OVERCOMING THE DIFFERENCES IN POST-TRANSLATIONAL PROTEIN MODIFICATION MECHANISMS BETWEEN PROKARYOTES AND EUKARYOTES, SUCH AS GLYCOSYLATION AND DISULFIDE BOND FORMATION, HAVE BEEN A TECHNOLOGICAL OBSTACLE PREVENTING THE STRUCTURALLY ACCURATE ‘ANTIGENIC’ EXPRESSION OF VIRAL EPITOPES IN BACTERIA. THIS PROBLEM IS EXACERBATED IN THE CASE OF INFLUENZA’S HEMAGGLUTININ (HA) SPIKE PROTEIN AS ITS CORRECT ANTIGENIC STRUCTURE IS INFLUENCED BY TRIMERIZATION, GLYCOSYLATION, AND DI-SULFIDE BOND FORMATION. IF THIS SYSTEM STRUCTURAL INCOMPATIBILITY CAN BE OVERCOME, PROBIOTIC BACTERIA COULD BECOME A LOW-COST, SHELF-STABLE ANIMAL VACCINE THAT COULD BE ADMINISTERED THROUGH ANIMAL FEED AND ELICIT PROTECTIVE ANTIBODIES AGAINST INFLUENZA IN GENERAL AND HIGHLY PATHOGENIC AVIAN INFLUENZA (H5N1) IN PARTICULAR. INDEED, A MULTIVALENT VACCINE MADE UP OF MIXES OF ENGINEERED PROBIOTIC BACTERIA IS POSSIBLE. DURING THIS PROJECT, WE WILL DEVELOP AND COMBINE METHODS FROM ARTIFICIAL INTELLIGENCE AND BIOMOLECULAR MODELING, SUCH AS MULTI-AGENT FRAMEWORK BASED ON LARGE LANGUAGE MODELS (LLMS) AND THE NOBEL-PRIZEWINNING ALPHAFOLD PROGRAM, TO MINE HISTORICAL HA SEQUENCE AND ANTIGENIC STRUCTURE INFORMATION. THIS WILL ENABLE THE IN SILICO EVALUATION AND MODELING OF POTENTIAL MUTATIONS OR AMINO ACID SUBSTITUTIONS NEEDED TO COMPENSATE FOR THE DIFFERENTIAL BACTERIA PROTEIN EXPRESSION SYSTEM TO REPRODUCE KEY ANTIGENIC STRUCTURES. THESE CANDIDATE SEQUENCES MAY BE IN THE FORM OF A MONOMER, TRIMERS, ANTIGEN-SCAFFOLD FUSIONS, AND FUSIONS TO ORTHOGONAL BACTERIAL MEMBRANE PROTEIN DOMAINS. SUCCESSFUL IN SILICO-DEVELOPED HA SEQUENCES WILL BE SYNTHESIZED AS DEGENERATE OLIGO POOLS, CLONED INTO EXPRESSION PLASMIDS, AND TESTED IN PARALLEL IN VITRO WITH A DEVELOPED HIGH THROUGHPUT BACTERIAL OUTER MEMBRANE PROTEIN PLASMID-BASED EXPRESSION AND SCREENING SYSTEM. THIS SYSTEM CAN SCREEN MILLIONS OF CANDIDATE SEQUENCES SIMULTANEOUSLY THROUGH DOWN SELECTION WITH KNOWN HA-INHIBITING ANTIBODIES. PROTEIN SEQUENCES THAT SUCCESSFULLY PRESENT ANTIGEN IN THE PLASMID EXPRESSION SYSTEM WILL THEN BE INSERTED DIRECTLY INTO THE BACTERIAL GENOME ELIMINATING THE NEED FOR PLASMIDS, WHICH CARRY UNWANTED ARTIFICIAL ANTIBIOTIC MARKERS AND OTHER UNNATURAL GENETIC SEQUENCES. THE ULTIMATE OBJECTIVE OF THIS PROJECT IS TO DEVELOP A POULTRY VACCINE PRODUCTION SYSTEM, THAT IS LOW COST WITH A LONG SHELF LIFE AT AMBIENT TEMPERATURES AND THAT CAN BE RAPIDLY MANUFACTURED AND DISTRIBUTED AS A PROBIOTIC FEED SUPPLEMENT. THIS SYSTEM WOULD BE HIGHLY MULTIVALENT AND RAPIDLY RECONFIGURABLE FOR NEW OR EMERGING VIRAL THREATS.

COLLABORATIVE RESEARCH: CLOUD TOP DISCHARGES AND THEIR PARENT STORMS -THUNDERSTORMS ARE HIGHLY-ELECTRIFIED PHENOMENA WHERE LIGHTNING IS FORMED FROM THE SEPARATION OF ELECTRICAL CHARGE WITHIN THE CLOUD. NOT ONLY DO THUNDERSTORMS PRODUCE CLOUD-TO-GROUND AND CLOUD-TO-CLOUD LIGHTNING, BUT THUNDERSTORMS ALSO CAN PRODUCE UPWARD-POINTED ELECTRICAL DISCHARGES FROM THE TOPS OF THE CLOUDS. THESE ARE KNOWN AS CLOUD TOP DISCHARGES AND HAVE MANY DIFFERENT NAMES BASED ON THEIR SIZE AND APPEARANCE, SUCH AS SPRITES, PIXIES, AND JETS. WHILE THESE DISCHARGES HAVE BEEN PHOTOGRAPHED, THERE ARE VERY FEW SCIENTIFIC RESEARCH-QUALITY DATA AVAILABLE FOR THESE EVENTS, AND A VARIETY OF QUESTIONS REMAIN UNANSWERED. THIS AWARD WILL INVOLVE THE DEVELOPMENT OF TWO NEW INSTRUMENTS AND A MULTI-YEAR OBSERVATIONAL EFFORT TO PROVIDE THE MOST COMPREHENSIVE DATA ON CLOUD TOP DISCHARGES AVAILABLE. THE RELEVANCE OF THIS PROJECT TO SOCIETY IS THAT UPWARD ELECTRICAL DISCHARGES ARE STRONG PRODUCERS OF NOX WHICH CAN AFFECT STRATOSPHERIC OZONE CHEMISTRY. THIS AWARD ALSO INCLUDES OUTREACH TO RURAL SCHOOLS IN GEORGIA TO PROVIDE SCIENCE EDUCATION ON THE TOPICS OF ELECTRICITY AND MAGNETISM. THE OVERALL OBJECTIVE OF THIS PROJECT IS TO INVESTIGATE THE ELECTRICAL NATURE OF CLOUD TOP DISCHARGES (CTDS) SUCH AS SPRITES, GNOMES, PIXIES, STARTERS, JETS AND GIGANTIC JETS AND HOW THUNDERSTORMS PRODUCE THESE UNIQUE EVENTS. OBSERVATIONS OF THESE CTDS ARE RARE DUE TO PRIOR OBSERVATIONAL CONSTRAINTS. IN THIS PROJECT, THE RESEARCH TEAM WILL DEVELOP AND DEPLOY A NEW GROUND-BASED SPECTROSCOPIC IMAGING SYSTEM CONSISTING OF TWO BLUE-SENSITIVE HIGH-SPEED CAMERAS. THE BLUE ASPECT IS IMPORTANT BECAUSE CTDS ARE CHARACTERIZED BY THEIR PREDOMINANTLY BLUE COLOR, BUT BLUE AND ULTRAVIOLET (UV) LIGHT IS HEAVILY SCATTERED BY THE LOWER ATMOSPHERE AND MOST PRIOR OPTICAL IMAGERS WERE NOT SENSITIVE TO BLUE/UV LIGHT. THE NEW SYSTEM WILL BE DEPLOYED IN TEXAS AND COMBINED WITH ADDITIONAL OBSERVATIONAL INFORMATION FROM VERY LOW FREQUENCY (VLF)/LOW FREQUENCY (LF) RADIO RECEIVERS, LIGHTNING MAPPING ARRAYS (LMAS), AND TRADITIONAL METEOROLOGICAL DATA TO ANSWER KEY QUESTIONS ABOUT THE ELECTRICAL AND MORPHOLOGICAL PROPERTIES OF CTDS AND THE CHARACTERIZATION OF CTD THUNDERSTORM CHARGE STRUCTURES. SPECIFIC RESEARCH QUESTIONS INCLUDE: ? WHAT ARE THE LEADER AND STREAMER DYNAMICS OF CTDS, SUCH AS EVOLUTION OF THERMAL TEMPERATURE FOR LEADERS AND ELECTRON ENERGY FOR STREAMERS? ? WHAT ARE QUANTITATIVE CHARACTERISTICS OF THE LEADER-TO-STREAMER TRANSITION REGION? ? HOW IS THE VERY HIGH FREQUENCY (VHF) MEASURED BY LMAS PRODUCED ABOVE THE CLOUD? ? WHAT PARAMETERS DICTATE THE TERMINAL ALTITUDE OF A CTD? ? WHAT ARE THE PARENT STORM CHARGE STRUCTURES AND WHAT METEOROLOGICAL PROCESSES FORM THEM? ? WHAT IS THE RELATIONSHIP BETWEEN CLOUD TOP MIXING AND DIVERGENCE, THE THUNDERSTORM CHARGE STRUCTURE, AND EVENT FORMATION? ? HOW DOES THE STORM STRUCTURE VARY FOR DIFFERENT CTDS? THIS AWARD REFLECTS NSF'S STATUTORY MISSION AND HAS BEEN DEEMED WORTHY OF SUPPORT THROUGH EVALUATION USING THE FOUNDATION'S INTELLECTUAL MERIT AND BROADER IMPACTS REVIEW CRITERIA.- SUBAWARDS ARE NOT PLANNED FOR THIS AWARD.

COLLABORATIVE RESEARCH: CEDAR: INVESTIGATION OF GIGANTIC JETS, THEIR IONOSPHERIC EFFECTS, AND HOW THEY COUPLE THE TROPOSPHERE AND IONOSPHERE -THIS AWARD SEEKS TO ADVANCE THE SCIENCE OF HOW CLOUD-TO-IONOSPHERE ELECTRICAL DISCHARGES KNOWN AS GIGANTIC JETS (GJS) COUPLE THE TROPOSPHERE AND IONOSPHERE. GJS ARE LARGE ELECTRICAL DISCHARGES THAT BEGIN INSIDE A THUNDERSTORM, EMERGE FROM THE TOP OF THE CLOUD, AND CONNECT WITH THE LOWER IONOSPHERE (80 -100 KM ALTITUDE). THEY ARE CAPABLE OF TRANSFERRING HUNDREDS OF COULOMBS OF CHARGE (10 TIMES MORE THAN TYPICAL LIGHTNING) BETWEEN THE TROPOSPHERE AND IONOSPHERE, DIRECTLY COUPLING THESE ATMOSPHERIC REGIONS. DUE TO INFREQUENT OBSERVATIONS FROM PAST OBSERVATIONAL TECHNIQUES, MANY MYSTERIES REMAIN REGARDING THESE EVENTS, SUCH AS THEIR EFFECT ON THE GLOBAL ELECTRIC CIRCUIT (GEC), HOW THEY PERTURB THE UPPER ATMOSPHERE, AND HOW THEY PROPAGATE TO SUCH HIGH ALTITUDES ABOVE THE CLOUD TOP. THE WORK UNDER THIS AWARD WILL INVOLVE DETECTING GJS ON NEARLY A HEMISPHERIC SCALE USING OPTICAL DATA FROM THE GEOSTATIONARY LIGHTNING MAPPERS (GLM) AND MACHINE LEARNING TECHNIQUES, IN ADDITION TO MULTI-STEP VALIDATION. THE DETECTION PIPELINE WILL HAVE THE CAPABILITY TO IDENTIFY THOUSANDS OF GJS PER YEAR, ORDERS OF MAGNITUDE MORE THAN PREVIOUS OBSERVATIONS. THE LARGE-SCALE DATABASE OF DETECTIONS WILL BE MADE PUBLICLY AVAILABLE AND WIDELY DISSEMINATED, ALLOWING HIGH IMPACT ON OTHER FIELDS OF RESEARCH SUCH AS AERONOMY (GEC), ATMOSPHERIC CHEMISTRY, AND METEOROLOGY. AS PART OF THE BROADER EDUCATIONAL OUTREACH FOR THIS AWARD, K-12 TEACHERS FROM TITLE 1 SCHOOLS (THOSE WITH HIGH PERCENTAGES OF CHILDREN IN POVERTY) VIA AN EXISTING NSF-SPONSORED PROGRAM (RESEARCH EXPERIENCE FOR TEACHERS) WILL WORK ON A RESEARCH PROJECT EACH SUMMER AND INCORPORATE WHAT THEY LEARNED IN THEIR CURRICULUM. GRADUATE STUDENTS WILL BE INVOLVED IN THE PROJECT AS WELL. THE THREE MAIN GOALS OF THIS PROJECT ARE: 1) CONSTRUCT CLIMATOLOGIES OF GJS AFTER DEVELOPING A ROBUST PIPELINE TO DETECT THEM BY THE THOUSANDS USING GLM DATA AND A MACHINE LEARNING CLASSIER. 2) QUANTIFY WHETHER AND HOW GJS PERTURB THE D-REGION IONOSPHERE. 3) INVESTIGATE THE PHYSICAL CHARACTERISTICS OF GJS. THE LARGE-SCALE DETECTION WILL BE PERFORMED BY A PIPELINE THAT USES GLM IN CONJUNCTION WITH MACHINE LEARNING ALGORITHMS, AND MULTI-STEP VALIDATION WITH GROUND-BASED RADIO NETWORKS. THE MULTI-STEP VALIDATION CONSISTS OF: CORRELATING POTENTIAL GJS WITH A LOW FREQUENCY (LF) LIGHTNING NETWORK IN SPACE AND TIME TO FILTER OUT NON-LIGHTNING EVENTS; VALIDATING WITH A STEREO ALTITUDE GLM MODEL (DEVELOPED AS PART OF THIS WORK); AND VALIDATING WITH AN ELF RADIO MODEL (DEVELOPED AS PART OF THIS WORK). USING VLF REMOTE SENSING ON A SUBSET OF EVENTS THAT PASS WITHIN AN EXISTING VLF RADIO NETWORK, CHANGES TO THE ELECTRON DENSITY PROFILE IN THE D-REGION WILL BE QUANTIFIED USING TEMPORAL-SPATIAL MAPPING METHODS AND CORRELATED TO PROPERTIES MEASURED BY THE DUKE ELF RADIO NETWORK. THE DETECTIONS WILL ALSO BE CORRELATED WITH GROUND-BASED INSTRUMENTS SUCH AS VERY HIGH FREQUENCY (VHF) LIGHTNING MAPPING ARRAYS (LMA) AND WITH INSTRUMENTS IN LOW EARTH ORBIT SUCH AS THE ATMOSPHERIC SPACE INTERACTIONS MONITOR (ASIM) TO UNDERSTAND THE DISCHARGE PHYSICS OF THE EVENTS, SUCH AS THE LEADER AND STREAMER PORTIONS OF THE DISCHARGE AND IF THEY ARE ASSOCIATED WITH GAMMA RAYS. THE RESEARCH SUPPORTED BY THIS AWARD WILL SIGNIFICANTLY ADVANCE THE SCIENCE OF HOW THE IONOSPHERE RESPONDS TO ELECTRICAL IMPULSES FROM BELOW. THIS PROJECT IS CO-FUNDED BY A COLLABORATION BETWEEN THE DIRECTORATE FOR GEOSCIENCES AND OFFICE OF ADVANCED CYBERINFRASTRUCTURE TO SUPPORT AI/ML AND OPEN SCIENCE ACTIVITIES IN THE GEOSCIENCES. THIS AWARD REFLECTS NSF'S STATUTORY MISSION AND HAS BEEN DEEMED WORTHY OF SUPPORT THROUGH EVALUATION USING THE FOUNDATION'S INTELLECTUAL MERIT AND BROADER IMPACTS REVIEW CRITERIA.

THIS RESEARCH WILL DEVELOP A TRANSLATION TO THE ALLOY ANALYZER (A LOGICAL SOLVER) THAT VALIDATES THIS APPROACH AND PROVIDES A SECOND SOFTWARE IMPLEMENTATION REQUIRED BY THE STANDARDS DEVELOPING ORGANIZATION BEFORE PROCEEDING WITH THE UPDATE TO SYSML.ACTIVITIES TO BE PERFORMED: THE RESEARCH WILL IMPROVE INTEGRATED SYSTEMS ANALYSIS FOR SMART MANUFACTURING SYSTEMS THROUGH THE FOLLOWING ACTIVITIES:1.DEVELOP AN AUTOMATED TRANSLATOR THAT READS IN SYSML-SPECIFIED FILE AND CONVERTS IT INTO AN ALLOY MODEL.2.INVESTIGATE THE SCALABILITY/TRACTABILITY OF SYSML MODELS TRANSLATED TO ALLOY.3.INVESTIGATE POSSIBILITIES FOR ADDITIONAL MODELING CAPABILITIES.4.DISSEMINATE FINDINGS AND MODELING RATIONALE.EXPECTED OUTCOMES: THE WORK WILL STRENGTHEN THE FOUNDATIONS OF SYSML FOR IMPROVED ANALYSIS AND INTEROPERABILITY, THROUGH CLEAR AND VERIFIABLE RULES REGARDING ALLOWABLE BEHAVIOR AND THE EXPECTED TRAJECTORY OF SYSTEM STATES. INTENDED BENEFICIARIES: THE TECHNOLOGY THAT IS DEVELOPED THROUGH THIS PROJECT WILL SUPPORT AN UPDATE OF THE SYSML STANDARD THAT WILL HAVE DIRECT RELEVANCE TO ENTERPRISES THAT DESIGN, MANUFACTURE AND MAINTAIN COMPLEX CYBER-PHYSICAL SYSTEMS. SYSML IS WIDELY USED AMONG AEROSPACE AND DEFENSE ENTERPRISES THAT BUILD COMPLEX, LONG-LIVED SYSTEMS. ONTOLOGICAL BEHAVIOR MODELLING HAS THE POTENTIAL TO ANALYZE MANUFACTURING SCENARIOS AT A MUCH HIGHER LEVEL THAN IS CURRENTLY POSSIBLE, ALLOWING COSTLY MISTAKES TO BE FOUND SOONER. THIS WORK ENABLES EFFECTIVE CAPTURE OF THE REASONING BEHIND DESIGN DECISIONS, WHICH IS CRITICAL FOR LONG-LIVED SYSTEMS. THE PROPOSED WORK WILL LEVERAGE THE LONG-TERM INVESTMENT THAT INDUSTRY HAS ALREADY MADE IN SYSML TO STRENGTHEN THE FOUNDATIONS OF SYSML FOR IMPROVED ANALYSIS AND INTEROPERABILITY. THIS NEW TECHNOLOGY WILL ENABLE SYSTEMS ENGINEERS USING SYSML TO PRODUCE SAFE, SECURE CYBER-PHYSICAL SYSTEMS OUT OF COMPONENTS FROM MULTIPLE SOURCES, RESULTING IN SIGNIFICANT PUBLIC BENEFIT OVER MANY YEARS.SUBRECIPIENT ACTIVITIES: THERE ARE NO SUBAWARDS IN THIS PROJECT.

THE FREQUENT LAKE-EFFECT SNOWSTORMS IN THIS AREA PRODUCE SEVERAL THUNDERSNOW EVENTS EACH YEAR, AND THIS PROJECT WILL MAKE THE FIRST EVER MEASUREMENTS OF THE ELECTRICAL STRUCTURE OF LAKE-EFFECT SNOW CLOUDS AND INFER HOW LIGHTNING WITHIN THEM IS RELATED TO PRECIPITATION PROCESSES IN THE CLOUDS. DURING ANOTHER RECENT NSF-FUNDED PROJECT, THE ONTARIO WINTER LAKE-EFFECT SYSTEMS (OWLES) FIELD CAMPAIGN DURING THE 2013-14 WINTER SEASON, ALL LAKE-EFFECT LIGHTNING OCCURRED INLAND AND MANY FLASHES WERE ASSOCIATED WITH THE MAPLE RIDGE WIND FARM, COMPOSED OF APPROXIMATELY 200 TURBINES OVER 100 M TALL. LIGHTNING IS A SIGNIFICANT CAUSE OF WIND TURBINE DAMAGE (E.G., TO THE BLADES), INCREASING WIND ENERGY GENERATION COSTS. THE STUDY REGION IS THEREFORE AN IDEAL NATURAL LABORATORY WITHIN WHICH BASIC UNDERSTANDING OF THE ELECTRICAL STRUCTURE OF CLOUDS CAN BE ADVANCED WHILE ALSO IMPROVING FORECASTS OF SUCH EVENTS AND UNDERSTANDING THEIR IMPACTS ON ENERGY INFRASTRUCTURE. THIS GRANT WILL INVOLVE MANY UNDERGRADUATE AND GRADUATE STUDENTS IN THE COLLECTION AND ANALYSIS OF DATA, DEVELOPING MEASUREMENT, INSTRUMENTATION AND DATA ANALYSIS SKILLS WHILE INSPIRING THEIR FURTHER EDUCATION AND INTEREST IN RESEARCH CAREERS. THE LAKE-EFFECT ELECTRIFICATION (LEE) PROJECT IS FOCUSED OVER AND EAST (THE TYPICAL DOWNWIND/LEE SIDE) OF LAKE ONTARIO DURING THE COOL SEASON. PROJECT LEE AIMS TO DOCUMENT, FOR THE FIRST TIME, THE TOTAL LIGHTNING AND ELECTRICAL CHARGE STRUCTURES OF LAKE-EFFECT STORMS AND THE ASSOCIATED STORM ENVIRONMENT USING A LIGHTNING MAPPING ARRAY (LMA), A DUAL-POLARIZATION X-BAND RADAR, AND BALLOON SOUNDINGS THAT WILL MEASURE VERTICAL PROFILES OF TEMPERATURE, HUMIDITY, WIND, ELECTRIC FIELD, AND HYDROMETEOR TYPES. PREVIOUS WORK HAS SHOWN THAT THE GREAT LAKES, ESPECIALLY LAKE ONTARIO, INITIATE LIGHTNING IN A MIX OF PRECIPITATION TYPES DURING LAKE-EFFECT STORMS. MOST OF THE LAKE ONTARIO LIGHTNING OCCURS DURING SINGLE, LONG-AXIS PRECIPITATION BANDS. SEVERAL QUESTIONS STILL REMAIN, SUCH AS EXPLAINING THE PREPONDERANCE OF POSITIVE POLARITY LIGHTNING IN SOME LAKE-EFFECT AND SIMILAR SEA-EFFECT STORMS (E.G., JAPAN), AND WHY THERE HAS BEEN A CLIMATOLOGICAL SHIFT IN MAXIMUM LAKE-EFFECT LIGHTNING OCCURRENCE FROM OVER LAKE ONTARIO TO FARTHER INLAND. THIS IS LIKELY DUE TO THE RECENT WIND FARM CONSTRUCTION IN THIS AREA, BUT THERE ARE STILL MANY UNKNOWNS ON HOW THESE TURBINES PRODUCE LIGHTNING. PROJECT LEE ALSO AFFORDS THE OPPORTUNITY TO IMPROVE OBSERVATIONS OF CONVECTIVE-TO-STRATIFORM ELECTRICAL DEVELOPMENT DUE TO THE SHALLOWNESS OF LAKE-EFFECT STORMS AND THE PROXIMITY OF THESE PROCESSES TO THE GROUND. FINALLY, LAKE-EFFECT STORM CONDITIONS REPRESENT MINIMAL THRESHOLDS FOR LIGHTNING INITIATION AS MANY OF THESE STORMS DO NOT PRODUCE LIGHTNING. THIS AWARD REFLECTS NSF'S STATUTORY MISSION AND HAS BEEN DEEMED WORTHY OF SUPPORT THROUGH EVALUATION USING THE FOUNDATION'S INTELLECTUAL MERIT AND BROADER IMPACTS REVIEW CRITERIA.
