IT ADVERSELY AFFECTS PATIENTS BY CAUSING SENSORY PAIN IN THE LOWER LIMBS, MUSCULAR WEAKNESS, PREDISPOSITION TO FALLS, DECREASED QUALITY OF LIFE, AND SIGNIFICANT PATIENT MORBIDITY AS A RESULT OF FOOT ULCERATIONS, AMPUTATIONS, AND DISABILITY. PDPN IS ESTIMATED TO COST $10.9 BILLION PER YEAR IN THE US ALONE, WITH MOST COSTS ATTRIBUTED TO THE TREATMENT OF FOOT ULCERS AND SUPERIMPOSED INFECTIONS. TREATMENT FOR PDPN REMAINS LIMITED DESPITE THE SIGNIFICANT ECONOMIC AND MEDICAL IMPACT ON PATIENTS AND THE U.S. HEALTHCARE SYSTEM. ESTABLISHED GUIDELINES ADVOCATE THE USE OF THERAPIES TARGETING THE SYMPTOMS OF PDPN, AND MAY INCLUDE ADMINISTRATION OF ANTICONVULSANTS, ANTIDEPRESSANTS, AND OPIOIDS, AS FIRST-LINE THERAPIES. YET, THESE THERAPIES ONLY OFFER MODERATE PAIN RELIEF, WITH ADVERSE EVENTS OUTWEIGHING PAIN REDUCTION. THE FDA HAS APPROVED VIRTUALLY NO NEW CATEGORIES OF ANALGESICS FOR THE TREATMENT OF NEUROPATHIC PAIN OVER THE LAST TWO DECADES, CREATING A GAPING HOLE IN THE DEVELOPMENT OF EFFECTIVE AND SAFE PDPN-SPECIFIC ALTERNATIVE THERAPIES. THIS PROJECT INTRODUCES A SIGNIFICANT INNOVATION IN THE TREATMENT OF PDPN THROUGH OUR CLINICALLY-DEMONSTRATED NOVEL WATER-SOLUBILIZED CANNABIDIOL-PALMITOYLETHANOLAMIDE (CBD-PEA) TECHNOLOGY. THE FORMULATION AVOIDS FIRST-PASS METABOLISM AND MEDIATES MULTIPLE PAIN PATHWAYS TO PROVIDE THE BEST ANTI-INFLAMMATORY AND ANTIOXIDANT CLINICAL OUTCOMES. OUR CBD-PEA DRUG MAY ELIMINATE THE ADVERSE EFFECTS OF CURRENT THERAPIES, THEREFORE ALLOWING ALL OR NEARLY ALL PDPN PATIENTS TO EXPERIENCE PAIN RELIEF INSTEAD OF ONLY 50% OF THE PATIENTS. PURE GREEN PHARMACEUTICALS (THE COMPANY) HAS SHOWN PRELIMINARY SUCCESS IN PUBLISHED CLINICAL DATA DEMONSTRATING THAT SUBLINGUAL ADMINISTRATION OF LOW-DOSE, WATER-SOLUBILIZED SUBLINGUAL TABLETS CONTAINING CBD (60 MG/DAY) AND PEA (300 MG/DAY), TO PATIENTS WITH CHRONIC DIABETIC NEUROPATHY, SIGNIFICANTLY REDUCED PAIN, IMPROVED SLEEP QUALITY, AND REDUCED ANXIETY [P< 0.0001 IN AN OPEN-LABEL TRIAL, N=31]. SIMILAR RESULTS WERE SEEN IN A CONFIRMATORY DOUBLE-BLIND, PLACEBO-CONTROLLED TRIAL [P<0.0001, N=54]. NO ADVERSE EVENTS WERE REPORTED. THE PROPOSED PROJECT SUBSTANTIALLY EXPANDS THE COMPANY’S CAPABILITIES DEMONSTRATED IN TWO RECENTLY PUBLISHED FOUNDATIONAL PHASE 2 CLINICAL TRIALS THAT STRONGLY SUGGESTED CLINICAL AND STATISTICAL EVIDENCE FOR THE EFFICACY AND SAFETY OF CBD-PEA IN PDPN. THIS STUDY WILL DETERMINE THE IMPACT AND EFFICACY OF THE COMPANY’S WATER-SOLUBLE 20 MG CBD AND MICRONIZED 100 MG PEA SUBLINGUAL TABLET IN PATIENTS SUFFERING FROM PDPN. THIS DIRECT-TO-PHASE II IS A MILESTONE-DRIVEN CLINICAL TRIAL TO VALIDATE THE EFFICACY OF PG PHARMA’S UNIQUE FORMULATION IN PDPN REDUCTION, BY MEASURING PAIN REDUCTION, PATIENTS’ ANXIETY AND SLEEP QUALITY COMPARED TO PLACEBO CONTROL. SEMICONDUCTOR FABRICATION REQUIRES LARGE VOLUMES OF ULTRAPURE WATER, WHICH IS PRODUCED USING EXTENSIVE FILTRATION AND CHEMICAL TREATMENT TO REMOVE CONTAMINANTS THAT CAN AFFECT MANUFACTURING YIELD AND PRODUCT RELIABILITY. CURRENT METHODS, SUCH AS REVERSE OSMOSIS, REQUIRE SIGNIFICANT PRE- AND POST-TREATMENT, INCREASING COSTS, ENERGY CONSUMPTION, AND OPERATIONAL COMPLEXITY. IN ADDITION TO BENEFITING SEMICONDUCTOR MANUFACTURING, THIS TECHNOLOGY HAS THE POTENTIAL TO IMPROVE DESALINATION AND WASTEWATER REUSE, PROVIDING A MORE EFFICIENT AND SCALABLE SOLUTION FOR PRODUCING CLEAN WATER IN REGIONS FACING WATER SHORTAGES. BY SIMPLIFYING WATER PURIFICATION, REDUCING ENERGY CONSUMPTION, AND ELIMINATING COSTLY CHEMICAL TREATMENTS, THIS INNOVATION COULD LOWER OPERATING COSTS FOR INDUSTRIAL WATER USERS, SUPPORT DOMESTIC SEMICONDUCTOR PRODUCTION, AND HELP ADDRESS CRITICAL WATER CHALLENGES WORLDWIDE. THIS PROJECT INTRODUCES A NOVEL PRESSURE-DRIVEN DISTILLATION MEMBRANE THAT ENABLES WATER PURIFICATION WITHOUT THE NEED FOR HIGH THERMAL ENERGY INPUT OR EXTENSIVE CHEMICAL PRE-TREATMENT. UNLIKE TRADITIONAL FILTRATION MEMBRANES, THIS APPROACH USES APPLIED PRESSURE TO DRIVE PHASE SEPARATION, SELECTIVELY REMOVING CONTAMINANTS SUCH AS BORON, UREA, SILICA, AND DISINFECTION BYPRODUCTS THAT ARE DIFFICULT TO FILTER WITH EXISTING MEMBRANE TECHNOLOGIES. THE RESEARCH WILL FOCUS ON OPTIMIZING MEMBRANE FABRICATION, ASSESSING SELECTIVITY AND DURABILITY UNDER OPERATIONAL CONDITIONS, AND INTEGRATING THE MEMBRANE INTO A SCALABLE MODULE SUITABLE FOR SEMICONDUCTOR MANUFACTURING, DESALINATION, AND WASTEWATER REUSE APPLICATIONS. WATERBORNE VIRUSES ARE PRESENT IN LOW CONCENTRATIONS AND THEIR DETECTION REQUIRES THE ANALYSIS OF LARGE SAMPLE VOLUMES. HOWEVER, CURRENT TOOLS ARE DESIGNED FOR SMALL VOLUMES, RESULTING IN LOW SENSITIVITY AND HIGH VARIABILITY IN TESTING RESULTS. THE GOAL OF THIS PROJECT IS TO DEVELOP A COMPREHENSIVE TECHNOLOGICAL SOLUTION TO ENHANCE THE RELIABILITY AND STANDARDIZATION OF WASTEWATER EPIDEMIOLOGY AND DRINKING WATER TESTING, A PRESSING NEED IDENTIFIED BY HEALTH AGENCIES TO IMPROVE DATA QUALITY AND COMPARABILITY. THIS INNOVATION AIMS TO INCREASE THE RESOLUTION AND VOLUME OF CRITICAL DATA BY FOUR TIMES, AIDING SYSTEM OPERATORS IN OPTIMIZING MEMBRANE AND SYSTEM OPERATIONS, RECOVERY, EFFICIENCY, AND MAINTENANCE. THE SIRO TECHNOLOGY INCORPORATES PBT?S PROPRIETARY THIN, FLEXIBLE, FLAT, AND INEXPENSIVE MULTI-SENSOR TECHNOLOGY, ENABLING PRECISE MEASUREMENT AND REPORTING OF WATER QUALITY AND INDIVIDUAL MEMBRANE PERFORMANCE AND HEALTH. IN COLLABORATION WITH THE UNITED STATES BUREAU OF RECLAMATION'S (USBR) WATER QUALITY IMPROVEMENT CENTER (WQIC) IN YUMA, ARIZONA, PBT WILL CONDUCT A 30-90 DAY FIELD PILOT TO EVALUATE THE SIRO TECHNOLOGY'S PERFORMANCE, INSTALLATION, AND USABILITY. DATA MEASURED AND MONITORED BY THE SYSTEM WILL INCLUDE CRITICAL MEMBRANE ELEMENT AND SYSTEM PERFORMANCE, HEALTH, PRODUCTIVITY, EFFICIENCY, AND RECOVERY INFORMATION--DATASETS CURRENTLY UNAVAILABLE WITHIN PRESSURE VESSELS. THIS PROJECT WILL SIGNIFICANTLY IMPROVE REAL-TIME DATA ACQUISITION, SUPPORTING OPERATORS IN OPTIMIZING RESOURCE USAGE, PROCESS CONTROL, MAINTENANCE, AND CLEANING PROCEDURES BY ENHANCING DECISION SUPPORT WITH HIGHER RESOLUTION DATA AND NEW DATA. IT ALSO AIMS TO PROVIDE INTELLIGENCE REGARDING MEMBRANE FOULING AND SCALING AND IDENTIFY FAILED MEMBRANE ELEMENTS WITHIN LARGE-SCALE OPERATIONS. SBIR PHASE I: A VERSATILE NUCLEIC ACID COLLECTION AND PURIFICATION TECHNOLOGY FOR WASTEWATER-BASED EPIDEMIOLOGY -THE BROADER IMPACT/COMMERCIAL POTENTIAL OF THIS SMALL BUSINESS INNOVATION RESEARCH (SBIR) PHASE I PROJECT IS TO PROVIDE PRIVATE AND PUBLIC STAKEHOLDERS WITH NEW TECHNOLOGICAL TOOLS TO BETTER MONITOR AND PREDICT PANDEMICS. THIS PROJECT IS EXPECTED TO MODERNIZE THE TOOLS USED TODAY BY PRIVATE AND PUBLIC LABORATORIES TO COLLECT AND PURIFY PATHOGENS, PARTICULARLY HUMAN CORONAVIRUSES FROM COMMUNITY WASTEWATER, FOR TESTING AND DIAGNOSTIC PURPOSES. THE NEW TECHNOLOGICAL TOOLS ARE EXPECTED TO SIMPLIFY THE WORKFLOW, REDUCE COSTS AND TIME, AND ENABLE THE PREDICTION OF COVID-19 OUTBREAKS AND OTHER PANDEMICS SEVERAL WEEKS BEFORE OBSERVING CLINICAL CASES. THE TECHNOLOGICAL PRODUCTS OF THIS PROJECT ARE EXPECTED TO EMPOWER THE GROWING NUMBER OF COMPANIES AND LABORATORIES OFFERING WASTEWATER-BASED EPIDEMIOLOGY (WBE) SERVICES AND HELP ESTABLISH WBE AS A ROUTINE, COST-EFFECTIVE AND RELIABLE TOOL FOR PUBLIC HEALTH MONITORING. THIS SMALL BUSINESS INNOVATION RESEARCH (SBIR) PHASE I PROJECT WILL ADDRESS A MAJOR TECHNOLOGICAL BARRIER FOR THE DETECTION OF VIRUSES SUCH AS SARS-COV-2 IN WASTEWATER. COMMERCIALLY AVAILABLE NUCLEIC ACID COLLECTION AND PURIFICATION KITS ARE DESIGNED FOR CLINICAL SAMPLES WITH SMALL VOLUMES. THESE KITS ARE NOT GENERALLY USED FOR LARGE WASTEWATER VOLUMES WHERE THE VIRUS IS PRESENT AT LOW CONCENTRATIONS. AS A RESULT, CURRENT PROCESSES ARE TIME-CONSUMING, AND RESULT IN THE RECOVERY OF LESS THAN 30% OF VIRUSES AND NUCLEIC ACIDS, SIGNIFICANTLY REDUCING THE SENSITIVITY. THE GOAL OF THIS SBIR PHASE I PROJECT IS TO DEMONSTRATE THE FEASIBILITY OF A NOVEL VIRUS AND NUCLEIC ACID COLLECTION AND PURIFICATION TECHNOLOGY FROM WASTEWATER. SPECIFICALLY, THE PROJECT TASKS AIM AT ENHANCING UNDERSTANDING OF VIRUS PROPERTIES, PARTICULARLY HUMAN CORONAVIRUSES IN WASTEWATER AND THEIR INTERACTIONS WITH FILTER MEDIA. THE PROJECT?S INNOVATIVE APPROACH IS TO DESIGN A STREAMLINED WORKFLOW THAT INCLUDES ALL THE STEPS FROM SAMPLE COLLECTION TO DETECTION, IN A SINGLE DISPOSABLE CARTRIDGE CONTAINING NOVEL FILTERS WITH HIGH AFFINITY TO VIRUSES. THIS CAPABILITY RESOLVES COMPLEX GENOMIC REGIONS SUCH AS REPETITIVE SEQUENCES, STRUCTURAL VARIATIONS, AND LONG-RANGE HAPLOTYPES OFTEN INACCESSIBLE WITH SHORT READS. LONG-READ TECHNOLOGIES, SUCH AS PACBIO AND OXFORD NANOPORE (ONT), EXCEL IN CAPTURING COMPLEX GENOMIC FEATURES BUT ARE MOST IMPACTFUL WHEN PROCESSING LARGER DNA MOLECULES. AS THESE METHODS IMPROVE IN COST AND ACCURACY, THEIR ADOPTION IS POISED TO ACCELERATE, ADVANCING OUR UNDERSTANDING OF GENOMIC ARCHITECTURE AND ITS RELEVANCE TO HEALTH AND DISEASE. HOWEVER, SAMPLE PREP, PARTICULARLY SIZE SELECTION, REMAINS A MAJOR BOTTLENECK FOR BOTH SEQUENCING METHODS. CURRENT TECHNIQUES, RELYING ON BEADS AND GELS, ARE LABOR-INTENSIVE, ERROR-PRONE, AND COSTLY, HEIGHTENING RISKS OF SAMPLE LOSS AND CONTAMINATION. ACCURATE SIZE SELECTION IS CRUCIAL, ESPECIALLY FOR PACBIO, WHERE FRAGMENT SIZES UNDER 10 KB COMPROMISE PERFORMANCE. TO ADDRESS THIS, MICROPURE GENOMICS INC. PROPOSES AN AUTOMATED, BEAD-LESS, GEL-LESS MICROFLUIDIC SIZE SELECTION APPROACH. THIS INNOVATION SIMPLIFIES WORKFLOWS, REDUCES COSTS, IMPROVES REPRODUCIBILITY, AND ENHANCES SEQUENCING PERFORMANCE, PAVING THE WAY FOR ACCURATE AND COMPREHENSIVE GENOMIC ANALYSES. USING PATENTED ELECTRO-HYDRODYNAMIC (EH) TRAPPING TECHNOLOGY, THE ΜPREP ENABLES GENTLE PURIFICATION AND MICROFLUIDIC HANDLING WITHOUT BEADS, MINIMIZING SAMPLE DEGRADATION. GIVEN THE ENVIRONMENTAL PERSISTENCE OF THESE CONTAMINANTS AND THEIR ESTABLISHED LINKAGE TO SERIOUS HEALTH RISKS, IT IS IMPERATIVE THAT SAFE, EFFICIENT, AND COST-EFFECTIVE PFAS REMEDIATION TECHNOLOGIES BE DEVELOPED THAT CAN ELIMINATE THESE CONTAMINANTS FROM THE U.S. WATER SUPPLIES. WITH THE TIGHTENING OF HEALTH-BASED CONTAMINATION LIMITS AND THE EXPANSION OF REGULATED PFAS STRUCTURES, THE IMPACTS OF PFAS CONTAMINATION ARE NOW ESTIMATED TO AFFECT OVER 200 MILLION AMERICANS. NIEHS SPONSORED DEXSORB ADSORBENT REPRESENTS A BREAKTHROUGH SOLUTION FOR THE SAFE, RAPID, AND COST-EFFECTIVE REMOVAL OF SHORT- AND LONG- CHAIN PFAS COMPOUNDS, INCLUDING PFOA AND PFOS FROM WATER. IT TAKES EXACTING TECHNOLOGY TO REMOVE PFAS COMPOUNDS AT THE TRACE LEVELS BEING MANDATED BY STATES AND THE EPA. MADE FROM RENEWABLE CYCLODEXTRINS, DEXSORB’S ABILITY TO SELECTIVELY TARGET PFAS COMPOUNDS AT TRACE CONCENTRATIONS WITH RAPID UPTAKE, HIGH CAPACITY, AND RESISTANCE TO FOULING MAKE IT IDEALLY SUITED FOR USE IN PRIVATE AND MUNICIPAL WATER TREATMENT SYSTEMS. WORKING WITH ENVIRONMENTAL PARTNERS, CYCLOPURE HAS DEMONSTRATED THE SUPERIOR PERFORMANCE AND FAVORABLE LIFE CYCLE ECONOMIES OF DEXSORB IN PILOT WATER TREATMENT ACTIVITIES. PRIOR TO SEQUENCING A BIOLOGICAL SAMPLE, DNA MUST BE LIBERATED FROM CELLS AND SEPARATED FROM PROTEINS AND OTHER UNWANTED DEBRIS, AND THEN MIXED WITH SPECIALTY BUFFERS AND CHEMICAL AGENTS. THIS SKILLED TASK IS CURRENTLY CARRIED OUT BY TRAINED SCIENTISTS USING LARGELY MANUAL MANIPULATIONS OF THE SAMPLES AND EXPENSIVE EQUIPMENT. THE ALTERNATIVE METHOD PROPOSED IN THIS PROJECT WILL SPEED-UP DIAGNOSES FROM GENOMIC SEQUENCING BY SIGNIFICANTLY REDUCING PREPARATION TIME WHILE ALSO MAKING PREPARATION MORE RELIABLE THROUGH AUTOMATION. SBIR PHASE I: ALGAE CULTIVATION TECHNOLOGY FOR VALUE ADDED CO-PRODUCTS FROM WASTE WATER TREATMENT EFFLUENT -THE BROADER IMPACTS OF THIS SMALL BUSINESS INNOVATIVE RESEARCH (SBIR) PHASE I PROJECT IS TO IMPROVE REUSE OF WASTEWATER. THIS PROJECT ADVANCES A NEW ALGAE REACTOR TO REDUCE BIOLOGICAL NUTRIENT POLLUTION. THIS WILL IMPROVE HEALTH, POTENTIAL LOSS OF COMMERCIAL FISHING RESOURCES, WATER TOURISM, AND PROPERTY VALUE DECLINE. THE DIVERSITY OF PRODUCTS THAT CAN BE GENERATED FROM ALGAE CULTIVATION IMPROVE THE ROBUSTNESS OF THE GLOBAL FOOD CHAIN AND THE SECURITY OF DOMESTIC FEEDSTOCKS, E.G. HIGH NUTRITION FEEDS FOR LIVESTOCK AND AQUACULTURE AND A NEW SOURCE OF CHEMICALS AND BIOFUELS. THE PROPOSED DESIGN FOR MAXIMUM RESOURCE EFFICIENCY IN WATER AND ENERGY ENABLES VALUE EXTRACTION FROM MARGINAL LANDS. THE PROPOSED PROJECT AIMS TO REDUCE LEVELS OF BIOLOGICAL NUTRIENT POLLUTANTS IN THE EFFLUENT OF WASTEWATER TREATMENT FACILITIES?BY THE DEVELOPMENT OF AN INDUSTRIAL SCALE, ALGAE PHOTOBIOREACTOR, DESIGNED TO OPERATE IN A HIGH-VOLUME, CONTINUOUS FLOW PROCESS. ANTICIPATED ADVANTAGES OF THE NEW TECHNOLOGY ARE A REDUCTION IN CAPITAL AND OPERATING EXPENSE COMPARED TO EXISTING NUTRIENT REMOVAL METHODS AND THE CAPACITY TO GENERATE, HIGH-PURITY, ALGAE COPRODUCTS WITH COMMERCIAL VALUE. RESEARCH OBJECTIVES INCLUDE THE FOLLOWING: (1) CREATE A COMPUTATIONAL FLUID DYNAMICS (CFD) MODEL OF THE REACTOR UNDER VARIOUS FLOW CONDITIONS; (2) PERFORM DYE TRACER EXPERIMENTS IN THE REACTOR AS VALIDATION OF THE MODELING RESULTS; (3) INSTALL THE REACTOR AS PART OF AN INTEGRATED NUTRIENT REMOVAL SYSTEM INCLUDING ULTRAFILTRATION, PRODUCT RECYCLE STREAM, NUTRIENT SENSORS AND PROCESS FEEDBACK CONTROL; (4) EVALUATE THE NUTRIENT REMOVAL CAPABILITIES OF THE SYSTEM USING A LIVE ALGAE CULTURE AND EFFLUENT FROM AN ADJACENT WASTEWATER TREATMENT FACILITY; AND (5) PERFORM A TECHNOECONOMIC ANALYSIS OF THE PROPOSED PROCESS. RESULTS OF THE PROJECT WILL PROVIDE A BETTER UNDERSTANDING OF THE EFFECT OF REACTOR DESIGN AND OPERATING PARAMETERS ON THE HYDRAULIC AND SOLIDS RESIDENCE TIME DISTRIBUTIONS, HOW THESE INFLUENCE THE WASTE REMOVAL PERFORMANCE OF THE SYSTEM, AND WHAT DESIGN PARAMETERS EXERT THE MOST CONTROL OVER ECONOMIC FEASIBILITY. THIS AWARD REFLECTS NSF'S STATUTORY MISSION AND HAS BEEN DEEMED WORTHY OF SUPPORT THROUGH EVALUATION USING THE FOUNDATION'S INTELLECTUAL MERIT AND BROADER IMPACTS REVIEW CRITERIA.
