| National Science Foundation | COLLABORATIVE RESEARCH: UNPACKING COMPUTATIONAL THINKING FOR ELEMENTARY TEACHERS AND LEARNERS -THE GOAL OF THIS PROJECT IS TO INVESTIGATE THE INTEGRATION OF COMPUTATIONAL THINKING (CT) INTO ELEMENTARY SCHOOL CURRICULA BY STUDYING HOW TEACHERS DEVELOP EXPERTISE IN INTEGRATING CT ACTIVITIES THAT ALIGN WITH INTERDISCIPLINARY STANDARDS AND EXISTING CURRICULA. THIS PROJECT ADDRESSES THE CRITICAL NEED FOR SCIENCE, TECHNOLOGY, ENGINEERING, AND MATHEMATICS (STEM) LITERACY BY INVESTIGATING EFFECTIVE MODELS OF PROFESSIONAL DEVELOPMENT (PD) THAT IMPROVE STEM TEACHING AND LEARNING. LEVERAGING AN ASSET-BASED APPROACH, THE PROJECT WILL PROVIDE OPPORTUNITIES TO BROADEN PARTICIPATION IN COMPUTER SCIENCE EDUCATION THROUGH BUILDING A COMMUNITY OF PRACTICE FOR TEACHERS AND DESIGNING CT-INFUSED CURRICULA. TO ENSURE THAT ALL TEACHERS HAVE THE NECESSARY SUPPORT TO PARTICIPATE IN NEW PEDAGOGICAL PRACTICES OF INCREASING COMPLEXITY, THE PROJECT WILL UTILIZE A COLLABORATIVE TEACHER PD AND MENTORSHIP STRUCTURE IN WHICH TEACHERS WILL WORK CLOSELY WITH PEERS AND PROJECT TEAM MEMBERS TO PREPARE, TEACH, AND REFLECT UPON LESSONS THROUGH MULTI-YEAR PARTICIPATION. THE RESEARCH WILL DOCUMENT HOW TEACHERS COME TO UNDERSTAND CT AND TAKE UP THESE PRACTICES IN THEIR CLASSROOMS. THE DISCOVERY RESEARCH PREK-12 PROGRAM (DRK-12) SEEKS TO SIGNIFICANTLY ENHANCE THE LEARNING AND TEACHING OF STEM BY PREK-12 STUDENTS AND TEACHERS, THROUGH RESEARCH AND DEVELOPMENT OF INNOVATIVE RESOURCES, MODELS AND TOOLS. PROJECTS IN THE DRK-12 PROGRAM BUILD ON FUNDAMENTAL RESEARCH IN STEM EDUCATION AND PRIOR RESEARCH AND DEVELOPMENT EFFORTS THAT PROVIDE THEORETICAL AND EMPIRICAL JUSTIFICATION FOR PROPOSED PROJECTS. OVER FOUR YEARS, THIS PROJECT WILL DIRECTLY IMPACT 135 TEACHERS IN MULTI-YEAR PARTICIPATION IN PROFESSIONAL DEVELOPMENT AND APPROXIMATELY 12,000 STUDENTS INDIRECTLY IN THEIR CLASSROOMS. THIS PROJECT WILL INITIALLY FOCUS ON SCHOOLS IN SOUTH CAROLINA, ESPECIALLY IN RURAL DISTRICTS. THE PROJECT WILL BE GUIDED BY THREE PRIMARY RESEARCH QUESTIONS: 1) HOW DO PREK-5 TEACHERS INTEGRATE CT INTO THEIR DISCIPLINARY TEACHING?; 2) WHAT ARE THE PERSONAL LEARNING TRAJECTORIES THAT TEACHERS MOVE THROUGH AS THEY WORK TOWARDS CT INTEGRATION?; 3) WHAT ARE THE BARRIERS AND ACCELERATORS FOR TEACHERS? IMPLEMENTATION OF CT-INFUSED LESSONS? THIS PROJECT IS GROUNDED IN THE IDEA THAT INCREMENTAL, SUSTAINABLE, AND SUBSTANTIAL TEACHER LEARNING COMES WHEN TEACHERS ARE FULLY INVESTED IN THE CYCLE OF LEARNING AND HAVE OPPORTUNITIES TO BUILD AND STRENGTHEN THEIR COMMUNITIES OF PRACTICE. IN REFLECTIVE PD SESSIONS HELD THROUGHOUT THE ACADEMIC YEAR AND SUMMER, TEACHERS WILL EXPLORE, CO-DESIGN, IMPLEMENT, AND REFINE STUDENT LEARNING ACTIVITIES THAT INTEGRATE CT INTO EXISTING CONTENT AREA STANDARDS AND CURRICULA. THE CT ACTIVITIES, WHICH WILL INCLUDE STANDARDS-BASED LESSON PLANS, ASSESSMENTS, AND SUPPLEMENTAL RESOURCES, WILL OFFER BOTH PLUGGED AND UNPLUGGED COMPONENTS, SO THAT STUDENTS AND TEACHERS HAVE OPPORTUNITIES TO UNDERSTAND THE CONCEPTUAL UNDERPINNINGS OF COMPUTATIONAL THINKING WITH AND WITHOUT COMPUTERS. A MINIMUM OF 120 LESSONS AND RELATED PEDAGOGICAL MATERIALS MAPPED TO CORE-CONTENT STANDARDS WILL BE DEVELOPED AND MADE FREELY AVAILABLE TO THE PUBLIC ON THE PROJECT WEBSITE. USING A MIXED METHODS APPROACH, THIS STUDY WILL DOCUMENT THE TRAJECTORIES THROUGH WHICH ELEMENTARY TEACHERS MOVE AS THEY BEGIN TO INTEGRATE CT INTO DISCIPLINARY TEACHING, AS WELL AS BARRIERS AND ACCELERATORS FACED AS THEY INTEGRATE CT INTO DISCIPLINARY TEACHING. QUANTITATIVE SURVEY METHODOLOGIES AND MULTIMODAL ANALYSIS OF NARRATIVE INTERVIEWS, CLASSROOM VIDEO, TEACHER REFLECTIONS, AND PEDAGOGICAL ARTIFACTS WILL BE USED TO ANALYZE TEACHERS? SENSE MAKING ABOUT CT OVER TIME AND THE UPTAKE OF NEW PEDAGOGICAL APPROACHES IN THEIR TEACHING. ULTIMATELY, THIS STUDY WILL PRODUCE A SET OF EMPIRICALLY-TESTED LEARNING TRAJECTORIES THAT CAN BE USED TO SUPPORT TEACHERS IN INTEGRATING CT INTO THEIR CLASSROOMS. | National Science Foundation | COLLABORATIVE RESEARCH: ENGAGING K-8 TEACHERS IN INTEGRATING COMPUTER SCIENCE, AI, AND CYBERSECURITY INTO STANDARD-BASED TEACHING -AS ARTIFICIAL INTELLIGENCE (AI) BECOMES INCREASINGLY EMBEDDED IN THE TECHNOLOGIES USED BY BOTH STUDENTS AND TEACHERS, IT IS ESSENTIAL FOR THEM TO UNDERSTAND HOW TO BE SAFE WHILE USING AI. FURTHERMORE, AI AND CYBERSECURITY TECHNOLOGY TOGETHER ARE BETTER AT DETECTING MALICIOUS ACTIVITIES THAN CONVENTIONAL SECURITY SYSTEMS. THE NEED TO BLEND THE TWO DISCIPLINES INTO A SINGLE, INTEGRATED CURRICULUM FOR K-8 EDUCATION IS HIGHLIGHTED BY THE INTERCONNECTEDNESS OF AI AND CYBERSECURITY AS COMPLEMENTARY SYSTEMS. THIS PROJECT WILL PLANT THE SEEDS OF THESE LITERACIES BY SPIRALING CONTENT ON TOPICS FROM COMPUTER PROGRAMMING, INTERNET FUNDAMENTALS, AND INTRODUCTION TO DATA AND AI ALONG WITH CYBERSECURITY TOPICS IN SMALL DOSES THROUGHOUT STUDENTS' K-8 EDUCATION. THIS PROJECT WILL LAY THE FOUNDATION FOR THE STUDENTS TO EVENTUALLY DEVELOP A COMPREHENSIVE UNDERSTANDING OF HOW DIFFERENT TECHNOLOGIES WORK AND INTERACT. THIS PROJECT AIMS TO INVESTIGATE HOW TEACHERS FROM RURAL SCHOOLS APPROACH THE INTEGRATION OF COMPUTER SCIENCE (CS), AND IN PARTICULAR, AI AND CYBERSECURITY COMPONENTS, INTO THEIR EXISTING CURRICULA. THIS PROJECT WILL DIRECTLY IMPACT AT LEAST 82 RURAL TEACHERS FROM SOUTH CAROLINA THROUGH MULTI-YEAR PARTICIPATION IN PROFESSIONAL DEVELOPMENT AND COACHING AND APPROXIMATELY 8,100 STUDENTS INDIRECTLY IN THEIR CLASSROOMS. RESOURCES, INCLUDING LESSON PLANS, DEVELOPED THROUGH THE PROJECT WILL BE MADE AVAILABLE AS OPEN ACCESS ON THE PROJECT WEBSITE, THROUGH PARTNERSHIPS, AND VIA CONFERENCE PRESENTATIONS AND PUBLICATIONS. THE OVERARCHING GOAL OF THIS PROJECT IS TO IMPROVE STEM TEACHING AND LEARNING BY EXAMINING SUCCESSFUL TEACHER PROFESSIONAL DEVELOPMENT (PD) MODELS IN RESPONSE TO A NEED FOR PREPARING STUDENTS WITH BOTH STEM LITERACY SKILLS AND OTHER TECHNICAL COMPETENCIES NEEDED TO BE INFORMED CITIZENS AND CONTRIBUTORS TO SOCIETY. THE PROJECT WILL BLEND AI AND CYBERSECURITY INTO A UNIFIED CURRICULA, SUPPORTED BY THE FOLLOWING OBJECTIVES: (1) ANALYZE HOW K-8 EDUCATORS INCORPORATE AI AND CYBERSECURITY TOPICS INTO DISCIPLINARY TEACHING COMPONENTS; (2) EXPLORE THE LEARNING SUPPORTS THAT K-8 EDUCATORS NEED TO EFFECTIVELY INCORPORATE AI AND CYBERSECURITY TOPICS INTO CURRENT CURRICULA TO ASSIST STUDENTS IN MEETING CONTENT-FOCUSED LEARNING OBJECTIVES; AND (3) COMPARE THE INTEGRATION OPPORTUNITIES AVAILABLE TO K-8 EDUCATORS IN RURAL AREAS OF SOUTH CAROLINA. IN THE END, HYPOTHESIZING THAT THIS RESEARCH CAN FORM THE BASIS FOR THE CREATION OF MORE SCALABLE PD PROGRAMS FOR K-8 TEACHERS AS WELL AS CONTINUOUS ASSISTANCE WITH CS INTEGRATION. THE RESEARCH WILL FOLLOW A MIXED-METHODS DESIGN, COMBINING SURVEY DATA, CLASSROOM OBSERVATIONS, AND INTERVIEWS TO EXAMINE HOW EDUCATORS MAKE SENSE OF AND APPLY THESE CONCEPTS OVER TIME. STATISTICAL ANALYSES WILL IDENTIFY SIGNIFICANT PATTERNS ACROSS TEACHER IMPLEMENTATION CONTEXTS. QUALITATIVE DATA--DRAWN FROM SEMI-STRUCTURED INTERVIEWS, TEACHING ARTIFACTS, VIDEO DOCUMENTATION, AND FIELD NOTES--WILL BE CODED TO SURFACE PEDAGOGICAL STRATEGIES, CHALLENGES, AND EVOLVING TEACHER UNDERSTANDINGS. IN ADDITION, AN APP WILL BE CREATED TO ALLOW TEACHERS TO DOCUMENT REAL-TIME CLASSROOM INTEGRATION OF AI AND CYBERSECURITY, PROVIDING A ROBUST, LONGITUDINAL DATASET. THE FINDINGS WILL INFORM SCALABLE, CONTEXT-SENSITIVE PROFESSIONAL DEVELOPMENT MODELS AND CONTRIBUTE TO A BROADER UNDERSTANDING OF HOW TO SUPPORT INTERDISCIPLINARY CS LEARNING IN K-8 SETTINGS. ALL DATA COLLECTION WILL BE DESIGNED TO ENSURE METHODOLOGICAL RIGOR AND TRUSTWORTHINESS THROUGH TRIANGULATION AND ONGOING MEMBER CHECKS WITH PARTICIPATING EDUCATORS. | Department of Health and Human Services | A NOVEL SMALL MOLECULE FOR THE PREVENTION AND TREATMENT OF DIABETIC RETINOPATHY - SUMMARY DIABETIC RETINOPATHY (DR) IS A LEADING CAUSE OF BLINDNESS IN THE UNITED STATES. CURRENTLY, MOST TREATMENTS FOR DR ARE ONLY APPLICABLE AT THE LATE STAGE AND ARE INVASIVE. ANTI-VEGF (VASCULAR ENDOTHELIAL GROWTH FACTOR) AGENTS HAVE ACHIEVED IMPRESSIVE THERAPEUTIC EFFECTS AGAINST DR AND NEOVASCULARIZATION (NV). HOWEVER, 40% OF PATIENTS WITH DR HAVE INADEQUATE RESPONSES TO THE TREATMENT. GIVEN THAT DR IS A MULTI-FACTORIAL DISEASE, THERAPIES THAT TARGET ONE SINGLE PATHOGENIC FACTOR (SUCH AS VEGF) ARE CLEARLY INSUFFICIENT TO ACHIEVE DESIRED EFFICACY IN ALL DR PATIENTS. THEREFORE, THE DEVELOPMENT OF NOVEL, NON-INVASIVE, LONG-TERM AND MORE EFFECTIVE PHARMACOLOGICAL TREATMENTS FOR DR IS URGENTLY REQUIRED, PARTICULARLY AT AN EARLY STAGE OF THE DISEASE. THE OVERALL OBJECTIVE OF THIS PHASE II PROJECT IS TO DEVELOP A NOVEL DRUG FOR THE ORAL PHARMACOTHERAPY OF DR. PEROXISOME PROLIFERATOR-ACTIVATED RECEPTOR A (PPARA) IS A LIGAND-ACTIVATED NUCLEAR RECEPTOR THAT REGULATES MITOCHONDRIAL LIPID SS OXIDATION AND PEROXISOME SCAVENGE HYDROGEN PEROXIDE IN THE CYTOPLASM. TWO LARGE AND LONGITUDINAL CLINICAL STUDIES REPORTED INDEPENDENTLY THAT ORAL ADMINISTRATION OF PPARA AGONIST FENOFIBRATE HAS ROBUST THERAPEUTIC EFFECT ON DR IN TYPE 2 DIABETIC PATIENTS. THIS IS THE FIRST ORAL DRUG WITH PROVEN EFFECT IN DR PATIENTS. IN PRECLINICAL STUDIES WITH ANIMAL MODELS OF DR, PHARMACOLOGICAL ACTIVATION OF PPARA HAS BEEN SHOWN TO DECREASE RETINAL PATHOLOGIES TYPICALLY OBSERVED IN DR (E.G. PATHOGENIC NEOVASCULARIZATION, VASCULAR LEAKAGE AND INFLAMMATION IN THE RETINA), RESULTING IN A PROFOUND THERAPEUTIC EFFECT. THUS, PPARA REPRESENTS A NOVEL THERAPEUTIC TARGET FOR THE TREATMENT OF DR. HOWEVER, THERE ARE CURRENTLY NO FDA-APPROVED PPARA DRUGS FOR DR. OUR MEDICINAL CHEMISTRY TEAM HAS SYNTHESIZED AND IDENTIFIED A NON-FIBRATE, QUINOLINE-DERIVED SMALL MOLECULE A190, A NOVEL PPARA AGONIST WITH MORE POTENT ACTIVITY. IN SBIR PHASE I STUDIES, WE HAVE COMPLETED PROOF-OF-PRINCIPLE STUDIES DEMONSTRATING THAT SYSTEMIC ADMINISTRATION OF A190 IS MORE EFFECTIVE THAN FENOFIBRATE ON TWO DIABETIC ANIMAL MODELS. THESE STUDIES ESTABLISHED A190 AS A PROMISING DRUG CANDIDATE FOR FURTHER DEVELOPMENT. THE PROPOSED RESEARCH IN PHASE II WILL FOCUS ON EVALUATING IN VIVO SAFETY AND TOXICOLOGY OF A190 IN TWO SPECIES (RATS AND RABBITS). IN ADDITION, THE LONG- TERM EFFICACY OF ORALLY ADMINISTERED A190 WILL BE TESTED IN DR MODELS. THIS PHASE II PROGRAM INCLUDES THREE SPECIFIC AIMS. CCBHC WILL CHANGE THAT, OFFERING A RANGE OF COORDINATED SERVICES SUCH AS PRIMARY CARE SCREENING, CRISIS MENTAL HEALTH SERVICES AND CASE MANAGEMENT IN ADDITION TO MENTAL AND CHEMICAL HEALTH CARE. IN SHORT, CCBHC WILL SERVE AS A "ONE-STOP-SHOP" FOR BOTH ADULTS AND CHILDREN WHO HAVE TROUBLE OTHERWISE GETTING THE HELP THEY NEED. CCBHC WILL IMPROVE ACCESS, QUALITY, COORDINATION AND CONTINUUM OF CARE TO A MINIMUM OF 500 CLIENTS ANNUALLY, THROUGH THE USE OF EVIDENCE-BASED STANDARDIZED ASSESSMENT TOOLS AND PRACTICES; IMPLEMENTATION OF PERSON CENTERED AND/OR FAMILY CENTERED INFORMED CARE; INCREASE OF CARE COORDINATION AND RECOVERY SUPPORT SERVICES; AND INCLUSION OF YOUTH AND FAMILY/CAREGIVER PARTICIPATION AT ALL LEVELS. PEOPLE WHO UTILIZE CCBHC MAY RECEIVE COMPREHENSIVE OUTPATIENT MENTAL HEALTH AND SUBSTANCE USE SERVICES, INCLUDING PROVISION OF PSYCHOTROPIC/ANTI-PSYCHOTIC MEDICATIONS; PSYCHO-THERAPEUTIC INTERVENTIONS; PRIMARY CARE SCREENING AND MONITORING (INCLUDING HIV AND VIRAL HEPATITIS SCREENING); BEHAVIORAL HEALTH SCREENING, ASSESSMENT AND DIAGNOSIS, INCLUDING RISK MANAGEMENT; PSYCHIATRIC REHABILITATION SERVICES; INCLUDING 24-HOUR MOBILE CRISIS TEAMS, EMERGENCY CRISIS INTERVENTION AND CRISIS STABILIZATION; PATIENT-CENTERED TREATMENT/CRISIS PLANNING; TARGETED CASE MANAGEMENT; PEER AND FAMILY SUPPORT; INTENSIVE COMMUNITY BASED TREATMENT SERVICES FOR VETERANS AND MEMBERS OF THE ARMED FORCES; ASSERTIVE COMMUNITY TREATMENT; SOCIAL SUPPORT SERVICES AND/OR CONNECTIONS WITH OTHER SERVICE PROVIDERS. THE OVERALL OBJECTIVE OF THE PROJECT IS TO REDUCE DISPARITIES IN MENTAL HEALTH AND IMPROVE HEALTH OUTCOMES. ADDITIONALLY, THIS PROJECT AIMS TO SUPPORT 15 FELLOWS BY PROVIDING GRADUATE COURSEWORK CULMINATING IN A SOUTH CAROLINA ADD-ON CERTIFICATION ENDORSEMENT IN COMPUTER SCIENCE. THE PROJECT INTENDS TO ALLOW TEACHERS TO COLLABORATE WITH OTHER TEACHERS AND PROJECT STAFF TO CREATE A STATEWIDE RURAL COMPUTER SCIENCE TEACHER NETWORK, A TEACHER-DRIVEN PROFESSIONAL COMMUNITY FOCUSED ON ADDRESSING THE UNIQUE CHALLENGES AND OPPORTUNITIES FACED BY RURAL COMPUTER SCIENCE TEACHERS. THE PROPOSED PROJECT COMPONENTS ARE POSITIONED TO ENABLE HIGH-ACHIEVING PRACTICING TEACHERS TO BECOME COMPUTER SCIENCE TEACHER-LEADERS IN THEIR RURAL COMMUNITIES. THIS PROJECT AT THE CITADEL IS A COLLABORATION BETWEEN THE CITADEL?S ZUCKER FAMILY SCHOOL OF EDUCATION AND SWAIN FAMILY SCHOOL OF SCIENCE AND MATHEMATICS, AS WELL AS ORANGEBURG COUNTY SCHOOL DISTRICT AND THE COMPUTER SCIENCE TEACHERS ASSOCIATION. THIS PROJECT IS DESIGNED TO ENHANCE CS EDUCATION IN SOUTH CAROLINA'S RURAL, HIGH-NEEDS SCHOOLS BY DEVELOPING 15 RURAL TEACHER LEADERS WITH ADVANCED PEDAGOGICAL AND LEADERSHIP SKILLS. PROFESSIONAL DEVELOPMENT (PD) CONTENT PLANS TO BE TAILORED TO THE SPECIFIC NEEDS OF THESE TEACHERS AND THEIR STUDENTS, PROPOSING TO IMPROVE CRITICAL THINKING, PROBLEM-SOLVING SKILLS, AND STUDENT MOTIVATION IN COMPUTER SCIENCE. THE PROJECT AIMS TO CREATE VIRTUAL PD, CURRICULUM RESOURCES, AND A STATEWIDE CS4SC TEACHER HUB TO SUPPORT SUSTAINABLE AND SCALABLE INTEGRATION OF THESE EFFORTS. EVEN AFTER PATIENTS ARE INITIALLY RESUSCITATED, MANY DIE WITHIN A WEEK FROM SEVERE BRAIN INJURY. THE GOAL OF THIS APPLICATION IS TO IMPROVE NEUROLOGICALLY-INTACT SURVIVAL RATES AFTER PRE-HOSPITAL AND IN-HOSPITAL SCA THROUGH THE COMMERCIALIZATION OF A NOVEL METHOD OF CPR CALLED HEAD UP POSITION (HUP) CPR. HUP CPR WORKS BY HARNESSING GRAVITY TO ENHANCE VENOUS BLOOD FLOW FROM THE BRAIN TO THE HEART, LOWER INTRACRANIAL PRESSURE, AND ENHANCE CARDIAC OUTPUT. THE FIRST CLINICAL HUP CPR DEVICE, THE ELEGARDTM, WAS DESIGNED, BUILT, AND TESTED WITH PHASE I FUNDING SUPPORT. THE ELEGARD HAS BEEN USED TO HELP TREAT >4500 SCA PATIENTS TO DATE. BASED UPON EARLY POSITIVE CLINICAL OUTCOMES, PHASE II FUNDING SUPPORTED THE DEVELOPMENT OF A REFINED AND MORE PORTABLE DEVICE TO ACCELERATE ADOPTION (ELEGARD 3.0). A SUBSEQUENT LIMITED LAUNCH EVALUATION IN 2024 WAS HIGHLY FAVORABLE. HOWEVER, IT REVEALED A COMPATIBILITY ISSUE WHERE SOME SHORTER PATIENTS WERE SLIPPING OUT OF POSITION DURING ELEVATION, LIKELY DUE TO EXPERIENCING GREATER HEAD AND THORAX ELEVATION COMPARED TO TALLER PATIENTS. AS A RESULT, POST-AWARD EFFORTS HAVE FOCUSED ON REFINING THE HEAD CRADLING MECHANISM TO ENSURE CONSISTENT HEAD ELEVATION AND MINIMIZE SLIPPAGE FOR PATIENTS OF ALL HEIGHTS. ADDITIONALLY, DURING THIS PERIOD, THE DEVICE’S CONTRACT MANUFACTURER SUDDENLY CLOSED, REQUIRING THE TRANSFER OF ELEGARD 3.0 ASSEMBLY AND PROCESSES TO A NEW PRODUCTION FACILITY. WHICH CLING TO SOFT EPIDERMIS BY MEANS OF SKIN-PINCHING MECHANICS; AND ?GOUGERS? WHICH CAN PENETRATE TISSUE, SCUTES, SCALES, AND SOMETIMES TO THE UNDERLYING BONE. THIS RESEARCH WILL MAP GENETIC TAGS FROM BARNACLES COLLECTED WORLDWIDE TO 1) STUDY HOST CHOICE AND THE EVOLUTIONARY HISTORY OF BARNACLE ATTACHMENT, 2) DESCRIBE TO WHAT EXTENT BARNACLES MAY BE A ?FINGERPRINT? MANAGEMENT TOOL FOR TRACKING SEA TURTLE MOVEMENTS, AND 3) ESTABLISH A BASELINE DISTRIBUTION OF BARNACLE GENETIC TYPES FOR MONITORING ALTERATIONS IN SPECIES RANGES DUE TO CLIMATE CHANGE. THIS RESEARCH WILL TRAIN UNDERGRADUATE AND GRADUATE STUDENTS IN GENETICS AND EVOLUTIONARY ANALYSES AS WELL AS FIELD METHODS IN MARINE BIOLOGY VIA A FIELD COURSE IN COSTA RICA. OUTREACH INCLUDES THE DEVELOPMENT OF MUSEUM EXHIBITS AND THE PUBLICATION OF POPULAR ARTICLES FOR A BROAD AUDIENCE. THE EVOLUTIONARY HISTORY AND SPECIES DELINEATIONS OF THE CORONULOID BARNACLES (ARTHROPODA: CRUSTACEA: CIRRIPEDIA) IS UNCERTAIN. SPECIFICALLY, LITTLE IS KNOWN OF THEIR ROUTE TO EPIZOISM, THEIR DEGREE OF DEPENDENCE ON HOSTS FOR DISPERSAL AND DIVERSIFICATION, THEIR PATHWAYS TO VARIABLE ATTACHMENT MECHANISMS, AND THEIR CONTEMPORARY PHYLOGEOGRAPHIC DISTRIBUTIONS. THE RESEARCHERS AIM TO ADDRESS THESE DEFICIENCIES THROUGH PHYLOGENETIC AND POPULATION GENOMIC ANALYSIS WITH THREE PRINCIPLE OBJECTIVES: 1) CONSTRUCT A DEFINITIVE PHYLOGENY FOR THE WHALE AND TURTLE BARNACLES AND MAP TO IT THE EVOLUTIONARY CHRONOLOGY OF THEIR GLUING, GRIPPING, AND GOUGING ATTACHMENT MODES; 2) COMPARE GLOBAL PHYLOGEOGRAPHIC PATTERNS USING GENOME-WIDE MARKERS FOR THE THREE MOST-COMMON BARNACLE SPECIES FROM THE FOUR MOST-COMMON SEA TURTLE SPECIES, ASSESSING POPULATION CONNECTIVITY IN THE CONTEXT OF OCEANOGRAPHIC LARVAL DISPERSAL (GEOGRAPHIC LOCALITIES) VS. HOST TRANSPORT (HOST LINEAGES); AND 3) TEST WHETHER BARNACLE GENETIC LINEAGES WITHIN SPECIES ASSORT WITH RESPECT TO SEA TURTLE SPECIES. OF THOSE ONLY ABOUT 50,000 WILL HAVE A FULL RECOVERY. MORE THAN 750,000 PATIENTS WILL HAVE NO HOPE FOR MEANINGFUL SURVIVAL. THAT IS BECAUSE THERE IS NO CURRENTLY DEMONSTRABLE EFFECTIVE TREATMENT FOR REFRACTORY (R)-CA PATIENTS, THE ENORMOUS SUBGROUP OF PATIENTS WHO INVARIABLY DIE ANNUALLY FROM THIS LEADING CAUSE OF DEATH. DEPENDING UPON THE PRESENTING RHYTHM, CA BECOMES REFRACTORY TO CURRENT INTERVENTIONS, HEREIN DEFINED AS A <2% SURVIVAL, <10 MINUTES AFTER STARTING CONVENTIONAL (C) CPR. THIS APPLICATION IS FOCUSED ON A BETTER TREATMENT FOR PATIENTS WITH CA REFRACTORY TO CURRENT MANAGEMENT STRATEGIES, REGARDLESS OF THE INITIAL RHYTHM PRESENTATION. PATIENTS IN R-CA CANNOT BE RESUSCITATED WITH C-CPR, DEFIBRILLATION FOR A SHOCKABLE RHYTHM, AND VASOPRESSOR THERAPY, SUCH AS EPINEPHRINE, FLUIDS, AND ANTI-ARRHYTHMIC DRUGS. FOR OVER A HALF CENTURY WE HAVE FAILED TO EFFECTIVELY TREAT R-CA: WE HAVE NOT KNOWN HOW. OCCASIONALLY TRANSCUTANEOUS CARDIAC PACING, ECMO AND/OR DOUBLE SEQUENTIAL DEFIBRILLATION CAN BE USED WITH SOME BENEFIT FOR THE VERY SMALL PERCENTAGE OF PATIENTS THAT REMAIN IN REFRACTORY VENTRICULAR FIBRILLATION (R-VF). HOWEVER, IN GENERAL THESE TREATMENT STRATEGIES RARELY WORK FOR R-CA, ESPECIALLY IN PATIENTS WITH A NON-SHOCKABLE PRESENTING RHYTHM THAT COMPRISE 75-85% OF ALL CA CASES. HOWEVER, THE APPLICANTS RECENTLY OBSERVED THAT THE ADDITION OF RESUSCITATIVE ENDOVASCULAR BALLOON OCCLUSION OF THE AORTA (REBOA) TO AUTOMATED HEAD UP POSITION (AHUP) CPR RESULTED IN A SYNERGISTIC AND REMARKABLE IMPROVEMENT IN CORONARY AND CEREBRAL PERFUSION PRESSURES AFTER MORE THAN 30 MINUTES OF AHUP-CPR IN PIGS. THIS DISCOVERY SUGGESTS THAT REBOA + AHUP CPR MAY PROVIDE MORE DEFINITIVE TREATMENT FOR R-CA OR, AT A MINIMUM, A BRIDGE TO MORE DEFINITIVE THERAPY. | Department of Health and Human Services | A NOVEL SMALL MOLECULE FOR THE PREVENTION AND TREATMENT OF AGE-RELATED MACULAR DEGENERATION - SUMMARY AGE-RELATED MACULAR DEGENERATION (AMD) IS THE LEADING CAUSE OF BLINDNESS IN THE ELDERLY IN THE U.S. WET AMD ACCOUNTS FOR UP TO 90% OF VISION LOSS IN AMD PATIENTS, WHICH IS CHARACTERIZED BY CHOROIDAL NEOVASCULARIZATION (CNV). CURRENTLY, NO CURE EXISTS FOR AMD, AND AVAILABLE DRUG THERAPIES FOCUS ON INTRAVITREAL INJECTION OF ANTI- VASCULAR ENDOTHELIAL GROWTH FACTOR (VEGF) AGENTS. THESE ANTI-VEGF DRUGS HAVE ACHIEVED IMPRESSIVE THERAPEUTIC EFFECTS ON CNV IN APPROXIMATELY 70% OF AMD PATIENTS; HOWEVER, THE OTHER 30% OF PATIENTS REMAIN UNRESPONSIVE TO ANTI-VEGF THERAPIES. AMD IS A MULTI-FACTORIAL DISEASE,THERAPIES THAT ONLY TARGET A SINGLE MOLECULE (SUCH AS VEGF) DO NOT BENEFIT FOR ALL PATIENTS WITH AMD. THEREFORE, THE DEVELOPMENT OF NEW DRUGS TO TARGET MULTIPLE PATHOGENIC FACTORS WITH A SINGLE COMPOUND, IS AN UNMET CLINICAL NEED. PEROXISOME PROLIFERATOR-ACTIVATED RECEPTOR A (PPARA) IS A WIDELY EXPRESSED, LIGAND-ACTIVATED TRANSCRIPTIONAL FACTOR. RECENT STUDIES PROVIDE STRONG EVIDENCE SUPPORTING THAT PPARA AGONIST COULD SUPPRESS THE VICIOUS CYCLE OF CHRONIC INFLAMMATION IN THE RPE AND RETINA, A KEY CAUSE IN THE PATHOGENESIS OF AMD. IN ADDITION, CLINICAL STUDIES HAVE DEMONSTRATED THAT ORAL TREATMENT OF FENOFIBRATE (AN AGONIST OF PPARA) COULD SIGNIFICANTLY REDUCE THE NEED OF LASER PHOTOCOAGULATION FOR RETINAL NEOVASCULARIZATION BY 37-40%. PPARA AGONIST INHIBITS NEOVASCULARIZATION AND OVERPRODUCTION OF MULTIPLE INFLAMMATORY FACTORS IN AMD ANIMAL MODELS. MEANWHILE, AS SMALL MOLECULES, PPARA AGONISTS CAN PENETRATE THROUGH THE BLOOD-RETINAL BARRIER, MAKING IT A PROMISING NOVEL ORAL OPTION FOR THE PREVENTION AND TREATMENT OF AMD. HOWEVER, FENOFIBRATE’S LOW SPECIFICITY, LOW POTENCY, AND DOSE-LIMITING TOXICITIES MAKE IT A NON-IDEAL TREATMENT OPTION FOR AMD. RECENTLY, OUR TEAM HAS INDEPENDENTLY DESIGNED, SYNTHESIZED, AND SCREENED MORE THAN 200 NEW PPARA AGONISTS, FROM WHICH WE IDENTIFIED A NOVEL NON-FIBRATE COMPOUND A190, WITH SIGNIFICANTLY IMPROVED POTENCY AND SELECTIVITY FOR PPARA AGONISM (EC50 = ~37 NM). IT ACTIVATES PPARA AND UPREGULATES EXPRESSION OF PPARA TARGET GENES IN RETINAL CELLS, AND CONFERS ANTI-ANGIOGENIC AND ANTI-INFLAMMATORY, AND NEUROPROTECTIVE EFFECTS SUPERIOR TO FENOFIBRATE. IT ALSO REDUCES RETINAL INFLAMMATION AND VASCULAR LEAKAGE IN AMD MODELS. WE WILL EVALUATE THE EFFECTS OF A190 ON RETINAL NV AND INFLAMMATION IN AMD ANIMAL MODELS. THIS BIOMASS CAN THEN BE USED FOR OTHER PURPOSES, SUCH AS BIO-DIESEL FUEL. TO DATE, HIGH PH ALGAL GROWTH SYSTEMS HAVE NOT BEEN ADEQUATELY EXPLORED EVEN THOUGH, AT THESE CONDITIONS, CARBON DIOXIDE IS CHEMICALLY CONSUMED AT THE AIR-WATER INFERENCE. TO BETTER UNDERSTAND THE POTENTIAL OF THE HIGH PH/ALGAL SYSTEM, EXPERIMENTS AND MATHEMATICAL MODELING OF THE DATA WILL BE USED TO EXPLORE HIGH PH/ALGAL CARBON CAPTURE PROCESSES, FEASIBILITY, AND PERFORMANCE. THE MODEL CAN BE USED BY RESEARCHERS AND PRACTITIONERS TO ESTIMATE THE CARBON OFFSETS OF LARGE-SCALE CULTURES AIMED AT PRODUCTION OF HIGH-VALUE PRODUCES (E.G., BIOFUELS). THROUGH MODEL DEVELOPMENT, THE RESEARCH WILL CLARIFY THE ROLE OF CARBONATE AND DESCRIBE HOW TOTAL DISSOLVED INORGANIC CARBON SPECIES KINETICALLY INTERACT DURING ALGAL GROWTH. THE IMPACTS OF NITROGEN-SOURCE CYCLING AS A NATURAL, COST-EFFECTIVE STRATEGY TO MANAGE PH AND IMPROVE CARBON CAPTURE WILL ALSO BE EXPLORED. BROADER IMPACTS OF THE WORK INCLUDE BENEFITS TO SOCIETY BY SUPPORTING CLIMATE CHANGE MANAGEMENT AND WORKFORCE DEVELOPMENT THROUGH DESIGNING ALGAL PRODUCTION SYSTEMS THAT MAXIMIZE ATMOSPHERIC CARBON DIOXIDE REMOVAL. THIS CAN SERVE AS PART OF AN INTEGRATED CARBON MANAGEMENT PLAN TO MINIMIZE A MYRIAD OF ECONOMIC, ECOLOGICAL AND SOCIAL CONSEQUENCES OF RISING TEMPERATURES DUE TO HIGH CONCENTRATIONS OF GREENHOUSE GASES. THIS RESEARCH ENTAILS EXPERIMENTS WITH MIXED ALGAL CULTURES AND THE PRIMARY PRODUCT WILL BE AN EXPANDED, VALIDATED, ALGAL GROWTH MODEL FOR PREDICTING ALGAL BIOMASS PRODUCTION AND CARBON CAPTURE AS A FUNCTION OF INTERCONNECTED PARAMETERS OF TOTAL DISSOLVED INORGANIC CARBON AQUEOUS SPECIES AVAILABILITY, NITROGEN SOURCE TYPE, AND CULTURE PH. HIGH PH IS BEING INVESTIGATED BECAUSE AT HIGH PH MOST DISSOLVED TOTAL INORGANIC CARBON IS CONVERTED TO DISSOLVED CARBONATE, WHOSE SUITABILITY AS AN INORGANIC CARBON SOURCE HAS RECEIVED LITTLE ATTENTION. THUS, THIS RESEARCH SEEKS TO EXPAND AND VALIDATE A MATHEMATICAL MODEL TO QUANTIFY MIXED FRESHWATER ALGAL GROWTH IN HIGH PH SYSTEMS TO INFORM FUTURE CARBON CAPTURE EXPERIMENTS. THE EXPERIMENTS WILL BE CONDUCTED AT THE CLEMSON UNIVERSITY'S PARTITIONED AQUACULTURE SYSTEM. PROJECT GOALS ARE TO CHARACTERIZE AND MODEL TOTAL DISSOLVED INORGANIC CARBON-LIMITED GROWTH IN BATCH REACTORS THROUGH THE LENS OF FOUR OBJECTIVES, THE FIRST BEING TO IDENTIFY AND QUANTIFY THE ACCURACY OF ANALYTICAL METHODS FOR MEASURING INORGANIC CARBON SPECIES AT HIGH PH. THE SECOND AND THIRD ARE TO DETERMINE TO WHAT EXTENT CARBONATE KINETICALLY IMPACTS ALGAL GROWTH AND DETERMINE AN APPROPRIATE FORMULATION OF THE MONOD-TOTAL DISSOLVED INORGANIC CARBON-LIMITED GROWTH RATE OF MIXED CULTURE ALGAE. THE FOURTH GOAL WILL BE TO QUANTITATIVELY MEASURE CARBON CAPTURE BY THE ALGAL CULTURES IN OPEN BATCH REACTORS, DATA THAT WILL BE USED TO SUPPORT MODEL VALIDATION. ENGINEERS, CONSTRUCTION INDUSTRY SPECIALISTS, AND PROJECT MANAGERS ALL WORK TOGETHER TO PROVIDE COMMUNITIES AND PRIVATE SECTOR OWNERS WITH MORE RELIABLE AND SUSTAINABLE COMPLETED PROJECTS THAT MEET EVER-INCREASING MARKET DEMANDS AND STRENGTHEN THE FABRIC OF FLOURISHING COMMUNITIES. COLLABORATIVE RESEARCH: UNPACKING COMPUTATIONAL THINKING FOR ELEMENTARY TEACHERS AND LEARNERS -THE GOAL OF THIS PROJECT IS TO INVESTIGATE THE INTEGRATION OF COMPUTATIONAL THINKING (CT) INTO ELEMENTARY SCHOOL CURRICULA BY STUDYING HOW TEACHERS DEVELOP EXPERTISE IN INTEGRATING CT ACTIVITIES THAT ALIGN WITH INTERDISCIPLINARY STANDARDS AND EXISTING CURRICULA. THIS PROJECT ADDRESSES THE CRITICAL NEED FOR SCIENCE, TECHNOLOGY, ENGINEERING, AND MATHEMATICS (STEM) LITERACY BY INVESTIGATING EFFECTIVE MODELS OF PROFESSIONAL DEVELOPMENT (PD) THAT IMPROVE STEM TEACHING AND LEARNING. LEVERAGING AN ASSET-BASED APPROACH, THE PROJECT WILL PROVIDE OPPORTUNITIES TO BROADEN PARTICIPATION IN COMPUTER SCIENCE EDUCATION THROUGH BUILDING A COMMUNITY OF PRACTICE FOR TEACHERS AND DESIGNING CT-INFUSED CURRICULA. TO ENSURE THAT ALL TEACHERS HAVE THE NECESSARY SUPPORT TO PARTICIPATE IN NEW PEDAGOGICAL PRACTICES OF INCREASING COMPLEXITY, THE PROJECT WILL UTILIZE A COLLABORATIVE TEACHER PD AND MENTORSHIP STRUCTURE IN WHICH TEACHERS WILL WORK CLOSELY WITH PEERS AND PROJECT TEAM MEMBERS TO PREPARE, TEACH, AND REFLECT UPON LESSONS THROUGH MULTI-YEAR PARTICIPATION. THE RESEARCH WILL DOCUMENT HOW TEACHERS COME TO UNDERSTAND CT AND TAKE UP THESE PRACTICES IN THEIR CLASSROOMS. THE DISCOVERY RESEARCH PREK-12 PROGRAM (DRK-12) SEEKS TO SIGNIFICANTLY ENHANCE THE LEARNING AND TEACHING OF STEM BY PREK-12 STUDENTS AND TEACHERS, THROUGH RESEARCH AND DEVELOPMENT OF INNOVATIVE RESOURCES, MODELS AND TOOLS. PROJECTS IN THE DRK-12 PROGRAM BUILD ON FUNDAMENTAL RESEARCH IN STEM EDUCATION AND PRIOR RESEARCH AND DEVELOPMENT EFFORTS THAT PROVIDE THEORETICAL AND EMPIRICAL JUSTIFICATION FOR PROPOSED PROJECTS. OVER FOUR YEARS, THIS PROJECT WILL DIRECTLY IMPACT 135 TEACHERS IN MULTI-YEAR PARTICIPATION IN PROFESSIONAL DEVELOPMENT AND APPROXIMATELY 12,000 STUDENTS INDIRECTLY IN THEIR CLASSROOMS. THIS PROJECT WILL INITIALLY FOCUS ON SCHOOLS IN SOUTH CAROLINA, ESPECIALLY IN RURAL DISTRICTS. THE PROJECT WILL BE GUIDED BY THREE PRIMARY RESEARCH QUESTIONS: 1) HOW DO PREK-5 TEACHERS INTEGRATE CT INTO THEIR DISCIPLINARY TEACHING?; 2) WHAT ARE THE PERSONAL LEARNING TRAJECTORIES THAT TEACHERS MOVE THROUGH AS THEY WORK TOWARDS CT INTEGRATION?; 3) WHAT ARE THE BARRIERS AND ACCELERATORS FOR TEACHERS? IMPLEMENTATION OF CT-INFUSED LESSONS? THIS PROJECT IS GROUNDED IN THE IDEA THAT INCREMENTAL, SUSTAINABLE, AND SUBSTANTIAL TEACHER LEARNING COMES WHEN TEACHERS ARE FULLY INVESTED IN THE CYCLE OF LEARNING AND HAVE OPPORTUNITIES TO BUILD AND STRENGTHEN THEIR COMMUNITIES OF PRACTICE. IN REFLECTIVE PD SESSIONS HELD THROUGHOUT THE ACADEMIC YEAR AND SUMMER, TEACHERS WILL EXPLORE, CO-DESIGN, IMPLEMENT, AND REFINE STUDENT LEARNING ACTIVITIES THAT INTEGRATE CT INTO EXISTING CONTENT AREA STANDARDS AND CURRICULA. THE CT ACTIVITIES, WHICH WILL INCLUDE STANDARDS-BASED LESSON PLANS, ASSESSMENTS, AND SUPPLEMENTAL RESOURCES, WILL OFFER BOTH PLUGGED AND UNPLUGGED COMPONENTS, SO THAT STUDENTS AND TEACHERS HAVE OPPORTUNITIES TO UNDERSTAND THE CONCEPTUAL UNDERPINNINGS OF COMPUTATIONAL THINKING WITH AND WITHOUT COMPUTERS. A MINIMUM OF 120 LESSONS AND RELATED PEDAGOGICAL MATERIALS MAPPED TO CORE-CONTENT STANDARDS WILL BE DEVELOPED AND MADE FREELY AVAILABLE TO THE PUBLIC ON THE PROJECT WEBSITE. USING A MIXED METHODS APPROACH, THIS STUDY WILL DOCUMENT THE TRAJECTORIES THROUGH WHICH ELEMENTARY TEACHERS MOVE AS THEY BEGIN TO INTEGRATE CT INTO DISCIPLINARY TEACHING, AS WELL AS BARRIERS AND ACCELERATORS FACED AS THEY INTEGRATE CT INTO DISCIPLINARY TEACHING. QUANTITATIVE SURVEY METHODOLOGIES AND MULTIMODAL ANALYSIS OF NARRATIVE INTERVIEWS, CLASSROOM VIDEO, TEACHER REFLECTIONS, AND PEDAGOGICAL ARTIFACTS WILL BE USED TO ANALYZE TEACHERS? SENSE MAKING ABOUT CT OVER TIME AND THE UPTAKE OF NEW PEDAGOGICAL APPROACHES IN THEIR TEACHING. ULTIMATELY, THIS STUDY WILL PRODUCE A SET OF EMPIRICALLY-TESTED LEARNING TRAJECTORIES THAT CAN BE USED TO SUPPORT TEACHERS IN INTEGRATING CT INTO THEIR CLASSROOMS. AS A MEANS OF EXPANDING THE COMMUNITY OF PRACTICE, SHARING RESOURCES, AND DISSEMINATING PROJECT FINDINGS TO A NATIONWIDE AUDIENCE, THE PROJECT ALSO INCORPORATES A VIRTUAL PD COMPONENT TO SUPPORT PD ACCESS TO ELEMENTARY SCHOOL TEACHERS FROM UNDERREPRESENTED AND UNDERSERVED SCHOOLS ACROSS THE UNITED STATES. COLLABORATIVE RESEARCH: ENGAGING K-8 TEACHERS IN INTEGRATING COMPUTER SCIENCE, AI, AND CYBERSECURITY INTO STANDARD-BASED TEACHING -AS ARTIFICIAL INTELLIGENCE (AI) BECOMES INCREASINGLY EMBEDDED IN THE TECHNOLOGIES USED BY BOTH STUDENTS AND TEACHERS, IT IS ESSENTIAL FOR THEM TO UNDERSTAND HOW TO BE SAFE WHILE USING AI. FURTHERMORE, AI AND CYBERSECURITY TECHNOLOGY TOGETHER ARE BETTER AT DETECTING MALICIOUS ACTIVITIES THAN CONVENTIONAL SECURITY SYSTEMS. THE NEED TO BLEND THE TWO DISCIPLINES INTO A SINGLE, INTEGRATED CURRICULUM FOR K-8 EDUCATION IS HIGHLIGHTED BY THE INTERCONNECTEDNESS OF AI AND CYBERSECURITY AS COMPLEMENTARY SYSTEMS. THIS PROJECT WILL PLANT THE SEEDS OF THESE LITERACIES BY SPIRALING CONTENT ON TOPICS FROM COMPUTER PROGRAMMING, INTERNET FUNDAMENTALS, AND INTRODUCTION TO DATA AND AI ALONG WITH CYBERSECURITY TOPICS IN SMALL DOSES THROUGHOUT STUDENTS' K-8 EDUCATION. THIS PROJECT WILL LAY THE FOUNDATION FOR THE STUDENTS TO EVENTUALLY DEVELOP A COMPREHENSIVE UNDERSTANDING OF HOW DIFFERENT TECHNOLOGIES WORK AND INTERACT. THIS PROJECT AIMS TO INVESTIGATE HOW TEACHERS FROM RURAL SCHOOLS APPROACH THE INTEGRATION OF COMPUTER SCIENCE (CS), AND IN PARTICULAR, AI AND CYBERSECURITY COMPONENTS, INTO THEIR EXISTING CURRICULA. THIS PROJECT WILL DIRECTLY IMPACT AT LEAST 82 RURAL TEACHERS FROM SOUTH CAROLINA THROUGH MULTI-YEAR PARTICIPATION IN PROFESSIONAL DEVELOPMENT AND COACHING AND APPROXIMATELY 8,100 STUDENTS INDIRECTLY IN THEIR CLASSROOMS. RESOURCES, INCLUDING LESSON PLANS, DEVELOPED THROUGH THE PROJECT WILL BE MADE AVAILABLE AS OPEN ACCESS ON THE PROJECT WEBSITE, THROUGH PARTNERSHIPS, AND VIA CONFERENCE PRESENTATIONS AND PUBLICATIONS. THE OVERARCHING GOAL OF THIS PROJECT IS TO IMPROVE STEM TEACHING AND LEARNING BY EXAMINING SUCCESSFUL TEACHER PROFESSIONAL DEVELOPMENT (PD) MODELS IN RESPONSE TO A NEED FOR PREPARING STUDENTS WITH BOTH STEM LITERACY SKILLS AND OTHER TECHNICAL COMPETENCIES NEEDED TO BE INFORMED CITIZENS AND CONTRIBUTORS TO SOCIETY. THE PROJECT WILL BLEND AI AND CYBERSECURITY INTO A UNIFIED CURRICULA, SUPPORTED BY THE FOLLOWING OBJECTIVES: (1) ANALYZE HOW K-8 EDUCATORS INCORPORATE AI AND CYBERSECURITY TOPICS INTO DISCIPLINARY TEACHING COMPONENTS; (2) EXPLORE THE LEARNING SUPPORTS THAT K-8 EDUCATORS NEED TO EFFECTIVELY INCORPORATE AI AND CYBERSECURITY TOPICS INTO CURRENT CURRICULA TO ASSIST STUDENTS IN MEETING CONTENT-FOCUSED LEARNING OBJECTIVES; AND (3) COMPARE THE INTEGRATION OPPORTUNITIES AVAILABLE TO K-8 EDUCATORS IN RURAL AREAS OF SOUTH CAROLINA. IN THE END, HYPOTHESIZING THAT THIS RESEARCH CAN FORM THE BASIS FOR THE CREATION OF MORE SCALABLE PD PROGRAMS FOR K-8 TEACHERS AS WELL AS CONTINUOUS ASSISTANCE WITH CS INTEGRATION. THE RESEARCH WILL FOLLOW A MIXED-METHODS DESIGN, COMBINING SURVEY DATA, CLASSROOM OBSERVATIONS, AND INTERVIEWS TO EXAMINE HOW EDUCATORS MAKE SENSE OF AND APPLY THESE CONCEPTS OVER TIME. STATISTICAL ANALYSES WILL IDENTIFY SIGNIFICANT PATTERNS ACROSS TEACHER IMPLEMENTATION CONTEXTS. QUALITATIVE DATA--DRAWN FROM SEMI-STRUCTURED INTERVIEWS, TEACHING ARTIFACTS, VIDEO DOCUMENTATION, AND FIELD NOTES--WILL BE CODED TO SURFACE PEDAGOGICAL STRATEGIES, CHALLENGES, AND EVOLVING TEACHER UNDERSTANDINGS. IN ADDITION, AN APP WILL BE CREATED TO ALLOW TEACHERS TO DOCUMENT REAL-TIME CLASSROOM INTEGRATION OF AI AND CYBERSECURITY, PROVIDING A ROBUST, LONGITUDINAL DATASET. THE FINDINGS WILL INFORM SCALABLE, CONTEXT-SENSITIVE PROFESSIONAL DEVELOPMENT MODELS AND CONTRIBUTE TO A BROADER UNDERSTANDING OF HOW TO SUPPORT INTERDISCIPLINARY CS LEARNING IN K-8 SETTINGS. ALL DATA COLLECTION WILL BE DESIGNED TO ENSURE METHODOLOGICAL RIGOR AND TRUSTWORTHINESS THROUGH TRIANGULATION AND ONGOING MEMBER CHECKS WITH PARTICIPATING EDUCATORS. A NOVEL SMALL MOLECULE FOR THE PREVENTION AND TREATMENT OF DIABETIC RETINOPATHY - SUMMARY DIABETIC RETINOPATHY (DR) IS A LEADING CAUSE OF BLINDNESS IN THE UNITED STATES. CURRENTLY, MOST TREATMENTS FOR DR ARE ONLY APPLICABLE AT THE LATE STAGE AND ARE INVASIVE. ANTI-VEGF (VASCULAR ENDOTHELIAL GROWTH FACTOR) AGENTS HAVE ACHIEVED IMPRESSIVE THERAPEUTIC EFFECTS AGAINST DR AND NEOVASCULARIZATION (NV). HOWEVER, 40% OF PATIENTS WITH DR HAVE INADEQUATE RESPONSES TO THE TREATMENT. GIVEN THAT DR IS A MULTI-FACTORIAL DISEASE, THERAPIES THAT TARGET ONE SINGLE PATHOGENIC FACTOR (SUCH AS VEGF) ARE CLEARLY INSUFFICIENT TO ACHIEVE DESIRED EFFICACY IN ALL DR PATIENTS. THEREFORE, THE DEVELOPMENT OF NOVEL, NON-INVASIVE, LONG-TERM AND MORE EFFECTIVE PHARMACOLOGICAL TREATMENTS FOR DR IS URGENTLY REQUIRED, PARTICULARLY AT AN EARLY STAGE OF THE DISEASE. THE OVERALL OBJECTIVE OF THIS PHASE II PROJECT IS TO DEVELOP A NOVEL DRUG FOR THE ORAL PHARMACOTHERAPY OF DR. PEROXISOME PROLIFERATOR-ACTIVATED RECEPTOR A (PPARA) IS A LIGAND-ACTIVATED NUCLEAR RECEPTOR THAT REGULATES MITOCHONDRIAL LIPID SS OXIDATION AND PEROXISOME SCAVENGE HYDROGEN PEROXIDE IN THE CYTOPLASM. TWO LARGE AND LONGITUDINAL CLINICAL STUDIES REPORTED INDEPENDENTLY THAT ORAL ADMINISTRATION OF PPARA AGONIST FENOFIBRATE HAS ROBUST THERAPEUTIC EFFECT ON DR IN TYPE 2 DIABETIC PATIENTS. THIS IS THE FIRST ORAL DRUG WITH PROVEN EFFECT IN DR PATIENTS. IN PRECLINICAL STUDIES WITH ANIMAL MODELS OF DR, PHARMACOLOGICAL ACTIVATION OF PPARA HAS BEEN SHOWN TO DECREASE RETINAL PATHOLOGIES TYPICALLY OBSERVED IN DR (E.G. PATHOGENIC NEOVASCULARIZATION, VASCULAR LEAKAGE AND INFLAMMATION IN THE RETINA), RESULTING IN A PROFOUND THERAPEUTIC EFFECT. THUS, PPARA REPRESENTS A NOVEL THERAPEUTIC TARGET FOR THE TREATMENT OF DR. HOWEVER, THERE ARE CURRENTLY NO FDA-APPROVED PPARA DRUGS FOR DR. OUR MEDICINAL CHEMISTRY TEAM HAS SYNTHESIZED AND IDENTIFIED A NON-FIBRATE, QUINOLINE-DERIVED SMALL MOLECULE A190, A NOVEL PPARA AGONIST WITH MORE POTENT ACTIVITY. IN SBIR PHASE I STUDIES, WE HAVE COMPLETED PROOF-OF-PRINCIPLE STUDIES DEMONSTRATING THAT SYSTEMIC ADMINISTRATION OF A190 IS MORE EFFECTIVE THAN FENOFIBRATE ON TWO DIABETIC ANIMAL MODELS. THESE STUDIES ESTABLISHED A190 AS A PROMISING DRUG CANDIDATE FOR FURTHER DEVELOPMENT. THE PROPOSED RESEARCH IN PHASE II WILL FOCUS ON EVALUATING IN VIVO SAFETY AND TOXICOLOGY OF A190 IN TWO SPECIES (RATS AND RABBITS). IN ADDITION, THE LONG- TERM EFFICACY OF ORALLY ADMINISTERED A190 WILL BE TESTED IN DR MODELS. THIS PHASE II PROGRAM INCLUDES THREE SPECIFIC AIMS. SPECIFIC AIM 1: EVALUATE AND OPTIMIZE THE EFFICACY OF A190 ON RETINAL VASCULAR LEAKAGE IN DR MODELS. SPECIFIC AIM 2: DETERMINE THE PHARMACOKINETICS OF A190 AND ITS OCULAR TISSUE DISTRIBUTION. CITADEL COMMUNITY MENTAL HEALTH CLINIC - CITADEL COMMUNITY BEHAVIORAL HEALTH CLINIC (CCBHC) IS DESIGNED TO NOT ONLY BRING TOGETHER CHEMICAL AND MENTAL HEALTH CARE,BUT TO COORDINATE A PERSON'S TOTAL HEALTH AND SOCIAL SERVICE NEEDS, INCLUDING PHYSICAL HEALTH CARE. IN MANY COMMUNITIES, A PERSON WITH A MENTAL ILLNESS WOULD NEED TO CONTACT SEVERAL DIFFERENT AGENCIES TO OBTAIN SERVICES, AND RARELY COULD THEY OBTAIN BOTH MENTAL AND SUBSTANCE USE DISORDER TREATMENT THROUGH THE SAME AGENCY. CCBHC WILL CHANGE THAT, OFFERING A RANGE OF COORDINATED SERVICES SUCH AS PRIMARY CARE SCREENING, CRISIS MENTAL HEALTH SERVICES AND CASE MANAGEMENT IN ADDITION TO MENTAL AND CHEMICAL HEALTH CARE. IN SHORT, CCBHC WILL SERVE AS A "ONE-STOP-SHOP" FOR BOTH ADULTS AND CHILDREN WHO HAVE TROUBLE OTHERWISE GETTING THE HELP THEY NEED. CCBHC WILL IMPROVE ACCESS, QUALITY, COORDINATION AND CONTINUUM OF CARE TO A MINIMUM OF 500 CLIENTS ANNUALLY, THROUGH THE USE OF EVIDENCE-BASED STANDARDIZED ASSESSMENT TOOLS AND PRACTICES; IMPLEMENTATION OF PERSON CENTERED AND/OR FAMILY CENTERED INFORMED CARE; INCREASE OF CARE COORDINATION AND RECOVERY SUPPORT SERVICES; AND INCLUSION OF YOUTH AND FAMILY/CAREGIVER PARTICIPATION AT ALL LEVELS. THE TARGET POPULATION IS ADULTS AND/OR YOUTH WHO ARE LOCALLY IDENTIFIED WITH PRIORITY GOING TO ADULTS AND/OR YOUTH, AS WELL AS YOUTH INVOLVED IN MULTIPLE SYSTEMS, SUCH AS WELFARE AND JUSTICE SYSTEMS. SERVICES WILL BE DELIVERED THROUGH COLLABORATIONS/PARTNERSHIPS BETWEEN CCBHC AND OTHER COMMUNITY-BASED TREATMENT PROVIDERS AND/OR DCO'S IN THE RIVERSIDE-SAN BERNARDINO-ONTARIO MSA. PEOPLE WHO UTILIZE CCBHC MAY RECEIVE COMPREHENSIVE OUTPATIENT MENTAL HEALTH AND SUBSTANCE USE SERVICES, INCLUDING PROVISION OF PSYCHOTROPIC/ANTI-PSYCHOTIC MEDICATIONS; PSYCHO-THERAPEUTIC INTERVENTIONS; PRIMARY CARE SCREENING AND MONITORING (INCLUDING HIV AND VIRAL HEPATITIS SCREENING); BEHAVIORAL HEALTH SCREENING, ASSESSMENT AND DIAGNOSIS, INCLUDING RISK MANAGEMENT; PSYCHIATRIC REHABILITATION SERVICES; INCLUDING 24-HOUR MOBILE CRISIS TEAMS, EMERGENCY CRISIS INTERVENTION AND CRISIS STABILIZATION; PATIENT-CENTERED TREATMENT/CRISIS PLANNING; TARGETED CASE MANAGEMENT; PEER AND FAMILY SUPPORT; INTENSIVE COMMUNITY BASED TREATMENT SERVICES FOR VETERANS AND MEMBERS OF THE ARMED FORCES; ASSERTIVE COMMUNITY TREATMENT; SOCIAL SUPPORT SERVICES AND/OR CONNECTIONS WITH OTHER SERVICE PROVIDERS. THE OVERALL OBJECTIVE OF THE PROJECT IS TO REDUCE DISPARITIES IN MENTAL HEALTH AND IMPROVE HEALTH OUTCOMES. ADDITIONALLY, THIS PROJECT AIMS TO SUPPORT 15 FELLOWS BY PROVIDING GRADUATE COURSEWORK CULMINATING IN A SOUTH CAROLINA ADD-ON CERTIFICATION ENDORSEMENT IN COMPUTER SCIENCE. THE PROJECT INTENDS TO ALLOW TEACHERS TO COLLABORATE WITH OTHER TEACHERS AND PROJECT STAFF TO CREATE A STATEWIDE RURAL COMPUTER SCIENCE TEACHER NETWORK, A TEACHER-DRIVEN PROFESSIONAL COMMUNITY FOCUSED ON ADDRESSING THE UNIQUE CHALLENGES AND OPPORTUNITIES FACED BY RURAL COMPUTER SCIENCE TEACHERS. THE PROPOSED PROJECT COMPONENTS ARE POSITIONED TO ENABLE HIGH-ACHIEVING PRACTICING TEACHERS TO BECOME COMPUTER SCIENCE TEACHER-LEADERS IN THEIR RURAL COMMUNITIES. THIS PROJECT AT THE CITADEL IS A COLLABORATION BETWEEN THE CITADEL?S ZUCKER FAMILY SCHOOL OF EDUCATION AND SWAIN FAMILY SCHOOL OF SCIENCE AND MATHEMATICS, AS WELL AS ORANGEBURG COUNTY SCHOOL DISTRICT AND THE COMPUTER SCIENCE TEACHERS ASSOCIATION. PROJECT GOALS INCLUDE: 1) BUILDING MASTER TEACHER FELLOWS' (MTFS) PEDAGOGICAL AND CS CONTENT KNOWLEDGE; 2) BUILDING MTFS' TEACHER LEADERSHIP SKILLS; AND 3) SUPPORTING MTFS TO SUSTAIN THE SOUTH CAROLINA CHAPTER OF THE CSTA. HOW DOES PARTICIPATION IN THE PROJECT IMPACT TEACHER SELF-EFFICACY AND IDENTITY AS A TEACHER LEADER? RQ3. HOW DOES A DISTRICT TEAM APPROACH COMPARE TO THE EXPERIENCES AND OUTCOMES OF SINGLE TEACHERS FROM DIFFERENT DISTRICTS IN DEVELOPING HIGHLY EFFECTIVE TEACHER LEADERS? THIS PROJECT IS DESIGNED TO ENHANCE CS EDUCATION IN SOUTH CAROLINA'S RURAL, HIGH-NEEDS SCHOOLS BY DEVELOPING 15 RURAL TEACHER LEADERS WITH ADVANCED PEDAGOGICAL AND LEADERSHIP SKILLS. PROFESSIONAL DEVELOPMENT (PD) CONTENT PLANS TO BE TAILORED TO THE SPECIFIC NEEDS OF THESE TEACHERS AND THEIR STUDENTS, PROPOSING TO IMPROVE CRITICAL THINKING, PROBLEM-SOLVING SKILLS, AND STUDENT MOTIVATION IN COMPUTER SCIENCE. THE PROJECT AIMS TO CREATE VIRTUAL PD, CURRICULUM RESOURCES, AND A STATEWIDE CS4SC TEACHER HUB TO SUPPORT SUSTAINABLE AND SCALABLE INTEGRATION OF THESE EFFORTS. COMMERCIALIZATION OF THE HEAD UP CPR SYSTEM FOR IMPROVING SURVIVAL AFTER CARDIAC ARREST - ABSTRACT SUDDEN CARDIAC ARREST (SCA) IS A LEADING CAUSE OF DEATH IN THE USA, WITH ONLY 3-20% NEUROLOGICALLY INTACT SURVIVAL FOR >350,000 OUT-OF-HOSPITAL SCA PATIENTS EACH YEAR. EVEN AFTER PATIENTS ARE INITIALLY RESUSCITATED, MANY DIE WITHIN A WEEK FROM SEVERE BRAIN INJURY. THE GOAL OF THIS APPLICATION IS TO IMPROVE NEUROLOGICALLY-INTACT SURVIVAL RATES AFTER PRE-HOSPITAL AND IN-HOSPITAL SCA THROUGH THE COMMERCIALIZATION OF A NOVEL METHOD OF CPR CALLED HEAD UP POSITION (HUP) CPR. HUP CPR WORKS BY HARNESSING GRAVITY TO ENHANCE VENOUS BLOOD FLOW FROM THE BRAIN TO THE HEART, LOWER INTRACRANIAL PRESSURE, AND ENHANCE CARDIAC OUTPUT. THE FIRST CLINICAL HUP CPR DEVICE, THE ELEGARDTM, WAS DESIGNED, BUILT, AND TESTED WITH PHASE I FUNDING SUPPORT. THE ELEGARD HAS BEEN USED TO HELP TREAT >4500 SCA PATIENTS TO DATE. BASED UPON EARLY POSITIVE CLINICAL OUTCOMES, PHASE II FUNDING SUPPORTED THE DEVELOPMENT OF A REFINED AND MORE PORTABLE DEVICE TO ACCELERATE ADOPTION (ELEGARD 3.0). A SUBSEQUENT LIMITED LAUNCH EVALUATION IN 2024 WAS HIGHLY FAVORABLE. HOWEVER, IT REVEALED A COMPATIBILITY ISSUE WHERE SOME SHORTER PATIENTS WERE SLIPPING OUT OF POSITION DURING ELEVATION, LIKELY DUE TO EXPERIENCING GREATER HEAD AND THORAX ELEVATION COMPARED TO TALLER PATIENTS. AS A RESULT, POST-AWARD EFFORTS HAVE FOCUSED ON REFINING THE HEAD CRADLING MECHANISM TO ENSURE CONSISTENT HEAD ELEVATION AND MINIMIZE SLIPPAGE FOR PATIENTS OF ALL HEIGHTS. ADDITIONALLY, DURING THIS PERIOD, THE DEVICE’S CONTRACT MANUFACTURER SUDDENLY CLOSED, REQUIRING THE TRANSFER OF ELEGARD 3.0 ASSEMBLY AND PROCESSES TO A NEW PRODUCTION FACILITY. THIS TRANSITION REQUIRES SIGNIFICANT ADDITIONAL RESOURCES TO INTEGRATE THE DEVICE INTO A NEW PRODUCTION FACILITY AND QUALITY SYSTEM. GIVEN THAT THE SBIR CRP PROGRAM SUPPORTS LATE-STAGE DEVELOPMENT ACTIVITIES, RESUSCITATION INNOVATIONS (RI) HOPES TO SECURE FUNDING TO 1) OPTIMIZE, MANUFACTURE, AND VALIDATE AN UPDATED ELEGARD 3.0 HEAD CRADLING MECHANISM AND CARRYING CASE; AND 2) TRANSFER MANUFACTURING OF ELEGARD 3.0 TO A NEW ISO13854 CERTIFIED PRODUCTION FACILITY AND COMPLETE PROCESS VALIDATIONS AND DESIGN CONTROL TRANSFER FOR COMMERCIAL LAUNCH. SUCCESSFUL COMPLETION OF THESE AIMS WILL ACCELERATE COMMERCIALIZATION AND ADOPTION OF THIS INNOVATIVE TECHNOLOGY TO IMPROVE NEUROLOGICALLY-INTACT SURVIVAL RATES AFTER SCA.

THESE STATIONARY CRUSTACEANS HAVE EVOLVED TO SEEK OUT SPECIFIC HOSTS AS MOBILE HOMES BECAUSE THEY PROVIDE BARNACLES AN EASY MEANS OF DISPERSING AND FEEDING, WHILE GIVING THEM PROTECTION FROM PREDATORS. BARNACLES ATTACH TO TURTLES AND WHALES BY THREE MECHANISMS: THERE ARE ?GLUERS? WHICH CEMENT THEMSELVES TO SHELL AND OTHER FIRM BODY PARTS WITH MINIMAL TO NO INVASIVE ACTION; ?GRIPPERS? WHICH CLING TO SOFT EPIDERMIS BY MEANS OF SKIN-PINCHING MECHANICS; AND ?GOUGERS? WHICH CAN PENETRATE TISSUE, SCUTES, SCALES, AND SOMETIMES TO THE UNDERLYING BONE. THIS RESEARCH WILL MAP GENETIC TAGS FROM BARNACLES COLLECTED WORLDWIDE TO 1) STUDY HOST CHOICE AND THE EVOLUTIONARY HISTORY OF BARNACLE ATTACHMENT, 2) DESCRIBE TO WHAT EXTENT BARNACLES MAY BE A ?FINGERPRINT? MANAGEMENT TOOL FOR TRACKING SEA TURTLE MOVEMENTS, AND 3) ESTABLISH A BASELINE DISTRIBUTION OF BARNACLE GENETIC TYPES FOR MONITORING ALTERATIONS IN SPECIES RANGES DUE TO CLIMATE CHANGE. THIS RESEARCH WILL TRAIN UNDERGRADUATE AND GRADUATE STUDENTS IN GENETICS AND EVOLUTIONARY ANALYSES AS WELL AS FIELD METHODS IN MARINE BIOLOGY VIA A FIELD COURSE IN COSTA RICA. OUTREACH INCLUDES THE DEVELOPMENT OF MUSEUM EXHIBITS AND THE PUBLICATION OF POPULAR ARTICLES FOR A BROAD AUDIENCE. THE EVOLUTIONARY HISTORY AND SPECIES DELINEATIONS OF THE CORONULOID BARNACLES (ARTHROPODA: CRUSTACEA: CIRRIPEDIA) IS UNCERTAIN. SPECIFICALLY, LITTLE IS KNOWN OF THEIR ROUTE TO EPIZOISM, THEIR DEGREE OF DEPENDENCE ON HOSTS FOR DISPERSAL AND DIVERSIFICATION, THEIR PATHWAYS TO VARIABLE ATTACHMENT MECHANISMS, AND THEIR CONTEMPORARY PHYLOGEOGRAPHIC DISTRIBUTIONS. THE RESEARCHERS AIM TO ADDRESS THESE DEFICIENCIES THROUGH PHYLOGENETIC AND POPULATION GENOMIC ANALYSIS WITH THREE PRINCIPLE OBJECTIVES: 1) CONSTRUCT A DEFINITIVE PHYLOGENY FOR THE WHALE AND TURTLE BARNACLES AND MAP TO IT THE EVOLUTIONARY CHRONOLOGY OF THEIR GLUING, GRIPPING, AND GOUGING ATTACHMENT MODES; 2) COMPARE GLOBAL PHYLOGEOGRAPHIC PATTERNS USING GENOME-WIDE MARKERS FOR THE THREE MOST-COMMON BARNACLE SPECIES FROM THE FOUR MOST-COMMON SEA TURTLE SPECIES, ASSESSING POPULATION CONNECTIVITY IN THE CONTEXT OF OCEANOGRAPHIC LARVAL DISPERSAL (GEOGRAPHIC LOCALITIES) VS. HOST TRANSPORT (HOST LINEAGES); AND 3) TEST WHETHER BARNACLE GENETIC LINEAGES WITHIN SPECIES ASSORT WITH RESPECT TO SEA TURTLE SPECIES. A NETWORK OF INTERNATIONAL COLLABORATORS AT 16 GLOBAL LOCALITIES WILL HELP COLLECT, FROM MORE THAN 130 SEA TURTLES, ~2,200 TOTAL BARNACLE SPECIMENS FOR POPULATION GENOMIC ANALYSIS. HEAD UP CPR AND AORTIC OCCLUSION TO IMPROVE SURVIVAL AFTER REFRACTORY CARDIAC ARREST - ABSTRACT IN 2024 THERE WILL BE NEARLY 800,000 CARDIAC ARREST CASES IN THE US ALONE. OF THOSE ONLY ABOUT 50,000 WILL HAVE A FULL RECOVERY. MORE THAN 750,000 PATIENTS WILL HAVE NO HOPE FOR MEANINGFUL SURVIVAL. THAT IS BECAUSE THERE IS NO CURRENTLY DEMONSTRABLE EFFECTIVE TREATMENT FOR REFRACTORY (R)-CA PATIENTS, THE ENORMOUS SUBGROUP OF PATIENTS WHO INVARIABLY DIE ANNUALLY FROM THIS LEADING CAUSE OF DEATH. DEPENDING UPON THE PRESENTING RHYTHM, CA BECOMES REFRACTORY TO CURRENT INTERVENTIONS, HEREIN DEFINED AS A <2% SURVIVAL, <10 MINUTES AFTER STARTING CONVENTIONAL (C) CPR. THIS APPLICATION IS FOCUSED ON A BETTER TREATMENT FOR PATIENTS WITH CA REFRACTORY TO CURRENT MANAGEMENT STRATEGIES, REGARDLESS OF THE INITIAL RHYTHM PRESENTATION. PATIENTS IN R-CA CANNOT BE RESUSCITATED WITH C-CPR, DEFIBRILLATION FOR A SHOCKABLE RHYTHM, AND VASOPRESSOR THERAPY, SUCH AS EPINEPHRINE, FLUIDS, AND ANTI-ARRHYTHMIC DRUGS. FOR OVER A HALF CENTURY WE HAVE FAILED TO EFFECTIVELY TREAT R-CA: WE HAVE NOT KNOWN HOW. OCCASIONALLY TRANSCUTANEOUS CARDIAC PACING, ECMO AND/OR DOUBLE SEQUENTIAL DEFIBRILLATION CAN BE USED WITH SOME BENEFIT FOR THE VERY SMALL PERCENTAGE OF PATIENTS THAT REMAIN IN REFRACTORY VENTRICULAR FIBRILLATION (R-VF). HOWEVER, IN GENERAL THESE TREATMENT STRATEGIES RARELY WORK FOR R-CA, ESPECIALLY IN PATIENTS WITH A NON-SHOCKABLE PRESENTING RHYTHM THAT COMPRISE 75-85% OF ALL CA CASES. HOWEVER, THE APPLICANTS RECENTLY OBSERVED THAT THE ADDITION OF RESUSCITATIVE ENDOVASCULAR BALLOON OCCLUSION OF THE AORTA (REBOA) TO AUTOMATED HEAD UP POSITION (AHUP) CPR RESULTED IN A SYNERGISTIC AND REMARKABLE IMPROVEMENT IN CORONARY AND CEREBRAL PERFUSION PRESSURES AFTER MORE THAN 30 MINUTES OF AHUP-CPR IN PIGS. THIS DISCOVERY SUGGESTS THAT REBOA + AHUP CPR MAY PROVIDE MORE DEFINITIVE TREATMENT FOR R-CA OR, AT A MINIMUM, A BRIDGE TO MORE DEFINITIVE THERAPY. BASED UPON THESE NEW FINDINGS, THE APPLICANTS INTEND TO TEST THE HYPOTHESIS THAT THE ADDITION OF REBOA TO AHUP-CPR FOR R- CA WILL SIGNIFICANTLY IMPROVE BRAIN AND VITAL ORGAN BLOOD FLOW AND INCREASE THE LIKELIHOOD OF NEUROLOGICALLY- FAVORABLE SURVIVAL IN A WELL-ESTABLISHED PRE-CLINICAL MODEL.

A NOVEL SMALL MOLECULE FOR THE PREVENTION AND TREATMENT OF AGE-RELATED MACULAR DEGENERATION - SUMMARY AGE-RELATED MACULAR DEGENERATION (AMD) IS THE LEADING CAUSE OF BLINDNESS IN THE ELDERLY IN THE U.S. WET AMD ACCOUNTS FOR UP TO 90% OF VISION LOSS IN AMD PATIENTS, WHICH IS CHARACTERIZED BY CHOROIDAL NEOVASCULARIZATION (CNV). CURRENTLY, NO CURE EXISTS FOR AMD, AND AVAILABLE DRUG THERAPIES FOCUS ON INTRAVITREAL INJECTION OF ANTI- VASCULAR ENDOTHELIAL GROWTH FACTOR (VEGF) AGENTS. THESE ANTI-VEGF DRUGS HAVE ACHIEVED IMPRESSIVE THERAPEUTIC EFFECTS ON CNV IN APPROXIMATELY 70% OF AMD PATIENTS; HOWEVER, THE OTHER 30% OF PATIENTS REMAIN UNRESPONSIVE TO ANTI-VEGF THERAPIES. AMD IS A MULTI-FACTORIAL DISEASE,THERAPIES THAT ONLY TARGET A SINGLE MOLECULE (SUCH AS VEGF) DO NOT BENEFIT FOR ALL PATIENTS WITH AMD. THEREFORE, THE DEVELOPMENT OF NEW DRUGS TO TARGET MULTIPLE PATHOGENIC FACTORS WITH A SINGLE COMPOUND, IS AN UNMET CLINICAL NEED. PEROXISOME PROLIFERATOR-ACTIVATED RECEPTOR A (PPARA) IS A WIDELY EXPRESSED, LIGAND-ACTIVATED TRANSCRIPTIONAL FACTOR. RECENT STUDIES PROVIDE STRONG EVIDENCE SUPPORTING THAT PPARA AGONIST COULD SUPPRESS THE VICIOUS CYCLE OF CHRONIC INFLAMMATION IN THE RPE AND RETINA, A KEY CAUSE IN THE PATHOGENESIS OF AMD. IN ADDITION, CLINICAL STUDIES HAVE DEMONSTRATED THAT ORAL TREATMENT OF FENOFIBRATE (AN AGONIST OF PPARA) COULD SIGNIFICANTLY REDUCE THE NEED OF LASER PHOTOCOAGULATION FOR RETINAL NEOVASCULARIZATION BY 37-40%. PPARA AGONIST INHIBITS NEOVASCULARIZATION AND OVERPRODUCTION OF MULTIPLE INFLAMMATORY FACTORS IN AMD ANIMAL MODELS. MEANWHILE, AS SMALL MOLECULES, PPARA AGONISTS CAN PENETRATE THROUGH THE BLOOD-RETINAL BARRIER, MAKING IT A PROMISING NOVEL ORAL OPTION FOR THE PREVENTION AND TREATMENT OF AMD. HOWEVER, FENOFIBRATE’S LOW SPECIFICITY, LOW POTENCY, AND DOSE-LIMITING TOXICITIES MAKE IT A NON-IDEAL TREATMENT OPTION FOR AMD. RECENTLY, OUR TEAM HAS INDEPENDENTLY DESIGNED, SYNTHESIZED, AND SCREENED MORE THAN 200 NEW PPARA AGONISTS, FROM WHICH WE IDENTIFIED A NOVEL NON-FIBRATE COMPOUND A190, WITH SIGNIFICANTLY IMPROVED POTENCY AND SELECTIVITY FOR PPARA AGONISM (EC50 = ~37 NM). IT ACTIVATES PPARA AND UPREGULATES EXPRESSION OF PPARA TARGET GENES IN RETINAL CELLS, AND CONFERS ANTI-ANGIOGENIC AND ANTI-INFLAMMATORY, AND NEUROPROTECTIVE EFFECTS SUPERIOR TO FENOFIBRATE. IT ALSO REDUCES RETINAL INFLAMMATION AND VASCULAR LEAKAGE IN AMD MODELS. WE WILL EVALUATE THE EFFECTS OF A190 ON RETINAL NV AND INFLAMMATION IN AMD ANIMAL MODELS. THE PROJECT WILL BE ACHIEVED BY TWO SPECIFIC AIMS. SPECIFIC AIM 1: DETERMINE WHETHER A190 SUPPRESSES SUBRETINAL NV. SPECIFIC AIM 2: DETERMINE WHETHER A190 INHIBITS OXIDATIVE STRESS/INFLAMMATION. THE PROPOSED STUDIES WILL PROVE THE THERAPEUTIC EFFECT OF THIS COMPOUND ON AMD MODELS AND WILL LAY A SOLID GROUND FOR FURTHER PK, TOXICITY, MECHANISM STUDIES IN FUTURE PHASE II PROJECT.

THIS RESEARCH EXAMINES, THROUGH EXPERIMENT AND MODELING, THE POTENTIAL OF HIGH PH ALGAL CULTURES TO MAXIMIZE THE AMOUNT OF INORGANIC CARBON (THROUGH DISSOLVED CARBON SPECIES DERIVED FROM ATMOSPHERIC CO2) AVAILABLE FOR CONVERSION OF CO2 TO ALGAL BIOMASS. THIS BIOMASS CAN THEN BE USED FOR OTHER PURPOSES, SUCH AS BIO-DIESEL FUEL. TO DATE, HIGH PH ALGAL GROWTH SYSTEMS HAVE NOT BEEN ADEQUATELY EXPLORED EVEN THOUGH, AT THESE CONDITIONS, CARBON DIOXIDE IS CHEMICALLY CONSUMED AT THE AIR-WATER INFERENCE. TO BETTER UNDERSTAND THE POTENTIAL OF THE HIGH PH/ALGAL SYSTEM, EXPERIMENTS AND MATHEMATICAL MODELING OF THE DATA WILL BE USED TO EXPLORE HIGH PH/ALGAL CARBON CAPTURE PROCESSES, FEASIBILITY, AND PERFORMANCE. THE MODEL CAN BE USED BY RESEARCHERS AND PRACTITIONERS TO ESTIMATE THE CARBON OFFSETS OF LARGE-SCALE CULTURES AIMED AT PRODUCTION OF HIGH-VALUE PRODUCES (E.G., BIOFUELS). THROUGH MODEL DEVELOPMENT, THE RESEARCH WILL CLARIFY THE ROLE OF CARBONATE AND DESCRIBE HOW TOTAL DISSOLVED INORGANIC CARBON SPECIES KINETICALLY INTERACT DURING ALGAL GROWTH. THE IMPACTS OF NITROGEN-SOURCE CYCLING AS A NATURAL, COST-EFFECTIVE STRATEGY TO MANAGE PH AND IMPROVE CARBON CAPTURE WILL ALSO BE EXPLORED. BROADER IMPACTS OF THE WORK INCLUDE BENEFITS TO SOCIETY BY SUPPORTING CLIMATE CHANGE MANAGEMENT AND WORKFORCE DEVELOPMENT THROUGH DESIGNING ALGAL PRODUCTION SYSTEMS THAT MAXIMIZE ATMOSPHERIC CARBON DIOXIDE REMOVAL. THIS CAN SERVE AS PART OF AN INTEGRATED CARBON MANAGEMENT PLAN TO MINIMIZE A MYRIAD OF ECONOMIC, ECOLOGICAL AND SOCIAL CONSEQUENCES OF RISING TEMPERATURES DUE TO HIGH CONCENTRATIONS OF GREENHOUSE GASES. TO CARRY OUT THE WORK, STUDENTS FROM UNDERREPRESENTED GROUPS IN THE SCIENCES WILL BE ACTIVELY RECRUITED. THIS RESEARCH ENTAILS EXPERIMENTS WITH MIXED ALGAL CULTURES AND THE PRIMARY PRODUCT WILL BE AN EXPANDED, VALIDATED, ALGAL GROWTH MODEL FOR PREDICTING ALGAL BIOMASS PRODUCTION AND CARBON CAPTURE AS A FUNCTION OF INTERCONNECTED PARAMETERS OF TOTAL DISSOLVED INORGANIC CARBON AQUEOUS SPECIES AVAILABILITY, NITROGEN SOURCE TYPE, AND CULTURE PH. HIGH PH IS BEING INVESTIGATED BECAUSE AT HIGH PH MOST DISSOLVED TOTAL INORGANIC CARBON IS CONVERTED TO DISSOLVED CARBONATE, WHOSE SUITABILITY AS AN INORGANIC CARBON SOURCE HAS RECEIVED LITTLE ATTENTION. THUS, THIS RESEARCH SEEKS TO EXPAND AND VALIDATE A MATHEMATICAL MODEL TO QUANTIFY MIXED FRESHWATER ALGAL GROWTH IN HIGH PH SYSTEMS TO INFORM FUTURE CARBON CAPTURE EXPERIMENTS. THE EXPERIMENTS WILL BE CONDUCTED AT THE CLEMSON UNIVERSITY'S PARTITIONED AQUACULTURE SYSTEM. PROJECT GOALS ARE TO CHARACTERIZE AND MODEL TOTAL DISSOLVED INORGANIC CARBON-LIMITED GROWTH IN BATCH REACTORS THROUGH THE LENS OF FOUR OBJECTIVES, THE FIRST BEING TO IDENTIFY AND QUANTIFY THE ACCURACY OF ANALYTICAL METHODS FOR MEASURING INORGANIC CARBON SPECIES AT HIGH PH. THE SECOND AND THIRD ARE TO DETERMINE TO WHAT EXTENT CARBONATE KINETICALLY IMPACTS ALGAL GROWTH AND DETERMINE AN APPROPRIATE FORMULATION OF THE MONOD-TOTAL DISSOLVED INORGANIC CARBON-LIMITED GROWTH RATE OF MIXED CULTURE ALGAE. THE FOURTH GOAL WILL BE TO QUANTITATIVELY MEASURE CARBON CAPTURE BY THE ALGAL CULTURES IN OPEN BATCH REACTORS, DATA THAT WILL BE USED TO SUPPORT MODEL VALIDATION. AN ADDITIONAL OBJECTIVE WILL BE TO EXPLORE HOW NITROGEN SOURCE CYCLING COULD BE USED TO MANAGE PH FOR CARBON CAPTURE IN THE CULTURING REACTOR SO IMPACTS OF NITROGEN ON PH AND BIOMASS PRODUCTION CAN BE ESTIMATED AND TO DELIVER ESTIMATES OF CARBON CAPTURE BASED ON SEVERAL NUTRIENT LOADING SCENARIOS. DURING THE PROJECT, A GRADUATE STUDENT AND SEVERAL ADDITIONAL STUDENTS WILL BE ENGAGED. UNDER THIS COOPERATIVE AGREEMENT, IAF PARTNER ASOCIACIÓN DE CAPACITACIÓN E INVESTIGACIÓN PARA LA SALUD MENTAL (ACISAM) WILL WORK IN FOUR COUNTRIES TO FURTHER DEVELOP A NETWORK OF NINE GRASSROOTS ASSOCIATIONS FOCUSING ON SERVICES FOR PEOPLE WITH PSYCHIATRIC DISABILITIES AND FOR THEIR CAREGIVERS. THE PARTNER AND THE ASSOCIATIONS WILL COLLABORATE ON ORGANIZING EDUCATIONAL EVENTS; FACILITATING COMMUNICATIONS; IMPROVING THEIR ADMINISTRATION, LEADERSHIP AND PARTNERSHIPS; AND MANAGING A SUBGRANT FUND FINANCING ECONOMIC INITIATIVES, TRAINING AND OUTREACH. IN THIS COOPERATIVE AGREEMENT, ACISAM WILL WORK WITH THE IAF ON REGIONAL EVENTS, SUBGRANT REVIEW AND DISSEMINATION OF LESSONS. SOME 800 SALVADORANS, NICARAGUANS, PANAMANIANS AND COSTA RICANS WILL BENEFIT DIRECTLY AND ANOTHER 7,000 INDIRECTLY.

(CICADES) WILL WORK WITH FARMERS OF THE INTEGRADORA DE CAFÉS DE ESPECIALIDAD DE LAS ALTAS MONTAÑAS (INCAFESAM) IN OAXACA, PUEBLA AND VERACRUZ TO STRENGTHEN THESE GROWERS’ COMPETITIVENESS AND SUSTAINABILITY. PROJECT ACTIVITIES WILL HELP IMPROVE ORGANIC COFFEE PRODUCTION, PROCESSING, AND SALES FOR SPECIALTY EXPORT MARKETS AND TO SATISFY DOMESTIC DEMAND FOR ROASTED COFFEE. FARMERS WILL USE ORGANIC FERTILIZERS AND LEARN ABOUT TECHNIQUES TO PRODUCE HIGH QUALITY FUNGUS-RESISTANT COFFEE PLANTS. THIS PROJECT WILL ALSO PROMOTE THE ORGANIZATIONAL GROWTH FOR INCAFESAM AND ENCOURAGE YOUTH PARTICIPATION THROUGH TRAINING AND OPPORTUNITIES IN SALES AND COFFEE CUPPING. THE GRANT WILL BENEFIT 490 PEOPLE DIRECTLY AND 2,000 INDIRECTLY.
