PURPOSE: THE AEROSPACE EDUCATION HUB IS THE SECOND FACILITY OF THE AEROSPACE INNOVATION RAMP AT KANSAS STATE UNIVERSITY SALINA, A CLUSTER OF INDUSTRY-CONNECTED TEACHING AND RESEARCH SPACES THAT SUPPORT THE CAMPUS' VISION OF BEING THE MOST TECHNICALLY PROGRESSIVE OPERATIONAL AEROSPACE EDUCATION AND TRAINING INSTITUTION IN THE NATION.ACTIVITIES TO BE PERFORMED: THE AEROSPACE EDUCATION HUB WILL HOUSE THE APPLIED AEROSPACE RESEARCH CENTER, THE AVIATION MAINTENANCE TRAINING CENTER, AND THE ADVANCED COMPOSITES LAB. THE APPLIED AEROSPACE RESEARCH CENTER (AARC) IS AT THE FOREFRONT OF UAS RESEARCH THAT INFLUENCES AND ENHANCES UAS REGULATIONS AND INCREASES CAPABILITIES, FOUNDED ON MANNED AVIATION?S CULTURE OF SAFETY. THE AARC STAFF IS COMPRISED OF SUBJECT-MATTER EXPERTS, CONNECTING WITH INDUSTRY TO PROVIDE TRAINING, CONSULTING, COLLABORATION AND RESEARCH. THE AVIATION MAINTENANCE TRAINING CENTER PROVIDES HANDS-ON TRAINING OF THE MOST UP-TO-DATE MAINTENANCE PROCEDURES IN ORDER TO PRODUCE GRADUATES PREPARED TO OBTAIN FAA LICENSURE AND BEGIN LUCRATIVE CAREERS AT THE MAJOR AIRLINES AND MRO FACILITIES AROUND THE COUNTRY. THE ADVANCE COMPOSITES LAB NOT ONLY TRAINS STUDENTS IN THE ART AND SCIENCE OF COMPOSITE REPAIR AND FABRICATION, BUT ALSO CONDUCTS RESEARCH ON FUTURE COMPOSITE TECHNOLOGIES. TAKEN TOGETHER, THE AEROSPACE EDUCATION HUB WILL BE THE MOST ADVANCED TRAINING AND RESEARCH FACILITY OF ITS KIND IN THE COUNTRY.EXPECTED OUTCOMES: SUPPORT THE DESIGN, CONSTRUCTION, AND OUTFITTING OF THE AEROSPACE EDUCATION HUB. THE FACILITY WILL ENCOMPASS APPROXIMATELY 62,000 GSF OF CLASSROOM, OFFICE, LAB, AND HANGAR SPACE.INTENDED BENEFICIARIES: THE AEROSPACE EDUCATION HUB'S OBJECTIVE IS TO PROVIDE A COLLABORATIVE, TECHNICALLY PROGRESSIVE HOME TO KEY DISCIPLINES FOR THE AEROSPACE INDUSTRY TO FURTHER EDUCATION AND RESEARCH IN THE AEROSPACE SECTOR. BY CO-LOCATING UAS, COMPOSITE, AND AVM EDUCATION AND RESEARCH, K-STATE SALINA IS POISED TO BE A LEADER IN THE FUTURE OF AVIATION, AEROSPACE, AND ADVANCED AIR MOBILITY. THE FACILITY WILL HOUSE K-STATE SALINA FACULTY AND RESEARCH STAFF IN UAS, COMPOSITES, AND AVIATION MAINTENANCE. THE INITIAL OCCUPANCY IS ESTIMATED AT APPROXIMATELY 30 INDIVIDUALS. THIS ENVIRONMENT AMPLIFIES YIELD VARIABILITY POSING A RISK TO THE FOOD SUPPLY AND PRODUCER'S INCOME. NOVEL CROPLAND OBSERVATION NODES (CRONOS) WILL DELIVER REAL-TIME SOIL MOISTURE INFORMATION TO PRODUCERS AND EXTENSION AGENTS. DATA FROM FIELD EXPERIMENTS WILL CONSTRUCT DETAILED WATER AND NUTRIENT BUDGETS OF EXISTING AND IMPROVED CROPPING SYSTEMS. INTEGRATED MODELING, REMOTE SENSING, AND GROUND-BASED MONITORING WILL SIMULATE INNOVATIVE CROP-LIVESTOCK PRODUCTION SYSTEMS AND GENERATE EFFECTIVE DECISION SUPPORT TOOLS. ON-FARM SITES WILL EVALUATE PRODUCTION PRACTICES FOR SUCCESSFUL ADOPTION OF INTENSIFIED/DIVERSIFIED SYSTEMS FOR ENHANCED WATER AND N USE. THIS PROJECT WILL PROVIDE TRAINING TO UNDERGRADUATE AND GRADUATE STUDENTS, WHO WILL GAIN HANDS-ON EXPERIENCE REGARDING SUSTAINABILITY OFRAINFED AGRICULTURAL SYSTEMS.

THIS PLAN WOULD INCLUDE A RANGE OF ACTIVITIES TO SUPPORT SUCH EFFORTS AS: 1) INCREASING INSTITUTIONAL AND HUMAN CAPACITY THAT BETTER MEET THE DEMANDS OF THE AGRICULTURAL ECONOMY, 2) DEVELOPING REVENUE-GENERATING SERVICES TO PROVIDE TO THE REGION, AND 3) IMPROVING LINKAGES BETWEEN UNIVERSITIES, EXTENSIONISTS (PUBLIC AND PRIVATE), PRIVATE SECTOR, AND FARMERS. THE GOAL IS TO DEVELOP AN EASY-TO-FOLLOW AND REALISTIC PLAN THAT FOCUSES ON BETTER CONNECTING UNIVERSITIES TO FARMERS AND FARMS AND TRAINING INDIVIDUALS WHO WILL WORK IN THE SECTOR. ACTIVITIES TO BE PERFORMED: THE PRIMARY ACTIVITY WILL BE THE SELECTION AND PURCHASE OF THE EXPERIMENTAL AND COMPUTATION EQUIPMENT NEEDED TO PERFORM STATE-OF-THE-ART STEEL RESEARCH WITHIN THE CENTER. RESEARCH WILL BE FOCUSED ON ALLOY DESIGN, ALLOY FABRICATION AND HEAT TREATMENT FACILITIES (PROCESSING) AND PROPERTY ANALYSIS. THE EQUIPMENT SELECTION WILL INVOLVE REPRESENTATIVES OF LOCAL STEEL INDUSTRY, WHICH INCLUDE NUCOR, US STEEL AND OTHERS, TO ENSURE THE NEW CENTER MEETS THE NEEDS OF THESE COMPANIES. CLIMATE-CHANGE INDUCED DROUGHTS AND HEAT STRESS ACROSS THE REGION WILL INTENSIFY OVER THE NEXT 50 YEARS RAMPING UP PRESSURE ON AN ALREADY STRAINED AQUIFER SYSTEM. THE TRANSITION FROM FULLY IRRIGATED PRODUCTION TO CLIMATE-SMART SYSTEMS PRESENTS SIGNIFICANT BIOPHYSICAL SOCIOLOGICAL AND ECONOMIC IMPACTS FOR THE REGION. TO ADDRESS THESE CHALLENGES WE WILL APPLY AN INTEGRATED APPROACH COMPOSED OF RESEARCH CLASSROOM AND PRODUCER-LED EDUCATION AND EXTENSION PROGRAMS ACROSS THREE AIMS: 1) SOCIO-ECONOMIC 2) TECHNICAL AND 3) EDUCATION AND OUTREACH. OPENET A SATELLITE-BASED EVAPOTRANSPIRATION (ET) MODELING DATA VISUALIZATION AND ACCESS TOOL CAN PLAY A CRUCIAL ROLE IN ENABLING INNOVATIVE APPROACHES TO IRRIGATION PLANNING AND SCHEDULING DECISIONS BY PRODUCERS AND IDENTIFYING AREAS OF DIFFERENTIAL IRRIGATION EFFICIENCY USING BOTH FIELD-SCALE AND HISTORICAL DATA. SIMILARLY THE 'Q-STABLE' APPROACH CAN PROVIDE DATA-DRIVEN INSIGHTS INTO PUMPING LEVELS REQUIRED FOR AQUIFER SUSTAINABILITY. BY COLLABORATING WITH STAKEHOLDERS TO IMPROVE AND USE THESE TOOLS IN NEW WAYS THE PROJECT WILL ADDRESS THE SOCIO-ECONOMIC CHALLENGES OF INCREASING WATER USE EFFICIENCY AND REDUCING WATER USE TO LEVELS NEEDED TO STABILIZE AQUIFER LEVELS. THE KSRE NETWORK INCLUDES OFFICES IN ALL 105 COUNTIES, ALONG WITH FOUR REGIONAL EXTENSION OFFICES, FIVE AGRICULTURAL RESEARCH CENTERS DISTRIBUTED THROUGHOUT THE STATE TO ENCOMPASS VARIABILITY IN CLIMATE AND SOILS, FOUR SATELLITE UNITS, FOUR AGRONOMY EXPERIMENT FIELDS AND FOUR HORTICULTURE CENTERS AND EXPERIMENT FIELDS. IN ADDITION, KSRE FUNDS RESEARCH PROJECTS IN 20 DEPARTMENTS ACROSS FIVE COLLEGES. A UNIQUE FEATURE WITHIN THE K-STATE RESEARCH AND EXTENSION ORGANIZATION IS THE CLOSE ALIGNMENT OF RESEARCH AND EXTENSION. IN 1996, K-STATE RESEARCH AND EXTENSION WAS FORMED BY ALIGNING THE KANSAS AGRICULTURAL EXPERIMENT STATION (KAES) AND THE KANSAS COOPERATIVE EXTENSION SERVICE. THE STRATEGIC INTENT OF THIS ALIGNMENT WAS TO ACHIEVE GREATER EFFICIENCY AND SYNERGY BETWEEN DISCOVERY AND OUTREACH EFFORTS. ONE ANCILLARY BENEFIT OF THIS ALIGNMENT IS THAT BASIC AND APPLIED RESEARCH WILL LEAD TO RAPID EXTENSION PROGRAMMING AND APPLICATION IN THE FIELD. THIS ALIGNMENT ALSO ALLOWS MORE FLUID COMPLIANCE WITH MULTI-STATE AND INTEGRATED COMPONENTS OF AREERA. K-STATE RESEARCH AND EXTENSION IS FOCUSING ITS EFFORTS ON FIVE GRAND CHALLENGES (CRITICAL ISSUES) THAT DIRECTLY OR INDIRECTLY AFFECT ALL KANSANS: GLOBAL FOOD SYSTEMS, WATER AND NATURAL RESOURCES, HEALTH, DEVELOPING TOMORROWS LEADERS, AND COMMUNITY VITALITY. THESE GRAND CHALLENGES WERE DETERMINED THROUGH A SERIES OF MEETINGS WITH STAKEHOLDERS AND DEFINE AREAS OF EMPHASIS FOR AGENTS, SPECIALISTS, AND RESEARCHERS. EXTENSION PROFESSIONALS AT THE LOCAL, REGIONAL AND STATE LEVEL WORK TO DEVELOP REGIONAL AND STATEWIDE SIGNATURE PROGRAMS ALONG WITH PROFESSIONAL DEVELOPMENT OPPORTUNITIES FOR ITS MEMBERS THROUGH 11 PROGRAM FOCUS TEAMS ADULT DEVELOPMENT AND AGING; COMMUNITY VITALITY; CROP PRODUCTION; FAMILY AND CHILD DEVELOPMENT; FAMILY RESOURCE MANAGEMENT; FARM MANAGEMENT; HORTICULTURE; LIVESTOCK PRODUCTION; NATURAL RESOURCES; NUTRITION, FOOD SAFETY AND HEALTH; AND YOUTH DEVELOPMENT. MANY OF THESE ISSUES, SUCH AS MORE SEVERE WEATHER, WILL REQUIRE THEM TO CHANGE HOW THEY FARM. CREATING A PROTECTIVE LAYER OVER SOIL WHEN GROWING FIELD CROPS COULD HELP FARMERS BETTER MANAGE MANY ISSUES AT ONCE. COVERING SOIL WITH SHEET PLASTIC PREVENTS WEED GROWTH, EROSION, AND MOISTURE LOSS. BUT, USING LARGE AMOUNTS PLASTIC CREATES WASTE, IS NOT ECO-FRIENDLY, AND TOO COSTLY FOR FIELD CROPS. NEW LOCALLY SOURCED TYPES OF BIOPLASTICS THAT FULLY BREAK DOWN INTO TO SAFE BY-PRODUCTS CAN BE MADE. THESE NEW MATERIALS COULD PROVIDE FARMERS WITH A GREEN WAY TO CONTROL WEEDS, FERTILIZE CROPS, PROTECT SOIL AND WATER RESOURCES, AND WORK WITH NATURE TO BETTER MANAGE THEIR FIELDS. THIS NATIONAL SCIENCE FOUNDATION RESEARCH INFRASTRUCTURE IMPROVEMENT TRACK-2 FOCUSED EPSCOR COLLABORATIONS (NSF RII TRACK-2 FEC) AWARD WILL BE A NEW PARTNERSHIP CALLED BIOWRAP BETWEEN KANSAS STATE UNIVERSITY, SOUTH DAKOTA SCHOOL OF MINES AND TECHNOLOGY, AND UNIVERSITY OF NEBRASKA. THIS PROJECT WILL BE FOCUSED ON ADVANCING FARM USE OF BIOPLASTICS. TEAMS WILL DO THIS BY LINKING NATURAL AND MAN-MADE SYSTEMS, STOPPING HARMFUL WEED-CROP-TILL CYCLES, AND HELPING BUILD GREEN RURAL INDUSTRIES. THE TEAM WILL USE LAB AND FIELD TRIALS, NOVEL ANALYTICS AND PRODUCTS, AND THE LINKS BETWEEN PEOPLE, FARMS, INDUSTRIES, AND NATURAL SYSTEMS TO HELP CHANGE HOW WE GROW FIELD CROPS FOR THE BETTER. THIS RESEARCH INFRASTRUCTURE IMPROVEMENT TRACK-2 FOCUSED EPSCOR COLLABORATIONS (RII TRACK-2 FEC) AWARD WILL BE A NEW PARTNERSHIP FOCUSED ON A PROJECT CALLED BIOWRAP (BIOPLASTICS WITH REGENERATIVE AGRICULTURAL PROPERTIES) BETWEEN KANSAS STATE UNIVERSITY, SOUTH DAKOTA SCHOOL OF MINES AND TECHNOLOGY, AND UNIVERSITY OF NEBRASKA. THE OVERARCHING GOAL OF THIS PROJECT IS TO REDUCE THE USE OF CONVENTIONAL PLASTICS, HERBICIDES, AND THEIR ASSOCIATED ENVIRONMENTAL IMPACTS IN AGRICULTURAL PRODUCTION. THIS GOAL WILL BE ACCOMPLISHED THROUGH THE FOLLOWING INTEGRATED THRUST AREAS: (I) CREATE A SET OF BIO-BASED POLYMERS THAT CAN BE USED IN AGRICULTURAL PRODUCTION; (II) MEASURE THE AGRICULTURAL EFFECTIVENESS AND ASSOCIATED ENVIRONMENTAL IMPACTS OF THE TECHNOLOGY; AND (III) ESTIMATE THE POTENTIAL SOCIAL AND ECONOMIC IMPACTS ON COMMUNITIES AND THE AGRICULTURAL ECONOMY OF THE PROPOSED TECHNOLOGY ALONG WITH ESTABLISHING A CONSORTIUM FOR SUSTAINED COLLABORATIONS AMONG UNIVERSITY PARTNERS IN KANSAS, SOUTH DAKOTA, AND NEBRASKA IN THE FIELD OF SPRAY-ON BIOPOLYMER BASED FILMS FOR AGRICULTURAL PRODUCTION. THE RESEARCH WILL INCLUDE: ENGINEERING BIOPOLYMERS INCLUDED FUNCTIONALIZED PHA-BASED BIOPOLYMERS, PROTEIN BASED SPRAYABLE BIOPOLYMERS, AND HYBRID POLYMER SYSTEMS; STUDYING THE IMPACT OF THIS TECHNOLOGY ON STRENGTHENING OF AGROECOSYSTEMS BY EVALUATION OF BIOPOLYMER WITH RESPECT TO WEED SUPPRESSION AND CROP PRODUCTION EFFICIENCY, NUTRIENT CYCLING, SOIL MOISTURE RETENTION AND LIFE CYCLE IMPACTS ALONG WITH ENVIRONMENTAL ACCOUNTING; AND ASSESSING THE BROADER IMPACTS OF THE NEW BIOWRAP TECHNOLOGY USING SOCIO-ECONOMIC ANALYSIS OF PRODUCT ADOPTION AND RURAL SUSTAINABILITY ALONG WITH VALUING ECOSYSTEM SERVICES PROVIDED BY BIOWRAP, ECOSYSTEM SERVICE MARKET DESIGN AND PARTICIPATION AND ITS IMPACT ON SOCIO-ECONOMICS OF PARTICIPATING EPSCOR STATES AS WELL AS OVERALL SUSTAINABLE DATA PROVISION OF THE PROJECT. ACTIVITIES TO BE PERFORMED - THE KITE PROJECT WILL BUILD A HANGAR SPACE TO ACCOMMODATE HIGH-DEFINITION LARGE FORMAT DISPLAYS, MULTI-PURPOSE RECONFIGURABLE LED SETS, MOTION PLATFORMS SUITES, AN AUGMENTED REALITY/VIRTUAL REALITY DEVELOPMENT LABORATORY, A HAPTICS LABORATORY, A HOLOGRAPHIC AUDITORIUM, 3-D MODEL AND RENDERING STATIONS, MOTION CAPTURE CAPABILITIES, AND A LASER PROJECTION CAVE. STUDENT CLASSROOMS, COLLABORATIVE RESEARCH SPACES, AND A K-12 STEM RECRUITMENT ATTRACTION ARE ALSO TO BE INCLUDED IN THE HANGAR. THE CONGRESSIONALLY DIRECTED SPENDING ALLOCATED THROUGH NIST PROVIDES INITIAL FUNDING TOWARDS THE DESIGN FEES ASSOCIATED WITH THE FACILITY. FACTORS SUCH AS FREQUENT RELOCATION,REPEATED AND LENGTHY DEPLOYMENT, ISSUES ASSOCIATED WITH REUNION AND REINTEGRATION, SINGLE PARENT HOUSEHOLDS, LOWER INCOME,AND FAMILY MEMBERS RETURNING HOME WOUNDED OR NOT AT ALL PUT THESE YOUTH AND FAMILIES AT RISK. IT IS IMPORTANT FOR YOUTH TO HAVEOPPORTUNITIES TO DEVELOP SELF-CONFIDENCE, BUILD RESILIENCY, AND EXPERIENCE PERSONAL GROWTH SUPPORTED BY PEOPLE AND THEIR ENVIRONMENT. 4-H SUPPORTS THE DEVELOPMENT OF LIFE SKILLS THAT HELP YOUTH AND FAMILIES TACKLE THESE ISSUES WITH MEANING AND PURPOSE. THIS PROGRAM WILL PROVIDE QUALITY, EXPERIENTIALLY DELIVERED, AGE APPROPRIATE, POSITIVE YOUTH DEVELOPMENT PROGRAMMING TO MEET THE CHANGING NEEDS OF MILITARY YOUTH AND FAMILIES. SUB-AWARDS TO STATES/TERRITORIES WILL SUPPORT 4-H YOUTH DEVELOPMENT PROGRAMS FOR MILITARY CONNECTED YOUTH THAT INCLUDE 4-H CLUBS, CAMPS, SPECIAL PROJECTS, AFTER SCHOOL PROGRAMS, AND OTHER 4-HOPPORTUNITIES. 4-H YOUTH CAMPS WILL PROVIDE OPPORTUNITIES FOR ARMY AND AIR FORCE YOUTH TO DEVELOP LEADERSHIP, COMMUNICATION AND TEAMWORK SKILLS. PROGRAM EVALUATIONS, IMPACT STATEMENTS, AND REPORT DATA FROM THE CONSORTIUM OF LAND GRANT UNIVERSITIES WILL BE SUMMARIZED IN AN ANNUAL REPORT. THE OVERALL GOAL OF THIS PROGRAM IS TO CREATE ENVIRONMENTS AND OFFER EXPERIENCES THAT PROMOTE POSITIVE GROWTH AND DEVELOPMENT FOR MILITARY CONNECTED CHILDREN ANDYOUTH. COMMUNITY PROJECT FUNDING/CONGRESSIONALLY DIRECTED SPENDING - CONSTRUCTION - TO MEET THE WORKFORCE NEEDS OF BIOMANUFACTURING FIRMS IN KANSAS, INCLUDING THE RECENTLY ANNOUNCED DECISION BY SCORPIUS BIOMANUFACTURING TO LOCATE ITS COMMERCIAL-SCALE BIOMANUFACTURING FACILITY IN MANHATTAN, K-STATE PROPOSES TO CREATE A BIOMANUFACTURING TRAINING AND EDUCATION CENTER (BTEC). THE CENTER WILL INCLUDE BIOMANUFACTURING EQUIPMENT THAT SUPPORTS TRAINING FOR ALL PHASES OF MODERN BIOMANUFACTURING PROCESSES. THE BTEC WILL BE CONFIGURED WITH STATE-OF-THE-ART EQUIPMENT AND SOFTWARE THAT PREPARES STUDENTS TO TRANSFER IMMEDIATELY TO JOBS ACROSS THE BIOMANUFACTURING AND BIOPROCESSING SECTORS. THIS INCLUDES UPSTREAM PROCESSING, THE FIRST STAGE OF BIOMANUFACTURING THAT FOCUSES ON CELL LINE DEVELOPMENT AND CULTIVATION, AS WELL AS DOWNSTREAM PROCESSING, WHERE THE HIGHLY VALUABLE BIOLOGICAL PRODUCTS THAT RESULT ARE PRODUCED AT SCALE. MULTIPLE LEVELS OF TRAINING AND EDUCATION ARE NEEDED FOR AVAILABLE AND FUTURE JOBS IN BIOMANUFACTURING. K-STATE IS WORKING CLOSELY WITH SCORPIUS AND OTHER COMPANIES TO UNDERSTAND THOSE NEEDS AND IS WORKING IN PARTNERSHIP WITH MANHATTAN AREA TECHNICAL COLLEGE TO DELIVER TRAINING PROGRAMS THAT SPAN PRE-COLLEGE, ASSOCIATE'S LEVEL, BACHELOR'S LEVEL, AND GRADUATE LEVEL PROGRAMS AS WELL AS NON-DEGREE, CERTIFICATE, AND SKILLS-BASED TRAINING. NOT ONLY WILL THE CREATION OF A DEDICATED BIOMANUFACTURING TRAINING AND EDUCATION CENTER (FACILITIES AND PROGRAMS) SUPPORT THE RAPIDLY GROWING NEEDS OF BIOMANUFACTURING COMPANIES IN KANSAS, BUT IT WILL ALSO ESTABLISH A PATHWAY AND A MEANS TO RETAIN STUDENTS IN THE STATE BY PREPARING THEM FOR WELL-PAYING JOBS. K-STATE PLANS TO RENOVATE AN EXISTING STRUCTURE, BUILT IN 1949, TO UPFIT IT INTO A BIOMANUFACTURING TRAINING FACILITY AND WORKING BIOMANUFACTURING FACILITY THAT CAN PRODUCE VACCINES AND OTHER BIOLOGICS FOR INDUSTRY. THE NEW FACILITY WILL MEET NIH REQUIREMENTS FOR BSL-2 CONTAINMENT AND BE DESIGNED AND ENGINEERED IN ACCORDANCE WITH ALL RELEVANT CODES, STANDARDS, AND GUI DES TO MEET USDA CLEAN ROOM PRODUCTION STANDARDS. THE FACILITY WILL BE MADE ADA ACCESSIBLE AND HAVE DEDICATED SPACE FOR PRODUCTION AND TRAINING. SUSTAINABLE DESIGN PRINCIPLES WILL BE FOLLOWED THROUGHOUT THE PROJECT.

THIS AGREEMENT WILL PROVIDE FUNDING FOR KANSAS STATE UNIVERSITY (KSU) TO CONDUCT ELIGIBLE TECHNICAL ASSISTANCE-RELATED ACTIVITIES AS AUTHORIZED BY CERCLA 104(K)(7). ACTIVITIES:THIS AGREEMENT PROVIDES FINANCIAL ASSISTANCE TO KANSAS STATE UNIVERSITY (KSU) TO IMPLEMENT ITS OWN PROJECT TO PROVIDE TECHNICAL ASSISTANCE TO TRIBAL NATIONS AND ENTITIES IN BROWNFIELDS COMMUNITIES BY DEVELOPING RESOURCES AND OFFERING DIRECT ASSISTANCE. KSU'S PROJECT WILL PROVIDE A FOUR-PRONGED APPROACH: FIRST, THEY WILL HELP TRIBAL COMMUNITIES BUILD CAPACITY THROUGH FREE E-TOOLS, WEB RESOURCES, EVENTS, AND SERVICES. TOOLS AND INITIATIVES WILL SUPPORT TRIBAL RESPONSE PROGRAMS, COMMUNITY ENGAGEMENT, SOCIAL-ECOLOGICAL FRAMEWORKS, INCORPORATING INDIGENOUS KNOWLEDGE AND CULTURAL RESILIENCE, AND WILL BE PROVIDED ON A VARIETY OF PLATFORMS. SECOND, THEY WILL EXECUTE A SERIES OF TRAININGS, OUTREACH, AND EDUCATIONAL PROGRAMMING IN-PERSON, LIVE, AND ASYNCHRONOUSLY TO MAXIMIZE ACCESS TO TECHNICAL ASSISTANCE RESOURCES. THIRD, THEY WILL CONDUCT ONE-ON-ONE TECHNICAL ASSISTANCE FOR MORE INVOLVED SUPPORT ON CAPACITY BUILDING TASKS AND FOR COMMUNITY-LEVEL ASSISTANCE. FINALLY, KSU WILL EVALUATE ITS PERFORMANCE AND PROGRESS, AS WELL AS DEVELOP AN OUTREACH STRATEGY TO INCREASE AWARENESS OF SERVICES AND REACH COMMUNITIES WITH THE HIGHEST DEGREE OF BURDEN. SUBRECIPIENT:THERE WILL BE SUBAWARDS AWARDED TO UNIVERSITIES AND OTHER PARTNERS, INCLUDING NORTHERN ARIZONA UNIVERSITY INSTITUTE FOR ENVIRONMENTAL PROFESSIONALS (ITEP) AND ALASKAN NATIVE TRIBAL HEALTH CONSORTIUM (ANTHC). KSU WILL SELECT AND ISSUE SUBAWARDS FOR UNIVERSITIES AND OTHER PARTNERS WHO HAVE EXPRESSED INTEREST IN COLLABORATING AND WILL INVOLVE SUBAWARDS ON AN AS NEEDED BASIS FOLLOWING PROPER FEDERAL EPA SUBAWARD POLICY. THESE SUBAWARDEES WILL ACT AS A CLOSE PARTNER IN THE DEVELOPMENT AND MANAGEMENT OF THE TRAINING AND DELIVERY OF TECHNICAL ASSISTANCE. THE SUBAWARD WILL TOTAL $1,025,000 OVER THE FIVE YEARS. OUTCOMES:THE FIRST DELIVERABLE CATEGORY INVOLVES HELPING BUILD CAPACITY THROUGH FREE E-TOOLS, WEB RESOURCES, EVENTS AND SERVICES. KSU PLANS TO INITIATE FOUR GATHERINGS PER YEAR WITH THE NTBWG/TBIC PARTNERSHIP, AND INCREASE THE NUMBER OF E-TOOLS AND VIRTUAL RESOURCES PROVIDED THROUGH THIS TASK. THESE INCLUDE SEVERE ACUTE RESPIRATORY CORONAVIRUS (SARS-COV), MIDDLE EAST RESPIRATORY SYNDROME CORONAVIRUS (MERS-COV) AND, MOST RECENTLY, SARS-COV-2, THE CAUSATIVE AGENT OF CORONAVIRUS DISEASE 2019 (COVID-19). CURRENTLY, THERE ARE NO LICENSED VACCINES OR ANTIVIRAL DRUGS AGAINST THESE VIRUSES, UNDERSCORING AN URGENT NEED FOR THE DEVELOPMENT OF PREVENTIVE AND THERAPEUTIC MEASURES AGAINST CORONAVIRUSES. OUR FORAY IN THIS AREA HAS RESULTED IN THE DISCOVERY OF BROAD-SPECTRUM INHIBITORS OF MULTIPLE VIRUSES, INCLUDING CORONAVIRUSES AND NOROVIRUSES THAT ENCODE 3CLPRO, AS WELL AS THE FIRST DEMONSTRATION OF CLINICAL EFFICACY BY A FELINE CORONAVIRUS 3CLPRO INHIBITOR. RECENTLY, WE HAVE DEMONSTRATED THAT A DIPEPTIDYL SERIES OF COMPOUNDS POTENTLY INHIBIT HUMAN CORONAVIRUSES, INCLUDING MERS-COV AND SARS-COV-2 IN CELL CULTURE, AND DISPLAY IN VIVO EFFICACY IN THE DPP4-KI MOUSE MODEL OF MERS-COV INFECTION. THE ANTIVIRAL TARGET OF THE COMPOUNDS WAS VALIDATED BY OBTAINING HIGH RESOLUTION CRYSTAL STRUCTURES 3CLPRO-INHIBITOR COMPLEXES FROM SARS-COV, SARS- COV-2 AND MERS-COV. WE HYPOTHESIZE HEREIN THAT THE IDENTIFIED SERIES CAN SERVE AS A LAUNCHING PAD FOR THE DEVELOPMENT OF SARS-COV-2-SPECIFIC ANTIVIRALS. THE IMMEDIATE AND OVERARCHING GOAL OF THE PROPOSED STUDIES IS TO FURTHER OPTIMIZE THE PHARMACOLOGICAL ACTIVITY PK PARAMETERS OF IDENTIFIED LEAD INHIBITORS OF SARS-COV-2 3CLPRO AND THE DEMONSTRATION OF IN VIVO EFFICACY AGAINST SARS-COV-2. THE EXPECTED OUTCOME OF OUR STUDIES IS THE SELECTION OF A PRECLINICAL CANDIDATE (AND 1-2 BACKUP COMPOUNDS) THAT IS WELL-SUITED TO CONDUCTING FURTHER PRECLINICAL STUDIES, ULTIMATELY LEADING TO THE DEVELOPMENT OF A COVID-19-SPECIFIC ANTIVIRAL THERAPEUTIC.

K-STATE RESEARCH AND EXTENSION (KSRE) IS A STATEWIDE NETWORK OF EDUCATORS SHARING UNBIASED, RESEARCH-BASED INFORMATION AND EXPERTISE ON ISSUES IMPORTANT TO KANSANS. THE KSRE NETWORK INCLUDES OFFICES IN ALL 105 COUNTIES, ALONG WITH FOUR REGIONAL EXTENSION OFFICES, FIVE AGRICULTURAL RESEARCH CENTERS DISTRIBUTED THROUGHOUT THE STATE TO ENCOMPASS VARIABILITY IN CLIMATE AND SOILS, FOUR SATELLITE UNITS, FOUR AGRONOMY EXPERIMENT FIELDS AND FOUR HORTICULTURE CENTERS AND EXPERIMENT FIELDS. IN ADDITION, KSRE FUNDS RESEARCH PROJECTS IN 20 DEPARTMENTS ACROSS FIVE COLLEGES. A UNIQUE FEATURE WITHIN THE K-STATE RESEARCH AND EXTENSION ORGANIZATION IS THE CLOSE ALIGNMENT OF RESEARCH AND EXTENSION. IN 1996, K-STATE RESEARCH AND EXTENSION WAS FORMED BY ALIGNING THE KANSAS AGRICULTURAL EXPERIMENT STATION (KAES) AND THE KANSAS COOPERATIVE EXTENSION SERVICE. THE STRATEGIC INTENT OF THIS ALIGNMENT WAS TO ACHIEVE GREATER EFFICIENCY AND SYNERGY BETWEEN DISCOVERY AND OUTREACH EFFORTS. ONE ANCILLARY BENEFIT OFTHIS ALIGNMENT IS THAT BASIC AND APPLIED RESEARCH WILL LEAD TO RAPID EXTENSION PROGRAMMING AND APPLICATION IN THE FIELD. THIS ALIGNMENT ALSO ALLOWS MORE FLUID COMPLIANCE WITH MULTI-STATE AND INTEGRATED COMPONENTS OF AREERA. THESE GRAND CHALLENGES WERE DETERMINED THROUGH A SERIES OF MEETINGS WITH STAKEHOLDERS AND DEFINE AREAS OF EMPHASIS FOR AGENTS, SPECIALISTS, AND RESEARCHERS. EXTENSION PROFESSIONALS AT THE LOCAL, REGIONAL AND STATE LEVEL WORK TO DEVELOP REGIONAL AND STATEWIDE SIGNATURE PROGRAMS ALONG WITH PROFESSIONAL DEVELOPMENT OPPORTUNITIES FOR ITS MEMBERS THROUGH 11 PROGRAM FOCUS TEAMS - ADULT DEVELOPMENT AND AGING; COMMUNITY VITALITY; CROP PRODUCTION; FAMILY AND CHILD DEVELOPMENT; FAMILY RESOURCE MANAGEMENT; FARM MANAGEMENT; HORTICULTURE; LIVESTOCK PRODUCTION; NATURAL RESOURCES; NUTRITION, FOOD SAFETY AND HEALTH; AND YOUTH DEVELOPMENT. CONSOLIDATED BIOMEDICAL CORE FACILITIES SUPPORTING A CENTER ON EMERGING AND ZOONOTIC INFECTIOUS DISEASE RESEARCH - SUMMARY: KANSAS STATE UNIVERSITY (KSU) IS WELL-POSITIONED TO BECOME THE PREEMINENT INSTITUTION TO ADVANCE THE DISCOVERY AND DEVELOPMENT OF BIOSECURITY STRATEGIES FOR EMERGING AND ZOONOTIC INFECTIOUS DISEASES. WITH KSU’S BIOSECURITY RESEARCH INSTITUTE (BSL-3), USDA’S ARTHROPOD-BORNE ANIMAL DISEASE RESEARCH UNIT, AND USDA’S NATIONAL BIO AND AGRO-BIODEFENSE FACILITY (BSL-3/BSL-4), KSU IS THE ONLY U.S. UNIVERSITY WITH A FULL CONTINUUM OF BIOSECURITY LEVEL 1 (BSL-1) THROUGH BSL-4 FACILITIES COLLOCATED ON ONE CAMPUS. THE PROPOSED CORE RESEARCH FACILITY REPRESENTS A CRITICAL COMPONENT OF OUR RESEARCH INFRASTRUCTURE TO SUPPORT INFECTIOUS DISEASE STUDIES AND PROVIDE DIRECT SUPPORT OF BOTH KSU’S 2020 COBRE CENTER AND COLLOCATED FEDERAL FACILITIES. THE CORE FACILITY WILL STRATEGICALLY COMBINE FIVE KEY DISCIPLINES: ANIMAL MODEL/PATHOLOGY, MOLECULAR AND CELLULAR BIOLOGY, MICROSCOPIC IMAGING, FLOW CYTOMETRY AND CELL SORTING, AND NEXT-GENERATION SEQUENCING. INVESTIGATORS AT KSU AND IN THE REGION RELY ON THESE TECHNOLOGIES TO PERFORM HIGH-IMPACT RESEARCH AND COMPLETE EXPERIMENTS OUTLINED IN CURRENT AND FUTURE RESEARCH PROJECTS. HOWEVER, EXISTING FACILITIES FOR THESE KEY TECHNOLOGIES ARE INADEQUATE IN SIZE, LOCATION, FUNCTION, AND FLEXIBILITY. EXISTING LABORATORIES ARE ISOLATED FROM EACH OTHER, SPREAD ACROSS THREE BUILDINGS, AND IN SOME CASES, ARE HOSTED BY INDIVIDUAL FACULTY MEMBERS, WHICH LIMITS ACCESS AND RESEARCH PRODUCTIVITY. THE PROPOSED CORE-FACILITY SUITE IS THE FINAL ELEMENT OF A THREE-PHASE RENOVATION OF THE KSU VETERINARY COMPLEX. PHASE 1 DELIVERED A CONTEMPORARY 220-SEAT AUDITORIUM (8,200 GSF) ADJACENT TO MOSIER HALL. PHASE 2 REPLACED AN OUTDATED AUDITORIUM WITH A PRIMARY-CARE CLINICAL TRAINING FACILITY OCCUPYING THE 1ST FLOOR OF MOSIER HALL. PHASE 3, THE FOCUS OF THIS APPLICATION, PROPOSES TO BUILD A COLLABORATIVE, UNIVERSITY-WIDE CORE RESEARCH FACILITY OCCUPYING THE 2ND FLOOR OF THE DECONSTRUCTED AUDITORIUM SPACE. CURRENTLY, THE 2ND FLOOR IS 5,000 GSF OF LABORATORY-CONDITIONED SHELL SPACE. THIS PROPOSAL WILL STRENGTHEN OUR RESEARCH CAPACITY AND INFRASTRUCTURE BY CREATING A CENTRALLY ORGANIZED, INTEGRATED TECHNOLOGY PIPELINE, WITH PROXIMATE EXPERT ASSISTANCE AND TRAINING SUPPORT TO FACILITATE EFFICIENT USE OF CONTEMPORARY TECHNOLOGY IN CONFOCAL MICROSCOPY, LIVE-CELL AND IN VIVO IMAGING, LASER CAPTURE MICRODISSECTION, FLOW CYTOMETRY, CELL SORTING, DNA/RNA SEQUENCING AND CRISPR TECHNOLOGY. THE PROPOSED PURPOSE-BUILT, CORE-FACILITY SUITE WILL ASSEMBLE STATE-OF-THE-ART TECHNOLOGIES IN A SINGLE LOCATION TO DELIVER EFFICIENT, COORDINATED SERVICES FOR ACADEMIC, CORPORATE AND FEDERAL RESEARCHERS IN IMAGING AND MOLECULAR ANALYSES, THEREBY PROVIDING A COMPLETE RANGE OF SERVICES FROM WHOLE TISSUES TO SINGLE-CELL NUCLEIC ACID ANALYSES. A DEDICATED MODERN BIOMEDICAL RESEARCH FACILITY WITH ADVANCED INSTRUMENTATION AND TECHNICAL SUPPORT WILL FOSTER COLLABORATIVE, SYNERGISTIC AND TRANSDISCIPLINARY SCIENCE. THIS IS CRITICAL TO PROMOTING A ROBUST RESEARCH AND TRAINING ENVIRONMENT WHERE RESEARCHERS CAN ANSWER THE MOST CHALLENGING AND URGENT BIOMEDICAL QUESTIONS OF OUR TIME.

FOR EXAMPLE, ANIMAL HEALTH R&D AND MANUFACTURING FIRMS FORM THE WELL-KNOWN ANIMAL HEALTH CORRIDOR, STRETCHING ALONG I-70 FROM MANHATTAN, KS IN THE WEST TO COLUMBIA, MO IN THE EAST. K-STATE'S BIOSECURITY RESEARCH INSTITUTE, A BIOSAFETY LEVEL 3 RESEARCH FACILITY IS AUTHORIZED TO WORK WITH PATHOGENIC AGENTS HAVING PANDEMIC AND BIOWEAPON POTENTIAL SUCH AS HIGHLY PATHOGENIC AVIAN INFLUENZA AND WITH ANIMAL PATHOGENS THAT CAN CAUSE BILLIONS OF DOLLARS IN ECONOMIC LOSSES, SUCH AS AFRICAN SWINE FEVER VIRUS. K-STATE RESEARCHERS WORKING AT THE BRI AND IN THE COLLEGE OF VETERINARY MEDICINE HAVE DEVELOPED POTENTIAL VACCINES AND THERAPEUTICS THAT HAVE BEEN LICENSED FOR COMMERCIAL DEVELOPMENT. TO BETTER CAPITALIZE ON THE INTELLECTUAL PROPERTY DEVELOPED BY OUR SCIENTISTS, K-STATE PLANS TO DEVELOP A PILOT-SCALE MANUFACTURING FACILITY THAT CAN BE USED BY K-STATE RESEARCHERS AS WELL AS CORPORATE AND PRIVATE PARTNERS TO DEVELOP PILOT-SCALE PRODUCTION OF THEIR DISCOVERIES. THIS IS THE FIRST STEP TOWARD COMMERCIAL-SCALE PRODUCTION. THE PROPOSED PHARMA PILOT PLANT (PPP) AT K-STATE ALSO WILL PROVIDE EARLY-STAGE DRUG DISCOVERY COMPANIES AS WELL AS LARGER ESTABLISHED PHARMA COMPANIES WITH ACCESS TO "SCALE-UP" CAPABILITIES AS THEY MOVE FROM BENCH-SCALE TO COMMERCIAL-SCALE PRODUCTION OF THEIR PHARMA PRODUCTS. SUCH A FACILITY, SOMETIMES CALLED A "BIOLOGICS DEVELOPMENT MODULE" OR BDM, IS AN ESSENTIAL STEP IN SCALING TOWARD FULL-SCALE BIOMANUFACTURING AND PRODUCTION OF VACCINES AND THERAPEUTICS. THE PHARMA PILOT PLANT (PPP) WILL HELP TO ATTRACT COMPANIES AND JOBS TO KANSAS, AS PART OF K-STATE'S BROADER SUITE OF BIODEFENSE, BIOSECURITY, AND BIOMANUFACTURING EFFORTS. THE FACIL ITY WILL GROW INDUSTRY-UNIVERSITY RESEARCH AND SUPPORT THE MANUFACTURING SCALE-UP OF DISCOVERIES AT K-STATE, NBAF, AND IN THE PRIVATE SECTOR. K-STATE PLANS TO RENOVATE AN EXISTING STRUCTURE, BUILT IN 1949, TO UPFIT IT INTO A BIOMANUFACTURING TRAINING FACILITY AND WORKING BIOMANUFACTURING FACILITY THAT CAN PRODUCE VACCINES AND OTHER BIOLOGICS FOR INDUSTRY. THE NEW FACILITY WILL MEET NIH REQUIREMENTS FOR BSL-2 CONTAINMENT AND BE DESIGNED AND ENGINEERED IN ACCORDANCE WITH ALL RELEVANT CODES, STANDARDS, AND GUIDES TO MEET USDA CLEAN ROOM PRODUCTION STANDARDS. THE FACILITY WILL BE MADE ADA ACCESSIBLE AND HAVE DEDICATED SPACE FOR PRODUCTION AND TRAINING. SUSTAINABLE DESIGN PRINCIPLES WILL BE FOLLOWED THROUGHOUT THE PROJECT.

AGRICULTURE IS RESPONSIBLE FOR DIRECT ECONOMIC IMPACTS OF $21.3B, $26.3B, AND $7.6B IN KANSAS, NEBRASKA, AND OKLAHOMA, RESPECTIVELY (USDA ERS DATA FOR 2021). THIS AGRICULTURAL REGION IS FACING INCREASED FREQUENCY AND SEVERITY OF DROUGHT DUE TO CLIMATE CHANGE; AS AN EXAMPLE, IN OCTOBER 2022, 100% OF NEBRASKA, 98.5% OF KANSAS, AND 100% OF OKLAHOMA EXPERIENCED DROUGHT CONDITIONS, ACCORDING TO THE UNIVERSITY OF NEBRASKA-LINCOLN: U.S. DROUGHT MONITOR. THE MICRA PROJECT BRINGS TOGETHER RESEARCHERS FROM KANSAS STATE UNIVERSITY (KSU), THE UNIVERSITY OF NEBRASKA-LINCOLN (UNL), AND LANGSTON UNIVERSITY (LU) WITH A WIDE RANGE OF EXPERTISE (E.G., ENGINEERING, SCIENCE, AGRICULTURAL SCIENCE, AND SOCIAL SCIENCE). THE PROJECT TEAM WILL CONDUCT RESEARCH WITH THE GOAL TO PRESERVE SOIL MOISTURE AND IMPROVE WATER QUALITY UNDER DROUGHT CONDITIONS. CORN IS THE TARGET CROP FOR STUDY DUE TO ITS ECONOMIC IMPORTANCE IN THE MIDWEST, AND ITS SUBSTANTIAL DEMAND FOR WATER AND NUTRIENTS. RESEARCHERS WILL INVESTIGATE THE IMPACTS OF ADDING AMENDMENTS TO IMPROVE THE SOIL (I.E., SOIL WETTING BACTERIA, BACILLUS SUBTILIS, AND BIOCHAR); UNDERSTAND PLANTS? BEHAVIOR IN RESPONSE TO DROUGHT; UNDERSTAND THE IMPACTS OF CLIMATE CHANGE ON AGRICULTURAL WORKERS; AND DETERMINE FARMERS' WILLINGNESS TO ADOPT NEW IRRIGATION PRACTICES RELATIVE TO DIFFERING WATER RIGHTS AND POLICIES. THE MICRA PROJECT WILL INVESTIGATE THE IMPACTS OF SOIL AMENDMENTS AT THE LAB-SCALE, GREENHOUSE-SCALE, AND FIELD-SCALE, AND WILL CONDUCT SURVEYS AND FOCUS GROUPS. THE MICRA TEAM WILL RECRUIT STUDENTS FROM HISTORICALLY EXCLUDED GROUPS; MENTOR AND TRAIN UNDERGRADUATE AND GRADUATE STUDENTS TO CONDUCT INTERDISCIPLINARY RESEARCH; TEACH INTERDISCIPLINARY COURSES AT THE GRADUATE LEVEL; SUPPORT EARLY CAREER FACULTY AT KSU, UNL, AND LU; AND CONDUCT EDUCATIONAL OUTREACH TO RURAL K-12 SCIENCE TEACHERS. THE MICRA TEAM WILL GENERATE FUNDAMENTAL KNOWLEDGE REGARDING THE IMPACTS OF SOIL AMENDMENTS (I.E., SOIL WETTING BACTERIA, B. SUBTILIS, AND BIOCHAR) TO INCREASE THE WATER HOLDING CAPACITY OF SOIL, THEREBY IMPROVING AGROECOSYSTEM PRODUCTIVITY AS WATER BECOMES MORE LIMITED DUE TO CLIMATE-CHANGE-DRIVEN DROUGHTS. INTERDISCIPLINARY RESEARCH IS FOCUSED IN THREE RESEARCH THRUSTS. RESEARCH THRUST 1 WILL TEST THE HYPOTHESIS THAT SPATIAL AND TEMPORAL SOIL WETTING BEHAVIOR OF B. SUBTILIS IS DRIVEN BY SOIL TYPE, INITIAL SOIL MOISTURE LEVELS, SOIL HYDROPHOBICITY, AND ADMINISTRATION METHODS, THEREBY GENERATING NEW KNOWLEDGE REGARDING SOIL WETTING, EVAPOTRANSPIRATION, AND DEVELOPMENT OF PREDICTIVE, GENOME-SCALE METABOLIC MODELS FOR SOIL-MICROBIAL INTERACTIONS. THE CENTRAL HYPOTHESIS OF THRUST 2 IS THAT B. SUBTILIS-MEDIATED SURFACTANT PRODUCTION (I.E., WHICH CHANGES THE SURFACE TENSION OF WATER), WHEN COMBINED WITH BIOCHAR-MEDIATED SOIL AGGREGATION, CAN SYNERGISTICALLY IMPROVE SOIL WATER RETENTION. THRUST 2 WILL DETERMINE THE IMPACTS OF SOIL WETTING BACTERIA AND BIOCHAR ON WATER-HOLDING CAPACITY, NUTRIENT TRANSFORMATION, AND THE RHIZOBIOME UNDER DEFICIT IRRIGATION. THRUST 3 FOCUSES ON THE HYPOTHESES THAT PRODUCERS (I.E., FARMERS) HAVING LOWER QUALITY SOILS (E.G., HIGH SAND CONTENT) WILL BE MORE WILLING TO ADOPT NEW PRECISION IRRIGATION TECHNOLOGIES AND PRACTICES, PARTICULARLY IN REGIONS WHERE CURTAILMENTS TO HISTORICAL WATER USE AMOUNTS HAVE BEEN PUT IN PLACE. FOCUS GROUP AND SURVEY DATA WILL YIELD FUNDAMENTAL INSIGHTS REGARDING IRRIGATOR?S PERCEPTIONS REGARDING SOIL AMENDMENTS AND IRRIGATION. THE SAME REGION ALSO ANNUALLY GENERATES MORE THAN 80% OF THE COUNTRY?S TOTAL LIVESTOCK WASTES (DAIRY, BEEF, SWINE), RAISING CONCERNS ABOUT THE IMPACT ON WATER QUALITY AND LIVING CONDITIONS. ABOUT 12? 35% OF THE WATER USED ANNUALLY FOR PRODUCTION-INTENSIVE AGRICULTURE IN KANSAS, NEBRASKA, AND OKLAHOMA COMBINED (8 M ACRE-FT/YR) CAN POTENTIALLY BE DERIVED BY RECOVERING AND TREATING WATER CONTAINED IN LIVESTOCK WASTE. A TEAM OF INTERDISCIPLINARY RESEARCHERS FROM KANSAS STATE UNIVERSITY (KSU), SEWARD COUNTY COMMUNITY COLLEGE (SCCC), OKLAHOMA STATE UNIVERSITY (OSU), AND THE UNIVERSITY OF NEBRASKA-LINCOLN (UNL), REPRESENTING THREE CONTIGUOUS EPSCOR JURISDICTIONS, WILL SYNERGIZE THEIR COMPLEMENTARY RESEARCH CAPACITIES TO ENABLE ADOPTION OF CIRCULAR WASTE RESOURCE RECOVERY AND WATER REUSE TECHNOLOGY PLATFORMS. THIS RESEARCH WILL ENHANCE ECONOMIC RESILIENCY, ENVIRONMENTAL SUSTAINABILITY, AND QUALITY OF LIFE IN GREAT PLAINS MICROPOLITAN COMMUNITIES. THE OVERALL PROJECT OBJECTIVE IS TO BUILD REGIONAL RESEARCH CAPACITY AND DEVELOP AN ECONOMICALLY VIABLE, SOCIALLY ACCEPTED, AND EFFICIENT CIRCULAR RESOURCE RECOVERY PLATFORM INTEGRATED WITH WATER REUSE FROM LIVESTOCK WASTES THAT ARE COPIOUSLY GENERATED IN THE REGION. THE PROPOSED WORK WOULD BUILD CAPACITY FOR USE-INSPIRED RESEARCH TO BE DEMONSTRATED FOR ADOPTION BY LIVESTOCK OPERATIONS IN SOUTHWEST KANSAS FIRST (LIBERAL, KS), IN COLLABORATION WITH SCCC, AND WITH REGIONWIDE ADOPTION POTENTIAL. THE PROJECT WILL INTEGRATE A WIDE ARRAY OF WORKFORCE DEVELOPMENT ACTIVITIES SUCH AS AN EARLY-CAREER FACULTY DEVELOPMENT PROGRAM AND TECHNICAL SKILLS TRAINING THROUGH EXCHANGE SITE VISITS. WORKFORCE DEVELOPMENT INITIATIVES WILL BE GUIDED BY AN INDUSTRY-GOVERNMENT ADVISORY COUNCIL COMPOSED OF LIVESTOCK AND AGRICULTURAL PRODUCERS, LOCAL ASSOCIATIONS AND COUNCILS, AND GOVERNMENT/POLICY REPRESENTATIVES. DURING THIS PROJECT, CRITICALLY IMPORTANT AND COMPLEX CONCEPTS SUCH AS RESOURCE RECOVERY WILL BE INTRODUCED TO PARTICIPATING STUDENTS AND THE PUBLIC THROUGH SCIENCE CAF?S, SUMMER RESEARCH FIELD EXPERIENCES, AND INTERACTIONS WITH PUBLIC UTILITIES TO REALIZE THE RESEARCH ADVANCES AT SCALE. THIS WILL ENABLE A HOLISTIC FRAMEWORK AND ENCOURAGE INCORPORATION OF THE CIRCULAR RESOURCE RECOVERY AND REUSE SYSTEMS INTO THE RURAL COMMUNITIES AND WORKFORCE. THIS COLLABORATIVE RESEARCH TEAM SEEKS TO ACHIEVE OPTIMAL CIRCULAR WASTE RESOURCE RECOVERY AND WATER REUSE TECHNOLOGY PLATFORMS THROUGH THREE INTERCONNECTED RESEARCH THRUSTS. RESEARCH THRUST 1 AIMS TO DEVELOP THE ANAEROBIC SEQUENCING BATCH REACTOR (ASBR), ANAEROBIC MEMBRANE BIOREACTOR (ANMBR), AND MICROBIAL ELECTROCHEMICAL CELL (MXC) PLATFORMS FOR HOLISTIC RECOVERY OF SWINE MANURE CO-DIGESTED WITH FATS, OILS, AND GREASE (FOG) TO PRODUCE METHANE OR ORGANIC ACIDS, HYDROGEN PEROXIDE, NUTRIENTS (N AND P) AS TUNABLE-RELEASE INORGANIC FERTILIZERS (OCTACALCIUM PHOSPHATE AND STRUVITE), AND TREATED WATER FOR REUSE. SUCH GROUNDBREAKING ADVANCEMENTS IN MEMBRANE SCIENCE WILL BE GUIDED BY ARTIFICIAL INTELLIGENCE/MACHINE LEARNING. RESEARCH THRUST 2 FOCUSES ON CIRCULAR WATER REUSE BY COMBINING ADVANCED OXIDATION AND MEMBRANE-BASED PROCESSES, INCLUDING USING WASTE-DERIVED HYDROGEN PEROXIDE TO PRODUCE HIGH-QUALITY WATER. SPECIFIC FOCUS WILL BE PLACED ON MITIGATING ANTIMICROBIAL RESISTANCE, A PREVALENT AND UNDERSTUDIED ISSUE IN RURAL WATER SUPPLIES. RESEARCH THRUST 3 WILL INTEGRATE TECHNO-ECONOMIC AND RISK SIMULATION WITH AGRIBUSINESS DECISION NODE MODELING FOR REGION-SPECIFIC ADOPTION OF THE CIRCULAR SYSTEMS. HUMAN DIMENSIONS, INCLUDING CULTURAL PERCEPTIONS, ASSESSMENTS OF SAFETY AND SECURITY RISKS, AND SOCIAL-ECONOMIC IMPACTS OF THE PROPOSED TECHNOLOGIES, WILL BE ANALYZED FROM REPRESENTATIVE COMMUNITIES. COLLECTIVE RESEARCH CAPACITY FROM THE CONTIGUOUS JURISDICTIONS WILL BE SYNERGIZED AND VERIFIED THROUGH A FIELD DEMONSTRATION OF THE ANMBR + ADVANCED OXIDATION UNIT AT LIBERAL, KS, IN YEAR 4 OF THE PROJECT. NEW AVENUES FOR CROSS-CONVERGENT RESEARCH BETWEEN APPLIED AND PURE SCIENCE-BASED RESEARCHERS AS WELL AS POTENTIAL MANUFACTURING AND INDUSTRY PARTNERS WILL BE ACHIEVED THROUGHOUT THIS PROPOSAL. SYNERGISTIC RESEARCH THAT CO-ADDRESSES ENGINEERING GRAND CHALLENGES AND SOCIETY-BASED SUSTAINABLE DEVELOPMENT GOALS, SUCH AS RESPONSIBLE CONSUMPTION AND PRODUCTION, CLEAN WATER AND SANITATION, WILL ALSO BE DEMONSTRATED. THIS PROJECT IS SUPPORTED BY THE EPSCOR RESEARCH INFRASTRUCTURE IMPROVEMENT PROGRAM: FOCUSED EPSCOR COLLABORATIONS PROGRAM (FEC). FEC SUPPORTS INTERJURISDICTIONAL TEAMS OF EPSCOR INVESTIGATORS TO PERFORM RESEARCH IN TOPICS THAT ALIGN WITH NSF PRIORITIES, WITH THE GOALS OF DRIVING DISCOVERY AND BUILDING SUSTAINABLE STEM CAPACITY. NOVEL ENZYME INHIBITORS IN THE INNATE IMMUNE EVASION REPERTOIRE OF STAPHYLOCOCCI - PROJECT ABSTRACT THE INNATE IMMUNE SYSTEM IS BEST KNOWN AS AN ESSENTIAL DEFENSE AGAINST INVADING MICROBES. IN HUMANS, THIS RESPONSE REQUIRES AN IMMEDIATE AND CONCERTED ACTION BY BOTH HUMORAL AND CELLULAR COMPONENTS, WHICH ARE REPRESENTED BY THE COMPLEMENT SYSTEM AND NEUTROPHILS, RESPECTIVELY. EFFICIENT KILLING OF MICROBES BY THIS SO- CALLED “COMPLEMENT/NEUTROPHIL AXIS” IS PREDICATED UPON A HIGHLY ORCHESTRATED AND STEPWISE SERIES OF MOLECULAR RECOGNITION EVENTS AND BIOCHEMICAL TRANSFORMATIONS, WHICH AT THEIR MOST FUNDAMENTAL LEVEL INVOLVE ENZYMES. AS A CONSEQUENCE OF HOST/PATHOGEN CO-EVOLUTION, THE GRAM-POSITIVE BACTERIUM STAPHYLOCOCCUS AUREUS HAS DEVELOPED A POWERFUL ARRAY OF SMALL PROTEIN INHIBITORS THAT BLOCK MANY OF THE CENTRAL ENZYMATIC PLAYERS OF THE INNATE IMMUNE RESPONSE. IN THIS REGARD, WE IDENTIFIED THREE SECRETED STAPHYLOCOCCAL PROTEINS, CALLED EAP, EAPH1, AND EAPH2 (DENOTED “EAP PROTEINS”), WHICH POTENTLY INHIBIT THREE DIFFERENT PROTEASES KNOWN AS NEUTROPHIL SERINE PROTEASES (NSPS) THAT ARE CRITICAL COMPONENTS OF THE NEUTROPHIL'S ANTI-BACTERIAL ARSENAL. IN ADDITION TO THIS, EAP ITSELF ALSO INHIBITS ASSEMBLY OF A MULTI-SUBUNIT PROTEASE SYSTEM THAT IS REQUIRED FOR FUNCTION OF THE CLASSICAL AND LECTIN COMPLEMENT PATHWAYS. SEPARATELY, WE ALSO IDENTIFIED A NEW STAPHYLOCOCCAL PROTEIN, CALLED “SPIN”, THAT IS A POTENT INHIBITOR OF THE HOCL-GENERATING MYELOPEROXIDASE (MPO) FOUND IN NEUTROPHILS. COLLECTIVELY, THESE S. AUREUS PROTEINS INTERFERE WITH BACTERIAL KILLING IN BOTH IN VITRO SYSTEMS AND ANIMAL MODELS. WHILE OUR INITIAL STUDIES ON EAP PROTEINS AND SPIN HAVE PROVIDED IMPORTANT INFORMATION ON THE STRUCTURE, FUNCTION, AND MECHANISM OF THESE NOVEL ENZYME INHIBITORS, MANY SIGNIFICANT QUESTIONS STILL REMAIN. IN THIS PROJECT, WE WILL EMPLOY A COMBINATION OF STRUCTURAL, BIOCHEMICAL, FUNCTIONAL, AND INFORMATICS APPROACHES TO ADDRESS THESE ISSUES. IN THE FIRST SERIES OF INVESTIGATIONS, WE WILL DETERMINE HOW THE INDIVIDUAL REPEATING DOMAINS OF S. AUREUS EAP INHIBIT NSPS. THIS WILL PROVIDE A MEANS FOR COMPARATIVE ANALYSIS TO THE EAP HOMOLOGS, EAPH1 AND EAPH2, WHICH ARE MORE EXTENSIVELY CHARACTERIZED. WE WILL ALSO WORK TO DEFINE THE STRUCTURAL DETERMINANTS WITHIN EAP THAT ALLOW THIS PROTEIN, BUT NOT EAPH1 OR EAPH2, TO INHIBIT THE COMPLEMENT SYSTEM IN ADDITION TO NSPS. IN THE SECOND SERIES OF INVESTIGATIONS, WE WILL EXPLORE THE STRUCTURAL TRANSITIONS THAT ALLOW SPIN TO ADOPT AN INHIBITORY CONFORMATION UPON BINDING TO MPO. WE WILL ALSO DEFINE THE STRUCTURAL DETERMINANTS WITHIN SPIN PROTEINS THAT PROVIDE AN EXQUISITE LEVEL OF SELECTIVITY FOR MPO WHEN COMPARED TO CLOSELY RELATED HEME PEROXIDASES. IN OUR FINAL SERIES OF STUDIES, WE WILL LEVERAGE OUR EXTENSIVE STRUCTURAL AND FUNCTIONAL DATA ON SPINS AND STAPHYLOCOCCAL COMPLEMENT INHIBITORS TOWARD ESTABLISHING A PARADIGM FOR UNDERSTANDING THE PHYSICAL BASIS FOR HOST SPECIES SPECIFICITY OF VIRULENCE PROTEINS. BY COMPLETING THIS RESEARCH PLAN, WE WILL FURTHER OUR UNDERSTANDING OF TWO NOVEL CLASSES OF ENZYME INHIBITORS THAT FUNCTION AT THE HOST/PATHOGEN INTERFACE, AND LAY THE BASIC SCIENCE FOUNDATION FOR FUTURE DEVELOPMENT OF ANTI-BACTERIAL AND ANTI-INFLAMMATORY THERAPIES ARISING FROM THE INFORMATION WE UNCOVER.

SIGNALING PATHWAYS REGULATING MECHANOREFLEX SENSITIZATION IN CARDIOVASCULAR DISEASE - PROJECT SUMMARY HALLMARK FEATURES OF MANY FORMS OF CARDIOVASCULAR DISEASE INCLUDE EXERCISE INTOLERANCE AND A HIGH RISK OF SUFFERING AN ADVERSE ISCHEMIC CARDIOVASCULAR EVENT SUCH AS CARDIAC FIBRILLATION, MYOCARDIAL INFARCTION, OR STROKE DURING PHYSICAL EXERTION. AN EXAGGERATED INCREASE IN SYMPATHETIC NERVOUS SYSTEM ACTIVITY DURING EXERCISE (I.E., SYMPATHOEXCITATION) IS A DIRECT CONTRIBUTOR TO EXERCISE INTOLERANCE AND THE RISK OF SUCH ADVERSE EVENTS. DURING EXERCISE, MECHANICAL FORCES ASSOCIATED WITH CONTRACTING SKELETAL MUSCLES STIMULATE SMALL DIAMETER MUSCLE SENSORY NEURONS (GROUP III/IV MUSCLE AFFERENTS) WHICH PLAYS AN INTEGRAL ROLE IN THE ACTIVATION OF A REFLEX, THE EXERCISE PRESSOR REFLEX, THAT CONTRIBUTES TO INCREASED SYMPATHETIC NERVOUS SYSTEM ACTIVITY DURING EXERCISE. THE GOAL OF THIS PROJECT IS TO INVESTIGATE THE CELLULAR SIGNALING MECHANISMS THAT RESULT IN EXAGGERATED MECHANICALLY- ACTIVATED EXERCISE PRESSOR REFLEX SYMPATHETIC CONTROL SIGNALS IN CARDIOVASCULAR DISEASE. SPECIFICALLY, WE WILL USE A RAT MODEL OF LIMB ISCHEMIA IN WHICH A FEMORAL ARTERY IS CHRONICALLY LIGATED (A MODEL OF SIMULATED PERIPHERAL ARTERY DISEASE) AND A COMPLEMENTARY BLEND OF MOLECULAR AND WHOLE-ANIMAL APPROACHES TO INVESTIGATE THE SIGNALING PATHWAYS WITHIN MUSCLE SENSORY NEURONS THAT RESULT IN A PATHOPHYSIOLOGICAL ENHANCEMENT OF MECHANICALLY-ACTIVATED CHANNEL FUNCTION. THE INCORPORATION OF MULTIPLE EXPERIMENTAL TECHNIQUES INCLUDING MOLECULAR, ELECTROPHYSIOLOGY, REFLEX, AND CONSCIOUS EXERCISE EXPERIMENTS WILL ENSURE THAT OUR FINDINGS ARE ROBUST, INTEGRATIVE, AND TRANSLATIONAL IN NATURE. IN AIM 1, WE WILL INVESTIGATE THE ROLE PLAYED BY INOSITOL 1,4,5-TRISPHOSPHATE (IP3) RECEPTORS AND ALTERED CALCIUM SIGNALING IN GROUP III/IV MUSCLE AFFERENTS IN THE EXAGGERATED MECHANICAL COMPONENT OF THE EXERCISE PRESSOR REFLEX (I.E., THE MECHANOREFLEX) IN RATS WITH CHRONIC LIMB ISCHEMIA. IN AIM 2, WE WILL INVESTIGATE THE ROLE PLAYED BY EXCHANGE PROTEIN ACTIVATED BY CAMP (EPAC), ESPECIALLY EPAC 1, SIGNALING IN GROUP III/IV MUSCLE AFFERENTS IN THE EXAGGERATED MECHANOREFLEX IN RATS WITH CHRONIC LIMB ISCHEMIA. IN AIM 3, WE WILL INVESTIGATE THE ROLE PLAYED BY PROTEIN KINASE C (PKC), ESPECIALLY PKCE, SIGNALING IN GROUP III/IV MUSCLE AFFERENTS IN THE EXAGGERATED MECHANOREFLEX IN RATS WITH CHRONIC LIMB ISCHEMIA. THIS PROJECT IS INNOVATIVE BECAUSE IT IS THE FIRST TO SYSTEMATICALLY INVESTIGATE THE SIGNALING MECHANISMS THAT MODULATE THE FUNCTION OF MECHANICALLY ACTIVATED CHANNELS THAT CONTRIBUTE TO SYMPATHOEXCITATION DURING EXERCISE IN CARDIOVASCULAR DISEASE. THIS PROJECT IS SIGNIFICANT BECAUSE THE EXPERIMENTS MAY IDENTITY THREE NOVEL TARGETS (IP3 RECEPTORS, EPAC1, AND PKCE) FOR THERAPIES AIMED AT MITIGATING SYMPATHOEXCITATION, EXERCISE INTOLERANCE, AND/OR ELEVATED RISK OF ISCHEMIC EVENTS IN CARDIOVASCULAR DISEASE PATIENTS.

FUNCTION OF HEMOLYMPH PLASMA PROTEINS IN INSECT IMMUNE RESPONSES - INSECTS HAVE AN INNATE IMMUNE SYSTEM THAT PROTECTS AGAINST INFECTIONS BY PATHOGENS AND PARASITES. THE OVERALL GOAL OF THIS PROJECT IS TO INVESTIGATE PROTEINS IN INSECT HEMOLYMPH PLASMA THAT PARTICIPATE IN DIFFERENT ASPECTS OF INSECT IMMUNITY. PREVIOUS WORK FROM THIS LABORATORY HAS ESTABLISHED THE LEPIDOPTERAN INSECT, MANDUCA SEXTA, AS A MODEL SYSTEM WELL SUITED FOR BIOCHEMICAL CHARACTERIZATION OF THE COMPLEX FUNCTIONS OF HEMOLYMPH PROTEINS. WE PLAN TO INVESTIGATE THREE ASPECTS OF IMMUNITY THAT ARE MEDIATED BY PLASMA PROTEINS. (1) WE WILL CHARACTERIZE INTERACTIONS OF PLASMA PROTEINS WITH PEPTIDOGLYCAN FROM GRAM-NEGATIVE AND GRAM- POSITIVE BACTERIA THAT TRIGGER ACTIVATION OF A SERINE PROTEASE, THEREBY INITIATING PROTEASE CASCADES THAT RESULT IN ACTIVATION OF PHENOLOXIDASE AND THE TOLL PATHWAY. (2) WE WILL INVESTIGATE HOW THE REDOX ENVIRONMENT OF HEMOLYMPH IS REGULATED DURING AN IMMUNE RESPONSE AND TEST A HYPOTHESIS THAT IMMUNE PROTEINS MAY BE REGULATED BY THIOL MODIFICATIONS. (3) MAMMALS HAVE WELL-CHARACTERIZED MECHANISMS FOR DEPRIVING INVADING MICROORGANISMS OF IRON; HOWEVER, IMMUNE-RELATED IRON-WITHHOLDING IN INSECTS IS STILL POORLY UNDERSTOOD. WE WILL USE M. SEXTA AS A MODEL FOR DISCOVERING CONSERVED IRON-WITHHOLDING MECHANISMS IN INSECTS. THE FUNDAMENTAL BIOCHEMICAL KNOWLEDGE GAINED AS A RESULT OF THIS RESEARCH IS EXPECTED TO STIMULATE BETTER UNDERSTANDING OF REGULATION OF PROTEASE CASCADE PATHWAYS AND MOLECULES IN INSECT HEMOLYMPH THAT CONTRIBUTE TO OXIDATIVE MODULATION DURING IMMUNITY, AND LEAD TO UNDERSTANDING OF HOW IRON INFLUENCES HUMAN DISEASE TRANSMISSION BY INSECT VECTORS. THE ONGOING SPREAD OF HIGHLY PATHOGENIC AVIAN INFLUENZA (HPAI) H5N1 VIRUS POSES A SERIOUS THREAT TO THE POULTRY INDUSTRY, A VITAL COMPONENT OF U.S. AGRICULTURE. THE VIRUS ENDANGERS NOT ONLY POULTRY PRODUCTION, BUT ALSO FOOD SECURITY AND PUBLIC HEALTH. THUS, DEVELOPMENT OF SAFE, EFFECTIVE, SCALABLE, AND AFFORDABLE COUNTERMEASURES IS URGENTLY NEEDED TO PROTECT THIS CRITICAL SECTOR. WE HAVE DEVELOPED A NOVEL LIVE-ATTENUATED VACCINE PLATFORM BASED ON BOVINE PARAINFLUENZA VIRUS TYPE 3, GENOTYPE C (BPI3VC), ENGINEERED TO EXPRESS KEY PROTECTIVE ANTIGENS FROM H5N1. THE VECTOR DISPLAYS HEMAGGLUTININ (HA), NEURAMINIDASE (NA), AND MATRIX PROTEIN 2 ECTODOMAIN (M2E) ON THE SURFACE OF INFECTED CELLS, AND NUCLEOPROTEIN (NP) IN THE CYTOSOL. THE RECOMBINANT VACCINE, DESIGNATED BPI3VC-H5N1, REPLICATES EFFICIENTLY IN PRIMARY CHICKEN EMBRYO FIBROBLAST CELLS AND EXPRESSES THE TRANSGENES ROBUSTLY, AS CONFIRMED USING CONVALESCENT H5N1 SERA. SINCE BPI3VC IS NOT A CHICKEN PATHOGEN, IT ALLOWS FOR THE DEVELOPMENT OF A LATERAL FLOW DEVICE (LFD) CAPABLE OF DIFFERENTIATING INFECTED FROM VACCINATED ANIMALS (DIVA) BY DETECTING IMMUNE RESPONSES SPECIFIC TO BOTH THE VACCINE VECTOR AND H5N1 ANTIGENS. THIS PROJECT HAS TWO MAIN AIMS: 1) TO ASSESS THE SAFETY, IMMUNOGENICITY, AND EFFICACY OF THE BPI3VC-H5N1 VACCINE IN CHICKENS; AND 2) TO DEVELOP AND VALIDATE A COMPANION DIVA DIAGNOSTIC LFD. THE VACCINE IS READY FOR SCALE-UP IN READINESS FOR IN OVO VACCINATION, AND ADMINISTRATION TO DAY-OLD CHICKS AND PULLETS THROUGH SPRAY OR WATER. CHALLENGE STUDIES WITH CONTEMPORARY HPAI H5N1 WILL ASSESS PROTECTIVE EFFICACY, VIRAL SHEDDING, AND TRANSMISSION. IN PARALLEL, WE WILL DEVELOP AN LFD INCORPORATING THE BPI3VC FUSION (F) PROTEIN AND H5 HA ANTIGEN AS DUAL TEST LINES. THE DEVICE'S DIAGNOSTIC PERFORMANCE WILL BE VALIDATED USING STUDY AND FIELD SAMPLES. THE ANTI CIPATED OUTCOMES INCLUDE DELIVERY OF A H5N1 VACCINE FOR POULTRY AND A DIVA-COMPATIBLE LFD DIAGNOSTIC TOOL. THE VACCINE'S FLEXIBILITY FOR ADMINISTRATION IN OVO, VIA SPRAY, OR WATER, MAKES IT WELL SUITED FOR INTEGRATION INTO EXISTING HATCHERY AND FARM-BASED MASS VACCINATION SYSTEMS. EARLY IMMUNIZATION WILL PROVIDE PROTECTION TO BROILERS, LAYER CHICKS, AND PULLETS, ENSURING BROAD APPLICABILITY ACROSS PRODUCTION TYPES. IT DEVELOPS CUTTING-EDGE SENSORS THAT CAN DETECT NUTRIENTS, CHEMICAL COMPOUNDS, SOIL MICROBIOMES, AND GHGS. COLLECTING THESE DATA IN REAL TIME IS ESSENTIAL FOR ADVANCING PRECISION AGRICULTURE AND PROMOTING SUSTAINABLE PRACTICES. BY FOCUSING ON HOW SOIL, MICROBES, AND PLANTS INTERACT, INNOVATIVE TECHNOLOGIES ARE CONCEPTUALIZED AND GENERATED TO HELP FARMERS MANAGE THEIR CROPS MORE EFFECTIVELY AND SUSTAINABLY. THE TRANSDISCIPLINARY TEAM INCLUDES EXPERTS IN CHEMISTRY, COMPUTATIONAL BIOLOGY, OMICS, MICROBIOLOGY, AND METABOLIC ENGINEERING. THEY DEVELOP AND TRANSLATE TO THE AGRICULTURAL SECTOR NEW NITROGEN MANAGEMENT STRATEGIES AND GHG EMISSION MITIGATION TECHNOLOGIES. THE RESEARCH TAKES PLACE IN KEY AGRICULTURAL REGIONS WITH PRIMARY TEST SITES IN THE U.S. (CORN RESEARCH SITES IN KANSAS). THE MID-TERM GOAL IS TO EXTEND THIS EFFORT TO POTENTIAL PARTNER COUNTRIES, CANADA (WHEAT SITES IN QUEBEC) AND THE UK (WHEAT SITES IN SHEFFIELD), AND ULTIMATELY GLOBALLY. THIS PROJECT INTEGRATES RESEARCH, EDUCATION, STAKEHOLDER ENGAGEMENT AND OUTREACH. IT PROVIDES SUPPORT AND TRAINING TO UNDERGRADUATE AND GRADUATE STUDENTS, NOTABLY FROM UNDERREPRESENTED GROUPS IN STEM, AS WELL AS OUTREACH TOWARD K-12 STUDENTS, FARMERS AND PRODUCERS, AND INDUSTRY PARTNERS. THE RESEARCHERS FOCUS ON THE OVERALL UNDERSTANDING OF FERTILIZER, PLANT, AND SOIL INTERACTIONS. THEY PRIORITIZE THREE RESEARCH THRUSTS: (1) CREATING INNOVATIVE SENSORS TO RELIABLY SENSE AND MEASURE KEY NUTRIENTS (AMMONIUM AND NITRATE) AND GHG EMISSIONS (NITROUS OXIDE AND METHANE) BY ACTUATING PROCESSES WITHIN THE SOIL-MICROBE-PLANT-ENVIRONMENT CONTINUUM; (2) DESIGNING GREENHOUSE GAS (GHG) MODELING SYSTEMS AND ADVANCED MACHINE AND STATISTICAL LEARNING TECHNIQUES; (3) ESTABLISHING AND TESTING A BIOSENSOR NETWORK TO MONITOR PLANTS WITH BIOLOGICAL NITRIFICATION INHIBITORS (BNI) TO SUPPORT SUSTAINABLE AGRICULTURE. ADDITIONALLY, THEY ENGAGE IN CROSSCUTTING ACTIVITIES THAT INCLUDE (4) STAKEHOLDER ENGAGEMENT AND COLLABORATIVE RESEARCH INITIATIVES TO STRENGTHEN THE GLOBAL SCIENCE AND TECHNOLOGY COMMUNITY, AND (5) WORKFORCE DEVELOPMENT AND EDUCATIONAL PROGRAMS. WITH COLLABORATORS IN CANADA AND THE UK, THE TEAM DEVELOPS ALTERNATIVE NITROGEN FERTILIZERS INCLUDING BIOFERTILIZERS, BIOSTIMULANTS, AND FERTILIZERS DERIVED FROM WASTE RECOVERY. THIS PROJECT USES CRISPR-BASED GENOME EDITING TOOLS TO EXPRESS KEY HPAI ANTIGENS WITHIN THE TISSUES OF TWO INSECT SPECIES - THE RED FLOUR BEETLE TRIBOLIUM CASTANEUM (TCA), AND THE YELLOW MEALWORM TENEBRIO MOLITOR (TMO). EFFECTIVE RESPONSES TO DISEASES AND DAMAGING SPECIES HINGE ON UNDERSTANDING THEIR CAUSES AND IMPACTS. THE ROLE OF PRIOR KNOWLEDGE AND EVENT SEGMENTATION IN AGE- AND ALZHEIMER'S-RELATED CHANGES IN EVENT MEMORY - PROJECT SUMMARY THE HALLMARK SYMPTOM OF ALZHEIMER'S DISEASE (AD) AND MOST COMMON COMPLAINT AMONG COGNITIVELY HEALTHY OLDER ADULTS IS MEMORY LOSS, ESPECIALLY FOR EVERYDAY EVENTS SUCH AS CONVERSATIONS WITH A LOVED ONE, A MEAL WITH FRIENDS, A TRIP TO THE GROCERY STORE. HOWEVER, THERE IS A CRITICAL DIFFERENCE BETWEEN REMEMBERING THESE TYPES OF EVENTS AND THOSE THAT ARE OFTEN MEASURED IN THE LABORATORY OR IN NEUROPSYCHOLOGICAL BATTERIES, WHICH OFTEN LACK REAL-WORLD CONTEXTUAL MEANING. THIS GOAL IS HIGHLY RELEVANT TO THE NIH CORE MISSION “TO SEEK FUNDAMENTAL KNOWLEDGE ABOUT THE NATURE AND BEHAVIOR OF LIVING SYSTEMS AND THE APPLICATION OF THAT KNOWLEDGE TO ENHANCE HEALTH, LENGTHEN LIFE, AND REDUCE ILLNESS AND DISABILITY.” AIM 1 OF THIS PROJECT WILL DETERMINE WHETHER DEFICITS IN EVENT MEMORY ARE EXPLAINED BY AGE-RELATED DIFFERENCES IN MAINTAINING STABLE MENTAL REPRESENTATIONS DURING AN EXPERIENCE. AIM 2 WILL DETERMINE WHETHER OLDER ADULTS CAN EFFECTIVELY INTEGRATE NEW EVENT INFORMATION INTO EXISTING KNOWLEDGE STRUCTURES. AIM 3 WILL DETERMINE WHETHER CUEING PRIOR KNOWLEDGE IMPROVES EVENT MEMORY IN EARLY-STAGE AD. THE PROJECT WILL USE AN INNOVATIVE COMBINATION OF BEHAVIORAL AND NEUROPHYSIOLOGICAL MEASURES OF EVENT ENCODING TO ADDRESS THESE AIMS. IT WILL ALSO USE DYNAMIC ACTIVITIES THAT ARE OFTEN ENCOUNTERED IN DAILY LIFE—THE KIND THAT OLDER ADULTS REPORT HAVING DIFFICULTIES REMEMBERING. WE PREDICT THAT MPFC-MEDIATED PRIOR KNOWLEDGE WILL FACILITATE THE INTEGRATION OF NEW EVENT INFORMATION WITH PRIOR KNOWLEDGE RESULTING IN BETTER SEGMENTATION AND EVENT MEMORY FOR OLDER ADULTS. HOWEVER, WHEN NO PRIOR KNOWLEDGE IS AVAILABLE, THE EXTENT TO WHICH PEOPLE CAN EFFECTIVELY SEGMENT AN ACTIVITY (SUPPORTED BY MPFC-HIPPOCAMPAL COUPLING) WILL PREDICT MEMORY PERFORMANCE. FURTHER, WE PREDICT THAT CUEING RELEVANT KNOWLEDGE WILL SCAFFOLD THE LEARNING OF NEW SCHEMA-CONSISTENT EVENT INFORMATION IN COGNITIVELY HEALTHY OLDER ADULTS AND THOSE WITH EARLY AD. OUR GOAL OF IMPROVING OLDER ADULTS' ABILITY TO ENCODE AND RETRIEVE EVERYDAY ACTIVITIES IS ALIGNED WITH THE NIA'S VISION FOR OLDER ADULTS TO “ENJOY ROBUST HEALTH AND INDEPENDENCE, REMAIN PHYSICALLY AND MENTALLY ACTIVE, AND CONTINUE TO MAKE POSITIVE CONTRIBUTIONS TO THEIR FAMILIES AND COMMUNITIES.”

THE ARKANSAS STATE UNIVERSITY (A-STATE) POSITIVE BEHAVIORAL INTERVENTIONS AND SUPPORTS (PBIS) RESOURCE CENTER (THE CENTER) IN JONESBORO, ARKANSAS, PROPOSES PROJECT PEACE (PREVENTION, ENGAGEMENT, AND CONNECTION IN EDUCATION). THIS PROJECT WILL ADDRESS TWO FUNDING AREAS: (1) TRAIN SCHOOL PERSONNEL AND STUDENTS ON PREVENTING SCHOOL VIOLENCE. (2) PROVIDE SPECIALIZED TRAINING OR CREATE SPECIALIZED NON-TRAINING POLICIES FOR LAW ENFORCEMENT WHO WORK IN SCHOOLS AND/OR WITH SCHOOL-AGE POPULATIONS SUCH AS SCHOOL RESOURCE OFFICERS (SROS) AND PROBATION OFFICERS. THE BENEFICIARIES OF PROJECT PEACE ARE SEVEN ARKANSAS MIDDLE AND JUNIOR HIGH SCHOOLS LOCATED IN NORTHEAST ARKANSAS (JONESBORO AND BLYTHEVILLE), NORTHWEST ARKANSAS (FAYETTEVILLE), AND CENTRAL ARKANSAS (LITTLE ROCK). IN YEAR 1, FIVE SCHOOLS WILL PARTICIPATE: JONESBOROS ANNIE CAMP JR. HIGH, BLYTHEVILLE MIDDLE SCHOOL, FAYETTEVILLES RAMAY JR. HIGH, HOLT MIDDLE SCHOOL, AND JOHN L. COLBERT MIDDLE SCHOOL. IN YEAR 2, TWO MORE SCHOOLS WILL JOIN THE PROJECT: LISA ACADEMYS NORTH MIDDLE SCHOOL AND WEST MIDDLE SCHOOL IN LITTLE ROCK. ACTIVITIES INCLUDE TRAINING AND COACHING SCHOOL LEADERSHIP TEAMS AND SROS IN IMPLEMENTING PBIS, A MULTITIERED SYSTEMS OF SUPPORT (MTSS) FRAMEWORK; DEVELOPING SCHOOLWIDE ACTION PLANS TO IMPROVE SCHOOL CLIMATE; ENGAGING STUDENTS IN TOWN HALLS; TRAINING SCHOOL STAFF, FAMILIES, AND COMMUNITY GROUPS ON YOUTH MENTAL HEALTH FIRST AID (YMHFA) AND SUICIDE PREVENTION; AND BUILDING CAPACITY FOR THE CENTER TO PROVIDE MORE NEEDED TRAINING, SERVICES, AND COLLABORATIVE OPPORTUNITIES FOR SCHOOLS ACROSS ARKANSAS.

COLLABORATIVE RESEARCH: RESEARCH-PGR: PLANTTRANSFORM: THE GENETIC BASIS OF MAIZE REGENERATION AND APPLICATIONS TO PLANT TRANSFORMATION -GENETIC ENGINEERING OF PLANTS IS CRITICAL FOR CROP IMPROVEMENT AND REQUIRES REGENERATION OF WHOLE PLANTS FROM GENETICALLY MODIFIED CELLS. WHILE ALL PLANT SPECIES POSSESS GENETIC COMPONENTS TO FORM WHOLE PLANTS FROM SINGLE CELLS, MANY CROP PLANTS, INCLUDING MAIZE, HAVE A LIMITED ABILITY TO REGENERATE FROM CELLS THAT ARE SUBJECTED TO GENETIC MODIFICATION. MASTER GENES ARE HYPOTHESIZED TO BE INVOLVED IN THE PROCESS OF PLANT REGENERATION. THIS IS STRONGLY RELATED TO THE ABILITY OF CELL PROLIFERATION, AN INCREASE IN THE NUMBER OF CELLS AS A RESULT OF GROWING AND MULTIPLYING OF CELLS. IN THIS PROJECT, MAIZE GENES RESPONSIBLE FOR CONTROLLING CELL GROWTH AND MULTIPLICATION WILL BE IDENTIFIED USING A NOVEL HIGH-THROUGHPUT SCREENING APPROACH. VALIDATING KEY CANDIDATE GENES IN REGULATING MAIZE REGENERATION WILL BE CARRIED OUT THROUGH FURTHER TESTING. THE PRIMARY GOAL OF THIS PROJECT IS TO BETTER UNDERSTAND THE GENETIC BASIS OF PLANT REGENERATION FOR IMPROVED GENETIC ENGINEERING STRATEGIES. THIS PROJECT WILL PROVIDE NEW WAYS OF ANALYZING AND UNDERSTANDING PLANT GENETICS THROUGH THE USE OF NEW GENOME EDITING TECHNOLOGIES, LARGE AMOUNTS OF DATA, AND COMPUTATIONAL TOOLS. IT WILL ALSO OFFER TRAINING OPPORTUNITIES IN VARIOUS FIELDS SUCH AS PLANT GENETICS, GENOMICS, MOLECULAR BIOLOGY, GENOME EDITING, BIOINFORMATICS, AND MACHINE LEARNING. PLANT TRANSFORMATION AND REGENERATION ARE KEY PROCEDURES OF GENOME ENGINEERING. THE OVERARCHING GOAL OF THIS PROPOSAL IS TO IMPROVE OUR UNDERSTANDING OF THE GENETIC BASIS OF PLANT REGENERATION. AS EACH PLANT GENOME POSSESSES TOTIPOTENT COMPONENTS TO FORM AN ENTIRE PLANT BUT THE CAPABILITY TO REGENERATE IS AT LOW LEVELS IN MANY CROP SPECIES, IT IS HYPOTHESIZED THAT CONSERVED MASTER GENES FOR TOTIPOTENCY GOVERNING EMBRYOGENESIS EXIST AND THAT ADDITIONAL NON-CONSERVED GENES ACCOUNT FOR DIFFERING REGENERATION ABILITY, AND THESE GENES ARE ASSOCIATED WITH THE ABILITY OF CELL PROLIFERATION AND GROWTH. IN THIS PROJECT, MAIZE GENES CONTROLLING CELL PROLIFERATION AND GROWTH WILL BE IDENTIFIED BY CONDUCTING ANALYSIS OF TRANSCRIPTOMIC GENE NETWORKS, GENOMIC COMPARISONS, HIGH-THROUGHPUT CRISPR SCREENING, AND FUNCTIONAL VALIDATION OF INDIVIDUAL GENES. THE PROJECT WILL PROVIDE INNOVATIVE APPROACHES, MASSIVE DATA, AND COMPUTATIONAL TOOLS FOR INTEGRATIVE GENOMIC ANALYSIS AND DISCOVERIES. T THIS PROJECT IS JOINTLY FUNDED BY THE PLANT GENOME RESEARCH PROGRAM IN THE DIRECTORATE FOR BIOLOGICAL SCIENCES AND THE ESTABLISHED PROGRAM TO STIMULATE COMPETITIVE RESEARCH (EPSCOR).
