WE HAVE INITIATED A PIPELINE FOR DEVELOPING PRODUCTS FOR HLB TREATMENT, HLB PREVENTION, AND ACP CONTROL AND PROPOSE TO EXPAND IT UNDER THIS CAP. TOWARD THIS GOAL, WE HAVE ASSEMBLED AN ENTREPRENEURIAL TEAM OF SCIENTISTS WITH EXPERTISE IN DISEASE BIOLOGY AND THERAPY, REGULATORY CONSULTANTS WITH EXPERIENCE IN OBTAINING PRODUCT APPROVAL FROM FEDERAL AGENCIES, AND BUSINESS EXPERTS WITH A TRACK RECORD IN COMMERCIALIZATION OF AGRICULTURAL PRODUCTS. OUR TEAM WILL REGISTER A PRODUCT FOR HLB TREATMENT IN 3 YEARS. WE WILL BE ABLE TO REGISTER A TRANSGENIC PRODUCT FOR HLB PREVENTION IN 7 YEARS FIRST BY COLLECTING EFFICACY, TOXICITY, AND SAFETY DATA IN 5 YEARS (2 YEARS AFTER THIS PILOT CAP) AND THEN BY COLLECTING CITRUS HEALTH AND FRUIT QUALITY DATA IN NEXT 2 YEARS. IN ADDITION, WE WILL CREATE A KNOWLEDGE BASE FOR RESEARCHERS, WHICH WILL DESCRIBE OUR CONCEPT AND ITS APPLICATION IN THERAPY OF HLB AND OTHER PLANT DISEASES. FINALLY, OUR CAP WILL INCLUDE A COMPONENT ON EDUCATION AND OUTREACH THAT WILL INVOLVE TWO IMPORTANT FUNCTIONS: TRAINING OF STUDENTS, TECHNICIANS, AND EXTENSION PROFESSIONALS ON THE KNOWLEDGE BASE AND HOLDING ONSITE AND ONLINE MEETINGS/WORKSHOPS TO COMMUNICATE TO THE GROWERS AND STAKEHOLDERS THE SCOPE, DELIVERABLES, AND IMPACT OF OUR CAP.FOLLOWING THE RECOMMENDATION OF THE REVIEWERS, THE OBJECTIVES HAVE BEEN REVISED AS CONSTRAINED BY THE REDUCED TOTAL BUDGET (FROM 11.8M TO 4.75M) AND PROJECT DURATION (3 YEARS INSTEAD OF 5). HOWEVER, THE PROJECT THEME REMAINS THE SAME, I.E., TO DEMONSTRATE THAT ENHANCEMENT OF CITRUS INNATE IMMUNITY VIA DELIVERY OF THE CITRUS DERIVED HELIX-TURN-HELIX (HTH) PEPTIDES AND PROTEIN CHIMERAS LEADS TO EFFICACIOUS HLB THERAPY. LIGHT DETECTION AND RANGING (LIDAR) IS USEFUL FOR DEVELOPING HIGH RESOLUTION INPUTS TO COUPLED FIRE ATMOSPHERIC MODELING TO PLAN PRESCRIBED FIRES AND EVALUATE WILDFIRE RISK. THIS MODIFICATION WILL APPLY VEGETATION MEASURED THROUGH LIDAR AS INPUTS TO A NEW FIRE BEHAVIOR MODEL CALLED QUICFIRE ON USFWS PRIORITY LANDSCAPES ACROSS THE US INCLUDING ALASKA. THE OBJECTIVE OF THIS PROJECT WILL BE TO EVALUATE PRESCRIPTIONS USED TO TREAT FUELS FOR DESIRED OUTCOMES. THE USE OF LIDAR BEFORE AND AFTER FIRE WILL PROVIDE AN IDEAL MONITORING OF MODEL PERFORMANCE AT MANAGEMENT SCALES. SECONDLY THIS MODIFICATION WITH USFWS WILL DEVELOP A LIDAR FEATURE DETECTION WORKFLOW FOR ARCHAEOLOGICAL SITES ON THE SAME PRIORITY LANDSCAPES. USING EXISTING CULTURAL RESOURCE DATABASES AND DATASETS AND NEW TARGETED ARCHEOLOGICAL SURFACE SURVEYS WE WILL VALIDATE DETECTION RESULTS AND PROVIDE ADDITIONAL ASSISTANCE IN INTEGRATING THIS WORK INTO EXISTING CULTURAL RESOURCE MANAGEMENT PLANNING PROCEDURES FOR THE USFWS. FOLLOWING A SUCCESSFUL PILOT PROJECT LIDAR FEATURE DETECTION WORKFLOWS WILL BE AUGMENTED AND TESTED NATIONALLY (WITH OTHER FEDERAL PARTNERS) IN LOCALES WITH VARYING TOPOGRAPHIC CONDITIONS AND MULTIPLE CULTURAL RESOURCE SITE TYPES. ALTHOUGH IT IS GENERALLY ACCEPTED THAT CHORUS IS GENERATED BY THE UNSTABLE DISTRIBUTION OF ENERGETIC ELECTRONS FORMED DURING GEOMAGNETICALLY ACTIVE TIMES, KEY OBSERVATIONAL FEATURES OF THE CHORUS ARE NOT FULLY UNDERSTOOD. HERE THE TEAM WILL USE AN OPEN-SOURCE PARTICLE-IN-CELL (PIC) CODE TO CARRY OUT LARGE-SCALE FIRST-PRINCIPAL SIMULATIONS TO EXPLORE THE GENERATION MECHANISMS OF CHORUS AND PRODUCE FEATURES THAT CAN BE DIRECTLY COMPARED TO OBSERVATIONS. REALISTIC MAGNETIC FIELD TOPOLOGY AND PLASMA PARAMETERS DERIVED FROM OBSERVATIONS WILL BE USED IN OUR SIMULATIONS, AND CHORUS WITH RISING TONE, BAND GAPS AND REPETITIVE ELEMENTS CONSISTENT WITH OBSERVATIONS IS EXPECTED TO BE PRODUCED. WITH 2D AND 3D PIC SIMULATIONS, EXISTING THEORIES OF CHORUS GENERATION WILL BE TESTED, AND THE UNDERLYING NONLINEAR PHYSICS WILL BE REVEALED. THE STUDY WILL GREATLY ADVANCE OUR UNDERSTANDING OF WAVE-PARTICLE INTERACTION IN AN INHOMOGENEOUS MAGNETIC FIELD LIKE EARTH'S DIPOLE FIELD. THIS EFFORT WILL BE AN IMPORTANT STEP TOWARD BUILDING A PHYSICS-BASED CHORUS WAVE MODEL, WHICH IS CRITICAL FOR RELIABLE PREDICTION OF RADIATION BELT DYNAMICS AND THE NEAR-EARTH GEOSPACE ENVIRONMENT. THIS AWARD REFLECTS NSF'S STATUTORY MISSION AND HAS BEEN DEEMED WORTHY OF SUPPORT THROUGH EVALUATION USING THE FOUNDATION'S INTELLECTUAL MERIT AND BROADER IMPACTS REVIEW CRITERIA.

THESE RESULTS CONTRIBUTE DIRECTLY TO THE DECADAL SURVEY OBJECTIVE DISCOVER AND CHARACTERIZE FUNDAMENTAL PROCESSES THAT OCCUR BOTH WITHIN THE HELIOSPHERE AND THROUGHOUT THE UNIVERSE. FURTHERMORE BECAUSE PDI MAY PLAY A KEY ROLE IN HOW ALFVEN WAVES FROM THE SUN ARE EVOLVE AND POWER THE SOLAR WIND THE INSIGHTS GAINED FROM THIS PROPOSAL WILL ALSO HELP ADDRESS THE DECAL SURVEY OBJECTIVE DETERMINE THE INTERACTION OF THE SUN WITH THE SOLAR SYSTEM AND THE INTERSTELLAR MEDIUM. FROM CHANGES IN COMPOSITION ENERGY PITCH ANGLE ETC.). 2. QUANTIFY HOW THE IONS AFFECT THE RATE OF RECONNECTION AT THE DAYSIDE (E.G. BY CALCULATING THE PERCENTAGE REDUCTION IN THE RECONNECTION RATE AS A FUNCTION OF ACTIVITY AND SOLAR WIND CONDITIONS). 3. REVEAL THE RELATIVE EFFECTS OF THE ION PLASMA CLOAK THE PLASMASPHERIC PLUME AND THE LLBL PLASMA ON THE DAYSIDE RECONNECTION RATE (E.G. BY COMPARING THE RATE FOR PLUME PLASMA WITH THAT FOR CLOAK PLASMA AND LLBL PLASMA). THE PROPOSAL IS RELEVANT TO THREE DECADAL SURVEY GOALS: DETERMINE THE DYNAMICS AND COUPLING OF EARTH S MAGNETOSPHERE IONOSPHERE AND ATMOSPHERE AND THEIR RESPONSE TO SOLAR AND TERRESTRIAL INPUTS; DETERMINE THE INTERACTION OF THE SUN WITH THE SOLAR SYSTEM AND THE INTERSTELLAR MEDIUM; DISCOVER AND CHARACTERIZE FUNDAMENTAL PROCESSES THAT OCCUR BOTH WITHIN THE HELIOSPHERE AND THROUGHOUT THE UNIVERSE. .THE METHODOLOGY CONSISTS OF: (I) STATISTICAL STUDIES OF IONS IN THE INNER MAGNETOSPHERE AND AT THE RECONNECTION SITE (E.G. FOR THE CONJUNCTIONS WE DETERMINE AND QUANTIFY IF AND HOW THE ION POPULATION EVOLVES DURING ITS DRIFT. PURPOSE: THE VOLUNTEER INCOME TAX ASSISTANCE (VITA) GRANT WAS ESTABLISHED AS A MATCHING GRANT PROGRAM TO PROVIDE FUNDING FOR ORGANIZATIONS WHO SUPPORT COMMUNITY VOLUNTEER INCOME TAX ASSISTANCE. ACTIVITIES TO BE PERFORMED: THE VITA GRANT PROGRAM PROVIDES FINANCIAL SUPPORT TO ORGANIZATIONS WHO 1) EXTEND SERVICES TO UNDERSERVED POPULATIONS IN HARDEST TO REACH AREAS BOTH URBAN AND NON-URBAN; 2) INCREASE THE CAPACITY TO FILE RETURNS ELECTRONICALLY; 3) HEIGHTEN QUALITY CONTROL; 4) ENHANCE TRAINING OF VOLUNTEERS; AND 5) SIGNIFICANTLY IMPROVE THE ACCURACY RATE OF RETURNS PREPARED AT VITA SITES. END GOAL/EXPECTED OUTCOMES: VITA GRANT RECIPIENTS ARE EXPECTED TO 1) FOLLOW EXISTING GUIDANCE GOVERNING VITA SITE OPERATIONS; 2) ENSURE AT LEAST 90% OF RETURNS PREPARED ARE FOR INDIVIDUALS WHOSE INCOME IS EQUAL TO OR LESS THAN THE MAXIMUM EARNED INCOME TAX CREDIT (EITC) THRESHOLDS; 2) FILE ALL ELIGIBLE RETURNS ELECTRONICALLY; 3) ACHIEVE 100% OF THEIR RETURN PRODUCTION GOALS; 4) BECOME MORE EFFICIENT WITH GRANT FUNDS; AND 5) SHOW INCREMENTAL INCREASES IN RETURN PREPARATION EACH YEAR. INTENDED BENEFICIARIES: TAXPAYERS WHO ARE LOW TO MODERATE INCOME INDIVIDUALS, PERSONS WITH DISABILITIES, THOSE FOR WHOM ENGLISH IS A SECOND LANGUAGE, NATIVE AMERICANS, INDIVIDUALS LIVING IN RURAL AREAS, MEMBERS OF THE ARMED FORCES AND THEIR FAMILIES, AND THE ELDERLY. SUBRECIPIENT ACTIVITIES: SUBRECIPIENTS MAY BE UTILIZED BY GRANT RECIPIENTS TO HELP DELIVER KEY ELEMENTS OF THE PROGRAM AND MUST ADHERE TO GRANT PROGRAM GUIDELINES. COLLABORATIVE RESEARCH: SHINE: OBSERVATIONAL AND THEORETICAL STUDIES OF THE PARAMETRIC DECAY INSTABILITY IN THE LOWER SOLAR ATMOSPHERE -IT IS UNCLEAR HOW THE OUTER LAYER OF THE SUN, THE CORONA, IS HEATED TO MILLIONS OF DEGREES. ONE POTENTIAL MECHANISM IS THROUGH THE PARAMETRIC DECAY INSTABILITY (PDI). THIS PROJECT ADDRESSES THE SOLAR, HELIOSPHERIC, AND INTERPLANETARY ENVIRONMENT (SHINE) GOAL OF UNDERSTANDING THE SOLAR CORONA THROUGH NUMERICAL SIMULATIONS AND ANALYSIS OF SPACE-BASED AND NSF-FUNDED GROUND BASED SOLAR OBSERVATIONS. THE PROJECT ALSO SUPPORTS OUTREACH ACTIVITIES IN NEW YORK CITY AND NEW MEXICO. TWO POST-DOCTORAL RESEARCHERS WILL BE SUPPORTED, ALONG WITH UNDERGRADUATE STUDENTS AT COLUMBIA UNIVERSITY. THE PROJECT WILL DETERMINE WHETHER PDI IS AN IMPORTANT PROCESS IN THE SOLAR ATMOSPHERE. THE TEAM WILL ANALYZE OBSERVATIONS FROM THE CORONAL MULTICHANNEL POLARIMETER, THE EXTREME ULTRAVIOLET IMAGING SPECTROMETER, THE INTERFACE REGION IMAGING SPECTROMETER, AND THE NSF-FUNDED DANIEL K. INOUYE SOLAR TELESCOPE. NUMERICAL MODELING WILL BE USED TO BETTER UNDERSTAND THE BEHAVIOR OF PDI IN THE PRESENCE OF GRADIENTS IN THE PLASMA PROPERTIES OF THE TRANSITION REGION AND THE CORONA. MAGNETOHYDRODYNAMIC SIMULATIONS WILL BE CARRIED OUT TO STUDY ALFVEN WAVES AND PDI GROWTH IN THE LOWER SOLAR ATMOSPHERE, WHERE THE SCALE LENGTH OF GRADIENTS IS SMALLER THAN THE WAVELENGTH. THIS IS CRITICAL TO UNDERSTAND HOW DENSITY FLUCTUATIONS ARE GENERATED VERY CLOSE TO THE SUN. MOREOVER, ION HEATING ASSOCIATED WITH ACOUSTIC WAVES PRODUCED BY PDI WILL BE INVESTIGATED USING HYBRID SIMULATIONS, TO ESTIMATE WHAT FRACTION OF THE ALFVEN WAVE POWER CAN BE DISSIPATED IN THE LOWER ATMOSPHERE. BECAUSE CHROMOSOME FOLDING OCCURS OVER A WIDE RANGE OF LENGTH AND TIMES SCALES, A SOPHISTICATED COMBINATION OF EXPERIMENTAL AND COMPUTATIONAL APPROACHES IS REQUIRED TO UNRAVEL GENOME ORGANIZATION. IN THIS COLLABORATIVE PROJECT, A TEAM FROM NYU (TAMAR SCHLICK), HARVARD (ANNA LAPPALA AND JEANNIE LEE), AND NEW MEXICO CONSORTIUM (KARISSA SANBONMATSU) WILL COMBINE EXPERTISE IN MOLECULAR BIOLOGY, GENOMICS, COMPUTATIONAL BIOLOGY, BIOPHYSICS AND BIOMOLECULAR MODELING IN AN INTEGRATED EXPERIMENTAL AND COMPUTATIONAL EFFORT TO ADDRESS KEY MECHANISTIC QUESTIONS ABOUT X-CHROMOSOME STRUCTURE AND FUNCTION. INACTIVATION INFLUENCES X CHROMOSOME EXPRESSION IN FEMALES AND HAS PROFOUND HEALTH IMPLICATIONS. A BETTER UNDERSTANDING OF X-CHROMOSOME FOLDING AND GENE EXPRESSION WILL LEAD TO NEW AVENUES FOR TREATING OTHER DISEASES RELATED TO X-CHROMOSOME STATUS. THE RESEARCH PROGRAM WILL OFFER INTERDISCIPLINARY AND MULTIDISCIPLINARY TRAINING TO YOUNG SCIENTISTS IN STEM FIELDS, THROUGH HANDS ON MENTORING AND PARTICIPATION IN SUMMER CAMPS, PROMOTING NATIONAL NEEDS IN EDUCATION AND INNOVATION IN SCIENCE AND ENGINEERING. THE INVESTIGATORS WILL USE ADVANCED COMPUTER SIMULATIONS TO STUDY BLAZARS IN DETAIL TO UNLOCK THE SECRETS OF THESE SOURCES AND MAKE PREDICTIONS FOR FUTURE DISCOVERIES. UNDERGRADUATE STUDENTS AT PURDUE UNIVERSITY WILL HAVE THE CHANCE TO ACTIVELY TAKE PART IN THIS EXCITING RESEARCH. THE INVESTIGATORS WILL ALSO ENGAGE WITH HIGH SCHOOL STUDENTS THROUGH THE SATURDAY MORNING ASTROPHYSICS PROGRAM AT PURDUE UNIVERSITY. BLAZARS, RELATIVISTIC JETS FROM ACTIVE GALACTIC NUCLEI POINTING VERY CLOSE TO OUR LINE OF SIGHT, ARE AMONG THE MOST POWERFUL COSMIC PARTICLE ACCELERATORS IN THE UNIVERSE. THEY EXHIBIT VERY BRIGHT, HIGHLY VARIABLE, NONTHERMAL-DOMINATED EMISSION ACROSS THE ENTIRE ELECTROMAGNETIC SPECTRUM, INDICATING EXTREME PARTICLE ACCELERATION IN COMPACT REGIONS, OFTEN REFERRED TO AS THE BLAZAR ZONE. THIS PROPOSED PROGRAM WILL STUDY NEUTRINO BLAZARS COMBINING MHD, PIC, PARTICLE TRANSPORT, AND POLARIZED RADIATION TRANSFER SIMULATIONS. THESE SIMULATIONS ARE DESIGNED TO TRACK THE COSMIC RAY ACCELERATION AND TRANSPORT, THUS IDENTIFY TEMPORAL CORRELATION BETWEEN NEUTRINOS AND MULTI-WAVELENGTH SIGNATURES AS WELL AS CHARACTERISTIC RADIATION PATTERNS FOR NEUTRINO BLAZARS. THIS APPROACH THEREFORE LEVERAGES CURRENT OBSERVATIONAL EFFORTS TO REVEAL UNDERLYING BLAZAR ZONE PHYSICS, AND PROVIDES SOLID PREDICTIONS FOR FUTURE MULTI-MESSENGER DISCOVERIES. THE PROJECT WILL OFFER RESEARCH OPPORTUNITIES FOR UNDERGRADUATE STUDENTS AT PURDUE UNIVERSITY. THE WIDE CONTEXT OF THE PROPOSAL EQUALLY ALLOWS FOR ANALYTICAL AND NUMERICAL PROJECTS AS WELL AS COMPARISON WITH OBSERVATIONS, SO THAT UNDERGRADUATES CAN PICK THE OPPORTUNITY THAT FITS THEIR MAIN INTEREST. COLLABORATIVE RESEARCH: TESTING CONTROLS ON SOURCE, SINK, AND LIFETIME OF ATMOSPHERIC WATER WITH NUMERICAL TAGS AND STABLE ISOTOPE RATIOS -H2O IS INSTANTLY RECOGNIZABLE AS THE CHEMICAL FORMULA FOR WATER, BUT THIS SIMPLE FORMULA MASKS THE FACT THAT NOT ALL HYDROGEN ATOMS ARE ALIKE. A SMALL FRACTION OF THE WATER ON EARTH COULD BE MORE ACCURATELY CALLED HDO, WHERE D STANDS FOR DEUTERIUM, THE HEAVIER HYDROGEN ISOTOPE IN WHICH THE NUCLEUS CONTAINS A NEUTRON AS WELL AS A PROTON. LIKEWISE, SOME WATER MOLECULES CONTAIN 18O, AN ISOTOPE OF OXYGEN WHICH IS HEAVIER THAN ORDINARY OXYGEN BY THE ADDITION OF TWO NEUTRONS. THESE HEAVIER FORMS OF WATER SHARE A SUBTLE BUT IMPORTANT DIFFERENCE FROM ORDINARY H2O: THEY EVAPORATE MORE SLUGGISHLY AND CONDENSE MORE READILY, PREFERRING THE SOLID AND LIQUID PHASES TO THE VAPOR PHASE. WHEN WATER EVAPORATES FROM THE LAND OR OCEAN SURFACE THE VAPOR IS SLIGHTLY DEPLETED OF THE HEAVIER ISOTOPES, AND EACH TIME RAIN FALLS FROM A CLOUD THE WATER VAPOR REMAINING IN THE ATMOSPHERE IS SLIGHTLY LIGHTER THAN THE WATER THAT RAINED OUT. THE REMOVAL OF HEAVIER ISOTOPES BY PRECIPITATION SUGGESTS THAT THE ABUNDANCES OF HEAVIER ISOTOPES IN WATER VAPOR AND PRECIPITATED WATER CONTAIN IMPORTANT CLUES FOR UNDERSTANDING THE HYDROLOGICAL CYCLE AND ITS CONNECTIONS TO ENVIRONMENTAL CONDITIONS. RESULTS OF RESEARCH IN THIS AREA ARE PROMISING BUT THEY ARE COMPLICATED BY THE NUMEROUS FACTORS THAT CAN INFLUENCE ISOTOPIC COMPOSITION. AMONG THESE ARE THE TEMPERATURE AT THE LOCATION WHERE THE PRECIPITATION OCCURRED, THE HEIGHT OF THE CLOUD THAT PRODUCED THE PRECIPITATION, THE RELATIVE HUMIDITY OVER THE OCEAN WHERE THE WATER VAPOR EVAPORATED, AND THE EVAPORATION OF LIGHTER ISOTOPES FROM RAINDROPS AS THEY FALL THROUGH DRY AIR. MOREOVER, MUCH OF THE RESEARCH ON WATER ISOTOPES IS BASED ON CORRELATIONS BETWEEN ISOTOPIC COMPOSITION AND VARIABLES RELEVANT TO PHYSICAL MECHANISMS (TEMPERATURE IN THE REGION OF RAINOUT, FOR INSTANCE). THESE CORRELATIONS CAN BE QUITE INFORMATIVE BUT CORRELATIONS BY THEMSELVES CANNOT DEFINITIVELY ESTABLISH CAUSALITY. WORK UNDER THIS AWARD SEEKS TO ESTABLISH DIRECT, CAUSAL RELATIONSHIPS BETWEEN THE PHYSICAL MECHANISMS RELEVANT TO ISOTOPIC COMPOSITION AND THE ACTUAL COMPOSITION OF WATER VAPOR AND PRECIPITATED WATER. THE RESEARCH IS CONDUCTED USING A MODEL, THE COMMUNITY EARTH SYSTEM MODEL (CESM), IN WHICH THE MECHANISMS AND THEIR INFLUENCE ON ISOTOPIC COMPOSITION ARE KNOWN EXACTLY. CESM IS ALREADY EQUIPPED TO SIMULATE THE PHYSICAL PROCESSES THAT DETERMINE THE RATIO OF HEAVIER TO LIGHTER ISOTOPES, AND IT ALSO TRACKS THE ISOTOPIC COMPOSITION OF WATER VAPOR AS IT CIRCULATES IN THE SIMULATED ATMOSPHERE. THIS PROJECT DEVELOPS A NEW WATER TAGGING CAPABILITY WHICH ALLOWS SIMULATED WATER VAPOR TO RECORD INFORMATION ABOUT CONDITIONS UNDER WHICH IT EVAPORATED, FOR INSTANCE WHERE AND WHEN IT EVAPORATED AND WHETHER IT WAS PROCESSED THROUGH CLOUDS. THIS INFORMATION IS ALSO TRANSFERRED TO THE PRECIPITATION PRODUCED WHEN WATER VAPOR CONDENSES. THE TAG INFORMATION CAN BE DIRECTLY COMPARED TO ISOTOPIC COMPOSITION, ALLOWING THE INFLUENCE OF THE PHYSICAL CONDITIONS AND PROCESSES REPRESENTED BY THE TAGS TO BE DIRECTLY ASSESSED. SOME SIMULATIONS USE A DATA ASSIMILATION SYSTEM (KNOWN AS DART) TO ENSURE THAT THE SIMULATED CLIMATE AND CIRCULATION ACCURATELY REPRESENT THEIR REAL-WORLD COUNTERPARTS. ADDITIONAL WORK USES A ONE-DIMENSIONAL ENERGY BALANCE MODEL, WHICH PROVIDES A REDUCED-COMPLEXITY SETTING TO DEVELOP NEW IDEAS AND TEST HYPOTHESES. THE WORK HAS SCIENTIFIC BROADER IMPACTS DUE TO THE WIDESPREAD USE OF WATER ISOTOPES AS A WINDOW ON THE FUNCTIONING OF CLIMATE AND THE HYDROLOGICAL CYCLE. FOR INSTANCE ISOTOPE RATIOS IN GLACIAL ICE CORES HAVE BEEN USED TO ESTIMATE TEMPERATURE DIFFERENCES BETWEEN ICE AGES AND INTERGLACIAL WARM PERIODS, THUS PROVIDING A VALUABLE POINT OF REFERENCE FOR UNDERSTANDING CURRENT CLIMATE CHANGE. THE WATER TAGGING ALGORITHMS DEVELOPED HERE WILL BE MADE AVAILABLE TO THE WORLDWIDE USER COMMUNITY OF CESM, PROVIDING USERS WITH A POWERFUL TOOL FOR THE INTERPRETATION OF ISOTOPE DATA AND ITS APPLICATION TO A VARIETY OF RESEARCH QUESTIONS. IN ADDITION, RESEARCHERS SUPPORTED UNDER THIS AWARD CONDUCT EDUCATIONAL OUTREACH THROUGH A PROGRAM FOCUSED ON THE DECKER LAKE YOUTH CENTER, A FACILITY HOUSING POST-CONVICTION YOUTH IN THE SALT LAKE VALLEY. THE OUTREACH INVOLVES OZONE BIOINDICATOR GARDENS WHICH DEMONSTRATE THE HAZARDS OF OZONE POLLUTION, A COMMON PROBLEM IN THE SALT LAKE VALLEY. EDUCATIONAL OPPORTUNITIES IN CORRECTIONAL FACILITIES HAVE BEEN SHOWN TO REDUCE RECIDIVISM. MULTIPOINT MEASUREMENTS IN THE INNER MAGNETOSPHERE CAN ALLOW THE SPATIAL AND TEMPORAL EVOLUTION OF VARIOUS PARTICLE POPULATIONS AND WAVE MODES TO BE DISENTANGLED. IN PARTICULAR SPATIAL SCALES OF WAVE ACTIVE REGIONS HAVE RECENTLY BEEN ESTIMATED FOR A RANGE OF WAVE MODES INCLUDING WHISTLER MODE CHORUS ELECTROMAGNETIC ION CYCLOTRON (EMIC) AND QUASI-PERIODIC WAVES. UNDERSTANDING THE DRIVERS CONTROLLING THE SPATIAL SCALES OF WAVE ACTIVITY IN THE INNER MAGNETOSPHERE IS CRITICAL FOR UNCOVERING THE UNDERLYING PHYSICS BEHIND THE WAVE GENERATION AS WELL AS FOR BETTER PREDICTION OF WHERE AND WHEN WAVES WILL BE PRESENT. RECENT STUDIES OF EMIC WAVES REVEAL A DEPENDENCE OF THE WAVE SPATIAL EXTENT ON MAGNETIC LOCAL TIME (MLT) WAVE FREQUENCY AND L SHELL. VARIOUS HYPOTHESES HAVE BEEN PROPOSED TO EXPLAIN SOME OF THESE PATTERNS INCLUDING DIFFERENT SOURCES (AND SPATIAL EXTENTS) OF ION ANISOTROPY ON THE DAY VERSUS NIGHT SIDE COMPOSITIONAL VARIATIONS THROUGHOUT THE INNER MAGNETOSPHERE OR COLD PLASMA DENSITY STRUCTURE. STUDIES OF ION DYNAMICS IN THE INNER MAGNETOSPHERE HAVE SHOWN RAPID EVOLUTION IN SPATIAL STRUCTURES AND BOUNDARIES AS WELL AS COMPOSITION BUT THE RELATIONSHIP BETWEEN THESE VARIATIONS AND CHARACTERISTIC EMIC WAVE SCALES HAS YET TO BE EXPLORED. THIS WORK PROPOSES TO USE MULTIPOINT MEASUREMENTS AND ANALYSIS TECHNIQUES TO DETERMINE THE SPATIAL AND TEMPORAL EXTENTS AND EVOLUTION OF PLASMA STRUCTURES IN THE INNER MAGNETOSPHERE TO BETTER UNDERSTAND THE DRIVERS OF MAGNETOSPHERIC WAVE CHARACTERISTIC SCALES. IN PARTICULAR WE FOCUS ON EMIC WAVES AND THE PLASMA PARAMETERS IMPORTANT FOR THEIR GROWTH NAMELY COLD PLASMA DENSITY AND WARM ION DENSITY ANISOTROPY AND COMPOSITION. STRUCTURES AND BOUNDARIES IN THE LOCAL PLASMA POPULATIONS CAN HAVE DIRECT IMPACT ON WHERE AND WHEN WAVE GROWTH CAN OCCUR. THE PROPOSED WORK WILL ADDRESS THE IMPORTANT QUESTION OF WHAT PHYSICAL PARAMETERS CONTROL EMIC WAVE CHARACTERISTICS AND HOW DO THESE VARY THROUGHOUT THE INNER MAGNETOSPHERE. UTILIZING DATA FROM THE VAN ALLEN PROBES AND THEMIS MISSIONS WE PROPOSE TO: 1. INVESTIGATE SPATIAL STRUCTURES OF H+ HE+ AND O+ POPULATIONS IN THE INNER MAGNETOSPHERE. THE MULTIPLE PROBES CAN REVEAL THE TEMPORAL EVOLUTION OF THESE STRUCTURES AND HOW PERSISTENT VARIOUS PLASMA BOUNDARIES AND GRADIENTS ARE. 2. ESTIMATE ION ANISOTROPY AND THE SPATIAL SCALES OF ANISOTROPIC REGIONS. VARIATIONS IN THESE SCALES ACROSS L SHELL AND MLT MAY SUGGEST DIFFERENT PHYSICAL MECHANISMS GENERATING THESE ANISOTROPIES. 3. CHARACTERIZE COLD PLASMA DENSITY STRUCTURE INCLUDING FINE SCALE STRUCTURE AND ITS VARIABILITY ACROSS L SHELL AND MLT. DR. MICK DENTON CO I HAS STUDIED PLASMASPHERIC STRUCTURE AND REFILLING PROCESSES. RECENT WORK INCLUDES INVESTIGATIONS OF STORM-TIME ION DYNAMICS AND COMPOSITIONAL VARIATIONS IN EARTH S MAGNETOSPHERE. WE WILL CHARACTERIZE VEGETATION STRUCTURE AND COMPOSITION, AND SOIL THERMAL AND MOISTURE CONDITIONS AT SURVEYED SITES. FINALLY, WE WILL EVALUATE HOW ORNATE BOX TURTLE ABUNDANCES RELATE TO ENVIRONMENTAL CONDITIONS IN BURNED, TREATED, AND UNBURNED UNTREATED SITES. FROM THE DATA COLLECTED, WE EXPECT TO PUBLISH ONE SCIENTIFIC JOURNAL ARTICLE, GIVE ONE CONFERENCE PRESENTATION, AND, IN COLLABORATION WITH BLM STAFF, DEVELOP A GUIDANCE DOCUMENT THAT DETAILS TREATMENT APPROACHES THAT ARE MOST BENEFICIAL FOR RESTORING ORNATE BOX TURTLE HABITAT. THIS WORKSHOP AIMS TO ADVANCE MICROBIOME SCIENCE THROUGH COORDINATED DATA MANAGEMENT ACROSS RESEARCHERS, FUNDERS, DATA REPOSITORIES, AND PUBLISHERS. THE EVENT WILL ALSO BRING TOGETHER DIVERSE MICROBIOME SCIENCE STAKEHOLDERS THAT REPRESENT THE INTERDISCIPLINARY NATURE OF MICROBIOME RESEARCH, INCLUDING ECOLOGISTS, DATA SCIENTISTS, MODELERS, MICROBIOLOGISTS, AND BIOINFORMATICIANS, WITH THIS GROUP OF INVITEES INCLUDING GROUPS UNDERREPRESENTED IN SCIENCE AND ENGINEERING. THIS WORKSHOP WILL BE THE FIRST OF ITS KIND WITH PARTICIPANTS TASKED WITH IDENTIFYING LOCAL AND NATIONAL PRIORITIES AND FORMING A STRATEGY FOR IMPLEMENTATION OF ENVIRONMENTAL MICROBIOME DATA MANAGEMENT BEST PRACTICES ACROSS THE MICROBIOME RESEARCH ECOSYSTEM. RESULTS OF THIS PROPOSED WORKSHOP WILL FORM THE BASIS OF A FORMALIZED CHECKLIST TO BE IMPLEMENTED ACROSS JOURNALS AND REPOSITORIES FOR MICROBIOME DATA MANAGEMENT AND THESE BEST PRACTICES WILL DRIVE SCIENTIFIC INNOVATION NOW AND IN YEARS TO COME AS THIS DATA CONTINUES TO BE USED IN LARGE-SCALE MODELS AND MACHINE LEARNING EFFORTS. COLLABORATIVE RESEARCH: THE ROLE OF SUPRA-ARCADE DOWNFLOWS ON ENERGY TRANSFER IN SOLAR FLARES -SOLAR FLARES ARE THE MOST ENERGETIC PHENOMENA IN THE SOLAR SYSTEM, DURING WHICH THE FLARE ABOVE-THE-LOOP-TOP (ALT) REGIONS ARE THE KEY TO UNDERSTANDING ENERGY RELEASE IN SOLAR FLARES. THIS PROJECT AIMS TO OBTAIN A MORE COMPREHENSIVE PICTURE OF THE ENERGY RELEASE IN FLARE ALT REGIONS BY SYSTEMATICALLY INVESTIGATING THE ENERGY TRANSFER ASSOCIATED WITH SPATIALLY RESOLVED SUPRA ARCADE DOWNFLOWS (SADS). THE IMPROVED KNOWLEDGE OF THE ENERGY TRANSFER IN SOLAR FLARES IS CRITICAL TO UNDERSTANDING THE BASIC SCIENCE NEEDED TO MEET THE GOALS OF THE NATIONAL SPACE WEATHER STRATEGY AND ACTION PLAN, WHICH AIMS TO DEVELOP TOOLS TO FORECAST SPACE WEATHER AND MITIGATE ITS IMPACTS. THE PROJECT INVOLVES STATE-OF-ART MHD MODELS, INNOVATIVE PARTICLE SIMULATIONS, AND AN IMAGE SYNTHESIS APPROACH, TO COMPARE WITH OBSERVATIONS IN MULTIPLE WAVELENGTHS AND VIEWING PERSPECTIVES (INCLUDING MICROWAVE IMAGING SPECTROSCOPY IN FANS THAT HAVE BEEN RARELY STUDIED IN THE COMMUNITY). THIS PROJECT, LED BY SEVERAL EARLY-CAREER WOMEN PIS, WILL INVOLVE UNDERGRADUATE STUDENTS AND SUPPORT A GRADUATE STUDENT. THERE ARE OUTREACH ACTIVITIES AND DEVELOPING OPEN-SOURCE CODES PLANNED FOR THIS PROJECT. OVER THE DECADES, THE DEVELOPMENTS OF 2D STANDARD SOLAR FLARE MODELS HAVE SIGNIFICANTLY IMPROVED THE COMPREHENSION OF SOLAR ERUPTIONS. HOWEVER, RECENT OBSERVATIONS HAVE INDICATED THE IMPORTANCE OF SADS, WHICH ARE FREQUENTLY OBSERVED FROM A FACE-ON VIEWING PERSPECTIVE, TO ENERGY TRANSFER IN FLARE ALT REGIONS. THE PROJECT WILL THOROUGHLY EXPLORE THE ROLE OF SADS IN ENERGY TRANSFER IN THE ALT REGIONS BY COMBINING OBSERVATIONS AND SIMULATIONS. THE SCIENCE QUESTIONS TO BE ADDRESSED ARE: (1) HOW DO SADS ENGAGE IN ENERGY TRANSFER IN ALT REGIONS? (2) WHAT ARE THE STATISTICAL FEATURES OF PLASMA CONDITIONS AROUND SADS? AND (3) WHAT ARE OBSERVATIONAL SIGNATURES RELATED TO ENERGY TRANSFER PROCESSES OF SADS IN ALT REGIONS? FIRST, THEY WILL PERFORM A STATISTICAL STUDY ON THE THERMAL CHARACTERISTICS AND THE PHYSICAL RELATIONSHIP BETWEEN THESE CHARACTERISTICS AND THE DYNAMICS OF SADS. SECOND, THEY WILL PERFORM 3D MAGNETOHYDRODYNAMICS (MHD) SIMULATIONS TO INVESTIGATE THE ENERGY RELEASE IN FLARES. THEY WILL SYNTHESIZE THE EMISSIONS INCORPORATING DIFFERENT VIEWING PERSPECTIVES AND MULTI-WAVELENGTHS TO COMPARE WITH OBSERVATIONS, INCLUDING RADIO EMISSIONS IN FANS THAT ARE RARELY DISCUSSED IN THE LITERATURE. THIRD, THEY WILL CARRY OUT PARTICLE SIMULATIONS WITH TIME-RESOLVED DYNAMICS PROVIDED BY THE MHD FRAMEWORK TO EXPLORE THE IMPACT OF SADS ON ENERGETIC PARTICLES. HISTONE POST-TRANSLATIONAL MODIFICATIONS (PTMS) REPRESENT SOME OF THE MOST RELEVANT AND WIDELY STUDIED EPIGENOMIC FACTORS IN DISEASE. RECENT DEVELOPMENT OF SCCUT&TAG (SC CLEAVAGE UNDER TARGETS AND TAGMENTATION) PROVIDES THE FIRST TRACTABLE APPROACH TO MAP HISTONE PTMS AT SC RESOLUTION. THIS APPROACH USES ANTIBODIES TO BIND CHROMATIN PROTEINS IN SITU, AND THEN TETHERS A PROTEIN A-PROTEIN G AND HYPERACTIVE TN5 TRANSPOSASE (PAG-TN5) FUSION TO THESE SITES FOR TAGMENTATION. TARGETED DNA IS THEN AMPLIFIED AND SEQUENCED, DELIVERING A STREAMLINED, ULTRASENSITIVE ASSAY FOR HISTONE PTM MAPPING. DESPITE THIS PROGRESS, SC APPLICATIONS STILL PRESENT A UNIQUE CHALLENGE, AS THEY GENERATE INCREDIBLY SPARSE DATA (I.E. THIRD, SUCCESSFUL CANDIDATES ARE PURIFIED AND FURTHER ANALYZED FOR ANTIBODY SPECIFICITY AND EFFICIENCY USING DNA-BARCODED SNAP-CHIP® SPIKE-IN CONTROLS. IN PRELIMINARY STUDIES, WE USED OUR NOVEL APPROACH TO SELECT AND VALIDATE ULTRA-EFFICIENT ANTIBODIES FOR SEVERAL PTM TARGETS, DEVELOPING ANTIBODIES THAT EXHIBIT A >5-10X INCREASE IN NUCLEOSOME CAPTURE EFFICIENCY; THESE HIGH EFFICIENCY ANTIBODIES GENERATED FAR SUPERIOR SIGNAL-TO-NOISE (S/N) IN CUT&TAG ASSAYS (VS. CURRENT BEST-IN-CLASS ANTIBODIES). IN PHASE I, OUR GOAL IS TO DEMONSTRATE THAT ULTRA-EFFICIENT ANTIBODIES IMPROVE 1) S/N IN LOW INPUT BULK CUT&TAG ASSAYS, 2) THE NUMBER OF UNIQUE READS / CELL IN SCCUT&TAG ASSAYS AND 3) ENABLE ANALYSIS OF HIGH VALUE, LOW ABUNDANCE MARKS (E.G. H3K4ME3). IN PHASE II, WE WILL USE THIS NOVEL PIPELINE TO EXPAND DEVELOPMENT OF ULTRA-EFFICIENT HISTONE PTM ANTIBODIES AGAINST A BROAD RANGE OF HIGH VALUE TARGETS. THESE ANTIBODIES WILL ALSO BE USED TO DEVELOP LOW INPUT AND SC CUT&TAG ASSAY KITS TO FURTHER OUR UNDERSTANDING OF CHROMATIN BIOLOGY AND DEVELOP IMPROVED THERAPEUTICS AND DIAGNOSTICS.

LONG-TERM HUMAN FIRE MANAGEMENT AND ENVIRONMENTAL CHANGE IN HIGH-ELEVATION SOCIAL-ECOLOGICAL SYSTEMS -THIS PROJECT INVESTIGATES SOCIAL-ECOLOGICAL RELATIONSHIPS WITHIN PACIFIC NORTHWEST HIGH-ELEVATION ECOLOGICAL ZONES, EMPHASIZING LONG-TERM CULTURAL BURNING PRACTICED BY INDIGENOUS COMMUNITIES. THE PRIMARY OBJECTIVES OF THE PROJECT ARE TO CONDUCT ARCHAEOLOGICAL SURVEYS OF SUBALPINE ECOSYSTEMS; INTERPRET PROCESSES OF HIGH-ELEVATION SUBSISTENCE DURING THE EARLY HOLOCENE AND SUBSEQUENT CHANGES IN BURNING PRACTICES; AND INVESTIGATE HOW SUBSISTENCE LAND MANAGEMENT INFLUENCED HISTORICAL FIRE REGIMES AND LONG-TERM ECOSYSTEM STABILITY/CHANGE. THIS STUDY EMPLOYS AN INTERDISCIPLINARY APPROACH THAT INTERWEAVES ARCHAEOLOGY, PALEOECOLOGY, PARTICIPATORY MAPPING, AND COMPUTATIONAL MODELING TO EVALUATE HOW HUMAN-ENVIRONMENT INTERACTIONS SHAPE HIGH-ELEVATION LANDSCAPES. THIS RESEARCH DEVELOPS PARTNERSHIPS WITH FEDERAL LAND MANAGING AGENCIES AND INDIGENOUS PARTNERS IN THE CO-INTERPRETATION OF RESEARCH RESULTS. HISTORICALLY, FIRE HAS BEEN USED AS A PART OF A SET OF TOOLS TO MANAGE ECOSYSTEMS BUT RELATIVELY LITTLE IS KNOWN ABOUT THE INFLUENCE OF HISTORICAL PRACTICES OF BURNING IN HIGH-ELEVATION ECOSYSTEMS. THIS PROJECT EVALUATES THE SPATIAL DISTRIBUTION AND CHRONOLOGY OF HIGH ELEVATION LAND-USE, DOCUMENTS LONG-TERM TRENDS IN CLIMATE AND HUMAN INFLUENCE ON FIRE AND VEGETATION, AND RECORDS MATERIAL EVIDENCE FOR CULTURAL BURNING PRACTICES TO REJUVENATE OR MAINTAIN IMPORTANT PLANT SPECIES. THIS RESEARCH DEVELOPS A NOVEL PALEO-FIRE RECONSTRUCTION AND INTRODUCES NEW METHODS OF SOIL CHARCOAL ANALYSIS TO MAKE MORE SUBSTANTIAL CONNECTIONS BETWEEN PAST HUMAN LAND-USE, CULTURAL FIRE, AND VEGETATION CHANGE. DATASETS RESULTING FROM THIS PROJECT ARE EVALUATED USING A COMPUTATIONAL ?VIRTUAL LABORATORY? TO EXAMINE DYNAMICS AMONG CLIMATE AND CULTURAL BURNING THAT MAY NOT BE INTERPRETABLE FROM THE ARCHAEOLOGICAL AND FIRE HISTORY DATA ALONE. RESULTS FROM THIS PROJECT ADVANCE THE UNDERSTANDING OF SOCIAL-ENVIRONMENTAL CHANGE AND STABILITY IN HIGH-ELEVATION ECOSYSTEMS TO ENHANCE COOPERATIVE MANAGEMENT STRATEGIES (E.G., CO-STEWARDSHIP, CO-MANAGEMENT), MAINTAIN CULTURAL ECOSYSTEM SERVICES, REDUCE POTENTIAL FUTURE WILDFIRE SEVERITY, AND BUFFER AGAINST ECOSYSTEM LOSS IN THESE UNIQUE LANDSCAPES.
