CONTINUATION OF USE; 3) DECREASE WC YOUTH & YOUNG ADULT PRESCRIPTION MISUSE BY IMPLEMENTING EVIDENCE-BASED STRATEGIES WITHIN THE COMMUNITY AND SCHOOL DISTRICTS THAT ADDRESS BEHAVIORS THAT MAY LEAD TO THE INITIATION AND CONTINUATION OF USE; AND 4) INCREASE SUPPORT TO REDUCE MENTAL HEALTH INDICATORS AND SUBSTANCE USE AMONG LGB YOUTH. AS DEFINED BY THE 2021 CENSUS, WINDHAM COUNTY (WC) IS 70.94% RURAL. LOCATED IN SOUTH-EASTERN VERMONT, THE POPULATION OF WC IS 45,966. DUE TO ITS RURAL NATURE, WC IS DEFINED AS AN UNDERSERVED COMMUNITY. DATA PROVIDED DETAILS HIGHER PREVALENCE OF BOTH CANNABIS AND PRESCRIPTION DRUG USE AMONG RURAL WC YOUTH AND YOUNG ADULTS, LOW SES AND THE LGB POPULATION. WCPP WILL UTILIZE A COMPREHENSIVE APPROACH INCLUDING INFORMATION DISSEMINATION, EDUCATION, ALTERNATIVES, POLICY CHANGE, ID AND REFERRAL AND COMMUNITY-BASED PROCESSES. THIS PROJECT WILL ENGAGE THE STRATEGIC PREVENTION FRAMEWORK TO ADDRESS OUR COMMUNITY PREVENTION/MENTAL HEALTH PROMOTION PRIORITIES THROUGH ASSESSMENT, CAPACITY BUILDING, COMMUNITY PLANNING, DELIVERING SERVICES, AND EVALUATING OUTCOMES. ADDRESSING YOUTH ALCOHOL USE IN VERMONT'S WEST RIVER VALLEY - PROJECT NAME: ADDRESSING YOUTH ALCOHOL USE IN VERMONT'S WEST RIVER VALLEY THE WEST RIVER VALLEY THRIVES COALITION (THRIVES), SEEKS TO BUILD COMMUNITY CAPACITY TO PREVENT AND REDUCE YOUTH ALCOHOL USE IN THE RURAL COMMUNITIES OF VERMONT'S WEST RIVER VALLEY. LOCAL COMMUNITY DATA REFLECT HIGH RATES OF UNDERAGE DRINKING, LOW PERCEPTIONS OF HARM, ENVIRONMENTAL FACTORS AND COMMUNITY NORMS THAT CONTRIBUTE TO YOUTH ALCOHOL USE. THIS PROJECT SEEKS TO STRENGTHEN COMMUNITY CAPACITY TO ADDRESS THESE FACTORS THROUGH SHARED RESPONSIBILITY. SPECIFICALLY, THIS PROJECT SEEKS TO: 1.INCREASE COALITION AND COMMUNITY CAPACITY TO ADDRESS UNDERAGED DRINKING, AND 2. DECREASE UNDERAGED ALCOHOL USE THROUGH A MULTI-STRATEGY APPROACH BASED ON THE STRATEGIC PREVENTION FRAMEWORK. THE POPULATION OF FOCUS INCLUDES YOUTH 12-20 LIVING IN THE 7 TOWNS THAT MAKE UP THE WEST RIVER VALLEY, AS WELL AS ADULTS, PRIMARILY PARENTS AND CAREGIVERS, IN THESE TOWNS WITH WHOM THE COMMUNITY NORMS AROUND DRINKING AND YOUTH ACCESS TO ALCOHOL CAN BE ADDRESSED AND CHANGED. THE PROJECTED NUMBER OF PERSONS SERVED ANNUALLY INCLUDES: 500 PARENTS, 300 YOUTH 12 - 20 AND 2,000 COMMUNITY MEMBERS. WE ESTIMATE TOTALS REACHED DURING THE LIFE OF THE PROJECT TO BE: 2,000 PARENTS, 1,200 YOUTH AND 8,000 COMMUNITY MEMBERS. OBJECTIVES INCLUDE: INCREASE PARENT AND CARING ADULT INVOLVEMENT IN THE COALITION AND CAPACITY TO REDUCE UNDERAGE DRINKING; INCREASE PERCEPTION OF HARM AND LOWER EASE OF ACCESS TO DECREASE PAST 30-DAY USE; INCREASE ADULT AWARENESS OF RISKS AND CONSEQUENCES ASSOCIATED WITH, AND BEST PRACTICES TO PREVENT, YOUTH ACCESS TO ALCOHOL; ADDRESS ENVIRONMENTAL FACTORS AND COMMUNITY NORMS THAT CONTRIBUTE TO PERCEIVED ACCEPTANCE OF UNDERAGE DRINKING. TO ACHIEVE THESE GOALS AND OBJECTIVES, THRIVES WILL DEVELOP A 12-MONTH ACTION PLAN FOR EACH YEAR OF FUNDING BASED ON CURRENT CONDITION AND DATA. STRATEGIES WILL INCLUDE (NOT LIMITED TO): CAPACITY-BUILDING EDUCATION INCLUDING YOUTH AND ADULT TRAINING IN THE SPF TO FACILITATE ENGAGEMENT IN COALITION WORK; SOCIAL NORMS AND MARKETING CAMPAIGNS; DIRECT PREVENTION SERVICES TO YOUTH AND FAMILIES; CAPACITY-BUILDING STRATEGIES TO INCREASE COLLABORATION ACROSS GOVERNMENTAL, YOUTH PROVIDER AND EDUCATIONAL SYSTEMS; COLLABORATION WITH LAW ENFORCEMENT TO INCREASE ALCOHOL OUTLET COMPLIANCE CHECKS AND OTHER STRATEGIC INTERVENTIONS; A TARGETED AND INTENTIONAL EFFORT TO ENGAGE AND EDUCATE MORE PARENT AND CARING ADULTS, AND ADVOCACY FOR LOCAL POLITIES THAT RESTRICT ALCOHOL ACCESS IN PUBLIC SPACES, PARTICULARLY DURING FAMILY-FOCUSED EVENTS.
