A TOTAL OF 24 SCHOLARS PURSUING BACHELOR OF SCIENCE DEGREES IN APPLIED MATH, AQUATIC AND MARINE BIOLOGY, BIOCHEMISTRY, BIOLOGY, CHEMISTRY, COMPUTER INFORMATION SYSTEMS, COMPUTER SCIENCE, CYBERSECURITY, ENVIRONMENTAL SCIENCE, MATHEMATICS, MOLECULAR AND CELLULAR BIOLOGY, AND PHYSICS WILL RECEIVE SCHOLARSHIPS AVERAGING $12,500 PER YEAR FOR UP TO FIVE YEARS. SCHOLARS WILL RECEIVE FACULTY MENTORING, AND THE PROJECT WILL BUILD STRONG SCHOLAR COHORTS THROUGH PARTICIPATION IN A SUMMER BRIDGE PROGRAM, FRESHMAN AND JUNIOR SEMINAR COURSES, A STEM COLLOQUIUM, AND AN EXPLORATORY CURRICULUM. ADDITIONAL ACTIVITIES FOR SCHOLARS INCLUDE SUMMER RESEARCH EXPLORATION AND TUTORING SUPPORT. THE OVERALL GOAL OF THIS TRACK 2 SCHOLARSHIPS IN STEM PROJECT IS TO INCREASE STEM DEGREE COMPLETION OF ACADEMICALLY TALENTED, LOW-INCOME UNDERGRADUATE STUDENTS WITH DEMONSTRATED FINANCIAL NEED. THERE IS A SIGNIFICANT NATIONAL NEED TO GROW THE STEM WORKFORCE AND NURTURE KEY TALENT THAT WILL ENSURE ECONOMIC COMPETITIVENESS AND PROVIDE DOMESTIC LEADERSHIP ACROSS CRITICAL SECTORS. THIS PROJECT DIRECTLY SPEAKS TO THIS NEED BY SUPPORTING STEM STUDENT SUCCESS, WHICH WILL STRENGTHEN THE WORKFORCE IN BIOTECHNOLOGY, NANOTECHNOLOGY, DATA SCIENCE, AND OTHER KEY AREAS OF NEED. THE PROJECT WILL BE ASSESSED BY AN EXPERIENCED EVALUATOR THAT WILL INVESTIGATE THE EFFECTIVENESS OF THE PROPOSED INTERVENTIONS ON GRADUATION, RETENTION, AND SELF-EFFICACY OF THE S-STEM SCHOLARS COMPARED TO A MATCHED GROUP, AND THE DATA GENERATED WILL CONTRIBUTE TO THE KNOWLEDGE BASE REGARDING EFFECTIVE STRATEGIES TO SUPPORT TALENTED, LOW-INCOME STUDENTS IN STEM. SCC-CIVIC-FA TRACK A: ASSESSING THE PERFORMANCE OF GREEN STORMWATER INFRASTRUCTURE FOR CLIMATE ADAPTATION AND COASTAL RESILIENCE IN THE CITY OF CAPE CANAVERAL, FLORIDA -LIKE MANY COASTAL COMMUNITIES, THE CITY OF CAPE CANAVERAL, FLORIDA IS CONFRONTED WITH THE STRESSORS OF COASTAL FLOODING AND HARMFUL ALGAL BLOOMS (HABS) IN NEARSHORE WATERS. ANTIQUATED INFRASTRUCTURE IN THE BUILT ENVIRONMENT AND ACCELERATING CLIMATE CHANGE ARE TOGETHER AMPLIFYING THESE THREATS, WITH DELETERIOUS IMPACTS TO PUBLIC SAFETY, ECOSYSTEM HEALTH, AND PRIVATE PROPERTY. THE CAPE CANAVERAL MUNICIPAL GOVERNMENT ? IN COLLABORATION WITH ITS ACADEMIC AND COMMUNITY PARTNERS ? HAS IDENTIFIED GREEN STORMWATER INFRASTRUCTURE (GSI) BASED INTERVENTIONS AS ONE OF ITS PRIMARY STRATEGIES FOR CLIMATE AND WATER QUALITY MITIGATION. THIS SMART AND CONNECTED COMMUNITIES CIVIC INNOVATION CHALLENGE FULL AWARD (SCC-CIVIC-FA) PROJECT WILL ESTABLISH A PILOT GSI BIOSWALE AND ASSOCIATED HYDROLOGIC MONITORING SYSTEM AT VETERANS MEMORIAL PARK IN THE CITY OF CAPE CANAVERAL. PERFORMANCE DATA FROM THIS PILOT WILL DIRECTLY USED TO GUIDE DESIGN, IMPLEMENTATION, AND MONITORING PROTOCOLS WITHIN SUBSEQUENT GSI PROJECTS IN CAPE CANAVERAL AND OTHER SIMILARLY SITUATED COASTAL COMMUNITIES IN THE INDIAN RIVER LAGOON WATERSHED OF EASTERN FLORIDA. THIS INCLUDES CAPE CANAVERAL?S RECENT DEVELOPMENT OF AN AMBITIOUS 25-YEAR MASTER PLAN FOR GSI INTERVENTIONS WITHIN ITS PRESIDENTIAL STREETS NEIGHBORHOOD, A HOTSPOT FOR RECENT STORMWATER FLOODING EVENTS. GSI REFERS TO A SET OF STORMWATER MANAGEMENT PRACTICES THAT ARE DESIGNED TO MAINTAIN, RESTORE, AND/OR MIMIC PRE-DEVELOPMENT HYDROLOGIC CONDITIONS WITHIN BUILT ENVIRONMENTS. THESE PRACTICES ARE WIDELY PROMOTED AS A TOOL FOR REDUCING FLOOD RISK, IMPROVING WATER QUALITY, AND CREATING VALUABLE GREEN SPACE AMENITIES. HOWEVER, GSI HAS NOT YET BEEN ADOPTED AT SCALE IN MUCH OF THE U.S., INCLUDING AREAS ? LIKE MUCH OF COASTAL FLORIDA ? WHERE EXISTING STORMWATER INFRASTRUCTURE SYSTEMS ARE LARGELY MALADAPTED TO CLIMATE CHANGE-DRIVEN FLOOD RISKS AND OFTEN IDENTIFIED AS A PRIMARY POLLUTANT LOAD SOURCE. A RELATIVE LACK OF MONITORING AND PERFORMANCE DATA, AMBIGUOUS PERMITTING DESIGN STANDARDS, AND MISUNDERSTOOD MAINTENANCE PROTOCOLS ARE ALL COMMONLY CITED AS KEY BARRIERS TO THE WIDER APPLICATION OF GSI. TO HELP OVERCOME THESE BARRIERS, CAPE CANAVERAL IS MOTIVATED TO SERVE AS A PARTNER AND HOST FOR THIS PILOT PROJECT THAT WILL OBJECTIVELY TEST THE FIELD PERFORMANCE OF GSI INTERVENTIONS FOR MITIGATING STORMWATER-BASED FLOODING AND NON-POINT NUTRIENT LOAD ISSUES. PROJECT RESULTS ARE ANTICIPATED TO INFLUENCE BROADER MUNICIPAL CONSIDERATION AND ADOPTION OF SUITABLE GSI INTERVENTIONS WITHIN THE WATERSHED OF THE HIGHLY IMPAIRED INDIAN RIVER LAGOON, HOME TO 1.6 MILLION PEOPLE AND OFTEN DESCRIBED AS THE MOST BIODIVERSE ESTUARY IN NORTH AMERICA. UTILIZATION OF A NOVEL DISSIMILARITY APPROACH TO IDENTIFY A NOVEL MECHANISM OF MERKEL CELL POLYOMAVIRUS SMALL TUMOR ANTIGEN NUCLEAR IMPORT RESPONSIBLE FOR THE DEVELOPMENT OF MERKEL CELL CARCINOMA - PROJECT ABSTRACT MERKEL CELL CARCINOMA (MCC) IS A RARE BUT AGGRESSIVE SKIN CANCER WITH A MORTALITY RATE THREE TIMES HIGHER THAN MELANOMA. TREATMENT FOR MCC REMAINS A CHALLENGE, WITH ONLY APPROXIMATELY 50% OF INDIVIDUALS RESPONDING TO CURRENT THERAPIES. NOTABLY, 80% OF MCC CASES ARE CAUSED BY MERKEL CELL POLYOMAVIRUS (MCPYV); UNDERSTAND- ING THE MECHANISMS OF MCPYV TUMORIGENESIS IS THUS OF CRITICAL IMPORTANCE FOR THE DEVELOPMENT OF EFFECTIVE THERAPIES FOR MCC. IN MCPYV-POSITIVE MCC, THE SMALL (ST) AND LARGE (LT) TUMOR ANTIGENS ARE RESPONSIBLE FOR THE DEVELOPMENT OF MCC. MCPYV BELONGS TO A FAMILY OF 15 HUMAN POLYOMAVIRUSES (HPYV), ALL OF WHICH ENCODE THEIR OWN ST AND LT; HOWEVER, ONLY MCPYV IS ASSOCIATED WITH CANCER. SEEING THE LACK OF OTHER ONCOGENIC HPYVS FOR COMPARISON AS A LIMITATION, EARLIER RESEARCH HAS FOCUSED ON THE SIMILARITIES BETWEEN MCPYV AND TWO ONCOGENIC, NON-HUMAN POLYOMAVIRUSES (SIMIAN VIRUS 40 [SV40] AND MURINE POLYOMAVIRUS [MUPYV]). HOW- EVER, THIS APPROACH HAS BEEN LIMITED BY MCPYV, SV40, AND MUPYV EACH UTILIZING A DIFFERENT TUMOR ANTIGEN AS THEIR DOMINANT TRANSFORMING PROTEIN: ST IS THE DOMINANT TRANSFORMING PROTEIN OF MCPYV BUT IS ONLY AN ACCES- SORY PLAYER IN SV40- AND MUPYV- MEDIATED TRANSFORMATION. INSTEAD, THIS PROJECT USES AN INNOVATIVE “DISSIMI- LARITY APPROACH” TO EXPLOIT THE FACT THAT MCPYV IS UNIQUELY TUMORIGENIC AMONG HPYVS. BY IDENTIFYING THE DIF- FERENCES BETWEEN MCPYV AND TWO SKIN-TROPIC, NON-ONCOGENIC HPYVS, TRICHODYSPLASIA SPINULOSA POLYOMAVIRUS (TSPYV) AND HUMAN POLYOMAVIRUS 7 (HPYV7), ONE COULD IDENTIFY THE UNIQUE MECHANISMS RESPONSIBLE FOR MCPYV ONCOGENESIS. USING THIS APPROACH, IT WAS FOUND THAT MCPYV ST IS UNIQUELY CAPABLE OF NUCLEAR LOCALIZA- TION DESPITE THE ABSENCE OF A NUCLEAR LOCALIZATION SIGNAL (NLS) AND THAT NUCLEAR LOCALIZATION WAS NECESSARY FOR CELLULAR TRANSFORMATION. TO FURTHER INVESTIGATE MCPYV ST NUCLEAR LOCALIZATION AND ITS ROLE IN THE UNIQUE TRANS- FORMATIVE CAPABILITIES OF MCPYV ST, THE FOLLOWING AIMS ARE PROPOSED. THE FIRST AIM SEEKS TO IDENTIFY THE MCPYV ST NOVEL NLS (NVNLS) BY FOCUSING ON A REGION OF 30 AMINO ACIDS (MST30) RESPONSIBLE FOR NUCLEAR LOCALIZATION. THE SECOND AIM WILL DETERMINE WHETHER THE NVNLS IS RESPONSIBLE FOR THE UNIQUE TRANSFORMING ABILITIES OF MCPYV ST AMONG HPYVS AND WILL FACILITATE UNDERSTANDING OF THE HPYV DETERMINANTS IN INDUCING MALIGNANT OR NON-MALIGNANT DISEASE. THE THIRD AIM SEEKS TO FURTHER UNDERSTAND THE MECHANISM BY WHICH MCPYV ST CAN AC- COMPLISH NUCLEAR IMPORT BY INVESTIGATING INTERACTIONS WITH NUCLEAR TRANSPORT PROTEINS SUCH AS IMPORTIN AND NU- CLEAR PORE COMPLEX PROTEINS. TOGETHER, THE AIMS INVESTIGATED IN THIS PROPOSAL WILL FURTHER OUR UNDERSTANDING OF CELLULAR NUCLEAR IMPORT, DETERMINANTS OF HPYV ONCOGENESIS, AND THE MECHANISMS OF MCPYV ST-INDUCED MCC, WHICH MAY COLLECTIVELY PROVE INFLUENTIAL IN DEVELOPING NOVEL, EFFICACIOUS THERAPIES FOR MCC. ALL ASPECTS OF THESE INVESTIGATIONS WILL BE PERFORMED BY STETSON UNIVERSITY UNDERGRADUATE STUDENTS, THEREBY PROVIDING STUDENTS WITH INVALUABLE BIOMEDICAL RESEARCH EXPERIENCE, AND STRENGTHENING THE RESEARCH CULTURE OF STETSON UNIVERSITY.

BRC-BIO: INVESTIGATING THE MOLECULAR MECHANISMS OF FUNGAL CELL FUSION -THE PROCESS OF TWO CELLS COMBINING INTO ONE, KNOWN AS CELL FUSION, HAPPENS FREQUENTLY THROUGHOUT THE DEVELOPMENT OF MAMMALS, INCLUDING HUMANS. DURING REPRODUCTION, SPERM AND EGG CELLS MUST FUSE FOR FERTILIZATION TO OCCUR, AND LATER IN FETAL DEVELOPMENT MUSCLE CELLS FUSE INTO LONG SKELETAL MUSCLE FIBERS THAT ALLOW FOR MOVEMENT. THESE ARE JUST TWO EXAMPLES OF THE MANY FUSION EVENTS THAT OCCUR AND HIGHLIGHT THE IMPORTANCE OF THIS GENERAL PROCESS. CELL FUSION DOES NOT HAPPEN SPONTANEOUSLY AND IS HIGHLY REGULATED BY PROTEINS THAT ENSURE THE PROCESS OCCURS AT THE CORRECT TIME AND PLACE. IDENTIFYING THESE REGULATORS AND DETERMINING HOW THEY FUNCTION IS CRUCIAL FOR UNDERSTANDING CELL FUSION BUT HAS PROVEN DIFFICULT TO STUDY IN MAMMALS. THE YEAST, SACCHAROMYCES CEREVISIAE, UNDERGOES A FUSION EVENT SIMILAR TO FERTILIZATION DURING THEIR LIFE CYCLE MAKING THEM AN EXCELLENT MODEL SYSTEM TO STUDY FUSION. YEAST ARE EASY TO GROW AND MANIPULATE AND CONTAIN MANY OF THE SAME GENES AND PROTEINS AS HUMAN CELLS. THIS PROJECT AIMS TO STUDY THE MECHANISMS OF CELL FUSION IN S. CEREVISIAE BY IDENTIFYING NOVEL REGULATORS AND ANALYZING KNOWN PROTEINS REQUIRED FOR THE PROCESS. THIS WORK WILL BE PERFORMED BY UNDERGRADUATES THROUGH COURSE-BASED RESEARCH EXPERIENCES AND YEAR-LONG INDEPENDENT RESEARCH OPPORTUNITIES. STUDENTS WILL GAIN EXPERIENCE GENETICALLY MANIPULATING YEAST AND PERFORMING CUTTING EDGE MICROSCOPY TECHNIQUES. THIS PROJECT WILL ENCOURAGE THE NEXT GENERATION OF SCIENTISTS BY ALLOWING THEM ACCESS TO RESEARCH EXPERIENCES WHERE THEY WILL IDENTIFY MECHANISMS OF YEAST FUSION THAT CAN AID IN THE UNDERSTANDING OF CELL FUSION IN MAMMALS. YEAST CELLS HAVE A CELL WALL SURROUNDING THEIR PLASMA MEMBRANE, MEANING THAT ONCE TWO CELLS COME INTO CONTACT, THEY MUST FIRST DEGRADE THEIR CELL WALLS SPECIFICALLY WHERE THEY WILL FUSE, SIMILAR TO THE DEGRADATION OF EXTRACELLULAR MATRIX REQUIRED IN MAMMALIAN SYSTEMS. ONCE THIS OCCURS, PROTEINS THAT PROMOTE PLASMA MEMBRANE FUSION CAN ACT, LEADING TO CYTOPLASMIC MIXING. WHILE SOME REGULATORS AND MORPHOLOGICAL EVENTS OF YEAST FUSION HAVE BEEN IDENTIFIED, THERE ARE STILL UNANSWERED QUESTIONS ABOUT THE PROCESS. OVERALL, THIS PROPOSAL AIMS TO UNCOVER HOW CELL WALL DEGRADATION AND PLASMA MEMBRANE FUSION ARE REGULATED THROUGH MUTATIONAL ANALYSIS OF AN UNDERSTUDIED, KEY REGULATOR OF CELL WALL DEGRADATION, VISUALIZATION OF ENZYME SECRETION DURING FUSION, AND IDENTIFICATION OF NOVEL PLASMA MEMBRANE FUSION REGULATORS. FLUOROGEN-ACTIVATING TAGS WILL BE USED TO DEVELOP A MICROSCOPIC ASSAY FOR FUSION-SPECIFIC SECRETION, ADDRESSING A LONG-STANDING HYPOTHESIS IN THE FIELD. ADDITIONALLY, SUPPRESSOR ANALYSIS OF A MEMBRANE FUSION MUTATION RECENTLY IDENTIFIED BY A STUDENT RESEARCHER WILL ALLOW FOR IDENTIFICATION OF NOVEL PLASMA MEMBRANE FUSION REGULATORS, WHICH HAVE LONG-ELUDED SCIENTISTS. CIVIC-PG TRACK A: ASSESSING THE PERFORMANCE OF GREEN STORMWATER INFRASTRUCTURE FOR CLIMATE ADAPTATION AND COASTAL RESILIENCE IN THE CITY OF CAPE CANAVERAL, FLORIDA -THIS PROJECT PLANS TO ESTABLISH AN URBAN HYDROLOGY MONITORING NETWORK WITHIN THE CITY OF CAPE CANAVERAL, FLORIDA, THROUGH A PARTICIPATORY CHARRETTE PROCESS. LIKE MANY COASTAL COMMUNITIES, CAPE CANAVERAL IS CONFRONTED WITH THE STRESSORS OF GROWING FLOOD RISK AND HARMFUL ALGAL BLOOMS IN NEARSHORE WATERS. A COMBINATION OF ANTIQUATED INFRASTRUCTURE IN THE BUILT ENVIRONMENT AND ACCELERATING CLIMATE CHANGE IS AMPLIFYING THE IMPACTS OF BOTH URBAN FLOODING AND ALGAL BLOOMS, RESULTING IN INCREASING THREATS TO PUBLIC SAFETY, ECOSYSTEM HEALTH, AND PRIVATE PROPERTY. THE CAPE CANAVERAL MUNICIPAL GOVERNMENT, IN COLLABORATION WITH ITS ACADEMIC AND COMMUNITY PARTNERS, IS PLANNING TO DESIGN, IMPLEMENT, MONITOR, AND ASSESS NATURE-BASED GREEN STORMWATER INFRASTRUCTURE (GSI) AS AN APPROACH FOR MITIGATING THESE CURRENT AND FUTURE STRESSORS. THIS GSI MONITORING NETWORK WILL PROVIDE IMPORTANT BENEFITS FOR CAPE CANAVERAL AND THE BROADER INDIAN RIVER LAGOON WATERSHED OF EASTERN FLORIDA, WHILE ALSO DEMONSTRATING OPPORTUNITIES FOR OPTIMIZING AND ASSESSING GSI PERFORMANCE IN SIMILARLY VULNERABLE COASTAL LOCATIONS. GSI REFERS TO A SET OF STORMWATER MANAGEMENT PRACTICES THAT ARE DESIGNED TO MAINTAIN, RESTORE, AND/OR MIMIC PRE-DEVELOPMENT HYDROLOGIC CONDITIONS WITHIN BUILT ENVIRONMENTS. THESE PRACTICES ARE BEING INCREASINGLY PROMOTED AS A TOOL FOR REDUCING FLOOD RISK, IMPROVING WATER QUALITY, ENHANCING URBAN HABITAT, AND CREATING VALUABLE GREEN SPACE AMENITIES. HOWEVER, GSI HAS NOT YET BEEN ADOPTED AT SCALE IN MUCH OF THE U.S., INCLUDING AREAS ? LIKE MUCH OF COASTAL FLORIDA ? WHERE EXISTING STORMWATER INFRASTRUCTURE SYSTEMS ARE LARGELY MALADAPTED TO CLIMATE CHANGE-DRIVEN FLOOD RISKS AND OFTEN IDENTIFIED AS A PRIMARY POLLUTANT LOAD SOURCE INTO IMPAIRED AQUATIC ECOSYSTEMS. A RELATIVE LACK OF MONITORING AND PERFORMANCE DATA, AMBIGUOUS PERMITTING DESIGN STANDARDS, AND MISUNDERSTOOD MAINTENANCE PROTOCOLS ARE ALL COMMONLY CITED AS KEY BARRIERS TO THE WIDER APPLICATION OF GSI, IN FLORIDA AND ELSEWHERE. TO HELP OVERCOME THESE BARRIERS, CAPE CANAVERAL IS MOTIVATED TO SERVE AS A PARTNER AND HOST FOR AN APPLIED RESEARCH PROGRAM THAT COMPREHENSIVELY TESTS THE FIELD PERFORMANCE OF GSI INTERVENTIONS FOR MITIGATING LOCALLY SEVERE STORMWATER-BASED FLOODING AND NON-POINT NUTRIENT LOAD ISSUES. KEY RESEARCH GOALS FOR THE FULLY IMPLEMENTED PROJECT INCLUDE CHARACTERIZATION OF STORMWATER INFILTRATION RATE-CAPACITY, AREAL NUTRIENT-LOAD REDUCTION PERFORMANCE, AND PUBLIC VALUATION OF GSI SYSTEMS AS A MUNICIPAL AMENITY. RESULTS ARE ANTICIPATED TO FACILITATE GREATER GSI RESEARCH AND ADOPTION IN THE IMPERILED INDIAN RIVER LAGOON WATERSHED, HOME TO 1.5 MILLION PEOPLE AND OFTEN DESCRIBED AS THE MOST BIODIVERSE ESTUARY IN NORTH AMERICA, AND BROADLY IMPACT THE EVOLUTION OF GSI MONITORING PRACTICES IN URBANIZED COASTAL COMMUNITIES ACROSS THE WORLD. THIS PROJECT IS IN RESPONSE TO THE CIVIC INNOVATION CHALLENGE PROGRAM?TRACK A.
