SPRINGDALE, AR 72764 PROJECT DIRECTOR: MELISSA GREENSLADE VICE PRESIDENT OF INSTRUCTION, NORTHWEST TECHNICAL INSTITUTE MGREENSLADE@NWTI.EDU OFFICE: 479-751-8824 EXT. 105 THE NWTI EDUCATION FOUNDATION IS REQUESTING $10,000,000 IN FUNDING FROM THE U.S. DEPARTMENT OF HEALTH AND HUMAN SERVICES, HEALTH RESOURCES AND SERVICES ADMINISTRATION (HRSA) COMMUNITY PROJECT FUNDING/CONGRESSIONALLY DIRECTED SPENDING PROJECT GRANT. THIS $10,000,000 GRANT WILL ASSIST IN THE CONSTRUCTION OF A 45,000 SQ. FT. MEDICAL EDUCATION BUILDING ON THE CAMPUS OF NORTHWEST TECHNICAL INSTITUTE (NWTI) CAMPUS LOCATED IN SPRINGDALE, ARKANSAS. THIS FACILITY WOULD ACCOMMODATE NWTI’S ALLIED HEALTH PROGRAMS IN LICENSED PRACTICAL NURSING (LPN), SURGICAL TECHNOLOGY, CERTIFIED NURSING ASSISTANT (CNA), STERILE PROCESSING, AS WELL AS ADDITIONAL ALLIED HEALTH PROGRAMS BEING CONSIDERED FOR FUTURE ENROLLMENT. THIS NEW MEDICAL EDUCATION BUILDING WOULD ALLOW NWTI TO INCREASE IT ALLIED HEALTH STUDENT ENROLLMENT FROM 100 STUDENTS TO 300 STUDENTS EACH YEAR. THE GRADUATES OF THESE PROGRAMS WILL SIGNIFICANTLY REDUCE THE WORKFORCE GAP IN THE MEDICAL INDUSTRY ACROSS THE NORTHWEST ARKANSAS REGION. THE CONSTRUCTION OF THIS FACILITY AT NWTI WILL ADDRESS THE NUMBER OF TRAINED HEALTHCARE WORKERS THAT WILL ENTER THE WORKFORCE ANNUALLY WHICH WILL SIGNIFICANTLY REDUCE THE WORKFORCE GAP IN THE MEDICAL INDUSTRY ACROSS NORTHWEST ARKANSAS. BASED ON DATA FROM THE NORTHWEST ARKANSAS COUNCIL, THERE ARE APPROXIMATELY 3600 UNFILLED MEDICAL POSITIONS IN THE REGION CURRENTLY AND ABOUT 85% OF THESE POSITIONS ARE IN THE MIDDLE-SKILL DEVELOPMENT AREA. NWTI IS THE PRIMARY EDUCATIONAL FACILITY IN NORTHWEST ARKANSAS THAT IS FOCUSED ON MIDDLE-SKILL DEVELOPMENT TRAINING AND THE PROPOSED MEDICAL EDUCATION BUILDING WILL ENSURE OPPORTUNITY FOR CAREER AND TECHNICAL EDUCATION (CTE) TRAINING THAT WILL SATISFY THIS EMPLOYMENT NEED. BASED ON LABOR MARKET PROJECT IONS, THERE IS AN EMPLOYMENT SHORTFALL OF LICENSED PRACTICAL NURSES, SURGICAL TECHNOLOGISTS, AND CERTIFIED NURSING ASSISTANTS, ETC. IN A MEDICAL INDUSTRY THAT HAS BEEN DECIMATED BY THE COVID PANDEMIC. THE INCREASED ENROLLMENT IN NWTI ALLIED HEALTH PROGRAMS WILL SIGNIFICANTLY DECREASE THE EMPLOYMENT SHORTFALL IN THE REGIONAL MEDICAL INDUSTRY. ADDITIONALLY, NWTI SERVES AN AREA WITH INCREASING DIVERSITY AND HIGH POVERTY LEVELS. MIDDLE-SKILL LEVEL PROGRAMS, LIKE THOSE OFFERED BY NWTI HELP TO BREAK THE BONDS OF GENERATIONAL POVERTY IN THE REGION. NWTI OFFERS PROGRAMS AT A LOWER COST AND SHORTER TIME FRAMES THAN TRADITIONAL HIGHER EDUCATION, MAKING ITS PROGRAMS MORE ACCESSIBLE TO MANY OF THESE STUDENTS. PARTNERSHIPS WITH LOCAL COMMUNITY LEADERS, CHARITABLE ORGANIZATIONS, AND OTHERS HAVE INCREASED SCHOLARSHIP OPPORTUNITIES THAT TARGET STUDENTS IN THIS DEMOGRAPHIC OFFERING THEM THE CHANCE TO PURSUE HIGHER EDUCATION AND RAISE THE ECONOMIC STANDING OF MANY FAMILIES IN THE REGION, AND AS A RESULT, THE ADVANCED TRAINING OF THESE ALLIED HEALTH GRADUATES WILL SIGNIFICANTLY CONTRIBUTE TO ECONOMIC DEVELOPMENT ACROSS NORTHWEST ARKANSAS. NWTI BEGAN THE PROCESS OF PURSUING CONSTRUCTION OF THE MEDICAL EDUCATION BUILDING APPROXIMATELY TWO YEARS AGO BY CONSULTING WITH HOSPITALS, CLINICS, AND OTHER MEDICAL PROVIDERS IN THE REGION, ALONG WITH EDUCATORS, CHARITABLE ORGANIZATIONS AND REGIONAL CHAMBERS OF COMMERCE AND OTHER WORKFORCE GROUPS TO DETERMINE THE NEEDS FOR EXPANSION IN THE MEDICAL AREA. WITH THE SUPPORT OF THESE GROUPS, NWTI BEGAN SEEKING FUNDS FOR THE CONSTRUCTION OF THE MEDICAL EDUCATION BUILDING. OVER A CENTURY AGO, SIMILAR ROD MICROGLIA WERE HAND DRAWN TO INDICATE NEUROPATHOLOGY IN GENERAL PARESIS, TYPHOID, AND CHEMICAL EXPOSURE, AND THEN LARGELY IGNORED. FOLLOWING OUR 2012 PUBLICATION, ROD MICROGLIA HAVE BEEN VISUALIZED IN POST-MORTEM TISSUE WITH CLINICAL AND NEUROPATHOLOGICAL CONFIRMATION OF ALZHEIMER’S DISEASE AND RELATED DEMENTIAS (ADRD), IN ADDITION TO EPILEPSY, MENTAL HEALTH DISORDERS, AND OTHERS. YET NO MOLECULAR TOOLS EXIST FOR PRECISE MOLECULAR INVESTIGATION OF ROD MICROGLIA. AS CELLULAR MECHANISMS OF ADRD AND INJURY INCLUDE NEUROINFLAMMATION AND CONCOMITANT MICROGLIA ACTIVATION, A CRITICAL PROBLEM EMERGES FROM OUR INABILITY TO DETECT AND ANALYZE ROD MICROGLIA WITHIN THE CONTEXT OF ADRD/BRAIN INJURY NEUROINFLAMMATION. THE BODY OF KNOWLEDGE ON ROD MICROGLIA RELIES SOLELY ON HISTOLOGICAL STAINS AND NON-SPECIFIC MICROGLIA ANTIBODIES (E.G., IBA1, CD68, CX3CR1) FOR POST-MORTEM MICROSCOPIC IDENTIFICATION. OUR MOST RECENT SINGLE-CELL RNASEQ DATA FROM 100 ALZHEIMER’S DISEASE (AD) AND CONTROL SUBJECTS IDENTIFIED A UNIQUE CLUSTER OF MICROGLIA DEFINED BY GENES FOR MOTILITY, CYTOSKELETAL ACTIN DYNAMICS, AND MOVEMENT OF MICROGLIAL PROCESSES; CONFIRMATION OF THIS CLUSTER AS ROD MICROGLIA CANNOT OCCUR WITHOUT NEW MOLECULAR TOOLS TO PHENOTYPE THIS ELUSIVE MICROGLIA VARIANT. FOR THIS BIOENGINEERING RESEARCH GRANT (PAR 19-158), COLLECTIONS OF MOLECULAR TOOLS AND MARKERS UNIQUE TO THE ROD MICROGLIA VARIANT WILL BE DEVELOPED, VALIDATED, AND VERIFIED TO ENABLE FUTURE DIAGNOSTIC, PROGNOSTIC, AND MECHANISTIC STUDIES OF NEUROINFLAMMATION PROGRESSION IN CASES OF ADRD AND BRAIN INJURY. TO DEVELOP THESE TOOLS, WE PROPOSE A DIFFUSE BRAIN-INJURY MODEL THAT GENERATES FIELDS OF ROD MICROGLIA ADJACENT TO NEURONAL DENDRITES. FROM THIS TISSUE, WE WILL CAPTURE (LCM) CX3CR1-EGFP ROD MICROGLIA AND NON-ROD MICROGLIA FOR PHAGE DISPLAY BIOPANNING FROM A LIBRARY OF HUMAN-DERIVED VARIABLE HEAVY-CHAIN BINDING DOMAIN ANTIBODIES (DAB). SYNTHETIC CYCLIZED HCDR3 PEPTIDES WILL BE DEVELOPED AS DAB MIMETICS UNIQUE TO ROD MICROGLIA (OBJECTIVE 1). SPATIAL TRANSCRIPTOMICS OF THE ROD MICROGLIA VARIANT USING THE NANOSTRING GEOMX DIGITAL SPATIAL PROFILER WILL DETERMINE DIFFERENTIALLY EXPRESSED GENES BETWEEN ROD AND NON-ROD MICROGLIA (OBJECTIVE 2). PATHWAY ANALYSIS WILL IDENTIFY CELLULAR PROCESSES ASSOCIATED WITH THE STRUCTURE, FUNCTION, MOTILITY, AND ORIGIN OF ROD MICROGLIA. FOR EACH OBJECTIVE, NEW MOLECULAR TOOLS ARE VALIDATED AND VERIFIED ACROSS TIME POST-INJURY, BETWEEN SEXES, AND IN EXPERIMENTAL MODELS OF MIXED PATHOLOGY TBI, STROKE, AND AD (FAMILIAL AND LATE-ONSET AMYLOIDOSIS), AS WELL AS POST-MORTEM HUMAN AD/ADRD TISSUE. AS MILITARY CONNECTED STUDENTS LEARN ABOUT THE ARTS,CUSTOMS,AND TRADITIONS OF THE PREDOMINANTLY HISPANIC REGION,THOSE WHO WERE UNFAMILIAR WITH THE CULTURE WILL GAIN EXPOSURE TO HELP THEM ACCLIMATE TO THE BROADER COMMUNITY. BRIEF SUMMARY OF THE PROPOSED PROJECT, INCLUDING BUT NOT LIMITED TO, THE SCOPE OF THE PROJECT AND PROPOSED OUTCOMES: RCYB IS A COMPREHENSIVE PROGRAM TO ASSIST YOUTH IN COMPLETING HIGH SCHOOL DIPLOMA OR HIGH SCHOOL EQUIVALENCY CREDENTIAL WHILE BEING TRAINED IN THE CONSTRUCTION AND LOGISTICS INDUSTRIES. PARTICIPANTS WILL EARN INDUSTRY-RECOGNIZED CREDENTIALS. HOME BUILDERS INSTITUTE PRE-APPRENTICESHIP CONSTRUCTION TRAINING WILL BE TAUGHT TO 29 YOUTH. RCYB PARTICIPANTS WILL CONSTRUCT HOUSING UNITS ON RIVERSIDE HABITAT FOR HUMANITY PROJECTS (CALVET REN) HOUSES FOR LOW-INCOME VETERANS AND THEIR FAMILIES. THE LOGISTICS PROGRAM WILL BE TAUGHT TO 28 YOUTH TO LEARN THE SKILLS NECESSARY FOR ENTRY-LEVEL EMPLOYMENT IN WAREHOUSING, DISTRIBUTION, MATERIAL HANDLING, AND DISTRIBUTION IN SUPPLY CHAIN FACILITIES. WORK EXPERIENCE TRAINING FOR LOGISTICS IS AT DECKERS, A LOGISTICS DISTRIBUTION CENTER. TO ADDRESS THIS PROBLEM, EL PASO ISD PROPOSES TO IMPLEMENT THE SAFE HORIZONS PROGRAM, AN INNOVATIVE, COMPREHENSIVE INITIATIVE AIMED AT PROMOTING A SAFE AND NURTURING ENVIRONMENT FOR STUDENTS. BY HOLISTICALLY ADDRESSING BOTH PHYSICAL AND PSYCHOLOGICAL SAFETY, THE PROGRAM SEEKS TO CREATE AN ATMOSPHERE WHERE STUDENTS CAN THRIVE ACADEMICALLY, SOCIALLY, AND EMOTIONALLY. FOUR OF EL PASO ISDS MOST ECONOMICALLY DISADVANTAGED AND LOWEST-PERFORMING SCHOOLS WILL PILOT THIS PROGRAM: BOWIE HIGH SCHOOL, GUILLEN MIDDLE SCHOOL, IRVIN HIGH SCHOOL, AND MAGOFFIN MIDDLE SCHOOL. SAFE HORIZONS IS BASED ON THE BUILDING ASSETS AND REDUCING RISKS (BARR) MODEL, DRAWING ON ITS EIGHT STRATEGIES TO DEVELOP A POSITIVE SCHOOL CULTURE, BUILD RELATIONSHIPS, AND REMOVE BARRIERS TO LEARNING. THIS PROGRAM WILL ADDRESS VIOLENCE BY TRAINING SCHOOL PERSONNEL AND EDUCATING STUDENTS ON VIOLENCE PREVENTION, IMPLEMENTING A TECHNOLOGY SOLUTION FOR REPORTING SAFETY CONCERNS, AND HIRING DEDICATED PERSONNEL TO IMPROVE SCHOOL CLIMATE AND SUPPORT STUDENTS. WITH LIMITED RESOURCES, THE DEPARTMENTS OF COUNSELING & ADVISING AND STUDENT AND PARENT SERVICES ARE NOT ABLE TO PROVIDE THE SUPPORT/GUIDANCE THAT CAMPUSES AND STUDENTS NEED. AS A RESULT, STUDENTS HAVE BEEN COPING WITH THE TRAUMA FROM BULLYING WHICH HAS AFFECTED THEIR PERFORMANCE IN SCHOOL. IN SOME CASES, BULLYING HAS INCREASED THE NUMBER OF STUDENTS JOINING GANGS LEADING THEM TO BULLY OTHERS AS A WAY TO COPE, SOME HAVE ISOLATED THEMSELVES FROM PEERS AND FAMILY, AND OTHER HAVE HAD THOUGHTS AND/OR ACTS OF SUICIDE. TO ENSURE EL PASO ISD PROVIDES ITS STUDENTS A QUALITY EDUCATION IN A SUITABLE AND SAFE ENVIRONMENT, THE DISTRICT WILL ESTABLISH A DISTRICT-LEVEL BULLYING PREVENTION COMMITTEE. THE COMMITTEE WILL HAVE CERTIFIED STAFF TO PROVIDE ON-GOING BULLYING PREVENTION THROUGHOUT THE FIVE GEOGRAPHICAL AREAS OF THE DISTRICT: EAST, CENTRAL, SOUTHSIDE, NORTHEAST AND WESTSIDE. ADDITIONALLY, THROUGH SENATE BILL 2050, THE DISTRICT-LEVEL COMMITTEE WILL ASSIST ALL EIGHTY CAMPUSES IN ESTABLISHING THEIR OWN BULLYING PREVENTION COMMITTEE. EACH COMMITTEE WILL ADOPT A BULLYING PREVENTION CURRICULUM THAT WILL BE EMBEDDED INTO THE STUDENTS’ DAILY INSTRUCTION. STUDENTS THAT CONTINUE TO BULLY WILL BE ASSIGNED TO COMPLETE ANTI-BULLYING LEARNING MODULES. PEERS ARE UNIQUELY SUITED TO PROVIDE THIS CRUCIAL SUPPORT AS THEY ARE PERSONALLY FAMILIAR WITH THE STRUGGLES OF ADDICTION AND RECOVERY. BY PROVIDING THESE THREE HEALTH SYSTEMS WITH THE INITIAL FUNDING TO ESTABLISH THESE PROGRAMS AND HIRE AND TRAIN PRS AND OTHER NEEDED SUPPORT STAFF, THE SYSTEMS WILL BE ABLE TO SUSTAIN THE WORK AFTER THE GRANT PERIOD ENDS BY UTILIZING THE VIRGINIA MEDICAID ADDICTION AND RECOVERY TREATMENT SERVICES (ARTS) WAIVER ALONG WITH OTHER FUNDING OPPORTUNITIES. THE HEALTH SYSTEMS COMMITTED TO ESTABLISHING HOSPITAL-BASED SUBSTANCE USE INTERVENTION PROGRAMS INCLUDE VALLEY HEATH, AUGUSTA HEALTH, AND CENTRA HEALTH WHICH SERVE THE POPULATIONS OF WINCHESTER, STAUNTON, FISHERSVILLE, FARMVILLE, AND LYNCHBURG. ALL THESE AREAS ARE CONSIDERED RURAL AND HAVE SIMILAR DEMOGRAPHICS INCLUDING A LESS THAN 40% BACHELOR'S DEGREE RATE, AN AVERAGE 29% MINORITY POPULATION, AND INCREASING RATES OF EMERGENCY DEPARTMENT VISITS RELATED TO OVERDOSE AND SUBSTANCE USE RELATED CO-MORBIDITIES. IN 2023, THE PARTICIPATING HEALTH SYSTEMS RECORDED A COMBINED TOTAL OF 2,161 EMERGENCY DEPARTMENT VISITS THAT RECOGNIZED SUBSTANCE USE-RELATED ADMISSION CODES. OF THOSE, 459 VISITS WERE FOR THE TREATMENT OF DRUG OVERDOSE. THIS REPRESENTS, ON AVERAGE, A 64% INCREASE IN OVERDOSE-RELATED VISITS ACROSS ALL THE PARTICIPATING HOSPITALS SINCE 2019, WITH CENTRA SOUTHSIDE COMMUNITY HOSPITAL REPORTING A 125% INCREASE IN THE PAST 5 YEARS. IT IS THE GOAL OF THIS PROJECT TO PROVIDE SUPPORT FOR AT LEAST 100 PROGRAM PARTICIPANTS OVER THE COURSE OF THE GRANT YEAR. THIS ALLOWS NECESSARY TIME FOR HIRING, ONBOARDING AND TRAINING SEVEN PEER RECOVERY SPECIALISTS, AND A GRADUAL RAMP-UP TO A FULL CASELOAD WHICH MAY BE ACHIEVED IN THE YEAR FOLLOWING GRANT FUNDING. ALTHOUGH THIS REPRESENTS A SMALL NUMBER OF THE TOTAL PATIENTS PRESENTING TO THE ED ANNUALLY, IT WILL SIGNIFICANTLY IMPACT THE SURVIVAL RATE OF PATIENTS WHO ENTER THE PROGRAM AND ALLOW FOR ADDITIONAL DATA COLLECTION THAT WILL ASSIST OTHER HEALTH SYSTEMS IN DEVELOPING SIMILAR PROGRAMS. PEER RECOVERY SPECIALISTS WILL BE EXPECTED TO COMPLETE THE VIRGINIA DEPARTMENT OF BEHAVIORAL HEALTH AND DEVELOPMENTAL SERVICES 72-HOUR PRS TRAINING PROGRAM AND WORK WITH THEIR SUPERVISORS TO ACCUMULATE THE 500 HOURS OF EXPERIENCE REQUIRED FOR VIRGINIA PRS CERTIFICATION. AFTER RECEIVING CERTIFICATION AND REGISTERING WITH THE BOARD OF COUNSELING, THE PEER RECOVERY SERVICES WILL QUALIFY FOR REIMBURSEMENT THROUGH THE ARTS WAIVER, ENSURING FISCAL SUSTAINABILITY FOR THE PROGRAM. THIS NEW MEDICAL EDUCATION BUILDING WOULD ALLOW NWTI TO INCREASE IT ALLIED HEALTH STUDENT ENROLLMENT FROM 100 STUDENTS TO 300 STUDENTS EACH YEAR. THE GRADUATES OF THESE PROGRAMS WILL SIGNIFICANTLY REDUCE THE WORKFORCE GAP IN THE MEDICAL INDUSTRY ACROSS THE NORTHWEST ARKANSAS REGION. NWTI IS THE PRIMARY EDUCATIONAL FACILITY IN NORTHWEST ARKANSAS THAT IS FOCUSED ON MIDDLE-SKILL DEVELOPMENT TRAINING AND THE PROPOSED MEDICAL EDUCATION BUILDING WILL ENSURE OPPORTUNITY FOR CAREER AND TECHNICAL EDUCATION (CTE) TRAINING THAT WILL SATISFY THIS EMPLOYMENT NEED. BASED ON LABOR MARKET PROJECT IONS, THERE IS AN EMPLOYMENT SHORTFALL OF LICENSED PRACTICAL NURSES, SURGICAL TECHNOLOGISTS, AND CERTIFIED NURSING ASSISTANTS, ETC. IN A MEDICAL INDUSTRY THAT HAS BEEN DECIMATED BY THE COVID PANDEMIC. THE INCREASED ENROLLMENT IN NWTI ALLIED HEALTH PROGRAMS WILL SIGNIFICANTLY DECREASE THE EMPLOYMENT SHORTFALL IN THE REGIONAL MEDICAL INDUSTRY. ADDITIONALLY, NWTI SERVES AN AREA WITH INCREASING DIVERSITY AND HIGH POVERTY LEVELS. MIDDLE-SKILL LEVEL PROGRAMS, LIKE THOSE OFFERED BY NWTI HELP TO BREAK THE BONDS OF GENERATIONAL POVERTY IN THE REGION. NWTI OFFERS PROGRAMS AT A LOWER COST AND SHORTER TIME FRAMES THAN TRADITIONAL HIGHER EDUCATION, MAKING ITS PROGRAMS MORE ACCESSIBLE TO MANY OF THESE STUDENTS. PARTNERSHIPS WITH LOCAL COMMUNITY LEADERS, CHARITABLE ORGANIZATIONS, AND OTHERS HAVE INCREASED SCHOLARSHIP OPPORTUNITIES THAT TARGET STUDENTS IN THIS DEMOGRAPHIC OFFERING THEM THE CHANCE TO PURSUE HIGHER EDUCATION AND RAISE THE ECONOMIC STANDING OF MANY FAMILIES IN THE REGION, AND AS A RESULT, THE ADVANCED TRAINING OF THESE ALLIED HEALTH GRADUATES WILL SIGNIFICANTLY CONTRIBUTE TO ECONOMIC DEVELOPMENT ACROSS NORTHWEST ARKANSAS. NWTI BEGAN THE PROCESS OF PURSUING CONSTRUCTION OF THE MEDICAL EDUCATION BUILDING APPROXIMATELY TWO YEARS AGO BY CONSULTING WITH HOSPITALS, CLINICS, AND OTHER MEDICAL PROVIDERS IN THE REGION, ALONG WITH EDUCATORS, CHARITABLE ORGANIZATIONS AND REGIONAL CHAMBERS OF COMMERCE AND OTHER WORKFORCE GROUPS TO DETERMINE THE NEEDS FOR EXPANSION IN THE MEDICAL AREA. WITH THE SUPPORT OF THESE GROUPS, NWTI BEGAN SEEKING FUNDS FOR THE CONSTRUCTION OF THE MEDICAL EDUCATION BUILDING. THE $10,000,000.00 REQUESTED IN THIS GRANT, ALONG WITH PREVIOUSLY RAISED FUNDS, WILL GO TOWARD THE CONSTRUCTION AND FURNISHING OF THIS MEDICAL EDUCATIONAL BUILDING, INCLUDING WORK TO PREPARE THE SITE FOR CONSTRUCTION, ARCHITECTURAL FEES, CONSTRUCTION MANAGEMENT FEES, INSTRUCTIONAL TECHNOLOGY EQUIPMENT, SPECIALTY MEDICAL TRAINING EQUIPMENT, AND INTERIOR AND EXTERIOR FURNISHINGS.

MOLECULAR TOOL DEVELOPMENT TO IDENTIFY, ISOLATE, AND INTERROGATE THE ROD MICROGLIA PHENOTYPE IN NEUROLOGICAL DISEASE AND INJURY - OUR PATHOLOGICAL EXAMINATION OF ACQUIRED NEUROLOGICAL INJURY UNCOVERED THE ROD MICROGLIA VARIANT OF ACTIVATED IMMUNE CELLS IN THE BRAIN. OVER A CENTURY AGO, SIMILAR ROD MICROGLIA WERE HAND DRAWN TO INDICATE NEUROPATHOLOGY IN GENERAL PARESIS, TYPHOID, AND CHEMICAL EXPOSURE, AND THEN LARGELY IGNORED. FOLLOWING OUR 2012 PUBLICATION, ROD MICROGLIA HAVE BEEN VISUALIZED IN POST-MORTEM TISSUE WITH CLINICAL AND NEUROPATHOLOGICAL CONFIRMATION OF ALZHEIMER’S DISEASE AND RELATED DEMENTIAS (ADRD), IN ADDITION TO EPILEPSY, MENTAL HEALTH DISORDERS, AND OTHERS. YET NO MOLECULAR TOOLS EXIST FOR PRECISE MOLECULAR INVESTIGATION OF ROD MICROGLIA. AS CELLULAR MECHANISMS OF ADRD AND INJURY INCLUDE NEUROINFLAMMATION AND CONCOMITANT MICROGLIA ACTIVATION, A CRITICAL PROBLEM EMERGES FROM OUR INABILITY TO DETECT AND ANALYZE ROD MICROGLIA WITHIN THE CONTEXT OF ADRD/BRAIN INJURY NEUROINFLAMMATION. THE BODY OF KNOWLEDGE ON ROD MICROGLIA RELIES SOLELY ON HISTOLOGICAL STAINS AND NON-SPECIFIC MICROGLIA ANTIBODIES (E.G., IBA1, CD68, CX3CR1) FOR POST-MORTEM MICROSCOPIC IDENTIFICATION. OUR MOST RECENT SINGLE-CELL RNASEQ DATA FROM 100 ALZHEIMER’S DISEASE (AD) AND CONTROL SUBJECTS IDENTIFIED A UNIQUE CLUSTER OF MICROGLIA DEFINED BY GENES FOR MOTILITY, CYTOSKELETAL ACTIN DYNAMICS, AND MOVEMENT OF MICROGLIAL PROCESSES; CONFIRMATION OF THIS CLUSTER AS ROD MICROGLIA CANNOT OCCUR WITHOUT NEW MOLECULAR TOOLS TO PHENOTYPE THIS ELUSIVE MICROGLIA VARIANT. FOR THIS BIOENGINEERING RESEARCH GRANT (PAR 19-158), COLLECTIONS OF MOLECULAR TOOLS AND MARKERS UNIQUE TO THE ROD MICROGLIA VARIANT WILL BE DEVELOPED, VALIDATED, AND VERIFIED TO ENABLE FUTURE DIAGNOSTIC, PROGNOSTIC, AND MECHANISTIC STUDIES OF NEUROINFLAMMATION PROGRESSION IN CASES OF ADRD AND BRAIN INJURY. TO DEVELOP THESE TOOLS, WE PROPOSE A DIFFUSE BRAIN-INJURY MODEL THAT GENERATES FIELDS OF ROD MICROGLIA ADJACENT TO NEURONAL DENDRITES. FROM THIS TISSUE, WE WILL CAPTURE (LCM) CX3CR1-EGFP ROD MICROGLIA AND NON-ROD MICROGLIA FOR PHAGE DISPLAY BIOPANNING FROM A LIBRARY OF HUMAN-DERIVED VARIABLE HEAVY-CHAIN BINDING DOMAIN ANTIBODIES (DAB). SYNTHETIC CYCLIZED HCDR3 PEPTIDES WILL BE DEVELOPED AS DAB MIMETICS UNIQUE TO ROD MICROGLIA (OBJECTIVE 1). SPATIAL TRANSCRIPTOMICS OF THE ROD MICROGLIA VARIANT USING THE NANOSTRING GEOMX DIGITAL SPATIAL PROFILER WILL DETERMINE DIFFERENTIALLY EXPRESSED GENES BETWEEN ROD AND NON-ROD MICROGLIA (OBJECTIVE 2). PATHWAY ANALYSIS WILL IDENTIFY CELLULAR PROCESSES ASSOCIATED WITH THE STRUCTURE, FUNCTION, MOTILITY, AND ORIGIN OF ROD MICROGLIA. FOR EACH OBJECTIVE, NEW MOLECULAR TOOLS ARE VALIDATED AND VERIFIED ACROSS TIME POST-INJURY, BETWEEN SEXES, AND IN EXPERIMENTAL MODELS OF MIXED PATHOLOGY TBI, STROKE, AND AD (FAMILIAL AND LATE-ONSET AMYLOIDOSIS), AS WELL AS POST-MORTEM HUMAN AD/ADRD TISSUE. THE NEW TOOLS PROPOSED IN THIS APPLICATION WILL BE ABLE TO IDENTIFY, ISOLATE, AND INTERROGATE THE ROD MICROGLIA VARIANT, WHILE EXPANDING THE CONCEPT OF NEUROINFLAMMATION IN AGING, INJURY, AND DISEASE TO INCLUDE THE OVERLOOKED ROD MICROGLIA VARIANT THAT PRESENTS ADJACENT TO NEURONAL DENDRITES. WITH REFINED MOLECULAR TOOLS, ROD MICROGLIA CAN BE INVESTIGATED ACROSS MANY NEUROLOGICAL CONDITIONS AND INCORPORATED INTO UNIFYING PRINCIPLES THAT EXPLAIN MICROGLIAL ACTIVATION AND NEUROINFLAMMATION. 33

HISTORICALLY THE DISTRICTS THREE THOUSAND EIGHT HUNDRED SEVENTY NINE MILITARY CONNECTED STUDENTS FROM FORT BLISS HAVE HAD LOWER PARTICIPATION IN DUAL LANGUAGE BECAUSE THEIR HIGH MOBILITY PROHIBITS THEM FROM TAKING PART IN THE COMPREHENSIVE ELEMENTARY PROGRAM. BLISS,HUGHEY,MILAM,AND POWELL ELEMENTARY SCHOOLS WILL PILOT THE ALTERNATIVE PROGRAM CONEXIONES THAT WILL PROVIDE K THROUGH 5 STUDENTS WITH SPANISH LANGUAGE EDUCATION COMPLEMENTED BY LESSONS ON HISPANIC CULTURE. AS MILITARY CONNECTED STUDENTS LEARN ABOUT THE ARTS,CUSTOMS,AND TRADITIONS OF THE PREDOMINANTLY HISPANIC REGION,THOSE WHO WERE UNFAMILIAR WITH THE CULTURE WILL GAIN EXPOSURE TO HELP THEM ACCLIMATE TO THE BROADER COMMUNITY. OUTSIDE OF REGULAR CLASSROOM ACTIVITIES,CONEXIONES WILL ENGAGE PARENTS THROUGH MONTHLY PROGRAM NEWSLETTERS AND FAMILY NIGHT ACTIVITIES CULMINATING IN THE ANNUAL WORLD LANGUAGE FESTIVAL,FURTHER CEMENTING THE CONNECTIONS BETWEEN MILITARY CONNECTED FAMILIES AND THE GREATER EL PASO COMMUNITY. THE PROGRAM WILL BE SUPPORTED BY PROFESSIONAL DEVELOPMENT FOR TEACHERS AND ADMINISTRATORS THAT WILL BUILD EL PASO ISD CAPACITY TO IMPLEMENT CONEXIONES AND EVENTUALLY EXPAND IT TO OTHER CAMPUSES AND STUDENT POPULATIONS. RCYB PARTICIPANTS WILL CONSTRUCT HOUSING UNITS ON RIVERSIDE HABITAT FOR HUMANITY PROJECTS (CALVET REN) HOUSES FOR LOW-INCOME VETERANS AND THEIR FAMILIES. THE LOGISTICS PROGRAM WILL BE TAUGHT TO 28 YOUTH TO LEARN THE SKILLS NECESSARY FOR ENTRY-LEVEL EMPLOYMENT IN WAREHOUSING, DISTRIBUTION, MATERIAL HANDLING, AND DISTRIBUTION IN SUPPLY CHAIN FACILITIES. WORK EXPERIENCE TRAINING FOR LOGISTICS IS AT DECKERS, A LOGISTICS DISTRIBUTION CENTER. SINCE THE START OF THE COVID-19 PANDEMIC, EL PASO INDEPENDENT SCHOOL DISTRICT (EL PASO ISD) SCHOOLS HAVE SEEN AN INCREASE IN BEHAVIORAL ISSUES. FIGHTING/MUTUAL COMBAT IS ONE OF THE MOST COMMON REASONS CITED FOR DISCIPLINARY ACTIONS. TO ADDRESS THIS PROBLEM, EL PASO ISD PROPOSES TO IMPLEMENT THE SAFE HORIZONS PROGRAM, AN INNOVATIVE, COMPREHENSIVE INITIATIVE AIMED AT PROMOTING A SAFE AND NURTURING ENVIRONMENT FOR STUDENTS. BY HOLISTICALLY ADDRESSING BOTH PHYSICAL AND PSYCHOLOGICAL SAFETY, THE PROGRAM SEEKS TO CREATE AN ATMOSPHERE WHERE STUDENTS CAN THRIVE ACADEMICALLY, SOCIALLY, AND EMOTIONALLY. FOUR OF EL PASO ISDS MOST ECONOMICALLY DISADVANTAGED AND LOWEST-PERFORMING SCHOOLS WILL PILOT THIS PROGRAM: BOWIE HIGH SCHOOL, GUILLEN MIDDLE SCHOOL, IRVIN HIGH SCHOOL, AND MAGOFFIN MIDDLE SCHOOL. SAFE HORIZONS IS BASED ON THE BUILDING ASSETS AND REDUCING RISKS (BARR) MODEL, DRAWING ON ITS EIGHT STRATEGIES TO DEVELOP A POSITIVE SCHOOL CULTURE, BUILD RELATIONSHIPS, AND REMOVE BARRIERS TO LEARNING. THIS PROGRAM WILL ADDRESS VIOLENCE BY TRAINING SCHOOL PERSONNEL AND EDUCATING STUDENTS ON VIOLENCE PREVENTION, IMPLEMENTING A TECHNOLOGY SOLUTION FOR REPORTING SAFETY CONCERNS, AND HIRING DEDICATED PERSONNEL TO IMPROVE SCHOOL CLIMATE AND SUPPORT STUDENTS. AFTER TWO YEARS, SAFE HORIZONS SCHOOLS SHOULD REPORT FEWER DISCIPLINARY ACTIONS AND INCREASED ATTENDANCE RATES, MATH AND READING SCORES, AND GRADE-LEVEL ADVANCEMENT.INDIVIDUAL STUDENTS ARE EXPECTED TO REPORT IMPROVED SELF-REGULATION SKILLS, TRUST IN TEACHERS AND PEERS, AND PERCEPTION OF SCHOOL CLIMATE.

EL PASO INDEPENDENT SCHOOL DISTRICT (EL PASO ISD) HAS NOTICED AN INCREASE IN THE NUMBER OF STUDENTS REPORTING EPISODES OF BULLYING. WITH LIMITED RESOURCES, THE DEPARTMENTS OF COUNSELING & ADVISING AND STUDENT AND PARENT SERVICES ARE NOT ABLE TO PROVIDE THE SUPPORT/GUIDANCE THAT CAMPUSES AND STUDENTS NEED. AS A RESULT, STUDENTS HAVE BEEN COPING WITH THE TRAUMA FROM BULLYING WHICH HAS AFFECTED THEIR PERFORMANCE IN SCHOOL. IN SOME CASES, BULLYING HAS INCREASED THE NUMBER OF STUDENTS JOINING GANGS LEADING THEM TO BULLY OTHERS AS A WAY TO COPE, SOME HAVE ISOLATED THEMSELVES FROM PEERS AND FAMILY, AND OTHER HAVE HAD THOUGHTS AND/OR ACTS OF SUICIDE. TO ENSURE EL PASO ISD PROVIDES ITS STUDENTS A QUALITY EDUCATION IN A SUITABLE AND SAFE ENVIRONMENT, THE DISTRICT WILL ESTABLISH A DISTRICT-LEVEL BULLYING PREVENTION COMMITTEE. THE COMMITTEE WILL HAVE CERTIFIED STAFF TO PROVIDE ON-GOING BULLYING PREVENTION THROUGHOUT THE FIVE GEOGRAPHICAL AREAS OF THE DISTRICT: EAST, CENTRAL, SOUTHSIDE, NORTHEAST AND WESTSIDE. ADDITIONALLY, THROUGH SENATE BILL 2050, THE DISTRICT-LEVEL COMMITTEE WILL ASSIST ALL EIGHTY CAMPUSES IN ESTABLISHING THEIR OWN BULLYING PREVENTION COMMITTEE. EACH COMMITTEE WILL ADOPT A BULLYING PREVENTION CURRICULUM THAT WILL BE EMBEDDED INTO THE STUDENTS’ DAILY INSTRUCTION. STUDENTS THAT CONTINUE TO BULLY WILL BE ASSIGNED TO COMPLETE ANTI-BULLYING LEARNING MODULES. BY THE END OF THE PROGRAM, EL PASO ISD WILL HAVE ADOPTED A COMPREHENSIVE BULLYING PREVENTION APPROACH THAT WILL CREATE A LONG-TERM CHANGE THAT CREATES A SAFE AND POSITIVE SCHOOL CLIMATE (OLWEUS, 2021).

HOSPITAL-BASED SUBSTANCE USE INTERVENTION PROGRAM - THE VIRGINIA HOSPITAL RESEARCH AND EDUCATION FOUNDATION, IN PARTNERSHIP WITH THREE MEMBER HEALTH SYSTEMS, WILL DEVELOP AND IMPLEMENT HOSPITAL-BASED SUBSTANCE USE INTERVENTION PROGRAMS WHICH UTILIZE THE EVIDENCE-BASED BEST PRACTICE OF CONNECTING PATIENTS IDENTIFIED WITH SUBSTANCE USE DISORDER TO A PEER RECOVERY SPECIALIST (PRS). THESE PEER RECOVERY SPECIALISTS WILL UNDERGO TRAINING AND CERTIFICATION TO FACILITATE THE IMPROVEMENT OF PATIENT OUTCOMES, STARTING WHEN A PATIENT PRESENTS IN THE EMERGENCY DEPARTMENT AND CONTINUING BY GUIDING THE PATIENT TO COMMUNITY RESOURCES AND THROUGH THE STAGES OF RECOVERY. PEERS ARE UNIQUELY SUITED TO PROVIDE THIS CRUCIAL SUPPORT AS THEY ARE PERSONALLY FAMILIAR WITH THE STRUGGLES OF ADDICTION AND RECOVERY. BY PROVIDING THESE THREE HEALTH SYSTEMS WITH THE INITIAL FUNDING TO ESTABLISH THESE PROGRAMS AND HIRE AND TRAIN PRS AND OTHER NEEDED SUPPORT STAFF, THE SYSTEMS WILL BE ABLE TO SUSTAIN THE WORK AFTER THE GRANT PERIOD ENDS BY UTILIZING THE VIRGINIA MEDICAID ADDICTION AND RECOVERY TREATMENT SERVICES (ARTS) WAIVER ALONG WITH OTHER FUNDING OPPORTUNITIES. IT IS THE GOAL OF THE FOUNDATION THAT THESE EARLY ADOPTERS WILL ASSIST IN ESTABLISHING A PEER RECOVERY SUPPORT MODEL THAT CAN BE EASILY DUPLICATED BY OTHER HOSPITALS WHICH ARE EXPERIENCING A GROWING RATE OF OPIOID RELATED EMERGENCY DEPARTMENT ADMISSIONS. THE HEALTH SYSTEMS COMMITTED TO ESTABLISHING HOSPITAL-BASED SUBSTANCE USE INTERVENTION PROGRAMS INCLUDE VALLEY HEATH, AUGUSTA HEALTH, AND CENTRA HEALTH WHICH SERVE THE POPULATIONS OF WINCHESTER, STAUNTON, FISHERSVILLE, FARMVILLE, AND LYNCHBURG. ALL THESE AREAS ARE CONSIDERED RURAL AND HAVE SIMILAR DEMOGRAPHICS INCLUDING A LESS THAN 40% BACHELOR'S DEGREE RATE, AN AVERAGE 29% MINORITY POPULATION, AND INCREASING RATES OF EMERGENCY DEPARTMENT VISITS RELATED TO OVERDOSE AND SUBSTANCE USE RELATED CO-MORBIDITIES. IN 2023, THE PARTICIPATING HEALTH SYSTEMS RECORDED A COMBINED TOTAL OF 2,161 EMERGENCY DEPARTMENT VISITS THAT RECOGNIZED SUBSTANCE USE-RELATED ADMISSION CODES. OF THOSE, 459 VISITS WERE FOR THE TREATMENT OF DRUG OVERDOSE. THIS REPRESENTS, ON AVERAGE, A 64% INCREASE IN OVERDOSE-RELATED VISITS ACROSS ALL THE PARTICIPATING HOSPITALS SINCE 2019, WITH CENTRA SOUTHSIDE COMMUNITY HOSPITAL REPORTING A 125% INCREASE IN THE PAST 5 YEARS. IT IS THE GOAL OF THIS PROJECT TO PROVIDE SUPPORT FOR AT LEAST 100 PROGRAM PARTICIPANTS OVER THE COURSE OF THE GRANT YEAR. THIS ALLOWS NECESSARY TIME FOR HIRING, ONBOARDING AND TRAINING SEVEN PEER RECOVERY SPECIALISTS, AND A GRADUAL RAMP-UP TO A FULL CASELOAD WHICH MAY BE ACHIEVED IN THE YEAR FOLLOWING GRANT FUNDING. ALTHOUGH THIS REPRESENTS A SMALL NUMBER OF THE TOTAL PATIENTS PRESENTING TO THE ED ANNUALLY, IT WILL SIGNIFICANTLY IMPACT THE SURVIVAL RATE OF PATIENTS WHO ENTER THE PROGRAM AND ALLOW FOR ADDITIONAL DATA COLLECTION THAT WILL ASSIST OTHER HEALTH SYSTEMS IN DEVELOPING SIMILAR PROGRAMS. PEER RECOVERY SPECIALISTS WILL BE EXPECTED TO COMPLETE THE VIRGINIA DEPARTMENT OF BEHAVIORAL HEALTH AND DEVELOPMENTAL SERVICES 72-HOUR PRS TRAINING PROGRAM AND WORK WITH THEIR SUPERVISORS TO ACCUMULATE THE 500 HOURS OF EXPERIENCE REQUIRED FOR VIRGINIA PRS CERTIFICATION. AFTER RECEIVING CERTIFICATION AND REGISTERING WITH THE BOARD OF COUNSELING, THE PEER RECOVERY SERVICES WILL QUALIFY FOR REIMBURSEMENT THROUGH THE ARTS WAIVER, ENSURING FISCAL SUSTAINABILITY FOR THE PROGRAM. LEGITIMIZING THE ROLE OF PEER SUPPORT IN THE RECOVERY PROCESS NOT ONLY INCREASES POSITIVE OUTCOMES FOR THOSE IN ACTIVE ADDICTION, BUT ALSO GIVES HOPE TO THOSE IN ACTIVE RECOVERY THAT THERE ARE OPPORTUNITIES FOR THEM TO GIVE BACK TO THEIR COMMUNITIES IN A MEASURABLE WAY.

UNFORTUNATELY MONOTHERAPIES TARGETING KNOWN DRIVERS OF GLIOBLASTOMAS SUCH AS HYPERACTIVE EGFR AND C-MET RTKS HAVE PROVEN TO BE INEFFECTIVE SUGGESTING REDUNDANT MECHANISMS OF ACTIVATION. WE HAVE IDENTIFIED CIRCULAR RNA-DERIVED PROTEINS WHICH ARE OVEREXPRESSED IN GLIOBLASTOMA AND THESE NOVEL PRODUCTS ARE CAPABLE OF STIMULATING RECEPTOR TYROSINE KINASE ACTIVITY LEADING TO ENHANCED PROLIFERATION, MOBILITY AND INVASIVE CHARACTERISTICS. IN THIS APPLICATION WE PROPOSE TO 1.) DETERMINE THE MECHANISM(S) OF CIRC-HGF MEDIATED ACTIVATION OF C-MET IN GLIOBLASTOMA, 2.) CLARIFY THE ROLE OF PKA/PTBP1 SIGNALING IN THE REGULATION OF CIRC-HGF AND CIRC-E-CAD TRANSLATION INITIATION AND EXPRESSION IN GBM, 3.) EVALUATE A NOVEL SMALL MOLECULE INHIBITOR OF CIRC-HGF AND CIRC-E-CAD IRES-DEPENDENT TRANSLATION IN ORTHOTOPIC GBM PDX MODELS. WE ALSO PROPOSE TO INVESTIGATE AND CHEMICALLY MODIFY THE INHIBITOR TO BUILD IN DESIRED ANTI-GLIOBLASTOMA EFFECTS.

THE STUDY WILL LEAD TO THE DEVELOPMENT OF BEST PRACTICES THAT SPUR STARTUP GROWTH THROUGH THE COMMERCIALIZATION PROCESS TO ENSURE THAT THE STATE OF TEXAS IS GIVING THE WORLD ACCESS TO CUTTING-EDGE TECHNOLOGIES DEVELOPED IN TEXAS UNIVERSITIES, WHILE CREATING A TECHNOLOGY BUSINESS-FRIENDLY AND COMPETITIVE ENVIRONMENT, ULTIMATELY CREATING HIGH-SKILL HIGH-VALUE JOBS THAT WILL HELP TO DIVERSIFY THE STATE-WIDE ECONOMY AND NATIONALLY.
