| Department of Health and Human Services | HUMAN-INFORMED DATA-DRIVEN DEVELOPMENT OF NEXT-GENERATION T CELL VACCINE AGAINST MALARIA - PA-19-077 DOOLAN: PROJECT SUMMARY MALARIA REMAINS A GLOBAL PUBLIC HEALTH PROBLEM WITH NEARLY HALF OF THE WORLD'S POPULATION AT RISK. AN EFFECTIVE MALARIA VACCINE WOULD PREVENT ALMOST HALF A MILLION DEATHS AND OVER 200 MILLION CLINICAL CASES EACH YEAR AND HELP ERADICATE THE DISEASE. THE MOST EFFECTIVE EXPERIMENTAL MALARIA VACCINATION REGIMENS TO DATE ARE RADIATION- ATTENUATED SPOROZOITES (RAS; PFSPZ) AND INFECTIOUS SPOROZOITES ADMINISTERED UNDER CHEMOPROPHYLAXIS (CPS). PROTECTIVE IMMUNITY IS BELIEVED TO BE MEDIATED BY CD8+ T CELLS WHICH ATTACK THE PARASITE DURING THE PRE- ERYTHROCYTIC (PE) LIVER-STAGE OF INFECTION. HOWEVER, THE KEY ANTIGENS UNDERLYING THIS PROTECTION ARE LARGELY UNKNOWN. WE HAVE DEVELOPED AND APPLIED A PROTEOME-WIDE T CELL SCREENING APPROACH TO IDENTIFY THE SUBSET OF KEY ANTIGENS TARGETED BY T CELL RESPONSES FROM THE COMPLETE PLASMODIUM FALCIPARUM PARASITE PROTEOME. WE HAVE SHOWN THAT THE ANTIGENS PREFERENTIALLY RECOGNIZED BY T CELLS ARE DISTINCT FROM ANTIBODY TARGETS. HERE, WE WILL CHARACTERIZE AND CREDENTIAL HIGHLY RANKED PRE-ERYTHROCYTIC P. FALCIPARUM T CELL ANTIGENS FROM OUR UNIQUE DATASET, FOCUSING ON THOSE THAT ARE CATEGORIZED AS EXCLUSIVELY T CELL TARGETS, SINCE T CELLS DIRECTED AGAINST LIVER- STAGE ANTIGENS ARE CONSIDERED THE PRIMARY IMMUNE EFFECTORS REQUIRED FOR STERILE IMMUNITY AGAINST MALARIA WHICH PREVENTS DISEASE AND STOPS TRANSMISSION. OUR SELECTION CRITERIA WILL CONSIDER EXTENT OF SEQUENCE CONSERVATION ACROSS PLASMODIUM STRAINS AND SPECIES, TO TARGET A VACCINE THAT CONFERS STRAIN TRANSCENDING AND CROSS-SPECIES PROTECTION. WE WILL USE CLINICALLY RELEVANT SELECTION CRITERIA GOVERNED BY THE CAPACITY OF THE ANTIGEN TO PROTECT AGAINST VIRULENT P. YOELII PARASITE CHALLENGE IN ESTABLISHED PRECLINICAL MODELS AND TO BE RECOGNIZED BY RECALL PLASMODIUM-SPECIFIC IMMUNE RESPONSES IN PROTECTIVE HUMAN MODELS. WE WILL USE TWO INNOVATIVE VACCINE DELIVERY PLATFORMS SELECTED FOR CAPACITY TO INDUCE ROBUST AND SUSTAINED T CELL RESPONSES, WITH A SPECIFIC FOCUS ON THE INDUCTION OF LIVER-SPECIFIC RESIDENT MEMORY T CELLS (TRM) TARGETING THE PUTATIVE SITE OF IMMUNE ACTION. OUR SELECTED REGIMENS INCLUDE “PRIME-TARGET” COMPRISING A DNA PRIME FOLLOWED BY ADENOVIRUS BOOST DELIVERED INTRAVENOUSLY TO TARGET THE LIVER, AND A LIPOSOMAL MRNA VACCINE PLATFORM WITH AN INCORPORATED AGONIST (CA) TO ACTIVATE NKT CELLS AND LINK INNATE AND ADAPTIVE IMMUNITY; BOTH PLATFORMS CAN INDUCE SUSTAINED T CELL RESPONSES AND ARE CAPABLE OF PROTECTING MICE AGAINST SPOROZOITE CHALLENGE. WE WILL ASSIGN PRIORITIES FOR VACCINE DEVELOPMENT ACCORDING TO THEIR CREDENTIALS, EVALUATE COMBINATIONS FOR SYNERGY, AND DOWN-SELECT FOR CLINICAL DEVELOPMENT THE SET OF ANTIGENS THAT HAVE A MAXIMUM LIKELIHOOD OF INDUCING ROBUST AND SUSTAINED PROTECTIVE IMMUNITY AGAINST MALARIA IN HUMANS. | Department of Health and Human Services | WHAT CAUSES LOW BACK PAIN TO FLARE: HAS A MAJOR OPPORTUNITY TO UNDERSTAND BACK PAIN BEEN MISSED? - PROJECT SUMMARY/ABSTRACT LOW BACK PAIN (LBP) IS TRADITIONALLY VIEWED AS EITHER A TRANSIENT CONDITION WITH ACUTE EPISODES OR A CHRONIC PERSISTING CONDITION. HOWEVER, IT IS INCREASINGLY RECOGNIZED THAT FOR MOST (>80% OF CASES), LBP HAS A VARIABLE AND ONGOING TRAJECTORY CHARACTERIZED BY FLUCTUATING SYMPTOMS – WITH PERIODS OF WORSE SYMPTOMS REFERRED TO AS “FLARES”. CURRENT WORK REGARDING RISK FACTORS FOR LBP ONLY CONSIDERS THOSE ASSOCIATED WITH A NEW EPISODE OR THE TRANSITION TO CHRONIC LBP. FINDINGS FROM THAT WORK HAS FAILED TO DELIVER EFFECTIVE STRATEGIES TO PREVENT NEW EPISODES OR CHRONICITY, NOR REDUCE THE EXCESSIVE BURDEN OF LBP. THE UNDERLYING PREMISE OF THIS PROPOSAL IS THAT FLUCTUATION OR FLARING OF LBP IS MEDIATED BY TRANSIENT/FLUCTUATING EXPOSURE TO A RANGE OF FACTORS ACROSS MULTIPLE DOMAINS, THAT DIFFER BETWEEN INDIVIDUALS, AND IDENTIFICATION OF THESE FACTORS COULD PROVIDE A BASIS FOR A COMPLETELY NEW MODEL TO PERSONALIZE CARE. THIS PROJECT WILL TAKE A FRESH LOOK AT LBP BY DIRECTLY ADDRESSING THIS QUESTION USING INNOVATIVE METHODS THAT ARE ENABLED BY RECENT ADVANCES IN TECHNOLOGY. THE WORK IS BASED ON OUR EXTENSIVE COLLABORATION WITH PEOPLE WITH LBP THAT HAS PROVIDED CRITICAL INSIGHT INTO HOW FLARES ARE DEFINED (WHICH SURPRISINGLY IS NOT BASED ON PAIN ALONE) AND THEIR PERSPECTIVE OF THE POTENTIAL CAUSES FOR LBP FLARES. OUR MULTIDISCIPLINARY RESEARCH TEAM PROPOSE AN INNOVATIVE CASE-CROSSOVER STUDY THAT WILL ADDRESS TWO AIMS. AIM 1 WILL EXAMINE, AT MULTIPLE TIMEPOINTS OVER A PERIOD OF 3 MONTHS, AN ARRAY OF CANDIDATE BIOLOGICAL/BEHAVIORAL, PSYCHOLOGICAL, SOCIAL, AND ENVIRONMENTAL RISK FACTORS FOR LBP FLARE THAT HAVE BEEN SELECTED BASED ON CONSUMER PERSPECTIVE, LITERATURE REVIEW AND OUR EXTENSIVE PRELIMINARY STUDIES. MEASURES WILL BE MADE IN THE “REAL WORD” USING ADVANCED WEARABLE AND ONLINE TECHNOLOGIES. AIM 2 WILL INVOLVE AN EMBEDDED STUDY OF INDIVIDUALS FROM THE MAIN PARTICIPANT COHORT WHO FLARE FREQUENTLY. FOR 7 DAYS, ELIGIBLE INDIVIDUALS WILL UNDERGO MORE INTENSIVE “REAL- WORLD” PHYSIOLOGICAL AND BIOMECHANICAL MEASURES USING AN INNOVATIVE COMBINATION OF WEARABLE SENSORS AND HOME-BASED SAMPLE/DATA COLLECTION TO INVESTIGATE POTENTIAL MECHANISMS UNDERLYING TWO SURPRISING PRELIMINARY OBSERVATIONS: (1) THAT POOR SLEEP IS A POTENT RISK FACTOR FOR A SUBSEQUENT FLARE (AS OPPOSED TO THE TRADITIONAL VIEW THAT PAIN INTERFERES WITH AND CAUSES POOR SLEEP), AND (2) THAT LOW RATHER THAN HIGH PHYSICAL ACTIVITY IS A RISK FACTOR FOR LBP FLARE. WE AIMS TO IDENTIFY NOT ONLY IF, BUT HOW, DAILY VARIATIONS IN SLEEP QUALITY AND PHYSICAL ACTIVITY – TWO OF THE MOST IMPORTANT, YET MODIFIABLE HEALTH BEHAVIORS – DRIVE FLARE OF LBP. TOGETHER THESE DATA WILL FILL A MAJOR GAP BY PROVIDING NEW UNDERSTANDING OF MECHANISMS THAT DRIVE THE FLUCTUATING EXPERIENCE OF LBP. WE WILL ALSO OVER- SAMPLE INDIVIDUALS WITH SEVERE AND ENDURING AN (SE-AN), WHOSE DNA MAY BE ENRICHED FOR CAUSATIVE ALLELES. WE HAVE DEVELOPED A NOVEL TOOL, NAMED NANOSCALE SPATIOTEMPORAL INDEXING CLUSTERING (NASTIC), (WALLIS ET AL., BIORXIV, 2021, REF. 7), WHICH OFFERS UNPRECEDENTED INSIGHTS INTO THE DYNAMICS OF PROTEINS UNDERGOING LIQUID-LIQUID PHASE SEPARATION/TRANSITION IN LARGE AND NANOSCALE BMCS, IN LIVE NEURONS. WE IDENTIFIED TAU AND SYNUCLEIN AS CANDIDATES FOR THEIR ABILITY TO GENERATE SYNAPTIC BMCS IN HIPPOCAMPAL NEURONS. NASTIC ANALYSIS REVEALS THAT TAU MOLECULES CAN INDEED FORM SMALL NANOCLUSTERS IN LIVE HIPPOCAMPAL NEURONS IN WHICH THEY EXHIBIT VERY LOW MOBILITY AND SENSITIVITY TO 1,6-HEXANEDIOL, AN ALIPHATIC ALCOHOL USED TO INHIBIT WEAK MOLECULAR INTERACTIONS THAT MEDIATE BMCS. HUMAN-INFORMED DATA-DRIVEN DEVELOPMENT OF NEXT-GENERATION T CELL VACCINE AGAINST MALARIA - PA-19-077 DOOLAN: PROJECT SUMMARY MALARIA REMAINS A GLOBAL PUBLIC HEALTH PROBLEM WITH NEARLY HALF OF THE WORLD'S POPULATION AT RISK. AN EFFECTIVE MALARIA VACCINE WOULD PREVENT ALMOST HALF A MILLION DEATHS AND OVER 200 MILLION CLINICAL CASES EACH YEAR AND HELP ERADICATE THE DISEASE. THE MOST EFFECTIVE EXPERIMENTAL MALARIA VACCINATION REGIMENS TO DATE ARE RADIATION- ATTENUATED SPOROZOITES (RAS; PFSPZ) AND INFECTIOUS SPOROZOITES ADMINISTERED UNDER CHEMOPROPHYLAXIS (CPS). PROTECTIVE IMMUNITY IS BELIEVED TO BE MEDIATED BY CD8+ T CELLS WHICH ATTACK THE PARASITE DURING THE PRE- ERYTHROCYTIC (PE) LIVER-STAGE OF INFECTION. HOWEVER, THE KEY ANTIGENS UNDERLYING THIS PROTECTION ARE LARGELY UNKNOWN. WE HAVE DEVELOPED AND APPLIED A PROTEOME-WIDE T CELL SCREENING APPROACH TO IDENTIFY THE SUBSET OF KEY ANTIGENS TARGETED BY T CELL RESPONSES FROM THE COMPLETE PLASMODIUM FALCIPARUM PARASITE PROTEOME. WE HAVE SHOWN THAT THE ANTIGENS PREFERENTIALLY RECOGNIZED BY T CELLS ARE DISTINCT FROM ANTIBODY TARGETS. HERE, WE WILL CHARACTERIZE AND CREDENTIAL HIGHLY RANKED PRE-ERYTHROCYTIC P. FALCIPARUM T CELL ANTIGENS FROM OUR UNIQUE DATASET, FOCUSING ON THOSE THAT ARE CATEGORIZED AS EXCLUSIVELY T CELL TARGETS, SINCE T CELLS DIRECTED AGAINST LIVER- STAGE ANTIGENS ARE CONSIDERED THE PRIMARY IMMUNE EFFECTORS REQUIRED FOR STERILE IMMUNITY AGAINST MALARIA WHICH PREVENTS DISEASE AND STOPS TRANSMISSION. OUR SELECTION CRITERIA WILL CONSIDER EXTENT OF SEQUENCE CONSERVATION ACROSS PLASMODIUM STRAINS AND SPECIES, TO TARGET A VACCINE THAT CONFERS STRAIN TRANSCENDING AND CROSS-SPECIES PROTECTION. WE WILL USE CLINICALLY RELEVANT SELECTION CRITERIA GOVERNED BY THE CAPACITY OF THE ANTIGEN TO PROTECT AGAINST VIRULENT P. YOELII PARASITE CHALLENGE IN ESTABLISHED PRECLINICAL MODELS AND TO BE RECOGNIZED BY RECALL PLASMODIUM-SPECIFIC IMMUNE RESPONSES IN PROTECTIVE HUMAN MODELS. WE WILL USE TWO INNOVATIVE VACCINE DELIVERY PLATFORMS SELECTED FOR CAPACITY TO INDUCE ROBUST AND SUSTAINED T CELL RESPONSES, WITH A SPECIFIC FOCUS ON THE INDUCTION OF LIVER-SPECIFIC RESIDENT MEMORY T CELLS (TRM) TARGETING THE PUTATIVE SITE OF IMMUNE ACTION. OUR SELECTED REGIMENS INCLUDE “PRIME-TARGET” COMPRISING A DNA PRIME FOLLOWED BY ADENOVIRUS BOOST DELIVERED INTRAVENOUSLY TO TARGET THE LIVER, AND A LIPOSOMAL MRNA VACCINE PLATFORM WITH AN INCORPORATED AGONIST (CA) TO ACTIVATE NKT CELLS AND LINK INNATE AND ADAPTIVE IMMUNITY; BOTH PLATFORMS CAN INDUCE SUSTAINED T CELL RESPONSES AND ARE CAPABLE OF PROTECTING MICE AGAINST SPOROZOITE CHALLENGE. WE WILL ASSIGN PRIORITIES FOR VACCINE DEVELOPMENT ACCORDING TO THEIR CREDENTIALS, EVALUATE COMBINATIONS FOR SYNERGY, AND DOWN-SELECT FOR CLINICAL DEVELOPMENT THE SET OF ANTIGENS THAT HAVE A MAXIMUM LIKELIHOOD OF INDUCING ROBUST AND SUSTAINED PROTECTIVE IMMUNITY AGAINST MALARIA IN HUMANS. THE OPTIMAL VACCINE CANDIDATE DEFINED WITH THIS PRECLINICAL DEVELOPMENT STRATEGY COULD BE TRANSITIONED DIRECTLY TO CLINICAL DEVELOPMENT WITH THE ULTIMATE GOAL OF DEPLOYING IN THE FIELD AN EFFECTIVE RATIONALLY-DESIGNED GENOME-BASED VACCINE THAT WOULD INDUCE DURABLE T-CELL MEDIATED PROTECTION AND AMELIORATE DISEASE IN ALL AT-RISK INDIVIDUALS.

WHAT CAUSES LOW BACK PAIN TO FLARE: HAS A MAJOR OPPORTUNITY TO UNDERSTAND BACK PAIN BEEN MISSED? - PROJECT SUMMARY/ABSTRACT LOW BACK PAIN (LBP) IS TRADITIONALLY VIEWED AS EITHER A TRANSIENT CONDITION WITH ACUTE EPISODES OR A CHRONIC PERSISTING CONDITION. HOWEVER, IT IS INCREASINGLY RECOGNIZED THAT FOR MOST (>80% OF CASES), LBP HAS A VARIABLE AND ONGOING TRAJECTORY CHARACTERIZED BY FLUCTUATING SYMPTOMS – WITH PERIODS OF WORSE SYMPTOMS REFERRED TO AS “FLARES”. CURRENT WORK REGARDING RISK FACTORS FOR LBP ONLY CONSIDERS THOSE ASSOCIATED WITH A NEW EPISODE OR THE TRANSITION TO CHRONIC LBP. FINDINGS FROM THAT WORK HAS FAILED TO DELIVER EFFECTIVE STRATEGIES TO PREVENT NEW EPISODES OR CHRONICITY, NOR REDUCE THE EXCESSIVE BURDEN OF LBP. THE UNDERLYING PREMISE OF THIS PROPOSAL IS THAT FLUCTUATION OR FLARING OF LBP IS MEDIATED BY TRANSIENT/FLUCTUATING EXPOSURE TO A RANGE OF FACTORS ACROSS MULTIPLE DOMAINS, THAT DIFFER BETWEEN INDIVIDUALS, AND IDENTIFICATION OF THESE FACTORS COULD PROVIDE A BASIS FOR A COMPLETELY NEW MODEL TO PERSONALIZE CARE. THIS PROJECT WILL TAKE A FRESH LOOK AT LBP BY DIRECTLY ADDRESSING THIS QUESTION USING INNOVATIVE METHODS THAT ARE ENABLED BY RECENT ADVANCES IN TECHNOLOGY. THE WORK IS BASED ON OUR EXTENSIVE COLLABORATION WITH PEOPLE WITH LBP THAT HAS PROVIDED CRITICAL INSIGHT INTO HOW FLARES ARE DEFINED (WHICH SURPRISINGLY IS NOT BASED ON PAIN ALONE) AND THEIR PERSPECTIVE OF THE POTENTIAL CAUSES FOR LBP FLARES. OUR MULTIDISCIPLINARY RESEARCH TEAM PROPOSE AN INNOVATIVE CASE-CROSSOVER STUDY THAT WILL ADDRESS TWO AIMS. AIM 1 WILL EXAMINE, AT MULTIPLE TIMEPOINTS OVER A PERIOD OF 3 MONTHS, AN ARRAY OF CANDIDATE BIOLOGICAL/BEHAVIORAL, PSYCHOLOGICAL, SOCIAL, AND ENVIRONMENTAL RISK FACTORS FOR LBP FLARE THAT HAVE BEEN SELECTED BASED ON CONSUMER PERSPECTIVE, LITERATURE REVIEW AND OUR EXTENSIVE PRELIMINARY STUDIES. MEASURES WILL BE MADE IN THE “REAL WORD” USING ADVANCED WEARABLE AND ONLINE TECHNOLOGIES. AIM 2 WILL INVOLVE AN EMBEDDED STUDY OF INDIVIDUALS FROM THE MAIN PARTICIPANT COHORT WHO FLARE FREQUENTLY. FOR 7 DAYS, ELIGIBLE INDIVIDUALS WILL UNDERGO MORE INTENSIVE “REAL- WORLD” PHYSIOLOGICAL AND BIOMECHANICAL MEASURES USING AN INNOVATIVE COMBINATION OF WEARABLE SENSORS AND HOME-BASED SAMPLE/DATA COLLECTION TO INVESTIGATE POTENTIAL MECHANISMS UNDERLYING TWO SURPRISING PRELIMINARY OBSERVATIONS: (1) THAT POOR SLEEP IS A POTENT RISK FACTOR FOR A SUBSEQUENT FLARE (AS OPPOSED TO THE TRADITIONAL VIEW THAT PAIN INTERFERES WITH AND CAUSES POOR SLEEP), AND (2) THAT LOW RATHER THAN HIGH PHYSICAL ACTIVITY IS A RISK FACTOR FOR LBP FLARE. WE AIMS TO IDENTIFY NOT ONLY IF, BUT HOW, DAILY VARIATIONS IN SLEEP QUALITY AND PHYSICAL ACTIVITY – TWO OF THE MOST IMPORTANT, YET MODIFIABLE HEALTH BEHAVIORS – DRIVE FLARE OF LBP. TOGETHER THESE DATA WILL FILL A MAJOR GAP BY PROVIDING NEW UNDERSTANDING OF MECHANISMS THAT DRIVE THE FLUCTUATING EXPERIENCE OF LBP. SUCH FACTORS WILL PROVIDE THE FOUNDATION TO DEVELOP AND TEST MORE TARGETED AND PERSONALIZED INTERVENTIONS TO REDUCE THE ENORMOUS – AND WORSENING – BURDEN OF THIS CONDITION ON SUFFERERS AND SOCIETY.

WE NOW UNITE THE FOUR ORIGINAL EDGI1 SITES WITH EXCEPTIONALLY PRODUCTIVE GLOBAL NEW SITES TO ADVANCE GENOMIC DISCOVERY ACROSS ALL MAJOR EATING DISORDERS (EDS) TO IDENTIFY BIOLOGICALLY, CLINICALLY, CULTURALLY, AND THERAPEUTICALLY MEANINGFUL AND ACTIONABLE INSIGHTS. AIM 1: EDGI2 EXTENDS OUR CORE BUSINESS BY INCREASING SAMPLE SIZE, DIVERSITY, AND ED PHENOTYPES. EDGI2 EMPHASIZES CO-PRODUCTION—WITH REPRESENTATIVES FROM MINORITIZED AND MARGINALIZED COMMUNITIES, WE WILL ASCERTAIN 30% OF SAMPLES FROM UNDERREPRESENTED GROUPS. WE WILL ALSO OVER- SAMPLE INDIVIDUALS WITH SEVERE AND ENDURING AN (SE-AN), WHOSE DNA MAY BE ENRICHED FOR CAUSATIVE ALLELES. AIM 2: WE WILL APPLY STATISTICAL GENETIC ANALYSES TO EXPLICATE HETEROGENEITY AND BIOLOGY OF EDS BY: CONDUCTING STANDARD GWAS ANALYSES ON DIAGNOSES, TRANS-DIAGNOSTIC BEHAVIORS, AND CONTINUOUS PHENOTYPES INCLUDING POLYGENIC RISK SCORE (PRS), AND RARE VARIANT CNV ANALYSES; IDENTIFYING CLINICALLY MEANINGFULLY PATIENT SUBSETS; AND INTENSIVELY EVALUATING OUR PROPOSAL THAT AN IS A METABO-PSYCHIATRIC DISORDERS USING LDSC, PRSET, PHEWAS, AND MENDELIAN RANDOMIZATION TO CLARIFY DIRECTION OF CAUSATION. AIM 4: TO DETERMINE WHERE IN THE BODY EDS “LIVE”, WE WILL IDENTIFY BRAIN CELL TYPES AND ANATOMICAL REGIONS IMPLICATED BY GENOMIC STUDIES OF EDS; PREDICT GENETICALLY REGULATED GENE EXPRESSION (GREX) IN BRAIN, GUT, ADIPOSE, AND OTHER ED-RELEVANT TISSUES; USE SNRNASEQ ATLASES TO SHARPEN PRELIMINARY GTEX AND TWAS ANALYSES TO IDENTIFY BRAIN CELL TYPES STRONGLY IMPLICATED BY THE GENOMICS OF EACH ED; EXPAND TO RELEVANT NON-BRAIN CELL TYPES (E.G., ADIPOSE, MUSCLE, LIVER, SALIVARY); AND USE DYNAMIC GREX TO MODEL GENE EXPRESSION IN ED-RELEVANT CONTEXTS (E.G., SEX, BMI, STRESS) ENABLING PRECISE AND PERSONALIZED MODELLING OF GENE EXPRESSION. AIM 5: EDGI2 WILL CULMINATE IN A TRANSLATIONAL SUMMIT UNITING FORWARD-THINKING STAKEHOLDERS FROM MULTIPLE SECTORS TO DEVELOP A TRANSLATIONAL ROADMAP FOR EVIDENCE-BASED ED PREVENTION AND TREATMENT. EDGI2 WILL YIELD CRITICAL KNOWLEDGE ABOUT GENETIC AND ENVIRONMENTAL RISK FOR EDS, REVEAL MECHANISMS THAT POTENTIATE OR PROTECT AGAINST GENETIC RISK, AND TRANSITION ED GENETICS FROM DISCOVERY TO CLINICAL TRANSLATION.

THE QUEENS MUSEUM OF ART WILL CARRY OUT A CONSERVATION PROJECT FOR THE PANORAMA OF THE CITY OF NEW YORK, A LARGE-SCALE MODEL OF NEW YORK CITY THAT WAS BUILT FOR THE 1964-1965 NEW YORK WORLD’S FAIR. THE MODEL WAS CREATED BY MORE THAN 100 ARTISANS, COVERS AN AREA OF 9,335 SQUARE FEET, AND CONTAINS 895,000 BUILDINGS. PROJECT ACTIVITIES WILL INCLUDE AN OVERALL ASSESSMENT OF THE MODEL’S 272 PANELS AS WELL AS REPAINTING AND REINFORCEMENT OF THE STRUCTURAL SUPPORT. CONSERVATORS WILL ALSO REPAIR THE LIGHTING SYSTEM WITHIN THE MODEL, WHICH CODES FOR DIFFERENT BUILDING FUNCTIONS. WHEN RESTORED, THE MODEL WILL BE A RESOURCE FOR HISTORIANS AND RESEARCHERS OF NEW YORK CITY HISTORY.

SINGLE MOLECULE BIOMOLECULAR CONDENSATE ANALYSIS IN NEURONS - PROJECT SUMMARY/ABSTRACT BIOMOLECULAR CONDENSATES (BMCS) DEFINE SMALL MEMBRANELESS CELLULAR COMPARTMENTS THAT SEGREGATE SPECIFIC PROTEINS AND NUCLEIC ACIDS TO GENERATE CELLULAR FUNCTIONS. FLUORESCENCE RECOVERY AFTER PHOTOBLEACHING (FRAP) IS CURRENTLY THE METHOD OF CHOICE TO CHARACTERIZE BMCS IN VITRO AND IN VIVO AND DERIVE THE AVERAGE DIFFUSION AND TRAPPING OF MOLECULES TRAPPED IN THESE CONDENSATES. ALTHOUGH FRAP IS A GOOD WAY TO RETRIEVE THESE IMPORTANT METRICS, IT GREATLY LIMITS OUR UNDERSTANDING OF BMCS NANOSCALE ORGANIZATION AND DYNAMICS IN LIVE CELLS AND PRECLUDES THE ANALYSIS OF NANOSCALE BMCS (BELOW THE DIFFRACTION OF LIGHT ~200 NM). SINGLE-MOLECULE IMAGING SUPER-RESOLUTION MICROSCOPY ALLOWS DIRECT IMAGING OF MOLECULES TRAPPED IN BMCS WITH MUCH GREATER SPATIOTEMPORAL RESOLUTION BOTH IN VITRO AND IN LIVE NEURONS, AND HAS THE POTENTIAL TO REVEAL NANOSCALE BMCS AND THEIR EVOLUTION IN THE CONTEXT OF AGEING AND ALZHEIMER’S DISEASE (AD). HOWEVER, THERE ARE CURRENTLY NO BIOINFORMATIC TOOLS TO SPECIFICALLY ANALYZE BMCS AT THE SINGLE-MOLECULE LEVEL. WE HAVE DEVELOPED A NOVEL TOOL, NAMED NANOSCALE SPATIOTEMPORAL INDEXING CLUSTERING (NASTIC), (WALLIS ET AL., BIORXIV, 2021, REF. 7), WHICH OFFERS UNPRECEDENTED INSIGHTS INTO THE DYNAMICS OF PROTEINS UNDERGOING LIQUID-LIQUID PHASE SEPARATION/TRANSITION IN LARGE AND NANOSCALE BMCS, IN LIVE NEURONS. WE IDENTIFIED TAU AND SYNUCLEIN AS CANDIDATES FOR THEIR ABILITY TO GENERATE SYNAPTIC BMCS IN HIPPOCAMPAL NEURONS. NASTIC ANALYSIS REVEALS THAT TAU MOLECULES CAN INDEED FORM SMALL NANOCLUSTERS IN LIVE HIPPOCAMPAL NEURONS IN WHICH THEY EXHIBIT VERY LOW MOBILITY AND SENSITIVITY TO 1,6-HEXANEDIOL, AN ALIPHATIC ALCOHOL USED TO INHIBIT WEAK MOLECULAR INTERACTIONS THAT MEDIATE BMCS. NASTIC WILL BE AT THE CORE OF OUR ABILITY TO MAKE A SIGNIFICANT CONTRIBUTION TO THE UNDERSTANDING OF BMCS IN NEURONS BECAUSE IT OFFERS UNPRECEDENTED OPPORTUNITIES TO EXAMINE THE SPATIOTEMPORAL BEHAVIOUR OF MOLECULES IN THESE CONDENSATES. NASTIC WILL BE DEVELOPED FURTHER TO ENCOMPASS TWO-COLOR SINGLE-MOLECULE ANALYSIS. THIS WILL BE INSTRUMENTAL TO ASSESS THE SPATIOTEMPORAL RELATIONSHIP OF THESE NANOSCALE BMCS WITH THEIR CELLULAR ENVIRONMENT IN 2 AND 3D. WE WILL VALIDATE OUR TWO-COLOR NASTIC IMPLEMENTATION WITH THE PAIRWISE DUAL IMAGING OF TAU WILDTYPE AND AD MUTANT P301L, AND A-SYNUCLEIN. THIS WILL ALLOW US TO DECIPHER THE CO-CLUSTERING DYNAMICS AND EXAMINE POTENTIAL HIERARCHICAL DEPENDENCY OF ONE NANOBMC UPON THE OTHER ALLOWING REFINED UNDERSTANDING OF THE GENERATION OF NANOSCALE BMCS IN SYNAPSES. WE ANTICIPATE THAT THE P301L MUTATION WILL LARGELY INCREASE THE SIZE AND LIFETIME OF THE TAU NANOBMCS AND ALTER THEIR RELATIONSHIP WITH OTHER SYNAPTIC MOLECULES. THE OUTCOME OF THIS GRANT WILL BE THE DEVELOPMENT AND VALIDATION OF AN ANALYTICAL PIPELINE THAT WILL ENABLE EXPLORATION OF THE SPATIOTEMPORAL RELATIONSHIP OF TAU AND A-SYNUCLEIN NANOBMCS IN LIVE HIPPOCAMPAL NEURONS. OUR PROJECT WILL THEREFORE GENERATE DATA AND A MUCH-NEEDED TECHNOLOGY OPENING NEW AVENUES FOR OUR UNDERSTANDING OF BMCS IN NEURONAL FUNCTION AND AD.
