THE ADAPTIVE AND RESILIENT INFRASTRUCTURES DRIVEN BY SOCIAL EQUITY (ARISE) PROJECT WILL DEVELOP A SOCIAL-EQUITY-DRIVEN PARADIGM FOR RESILIENCE ANALYSIS AS WELL AS A PIPELINE OF DECISION MAKERS AND COMMUNITY LEADERS FOR THE TRANSFORMATION OF HOW COMMUNITIES INVEST IN AND MANAGE PHYSICAL AND HUMAN INFRASTRUCTURE. FRAMED AROUND FOUR THEMES, ARISE RESEARCH AIMS TO INNOVATE AND TRANSFORM HOW COMMUNITIES IN KANSAS CONSIDER RESILIENCE IN THE FACE OF HAZARDS AND DISASTERS. THESE THEMES ARE SUPPORTED BY EXTENSIVE COMMUNITY ENGAGEMENT, COMMUNITY ENGAGED RESEARCH, AND EDUCATION AND OUTREACH COMPONENTS. THE ARISE PROJECT WILL INCREASE RESEARCH, EDUCATION, AND TRAINING CAPACITY IN DATA SCIENCE, MACHINE LEARNING, COMPLEX NETWORK MODELING, DECISION AND RISK MANAGEMENT SCIENCE, AND SOCIAL EQUITY. THIS PROJECT WILL ALSO DEVELOP THE FOUNDATION FOR ?SMART? COMMUNITIES AND CITIES WHILE CREATING NEW INDUSTRIES THAT RETAIN HIGHLY QUALIFIED KANSAS PROFESSIONALS. ARISE WILL BE ADMINISTERED BY THE UNIVERSITY OF KANSAS IN COLLABORATION WITH KANSAS STATE UNIVERSITY, WICHITA STATE UNIVERSITY, BAKER UNIVERSITY, DONNELLY COLLEGE, AND SEVEN KANSAS COMMUNITY COLLEGES. THE ADAPTIVE AND RESILIENT INFRASTRUCTURES DRIVEN BY SOCIAL EQUITY (ARISE) PROJECT IS FRAMED AROUND FOUR RESEARCH THEMES. THEME 1 FOCUSES ON UNDERSTANDING EQUITY MEASUREMENTS AND INTERDEPENDENT INFRASTRUCTURE SYSTEMS; THEME 2 DEVELOPS AN EVALUATION FRAMEWORK; THEME 3 CONSIDERS FOUR CASE STUDIES FOR HYPOTHESIS TESTING; AND THEME 4 INTEGRATES RESEARCH INTO DECISION-LEVER CASE STUDIES AND DECISION SUPPORT TOOLS. ARISE WILL BUILD ACADEMIC RESEARCH CAPACITY IN KANSAS FOR TECHNOLOGY-ENABLED ?SMART AND RESILIENT? INFRASTRUCTURE RESEARCH TO TRANSFORM RURAL AND VULNERABLE COMMUNITIES THAT ARE TYPICALLY UNDERREPRESENTED IN ?SMART CITIES? RESEARCH. THE PROJECT IS BASED ON THE RECOGNITION THAT DISASTER RECOVERY POLICIES AND PROGRAMS HAVE BEEN SYSTEMATICALLY BIASED AGAINST UNDERSERVED COMMUNITY MEMBERS SUCH AS THE HOMELESS, THE DISABLED, THOSE LIVING IN POVERTY, MINORITIES, AND ETHNIC AND INDIGENOUS POPULATIONS. THE MAJOR GAP THE PROJECT SEEKS TO ADDRESS IS THIS LACK OF ATTENTION TO EQUITY IN RESILIENCE FRAMEWORKS, POLICY, AND TOOLS. ARISE WILL EXAMINE THE RELATIONSHIP BETWEEN DISASTERS, RESOURCES, AND EQUITY AND WILL DEVELOP A STOCHASTIC HETERO-FUNCTIONAL, GRAPH-THEORETIC FRAMEWORK FOR UNDERSTANDING INTERDEPENDENT HUMAN AND PHYSICAL INFRASTRUCTURES. THE RESEARCH WILL APPLY A SIX-DIMENSIONAL APPROACH FOR MEASURING SOCIAL EQUITY (COMMUNITY CAPITALS). ARISE RESEARCH IS INTEGRATED WITH CAPACITY BUILDING AND OUTREACH ACTIVITIES INCLUDING INTERDISCIPLINARY SUMMER RESEARCH EXPERIENCES FOR UNDERGRADUATES, PARTNERSHIPS WITH MUSEUMS AND LIBRARIES IN UNDERSERVED COMMUNITIES TO PROMOTE YOUTH AND FAMILY STEM ENGAGEMENT, AND WORKFORCE DEVELOPMENT PROGRAMS FOR MUNICIPAL PLANNERS AND UTILITY OPERATORS. THE PROJECT WILL ENABLE RESEARCHERS AND EDUCATORS TO BE AT THE FOREFRONT OF THE RESILIENCE SCIENCE AND ENGINEERING ARENA, LEADING TO ENHANCED RESEARCH CAPACITY IN KANSAS. THIS AWARD REFLECTS NSF'S STATUTORY MISSION AND HAS BEEN DEEMED WORTHY OF SUPPORT THROUGH EVALUATION USING THE FOUNDATION'S INTELLECTUAL MERIT AND BROADER IMPACTS REVIEW CRITERIA.

MOST CURRENT REFRIGERANTS ARE HIGH-GLOBAL-WARMING-POTENTIAL (GWP) HYDROFLUOROCARBONS (HFCS) WITH UP TO 4000 TIMES THE IMPACT OF CO2. HIGH HVACR-ASSOCIATED ENERGY CONSUMPTION AND HFC LEAKS ACCOUNT FOR 7.8% OF TOTAL GREENHOUSE-GAS EMISSIONS. HFCS? CHEMICAL STABILITY, COMBINED WITH CHALLENGES ASSOCIATED WITH SEPARATING HFC REFRIGERANT BLENDS INTO COMPONENTS, MAKE SUSTAINABLE RECYCLING AND REPURPOSING OF THESE REFRIGERANTS DIFFICULT. IN ADDITION, TIGHTENING U.S. REGULATIONS HAVE CREATED A MARKET FOR ILLEGAL HFC IMPORTS. THE AMERICAN INNOVATION AND MANUFACTURING (AIM) ACT MANDATES AN 85% PHASEDOWN OF HFCS OVER THE NEXT TWO DECADES, BUT THESE CHALLENGES THREATEN THAT GOAL. EVEN IF THE TECHNICAL GOALS ARE MET, IMPLEMENTING SUCH SWEEPING CHANGES WILL REQUIRE SOCIETAL AND INDUSTRIAL ADOPTION AND A GREATLY EXPANDED U.S. HVACR WORKFORCE. THE ENVIRONMENTALLY APPLIED REFRIGERANT TECHNOLOGY HUB (EARTH) ENGINEERING RESEARCH CENTER (ERC) WILL USE A TEAM-SCIENCE APPROACH TO BRING TOGETHER TALENT IN ENGINEERING (CHEMICAL, ENVIRONMENTAL, MECHANICAL, AND MATERIALS), ARCHITECTURE, BUSINESS, CHEMISTRY, ECONOMICS, GEOGRAPHY, HISTORY, LAW, PSYCHOLOGY, AND ENTREPRENEURSHIP IN ONE INNOVATION ECOSYSTEM TO CO-CREATE CONVERGENT TECHNICAL AND SOCIETAL SOLUTIONS WITH INDUSTRY PARTNERS, TECHNICAL AND COMMUNITY COLLEGES, PROFESSIONAL ORGANIZATIONS, REGULATORS, AND END USERS. EARTH?S VISION IS TO CREATE A TRANSFORMATIVE ?SUSTAINABLE REFRIGERANT LIFECYCLE? TO ADDRESS THE HVACR ECOSYSTEM?S KEY TECHNICAL AND SOCIETAL CHALLENGES: (1) LOWERING HFC EMISSIONS, (2) CREATING SAFE, PROPERTY-BALANCED REPLACEMENT REFRIGERANTS, AND (3) INCREASING HVACR ENERGY EFFICIENCY. IN ADDITION, EARTH WILL WORK TOWARD WORKFORCE GOALS CO-CREATED WITH INDUSTRY TO INCREASE THE NUMBER OF HVACR ENGINEERING RESEARCHERS AND WILL INVOLVE COMMUNITY AND TECHNICAL COLLEGES TO ADDRESS WORKFORCE GAPS THROUGH COORDINATED OUTREACH AND TRAINING, WITH A FOCUS ON WOMEN, NATIVE PEOPLE, AND TRIBAL NATIONS. WITH STAKEHOLDER INPUT AND INTEGRATION ACROSS FUNDAMENTAL KNOWLEDGE, ENABLING TECHNOLOGIES, AND SYSTEM TESTBEDS, EARTH WILL CONVERGENTLY ADDRESS THE SOCIETAL PROBLEM OF REFRIGERANT ENVIRONMENTAL IMPACTS. REFRIGERANT LEAKING AND VENTING WILL BE ADDRESSED BY NEW SEPARATION, CONVERSION, SECURITY-TAGGING, AND WASTE-REFRIGERANT-REUSE TECHNOLOGIES, SPURRING SUSTAINABLE DECISION-MAKING AND NEW STARTUPS. NOVEL, SAFE, PROPERTY-BALANCED, LOW-FLAMMABILITY, LOW ENVIRONMENTAL-IMPACT REFRIGERANTS WILL BE EXPLORED WITH MOLECULAR SIMULATIONS OF CANDIDATE FLUIDS, DEVELOPMENT OF SOLID-STATE MATERIALS, REGULATORY-IMPACT ECONOMIC ANALYSIS, AND CORPORATE-INNOVATION INSIGHTS. HIGHER HVACR ENERGY EFFICIENCY WILL BE OBTAINABLE THROUGH NEW ENERGY-EFFICIENT DEHUMIDIFICATION MATERIALS, REFRIGERANT-SPECIFIC LEAK SENSORS, ALTERNATIVE REFRIGERATION CYCLES, SYSTEMS MODELING, LIFECYCLE ANALYSIS, TECHNOECONOMIC ANALYSIS, AND EXPLORATION OF CORPORATE, ENVIRONMENTAL, SOCIAL, AND GOVERNANCE ACTIVITIES. CROSSCUT THEMES - SYNTHESIS & CHARACTERIZATION, BEHAVIOR & POLICY, AND MODELING & ANALYSIS - WILL INTEGRATE RESEARCH WITH ALL ERC PILLARS. THE EPA ESTIMATES THE AIM ACT?S SUCCESSFUL IMPLEMENTATION FROM 2022 TO 2050 WILL NET BENEFITS OF $272 BILLION AND EMISSION REDUCTIONS OF 4.6 BILLION TONS OF CO2. IT WILL ALSO CREATE 150,000 NEW JOBS, INCREASE U.S. MANUFACTURING BY $39 BILLION, AND PREVENT A 0.5 ?C INCREASE IN GLOBAL TEMPERATURE. EARTH WILL ENABLE AND ACCELERATE THESE BENEFITS WITH NEW TECHNOLOGIES, DATA-INFORMED REGULATORY GUIDANCE, AND INDUSTRY AND STAKEHOLDER BUY-IN. BY ADDRESSING THE TECHNICAL, ENVIRONMENTAL, AND SOCIETAL CHALLENGES OF REPLACING HIGH-GWP REFRIGERANTS, EARTH?S RESEARCH WILL GENERATE SCIENTIFIC KNOWLEDGE, ENGINEERING PRODUCTS, ENVIRONMENTAL-POLICY RECOMMENDATIONS, AND INDUSTRY AND STAKEHOLDER BEHAVIORAL CHANGES TO STIMULATE THIS HVACR-ECOSYSTEM TRANSFORMATION. EARTH HAS IDENTIFIED OVER 70 INITIAL HVACR-ECOSYSTEM PARTNERS, INCLUDING COMPANIES, COMMUNITY COLLEGES AND TECHNICAL/TRADE SCHOOLS, NON-PROFIT ORGANIZATIONS, INTERNATIONAL UNIVERSITIES, AND FEDERAL LABS, TO AID WITH INDUSTRY ADOPTION OF NEW TECHNOLOGIES. EARTH WILL INCREASE THE HVACR INDUSTRY?S WORKFORCE CAPACITY ACROSS 6 CORE INSTITUTIONS (3 EPSCOR), IN PARTNERSHIP WITH NATIVE-HAWAI?IAN-SERVING AND TRIBAL COLLEGES/UNIVERSITIES, COMMUNITY COLLEGES, AND TECHNICAL/TRADE SCHOOLS. THIS AWARD REFLECTS NSF'S STATUTORY MISSION AND HAS BEEN DEEMED WORTHY OF SUPPORT THROUGH EVALUATION USING THE FOUNDATION'S INTELLECTUAL MERIT AND BROADER IMPACTS REVIEW CRITERIA.- SUBAWARDS ARE PLANNED FOR THIS AWARD.

LEVERAGING BIG DATA TO IMPROVE WOMEN'S HEALTH (BD-WH) - SUMMARY THROUGH THIS NEW NIH-FUNDED BIOMEDICAL CENTER AT THE UNIVERSITY OF KANSAS (KU), WE ASPIRE TO LEVERAGE BIG DATA TO IMPROVE WOMEN’S HEALTH. A STRONG AND DIVERSE INAUGURAL GROUP OF RESEARCH PROJECT LEADERS, SPANNING THE LABORATORY SCIENCES AND SOCIAL/BEHAVIORAL SCIENCES WOULD LEAD THE RESEARCH. THEIR GOALS INCLUDE: IMPROVING THE DETECTION AND TREATMENT OF OVARIAN CANCER; DEVELOPING A BETTER UNDERSTANDING OF ESTROGEN’S LINK TO NEURODEGENERATION; PROVIDING WOMEN, WHO ARE AT HIGHER RISK OF ALZHEIMER’S DISEASE, WITH TOOLS THEY CAN USE TO IMPROVE THEIR LIFESTYLE AND REDUCE DISEASE PROBABILITY; ADDITIONALLY, TWO DIFFERENT PROJECTS SEEK TO IMPROVE WOMEN’S HEALTH BY STUDYING LIFESTYLES, OVERALL HEALTH, AND THE IMPACT OF HEALTH-IMPROVING INNOVATIONS (I.E TELEMEDICINE) IN A GEOGRAPHIC AND SEX-DIFFERENTIATED CONTEXT SO THAT LIMITED PUBLIC HEALTH FUNDS CAN BE LEVERAGED FOR MAXIMAL PUBLIC GOOD. THE CENTER WOULD ENABLE PROGRESS ON THESE PROJECTS AND OTHERS AT THE INTERFACE OF DATA SCIENCE AND WOMEN’S HEALTH BY PROVIDING A STRONG ADMINISTRATIVE STRUCTURE ORGANIZED AROUND THE FOLLOWING SPECIFIC AIMS: 1) ADVANCE FIVE RPLS’ RESEARCH PROGRAMS (THROUGH MENTORING, GRANT-WRITING SUPPORT, AND PROFESSIONAL DEVELOPMENT) 2) ENGAGE ADDITIONAL RESEARCH FACULTY (THROUGH THREE NEW TENURE-TRACK HIRES AND A UNIQUE RESEARCH-ENGAGED FACULTY FELLOWS PROGRAM); 3)BOLSTER BIG DATA RESEARCH WITH CORE LAB SUPPORT (THROUGH TWO INITIATIVES, THE DEVELOPMENT OF A NEW CORE LAB FOCUSED ON BIOMEDICAL DATASETS AND SERVICES, AND A VOUCHER PROGRAM, SUPPORTED BY UNIVERSITY MATCHING FUNDS, THAT LEVERAGES KU’S 17 EXISTING CENTRALIZED RESEARCH CORE LAB FACILITIES.) THE WELL-PLANNED THEME, EXCELLENT INAUGURAL PROJECT LEADERS, CLOSE COORDINATION WITH UNIVERSITY ADMINISTRATION, AND EXPERIENCED CENTER DIRECTOR AND ASSOCIATE-DIRECTOR ALL LEND STRONG CREDIBILITY THAT THE GOALS WOULD BE ACHIEVED. MOLECULAR ANALYSIS OF DISEASE PATHWAYS - PROJECT SUMMARY – OVERALL THE COBRE CMADP WAS CREATED IN JULY 2012 TO ENCOURAGE BASIC RESEARCH SCIENTISTS TO DEVELOP AND IMPLEMENT NEW ENABLING TECHNOLOGIES FOR THE STUDY OF BIOLOGICAL PATHWAYS AND APPLIED PROCESSES RELATED TO DISEASE. OVER THE PAST NINE YEARS, CMADP INVESTIGATORS HAVE PURSUED HEALTH-RELATED RESEARCH RELATED TO THE MOLECULAR ANALYSIS OF DISEASE PATHWAYS. WE HAVE SUPPORTED 24 JUNIOR FACULTY TO DATE THROUGH RESEARCH SUPPORT AND MENTORING. THE 18 FACULTY THAT HAVE GRADUATED FROM THE PROGRAM THUS FAR HAVE GENERATED OVER $38.5 MILLION IN EXTERNAL FUNDING. WE ALSO ESTABLISHED THREE NEW CORE LABORATORIES FOCUSED ON ENABLING TECHNOLOGIES FOR THE STUDY OF DISEASE: THE GENOME SEQUENCING CORE, THE SYNTHETIC CHEMICAL BIOLOGY CORE AND THE KU NANOFABRICATION FACILITY (FORMERLY THE MICROFABRICATION & MICROFLUIDICS CORE). THESE THREE CORES BROUGHT NEW AND ESSENTIAL ENABLING TECHNOLOGIES TO KU, ALLOWING US TO ATTRACT HIGH-QUALITY BIOMEDICAL RESEARCHERS AND FACULTY TO KANSAS. AS A RESULT, CMADP STRENGTHENED BOTH THE HUMAN AND THE PHYSICAL RESOURCES FOR BIOMEDICAL RESEARCH IN THE STATE. THIS APPLICATION REQUESTS FIVE YEARS OF FUNDING FOR PHASE 3 OF OUR COBRE CENTER TO PURSUE THE FOLLOWING OBJECTIVES: (1) TO CONTINUE GROWING A CRITICAL MASS OF NEW AND CONTINUING INVESTIGATORS FOCUSED ON THE VERY BROAD THEME OF MOLECULAR ANALYSIS OF DISEASE PATHWAYS, (2) TO CONTINUE A SUCCESSFUL PROGRAM OF COBRE PILOT PROJECT GRANTS THAT UTILIZE OUR CORE LABS, AND (3) TO STRENGTHEN OUR EXISTING CORE LABS BY EXPANDING THEIR CAPABILITIES AND USER BASES, WORKING WITH THE KU OFFICE OF RESEARCH TO POSITION THEM FOR LONG-TERM SUSTAINABILITY BASED ON A COMBINATION OF FEES GENERATED PLUS A BASE OF SUPPORT FROM THE UNIVERSITY. THE RESULT WILL BE A VERY STRONG CADRE OF SUCCESSFUL MID-CAREER BIOMEDICAL RESEARCH FACULTY AND SELF-SUSTAINING CORE LABS THAT SUPPORT RESEARCH IN MOLECULAR ANALYSIS OF DISEASE PATHWAYS STATEWIDE. WE STRONGLY BELIEVE THAT THE OVERALL IMPACT OF CMADP ON BIOMEDICAL RESEARCH AT KU AND THE STATE OF KANSAS WILL THUS BE SUBSTANTIAL AND ENDURING.

RESHAPING APOE4 AND ALZHEIMER'S BRAINS WITH APOE2 - PROJECT SUMMARY THE RESEARCH WE PROPOSE IN THIS APPLICATION SEEKS TO TRANSLATE OUR RECENT UNDERSTANDING OF THE NEUROPROTECTIVE MECHANISM ASSOCIATED WITH THE HUMAN APOLIPOPROTEIN E2 (APOE2) GENOTYPE INTO A THERAPEUTIC OPPORTUNITY TO PREVENT AND TREAT ALZHEIMER'S DISEASE (AD). THE OVERARCHING HYPOTHESES ARE THAT INTRODUCTION OF HUMAN APOE2 PROTEIN INTO HIGH-RISK APOE4 AND AD BRAINS POSITIVELY ALTERS THE COURSE OF BRAIN AGING OR DISEASE PATHOGENESIS BY BOLSTERING BRAIN RESILIENCE THROUGH ENHANCED GLYCOLYTIC METABOLISM, WHICH SUBSEQUENTLY IMPROVES GLUCOSE UTILIZATION, PROTEIN HOMEOSTASIS, AND SYNAPTIC ACTIVITY. IN PREPARATION, WE HAVE ACHIEVED THREE MAJOR MILESTONES CRITICAL TO THE SUCCESS OF THIS TRANSLATIONAL ENDEAVOR: (1) DEVELOPMENT OF A METHOD FOR THE PRODUCTION OF PHYSIOLOGICALLY RELEVANT AND HUMAN-COMPATIBLE RECOMBINANT APOE2 (RHAPOE2) GLYCOPROTEIN THAT POSSESSES BIOLOGICAL FUNCTIONALITY COMPARABLE TO ENDOGENOUS HUMAN APOE2; (2) DEVELOPMENT OF A NONINVASIVE APPROACH FOR BRAIN DELIVERY OF RHAPOE2 GLYCOPROTEIN VIA MODULATION OF CADHERIN INTERACTIONS ON THE BLOOD-BRAIN BARRIER (BBB), WHICH INDUCES NEUROPROTECTIVE SIGNALING IN APOE4 BRAINS; AND (3) DEVELOPMENT OF NOVEL HUMANIZED KNOCK-IN MOUSE MODELS THAT RESPECTIVELY TARGET HUMAN SPORADIC (SAD) AND FAMILIAL AD (FAD), WHICH ARE EXPECTED TO PROVIDE HIGH PREDICTIVE VALIDITY FOR TRANSLATING BENCH SUCCESSES TO BEDSIDE. THE PROPOSED STUDIES WILL PURSUE THREE SPECIFIC AIMS. BUILDING ON OUR INITIAL SUCCESS, THE OBJECTIVE OF THE FIRST AIM IS TO DETERMINE THE THERAPEUTICALLY OPTIMAL REGIMEN FOR DELIVERY OF RHAPOE2 GLYCOPROTEIN THAT WILL RESULT IN DEPOSITION OF RHAPOE2 THROUGHOUT CORTICAL AND HIPPOCAMPAL REGIONS AND UPREGULATION OF BRAIN GLYCOLYTIC METABOLISM WITHOUT ELICITING ADVERSE REACTIONS. THE OBJECTIVE OF THE SECOND AIM IS TO EVALUATE THE THERAPEUTIC IMPACT OF RHAPOE2 DELIVERY, IN COMBINATION WITH AGE AND SEX, ON BRAIN CHANGES ASSOCIATED WITH SAD IN HUMANIZED MOUSE MODELS THAT EXPRESS PHYSIOLOGICAL LEVELS OF HUMAN APOE3 OR APOE4 AND HUMAN WILD-TYPE APP PROTEINS. THE THIRD AIM WILL BE INVESTIGATED IN HUMANIZED MOUSE MODELS THAT EXPRESS PHYSIOLOGICAL LEVELS OF HUMAN APOE3 OR APOE4 AND HUMAN MUTANT APP PROTEINS TO EVALUATE THE THERAPEUTIC IMPACT OF RHAPOE2 DELIVERY ON BRAIN CHANGES ASSOCIATED WITH FAD, AND HOW THE RHAPOE2-MEDIATED EFFECTS ARE MODIFIED BY A COMBINATION OF AGE, SEX, APOE GENOTYPE, AND DISEASE STATUS. OUR OVERALL GOALS FOR THE PROPOSED RESEARCH ARE TO ESTABLISH THE PLAUSIBILITY OF TARGETING BRAIN METABOLIC RESILIENCE AS A DISEASE-MODIFYING STRATEGY AND GENERATE PROOF OF CONCEPT AS TO WHETHER A RHAPOE2-BASED PROTEIN THERAPY CAN POTENTIALLY BE DEVELOPED INTO AN EFFECTIVE AND SAFE INTERVENTION FOR AD.

THIS SETS OFF A VICIOUS CYCLE INVOLVING SYNAPSES, NEURAL ACTIVITY, AND MITOCHONDRIA, WHICH MAY DIFFER DEPENDING ON AGE AND BRAIN REGION. THIS PROPOSAL SEEKS TO INVESTIGATE THE INTERCONNECTEDNESS OF THESE FACTORS IN AN AGE- DEPENDENT MANNER ACROSS MULTIPLE BRAIN REGIONS. BY UTILIZING MOUSE MODELS OF ALZHEIMER'S DISEASE, THE STUDY AIMS TO DETERMINE WHETHER MODULATION OF NEURAL ACTIVITY, SYNAPSES, OR MITOCHONDRIA COULD ENHANCE NEURAL PLASTICITY IN AMYLOIDOSIS. THROUGH A COMBINATION OF IN VIVO AND EX VIVO APPROACHES, THE PROPOSAL AIMS TO UNRAVEL THE RELATIONSHIP BETWEEN NEURAL ACTIVITY AND SYNAPTIC ABNORMALITIES AND TEST THE EFFICACY OF TARGETING CYCLOPHILIN D, A COMPONENT OF THE MITOCHONDRIAL MEMBRANE PERMEABILITY TRANSITION PORE, IN IMPROVING MITOCHONDRIAL HEALTH AND REVERSING SYNAPTIC AND NEURAL ACTIVITY DEFICITS.

RAMP: MAKING MEANINGFUL CONNECTIONS: FOSTERING THE INTEGRATION OF BIODIVERSITY PATTERNS WITH GENETIC EVOLUTIONARY MECHANISMS (BIOGEM) -GIVEN THE SEVERITY AND COMPLEXITY THAT A RAPIDLY CHANGING CLIMATE IMPOSES ON ALL LIFE ON EARTH, A MULTI-FACETED APPROACH TO ADDRESSING THE BIODIVERSITY CRISIS IS NEEDED. THIS PROJECT BRINGS TOGETHER A GROUP OF MENTORS WITH DIVERSE EXPERTISE, RANGING FROM GENOMICS TO PALEONTOLOGY, IN ORDER TO TRAIN POSTBACCALAUREATE SCHOLARS IN INTEGRATIVE AND INTERDISCIPLINARY ORGANISMAL BIOLOGY RESEARCH. SCHOLARS AND THEIR MENTORS WILL ENGAGE IN RESEARCH THAT SPANS TIME SCALES, BIOLOGICAL HIERARCHIES, AND ORGANISMS ACROSS THE TREE OF LIFE, TO ADDRESS SPECIFIC QUESTIONS REGARDING PAST, PRESENT, AND FUTURE ORGANISMAL RESPONSES TO A CHANGING ENVIRONMENT. THREE COHORTS OF TEN POSTBACCALAUREATE SCHOLARS EACH WILL BE RECRUITED TO BROADEN PARTICIPATION IN THE SCIENCES. NETWORK PARTNERS INCLUDE A LOCAL TRIBAL COLLEGE, REGIONAL PRIMARILY UNDERGRADUATE INSTITUTIONS, A BIOMEDICAL RESEARCH INSTITUTE, AND A NON-PROFIT ORGANIZATION THAT INTERFACES WITH INDUSTRY. IN BRINGING TOGETHER A DIVERSE GROUP OF MENTORS, NETWORK PARTNERS, AND POSTBACCALAUREATE SCHOLARS TO ENGAGE IN INTERDISCIPLINARY RESEARCH, THIS PROJECT WILL PROVIDE EXTENSIVE TRAINING THAT DIVERSIFIES THE WORKFORCE IN INTEGRATIVE ORGANISMAL BIOLOGY, WHILE LEADING TO IMPORTANT DISCOVERIES THAT HELP BETTER UNDERSTAND AND PREDICT BIOLOGICAL RESPONSES TO A CHANGING CLIMATE. THIS PROGRAM CENTERS ON INTEGRATING BIODIVERSITY SCIENCE AND GENETIC EVOLUTIONARY MECHANISMS (BIOGEM) TO INVESTIGATE BIOLOGICAL RESPONSES TO CLIMATE CHANGE. BIOGEM POSTBACCALAUREATE SCHOLARS WILL BE PRIMARILY BASED AT THE UNIVERSITY OF KANSAS (KU) AND UTILIZE KU?S WORLD-CLASS FACILITIES AND MUSEUM COLLECTIONS TO CONDUCT RESEARCH THAT INCORPORATES THE NETWORK?S EXPERTISE IN BIODIVERSITY SCIENCE (PHYLOGENETICS, PALEONTOLOGY, MORPHOMETRICS, BIOGEOGRAPHY, AND COMMUNITY ECOLOGY) AND GENETIC EVOLUTIONARY MECHANISMS (COMPARATIVE AND FUNCTIONAL GENOMICS, DEVELOPMENTAL BIOLOGY, AND QUANTITATIVE GENETICS). SPECIFICALLY, STUDYING SPECIATION/EXTINCTION/PERSISTENCE, PALEOBIOGEOGRAPHY, AND MORPHOLOGICAL DISPARITY AND INNOVATION WILL ALLOW US TO ADDRESS HOW ORGANISMS, SPECIES, AND CLADES WERE SHAPED BY THEIR PAST ENVIRONMENT; STUDYING POPULATION DYNAMICS, ECOLOGICAL INTERACTIONS, AND BIOGEOGRAPHIC PATTERNS WILL ALLOW US TO ADDRESS HOW ORGANISMS, POPULATIONS, AND SPECIES RESPOND TO THEIR PRESENT BIOTIC AND ABIOTIC ENVIRONMENT; AND STUDYING PLASTICITY, GENOMIC RESPONSES TO ADAPTATION, AND MECHANISMS OF GENETIC DIVERSITY WILL ALLOW US TO ADDRESS HOW GENETIC MECHANISMS EQUIP ORGANISMS TO RESPOND TO FUTURE ENVIRONMENTAL CHANGE. A KEY COMPONENT OF THE BIOGEM PROGRAM EXPERIENCE IS A WEEKLY PROFESSIONAL & SCHOLARLY ADVANCEMENT SERIES, INVOLVING THE MENTORS AND NETWORK PARTNERS, THAT WILL INCLUDE BOTH PROFESSIONAL DEVELOPMENT WORKSHOPS AND HANDS-ON MENTOR-LED TRAINING MODULES COVERING A WIDE RANGE OF RESEARCH METHODS. THE BIOGEM PROGRAM IS GEARED TOWARDS RECENT GRADUATES THAT HAVE HAD LIMITED OR NO PRIOR RESEARCH EXPERIENCE. THIS AWARD REFLECTS NSF'S STATUTORY MISSION AND HAS BEEN DEEMED WORTHY OF SUPPORT THROUGH EVALUATION USING THE FOUNDATION'S INTELLECTUAL MERIT AND BROADER IMPACTS REVIEW CRITERIA.

EXAMINING ADAPTIVE PEER-MEDIATED INTERVENTIONS FOR PRESCHOOLERS WITH AUTISM SPECTRUM DISORDER AND LIMITED OR NO SPOKEN LANGUAGE: A SEQUENTIAL MULTIPLE ASSIGNMENT RANDOMIZED TRIAL - PROJECT SUMMARY IMPAIRED SOCIAL COMMUNICATION IS A CORE FEATURE OF AUTISM SPECTRUM DISORDER (ASD). THIS PROPOSAL WILL EVALUATE A SERIES OF PEER-MEDIATED INTERVENTIONS FOR PRESCHOOL-AGE CHILDREN (3 TO 6 YEARS) WITH ASD AND LIMITED OR NO SPOKEN LANGUAGE, USING AN INNOVATIVE SEQUENTIAL MULTIPLE ASSIGNMENT RANDOMIZED TRIAL (SMART) DESIGN. AVAILABLE EVIDENCE SUPPORTS THE BENEFICIAL EFFECTS OF PEER-MEDIATED INTERVENTIONS FOR IMPROVING SOCIAL COMMUNICATION SKILLS IN CHILDREN WITH ASD.1, 2 PEER-RELATED SOCIAL COMPETENCE IS VITAL TO A WIDE RANGE OF CHILD OUTCOMES.3 CHILDREN WHO ACHIEVE POSITIVE PEER INTERACTIONS IN PRESCHOOL SHOW IMPROVED SOCIAL COMMUNICATION AND FEWER BEHAVIORAL PROBLEMS IN ELEMENTARY SCHOOL.4 IN SPITE OF CONSIDERABLE EARLY INTERVENTION EFFORTS, APPROXIMATELY 30% OF CHILDREN WITH ASD REMAIN MINIMALLY-VERBAL IN KINDERGARTEN5, RESTRICTING PARTICIPATION IN INCLUSIVE ACTIVITIES. RECENT STUDIES REPORT IMPROVED COMMUNICATION BETWEEN CHILDREN WITH ASD AND PEERS AFTER A SPEECH-GENERATING DEVICE (SGD) IS INCLUDED IN TREATMENT.6-9 A REPERTOIRE OF EFFECTIVE INTERVENTIONS THAT CAN BE APPLIED AND MODIFIED IS NECESSARY TO ENSURE OPTIMAL SOCIAL COMMUNICATION OUTCOMES WHEN CHILDREN DO NOT MAKE ANTICIPATED PROGRESS. A STRENGTH OF THE STUDY IS THAT THESE INTERVENTIONS CAN BE ADOPTED BY COMMUNITY- BASED, EARLY SERVICE PROVIDERS. ALL PARTICIPANTS WILL RECEIVE AN ADAPTED STAY-PLAY-TALK (SPT)7, 10-12 PEER- MEDIATED INTERVENTION THAT VARIES IN ACTIVE INGREDIENTS EXAMINED BY THE PI IN PRELIMINARY STUDIES. NOW, WITH SMART DESIGNS, IT IS POSSIBLE TO TEST AND IDENTIFY ALTERNATIVE COMBINATIONS OF PEER-MEDIATED APPROACHES, SUCH AS THE ADDITION OF A SGD TO IMPROVE SOCIAL COMMUNICATION OF CHILDREN WITH ASD AND COMPLEX COMMUNICATION NEEDS.7, 8, 13 IN THE CURRENT PROPOSAL, 132 PRESCHOOLERS WITH ASD (AND N=264 PEERS WITHOUT DISABILITIES) WILL BE INITIALLY RANDOMIZED TO SPT AND SGD WITH SPOKEN PEER INPUT ONLY (SPT BASIC; PEERS TAUGHT TO MODEL LANGUAGE) OR SPT AND SGD WITH AUGMENTED PEER INPUT IN WHICH PEERS ARE TAUGHT TO MODEL VERBAL RESPONSES WHILE SIMULTANEOUSLY SELECTING SCREEN ICONS (SPT PLUS; PEERS TAUGHT TO USE VERBAL LANGUAGE MODELS CONCURRENTLY WITH THE SGD). EACH CHILD’S RESPONSE TO TREATMENT AFTER 5 WEEKS WILL DETERMINE THAT CHILD’S NEXT PHASE IN THE SMART DESIGN. CHILDREN SHOWING A POSITIVE RESPONSE WILL CONTINUE IN THEIR ORIGINALLY ASSIGNED GROUP. CHILDREN WHO ARE SLOW RESPONDERS AFTER 5 WEEKS WILL BE RANDOMLY ASSIGNED TO RECEIVE ADDED TREATMENT COMPONENTS TO IMPROVE PEER-DIRECTED COMMUNICATION (EITHER SPT PLUS OR SPT ADVANCED). SPT ADVANCED ADDS DIRECT INSTRUCTION STRATEGIES FOR THE CHILDREN WITH ASD (I.E., ADULT PROMPTS, REINFORCERS, AND TEACHING TRIALS) SHOWN TO INCREASE CHILD VOCALIZATIONS IN SGD INTERVENTIONS.9, 14 THE USE OF A SMART DESIGN EXTENDS OUR PRIOR WORK BY TESTING THE SYSTEMATIC ADDITION OF SELECTED PEER-MEDIATED STRATEGIES IN COMBINATION WITH AN SGD8 THAT ALLOWS FOR FLEXIBLE APPLICATION OF INTERVENTIONS BASED ON CHILD RESPONSE MEASURED AT IMPORTANT NODAL POINTS.15-17 WE HAVE ASSEMBLED AN OUTSTANDING TEAM OF HIGHLY QUALIFIED INVESTIGATORS WITH COMPLEMENTARY SKILLS IN PRESCHOOL ASSESSMENT, LANGUAGE INTERVENTION, CLINICAL TRIALS, AND STATISTICS.

NEUROCIRCUIT OF PARTNER-SEEKING FOLLOWING SOCIAL LOSS - PROJECT SUMMARY/ABSTRACT THE DEATH OF A LOVED ONE IS MONUMENTALLY PAINFUL, IMPACTING 8 MILLION IN THE US ANNUALLY, AND IS A SIGNIFICANT CAUSE OF PSYCHIATRIC AND MEDICAL MORBIDITY, INCLUDING PSYCHIATRIC SEQUELAE SUCH AS DEPRESSION, ANXIETY, SUBSTANCE ABUSE, AND COMPLICATED GRIEF. RESEARCH EXPLORING THE ASSOCIATION BETWEEN SOCIAL LOSS AND BRAIN FUNCTION ARE SPARSE, AND DIAGNOSTIC CRITERIA FOR THE EMERGING DISORDERS ARE BASED ON SYMPTOM CLUSTERS RATHER THAN NEURAL OR GENETIC ABNORMALITIES, HINDERING PURSUITS OF NEW THERAPEUTIC TARGETS. THIS HIGHLIGHTS THE NEED FOR PROPER ANIMAL MODELS FOR SOCIAL LOSS TO MIRROR THE BEHAVIORAL SYMPTOMOLOGY TO EXPLORE THE NEUROBIOLOGICAL MECHANISMS. RECENTLY, WE HAVE DEVELOPED AN ANIMAL MODEL OF SOCIAL LOSS USING THE SOCIALLY MONOGAMOUS PRAIRIE VOLE (MICROTUS OCHROGASTER), WHICH MANIFEST SIMILAR BEHAVIORAL SYMPTOMOLOGY AND DISRUPTION TO NORMAL MESOLIMBIC REWARD PATHWAY THAT OCCURS IN SOCIAL LOSS IN HUMANS. OUR LONG-TERM GOALS ARE TO DISSECT THE NEUROCIRCUITRY OF SOCIAL LOSS (INCLUDING THE MESOLIMBIC REWARD AND CORTICOLIMBIC PAIN PATHWAYS), DEFINE GENETIC BIOMARKERS OF INDIVIDUAL VULNERABILITY AND RESILIENCY TO SOCIAL LOSS, AND DEVELOP THERAPEUTIC INTERVENTION OF NEURAL AND BEHAVIORAL SYMPTOMS THAT APPEAR PERSISTENT. OUR THEORETICAL NEUROCIRCUIT OF SOCIAL LOSS CENTERS IN THE LIMBIC SYSTEM, WHICH RECEIVES DOPAMINE (DA) SIGNALING FROM THE VENTRAL TEGMENTAL AREA (VTA) AND AUGMENTS MOTIVATIONAL STATE AND EMOTIONAL VALENCE. OUR SPECIFIC AIMS WILL TEST THE FOLLOWING HYPOTHESES: (AIM 1) LOSS OF SPECIFIC RELATIONSHIP TYPES CAN PROPAGATE PERSISTENT CHANGE TO THE FUNCTIONAL AND STRUCTURAL CONNECTIVITY OF MESOLIMBIC CIRCUITS; (AIM 2) BEHAVIORAL SYMPTOMS OF LONG-TERM SOCIAL LOSS ARE BASED ON DISRUPTION TO DA NEUROTRANSMISSION AND CAN BE ALLEVIATED WITH PHOTOSTIMULATION AND CHEMICAL INNERVATIONS ALONG SELECT MESOLIMBIC PATHWAYS; (AIM 3) CHANGES TO THE EXPRESSION AND ACTIVITY OF SPECIFIC DA RECEPTORS IN MESOLIMBIC REGIONS ARE COUPLED TO FLUCTUATING MOTIVATIONAL STATES DURING LOSS AND SEEKING OF PARTNER-ASSOCIATED CUES. THIS CONTRIBUTION IS SIGNIFICANT SINCE IT WILL ESTABLISH THAT SEVERAL PATHWAYS TARGETED BY PHOTOSTIMULATION AND PHARMACOLOGICAL APPROACHES HAVE THE POTENTIAL TO REGULATE BEHAVIORAL SYMPTOMOLOGY OF SOCIAL LOSS THROUGH MESOLIMBIC ACTIVITY. THE PROPOSED RESEARCH IS INNOVATIVE AS IT IS THE FIRST SYSTEMATIC EVALUATION OF NEUROMODULATION OF THE MESOLIMBIC SYSTEM DURING PARTNER LOSS IN AN ANIMAL MODEL. INSIGHT INTO THE NEUROMODULATION OF SOCIAL LOSS IS IMPACTFUL AS IT MAY EXPAND DIAGNOSTIC CRITERIA FOR THE EMERGING MENTAL HEALTH DISORDERS BEYOND SYMPTOM CLUSTERS AND IDENTIFIES NEUROCHEMICAL SUBSTRATES FOR NEW THERAPEUTIC TARGETS.

PSYCHOMETRICS AND PREDICTIVE VALIDITY OF INFANT LEARNING - PROJECT SUMMARY THE ABILITY TO DETECT SENSORIMOTOR CONTINGENCIES BETWEEN AN ORGANISM'S ACTIONS AND CHANGES IN THE ENVIRONMENT IS ESSENTIAL TO SURVIVAL, IMPACTING SOCIAL, MOTOR, AFFECTIVE, AND COGNITIVE DOMAINS. OPERANT CONDITIONING PARADIGMS PROVIDE A DIRECT MEASURE OF CONTINGENCY LEARNING, AND ARE AMENABLE FOR USE THROUGHOUT INFANCY. THE PRIMARY GOAL OF THIS RESEARCH IS TO DETERMINE WHETHER INDIVIDUAL VARIATIONS IN CONTINGENCY LEARNING ARE CONSISTENT OR CORRELATED ACROSS TASKS AND/OR ACROSS DEVELOPMENT, AND TO DETERMINE WHETHER THESE MEASURES PREDICT FUTURE STANDARDIZED MEASURES OF COGNITION. TO THIS END, WE WILL LONGITUDINALLY TEST INFANTS FROM 6-18 MONTHS USING TWO ESTABLISHED INFANT OPERANT CONDITIONING TASKS—THE SYNCHRONOUS REINFORCEMENT PARADIGM AND TRAIN TASK—EACH INVOLVING A DIFFERENT RESPONSE SYSTEM (LOOKING VS. LEVER PRESSING) AND REINFORCER. DESPITE THE FACT THAT LEARNING IS UNIVERSALLY REGARDED AS A FUNDAMENTAL COMPONENT OF HUMAN BEHAVIOR, AFTER OVER A CENTURY OF BEHAVIORAL SCIENCE, WE SIMPLY DO NOT KNOW WHETHER OR TO WHAT DEGREE INDIVIDUAL DIFFERENCES IN MEASURES OF LEARNING IN INFANCY ARE CONSISTENT PSYCHOMETRICALLY, OR WHETHER THEY ARE DIRECTLY ASSOCIATED WITH BROADER COGNITIVE AND LANGUAGE OUTCOMES. THERE IS SUBSTANTIAL DEMAND FOR THE INCLUSION OF MEASURES OF LEARNING IN CLINICAL TRIALS AS POTENTIAL OUTCOMES FOR EARLY INTERVENTIONS, BUT THESE FUNDAMENTAL ISSUES HAVE NEVER BEEN PREVIOUSLY ESTABLISHED OR INVESTIGATED IN INFANCY. THIS PROPOSAL FEATURES TWO INVESTIGATORS WITH DIRECT EXPERTISE IN OPERANT LEARNING PARADIGMS FOR INFANTS AND IN CONDUCTING LONGITUDINAL PROJECTS; HERE, THEY SEEK TO ESTABLISH THE FUNDAMENTAL PSYCHOMETRIC PROPERTIES OF INDIVIDUAL DIFFERENCES IN CLASSIC PARAMETERS OF OPERANT CONDITIONING WITHIN AGES ACROSS TASKS AND ACROSS AGES, AND TO ESTABLISH THE DEVELOPMENTAL COURSE OF THESE PARAMETERS. WE ALSO SEEK TO DETERMINE THE PREDICTIVE VALIDITY OF THESE INDIVIDUAL DIFFERENCES, WHETHER THE INDICATORS THEMSELVES OR THEIR DEVELOPMENTAL FUNCTIONS ARE PREDICTIVE OF STANDARDIZED 24-MONTH OUTCOMES. WE PROPOSE TO USE TWO PARADIGMS THAT UTILIZE DIFFERENT OPERANTS (LOOKING AND MOTOR ACTIONS) EACH OF WHICH HAVE PREVIOUSLY BEEN ESTABLISHED AS VALID AND WORKABLE IN PUBLISHED LITERATURE. LEARNING WITHIN TRADITIONAL CONDITIONING PARADIGMS HAS LONG BEEN NEGLECTED IN THE AREA OF HUMAN INFANCY; THIS STUDY HAS THE POTENTIAL TO RESURRECT LEARNING IN INFANT STUDIES AND DEVELOPMENTAL SCIENCE. GIVEN THAT DIFFERENCES IN CONTINGENCY LEARNING HAVE BEEN NOTED IN AT-RISK INFANT SAMPLES AND LEARNING IS OFTEN USED IN PRECLINICAL ANIMAL WORK, THIS WORK WOULD SERVE TO PROVIDE A CONNECTION BETWEEN PRECLINICAL STUDIES AND CLINICAL TRIALS. OUR FINDINGS WILL HAVE CRITICAL IMPLICATIONS FOR THE USE OF LEARNING IN CLINICAL TRIALS AND LONG-TERM OUTCOME STUDIES.

WHILE MANAGEMENT OF PESTS IN ANNUAL CROPS EMPHASIZES CONTINUED DISRUPTION OF PATHOGEN POPULATIONS, SUCH STRATEGIES HAVE LIMITED VALUE IN PERENNIAL PRODUCTION SYSTEMS. IT IS QUITE LIKELY THAT MORE DURABLE RESISTANCE TO PATHOGENS CAN BE GENERATED BY MIMICKING NATURE AND UTILIZING MIXED STRATEGIES THAT INCLUDE BREEDING FOR RESISTANCE AND PLANTING PERENNIAL CROPS IN MULTISPECIES POLYCULTURES. WE EVALUATE THIS POSSIBILITYAND APPLY THIS FRAMEWORK TO AN EMERGING PERENNIAL OILSEED CROP, THEREBY ILLUSTRATING POTENTIAL RELEVANCE TO OTHER PERENNIAL PRODUCTION SYSTEMS.WE HAVE FOUND THAT PATHOGEN IMPACTS DECLINE WITH SPECIES DIVERSITY IN PLANT COMMUNITIES, AND THAT THIS PATHOGEN DILUTION GENERATES SUBSTANTIAL INCREASES IN PRODUCTIVITY RELATIVE TO MONOCULTURE (I.E. OVERYIELDING). WE ALSO FOUND THAT PATHOGEN DILUTION OCCURS IN CONCERT WITH EVOLUTION OF GENETIC RESISTANCE TO DISEASE. OUR WORK TESTS THE JOINT IMPACTS OF THE GENETIC AND COMMUNITY DIVERSITY ON PLANT PATHOGEN SPREAD AND IMPACT BUT DEVELOPING AND TESTING A MATHEMATICAL FRAMEWORK THAT PREDICTS PATHOGEN DILUTION AND ITS IMPACTS ON PRODUCTIVITY. WE BUILD MORE SYNTHETIC UNDERSTANDINGOF THE ECOLOGICAL AND GENETIC CONTROLS OF PATHOGEN SPREAD, WHICH TOGETHERCAN ADVANCE PEST MANAGEMENT IN AGRICULTURE AND RANGELANDS. WE WILL ALSO TRAIN STUDENTS AND HIGH SCHOOL TEACHERS IN AGROECOLOGY, PLANT PATHOLOGY AND DISEASE ECOLOGY.

PARENT IMPLEMENTED TREATMENT FOR REPETITIVE BEHAVIORS IN CHILDREN WITH ASD: USING A NOVEL TELEHEALTH APPROACH TO INCREASE SERVICE ACCESS - PROJECT SUMMARY FAMILIES ARE FACING SIGNIFICANT CHALLENGES DURING THE COVID-19 PANDEMIC, ESPECIALLY THOSE CARING FOR A CHILD WITH AUTISM SPECTRUM DISORDER (ASD). A RECENT SPARK SURVEY1 (N = 8,000) FOUND THAT APPROXIMATELY 95% OF PARENTS REPORTED INCREASES IN THEIR CHILD’S CHALLENGING BEHAVIORS AS A RESULT OF COVID-19, AND THE RESTRICTED, REPETITIVE BEHAVIORS AND INTERESTS (RRBI) OF CHILDREN LIKELY FALL WITHIN THIS CATEGORY. SINCE ONLY 35% OF PARENTS WHO TOOK THE SPARK SURVEY INDICATED THEIR CHILD IS RECEIVING THERAPY SERVICES REMOTELY, THE COVID-19 PANDEMIC WILL LIKELY EXACERBATE THIS CORE SYMPTOM OF ASD. RRBIS ARE OFTEN THE TARGET FOR CLINICAL TREATMENT, AS THEY CAN SIGNIFICANTLY INTERFERE WITH LEARNING, AND ARE ASSOCIATED WITH LONG-TERM FUNCTIONAL IMPAIRMENT, AND PSYCHIATRIC COMORBIDITIES (E.G., ANXIETY). GIVEN CHILDREN WITH ASD ARE CURRENTLY OUT OF ROUTINE AND EXPERIENCING A SEVERE REDUCTION IN THERAPEUTIC AND EDUCATIONAL SERVICES, THE RRBIS AND COMORBIDITIES OF CHILDREN ARE LIKELY MAGNIFIED AT THIS TIME. YET, THERE ARE NO EFFECTIVE THERAPIES TO TREAT THE FULL RANGE OF REPETITIVE, INFLEXIBLE BEHAVIORS FOUND IN ASD, AND THERE HAVE BEEN NO LARGE-SCALE STUDIES OF BEHAVIORAL INTERVENTIONS SPECIFICALLY DEVELOPED TO TARGET THIS CORE SYMPTOM DOMAIN. THIS HAS RESULTED IN A SIGNIFICANT TREATMENT GAP BETWEEN THE NUMBER OF EFFECTIVE BEHAVIORAL THERAPIES USED TO TARGET ATYPICAL SOCIAL-COMMUNICATION BEHAVIORS IN AUTISM WHEN COMPARED TO THOSE DESIGNED TO TREAT REPETITIVE BEHAVIORS. THUS, OUR TEAM DEVELOPED AND PILOT-TESTED THE FAMILY-IMPLEMENTED TREATMENT FOR BEHAVIORAL INFLEXIBILITY (FITBI), DERIVED FROM NEUROBEHAVIORAL MODELS OF REWARD AND LEARNING. THE OVERALL GOAL OF FITBI IS TO TEACH PARENTS HOW TO IDENTIFY HIGH PROBABILITY CUES IN THE ENVIRONMENT THAT CAN ELICIT RRBI SYMPTOMS AND TEACH THEIR CHILD TO INHIBIT REPETITIVE BEHAVIORS AND INSTEAD REPLACE THEM WITH FLEXIBLE AND MORE FUNCTIONAL BEHAVIORS. SINCE OUR INITIAL PILOT STUDIES, THE NEED FOR TECHNOLOGY-BASED INTERVENTIONS HAS CERTAINLY INCREASED. WHILE THE EFFECTIVENESS OF TELEHEALTH INTERVENTIONS FOR CHILDREN WITH ASD IS EMERGING, EARLY RESEARCH SUGGESTS INTERVENTIONS DELIVERED VIA TELEHEALTH FOR CHILDREN WITH ASD MAY BE JUST AS EFFECTIVE AS IN-PERSON INTERVENTIONS. YET, WE CURRENTLY HAVE NO UNDERSTANDING OF THE EFFECTIVENESS OF RRBI INTERVENTIONS DELIVERED REMOTELY. THEREFORE, THE OVERALL GOAL OF THIS PROJECT IS TO CONDUCT A PARALLEL GROUP RANDOMIZED CONTROLLED TRIAL (RCT) COMPARING REMOTE DELIVERY OF FITBI (12-WEEK INTERVENTION + 3 BOOSTER SESSIONS OVER 6 MONTHS) TO REMOTE DELIVERY OF A PARENT EDUCATION ONLY (PE) CONTROL CONDITION IN A FINAL SAMPLE SIZE OF 100 (3-9 YEARS) CHILDREN WITH ASD AND HIGH RATES OF RITUALISTIC REPETITIVE BEHAVIORS. WE WILL USE TORSH, A COMPREHENSIVE SECURE ONLINE PLATFORM THAT ENHANCES THERAPIST-PARENT COACHING VIA TELEHEALTH. FURTHER, AN IMPORTANT OBJECTIVE OF THIS PROPOSAL IS TO EXAMINE CHILD AND PARENT FACTORS ASSOCIATED WITH TREATMENT RESPONSE AND UPTAKE IN ORDER TO ADVANCE TRANSLATIONAL RESEARCH AND KNOWLEDGE ON PERSONALIZED INTERVENTION APPROACHES.

A RESOURCE FOR THE GENETIC DISSECTION OF COMPLEX TRAITS - PROJECT SUMMARY A LARGE, DIVERSE SET OF COMMON HUMAN DISEASES, AND MOST OF THE BIOMEDICALLY-RELEVANT TRAITS THAT MODEL ORGANISM BIOLOGISTS ROUTINELY TARGET, ARE COMPLEX AND POLYGENIC. POPULATION VARIATION IN THESE TRAITS HAS A SIZEABLE GENETIC COMPONENT, AND DISSECTING THIS VARIATION CAN YIELD IMPROVED DIAGNOSTICS AND THERAPEUTICS, AND ENABLE DETAILED DESCRIPTIONS OF THE MOLECULAR AND CELLULAR PROCESSES UNDERLYING DISEASE AND TRAIT VARIATION. GENOMEWIDE ASSOCIATION STUDIES (GWAS) ARE LARGELY RESPONSIBLE FOR DRAMATIC PROGRESS IN THE ANALYSIS OF HUMAN COMPLEX TRAITS IN RECENT YEARS. GWAS HAVE LINKED MANY GENES TO VARIATION IN HUMAN HEALTH, AND IMPLY A SIGNIFICANT FRACTION OF TRAIT VARIATION IS CONTROLLED BY THOUSANDS OF TINY-EFFECT, PRIMARILY REGULATORY SITES SPREAD ALONG THE GENOME. DESPITE THIS CRITICAL INSIGHT, THERE REMAIN MANY GAPS IN OUR UNDERSTANDING OF THE NATURE OF CAUSATIVE LOCI. ONE NOTABLE CAVEAT OF THE GWAS APPROACH IS ITS INHERENT BIAS TOWARDS INTERMEDIATE- FREQUENCY ALLELES; GWAS ARE ILL-SUITED TO UNCOVERING RARE ALLELES OF LARGE EFFECT AND GENES HARBORING SEVERAL INDIVIDUALLY-RARE MUTATIONS, EVENTS THAT MUTATION-SELECTION BALANCE MODELS PREDICT CONTRIBUTE TO TRAIT VARIATION. THE DISSECTION OF COMPLEX TRAITS IN MODEL ORGANISMS OFFERS GREAT EXPERIMENTAL FLEXIBILITY, AND THE OPPORTUNITY TO DEPLOY METHODS SYNERGISTIC WITH THE DOMINANT GWAS PARADIGM, YIELDING A MORE COMPLETE PICTURE OF THE GENETIC BASIS OF TRAIT VARIATION. WITH THIS IN MIND WE ESTABLISHED THE DSPR (DROSOPHILA SYNTHETIC POPULATION RESOURCE) AS A SHAREABLE TOOLKIT FOR COMPLEX TRAIT ANALYSIS IN FLIES. THE SET OF 1600 DSPR STRAINS WERE DERIVED FROM 15 HIGHLY-CHARACTERIZED FOUNDER GENOTYPES, AND REPRESENT THE MOST EXTENSIVE MULTIPARENTAL POPULATION (MPP) AVAILABLE IN ANIMALS. THE DSPR IS USED BY MANY RESEARCH GROUPS, HAS ENABLED THE IDENTIFICATION OF THOUSANDS OF QTL, AND HAS ASSOCIATED RARE ALLELES WITH TRAIT VARIATION. THE DSPR COMPLEMENTS GWAS APPROACHES FOR UNCOVERING THE FULL SPECTRUM OF ALLELIC VARIATION CONTRIBUTING TO COMPLEX TRAITS. IN AIM 1 WE WILL VALIDATE AND REFINE THE GENOTYPES OF ALL DSPR STRAINS, STRENGTHENING THE LIVING RESOURCE WE WILL CONTINUE TO SHARE WITH THE DROSOPHILA COMMUNITY, AND ENHANCE THE USABILITY OF THE COLLECTION BY INTEGRATING ANALYTICAL ROUTINES INTO THE POWERFUL R/QTL2 SOFTWARE PLATFORM. IN AIM 2 WE WILL EXTEND THE DSPR, AND ENABLE EXTREME QTL (OR X-QTL) MAPPING. FOR MANY TRAITS, AN EFFICIENT AND USER-FRIENDLY STRATEGY TO IDENTIFY QTL IS TO COMPARE ALLELE FREQUENCIES IN PHENOTYPICALLY-EXTREME RECOMBINANT POPULATIONS TO THOSE IN CONTROL COHORTS. WE WILL DERIVE SUCH MIXED DSPR POPULATIONS, EXPERIMENTALLY REFINE OUR APPROACH, AND DISTRIBUTE POPULATIONS AND SOFTWARE TO FACILITATE NOVEL, INVESTIGATOR-DRIVEN RESEARCH. IN AIM 3 WE WILL BROADEN THE UTILITY OF THE DSPR TO EXPLORE THE NATURE OF EXPRESSION REGULATION. USING A HOST-PATHOGEN MODEL, WE WILL EXPLOIT THE RAPID REGULATORY CHANGE THAT OCCURS DURING THE IMMUNE RESPONSE TO DEVELOP DYNAMIC EQTL MAPPING METHODS, FINDING LOCI THAT CONTRIBUTE TO VARIATION IN THE TRAJECTORY OF THE REGULATORY REACTION. THESE GENERAL RESOURCES WILL ALLOW RESEARCHERS EMPLOYING ANY MPP TO MOVE BEYOND STATIC PICTURES OF GENE EXPRESSION IN POPULATIONS.

NSF CONVERGENCE ACCELERATOR TRACK G: COMBATING VULNERABILITY AND UNAWARENESS IN 5G NETWORK SECURITY -RELIABLE AND HIGH-RATE 5G WIRELESS ACCESS HAS BECOME A GLOBAL NECESSITY; HOWEVER, THE US HAS FALLEN BEHIND IN WIRELESS LEADERSHIP, LACKING MAJOR RADIO ACCESS NETWORK (RAN) OR CELLULAR NETWORK MANUFACTURERS. FURTHERMORE, CELLULAR NETWORKS HAVE NOT BEEN DESIGNED FOR MISSION-CRITICAL COMMUNICATIONS AND HAVE EXPOSED SEVERAL SECURITY VULNERABILITIES. CONSEQUENTLY, THE DEPARTMENT OF DEFENSE (DOD) FACES CHALLENGES IN USING COMMERCIAL OFF-THE-SHELF 5G PRODUCTS AND COMMERCIAL NETWORKS FOR US MILITARY OPERATIONS. THE ZERO TRUST X (ZTX) TEAM, A CONSORTIUM OF INTERDISCIPLINARY EXPERTS IN THE FIELD OF 5G AND SECURITY, WILL RESEARCH AND DEVELOP A FAMILY OF SECURITY SOLUTIONS TO ESTABLISH A ZERO TRUST CHAIN (ZTC) THAT ENABLES END-TO-END SECURITY AND PROTECTION FOR RELIABLE USE OF 5G NETWORKS FOR DOD USE CASES. THE PROPOSED EFFORT WILL GENERATE KNOWLEDGE AND RESEARCH OUTCOMES TAILORED FOR USE BY US INDUSTRY AND DOD. ADDITIONALLY, THE PROJECT WILL TRAIN A DIVERSE TEAM OF STUDENTS IN RESEARCH AND PROVIDE OPEN-SOURCE SOFTWARE THAT FACILITATES PORTABILITY, REPRODUCIBILITY, AND INTEGRATION WITH OTHER TRACK G SOLUTIONS OF THIS PROGRAM. THE PROJECT'S SPECIFIC GOAL IS TO DEVELOP THE ZTC SOFTWARE THAT ENABLES MILITARY SQUADS TO SECURELY SHARE SITUATIONAL AWARENESS IN THEIR OPERATIONS USING HIGH-PERFORMANCE, YET OFTEN UNTRUSTED, 5G NETWORKS. THE SOFTWARE SOLUTION LEVERAGES THE FLEXIBILITY OF THE 5G STANDARD AND IMPLEMENTS INNOVATIVE SECURITY SOLUTIONS AT DIFFERENT NETWORK NODES AND LAYERS TO EMPOWER DOD OPERATORS TO DETECT MALICIOUS ENTITIES IN NEAR-REAL TIME AND ESTABLISH COMMUNICATION MECHANISMS TO PREVENT ACCESS TO OR CONTROL OVER DOD TRAFFIC. SPECIFICALLY, THROUGH MINIMAL COOPERATION WITH 5G NETWORK OPERATORS, PART OF THE ZTC SOLUTION LEVERAGES OPEN-RAN (O-RAN) AND 5G CORE-CENTRIC APPROACHES FOR PRACTICAL THREAT MONITORING AND MITIGATION. THIS IS COMPLEMENTED BY DEVICE-CENTRIC SECURITY ENHANCEMENTS TO ENSURE THAT DOD DEVICES ALSO IMPLEMENT THEIR OWN LAYER OF SECURITY AND DO NOT SOLELY DEPEND ON THE SECURITY PROTOCOLS OF THE NETWORK PROVIDER. SIX KEY FEATURES SET ZTC APART FROM OTHER SOLUTIONS: (I) IT BUILDS ON THE OPEN ARTIFICIAL INTELLIGENCE CELLULAR (OAIC) PLATFORM FOR DEVELOPING O-RAN THREAT MONITORING AND MITIGATION THROUGH RAN INTELLIGENT CONTROLLERS; (II) IT OFFERS END-TO-END SECURE SLICING ACROSS THE 5G RAN AND CORE; (III) IT DETECTS THREATS AT USER DEVICES IN NEAR-REAL TIME; (IV) IT PROTECTS COMMUNICATION THROUGH INNOVATION AT THE APPLICATION LAYER RATHER THAN MODIFYING EXISTING 5G PHYSICAL LAYER PROTOCOLS AND ALGORITHMS; (V) IT ENSURES LOCATION PRIVACY AND RESILIENCY TO UNKNOWN/UNANTICIPATED DENIAL OF SERVICE (DOS) ATTACKS; AND (VI) IT DOES NOT REQUIRE MODIFICATIONS TO PUBLIC 5G/O-RAN NETWORKS AND STANDARDS, AND ONLY REQUIRES INSTALLATION OF LOW-OVERHEAD SOFTWARE MODULES ON 5G USER DEVICES AND COOPERATIVE 5G NETWORKS. THE ZTX TEAM WILL IMPLEMENT AND EXPERIMENTALLY EVALUATE THE PROPOSED ZTC INITIALLY ON A LABORATORY-SCALE INTEGRATED 5G/O-RAN TESTBED, AND SUBSEQUENTLY ON OTHER AVAILABLE TESTBEDS TO PREPARE FOR COMMERCIAL TRANSITION. THE TEAM WILL APPLY CONVERGENCE ACCELERATOR FUNDAMENTALS TO FOSTER PARTNERSHIPS AND TO DEVELOP A SUSTAINABILITY MODEL WITH AN IMPACT EXTENDING WELL BEYOND PHASE 2 OF THE PROGRAM. MRNA EXPRESSION PROFILING FROM EXTRACELLULAR VESICLES (EVS): GENERATING A RAPID DIAGNOSTIC FOR STROKE - PROJECT SUMMARY/ABSTRACT STROKE IS A DISORDER OF THE BRAIN BY WHICH A PART LOSES ITS BLOOD SUPPLY AND THE AFFECTED REGION RAPIDLY PROGRESSES TO DEATH IF BLOOD FLOW IS NOT RESTORED IN TIME. TREATMENTS TO RESTORE BLOOD FLOW AFTER ACUTE ISCHEMIC STROKE (AIS) ARE EFFECTIVE IF ADMINISTERED <4.5 H FROM THE ONSET OF THE STROKE EVENT. BUT, DUE IN PART TO THE LACK OF AN IN VITRO DIAGNOSTIC TEST FOR AIS, IMAGING (CT – CLINICAL SENSITIVITY ~26%) AT THE ATTENDING HOSPITAL IS REQUIRED FOR DIAGNOSIS. AS A RESULT, <7% OF AIS PATIENTS RECEIVE TREATMENT. THEREFORE, A CRITICAL NEED EXISTS TO DEVELOP STRATEGIES FOR DIAGNOSING STROKE IN NEAR REAL TIME THAT POTENTIALLY CAN ALLOW FOR POINT-OF-CARE TESTING (POCT). ONE APPROACH IS A PERIPHERAL BLOOD TEST USING MARKERS THAT QUICKLY RESPOND TO CHANGES IN THE BRAIN INDUCED BY STROKE. THE PROPOSED PROJECT WILL DEVELOP INNOVATIVE TECHNOLOGIES THAT USES PERIPHERAL BLOOD MARKERS FOR DIAGNOSING STROKE SYNDROMES (AIS AND HEMORRHAGIC STROKE) IN NEAR REAL TIME (~31 MIN FOR SAMPLE-TO-ANSWER) WITH AN LOD OF ~0.03 NG (TOTAL RNA). THE RESEARCH TEAM HAS FOUND THAT ALTERATIONS IN MRNA EXPRESSION SECURED FROM WHITE BLOOD CELL SUBSETS CAN BE USED FOR STROKE DIAGNOSIS AND APPEAR RAPIDLY IN PERIPHERAL BLOOD FOLLOWING A STROKE EVENT. RESULTING FROM A PRIOR R01, CD15+ AND CD8+ LEUKOCYTES WERE DISCOVERED AS A PREDOMINANT SOURCE OF STROKE- RELATED MRNA BIOMARKERS. THIS PROJECT SEEKS TO REALIZE THE DEVELOPMENT OF AN INNOVATIVE FLUIDIC CARTRIDGE AND THE ASSOCIATED ASSAY FOR THE MEASUREMENT OF STROKE-RELATED RNA MARKERS SOURCED FROM CD15 AND CD8 EXPRESSING EXTRACELLULAR VESICLES (EVS). THE UTILITY OF EVS AS A SOURCE OF STROKE-RELATED RNA BIOMARKERS IS BASED ON THEIR RAPID APPEARANCE AND HIGH ABUNDANCE IN PLASMA, POTENTIALLY PROVIDING EVEN FASTER STROKE DIAGNOSIS COMPARED TO THE CELLS FROM WHICH THEY ORIGINATE. THIS PROJECT WILL DISCOVER EV-RNA MARKERS WITH HIGH CLINICAL SENSITIVITY AND SPECIFICITY (>80%) FOR DIAGNOSING ISCHEMIC AND HEMORRHAGIC STROKE IN <3 H FROM STROKE ONSET. THE CARTRIDGE, WHICH CONSISTS OF TASK-SPECIFIC MODULES MADE FROM PLASTICS VIA REPLICATION (I.E., INJECTION MOLDING) CONNECTED TO A FLUIDIC MOTHERBOARD, WILL USE THE EV-RNA MARKERS EMANATING FROM THIS PROJECT. PLASMA WILL SERVE AS THE INPUT FROM WHICH SURFACE-AFFINITY SELECTION OF CD8 AND CD15 EVS WILL OCCUR USING A SPECIFICALLY DESIGNED MODULE. FOLLOWING EV RELEASE FROM THE CAPTURE SURFACE VIA A PHOTOCLEAVABLE LINKER (CLEAVED USING A BLUE-LIGHT LED), THE CARTRIDGE WILL QUANTIFY THE NUMBER OF EVS SELECTED USING A LABEL-FREE READOUT STRATEGY. THE FLUIDIC CARTRIDGE ALSO CONSISTS OF A MIXED-SCALE (NM ΜM) MODULE TO READ ELECTRICALLY COPY NUMBERS OF STROKE EV-RNA MARKERS IN A HIGHLY MULTIPLEXED FASHION (>24 TARGETS). THIS MODULE WILL CONSIST OF IN-PLANE NANOPORES MADE IN A PLASTIC VIA NANO-INJECTION MOLDING AND CAN IDENTIFY RNAS USING A DISTINCT OLIGONUCLEOTIDE PRIMER PAIR QUERYING A SPECIFIC RNA USING A SOLID-PHASE LIGASE DETECTION REACTION (SPLDR). SINGLE-MOLECULE READOUT WILL ALLOW FOR HIGH ANALYTICAL SENSITIVITY TO OBSERVE SUBTLE CHANGES IN EV-RNA MARKER COPY NUMBERS. THE UTILITY OF THIS CARTRIDGE WILL BE EVALUATED IN A CLINICAL SETTING. SUCCESS OF THE PROJECT IS LEVERAGED BY A STRONG MULTIDISCIPLINARY TEAM, WHOM HAVE A PRODUCTIVE RECORD OF COLLABORATION IN DEVELOPING STROKE MARKERS AND DIAGNOSTIC PLATFORMS FOR STROKE.

A KEY OUTCOME WILL BE THE EDUCATION OF FUTURE MATERIALS SCIENTISTS AND BUILDING OF A QUANTUM-READY WORKFORCE TO BOOST KANSAS AND NEBRASKA?S ECONOMY. EDUCATION AND OUTREACH EFFORTS WILL RAISE PUBLIC AWARENESS AND INSPIRE THE NEXT GENERATION OF SCIENTISTS AND ENGINEERS. THE COLLABORATION BETWEEN THE UNIVERSITY OF KANSAS AND UNIVERSITY OF NEBRASKA?LINCOLN WILL COMBINE COMPLEMENTARY EXPERTISE IN SYNTHESIS, NANOFABRICATION, CHARACTERIZATION, SENSING, AND THEORY TO ADVANCE BOTH THE SCIENCE AND DEVICE APPLICATIONS OF ARTIFICIAL TWO-DIMENSIONAL (2D) MAGNETIC MATERIALS. 2D MAGNETIC AND FERROELECTRIC MATERIALS, ALONG WITH THEIR LAYERED AND TWISTED HETEROSTRUCTURES, UNLOCK NOVEL QUANTUM STATES AND ENABLE APPLICATIONS THAT GO BEYOND TRADITIONAL SILICON-BASED ELECTRONICS. REALIZING THEIR POTENTIAL REQUIRES UNDERSTANDING OF STRUCTURE INCLUDING MOIR? MODULATIONS, ELECTRONIC AND MAGNETIC STATES, AND EMERGENT INTERFACIAL EFFECTS. THE TEAM WILL EMPLOY PROBES SENSITIVE TO STRUCTURAL, ELECTRONIC, AND MAGNETIC PROPERTIES ACROSS MULTIPLE LENGTH SCALES TO ESTABLISH AN INTEGRATED CYCLE OF NOVEL SYNTHESIS, CHARACTERIZATION, AND DEVICE FABRICATION. EXISTING INFRASTRUCTURE, SUCH AS THE NEBRASKA CENTER FOR MATERIALS AND NANOSCIENCE, WILL BENEFIT FROM THE PROJECT THROUGH INCREASE AND BROADENING OF THE USER BASE AND UTILIZATION OF UNIQUE INSTRUMENTATION INCLUDING THE FIRST COMMERCIAL NITROGEN VACANCY LOW TEMPERATURE SCANNING MICROSCOPE IN THE US. THE PROJECT IS ESSENTIAL TO EDUCATE A QUANTUM-READY WORKFORCE, NECESSARY FOR THE ECONOMIC GROWTH OF BOTH JURISDICTIONS, AND TO ATTRACT THE NEXT GENERATION OF SCIENTISTS AND QUANTUM MATERIALS ENGINEERS. THIS PROJECT IS SUPPORTED BY THE EPSCOR RESEARCH INFRASTRUCTURE IMPROVEMENT PROGRAM: FOCUSED EPSCOR COLLABORATIONS (FEC), WHICH SUPPORTS INTERJURISDICTIONAL TEAMS OF EPSCOR INVESTIGATORS TO PERFORM RESEARCH IN TOPICS THAT ALIGN WITH NSF PRIORITIES, WITH THE GOALS OF DRIVING DISCOVERY AND BUILDING SUSTAINABLE STEM CAPACITY. THE PURPOSE OF THIS APPLICATION IS TO ESTABLISH A CENTER FOR PUBLIC SAFETY LEADERSHIP THAT WILL DEVELOP UNIVERSAL HIGH-QUALITY LEADERSHIP TRAINING THAT WILL ALSO SERVE THE UNIQUE NEEDS OF RURAL LAW ENFORCEMENT ACROSS THE MIDWEST. THE DELIVERY OF THIS TRAINING WILL USE INSTRUCTIONAL METHODOLOGIES THAT LEVERAGE ECONOMIES OF NATIONAL SCALE, DEVELOPED BY THE KANSAS LAW ENFORCEMENT TRAINING CENTER (KLETC) AND THE UNIVERSITY OF KANSAS (KU), THE STATE’S FLAGSHIP UNIVERSITY. AT KU, WE PUSH THE BOUNDARIES OF KNOWLEDGE, TRANSFORM THE ACADEMIC EXPERIENCE, AND CREATE SOLUTIONS THROUGH INNOVATIVE RESEARCH. KLETC IS WELL-SITUATED TO FACILITATE THIS CENTER WITH AN EXCELLENT INFRASTRUCTURE ON CAMPUS AND VAST RESOURCES AVAILABLE FROM KU. PERFORMANCE MEASURES WILL INCLUDE QUANTITATIVE AND QUALITATIVE METHODS. QUANTITATIVELY, THE NUMBER OF COURSES AND OFFICERS TRAINED WILL BE COUNTED AS AN OUTPUT OF THE TRAINING PROCESS. ADDITIONALLY, THE NUMBER OF RURAL LAW ENFORCEMENT AGENCIES SERVED WILL BE TRACKED. QUALITATIVELY, EVALUATIONS OF EACH COURSE DELIVERED, IN ANY FORMAT, WILL INCLUDE A STUDENT SURVEY TO MEASURE THEIR LEVEL OF SATISFACTION AS IT RELATES TO TRAINING CONTENT, TOPIC, AND DELIVERY. THE INSTRUCTOR’S PERFORMANCE AND ABILITY WILL ALSO BE MEASURED. PERFORMANCE MEASURES WILL BE CONTINUALLY EVALUATED AND ADJUSTED, BASED ON THE COURSES DELIVERED. THE CENTER WILL BE RUN BY A DIRECTOR UNDER THE GUIDANCE OF THE DIRECTOR OF POLICE TRAINING/KLETC EXECUTIVE DIRECTOR. AN ASSISTANT DIRECTOR FOR CURRICULUM DEVELOPMENT WILL DEVELOP AND MAINTAIN THE CENTER’S CURRICULUM AND TESTING PROGRAMS, AND AN ASSISTANT DIRECTOR FOR CURRICULUM DELIVERY WILL HAVE STRATEGIC AND OPERATIONAL OVERSIGHT WITH RESPECT TO PROGRAMMATIC FUNCTIONS WITHIN THE CENTER. THE DEVELOPMENT OF THE CENTER WILL BE IMPLEMENTED IN PHASES IN THE AREAS OF STAFFING, COURSE DEVELOPMENT AND COURSE OFFERINGS. SUCH RESERVE ANTIBIOTICS OFTEN CARRY THE RISK OF SIGNIFICANT TOXICITY AND SUBSEQUENT PATIENT HARM. THUS, THE TIMELY DEVELOPMENT OF NEW ANTIBACTERIAL AGENTS, PREFERABLY WITH MODES OF ACTION DISTINCT FROM ONES CURRENTLY EMPLOYED, IS ESSENTIAL FOR THE GLOBAL DECREASE OF HUMAN MORBIDITY AND MORTALITY. THE CHEMICAL SYNTHESIS OF KNOWN ANTIBACTERIAL NATURAL PRODUCTS IS A STRATEGY THAT HAS YIELDED GREAT DIVIDENDS IN THE PAST AND CONTINUES TO FURNISH IMPORTANT CLINICAL THERAPEUTICS IN THE PRESENT. OFTEN, WHILE ENTRENCHED IN THE CONSTRUCTION OF SUCH COMPLEX MOLECULES, THE SYNTHETIC PRACTITIONER IS CONFRONTED BY THE LIMITATIONS OF AVAILABLE TECHNOLOGY. THUS, THE DEVELOPMENT OF NEW ORGANIC METHODOLOGY IS OFTEN INTIMATELY JOINED WITH THE PURSUIT OF BIOLOGICALLY-ACTIVE NATURAL PRODUCTS. THE OVERARCHING THEMES GUIDING THIS R35 PROPOSAL ARE THE DEVELOPMENT OF NEW TETHERED AZA-WACKER CYCLIZATION REACTIONS AND THEIR USE IN THE SYNTHESIS OF NITROGENOUS ANTIBIOTICS. THE IMPORTANCE OF A TETHERED REACTION IS THAT IT FREES THE SYNTHETIC PRACTITIONER FROM THE CONSTRAINT OF NEEDING A PRE-EXISTING C–N BOND IN ORDER TO FORGE A NEW ONE, AND OUR LABORATORY HAS PUBLISHED SEVERAL STUDIES IN THIS AREA. WE PLAN TO APPLY THIS TECHNOLOGY FOR THE SYNTHESIS OF THE UNUSUAL DICHLORINATED POLYKETIDE ANTIBIOTICS BACTOBOLINS A-B AND ACYBOLIN A. BACTOBOLINS A AND B WERE FIRST ISOLATED FROM THE CULTURE BROTH OF PSEUDOMONAS YOSHIDOMIENSIS AND HAVE BEEN DEMONSTRATED TO HAVE BROAD ANTIBACTERIAL ACTIVITY AGAINST A VARIETY OF PATHOGENIC GRAM-POSITIVE AND GRAM-NEGATIVE ORGANISMS. RECENT WORK HAS PINPOINTED THE ANTIBACTERIAL ACTIVITY OF BACTOBOLIN A TO INHIBITION OF BACTERIAL PROTEIN TRANSLATION VIA BINDING TO AN UNPRECEDENTED SITE ON THE 50S PROKARYOTIC SUBUNIT (L2); A CRYSTAL STRUCTURE EXISTS SHOWING THIS INTERACTION. DESPITE THEIR RICH BIOLOGICAL ACTIVITY, THERE EXISTS ONLY ONE RACEMIC SYNTHESIS (WEINREB) AND ONE ENANTIOSPECIFIC SYNTHESIS (ŠVENDA) OF BACTOBOLIN A. ACYBOLIN A IS A RECENTLY ISOLATED ANTIBIOTIC WITH ACTIVITY AGAINST BACTOBOLIN-RESISTANT STRAINS OF B. SUBTILIS, SUGGESTING A DISPARATE MODE OF ACTION FROM BACTOBOLINS DESPITE STRUCTURAL HOMOLOGY. THERE CURRENTLY EXISTS NO SYNTHESIS OF THE ACYBOLIN FAMILY OF NATURAL PRODUCTS, NO EXPLORATION OF ANALOGUES, AND LIMITED ANALYSIS OF BIOLOGICAL ACTIVITY. A KEY STEP IN OUR PROPOSED SYNTHESIS OF THESE COMPOUNDS IS A REACTION RECENTLY DEVELOPED IN OUR LABORATORY, THE SULFAMATE AZA-WACKER CYCLIZATION. THE EXPECTED OUTCOMES OF THIS LINE OF RESEARCH INCLUDE THE DEVELOPMENT OF POWERFUL NEW METHODS FOR THE SITE-SELECTIVE AMINATION OF ALKENE MOIETIES AND ACCESS TO A VARIETY OF NEW ANTIBACTERIAL COMPOUNDS THROUGH PRECISE STEREOCONTROLLED SYNTHESIS. NEW CATALYTIC STRATEGIES TO MAKE NON-PROTEINOGENIC PEPTIDES - PROJECT SUMMARY/ABSTRACT G PROTEIN-COUPLED RECEPTORS (GPCRS) ARE VENERABLE TARGETS FOR DRUG DISCOVERY. ONE-THIRD OF ALL DRUGS IN CLINICAL USE TARGET A GPCR. THE ENDOGENOUS LIGANDS FOR MANY GPCRS ARE PEPTIDES, WHICH MAKE PEPTIDES IDEAL PROBES FOR EXPLORING THE STRUCTURE-ACTIVITY RELATIONSHIP OF THESE GPCRS, AND FOR DEVELOPING DRUGS THAT MODULATE THEIR ACTIVITY. WHILE A SELECT FEW NATIVE GPCR PEPTIDE LIGANDS HAVE BEEN SUCCESSFULLY TRANSLATED INTO HUMAN THERAPIES, THE VAST MAJORITY ARE PLAGUED BY INADEQUATE PK/PD PROPERTIES AND MAKE VERY POOR DRUGS. REPLACING THE NATURAL AMINO ACIDS (AAS) IN THE PEPTIDE WITH NON-PROTEINOGENIC AMINO ACIDS (NPAAS) CAN ENHANCE THE DRUG- LIKE PROPERTIES OF ORDINARY PEPTIDES, THEREBY IMPROVING THEIR PRACTICAL USE AS MEDICINES. 80% OF ALL GPCR PEPTIDE DRUGS CONTAIN NPAAS. DETERMINING WHICH NPAAS TO INCLUDE IN THE PEPTIDE AND WHERE TO PLACE THEM LARGELY DEPENDS ON TRIAL AND ERROR, EACH PEPTIDE VARIANT BEING MADE AS THE UNIQUE PRODUCT OF A SEPARATE MULTI-STEP SOLID- PHASE CHEMICAL SYNTHESIS PROCEDURE THAT RELIES ON A LIMITED POOL OF NPAAS WITH POOR SOLID-PHASE COUPLING EFFICIENCIES. NEW TECHNOLOGIES WHICH ENABLE NPAAS TO BE INTRODUCED INTO PEPTIDES IN A GENERAL, SYNTHETICALLY DIVERGENT, AND COST-EFFECTIVE MANNER WOULD (1) CONSIDERABLY IMPROVE GPCR PEPTIDE DRUG DEVELOPMENT AS IT IS PRACTICED TODAY, (2) ENABLE ACCESS TO COMPREHENSIVE PEPTIDE LIBRARIES TO THOROUGHLY EXPLORE STRUCTURE-ACTIVITY RELATIONSHIPS OF GPCRS, AND (3) PROMOTE THE DEVELOPMENT AND DEPLOYMENT OF NEW SYNTHETIC METHODS, WHICH WILL IN TURN ADVANCE THE FIELDS OF SYNTHETIC CHEMISTRY AND MEDICINAL PEPTIDE CHEMISTRY. OUR LABORATORY HAS DEVELOPED A NEW PARALLEL SYNTHESIS APPROACH THAT GENERATES ENTIRE LIBRARIES OF INDIVIDUAL PEPTIDE ANALOGS IN A SINGLE STEP. WE HAVE SHOWN THAT THIS STRATEGY CAN YIELD LIBRARIES OF PEPTIDES WHEREIN A SINGLE AMINO ACID IS TRANSFORMED TO ONE OF A MYRIAD OF NEW NPAA VARIANTS. TO FURTHER DEVELOP OUR STRATEGY AS A GENERAL TOOL FOR MEDICINAL CHEMISTRY AND DEMONSTRATE ITS ADVANTAGES FOR OPTIMIZING GPCR PEPTIDES WE PROPOSE HERE TO GENERATE AND TO EVALUATE LIBRARIES OF A NEW GPCR PEPTIDE LIGAND DESIGNED BY OUR LAB THAT HAS PROMISING ANTI-HIV ACTIVITY. BY PURSUING BOTH CHEMISTRY AND BIOLOGY IN PARALLEL WE WILL USE OUR NEW PEPTIDE AS A VEHICLE TO REFINE AND IMPROVE OUR NEWLY MINTED CHEMICAL METHODOLOGY TO PREPARE (1) PEPTIDES WITH MULTIPLE NPAAS, AND (2) PEPTIDES CONTAINING D-AMINO ACIDS, WHICH WILL ADVANCE OUR PEPTIDE AS AN ENTIRELY NEW ANTIRETROVIRAL DRUG AND WILL MAKE OUR SYNTHETIC PLATFORM MORE USEFUL FOR GPCR PEPTIDE DRUG DEVELOPMENT.
