UNFORTUNATELY, MOST MALARIA VACCINES THAT ENTERED LATE-STAGE CLINICAL DEVELOPMENT, HAVE SHOWN MODERATE EFFICACY IN LOW AND MIDDLE INCOME COUNTRIES. THROUGH A NUMBER OF STUDIES VACCINE EFFICACY WAS SHOWN TO VARY FROM 20% TO 100% WHEN USED IN DIFFERENT COUNTRIES AND POPULATIONS. BETTER UNDERSTANDING OF FACTORS THAT INFLUENCE THIS VARIATION IS URGENTLY NEEDED. EVEN WITHIN A COUNTRY SIGNIFICANT DIFFERENCES IN VACCINE EFFICACY IN RURAL AND URBAN AREAS ARE PRESENT. THIS IMPLIES THAT EXPOSURE TO ENVIRONMENTAL FACTORS PLAYS A MAJOR ROLE BESIDES GENETIC DETERMINANTS. HOW AND TO WHAT EXTENT ENVIRONMENTAL EXPOSURES CAN INFLUENCE IMMUNE PROFILES AND IN TURN AFFECT RESPONSES TO VACCINES? THIS QUESTION WILL BE ADDRESSED IN THE CURRENT PROPOSAL. TECHNOLOGICAL ADVANCES IN “OMICS” PLATFORMS HAVE IMPROVED OUR ABILITY TO EXAMINE THE IMMUNE SYSTEM IN A MORE UNBIASED MANNER. SUCH PLATFORMS, INVOLVING TRANSCRIPTOMICS, ARE BEING INCREASINGLY APPLIED TO UNDERSTAND VACCINE RESPONSES, WITH PROMISING RESULTS. HOWEVER, FOR MALARIA VACCINES, NO HARMONIZED APPROACH TO INTERROGATE IMMUNOLOGICAL REACTIVITIES AND PATHWAYS ACROSS COHORTS HAS BEEN DEVELOPED DESPITE ITS PUBLIC HEALTH IMPORTANCE AND AVAILABILITY OF COHORTS ASSESSED FOR A CLINICALLY RELEVANT OUTCOME (PREVENTION OF INFECTION OR DISEASE). EVEN LESS HAS BEEN DONE REGARDING INTEGRATED “OMICS”, COMPARING POPULATIONS LIVING IN DIFFERENT GEOGRAPHICAL LOCATIONS AND THEIR RESPONSE TO VACCINES. MOREOVER, THERE ARE VERY LITTLE DATA THAT LINK ENVIRONMENTAL EXPOSURES TO IN DEPTH CHANGES IN THE IMMUNE SYSTEM AND IF AVAILABLE, THE STUDIES OFTEN ADDRESS ONE ENVIRONMENTAL FACTOR AT A TIME. WE PROPOSE TO ADDRESS THIS AND NOT ONLY ASSESS CORRELATIONS IN MALARIA VACCINE COHORTS, BUT ALSO TO ADDRESS THE MECHANISMS UNDERLYING VACCINE (HYPO)RESPONSIVENESS. WE WILL BUILD ON AVAILABLE KNOWLEDGE OF IMMUNOLOGICAL PROCESSES THAT CAN AFFECT MALARIA VACCINE RESPONSES, AND USE SAMPLES FROM THE COHORTS WHERE HIGH DIMENSIONAL CYTOMETRY AND RNASEQ AND ANTIBODY INTERROGATIONS WILL HELP TO REFINE AND ENRICH THE QUESTIONS REGARDING MALARIA VACCINE (HYPO)RESPONSIVENESS. IN ADDITION, BY USING HUMAN PRIMARY HEPATOCYTES INFECTED WITH P. FALCIPARUM, WE WILL BRING THE RESEARCH CLOSER TO MECHANISMS OF TISSUE-SPECIFIC RESPONSES AND TO EXTRAPOLATE PRE-ERYTHROCYTIC IMMUNITY TO MALARIA IN THE LIVER. THIS IS OF PARTICULAR IMPORTANCE FOR PRE- ERYTHROCYTIC VACCINES, WHERE TISSUE-RESIDENT RESPONSES CAN PLAY AN IMPORTANT ROLE. BY GENERATING DATA ON THE SAME INDIVIDUALS, DATA INTEGRATION APPROACHES FOR HIGH DIMENSIONAL MEDIATION ANALYSIS, WILL BE USED TO PINPOINT THE SPECIFIC IMMUNOLOGICAL PATHWAYS AND MECHANISMS THAT RESULT IN MALARIA VACCINE (HYPO) RESPONSIVENESS. THIS INFORMATION CAN BE USED TO IMPROVE MALARIA VACCINE EFFICACY AND ALSO TO IDENTIFY INDIVIDUALS WHO WILL NOT BENEFIT FROM THE VACCINE REGIMENS USED SO FAR. A TOTAL OF 98 SCHOLARS PURSUING A TRANSFER DEGREE IN BIOLOGY, CHEMISTRY, DATA SCIENCE, ENGINEERING, GEOLOGY, MATHEMATICS, PHYSICS, AND COMPUTER SCIENCE WILL RECEIVE SCHOLARSHIPS AVERAGING $6554 FOR UP TO TWO YEARS. SCHOLARS WILL RECEIVE FACULTY AND PEER MENTORING, AND THE PROJECT WILL BUILD STRONG SCHOLAR COHORTS THROUGH SCHOLAR SPECIFIC SECTIONS OF A FIRST YEAR EXPERIENCE COURSE, FIELD TRIPS TO RELEVANT INDUSTRY LABORATORIES, AND A SPEAKER SERIES. TUTORING, STEM SPECIFIC ADVISING. ADDITIONAL ACTIVITIES FOR SCHOLARS INCLUDE TUTORING, STEM SPECIFIC ADVISING, AND TRANSFER SUPPORT. THE OVERALL GOAL OF THIS TRACK 2 SCHOLARSHIPS IN STEM PROJECT IS TO INCREASE STEM DEGREE COMPLETION OF ACADEMICALLY TALENTED, LOW-INCOME UNDERGRADUATES WITH DEMONSTRATED FINANCIAL NEED. THERE IS A SIGNIFICANT NATIONAL NEED TO GROW THE STEM WORKFORCE AND NURTURE KEY TALENT THAT WILL ENSURE ECONOMIC COMPETITIVENESS AND PROVIDE DOMESTIC LEADERSHIP ACROSS CRITICAL SECTORS. THIS PROJECT DIRECTLY SPEAKS TO THIS NEED BY SUPPORTING STEM STUDENT SUCCESS, WHICH WILL STRENGTHEN THE WORKFORCE IN BIOLOGY, CHEMISTRY, DATA SCIENCE, ENGINEERING, GEOLOGY, MATHEMATICS, PHYSICS, COMPUTER SCIENCE AND OTHER KEY AREAS OF NEED. THE PROJECT WILL BE ASSESSED BY AN EXPERIENCED EVALUATOR WHO WILL COLLECT BOTH FORMATIVE AND SUMMATIVE DATA, AND THE DATA GENERATED WILL CONTRIBUTE TO THE KNOWLEDGE BASE REGARDING EFFECTIVE STRATEGIES TO SUPPORT TALENTED, LOW-INCOME STUDENTS IN STEM. THIS PROJECT IS FUNDED BY NSF?S SCHOLARSHIPS IN SCIENCE, TECHNOLOGY, ENGINEERING, AND MATHEMATICS PROGRAM, WHICH SEEKS TO INCREASE THE NUMBER OF ACADEMICALLY TALENTED, LOW-INCOME STUDENTS WITH DEMONSTRATED FINANCIAL NEED WHO EARN DEGREES IN STEM FIELDS. WITH CANCER SURVIVORS ESTIMATED AT 19 MILLION IN THE USA BY 2025, DOX-INDUCED CARDIOTOXICITY IS CONSIDERED TO BE PART OF THE “CARDIO-ONCOLOGY EPIDEMIC”. THE DEVELOPMENT OF NEW APPROACHES TO REDUCE CHEMOTHERAPY-INDUCED CARDIOTOXICITY IS ESSENTIAL. DEXRAZOXANE (DEX), IS FDA APPROVED FOR THE SPECIFIC INDICATION OF “DOXORUBICIN ADMINISTRATION IN WOMEN WITH METASTATIC BREAST CANCER WHO HAVE RECEIVED A CUMULATIVE DOXORUBICIN DOSE OF 300 MG/M2 AND WHO WILL CONTINUE TO RECEIVE DOXORUBICIN THERAPY TO MAINTAIN TUMOR CONTROL”. LIGHTSEED HAS ASSEMBLED A HIGHLY EXPERIENCED TEAM (ONCOLOGIST-CANCER BIOLOGIST (WITH PRIOR BIOTECHNOLOGY COMPANY EXPERTISE), CARDIO-ONCOLOGIST, HIGH THROUGHPUT LEAD DISCOVERY EXPERT, IMMUNE CANCER BIOLOGIST- MOUSE GENETICIST) IN ORDER TO IDENTIFY AND REPURPOSE FDA-APPROVED COMPOUNDS WITH “DUAL FUNCTION” (ENHANCE CANCER CELL KILLING BY DOX BUT PROTECTION FROM DOX CARDIOTOXICITY). THREE COMPOUNDS FROM THE FDA- APPROVED COMPOUND LIBRARY HAVE BEEN VALIDATED. WE ARE SEEKING ADDITIONAL “DUAL FUNCTION” COMPOUNDS FROM THE LIBRARIES. WOMEN WITH DENSE BREASTS ARE MORE LIKELY TO BE DIAGNOSED IN THE “INTERVAL” BETWEEN SCREENING EXAMS, CONTRIBUTING TO INCREASED BREAST CANCER MORTALITY. HOWEVER, CURRENT SCREENING GUIDELINES FOR AVERAGE-RISK WOMEN DO NOT TAKE IMAGING RISK MARKERS INTO ACCOUNT. WOMEN WITH VERY HIGH RISK OF BREAST CANCER, REGARDLESS OF MD, RECEIVE AN INTENSIFIED SCREENING SCHEDULE WITH SUPPLEMENTAL CONTRAST-ENHANCED MAGNETIC RESONANCE IMAGING (MRI). ON MRI, RADIOLOGISTS ASSESS FGT AS WELL AS BACKGROUND PARENCHYMAL ENHANCEMENT (BPE). BPE IS THE DEGREE TO WHICH HEALTHY FGT BRIGHTENS DUE TO THE PRESENCE OF CONTRAST, REFLECTING THE METABOLIC ACTIVITY WITHIN THE FGT. WE RECENTLY DEMONSTRATED THAT BPE ON MRI WAS ASSOCIATED WITH BREAST CANCER RISK INDEPENDENTLY OF THE AMOUNT OF FGT. OTHER STUDIES CORROBORATE OUR FINDINGS, SUPPORTING OUR PREMISE THAT BPE IS A NEW MARKER OF BREAST CANCER RISK. HOWEVER, DUE TO THE SELECT POPULATION RECEIVING MRI, ALTERNATIVE MEASUREMENT APPROACHES ARE NEEDED TO INCREASE THE IMPACT OF BPE FOR WOMEN WHO ARE NOT ABLE TO RECEIVE SCREENING MRI. CONTRAST-ENHANCED MAMMOGRAPHY (CEM) IS AN INCREASINGLY-USED BREAST SCREENING MODALITY THAT SUBSTANTIALLY IMPROVES BREAST CANCER DETECTION, COMPARABLE IN SENSITIVITY TO MRI. AFTER INJECTION OF A CONTRAST AGENT, TWO MAMMOGRAMS AT HIGH AND LOW ENERGIES ARE TAKEN IN QUICK SUCCESSION. MD IS ASSESSED ON THE LOW-ENERGY IMAGE AND BPE IS ASSESSED SUBJECTIVELY BY RADIOLOGISTS AS “MINIMAL”, “MILD”, “MODERATE”, OR “MARKED” ON A PROCESSED (“RECOMBINED”) IMAGE PER THE BREAST IMAGING – REPORTING AND DATA SYSTEM (BI-RADS). BI-RADS BPE MEASURED ON CEM HAS GOOD AGREEMENT WITH BI-RADS BPE MEASURED ON MRI. HOWEVER, SUBJECTIVE MEASUREMENT OF BPE VARIES BETWEEN READERS, PREVENTING REPRODUCIBLE RISK ASSESSMENT RESEARCH. AN OBJECTIVE, QUANTITATIVE MEASURE OF BPE ON CEM IS NEEDED TO RIGOROUSLY EVALUATE THE RELATIONSHIP BETWEEN INCREASED BPE AND BREAST CANCER RISK. TO ADDRESS THIS RESEARCH GAP, WE PROPOSE TO ASSESS THE ASSOCIATION BETWEEN CANCER AND A FULLY AUTOMATED MEASURE OF BPE ON CEM. AIM 1. EVALUATE BPE AS A REPRODUCIBLE MARKER OF BREAST CANCER RISK: WE WILL IMPLEMENT A FULLY AUTOMATED MEASURE OF QUANTITATIVE BPE ON 462 CEMS, EVALUATE THE AGREEMENT WITH THE GOLD STANDARD BI-RADS MEASURE, AND COMPARE WITH BPE ON MRI. THIS SMALL BUSINESS INNOVATION RESEARCH (SBIR) PHASE I PROJECT AIMS TO DEVELOP A HIGHLY SENSITIVE, MINIMALLY INVASIVE LABORATORY TEST TO DETECT EARLY-STAGE OVARIAN CANCERS (OC) BY ASSAYING RELEVANT BIOMARKERS ON CIRCULATING EXTRACELLULAR VESICLES (EVS) IN PATIENT PLASMA. EVS PROVIDE SIGNIFICANT ADVANTAGES IN SENSITIVITY AND PATIENT OUTCOMES OVER TESTS USING CIRCULATING TUMOR CELLS OR CELL-FREE TUMOR DNA, AS THEY ARE SECRETED BY LIVING CANCER CELLS WITHIN THE PRIMARY TUMOR. THIS PROJECT WILL UTILIZE A NOVEL TECHNOLOGY TO IDENTIFY KEY PROTEINS IN EVS FROM VARIOUS OC SUBTYPES, ENGINEERING AN ASSAY TO DIFFERENTIATE BETWEEN PATIENTS WITH MALIGNANT TUMORS AND THOSE WITHOUT CANCER WITH HIGH SPECIFICITY. THE SECOND OBJECTIVE IS TO EMPLOY A MACHINE LEARNING TOOL TO PERFORM DIAGNOSTIC CLASSIFICATION, PROVIDING ACTIONABLE INFORMATION TO CLINICIANS REGARDING THE PRESENCE OF A TUMOR. BRINGING A SAFE AND EFFECTIVE NEW BIOLOGICAL-BASED PRODUCT TO THE MARKET WILL LOWER BEEKEEPER?S ANNUAL SPENDING AS WELL AS TIME AND EFFORT TO KEEP THEIR COLONIES ALIVE BY LEAVING HONEY BEE COLONIES FREE FROM THE DEVASTATING DOWNSTREAM EFFECTS OF TRADITIONAL PESTICIDES. IN ADDITION, THE SOLUTION PAVES THE WAY FOR MORE BIOLOGICALS TO BE DEVELOPED AND COMMERCIALIZED THROUGH THE NOVEL SCIENTIFIC APPROACH AND TECHNOLOGICAL ADVANCEMENT PROPOSED HEREIN, ESSENTIALLY REDUCING THE PERCEIVED RISK. BASELINE HOST AND ENVIRONMENTAL FACTORS THAT IMPACT PRE-ERYTHROCYTIC MALARIA VACCINE (HYPO)RESPONSIVENESS IN ENDEMIC REGIONS - ABSTRACT AN EFFECTIVE VACCINE WOULD COMPLEMENT OTHER PUBLIC HEALTH MEASURES AND IS LIKELY ESSENTIAL FOR PUTTING AN END TO THE HIGH BURDEN OF MALARIA WORLDWIDE. UNFORTUNATELY, MOST MALARIA VACCINES THAT ENTERED LATE-STAGE CLINICAL DEVELOPMENT, HAVE SHOWN MODERATE EFFICACY IN LOW AND MIDDLE INCOME COUNTRIES. THROUGH A NUMBER OF STUDIES VACCINE EFFICACY WAS SHOWN TO VARY FROM 20% TO 100% WHEN USED IN DIFFERENT COUNTRIES AND POPULATIONS. BETTER UNDERSTANDING OF FACTORS THAT INFLUENCE THIS VARIATION IS URGENTLY NEEDED. EVEN WITHIN A COUNTRY SIGNIFICANT DIFFERENCES IN VACCINE EFFICACY IN RURAL AND URBAN AREAS ARE PRESENT. THIS IMPLIES THAT EXPOSURE TO ENVIRONMENTAL FACTORS PLAYS A MAJOR ROLE BESIDES GENETIC DETERMINANTS. HOW AND TO WHAT EXTENT ENVIRONMENTAL EXPOSURES CAN INFLUENCE IMMUNE PROFILES AND IN TURN AFFECT RESPONSES TO VACCINES? THIS QUESTION WILL BE ADDRESSED IN THE CURRENT PROPOSAL. TECHNOLOGICAL ADVANCES IN “OMICS” PLATFORMS HAVE IMPROVED OUR ABILITY TO EXAMINE THE IMMUNE SYSTEM IN A MORE UNBIASED MANNER. SUCH PLATFORMS, INVOLVING TRANSCRIPTOMICS, ARE BEING INCREASINGLY APPLIED TO UNDERSTAND VACCINE RESPONSES, WITH PROMISING RESULTS. HOWEVER, FOR MALARIA VACCINES, NO HARMONIZED APPROACH TO INTERROGATE IMMUNOLOGICAL REACTIVITIES AND PATHWAYS ACROSS COHORTS HAS BEEN DEVELOPED DESPITE ITS PUBLIC HEALTH IMPORTANCE AND AVAILABILITY OF COHORTS ASSESSED FOR A CLINICALLY RELEVANT OUTCOME (PREVENTION OF INFECTION OR DISEASE). EVEN LESS HAS BEEN DONE REGARDING INTEGRATED “OMICS”, COMPARING POPULATIONS LIVING IN DIFFERENT GEOGRAPHICAL LOCATIONS AND THEIR RESPONSE TO VACCINES. MOREOVER, THERE ARE VERY LITTLE DATA THAT LINK ENVIRONMENTAL EXPOSURES TO IN DEPTH CHANGES IN THE IMMUNE SYSTEM AND IF AVAILABLE, THE STUDIES OFTEN ADDRESS ONE ENVIRONMENTAL FACTOR AT A TIME. WE PROPOSE TO ADDRESS THIS AND NOT ONLY ASSESS CORRELATIONS IN MALARIA VACCINE COHORTS, BUT ALSO TO ADDRESS THE MECHANISMS UNDERLYING VACCINE (HYPO)RESPONSIVENESS. WE WILL BUILD ON AVAILABLE KNOWLEDGE OF IMMUNOLOGICAL PROCESSES THAT CAN AFFECT MALARIA VACCINE RESPONSES, AND USE SAMPLES FROM THE COHORTS WHERE HIGH DIMENSIONAL CYTOMETRY AND RNASEQ AND ANTIBODY INTERROGATIONS WILL HELP TO REFINE AND ENRICH THE QUESTIONS REGARDING MALARIA VACCINE (HYPO)RESPONSIVENESS. IN ADDITION, BY USING HUMAN PRIMARY HEPATOCYTES INFECTED WITH P. FALCIPARUM, WE WILL BRING THE RESEARCH CLOSER TO MECHANISMS OF TISSUE-SPECIFIC RESPONSES AND TO EXTRAPOLATE PRE-ERYTHROCYTIC IMMUNITY TO MALARIA IN THE LIVER. BY GENERATING DATA ON THE SAME INDIVIDUALS, DATA INTEGRATION APPROACHES FOR HIGH DIMENSIONAL MEDIATION ANALYSIS, WILL BE USED TO PINPOINT THE SPECIFIC IMMUNOLOGICAL PATHWAYS AND MECHANISMS THAT RESULT IN MALARIA VACCINE (HYPO) RESPONSIVENESS. THIS INFORMATION CAN BE USED TO IMPROVE MALARIA VACCINE EFFICACY AND ALSO TO IDENTIFY INDIVIDUALS WHO WILL NOT BENEFIT FROM THE VACCINE REGIMENS USED SO FAR. IT CAN DIRECT, IN AN EVIDENCE BASED MANNER, ALTERATIONS TO THE VACCINATION DOSE, INTERVALS AND ADJUVANTS.

SUPPORTIVE COMMUNITY COLLEGE STEM STUDENTS DEVELOPMENT, PERSISTENCE, TRANSFER, AND SUCCESS -THIS PROJECT WILL CONTRIBUTE TO THE NATIONAL NEED FOR WELL-EDUCATED SCIENTISTS, MATHEMATICIANS, ENGINEERS, AND TECHNICIANS BY SUPPORTING THE RETENTION AND GRADUATION OF HIGH-ACHIEVING, LOW-INCOME STUDENTS WITH DEMONSTRATED FINANCIAL NEED AT CHEMEKETA COMMUNITY COLLEGE. A TOTAL OF 98 SCHOLARS PURSUING A TRANSFER DEGREE IN BIOLOGY, CHEMISTRY, DATA SCIENCE, ENGINEERING, GEOLOGY, MATHEMATICS, PHYSICS, AND COMPUTER SCIENCE WILL RECEIVE SCHOLARSHIPS AVERAGING $6554 FOR UP TO TWO YEARS. SCHOLARS WILL RECEIVE FACULTY AND PEER MENTORING, AND THE PROJECT WILL BUILD STRONG SCHOLAR COHORTS THROUGH SCHOLAR SPECIFIC SECTIONS OF A FIRST YEAR EXPERIENCE COURSE, FIELD TRIPS TO RELEVANT INDUSTRY LABORATORIES, AND A SPEAKER SERIES. TUTORING, STEM SPECIFIC ADVISING. ADDITIONAL ACTIVITIES FOR SCHOLARS INCLUDE TUTORING, STEM SPECIFIC ADVISING, AND TRANSFER SUPPORT. THE OVERALL GOAL OF THIS TRACK 2 SCHOLARSHIPS IN STEM PROJECT IS TO INCREASE STEM DEGREE COMPLETION OF ACADEMICALLY TALENTED, LOW-INCOME UNDERGRADUATES WITH DEMONSTRATED FINANCIAL NEED. THERE IS A SIGNIFICANT NATIONAL NEED TO GROW THE STEM WORKFORCE AND NURTURE KEY TALENT THAT WILL ENSURE ECONOMIC COMPETITIVENESS AND PROVIDE DOMESTIC LEADERSHIP ACROSS CRITICAL SECTORS. THIS PROJECT DIRECTLY SPEAKS TO THIS NEED BY SUPPORTING STEM STUDENT SUCCESS, WHICH WILL STRENGTHEN THE WORKFORCE IN BIOLOGY, CHEMISTRY, DATA SCIENCE, ENGINEERING, GEOLOGY, MATHEMATICS, PHYSICS, COMPUTER SCIENCE AND OTHER KEY AREAS OF NEED. THE PROJECT WILL BE ASSESSED BY AN EXPERIENCED EVALUATOR WHO WILL COLLECT BOTH FORMATIVE AND SUMMATIVE DATA, AND THE DATA GENERATED WILL CONTRIBUTE TO THE KNOWLEDGE BASE REGARDING EFFECTIVE STRATEGIES TO SUPPORT TALENTED, LOW-INCOME STUDENTS IN STEM. THIS PROJECT IS FUNDED BY NSF?S SCHOLARSHIPS IN SCIENCE, TECHNOLOGY, ENGINEERING, AND MATHEMATICS PROGRAM, WHICH SEEKS TO INCREASE THE NUMBER OF ACADEMICALLY TALENTED, LOW-INCOME STUDENTS WITH DEMONSTRATED FINANCIAL NEED WHO EARN DEGREES IN STEM FIELDS. IMPROVING OUTCOMES IN CANCER TREATMENT-RELATED CARDIOTOXICITY - PROJECT SUMMARY/ABSTRACT ANTHRACYCLINES, SUCH AS DOXORUBICIN (DOX), ARE EFFECTIVE FOR THE TREATMENT OF MANY CANCERS (OVARIAN, MULTIPLE MYELOMA, KAPOSI SARCOMA, LEUKEMIA, BONE SARCOMA, BREAST, ENDOMETRIAL, GASTRIC, LIVER, KIDNEY, AND OTHER CANCERS). THE GLOBAL DOX MARKET IS INCREASING ANNUALLY AND EXPECTED TO REACH $1.3B BY 2026. DOX TOXICITY IS THEREFORE RELEVANT TO A BROAD NUMBER OF CANCERS. HOWEVER, CHEMOTHERAPY INDUCED CARDIAC TOXICITY HAS SUBSTANTIAL MORBIDITY AND MORTALITY. CARDIOTOXICITY AND RECOVERY FROM DNA DAMAGING CHEMOTHERAPY IS DOSE-DEPENDENT. WITH CANCER SURVIVORS ESTIMATED AT 19 MILLION IN THE USA BY 2025, DOX-INDUCED CARDIOTOXICITY IS CONSIDERED TO BE PART OF THE “CARDIO-ONCOLOGY EPIDEMIC”. THE DEVELOPMENT OF NEW APPROACHES TO REDUCE CHEMOTHERAPY-INDUCED CARDIOTOXICITY IS ESSENTIAL. DEXRAZOXANE (DEX), IS FDA APPROVED FOR THE SPECIFIC INDICATION OF “DOXORUBICIN ADMINISTRATION IN WOMEN WITH METASTATIC BREAST CANCER WHO HAVE RECEIVED A CUMULATIVE DOXORUBICIN DOSE OF 300 MG/M2 AND WHO WILL CONTINUE TO RECEIVE DOXORUBICIN THERAPY TO MAINTAIN TUMOR CONTROL”. DEX HAS THE RISK OF MYELOTOXICITY(5). IN OFF LABEL USE OF DEX IN OTHER CANCER POPULATIONS, CARDIAC PROTECTION WAS INCOMPLETE. IN THE PEDIATRIC POPULATION DEX PROVIDED “'INCOMPLETE ACUTE CARDIOPROTECTION AND “NO IMPACT ON CHRONIC CARDIOPROTECTION OR OVERALL SURVIVAL”. LIGHTSEED HAS ASSEMBLED A HIGHLY EXPERIENCED TEAM (ONCOLOGIST-CANCER BIOLOGIST (WITH PRIOR BIOTECHNOLOGY COMPANY EXPERTISE), CARDIO-ONCOLOGIST, HIGH THROUGHPUT LEAD DISCOVERY EXPERT, IMMUNE CANCER BIOLOGIST- MOUSE GENETICIST) IN ORDER TO IDENTIFY AND REPURPOSE FDA-APPROVED COMPOUNDS WITH “DUAL FUNCTION” (ENHANCE CANCER CELL KILLING BY DOX BUT PROTECTION FROM DOX CARDIOTOXICITY). THREE COMPOUNDS FROM THE FDA- APPROVED COMPOUND LIBRARY HAVE BEEN VALIDATED. WE ARE SEEKING ADDITIONAL “DUAL FUNCTION” COMPOUNDS FROM THE LIBRARIES. WE HAVE DEPLOYED AN INNOVATIVE GENETICALLY MODIFIED MURINE TESTING SYSTEM WHICH: (I). ALLOWS COMPARISON WITH THE INCUMBENT TECHNOLOGY (DEX), (II). PROVIDES A NORMAL IMMUNE SYSTEM FOR THE TUMOR IMMUNE RESPONSE. (III). ALLOWS ANALYSIS OF THE HOST IMMUNE RESPONSE USING DOUBLE KNOCKIN FLUORESECENT REPORTER GENES. (IV). ALLOWS DETECTION OF THE TUMOR GROWTH USING A THIRD FLUORESCENT REPORT GENE FOR IN VIVO PROGRESSION ANALYSIS. LIGHTSEED WILL IDENTIFY ADDITIONAL “DUAL FUNCTION” COMPOUNDS BY HIGH THROUGHPUT SCREENING UNDER THE DIRECTION OF A CORE DIRECTOR WHO IS HIGHLY EXPERIENCED WITH HTS SCREENING (PREVIOUSLY HEAD OF LEAD DISCOVERY AT JANSSEN FOR 15 YRS). LIGHTSEED WILL USE CARDIOMYOCYTE CELLS DERIVED FROM POOLED IPSC-CM (IPSC-DERIVED CARDIOMYOCYTES) AND CANCER CELLS. THIS PROPOSAL WILL: (SA1). SCREEN A LIBRARY THAT CONSISTS OF FDA AND EU APPROVED COMPOUNDS, USING AN ASSAY THAT PROTECTS HUMAN CARDIAC MYOCYTES FROM DOX-INDUCED CARDIAC TOXICITY AND ENHANCES CANCER CELL KILLING. (SA2). VALIDATE THESE COMPOUNDS IN TISSUE CULTURE AND (SA3) IN A NOVEL “DUAL FUNCTION” REPORTER MOUSE SYSTEM.

QUANTITATIVE BACKGROUND PARENCHYMAL ENHANCEMENT, MEASURED ON CONTRAST-ENHANCED MAMMOGRAM, AS A NOVEL MARKER OF BREAST CANCER RISK - ROUTINE MAMMOGRAPHIC SCREENING FOR BREAST CANCER REDUCES BREAST CANCER MORTALITY. ON A MAMMOGRAM, RADIOLOGISTS ASSESS MAMMOGRAPHIC DENSITY (MD), OR THE AMOUNT OF FIBROGLANDULAR TISSUE (FGT) IN THE BREAST, WHICH IS A STRONG RISK FACTOR FOR BREAST CANCER. WOMEN WITH DENSE BREASTS ARE MORE LIKELY TO BE DIAGNOSED IN THE “INTERVAL” BETWEEN SCREENING EXAMS, CONTRIBUTING TO INCREASED BREAST CANCER MORTALITY. HOWEVER, CURRENT SCREENING GUIDELINES FOR AVERAGE-RISK WOMEN DO NOT TAKE IMAGING RISK MARKERS INTO ACCOUNT. WOMEN WITH VERY HIGH RISK OF BREAST CANCER, REGARDLESS OF MD, RECEIVE AN INTENSIFIED SCREENING SCHEDULE WITH SUPPLEMENTAL CONTRAST-ENHANCED MAGNETIC RESONANCE IMAGING (MRI). ON MRI, RADIOLOGISTS ASSESS FGT AS WELL AS BACKGROUND PARENCHYMAL ENHANCEMENT (BPE). BPE IS THE DEGREE TO WHICH HEALTHY FGT BRIGHTENS DUE TO THE PRESENCE OF CONTRAST, REFLECTING THE METABOLIC ACTIVITY WITHIN THE FGT. WE RECENTLY DEMONSTRATED THAT BPE ON MRI WAS ASSOCIATED WITH BREAST CANCER RISK INDEPENDENTLY OF THE AMOUNT OF FGT. OTHER STUDIES CORROBORATE OUR FINDINGS, SUPPORTING OUR PREMISE THAT BPE IS A NEW MARKER OF BREAST CANCER RISK. HOWEVER, DUE TO THE SELECT POPULATION RECEIVING MRI, ALTERNATIVE MEASUREMENT APPROACHES ARE NEEDED TO INCREASE THE IMPACT OF BPE FOR WOMEN WHO ARE NOT ABLE TO RECEIVE SCREENING MRI. CONTRAST-ENHANCED MAMMOGRAPHY (CEM) IS AN INCREASINGLY-USED BREAST SCREENING MODALITY THAT SUBSTANTIALLY IMPROVES BREAST CANCER DETECTION, COMPARABLE IN SENSITIVITY TO MRI. AFTER INJECTION OF A CONTRAST AGENT, TWO MAMMOGRAMS AT HIGH AND LOW ENERGIES ARE TAKEN IN QUICK SUCCESSION. MD IS ASSESSED ON THE LOW-ENERGY IMAGE AND BPE IS ASSESSED SUBJECTIVELY BY RADIOLOGISTS AS “MINIMAL”, “MILD”, “MODERATE”, OR “MARKED” ON A PROCESSED (“RECOMBINED”) IMAGE PER THE BREAST IMAGING – REPORTING AND DATA SYSTEM (BI-RADS). BI-RADS BPE MEASURED ON CEM HAS GOOD AGREEMENT WITH BI-RADS BPE MEASURED ON MRI. HOWEVER, SUBJECTIVE MEASUREMENT OF BPE VARIES BETWEEN READERS, PREVENTING REPRODUCIBLE RISK ASSESSMENT RESEARCH. AN OBJECTIVE, QUANTITATIVE MEASURE OF BPE ON CEM IS NEEDED TO RIGOROUSLY EVALUATE THE RELATIONSHIP BETWEEN INCREASED BPE AND BREAST CANCER RISK. TO ADDRESS THIS RESEARCH GAP, WE PROPOSE TO ASSESS THE ASSOCIATION BETWEEN CANCER AND A FULLY AUTOMATED MEASURE OF BPE ON CEM. AIM 1. EVALUATE BPE AS A REPRODUCIBLE MARKER OF BREAST CANCER RISK: WE WILL IMPLEMENT A FULLY AUTOMATED MEASURE OF QUANTITATIVE BPE ON 462 CEMS, EVALUATE THE AGREEMENT WITH THE GOLD STANDARD BI-RADS MEASURE, AND COMPARE WITH BPE ON MRI. AIM 2. DETERMINE WHETHER QUANTITATIVE BPE IMPROVES BREAST CANCER RISK ASSESSMENT: IN A MATCHED CASE-CONTROL STUDY OF 154 NEWLY DIAGNOSED BREAST CASES MATCHED TO 308 CANCER- FREE CONTROLS, WE WILL DETERMINE THE ASSOCIATION OF BREAST CANCER WITH BPE, ACCOUNTING FOR FGT, RACE/ETHNICITY, REPRODUCTIVE FACTORS, AND OTHER RISK FACTORS FOR BREAST CANCER. OUR LONG-TERM GOAL IS TO USE QUANTITATIVE BPE ON CEM TO IMPROVE BREAST CANCER RISK ASSESSMENT, THEREBY ENABLING PERSONALIZATION OF BREAST CANCER SCREENING TO IMPROVE SCREENING OUTCOMES AND REDUCE MORTALITY DUE TO BREAST CANCER.

THE SMALL EASTERN PACIFIC HAWKSBILL NESTING POPULATION WAS THOUGHT TO HAVE BEEN EXTIRPATED UNTIL REMNANT POPULATIONS WERE DISCOVERED IN REMOTE SITES UNTIL 2008. BAHIA DE JIQUILISCO ACCOUNTS FOR ABOUT 40 % OF ALL HAWKSBILL NESTING IN THE EAST PACIFIC. SPECIFIC ACTIVITIES INCLUDE: (1) CONDUCT COMMUNITY BASED PATROLS AND RELOCATION OF NESTS TO HATCHERIES; (2) CONDUCT OUTREACH ACTIVITIES SUCH AS A HAWKSBILL FESTIVAL, HAWKSBILL CUP COMPETITION AND 'DAY OF THE HAWKSBILL' EVENTS IN SCHOOLS TO RAISE AWARENESS ABOUT THREATS TO HAWKSBILLS; (3) CONDUCT YEAR ROUND FISHERIES BYCATCH MONITORING OF LOBSTER FISHERIES WITH ON BOARD OBSERVERS; (4) CONDUCT LED LIGHT TRIALS ON LOBSTER NETS TO DETERMINE DETERRENCE EFFECTIVENESS; AND (5) DEVELOP LOCAL CAPACITY TO STRENGTHEN A HAWKSBILL ECOTOURISM PROGRAM FOR THE BENEFIT OF LOCAL RESIDENTS.

THIS INITIATIVE INTRODUCES A PIONEERING METHOD FOR IDENTIFYING BIOMARKERS WITHIN EXTRACELLULAR VESICLES (EVS), EARLY MARKERS SPECIFIC TO CANCER CIRCULATING IN THE BLOODSTREAM. A SUCCESSFUL OUTCOME FROM THIS PROJECT WOULD POSITION ANY RESULTING PRODUCTS FAVORABLY IN THE BROADER LANDSCAPE OF CANCER DETECTION TECHNOLOGIES. THIS SMALL BUSINESS INNOVATION RESEARCH (SBIR) PHASE I PROJECT AIMS TO DEVELOP A HIGHLY SENSITIVE, MINIMALLY INVASIVE LABORATORY TEST TO DETECT EARLY-STAGE OVARIAN CANCERS (OC) BY ASSAYING RELEVANT BIOMARKERS ON CIRCULATING EXTRACELLULAR VESICLES (EVS) IN PATIENT PLASMA. CURRENTLY, BOTH COMMERCIAL AND HOBBYIST BEEKEEPERS MUST DECIDE BETWEEN LOSING A COLONY NOW TO VARROA DESTRUCTOR INFESTATION, OR LATER TO CONSEQUENT PESTICIDE EFFECTS. BRINGING A SAFE AND EFFECTIVE NEW BIOLOGICAL-BASED PRODUCT TO THE MARKET WILL LOWER BEEKEEPER?S ANNUAL SPENDING AS WELL AS TIME AND EFFORT TO KEEP THEIR COLONIES ALIVE BY LEAVING HONEY BEE COLONIES FREE FROM THE DEVASTATING DOWNSTREAM EFFECTS OF TRADITIONAL PESTICIDES. CURRENTLY, STUDENTS AT THE INSTITUTION'S WOODBURN CAMPUS MUST COMMUTE UP TO 90 MINUTES TO COMPLETE LABORATORY COURSES ON THE SALEM CAMPUS. TO ADDRESS THIS CHALLENGE, THE INSTITUTION IS RENOVATING EXISTING WOODBURN CLASSROOM SPACE TO OFFER LABORATORY SECTIONS FOR AT LEAST 6 COURSES. TO EQUIP THE NEW SPACE, THIS PROJECT WILL SECURE A BROAD COLLECTION OF EQUIPMENT, INCLUDING MICROSCOPES, INSTRUCTIONAL MODELS, DISSECTION TOOLS AND SUPPLIES, AND SAFETY EQUIPMENT. AN ESTIMATED 600 STUDENTS WILL UTILIZE THE PROJECT-FUNDED EQUIPMENT EACH YEAR, SAVING UP TO 900 HOURS OF STUDENT COMMUTING TIME. THE GOALS OF THIS PROJECT ARE TO ENRICH THE LEARNING AND EXPERIENCES OF UNDERGRADUATE STUDENTS BY PROVIDING CRITICAL EQUIPMENT FOR UNDERGRADUATE LABORATORY COURSES. THE PROJECT WILL FURTHERMORE REDUCE THE NEED FOR STUDENTS ON THE WOODBURN CAMPUS TO COMMUTE TO ENROLL IN KEY LABORATORY COURSES. THIS REMOVES A SIGNIFICANT OBSTACLE TO THEIR STEM EDUCATIONAL EXPERIENCES. THE PROJECT TEAM WILL ASSESS THE IMPACT OF THE PROJECT-FUNDED EQUIPMENT BY TRACKING THE GROWTH OF LABORATORY COURSE ENROLLMENT ON THE WOODBURN CAMPUS AND CAPTURING STUDENT USE OF THE EQUIPMENT. THIS PROJECT IS FUNDED BY THE HSI PROGRAM, WHICH AIMS TO ENHANCE UNDERGRADUATE STEM EDUCATION AND INCREASE CAPACITY TO ENGAGE IN THE DEVELOPMENT AND IMPLEMENTATION OF INNOVATIONS TO IMPROVE STEM TEACHING AND LEARNING AT HSIS. THE CHEMEKETA INCUBATOR FARM (CIF) PROJECT IS UNIQUE IN THAT IT IS A LAND-BASED EXPERIENTIAL LEARNING OPPORTUNITY THAT PROVIDES BEGINNING FARMERS A PLOT OF LAND ONTO WHICH THEY CAN ESTABLISH THEIR OWN INDEPENDENT FARM ENTERPRISES WITH ON-SITE SUPPORT FROM CHEMEKETA FACULTY AND STAFF. OREGON STATE UNIVERSITY EXTENSION WILL BE A VALUABLE PARTNER ON THIS PROJECT, SUPPORTING COLLABORATION WITH STATEWIDE PARTNERS AND RESOURCES. FARMER PARTICIPANTS CAN UTILIZE THE MANY SERVICES, CERTIFICATIONS, AND DEGREES THAT BOTH INSTITUTIONS OFFER.
