CAREER: SELECTION PIPELINE FOR ANCIENT AND MODERN GENOMES: IMPROVING ACCESS TO GENOMIC METHODS FOR STUDYING HUMAN ADAPTATIONS -AS MODERN HUMANS DISPERSED INTO NEW ENVIRONMENTS AROUND THE GLOBE, DIRECTIONAL SELECTION PLAYED AN IMPORTANT ROLE IN SHAPING THE GENETIC VARIANTS PRESENT ACROSS DIFFERENT POPULATIONS. MANY DIVERSE AND UNIQUE ADAPTATIONS EXIST, OFTEN PLAYING A LARGE ROLE IN HUMAN HEALTH AND CULTURE. TOOLS THAT CAPTURE GENOMIC PATTERNS OF SELECTION ARE INSTRUMENTAL FOR DETERMINING HOW DIRECTIONAL SELECTION HAS IMPACTED MODERN HUMAN POPULATIONS, AND RAPID ADVANCES IN SEQUENCING TECHNOLOGY HAVE GREATLY EXPANDED THE NUMBER OF ANCIENT AND PRESENT-DAY HUMAN GENOMES FOR STUDY. HOWEVER, THE NUMBER OF RESEARCHERS WITH THE BIOINFORMATIC EXPERTISE NEEDED TO USE THESE TOOLS IS LIMITED, AND REFINED DEMOGRAPHIC MODELS TO BETTER INFORM SELECTION INFERENCES ARE NOT WIDELY AVAILABLE FOR DIFFERENT HUMAN POPULATIONS. THIS PROJECT WILL DEVELOP SOFTWARE INFRASTRUCTURE FOR STUDYING ADAPTATIONS IN MODERN HUMANS USING ANCIENT AND PRESENT-DAY GENOMIC DATASETS THAT CAN BE IMPLEMENTED BY RESEARCHERS WITH LOW COMPUTATIONAL EXPERTISE. THIS PROJECT WILL IMPLEMENT THE PIPELINE IN COURSE-BASED UNDERGRADUATE RESEARCH EXPERIENCES (CURES) TO ACCELERATE DETECTION AND CHARACTERIZATION OF POSITIVE SELECTION IN GENOMIC REGIONS OF INTEREST FOR WORLDWIDE HUMAN POPULATIONS. IN ADDITION, THIS PROJECT WILL PROVIDE APPLIED COMPUTING TRAINING TO UNDERGRADUATE STUDENTS AND PROFESSIONAL DEVELOPMENT RELATED TO THE SELECTION SOFTWARE FOR HIGHER EDUCATION BIOLOGY FACULTY, WHICH WILL HELP MORE STUDENTS ACROSS THE NATION GAIN GENOMICS-BASED RESEARCH SKILLS. IN TOTAL, THIS PROJECT WILL SUPPORT FUTURE RESEARCHERS BY TRAINING ONE POSTDOCTORAL FELLOW, TWO POST-BACCALAUREATE STUDENTS, AND TEN UNDERGRADUATE RESEARCH STUDENTS, AS WELL AS APPROXIMATELY 60 UNDERGRADUATE STUDENTS AND 30 HIGHER EDUCATION BIOLOGY FACULTY IN RELATED DATA SCIENCE SHORT COURSES AND GENOMICS WORKSHOPS. THIS PROJECT WILL DEVELOP SOFTWARE INFRASTRUCTURE ESTABLISHING A FLEXIBLE, MODULARIZED, AND ACCESSIBLE SELECTION PIPELINE INCORPORATING ANCIENT AND PRESENT-DAY HUMAN GENOMIC DATASETS THAT CAN BE IMPLEMENTED BY RESEARCHERS WITH LIMITED COMPUTATIONAL BACKGROUND. A RELIABLE DEMOGRAPHIC MODEL FOR EAST AND SOUTHEAST ASIANS WILL BE CREATED AND INCORPORATED INTO THIS PIPELINE, AND THIS TOOL WILL BE USED TO CHARACTERIZE SELECTION AT PUTATIVELY ADAPTED GENES IN EAST AND SOUTHEAST ASIANS, WHICH WILL BE BENEFICIAL TO FURTHER RESEARCH ON HUMAN HEALTH AND EVOLUTIONARY HISTORY. TO FACILITATE DATA SCIENCE AND RESEARCH SKILLS ACROSS THE FUTURE STEM WORKFORCE, THIS RESEARCH WILL BE INTEGRATED WITH EDUCATION IN THE FOLLOWING WAYS. FIRST, UNDERGRADUATE STUDENTS WILL CONDUCT ORIGINAL RESEARCH PROJECTS USING THE SELECTION PIPELINE THROUGH INDEPENDENT SUMMER RESEARCH OR AN UPPER-LEVEL BIOLOGY COURSE. SECOND, THIS PROJECT WILL CREATE WORKSHOPS FOR HIGHER EDUCATION FACULTY FOCUSED ON THE CURRICULUM AND IMPLEMENTATION OF THE SELECTION PIPELINE AS A CURE, EXTENDING THIS OPPORTUNITY TO MORE UNDERGRADUATES ACROSS THE NATION. IN ADDITION TO THE RADIO WAVES THAT COME FROM BRIGHT OBJECTS IN SPACE THAT WE KNOW ABOUT, THERE IS ALSO DIFFUSE RADIO EMISSION WHICH COMES FROM EVERY DIRECTION IN THE SKY.? THIS DIFFUSE RADIO EMISSION CAN ONLY BE PROPERLY UNDERSTOOD BY OBSERVING IT OVER LARGE PORTIONS OF THE SKY AT DIFFERENT RADIO WAVELENGTHS.? THIS PROJECT WILL MAKE A NEW, MODERN MAP OF THESE SIGNALS BY UTILIZING THE UNIQUE FEATURES OF THE ROBERT C. BYRD GREEN BANK TELESCOPE ALONG WITH ADVANCED CUSTOM INSTRUMENTATION DEVELOPED SPECIFICALLY FOR THIS PROJECT.? THE INVESTIGATORS WILL LEVERAGE CUTTING-EDGE RADIO RECEIVER TECHNOLOGY AND CONTEMPORARY COMPUTING CAPABILITIES TO ACHIEVE UNPRECEDENTED ACCURACY.? THE RESULTING PICTURE OF THE RADIO EMISSION COMING FROM OUR UNIVERSE WILL BE AVAILABLE TO THE ASTRONOMICAL COMMUNITY AS AN IMPORTANT RESOURCE. THIS MAP OF THE WHOLE SKY NORTH OF -47 DEGREES DECLINATION, WITH ALL GALACTIC LATITUDES OBSERVED, WILL ALLOW A DETERMINATION, FOR THE FIRST TIME IN A SYSTEMATICALLY RELIABLE WAY, OF THE ABSOLUTE LEVEL OF DIFFUSE RADIO EMISSION IN THE UNIVERSE AND FROM GALACTIC STRUCTURE.? IT WILL PROVIDE AN IMPORTANT NEW RESOURCE FOR UNDERSTANDING AND CONSTRAINING ALMOST ALL GALACTIC AND EXTRAGALACTIC PHENOMENA THAT MANIFEST IN, OR DEPEND ON THE UNDERSTANDING OF, DIFFUSE RADIO EMISSION. SPECIFICALLY, THIS WORK WILL FOCUS ON HOW NEURONS POPULATE THEIR MEMBRANES WITH RECEPTORS, KNOWN AS ?AMPA? RECEPTORS, THAT CAN RESPOND TO NEUROTRANSMITTERS. THIS PROCESS REQUIRES INTRACELLULAR MEMBRANE VESICLES TO FUSE WITH THE PLASMA MEMBRANE OF NEURONS. THOUGH MEMBRANE FUSION HAS BEEN STUDIED FOR DECADES, THE REGULATION OF MEMBRANE FUSION IN AMPA RECEPTOR (AMPAR) DELIVERY IS STILL POORLY UNDERSTOOD. THIS PROJECT WILL DESCRIBE KEY STEPS IN THE DELIVERY PROCESS AND AIMS TO IDENTIFY NOVEL PROTEINS INVOLVED IN RECEPTOR TRAFFICKING DURING LEARNING AND MEMORY. TOGETHER, THESE AIMS WILL ADVANCE THE UNDERSTANDING OF HOW MEMORIES FORM. THIS WORK WILL BE PERFORMED PRIMARILY BY UNDERGRADUATES, BOTH IN MENTORED RESEARCH PROJECTS AND IN A COURSE-BASED UNDERGRADUATE RESEARCH EXPERIENCE (CURE). THESE EFFORTS WILL SIGNIFICANTLY EXPAND THE TRAINING OPPORTUNITIES AVAILABLE TO UNDERGRADUATE STUDENTS. AT THE NEURONAL SYNAPSE, MEMBRANE FUSION IS NOT ONLY ESSENTIAL FOR THE DELIVERY OF NEUROTRANSMITTER MOLECULES BUT ALSO FOR THE INSERTION OF NEUROTRANSMITTER RECEPTORS INTO THE PLASMA MEMBRANE. THOUGH MEMBRANE FUSION INVOLVED IN THE DELIVERY OF NEUROTRANSMITTERS IS WELL-CHARACTERIZED, COMPARATIVELY LITTLE IS KNOWN ABOUT THE REGULATION OF NEUROTRANSMITTER RECEPTOR DELIVERY. MEMBRANE FUSION DURING AMPAR INSERTION INTO THE PLASMA MEMBRANE IS DRIVEN BY SNARE PROTEINS, INCLUDING SYNTAXIN-3, SYNAPTOBREVIN-2, AND SNAP-47 (S47). THIS PROJECT WILL INVESTIGATE THE ROLE OF S47, SPECIFICALLY FOCUSING ON ITS POORLY CHARACTERIZED N-TERMINAL AND LINKER DOMAINS. AN IN VITRO FUSION ASSAY WILL BE USED TO DESCRIBE THE FUNCTION OF THE UNCHARACTERIZED S47 DOMAINS, AND SINGLE-PARTICLE CRYO-ELECTRON MICROSCOPY (CRYOEM) WILL BE USED TO SOLVE THE STRUCTURE OF THE SNARE PROTEIN COMPLEX INVOLVED IN AMPAR EXOCYTOSIS. THESE DATA WILL PROVIDE INSIGHT INTO THE REGULATION OF MEMBRANE FUSION DURING AMPAR DELIVERY TO THE PLASMA MEMBRANE. WHILE ONLY A SMALL PERCENTAGE OF TSETSE ARE INFECTED WITH TRYPANOSOMES, THEY ALL HOUSE A MIXED POPULATION OF INDIGENOUS ENDOSYMBIOTIC BACTERIA, INCLUDING SODALIS GLOSSINIDIUS. S. GLOSSINIDIUS RESIDES IN TSETSE’S MIDGUT, WHICH IS EXPOSED TO LARGE AMOUNTS OF IMBIBED BLOOD. THE CENTRAL HYPOTHESIS UNDERLYING THE WORK PROPOSED IN THIS APPLICATION IS THAT S. GLOSSINIDIUS COLONIZATION OF THE TSETSE GUT REQUIRES EXPRESSION OF A SUITE OF GENES THAT ENABLES THE BACTERIUM TO ACCESS NUTRIENTS FROM AND SURVIVE WITHIN THIS BLOOD-RICH ENVIRONMENT. THE SCIENTIFIC GOALS OF THIS PROJECT ARE TO EXAMINE THESE HYPOTHESES. IN SPECIFIC AIM 1, S. GLOSSINIDIUS TRANSPOSON LIBRARIES WILL BE SCREENED FOR CANDIDATE GENES THAT CONTRIBUTE TO FITNESS IN BLOOD. SOME INVASIVE SPECIES SHOW SIGNS OF RAPID ADAPTATION TO NEW HABITATS, BUT FEW STUDIES HAVE CAPTURED POST-INVASION ADAPTATION IN REAL TIME. ADDITIONALLY, INVASIVE SPECIES COLONIZING MULTIPLE HABITATS SIMULTANEOUSLY OFFER THE OPPORTUNITY TO STUDY THE LONGSTANDING QUESTION OF WHETHER EVOLUTION IS PREDICTABLE. TO UNDERSTAND HOW QUICKLY AND PREDICTABLY ORGANISMS ADAPT TO NEWLY COLONIZED ENVIRONMENTS, DETAILED GENOMIC STUDIES OF REPEATED INCIPIENT INVASIONS ARE NECESSARY. IN THIS STUDY, WE WILL GENETICALLY AND PHENOTYPICALLY CHARACTERIZE NORTH AMERICAN POPULATIONS OF THE AFRICAN FIG FLY, ZAPRIONUS INDIANUS, TO TEST FOR LOCAL ADAPTATION AND PARALLEL EVOLUTION IN ITS ONGOING INVASION. THIS TRACTABLE MODEL ORGANISM ARRIVED IN NORTH AMERICA LESS THAN TWO DECADES AGO AND RAPIDLY SPREAD. OUR DATA SUGGEST IT IS LOCALLY EXTIRPATED FROM TEMPERATE HABITATS EACH WINTER AND RE-COLONIZES THESE LOCALES EACH SPRING. OUR CENTRAL HYPOTHESIS IS THAT Z. INDIANUS UNDERGOES PREDICTABLE POST-COLONIZATION ADAPTATION ACROSS MULTIPLE LOCATIONS AND YEARS, RESULTING IN REPEATED CHANGES IN ALLELE FREQUENCIES AND ECOLOGICALLY RELEVANT PHENOTYPES. TO TEST THIS HYPOTHESIS, WE WILL COLLECT, SEQUENCE, AND PHENOTYPE ISOLATES FROM ALONG THE EAST COAST OF NORTH AMERICA AND FROM TWO FOCAL VIRGINIA ORCHARDS OVER MULTIPLE YEARS. IN AIM 1, WE WILL QUANTIFY GENETIC AND PHENOTYPIC VARIATION IN NORTH AMERICAN POPULATIONS BY GENERATING POOLED SEQUENCING OF POPULATIONS COLLECTED ALONG A LATITUDINAL GRADIENT TO TEST FOR CLINALLY VARYING POLYMORPHISMS OVER TWO SAMPLING YEARS. WE WILL ALSO PHENOTYPE LINES COLLECTED AT DIFFERENT LATITUDES TO TEST FOR CLINAL DIFFERENCES IN MORPHOLOGICAL, LIFE-HISTORY, AND STRESS TOLERANCE PHENOTYPES. REPEATED CLINAL AND PHENOTYPIC DIFFERENTIATION WOULD PRESENT EVIDENCE FOR RAPID ADAPTATION TO LOCAL ENVIRONMENTS. IN AIM 2, WE WILL SAMPLE AND SEQUENCE FLIES FROM TWO GEOGRAPHICALLY SEPARATE VIRGINIA ORCHARDS OVER THREE GROWING SEASONS TO TEST FOR ALLELES AND PHENOTYPES THAT PREDICTABLY CHANGE FOLLOWING INVASION. THESE EXPERIMENTS WILL ALLOW US TO IDENTIFY THE GENOMIC TARGETS OF POST-INVASION ADAPTATION AND QUANTIFY THE PREDICTABILITY OF EVOLUTION. IN AIM 3, WE WILL CHARACTERIZE THE PUTATIVE TARGET OF A RECENT SELECTIVE SWEEP IN NORTH AMERICAN FLIES: A 600 KB HAPLOTYPE THAT IS COMMON IN VIRGINIA, INTERMEDIATE FREQUENCY IN FLORIDA, AND NOT FOUND IN AFRICA. WE WILL GENERATE OUTBRED POPULATIONS FIXED FOR ALTERNATE ALLELES AND PERFORM RNASEQ TO TEST FOR DIFFERENTIAL GENE EXPRESSION ASSOCIATED WITH ALTERNATE HAPLOTYPES. PHENOTYPING EXPERIMENTS WILL EXPLORE THE POTENTIAL FUNCTIONAL TARGETS OF THIS SWEEP, ALLOWING US TO DISSECT THE BIOLOGICAL FUNCTION OF A GENOMIC SIGNAL OF SELECTION. COLLECTIVELY, THESE EXPERIMENTS WILL TAKE A MULTIPRONGED APPROACH TO TEST FOR POST-INVASION ADAPTATION AND PARALLEL EVOLUTION IN REAL TIME. THIS STUDY WILL PROVIDE A FRAMEWORK TO UNDERSTANDING RAPID ADAPTATION TO NOVEL ENVIRONMENTS AND PREDICTING EVOLUTIONARY TRAJECTORIES OF FUTURE INVASIVE SPECIES, WHICH MAY LEAD TO SOLUTIONS TO CONTROL THEIR SPREAD.

RUI: TOWARDS A BETTER UNDERSTANDING OF POLYRADICAL ELECTRONIC STRUCTURE: HARNESSING ORGANIC BUILDING BLOCKS FOR THE DESIGN OF MOLECULAR MAGNETS -IN THIS RUI PROJECT, FUNDED BY THE CHEMICAL MECHANISM, FUNCTION, AND PROPERTIES PROGRAM OF THE CHEMISTRY DIVISION, PROFESSOR CAROL PARISH OF THE DEPARTMENT OF CHEMISTRY AT THE UNIVERSITY OF RICHMOND WILL DEVELOP MODELS TO BETTER UNDERSTAND THE FUNDAMENTAL BEHAVIOR OF NON-METALLIC MOLECULE-BASED MAGNETS. MAGNETIC MATERIALS ARE CRITICALLY IMPORTANT IN THE DEVELOPMENT AND MANUFACTURE OF DEVICES FOR NAVIGATION, IMAGING, SENSING, RECORDING AND GEOLOCATION. MAGNETS ARE WIDELY USED IN APPLICATIONS RANGING FROM ELECTRIC MOTORS, TO HARD DRIVE DATA STORAGE, TO SOUND AND VIDEO RECORDING, TO MEDICAL IMAGING. THE MOST COMMON MAGNETS IN USE TODAY ARE METAL ALLOYS THAT EXTEND ACROSS LARGE, EXTENDED LATTICES. THESE CONVENTIONAL MAGNETS ARE RIGID, BULKY AND HEAVY. MAGNETS FORMED FROM ORGANIC MATERIALS ARE EXTREMELY FLEXIBLE, MALLEABLE AND LIGHTWEIGHT; HOWEVER, THEY ARE LESS WELL UNDERSTOOD. THIS PROJECT WILL HARNESS PROFESSOR PARISH?S EXPERTISE IN THE PROPER CHARACTERIZATION OF MOLECULES WITH UNPAIRED ELECTRONS TO DESIGN BETTER MATERIALS FOR MAGNETIC APPLICATIONS. THE RESEARCH WILL ADDRESS THREE MAJOR QUESTIONS IN MATERIALS SCIENCE: (1) HOW DO REMARKABLE PROPERTIES OF MATTER EMERGE FROM COMPLEX RELATIONSHIPS AT THE ATOMIC, MAGNETIC, AND ELECTRONIC LEVEL? (2) HOW DO WE CONTROL MATERIAL PROCESSES AT THE LEVEL OF ELECTRONS? (3) HOW DO WE DESIGN REVOLUTIONARY NEW FORMS OF MATTER WITH TAILORED PROPERTIES? THE PROJECT IS HIGHLY INTERDISCIPLINARY, UTILIZING CONCEPTS FROM PHYSICAL, MATERIALS AND ORGANIC CHEMISTRY, AND WILL PROVIDE AN IDEAL ENVIRONMENT IN WHICH TO EDUCATE AND TRAIN THE NEXT GENERATION OF SCIENTISTS. THE PROJECT TEAM WILL USE HIGHLY CORRELATED, MULTIREFERENCE METHODS TO CHARACTERIZE THE GROUND AND EXCITED STATES OF A VARIETY OF DI AND POLYRADICAL MOLECULES THAT SERVE AS MODELS FOR THE DEVELOPMENT OF MOLECULAR MAGNETS. PROFESSOR PARISH AND HER RESEARCH GROUP WILL CHARACTERIZE THE ELECTRONIC STRUCTURE AND MAGNETIC PROPERTIES OF THESE POLYRADICALS, AND USE THIS DATA, ALONG WITH MAGNETIC PROPERTIES OBTAINED FROM EXPERIMENTAL MEASUREMENTS, TO DEVELOP A PREDICTIVE MODEL FOR MOLECULAR MAGNETISM IN ORGANIC POLYRADICALS. THE RESULTS FROM THESE STUDIES WILL PROVIDE FUNDAMENTAL INFORMATION TO SUPPORT THE DESIGN OF ORGANIC MAGNETS THAT WILL BE MORE FLEXIBLE AND LIGHTER THAN CONVENTIONAL METAL-BASED MAGNETS WITH A SMALLER, LESS TOXIC ENVIRONMENTAL FOOTPRINT. BRC-BIO: LOOKING BEYOND THE CALORIE: NUTRITIONAL ECOLOGY IN A GENERALIST PREDATOR -CALORIES ARE OFTEN TREATED AS THE PRIMARY UNIT OF NUTRITION, BUT A GROWING BODY OF EVIDENCE SUGGESTS THAT THE SPECIFIC NUTRIENT COMPOSITION OF FOOD MAY BE A MORE IMPORTANT DRIVER OF FORAGING BEHAVIOR IN WILDLIFE. BECAUSE NUTRIENT COMPOSITION VARIES WITH ENVIRONMENTAL CONDITIONS, CLIMATE CHANGE IS LIKELY TO AFFECT NUTRIENT CONTENT OF FOOD. FOR PREDATORS, THIS MEANS THAT A CHANGING CLIMATE COULD ALTER THE NUTRITION OF THEIR PREY, AND THEREFORE, WHAT PREY THEY SELECT. HOWEVER, WITHOUT BASELINE KNOWLEDGE OF HOW NUTRIENTS SHIFT IN RESPONSE TO CHANGING ENVIRONMENTAL CONDITIONS, IT IS DIFFICULT TO MAKE PREDICTIONS ABOUT PREDATOR BEHAVIOR. SONGBIRDS ARE COMMON PREDATORS IN MANY ECOSYSTEMS THAT PREY ON A VARIETY OF INSECTS AND CAN EASILY BE CAPTURED AND OBSERVED FOR BEHAVIORAL STUDIES. THIS PROJECT WILL 1) ASSESS MACRONUTRIENTS (PROTEIN, CARBOHYDRATE, LIPID) IN PREY AND IDENTIFY SPECIFIC RATIOS OF MACRONUTRIENTS THAT MAXIMIZE NESTLING GROWTH, 2) EXAMINE HOW THE NUTRITIONAL VALUE OF PREY CHANGES WHEN REARED IN EXPERIMENTALLY MANIPULATED ENVIRONMENTAL CONDITIONS, AND 3) EXAMINE WHETHER CHANGES IN NUTRITION AFFECT BIRDS? PREFERENCE FOR PREY. BECAUSE INSECTS ARE CONSUMED BY MANY ANIMALS OUTSIDE OF BIRDS, THIS WORK WILL HAVE CONSERVATION IMPLICATIONS FOR A WIDE RANGE OF TAXA. THE RESEARCH TEAM WILL INCLUDE UNDERGRADUATE STUDENTS ENROLLED IN RELEVANT BIOLOGY COURSES AND PAID UNDERGRADUATE ASSISTANTS. STUDENTS WILL DEVELOP INDEPENDENT PROJECTS RELATED TO THE BROADER THEME AND TRAVEL TO NATIONAL CONFERENCES TO PRESENT THEIR WORK. THIS RESEARCH EXPERIENCE WILL EQUIP PARTICIPANTS WITH SKILLS IN LABORATORY AND FIELD WORK AND BUILD STUDENTS? CONFIDENCE TO PERSIST IN STEM CAREERS. USING A COMBINATION OF MANIPULATIVE LABORATORY EXPERIMENTS AND AN EXISTING NETWORK OF 100 EASTERN BLUEBIRD NEST-BOXES, THE RESEARCHERS WILL EXAMINE HOW ENVIRONMENTAL CONDITIONS AFFECT PREY NUTRIENT COMPOSITION AND WHETHER THESE CHANGES IMPACT PREDATOR PREFERENCE. THIS WORK WILL TEST THE USE OF THE NUTRITIONAL GEOMETRIC FRAMEWORK: A TOOL FOR ASSESSING NUTRITION BY UTILIZING RATIOS OF NUTRIENTS TO MODEL HOW FOODS ARE COMBINED TO REACH MULTIDIMENSIONAL NUTRITIONAL TARGETS THAT MAXIMIZE ORGANISM PERFORMANCE. THIS FRAMEWORK IS USEFUL IN MODELING HOW DIET CHANGES AS DIFFERENT FOOD ITEMS CHANGE IN AVAILABILITY OR QUALITY. BY MEASURING THE IMPACT OF PREY MACRONUTRIENTS ON NESTLING DEVELOPMENT IN OBJECTIVE 1, THE RESEARCHERS WILL IDENTIFY MULTIDIMENSIONAL NUTRITIONAL TARGETS THAT MAXIMIZE GROWTH IN NESTLINGS. IN OBJECTIVE 2, THESE DATA WILL BE USED TO TEST WHETHER THE ENVIRONMENTAL CONDITIONS OF PREY MEANINGFULLY IMPACT THEIR NUTRIENT CONTENT. ALTHOUGH SOME STUDIES INVESTIGATE HOW NUTRIENTS CHANGE IN RESPONSE TO TEMPERATURE OR DIET SHIFTS, VERY FEW EXAMINE THESE IN COMBINATION, EVEN THOUGH CLIMATE CHANGE IS PREDICTED TO SIMULTANEOUSLY AFFECT BOTH. EXPERIMENTS IN OBJECTIVE 2 MANIPULATING DIET AND REARING TEMPERATURE WILL ADVANCE OUR UNDERSTANDING OF HOW MULTIPLE ENVIRONMENTAL CHANGES INTERACT TO DETERMINE NUTRITIONAL VALUE. FINALLY IN OBJECTIVE 3, RESEARCHERS WILL EXAMINE BLUEBIRD NUTRITIONAL PREFERENCES USING FIELD-BASED CHOICE ASSAYS WITH NUTRITIONALLY ALTERED INSECT PREY. OUR FACULTY HAVE BEEN SUPPORTING THEIR WORK USING LAPTOPS, DESKTOPS, BORROWING COMPUTE CYCLES FROM COLLABORATORS, USING CLOUD RESOURCES AND SUPERCOMPUTER CENTERS. AS THE IMPORTANCE OF COMPUTING IN BIOMEDICAL RESEARCH INCREASES, THIS DEFICIENCY IS LIMITING THE SIZE AND NATURE OF THE PROJECTS THAT OUR FACULTY CAN ENVISION AND COMPLETE, PREVENTING THEM FROM ACHIEVING THEIR FULL POTENTIAL OR ENGAGING STUDENTS AS DEEPLY AS THEY COULD. THE ADDITION OF A NINE-NODE, HIGH-PERFORMANCE, SHARED COMPUTATIONAL RESOURCE CONTAINING CPUS, GPUS, LARGE AMOUNTS OF RAM, AND RAID-PROTECTED STORAGE WILL ENABLE OUR FACULTY AND THEIR STUDENTS TO PERFORM STATE-OF-THE-ART COMPUTATIONS. THIS HARDWARE WILL BE USED TO PERFORM GENOMIC ANALYSIS OF TICK-BORNE DISEASES, AND TO STUDY THE EVOLUTION OF HUMAN DEMOGRAPHICS AND POPULATIONS. THE HARDWARE WILL ALSO SUPPORT IMAGE AND/OR VIDEO ANALYSIS RELATED TO AN RNA HELICASE IMPORTANT IN TRANSLATION, A CARBON NANODOT DRUG DELIVERY SYSTEM, AND THE DEVELOPMENT OF A RODENT BASED BEHAVIORAL MODEL OF EFFORT-BASED REWARDS. THE RESOURCE WILL ALSO BE USED FOR MOLECULAR DYNAMICS SIMULATIONS THAT WILL UNRAVEL THE STRUCTURE-FUNCTION RELATIONSHIP IN VOLTAGE AND NON-VOLTAGE GATED POTASSIUM ION CHANNELS, RNA HELICASE, PROTEINS THAT MEDIATE CANCER PROGRESSION, THE BINDING AFFINITY OF CARBON NANODOTS WITH MINERALIZED BONES, AND AMYLOID PRECURSOR PROTEIN WITH A MOTOR PROTEIN, AND THE DESIGN OF A NOVEL DNA POLYMERASE. CURRENT REARING EFFORTS REQUIRE LIVE WOOD AND FOLIAGE TO MAINTAIN EAB REARING, WHICH IS DIFFICULT AND COSTLY. RELEASE OF BIOCONTROL AGENTS IS KEY TO THE RESTORATION OF ASH TREES AND MAINTENANCE OF FOREST ECOSYSTEMS ACROSS A LARGE PORTION OF THE UNITED STATES. STUDIES CONDUCTED FOR THIS AGREEMENT AIM TO INCREASE NEONATE TO LATE INSTAR LARVAL SURVIVAL ON EXTRUDED DIET, DEVELOP METHODS FOR EAB ADULT REARING ON DIET AND SUCCESSFUL PARASITISM OF DIET-REARED EAB LARVAE. ACTIVITIES TO BE PERFORMED: THE RECIPIENT WILL PROVIDE THREE TECHNICIANS (TWO FULL-TIME AND ONE PART-TIME). ONE FULL-TIME AND ONE PART-TIME TECHNICIAN WILL BE BASED AT THE APHIS EAB BIOCONTROL REARING FACILITY IN BRIGHTON, MI. ACTIVITIES WILL INCLUDE CONDUCTING RESEARCH EXAMINING EXTRUDED ARTIFICIAL DIET AND DEVELOPING EAB ADULT REARING METHODS AND DEVELOPING METHODS TO INCREASE PARASITISM RATES OF DIET-REARED EAB. THE OTHER FULL-TIME TECHNICIAN WILL BE STATIONED AT THE FOREST PEST METHODS LABORATORY. THEY WILL RUN THE EXTRUDER AND PRODUCE EXTRUDED DIETS FOR TESTING IN BRIGHTON. THE PRINCIPAL INVESTIGATOR WILL, IN COOPERATION WITH THE APHIS PROJECT MANAGER, PROVIDE A DETAILED PROJECT WORK PLAN, SUPERVISE THE TECHNICIANS, PURCHASE SUPPLIES, AND WRITE REPORTS. THE TECHNICIANS, PROJECT MANAGER, AND RECIPIENT WILL HAVE WEEKLY VIRTUAL MEETINGS TO DISCUSS PROGRESS AND FUTURE PLANNING THROUGHOUT THE PROJECT. DELIVERABLES AND EXPECTED OUTCOMES: A TWIN-SCREW EXTRUDED DIET REARING METHOD TO PRODUCE ADULT EAB AND A TESTABLE PROTOCOL TO DETERMINE WHETHER DIET-REARED EAB CAN BE INCORPORATED INTO THE MASS-REARING PROCEDURES. THE RECIPIENT WILL PROVIDE ONE TECHNICIAN WHO WILL REAR BOX TREE MOTH IN CAGES, CARE FOR BOX TREE PLANTS IN A GREENHOUSE, CONDUCT THE RESEARCH ON ALTERNATE FOODS AS LAID OUT IN THE WORK PLAN, RECORD AND ENTER DATA ONTO PAPER AND ELECTRONIC DATA FILES, AND SUMMARIZE THE DATA. THE PRINCIPAL INVESTIGATOR WILL, IN COOPERATION WITH THE APHIS PROJECT MANAGER, PROVIDE A DETAILED PROJECT WORK PLAN, SUPERVISE THE TECHNICIANS, AND PURCHASE SUPPLIES. THE RECIPIENT WILL PROVIDE ONE TECHNICIAN WHO WILL REAR BOX TREE MOTH IN CAGES, CARE FOR BOX TREE PLANTS IN A GREENHOUSE, CONDUCT THE RESEARCH ON ARTIFICIAL DIET AS LAID OUT IN THE WORK PLAN, RECORD AND ENTER DATA ONTO PAPER AND ELECTRONIC DATA FILES, AND SUMMARIZE THE DATA. THIS PROJECT DEVELOPS A PROTOTYPE OF AN OPEN-SOURCE SOFTWARE TOOL TO FACILITATE STUDYING RECORDED MUSIC PERFORMANCES. THE PURPOSE OF THIS AGREEMENT IS TO GENERATE DATA THAT SUPPORTS A NEW POST-HARVEST PHYTOSANITARY IRRADIATION TREATMENT FOR TWO-SPOT COTTON LEAFHOPPER, A DESTRUCTIVE AND INVASIVE PEST OF MANY AGRICULTURAL COMMODITIES, PARTICULARLY OKRA, EGGPLANT, AND COTTON. ADULT TWO-SPOT COTTON LEAFHOPPERS CAN DEPOSIT EGGS INTO OKRA FRUIT, WHICH INTRODUCES A RISK FOR DISPERSAL FROM ITS CURRENT RANGE TO KEY COTTON GROWING REGIONS IN THE UNITED STATES. DEVELOPING A NEW TREATMENT OPTION WILL IMPROVE PLANT PROTECTION AND QUARANTINE’S ABILITY TO LIMIT TWO-SPOT COTTON LEAFHOPPER SPREAD AND PROTECT AGRICULTURAL COMMODITIES WITHIN THE UNITED STATES. ACTIVITIES TO BE PERFORMED: ALL WORK WILL BE CONDUCTED WITHIN A SECURE QUARANTINE FACILITY AT THE FOREST PEST METHODS LABORATORY IN BUZZARDS BAY, MA, AND USING A LABORATORY TWO-SPOT COTTON LEAFHOPPER COLONY. INITIAL TESTING WILL FOCUS ON IDENTIFYING AN IRRADIATION TREATMENT DOSE THAT KILLS 99.9968% OF INDIVIDUALS, A STANDARD MEASURE FOR ESTABLISHING QUARANTINE SECURITY. FOR EACH LIFE STAGE OF THE TWO-SPOT COTTON LEAFHOPPER (EGGS, NYMPHS, AND ADULTS), 1000 INDIVIDUALS WILL BE EXPOSED TO ONE OF FIVE IRRADIATION DOSES RANGING FROM 0 TO 400 GY (200 INDIVIDUALS TESTED PER TREATMENT DOSE). AFTER IRRADIATION TREATMENT, INDIVIDUAL INSECTS WILL BE HELD AND MONITORED FOR (1) SURVIVAL TO ADULT EMERGENCE AND (2) PRODUCTION OF EGGS, WHICH WILL INDICATE IF THE TREATMENT HAD A STERILIZING EFFECT. DOSE RESPONSE CURVES WILL BE USED TO DETERMINE THE IRRADIATION DOSE THAT CAUSES 99.9968% MORTALITY AND STERILITY FOR EACH LIFE STAGE. FOR THE MOST RADIO-TOLERANT LIFE STAGE, ADDITIONAL TESTING WILL BE PERFORMED TO VALIDATE THE MINIMUM IRRADIATION TREATMENT DOSE THAT PROVIDES QUARANTINE SECURITY. A MINIMUM OF 10,000 INDIVIDUALS FROM THE LABORATORY COLONY WILL BE EXPOSED TO THE CANDIDATE TREATMENT DOSE, AND INSECTS WILL AGAIN BE MONITORED FOR SURVIVAL. DELIVERABLES AND EXPECTED OUTCOMES: DATA THAT IDENTIFIES THE MINIMUM IRRADIATION TREATMENT DOSE NEEDED TO KILL 99.9968% OF TSCL INDIVIDUALS. LARGE-SCALE VALIDATION DATASET TO CONFIRM THAT THE IDENTIFIED IRRADIATION TREATMENT DOSE WILL PROVIDE ACCEPTABLE QUARANTINE SECURITY LEVELS. THESE DATA WILL SUPPORT THE ADDITION OF A NEW TREATMENT SCHEDULE FOR TWO-SPOT COTTON LEAFHOPPER TO THE USDA TREATMENT MANUAL. INTENDED BENEFICIARY(IES): FLORIDA OKRA PRODUCERS AND EXPORTERS: IN MIAMI-DADE COUNTY, FLORIDA, WHERE TWO-SPOT COTTON LEAFHOPPER PRESSURE IS HIGH, OKRA (ABELMOSCHUS ESCULENTUS) IS A PREFERRED HOST PLANT FOR TSCL. OKRA IS AN IMPORTANT CROP COMMODITY FOR THE REGION; IT IS GROWN ON AN ESTIMATED 1,000 – 1,500 ACRES ACROSS MIAMI-DADE COUNTY, AND FRESH PODS ARE SOLD NATIONWIDE. IF QUARANTINE RESTRICTIONS FOR TWO-SPOT COTTON LEAFHOPPER ARE IMPLEMENTED IN FLORIDA, HAVING DATA-SUPPORTED PHYTOSANITARY TREATMENTS WILL BE CRITICAL FOR ENSURING THAT THIS MARKET REMAINS OPEN BOTH DOMESTICALLY AND INTERNATIONALLY. COTTON PRODUCERS: TWO-SPOT COTTON LEAFHOPPER IS A DEVASTATING PEST OF MULTIPLE CROPS, INCLUDING COTTON. PREVENTING ITS SPREAD WILL BENEFIT COTTON PRODUCERS WHO OPERATE OUTSIDE OF ITS CURRENT RANGE.
