THE HUMAN DRD4 GENE IS HIGHLY POLYMORPHIC, WITH DIFFERENT VARIANTS ASSOCIATED WITH VARIOUS NEUROPSYCHIATRIC DISORDERS. IN PARTICULAR, VNTR VARIANTS (THE THREE MAJOR ONES ARE TERMED D4.2R, D4.4R, AND D4.7R) ARE DIFFERENTIALLY ASSOCIATED WITH ADHD AND SUBSTANCE USE DISORDERS, WITH THE D4.7R RELIABLY ASSOCIATED WITH INCREASED PREVALENCE OF ADHD AND SUBSTANCE USE DISORDERS (SUDS). DETERMINING PRECISE PHYSIOLOGICAL ROLES OF D4R SIGNALING AS WELL AS UNDERSTANDING THE CONSEQUENCES OF D4R VARIANTS IN CONTRIBUTING TO DISEASE STATES HAS BEEN HAMPERED BY THE LACK OF SUITABLE COMPOUNDS WITH HIGH AFFINITY, SUFFICIENT SELECTIVITY, CHARACTERIZED AGONIST-ANTAGONIST RESPONSES, AND/OR SUITABLE IN VIVO ACTIVITY. CURRENTLY THERE ARE NO KNOWN COMPOUNDS WITH DIFFERENTIAL ACTIVITY AT D4R VARIANTS. THEREFORE, WE SEEK TO DEVELOP NEW, OPTIMIZED D4R LIGANDS WITH VARIANT-SPECIFIC ACTIVITY TO EXPAND OUR UNDERSTANDING OF D4R FUNCTION AND IN THE DEVELOPMENT OF POTENTIAL THERAPEUTIC COMPOUNDS. D4.7R HAS ALTERED ABILITY TO HETEROMERIZE WITH DOPAMINE D2 RECEPTORS (D2RS) OR A2A ADRENERGIC RECEPTORS (A2ARS), WHEN COMPARED TO THE MORE COMMON D4.4R. OUR TEAM HAS DISCOVERED THAT D4R AGONIST A-412997 PRODUCES DIFFERENTIAL SIGNALING AT D4.7R OR D4.4R HETEROMERIZED WITH D2R OR A2AR, AND DIFFERENTIAL SIGNALING AT GAI1 VERSUS GAO1. WE WILL LEVERAGE OUR EXISTING LIBRARY OF A-412997 ANALOGS, AND CREATE NEW RATIONALLY DESIGNED ANALOGS, TO IDENTIFY NEW PHARMACOLOGICAL TOOLS WILL HAVE THE POTENTIAL TO SURMOUNT CURRENT LIMITATIONS ON THE UNDERSTANDING OF THE FUNCTIONAL SIGNIFICANCE OF D4R VNTR POLYMORPHISMS. NOVEL D4R LIGANDS ALSO HOLD THE POTENTIAL TO BE USED AS PHARMACOTHERAPEUTICS FOR SUD. SINCE THERE ARE NO FDA-APPROVED MEDICATIONS FOR THE TREATMENT OF PSYCHOSTIMULANT USE DISORDERS, AND NO FDA-APPROVED MEDICATIONS SELECTIVE FOR D4R, NOVEL PHARMACOLOGICAL TOOLS TARGETING D4R VARIANTS CAN ADDRESS MULTIPLE NEEDS. THE PROPOSED RESEARCH SEEKS TO DESIGN, SYNTHESIZE, AND EVALUATE NEW LIGANDS USING COMPUTATIONAL MODELING APPROACHES THAT EXPLOIT UNDEREXPLORED REGIONS WITHIN THE D4R PROTEIN STRUCTURE TO DEVELOP SELECTIVE D4.2R, D4.4R, AND D4.7R LIGANDS.

EQUIPMENT: MRI: TRACK 1 ACQUISITION OF A MASKLESS PHOTOLITHOGRAPHY INSTRUMENT FOR UNDERGRADUATE RESEARCH AND TEACHING AT HIGH POINT UNIVERSITY -THIS MAJOR RESEARCH INSTRUMENTATION (MRI) AWARD IS FOR THE ACQUISITION OF A MASKLESS PHOTOLITHOGRAPHY TOOL FOR USE BY FACULTY AND UNDERGRADUATE RESEARCHERS IN THE DEPARTMENTS OF CHEMISTRY, PHYSICS AND ENGINEERING AT HIGH POINT UNIVERSITY (HPU) (NC) IN COLLABORATION WITH FACULTY AND UNDERGRADUATE RESEARCHERS FROM THE DEPARTMENT OF CHEMISTRY AT THE UNIVERSITY OF RICHMOND (VA), DEPARTMENT OF PHYSICS AT JAMES MADISON UNIVERSITY (VA) AND THE DEPARTMENT OF BIOLOGY AT THE UNIVERSITY OF VIRGINIA ALONG WITH OTHER REGIONAL UNDERGRADUATE INSTITUTIONAL PARTNERS. THE ABILITY TO RAPIDLY PROTOTYPE STRUCTURES AND DEVICES WITH DIMENSIONS SMALLER THAN A CELL IS A KEY ENABLING CAPABILITY FOR A VARIETY OF TECHNOLOGIES, AND THE TRAINING OF UNDERGRADUATE CHEMISTRY, PHYSICS AND ENGINEERING STUDENTS WILL HELP EXPAND THE PIPELINE OF FUTURE NANOTECHNOLOGISTS. MICROLITHOGRAPHY IS A CRITICAL PROCESSING STEP IN THE PRODUCTION OF INTEGRATED CIRCUITS AND DEVICES, AND DEVELOPING A TRAINED WORKFORCE IN NANOFABRICATION IS VITAL TO THE ECONOMIC AND NATIONAL SECURITY INTERESTS OF THE UNITED STATES. FINALLY, STRUCTURES CREATED USING THIS TOOL WILL BE USED IN CONJUNCTION WITH THE IMAGING LABORATORY AT HPU FOR SCIENTIFIC OUTREACH TO LOCAL SCHOOLS AND MUSEUMS IN THE PIEDMONT TRIAD REGION OF NORTH CAROLINA TO SPARK AN INTEREST IN THE NEXT GENERATION OF LEARNERS IN THE MICRO AND NANOSCOPIC WORLD. THIS INSTRUMENT WILL BE USED TO TRAIN UNDERGRADUATE STUDENTS HOW TO RAPIDLY PROTOTYPE MICRO- AND NANOSTRUCTURES USED IN CHEMICAL SENSING, OPTICS, NEUROSCIENCE STUDIES, SURFACE ENGINEERING AND BIOMEDICAL DEVICES. THIS MAJOR RESEARCH INSTRUMENTATION AWARD IS FOR THE ACQUISITION OF A DIRECT-WRITE MASKLESS PHOTOLITHOGRAPHY TOOL FOR USE BY FACULTY AND UNDERGRADUATE RESEARCHERS AT HIGH POINT UNIVERSITY IN COLLABORATION WITH FACULTY AND UNDERGRADUATE RESEARCHERS AT JAMES MADISON UNIVERSITY AND THE AND THE UNIVERSITY OF VIRGINIA. THIS LITHOGRAPHY TOOL WILL ENABLE THE RAPID PROTOTYPING OF MICROSTRUCTURES DOWN TO 0.6 ?M IN SUPPORT OF THE RESEARCHERS ON THE PROJECT TEAM AND WILL BE MADE AVAILABLE TO OTHER INVESTIGATORS AT REGIONAL PRIMARILY UNDERGRADUATE INSTITUTIONS (PUIS) THROUGH THE PRIMARILY UNDERGRADUATE NANOMATERIALS COOPERATIVE (PUNC) AS A RESOURCE TO HELP FURTHER DEVELOP THEIR MICROFABRICATION RESEARCH AND EDUCATIONAL PROJECTS. THIS PROJECT WILL DIRECTLY ENABLE THE RAPID PROTOTYPING OF MICROSTRUCTURES IN RESEARCH PROJECTS RANGING FROM POLYMER MATERIALS SCIENCE TO NEUROSCIENCE. SPECIFICALLY, THESE PROJECTS INCLUDE THE RAPID PROTOTYPING OF SILICONE-BASED MICROSTRUCTURES FOR MICROFLUIDIC APPLICATIONS AND BIOFILM PREVENTION, MICROCONTACT PRINTING (?-CP) OF SURFACES FOR GRAFTING-FROM CONDUCTIVE POLYMER SYNTHESIS, MICROFABRICATION OF SILICON-BASED PHOTODETECTORS, DESIGN AND PROTOTYPING OF NANOPARTICLE-ENHANCED CHEMICAL SENSING PLATFORMS, AND RAPID PROTOTYPING OF MICROFLUIDIC STRUCTURES FOR NEUROSCIENCE STUDIES. DIFFERENT TYPES OF IMAGES HAVE BEEN EMPLOYED IN VARIOUS AREAS OF STUDY. ONE EXAMPLE IS THE HYPERSPECTRAL IMAGES, WHICH HAVE HIGHER RESOLUTION WHEN COMPARED TO CONVENTIONAL CAMERA IMAGES. ANALYZING SUCH IMAGES HAS ITS CHALLENGES. FOR INSTANCE, IT IS COMPUTATIONALLY DEMANDING, AND TRADITIONAL METHODS HAVE SOME LIMITATIONS. THIS PROJECT PROVIDES AN EFFICIENT SOLUTION TO ANALYZE SUCH IMAGES, BY EXPLOITING HIGH-PERFORMANCE COMPUTING TOOLS AND MACHINE LEARNING TECHNIQUES. THE RESULTING METHODS ARE APPLIED TO IMAGE-BASED ATMOSPHERIC CLOUD DETECTION. THE PROJECT DEVELOPS A REAL TIME, MULTI-LAYER, AND MODULAR SEGMENTATION FRAMEWORK FOR HYPERSPECTRAL IMAGES. THE DEVELOPED FRAMEWORK AUTOMATICALLY IDENTIFIES VARIOUS REGIONS WITHIN A HYPERSPECTRAL IMAGE BY CLASSIFYING EACH PIXEL OF THE IMAGE AND ASSOCIATING THEM TO CLASS SEGMENTS. THE DEVELOPED SYSTEM IS MULTI-LAYER, WHERE EACH LAYER?S RESPONSIBILITY IS TO PERFORM AN OPERATION ON ITS INPUT, GENERATE REGION CLASSIFICATION DATA, AND PASS THE RESULTANT OUTPUT TO THE NEXT LAYER. IMPORTANTLY, EACH LAYER ANALYZES ITS INPUT FROM DISTINCT VIEWPOINTS, UTILIZING SPECTRAL AND SPATIAL DATA, RESULTING IN A MULTI-LAYER FRAMEWORK WHERE THE LAYERS COMPLEMENT EACH OTHER. ALSO, THIS PROJECT AIMS TO PROVIDE AN OPTIMIZED HIGH-PERFORMANCE (SPEED-UP AND ACCURACY) COMPUTATIONAL TOOL FOR REAL-TIME HYPERSPECTRAL IMAGE ANALYSIS. THIS IS ACHIEVED BY ADAPTING THE ALGORITHMS USED IN THE DIFFERENT PARTS OF THE MODEL FOR PARALLEL PROCESSING.
