THE RESILIENT EXTRATERRESTRIAL HABITATS RESEARCH INSTITUTE (RETHI) WILL HARNESS PROMISING NEXT-GENERATION TECHNOLOGICAL ADVANCES TO OVERCOME THE GRAND CHALLENGE OF DEEP SPACE HABITATION. BY LEVERAGING WORLD-LEADING EXPERTISE IN DEVELOPING CIVIL INFRASTRUCTURE RESPONSIVE TO CATASTROPHIC NATURAL HAZARDS AND MERGING IT WITH LEADERS IN THE FIELDS OF MODULAR AND AUTONOMOUS ROBOTICS PLATFORMS SMART BUILDINGS HYBRID SIMULATION COMPLEX SYSTEMS AND DIAGNOSTICS AND PROGNOSTICS WE WILL DEVELOP AND DEMONSTRATE THE TRANSFORMATIVE TECHNOLOGIES NEEDED TO DESIGN AND OPERATE RESILIENT DEEP SPACE HABITATS THAT CAN ADAPT ABSORB AND RAPIDLY RECOVER FROM EXPECTED AND UNEXPECTED DISRUPTIONS TO DEEP SPACE HABITAT SYSTEMS WITHOUT FUNDAMENTAL CHANGES IN FUNCTION OR SACRIFICES IN SAFETY. TO ACHIEVE THIS VISION THE RETHI TEAM WILL PURSUE THE FOLLOWING TIGHTLY-INTEGRATED RESEARCH OBJECTIVES: 1) ESTABLISH A COMPREHENSIVE RESILIENCE FRAMEWORK TO SUPPORT RESILIENT DESIGN OPERATION AND MANAGEMENT WHILE ANTICIPATING A HABITAT THAT EVOLVES AND GROWS OVER TIME; 2) CONDUCT THE RESEARCH AND DEVELOPMENT NECESSARY TO ESTABLISH SMART HABITATS THAT AUTONOMOUSLY SENSE ANTICIPATE AND RESPOND UNDER A VARIETY OF MANNED AND UNMANNED CONFIGURATIONS; 3) DEVELOP DECISION-MAKING TECHNIQUES THAT CAN WEIGH ALTERNATIVES FOR COMPLEX INTERCONNECTED INTERDEPENDENT HABITAT SYSTEMS WHILE ALSO UNDERSTANDING AND WHEN NECESSARY COMBATING THE NATURAL HUMAN INSTINCT TO INTERFERE OR OVERRIDE AUTOMATED SYSTEMS; AND 4) CONTRIBUTE TO THE NASA MISSION TO EDUCATE THE NEXT GENERATION OF ENGINEERS AND SCIENTISTS WHILE BUILDING PARTNERSHIPS WITH US INDUSTRIES AND ORGANIZATIONS AND OTHER NATIONS AND GENERATE DATA PRODUCTS THAT WILL INFORM AND GUIDE FUTURE RESEARCH AND DEVELOPMENT. TO ACCOMPLISH THESE GOALS WE WILL EMPLOY THE LATEST APPROACHES IN RESILIENT DESIGN AUTOMATED AND COORDINATED ROBOTICS MACHINE LEARNING INTELLIGENT SYSTEM MONITORING AND ADAPTIVE CONTROL. THESE PARALLEL BUT INTERCONNECTED EFFORTS WILL CONVERGE IN THE ESTABLISHMENT OF A CYBER-PHYSICAL TESTBED THAT INTEGRATES PHYSICAL MODELS WITH VIRTUAL MODELS TO SYSTEMATICALLY DEVELOP DEPLOY AND VALIDATE PRINCIPLES OF RESILIENCE AND AUTONOMOUS DETECTION AND CORRECTIVE CAPABILITIES. RETHI S HEADQUARTERS WILL BE AT PURDUE UNIVERSITY S DISCOVERY PARK. PARTICIPANT UNIVERSITIES WILL INCLUDE THE UNIVERSITY OF CONNECTICUT HARVARD UNIVERSITY AND THE UNIVERSITY OF TEXAS SAN ANTONIO. THROUGH THIS INTEGRATED EFFORT RETHI WILL FACILITATE DEEP SPACE HABITATS WITH EMBEDDED AUTONOMOUS CAPABILITIES THAT HAVE THE ABILITY TO OPERATE IN BOTH MANNED AND UNMANNED CONFIGURATIONS RAPIDLY AND SAFELY TRANSITIONING BETWEEN THOSE STATES WHEN NEEDED. RATHER THAN EVALUATING SUBSYSTEMS OR COMPONENTS INDEPENDENTLY RETHI WILL ESTABLISH A NEW APPROACH SYSTEM RESILIENCE THAT WILL BE THE TURNING POINT IN ACHIEVING PERMANENT DEEP SPACE HABITATS FOR LONG TERM HUMAN SETTLEMENT THAT ARE MORE COST-EFFECTIVE AND MORE RESPONSIVE TO UNFORESEEN EVENTS. THE INSTITUTE WILL SERVE AS A HUB FOR WORLD-CLASS RESEARCH PRIVATE AND PUBLIC PARTNERSHIPS AND PROJECT-BASED LEARNING BY DISTRIBUTED TEAMS OF STUDENTS ALL FOCUSED ON THE RESILIENCE OF COMPLEX SYSTEMS THAT ARE BUILT FROM AND DEPEND UPON THE SYNERGY OF COMPUTATIONAL AND PHYSICAL COMPONENTS.

AQUAPONICS INTENSIVELY PRODUCES A DIVERSE ARRAY OF NUTRITIOUS BLUE FOODS AND SPECIALTY CROPS, BUT THIS EMERGING FOOD PRODUCTION SYSTEM FACES INTERTWINED TECHNICAL, ECONOMIC, AND ENVIRONMENTAL CHALLENGES THAT ARE THE MAIN BARRIERS TO ITS WIDE ADOPTION. THE OVERARCHING HYPOTHESES OF THIS PURDUE UNIVERSITY-LED PROJECT ARE: 1) INTEGRATING AQUAPONICS WITH MICROALGAE CULTIVATION, ANAEROBIC DIGESTION AND BIOREFINING CAN FULLY UTILIZE NUTRIENTS AND PRODUCE ENERGY AND HIGH-VALUE BIOPRODUCTS; AND, 2) INTEGRATING RESEARCH WITH EXTENSION AND EDUCATION WILL INCREASE ADOPTION OF AQUAPONICS AND CONSUMER WILLINGNESS TO PURCHASE BLUE FOODS. SPECIFIC PROJECT OBJECTIVES INCLUDE: 1) IDENTIFYING BARRIERS AND OPPORTUNITIES FOR BLUE FOOD AND AQUAPONICS; 2) BUILDING A NOVEL, INTEGRATED, AQUAPONICS FOOD PRODUCTION SYSTEM; 3) DEVELOPING MULTI-DIMENSIONAL SUSTAINABILITY METRICS FOR SYSTEM ASSESSMENT AND MANAGEMENT; 4) SUPPORT AQUAPONICS AND BLUE FOOD INDUSTRY AND MARKET DEVELOPMENT; AND, 5) CREATING BLUE FOOD-RELATED EDUCATION MATERIALS. THIS PROJECT IS EXPECTED TO BUILD A ZERO-WASTE, GRID-INDEPENDENT, AND ECONOMICALLY VIABLE FOOD PRODUCTION SYSTEM, AND GENERATE AN EVIDENCE-BASED VISION AND FRAMEWORK FOR FACILITATING THE ADOPTION OF AQUAPONICS. MORE DETAILED AND ACCURATE INFORMATION IS NEEDED TO ALLOW LANDOWNERS, MANAGERS AND POLICYMAKERS TO MAKE ECONOMICAL AND DATA-DRIVEN DECISIONS THAT WILL AFFECT THE DELIVERY, SUSTAINABILITY AND RESILIENCE OF FOREST RESOURCES. THE GOAL OF THE PROMOTING ECONOMIC RESILIENCE AND SUSTAINABILITY OF THE EASTERN U.S. FORESTS (PERSEUS) PROJECT IS TO PROVIDE SOUND SCIENCE, OUTREACH AND EDUCATIONAL OPPORTUNITIES THAT LAY THE FOUNDATION FOR A MAJOR FUNDAMENTAL CHANGE IN FORESTRY. MORE SPECIFICALLY, PERSEUS AIMS TO (1) CREATE A SET OF DIGITAL TOOLS AND WALL-TO-WALL FOREST MAPPING PRODUCTS THAT WILL PROVIDE DETAILED AND NEAR REAL-TIME MEASUREMENTS, (2) BUILD AND APPLY A FRAMEWORK TO VISUALLY REPRESENT CURRENT AND FUTURE FOREST TRENDS, (3) ENGAGE WITH STAKEHOLDERS TO DEVELOP DATA-DRIVEN MANAGEMENT PRACTICES, AND (4) ESTABLISH EDUCATIONAL AND TRAINING PROGRAMS THAT SUPPORT STUDENTS AND PROFESSIONALS IN UNDERSTANDING AND MAKING USE OF DIGITAL TECHNOLOGIES. PERSEUS WILL PROVIDE A FOUNDATION FOR REDEFINING FOREST INVENTORY IN THE UNITED STATES WHILE ALSO PROVIDING THE MUCH-NEEDED ABILITY TO PREDICT FUTURE FOREST CONDITIONS. WORKING WITH STAKEHOLDERS, PERSEUS WILL PROVIDE (1) USER-FRIENDLY TOOLS AND PROGRAMS TO BETTER MEASURE AND MONITOR FORESTS LIKE NEVER BEFORE, (2) SUSTAINABLE FOREST MANAGEMENT PRACTICES, AND (3) A PLATFORM TO OPTIMIZE REGIONAL EFFORTS IN BUILDING ENVIRONMENTALLY AND ECONOMICALLY SUSTAINABLE FORESTS, ESPECIALLY ON PRIVATE LANDS IN THE RURAL UNITED STATES. MOREOVER, THE PROJECT WILL TRAIN COHORTS OF STUDENTS AND PROFESSIONALS TO BUILD A DIGITALLY COMPETENT WORKFORCE. SUCCESSFUL COMPLETION OF THE PROJECT WILL INCREASE PHYSICAL GOODS SUCH AS FOOD, WATER, FUEL AND TIMBER PROVIDED BY FOREST ECOSYSTEMS, AND IMPROVE OPERATIONAL EFFICIENCY IN FORESTS ACROSS THE EASTERN UNITED STATES. IN PARTICULAR, MEASUREMENT TOOLS DEVELOPED BY PERSEUS WILL MODERNIZE AND INFORM FOREST MANAGEMENT BY CREATING DIGITAL INVENTORY TOOLS THAT EMPOWER STAKEHOLDERS TO BETTER UNDERSTAND THE POTENTIAL IMPACTS OF THEIR DECISIONS ON A LOCAL AND REGIONAL SCALE. THE RESULTING FRAMEWORK WILL PROVIDE A DIGITAL BRIDGE, ENABLING LANDOWNERS AND STAKEHOLDERS WITH PRACTICAL TACTICS AND FOREST SIMULATION TO INFORM DECISION- AND POLICY-MAKING. TWO STRATEGIC PRIORITIES, DIVERSITY, EQUITY AND INCLUSION (DEI), AND COMMERCIALIZATION, GUIDE EXTENSION ACTIVITIES. EXTENSION WILL CULTIVATE STAKEHOLDER ENGAGEMENT TO PROMOTE A CULTURE OF DEI; FOSTER A CLIMATE OF BELONGING WHERE INDIVIDUALS FULLY PARTICIPATE, CONTRIBUTE, AND THRIVE; AND ENHANCE INNOVATIONS, COLLABORATIONS, AND SYNERGIES ACROSS ALL STAKEHOLDERS. SEVEN NEW CRITICAL ISSUES WERE IDENTIFIED TO SET PRIORITIES FOR EXTENSION PROGRAMS ACROSS INDIANA. IN A STATEWIDE SURVEY, OVER 4,000 STAKEHOLDERS SHARED INPUT ON THE IMPORTANCE OF THE CRITICAL ISSUES FOR THEIR COMMUNITIES. THIS PROJECT ADDRESSES TWO CRITICAL PUBLIC-HEALTH NEEDS REGARDING LANGUAGE DISORDERS: (1) CHARACTERIZING EARLY LEXICO-SEMANTIC SKILL TRAJECTORIES WITH THE GOAL OF (2) IMPROVING EARLY IDENTIFICATION. WE FOCUS ON SEVERAL PROMISING ASSESSMENTS OF LEXICO-SEMANTIC ABILITIES IN TODDLERS, AS LEXICO-SEMANTIC SKILLS ARE A COMPONENT OF LANGUAGE WHICH IS AFFECTED IN SCHOOL-AGE CHILDREN WITH DEVELOPMENTAL LANGUAGE DISORDERS, AND CAN ALSO BE RELIABLY MEASURED IN TODDLERS. WE PROPOSE TO RECRUIT A LARGE GROUP OF 18-MONTH-OLD TODDLERS WITH A DIVERSE SET OF LANGUAGE ABILITIES AND BACKGROUNDS AND THEN FOLLOW THESE CHILDREN LONGITUDINALLY TO ASSESS WHETHER AND HOW GROWTH IN THREE MAIN LEXICO-SEMANTIC SKILLS (STRUCTURE, LEARNING, AND PROCESSING) PREDICTS LANGUAGE OUTCOMES AT AGE 4, WHEN CLINICAL LANGUAGE DISORDER STATUS CAN INITIALLY BE ASSESSED. TO FULLY CHARACTERIZE HOW EARLY SKILLS AND TRAJECTORIES LEAD TO LATER OUTCOMES, WE CARRY OUT SEVERAL ANALYSES ON CONCURRENT AND PREDICTIVE RELATIONS BETWEEN SKILLS AND CREATE DETAILED MODELS OF CHILDREN'S EARLY LEXICONS. WITH THE BASIS OF THIS PROGRAM SUPPORTING THE DISCOVERY AND DEVELOPMENT OF MEDICATIONS TO PREVENT AND TREAT OPIOID USE DISORDERS AND OVERDOSE, RAPID ADVANCEMENT TOWARDS A VIABLE PRODUCT FOR NEW DOSE REGIMENS AND EASE OF ADMINISTRATION FOR INCREASED ADHERENCE SHOULD BE ONE OF THE FIRST, SCIENTIFICALLY SOUND, AND ROBUST CHOICES MOVING FORWARD. PLGA-BASED DRUG DELIVERY SYSTEMS HAVE BEEN USED SUCCESSFULLY IN A NUMBER OF SMALL MOLECULE PRODUCTS AND ARE THE MOST WIDELY UTILIZED AND STUDIED BIOCOMPATIBLE POLYMER SYSTEMS IN CONTROLLED RELEASE. THEREFORE, THE REGULATORY AND DEVELOPMENT HURDLES WITH THE FDA WILL BE `LOWER' THAN WITH OTHER NOVEL EXCIPIENTS OR TECHNOLOGIES. THE GOAL OF THIS RESEARCH AND PRODUCT DEVELOPMENT PLAN IS TO SUBMIT A PHASE I APPLICATION FOR A 2-MONTH NTX FORMULATION WITH FAVORABLE RELEASE KINETICS AND A PATIENT-FRIENDLY FORMAT. OUR PRELIMINARY DATA INDICATE TWO TYPES OF CURRENT, LABORATORY BASED SYSTEMS CAN PROVIDE BOTH A HIGH DRUG LOADING AND CONTROLLABLE RELEASE KINETICS RESULTING OF NTX FOR AT LEAST 2 MONTHS. THE SPECIFIC AIM OF THIS PROJECT IS TO OPTIMIZE AND BRIDGE OUR LABORATORY SCALE 2-MONTH INJECTABLE NTX DELIVERY FORMULATION TO PHASE 1 CLINICAL TRIALS USING 380 MG OF NTX WITH A MICROPARTICLE SIZE OF LESS THAN 100 ΜM FOR A LESS PAINFUL INJECTION THE SUB-AIMS FOR THE UG3 PHASE ARE: (I) ESTABLISH THE DESIGN SPACE FOR THE TWO 2-MONTH NTX (2M- NTX) FORMULATIONS: EARLY RELEASE (ER) AND DELAYED RELEASE (DR); (II) IN VIVO PHARMACOKINETIC EVALUATIONS OF 2M- NTX-ER AND 2M-NTX-DR FORMULATIONS; AND FOR THE UH3 PHASE (III) GMP MANUFACTURING SCALE-UP OF THE LEAD CANDIDATE FORMULATION; (IV) LEAD CANDIDATE FORMULATION NONCLINICAL CHARACTERIZATION; AND (V) 505B2 IND SUBMISSION FOR A PHASE I CLINICAL TRIAL. THE INNOVATION IN THIS TECHNOLOGY IS THE ABILITY TO CONTROL THE NTX RELEASE KINETICS WHILE ELIMINATING THE INITIAL BURST; BASED ON OUR MECHANISTIC UNDERSTANDING OF THE PLGA MICROPARTICLE FORMATION PROCESS, USING PLGAS WITH SPECIFIC MOLECULAR PROPERTIES, AND PROVIDING TIGHT CONTROL OVER THE MANUFACTURING CONDITIONS. THIS INNOVATION HAS ALLOWED US TO DESIGN TWO SPECIFIC TYPES OF FORMULATIONS TO AID IN COMBATING THE OPIOID EPIDEMIC: (1) ER PROVIDING NEAR ZERO-ORDER RELEASE KINETICS FOR TWO MONTHS AND (2) DR PROVIDING AN INITIAL LAG PHASE OF 7-10 DAYS, WHERE MINIMAL NTX RELEASE OCCURS, SO IT CAN BE ADMINISTERED TO PATIENTS WHO ARE STILL UNDER THE INFLUENCE OF OPIOIDS WITHOUT PRECIPITATING WITHDRAWAL SYMPTOMS. PLGA-BASED MICROPARTICLE FORMULATIONS HAVE PREVIOUSLY BEEN SCALED AND HAVE BEEN SHOWN TO BE SAFE BASED ON THE APPROXIMATE 20 FDA APPROVED PRODUCTS CURRENTLY ON THE MARKET. THE SIGNIFICANCE OF THIS RESEARCH AND PRODUCT DEVELOPMENT IS THE FINAL OUTCOME OF THIS PROJECT WILL ULTIMATELY PROVIDE A NEW, READILY VIABLE, ESSENTIAL TOOL TO HELP PATIENTS OVERCOME OPIOID DEPENDENCE.

SPECIFICALLY, WE SEEK TO OPTIMIZE MICROELECTRODE ARRAYS (MEAS) AND ULTRA-MICROELECTRODE ARRAYS (UMEAS) FOR LARGE-SCALE CIRCUIT MANIPULATION THAT WILL CONTROL NEURAL ACTIVITY AT CELLULAR RESOLUTION WITH HIGH TEMPORAL RESOLUTION. OUR GOALS ARE TO 1) ADVANCE CNS MEA AND UMEA ELECTRICAL MICROSTIMULATION BY TESTING THE SEPARATE HYPOTHESES THAT MEAS AND UMEAS CAN DELIVER SAFE, EFFECTIVE LEVELS OF CORTICAL ELECTRICAL STIMULATION AND 2) ADVANCE RESEARCH BY GENERATING TRANSFORMATIVE TOOLS AND TECHNOLOGIES THAT WILL BE WIDELY USED THROUGHOUT THE RESEARCH COMMUNITY. HERE WE PROPOSE COMBINING COMPUTATIONAL MODELING, ENGINEERING OPTIMIZATION, AND IN VIVO MEASUREMENT TO ADDRESS THESE CHALLENGES AND PRODUCE ADVANCES IN MICROSTIMULATION AND TOOLS FOR THE COMMUNITY. HIGH THROUGHPUT INFRASTRUCTURE FOR REACTION SCREENING AND BIOASSAYS - HIGH THROUGHPUT INFRASTRUCTURE FOR REACTION SCREENING AND BIOASSAYS MASS SPECTROMETRY (MS) IS A POWERFUL AND WIDELY APPLICABLE ANALYTICAL METHOD FOR QUALITATIVE AND QUANTITATIVE ANALYSIS OF COMPOUNDS OF ALL TYPES AND SIZES. DESORPTION ELECTROSPRAY IONIZATION (DESI) IS AN AMBIENT IONIZATION METHOD IN WHICH SAMPLES ARE ANALYZED IN THE OPEN AIR BY IMPACT OF PRIMARY DROPLETS. GIVEN THE ABILITY TO POSITION AN ARRAY OF SAMPLES RELATIVE TO THE MASS SPECTROMETER, DESI-MS BECOMES A HIGH THROUGHPUT (HT) CHEMICAL ANALYSIS METHOD. THE POWER OF MS AS AN ANALYTICAL TECHNIQUE IS WELL KNOWN BUT IT IS LESS COMMONLY REALIZED THAT MS CAN ALSO SERVE AS A PREPARATIVE METHOD, E.G. IT CAN BE USED TO DEPOSIT MASS-SELECTED IONS ON SURFACES TO CREATE NEW MATERIALS IN VACUO. OF SIGNIFICANT INTEREST TO ORGANIC SYNTHESIS, A UNIQUE FEATURE OF DESI IS THAT, UPON IMPACT, THE SPRAY OF SOLVENT USED TO ANALYZE A REACTION MIXTURE GENERATES SECONDARY MICRODROPLETS IN WHICH REACTIONS MAY BE ACCELERATED EN ROUTE TO THE MASS SPECTROMETER. IT IS THIS REMARKABLE FEATURE THAT MAKES DESI- MS A POWERFUL SYNTHETIC METHOD COMBINED WITH A BUILT-IN ANALYTICAL CAPABILITY. WITH SUPPORT OF DARPA, WE BUILT A HIGH THROUGHPUT SYSTEM AT PURDUE CAPABLE OF AUTOMATED REACTION SCREENING AT A RATE GREATER THAN 1 REACTION MIXTURE PER SECOND. WE NOW PROPOSE AN INTRAMURAL - EXTRAMURAL COLLABORATION BETWEEN PURDUE AND THE NCATS ASPIRE LABORATORY. THE UG3 COMPONENT OF THE COLLABORATION WILL FOCUS ON DESIGNING, FABRICATING, AND TESTING AN IMPROVED HIGH THROUGHPUT SYSTEM FOR REACTION SCREENING BASED ON DESI-MS. THE SYSTEM WILL REPLICATE THE CAPABILITIES OF THE EXISTING PURDUE SYSTEM AND ALSO INCLUDE NEW CAPABILITIES FOR SMALL-SCALE SYNTHESIS COMBINED WITH HIGH THROUGHPUT BIOASSAYS. IN THE UH3 PHASE OF THE PROPOSED STUDY, THE SYSTEM WILL BE TRANSFERRED TO NCATS AND USED IN COLLABORATION WITH THE INTRAMURAL GROUP. AS AN INITIAL DEMONSTRATION OF THE NEW HIGH THROUGHPUT PLATFORM CAPABILITIES, WE WILL PURSUE THE DISCOVERY OF NOVEL THERAPEUTICS FOR ADVANCED-STAGE PROSTATE CANCER, FOR WHICH CURRENT CHEMOTHERAPEUTIC AGENTS SHOW LIMITED EFFECTIVENESS. SPECIFICALLY, THIS EFFORT WILL ENTAIL LARGE-SCALE SCREENING AND SYNTHESIS OF POTENTIAL CHOLESTEROL SULFOTRANSFERASE (SULT2B1B, A CURRENTLY UNDRUGGED BIOLOGICAL TARGET) INHIBITORY COMPOUNDS, TOGETHER WITH LATE-STAGE FUNCTIONALIZATION OF BIOACTIVE SCAFFOLDS TO GENERATE A DIVERSE RANGE OF ANALOGS. THROUGH THIS EFFORT, THE SYSTEM WILL BE ESTABLISHED AS AN ALL-IN-ONE NEXT-GENERATION DRUG DISCOVERY PLATFORM, WITH INTEGRATED SCREENING, SYNTHESIS, AND BIOLOGICAL ASSAY CAPABILITIES. DURING THE LATTER STAGES OF THE UH3 PHASE, THE VERSATILITY OF THE SYSTEM WILL BE TESTED IN SEVERAL OTHER BIOLOGICAL APPLICATIONS, INCLUDING DIRECTED EVOLUTION AND FUNCTIONALIZATION OF ACETYLCHOLINESTERASE REACTIVATORS. SUCCESSFUL COMPLETION OF THESE TASKS WILL DEMONSTRATE THE NEWLY CONSTRUCTED HIGH THROUGHPUT DESI-MS PLATFORM TO BE AN EFFICIENT METHOD FOR THE DISCOVERY AND EXPANSION OF CHEMICAL SPACE TOWARDS CURRENTLY UNDRUGGED BIOLOGICAL TARGETS.

MODELING FUNCTIONAL GENOMICS OF SUSCEPTIBILITY TO THE PERSISTENT EFFECTS OF ENVIRONMENTAL TOXINS IN AN ELDERLY RURAL INDIANA NEURODEGENERATIVE COHORT - ALZHEIMER'S DISEASE (AD), AD-RELATED DEMENTIAS (ADRD) AND OTHER NEURODEGENERATIVE DISEASES SUCH AS PARKINSON’S DISEASE (PD) EXHIBIT PATHOGENIC GENE-ENVIRONMENT INTERACTIONS (GXE) WITH SYNERGISTIC EFFECTS OF EXPOSURE TO AN ENVIRONMENTAL CHEMICAL/PATHOGEN AND GENOTYPE. RECENT PROGRESS IN NEXT- GENERATION SEQUENCING HAS EXPEDITED THE DISCOVERY OF GENETIC RISK FACTORS ASSOCIATED WITH ALZHEIMER'S DISEASE, AD-RELATED DEMENTIAS AND PARKINSON’S DISEASE. YET, IDENTIFIED GENETIC FACTORS ONLY ACCOUNT FOR A FRACTION OF PATIENTS, AND NOT ALL PATIENTS WITH THE IDENTIFIED GENETIC RISKS DEVELOP DISEASE. THIS IS EXASPERATED FOR AD/ADRD/PD AS IT IS CONVOLUTED WITH MANY COVARIATES OVER A PERSON’S LIFETIME. TO ELUCIDATE THE CONTRIBUTION OF GXE TO DISEASE WE PROPOSE AN APPROACH BASED ON THE PRINCIPLES OF LATENT-PERSISTENT EFFECTS OF ENVIRONMENTAL NEUROTOXICANTS; AND THE DEVELOPMENTAL ORIGINS OF ADULT DISEASE HYPOTHESIS. WE SEEK TO TEST THE HYPOTHESIS THAT PERSISTENT NEUROTOXICITY IS DUE TO EXPOSURES ALTERING SELF-PERPETUATING HOMEOSTATIC PROCESSES THAT GIVE THE RESILIENCY AND PERPETUITY TO THE ADVERSE TOXICOLOGICAL PROCESSES BY EITHER GENETIC AND/OR EPIGENETIC MEANS. SPECIFICALLY, WE WILL COLLECT SAMPLES FROM SAME-SEX SIBLINGS IN NORTHERN INDIANA THAT PHENOTYPICALLY DIFFER IN AD/ADRD/PD RELEVANT COGNITIVE AND MOTOR DYSFUNCTION PHENOTYPES; REPROGRAM BLOOD CELLS INTO HUMAN INDUCED PLURIPOTENT STEM CELLS (HIPSCS); DIFFERENTIATE THEM INTO CORTICAL AND MIDBRAIN LINEAGES; AND CHARACTERIZE THEIR VULNERABILITY TO INDUCTION OF A PERSISTENT NEUROTOXIC STATE CAUSED BY NEURODEVELOPMENTAL EXPOSURES TO ENVIRONMENTAL HAZARD/TOXICANTS RELEVANT TO THIS REGION AND HISTORY. WE EXPECT TO IDENTIFY A PERSISTENT NEUROTOXIC STATE WITH A CONSERVED RESPONSE SHARED ACROSS INDIVIDUALS AT THE LEVEL OF THE GENETIC/EPIGENETIC PATHWAYS EVOKED. 'AGE-RELATED SIGNALING' NETWORKS HAVE BEEN DEFINED AS CENTRAL PATHWAYS REGULATING HEALTHY LIFESPAN AND AGING AND ARE THUS EXPECTED AS A SHARED NETWORK FEATURE CONFERRED BY GXE RISKS. OUR INNOVATIVE APPROACH WILL LEVERAGE SINGLE-CELL GENOMICS APPROACHES AND PATHWAY ANALYSIS TO IDENTIFY MECHANISMS ACTIVATED BY SUBJECT-SPECIFIC GXE TO CAUSE A PERSISTENT NEUROTOXIC STATE WITH ONLY A SMALL SUBJECT SIZE AND LOW FDR. THE FOLLOWING AIMS WILL TEST OUR HYPOTHESIS AND VALIDATE THE SHARED SIGNALING NETWORKS: AIM 1: IDENTIFY SUBJECT BY TOXICANT INTERACTIONS CONTRIBUTING TO PERSISTENT NEUROTOXICITY USING HUMAN CELLS DERIVED FROM PAIRED ALZHEIMER'S DISEASE, AD-RELATED DEMENTIAS AND PARKINSON’S DISEASE AND HEALTHY COHORT WITH COMPARABLE EXPOSURE HISTORIES; AIM 2: IDENTIFY GENETIC PATHWAYS ASSOCIATED WITH ESTABLISHING A PERSISTENT NEUROTOXIC STATE VIA SINGLE-CELL GENOMICS AND BIOINFORMATIC COMPARISONS TO POPULATION LEVEL DATA; AND AIM 3: VALIDATE THE GENETIC PATHWAYS OF GXE INDUCTION OF PERSISTENT NEUROTOXICITY WITH IN VIVO AND IN VITRO MODELS. BY LOWERING TECHNICAL BARRIERS AND MAKING DATA AI-READY, STREAMCI EMPOWERS DOMAIN EXPERTS TO BUILD INTELLIGENT AND RESPONSIVE APPLICATIONS THAT DRIVE FASTER DISCOVERIES AND MORE EFFECTIVE SOLUTIONS. THE PLATFORM ALSO SERVES AS AN OPEN EDUCATIONAL RESOURCE, TRAINING THE NEXT GENERATION OF DATA- AND AI-SAVVY RESEARCHERS TO CREATE DATA-DRIVEN SOLUTIONS FOR CRITICAL SOCIETAL NEEDS. STREAMCI IS DESIGNED TO STREAMLINE THE ENTIRE WORKFLOW FOR SENSOR DATA STREAMS, FROM CAPTURING RAW SENSOR DATA TO PROCESSING IT AT VARIOUS LEVELS OF FIDELITY, ANONYMIZATION, AND TRANSFORMATION; AND TO APPLY SUITABLE ML METHODS ACROSS DIFFERENT MODALITIES AND ENABLE FINDABLE, ACCESSIBLE, INTEROPERABLE, AND REUSABLE (FAIR) DATA SHARING FOR RESEARCH REPRODUCIBILITY AND CROSS-DOMAIN SCIENCE. THE CORE INNOVATIONS OF THE PROJECT INCLUDE HIGH-LEVEL DATA ABSTRACTIONS THAT ALLOW RESEARCHERS TO COMBINE DATA STREAMS ACROSS SOURCES; AI-READINESS THROUGH NOVEL TOOLS FOR DATA PREPARATION AND CANONICAL AND CUSTOMIZED DATA PIPELINES; LOW-CODE APPLICATION DEVELOPMENT AND INTEGRATION OF POWERFUL TOOLS; AND SCALABLE, SEAMLESS SERVICE DEPLOYMENT AND DELIVERY. STREAMCI IS USED ACROSS A WIDE RANGE OF DOMAINS, INCLUDING ENERGY, MANUFACTURING, TRANSPORTATION, AGRICULTURE, ECOLOGY, AUDIOLOGY, AND EDUCATION, DEMONSTRATING ITS BROAD APPLICABILITY AND VERSATILITY. COLLABORATIVE RESEARCH: RESEARCH INFRASTRUCTURE: CFS (TRACK III) COSMOGENIC NUCLIDES IN EARTH SCIENCE RESEARCH: ALLIED FACILITIES FOR SAMPLE PREPARATION, ANALYSIS, AND TRAINING -ACCELERATOR MASS SPECTROMETRY (AMS) IS A TECHNIQUE USED TO MEASURE EXCEEDINGLY RARE MATERIALS PRODUCED AS COSMIC RAYS INTERACT WITH ROCKS, SOIL, AND EARTH?S ATMOSPHERE. USING THESE DATA, SCIENTISTS CAN DETERMINE THE TIMING OF EVENTS THAT HAVE THE POTENTIAL TO IMPACT HUMAN ENDEAVORS. SOME CATASTROPHIC CHANGES, LIKE FLOODS OR EARTHQUAKES, OCCUR OVER SHORT TIME-SCALES, WHILE OTHERS, LANDSCAPE EROSION AND THE GROWTH AND DEMISE OF ICE SHEETS, OCCUR OVER LONGER TIME-SCALES, THOUSANDS TO MILLIONS OF YEARS. THESE CHANGES DIRECTLY IMPACT THE WELFARE OF HUMAN POPULATIONS WORLD-WIDE. THIS AWARD RENEWS FUNDING FOR THE PURDUE RARE ISOTOPE MEASUREMENT LABORATORY (PRIME LAB) AND THE UNIVERSITY OF VERMONT (UVM) COMMUNITY COSMOGENIC FACILITY. THESE TWO FACILITIES WILL COLLABORATE TO PRODUCE A NATIONAL, MULTI-USER FACILITY DEDICATED TO THE MEASUREMENT OF COSMOGENIC RADIONUCLIDES USING ACCELERATOR MASS SPECTROMETRY. DOING SO ALLOWS THE COMBINED FACILITY TO TRAIN STUDENTS AND RESEARCHERS FROM DOZENS OF UNIVERSITIES WITHIN THE US THAT DO NOT HAVE THESE CAPABILITIES OR THE FUNDS TO CREATE THEM. AT THE UNIVERSITY OF VERMONT, STUDENTS LEARN HOW TO PERFORM THE PHYSICAL AND CHEMICAL PREPARATION OF SAMPLES AND THEN PARTICIPATE IN THE MEASUREMENT OF THEIR SAMPLES ON THE PRIME LAB AMS. THE INTERACTION BETWEEN STUDENTS, FACULTY, AND PROFESSIONAL STAFF AT PRIME LAB AND UNIVERSITY OF VERMONT FACILITATES INTERACTION WITH GEOLOGISTS, CHEMISTS, AND PHYSICISTS, PROVIDING A UNIQUE OPPORTUNITY TO EXPAND THE EDUCATIONAL OPPORTUNITY BEYOND THE FOCUS OF SPECIFIC RESEARCH PROJECTS. AN ESSENTIAL COMPONENT OF THIS AWARD IS OUTREACH TO UNDERREPRESENTED COMMUNITIES, WHICH WILL PROVIDE STUDENTS HANDS-ON OPPORTUNITIES TO PARTICIPATE IN GEOLOGIC FIELD WORK, LABORATORY INSTRUCTION, AND EXPOSURE TO NUCLEAR PHYSICS MEASUREMENT TECHNIQUES. PRIME LAB HAS PROVIDED AMS MEASUREMENTS FOR THE GEOSCIENCE COMMUNITY SINCE THE EARLY 1990?S. PRIME LAB PROVIDES MEASUREMENTS OF 10BE, 14C, 26AL, 36CL, AND 129I IN TERRESTRIAL GEOLOGIC SAMPLES. PRIME LAB ROUTINELY MEASURES THOUSANDS OF UNKNOWNS ANNUALLY FOR RESEARCHERS, INCLUDING THOSE IN THE NSF COMMUNITY, BUT THE LABORATORY CAN NEITHER KEEP PACE WITH DEMAND AND ADEQUATELY TRAIN ENOUGH ?NEXT-GENERATION? COSMOGENIC NUCLIDES RESEARCHERS. ACCORDINGLY, A SIGNIFICANT GOAL OF THIS AWARD IS THE ESTABLISHMENT OF ADDITIONAL SAMPLE PREPARATION FACILITIES FOR THE GEOSCIENCE COMMUNITY. THE COMBINATION OF THE AMS MEASUREMENT CAPABILITIES AT PRIME LAB WITH THE SAMPLE PREPARATION FACILITIES AT UVM HAS VALUE BEYOND JUST THE COMBINATION OF TWO FACILITIES. THE NUMBER OF CYBERINFRASTRUCTURE (CI) PROFESSIONALS CRITICAL TO SUPPORT THESE ACTIVITIES WITHIN THE RESEARCH ENTERPRISE AT HIGHER EDUCATION INSTITUTIONS IS TOO FEW AND, FREQUENTLY, NOT WELL-INTEGRATED INTO REGIONAL AND NATIONAL EFFORTS THAT WOULD SERVE AS SOURCES OF INNOVATION. THIS PROJECT, CI PIVOT, WILL ADDRESS THESE WORKFORCE ISSUES BY CREATING SEVERAL TEAMS OF CI PROFESSIONALS TRAINED WITH TAILORED, BEST-OF-BREED EDUCATIONAL MATERIALS WHO WILL CONTRIBUTE, COLLABORATE, AND CO-CREATE AS PART OF LOCAL, REGIONAL, AND NATIONAL RESEARCH PARTNERSHIPS. THESE PARTNERSHIPS WILL PROMOTE THE PROGRESS OF SCIENCE AND RESULT IN SIGNIFICANT ADVANCES IN THE FIELDS OF GEOSCIENCE, ENGINEERING, AND SOCIAL BEHAVIORAL AND ECONOMIC SCIENCES. DESPITE THE EMERGING INTEREST AND ECONOMIC OPPORTUNITY OF LOCAL FOOD SYSTEMS, RESOURCES (E.G., RESEARCH, FUNDING) HAVE DISPROPORTIONATELY FAVORED CONVENTIONAL LARGE FARMS OVER THE MORE DIFFUSE NETWORK OF SMALL-MEDIUM SIZED FARMS THAT TAKE PART IN DIRECT-TO-CONSUMER (DTC) SALES. ONE OF THE CENTRAL TOOLS EMPLOYED BY DTC FARMERS ARE HIGH TUNNELS (HT), A FORM OF PROTECTED AGRICULTURE USED ALMOST EXCLUSIVELY TO PRODUCE SPECIALTY CROPS. LITTLE IS AVAILABLE IN THE WAY OF RESEARCH-BACKED RECOMMENDATIONS RELATED TO PEST MANAGEMENT IN HT CROPS, EVEN THOUGH WE KNOW FROM DISCUSSIONS WITH AND SURVEYS OF GROWERS THAT THIS IS A MAJOR FACTOR LIMITING PRODUCTION. IN THIS PROJECT, WE AIM TO UNDERSTAND THE IMPACTS OF CROP DIVERSITY (MONO- VS. POLYCUTLURE) ON PEST AND BENEFICIAL INSECTS ACROSS A RURAL TO URBAN GRADIENT AND ALL FOUR GROWING SEASON (I.E., WINTER PRODUCTION). WE WILL INVESTIGATE THE CONTRIBUTION OF NATURAL ENEMIES TO PEST SUPPRESSION THROUGH MOLECULAR GUT CONTENT ANALYSES AND EXPERIMENTAL MANIPULATIONS OF NATURAL ENEMY RELEASES ON GROWER AND RESEARCH FARMS. THEIR RAPID AND WIDESPREAD ADOPTION, INCLUDING REGISTRATION FOR USE ON VIRTUALLY ALL FIELD AND HORTICULTURAL CROPS, IS LARGELY DUE TO THEIR: A) HIGH SELECTIVITY TOWARDS INSECTS WITH LOW MAMMALIAN TOXICITY, MAKING NEONICS RELATIVELY SAFE TO HUMANS COMPARED WITH OLDER INSECTICIDE CLASSES; B) HIGH WATER SOLUBILITY, ALLOWING FOR SYSTEMIC UPTAKE AND CIRCULATION TO ALL PLANT PARTS; AND C) BROAD-SPECTRUM ACTIVITY THAT TARGETS A WIDE DIVERSITY OF INSECT PESTS. UNFORTUNATELY, THESE SAME ATTRIBUTES THAT MAKE NEONICS VERSATILE AND POWERFUL PEST MANAGEMENT TOOLS MAKE THEM PRONE TO NON-TARGET EFFECTS. THEIR SYSTEMIC ACTIVITY, FOR EXAMPLE, RESULTS IN THE PRESENCE OF ACTIVE INGREDIENTS IN POLLEN AND NECTAR, INCLUDING BOTH CROPS GROWN FROM NEONIC-TREATED SEED AND UNCULTIVATED WEEDS AND WILDFLOWERS, PROVIDING AN UNANTICIPATED EXPOSURE ROUTE FOR BENEFICIAL POLLINATING INSECTS. THUS, FARMERS GROWING POLLINATOR-DEPENDENT CROPS--REPRESENTING A LARGE FRACTION OF ALL FRUITS AND VEGETABLES--ARE CONFRONTED WITH A POTENTIAL TRADE-OFF BETWEEN TWO COMPETING ASPECTS OF CROP PRODUCTION: EFFECTIVE PEST SUPPRESSION AND SUCCESSFUL POLLINATION. IT REMAINS UNCLEAR, HOWEVER, TO WHAT DEGREE PEST AND POLLINATOR MANAGEMENT ARE MUTUALLY EXCLUSIVE IN SPECIALTY CROPS MANAGED WITH NEONICS AND WHETHER THIS TRADE-OFF CAN BE MITIGATED.THIS DILEMMA IS PARTICULARLY APPARENT IN CUCURBITS (COLLECTIVELY VALUED AT CA. $1.5 BILLION ANNUALLY IN THE U.S.), WHICH ARE ATTACKED BY SEVERAL INSECT PESTS THAT CONSTRAIN PRODUCTION. THESE PESTS ARE EFFECTIVELY MANAGED WITH NEONICS, BUT ARE ULTIMATELY DEPENDENT UPON BEES FOR POLLINATION AND FRUITSET. CUCURBITS PRESENT THE IDEAL FRAMEWORK TO ADDRESS A CONUNDRUM POSED BY NEONICS IN AGRICULTURE AT LARGE: CAN THESE POWERFUL AND UBIQUITOUS INSECTICIDES BE USED TO EFFECTIVELY TARGET PESTS WHILE SIMULTANEOUSLY PRESERVING KEY POLLINATORS? TO ADDRESS THIS BROAD GOAL, WE HAVE ASSEMBLED A DIVERSE TEAM OF RESEARCH AND EXTENSION SCIENTISTS, FROM ENTOMOLOGISTS TO ECONOMISTS, ACROSS SEVERAL MIDWESTERN AND NORTHEASTERN STATES, COMBINED WITH INPUT FROM A CONSORTIUM OF CUCURBIT GROWERS, BEEKEEPERS, AND PEST CONTROL REPRESENTATIVES. OUR LONG TERM GOAL IS TO DEVELOP HOLISTIC PEST MANAGEMENT REGIMES THAT ARE EFFECTIVE, PROFITABLE, AND SUSTAINABLE.
