THE PATHWAY TOOLS SOFTWARE IS A ROBUST AND COMPREHENSIVE SYSTEM FOR CONSTRUCTING SUCH ORGANISM-SPECIC DBS. PATH- WAY TOOLS ENABLES COMMUNITIES OF SCIENTISTS TO CREATE, QUERY, VISUALIZE, AND ANALYZE ORGANISM DBS, AND TO PUBLISH SUCH DBS ON THE WEB. THE SOFTWARE SUPPORTS CONSTRUCTION OF ORGANISM DBS THAT COMBINE A LARGE NUMBER OF BIOIN- FORMATICS DATATYPES, INCLUDING GENOME MAPS, GENES, OPERONS, RNAS, PROTEINS, CHEMICAL COMPOUNDS, BIOCHEMICAL REACTIONS, METABOLIC PATHWAYS, AND REGULATORY INTERACTIONS. PATHWAY TOOLS FURTHER ENABLES INFERENCE OF QUALITATIVE METABOLIC RECONSTRUCTIONS FROM SEQUENCED GENOMES, AND THE CREATION OF QUANTITATIVE METABOLIC UX MODELS FROM THOSE RECONSTRUCTIONS. SUCH METABOLIC MODELS ENABLE COMPUTATIONAL PREDICTIONS OF WHICH GENES AND REACTIONS WITHIN THE METABOLIC NETWORK ARE ESSENTIAL, THUS FACILITATING THE DESIGN OF DRUGS AGAINST DISEASE-CAUSING BACTERIA. WE PROPOSE TO RESEARCH SEVERAL NEW INFERENCE ALGORITHMS THAT DERIVE NEW INFORMATION FOR A SEQUENCED PATHOGEN GENOME FOR INCLUSION IN PATHWAY TOOLS DBS. ONE ALGORITHM WILL DETECT CROSS-FEEDING AND COFACTOR- EXCHANGE RELATIONSHIPS WITHIN A COMMUNITY OF ORGANISMS TO ADVANCE MECHANISTIC UNDERSTANDING OF THE HUMAN MICROBIOME. A SECOND ALGORITHM WILL ENABLE RAPID COMMUNICATION OF THE BIOCHEMICAL MACHINERY OF AN ORGANISM ENCODED IN ITS GENOME TO SCIENTISTS, SUCH AS DURING A DISEASE OUTBREAK. A THIRD ALGORITHM WILL IMPROVE THE PRE- DICTION OF OPERONS WITHIN BACTERIAL GENOMES BASED ON NEW FEATURES INCLUDING RNA-SEQ DATA. WE ALSO PROPOSE TO IMPLEMENT A SUITE OF CLOUD-BASED EDITING TOOLS FOR PATHWAY TOOLS DATABASES THAT EXECUTE WITHIN THE USER'S WEB BROWSER. THESE EDITING TOOLS WILL ENABLE COMMUNITIES OF SCIENTISTS TO WORK TOGETHER TO UPDATE AND SHARE KNOWL- EDGE ABOUT A GIVEN GENOME. PATHWAY TOOLS IS A MATURE AND PRODUCTION-GRADE SOFTWARE ENVIRONMENT THAT 10,800 RESEARCH GROUPS AROUND THE WORLD HAVE LICENSED, AND IT HAS BEEN CITED 1,650 TIMES. WE PROPOSE TO MAINTAIN THE SOFTWARE, SUCH AS BY INSTALLING NEW VERSIONS OF ITS MANY COMPONENTS, AND TO SUPPORT ITS USER COMMUNITY THROUGH TRAINING, DOCUMENTATION, AND BUG XING.

WILL DEVELOP INNOVATIVE EDGE-TO-CORE SECURITY SERVICES FOR THE NEXT GENERATION OF THE OPEN RADIO ACCESS NETWORK (O-RAN) COMPLIANT 5G+ MOBILE ARCHITECTURE. THIS COLLABORATIVE PROJECT, NAMED SECURITY-ENHANCED RADIO ACCESS NETWORK (SE-RAN), WILL FORTIFY 5G MOBILE INFRASTRUCTURES AGAINST A WIDE RANGE OF ATTACKS THAT TARGET VULNERABILITIES WITHIN 5G NETWORKS, PROTOCOLS, AND THEIR CONTROL-LAYER SERVICES. THE PROJECT'S CENTERPIECE IS A TRANSFORMATIVE NETWORK MANAGEMENT SERVICE, OFFERING 5G OPERATORS AN UNPRECEDENTED LEVEL OF THREAT IDENTIFICATION, POLICY ENFORCEMENT, AND COMPLIANCE MONITORING THROUGHOUT THEIR ENTIRE 5G NETWORK INFRASTRUCTURES. PROJECT SE-RAN SPECIFICALLY FOCUSES ON SAFEGUARDING MISSION-CRITICAL 5G NETWORKS, PROVIDING A COMPREHENSIVE PROTECTION ARCHITECTURE AGAINST SOPHISTICATED MOBILE-NETWORK ADVERSARIES. PROJECT SE-RAN WILL DELIVER AN O-RAN COMPLIANT 5G-NATIVE APPLICATION PROTECTION PLATFORM (5GNAPP) FOR MONITORING AND INLINE POLICY ENFORCEMENT ACROSS MOBILE DEVICES, BASE STATIONS, RAN OPERATIONS, AND THE 5G CONTROL PLANE. IT WILL SUBSTANTIALLY ENHANCE THE TRUSTWORTHINESS OF 5G NETWORKS, INCLUDING SECURITY WITH RESPECT TO MOBILE DEVICE PRIVACY, 5G COMMUNICATIONS CONFIDENTIALITY AND INTEGRITY, RESISTANCE TO ATTACKS, INCLUDING ATTEMPTS AT CONTROL-PLANE INFILTRATION, AND LIVE DETECTION OF ATTACKS AGAINST THE MOBILE INFRASTRUCTURE AND ITS USERS. THE PROJECT INVOLVES COLLABORATION WITH KEY OPEN-SOURCE STAKEHOLDERS TO INTEGRATE SECURITY SPECIFICATIONS AND MODULES WITH TOP-TIER 5G OPEN-SOURCE O-RAN PROJECTS. THE PROJECT WILL ALSO WORK WITH 5G INTEGRATORS THROUGHOUT THE GOVERNMENT TO TRANSITION MODULAR SECURITY SERVICES TO ADDRESS VARIOUS MISSION-CRITICAL USE CASES. FINALLY, THE PROJECT WILL FOSTER SUSTAINABLE IMPACTS ON THE U.S. INFORMATION TECHNOLOGY INDUSTRIES BY TRANSITIONING SE-RAN TECHNOLOGIES THROUGH STRATEGIC RELATIONSHIPS WITH STARTUPS, INDUSTRY LEADERS, AND INVESTORS ACTIVELY INVOLVED IN THE DEVELOPMENT OF NOVEL AND DISRUPTIVE 5G SECURITY AND PRIVACY TECHNOLOGIES. PROJECT SE-RAN WILL FOSTER A COMMUNITY THAT CREATES MODULAR O-RAN-COMPLIANT SECURITY COMPONENTS TO ENHANCE THE DEPLOYMENT AND RUNTIME MANAGEMENT OF MOBILE NETWORK INFRASTRUCTURES. THESE SOLUTIONS WILL EXTEND THE EXISTING O-RAN CONSORTIUM?S OPEN SOFTWARE ARCHITECTURE TO TACKLE AT LEAST TWO FUNDAMENTAL PROBLEMS. THE FIRST PROBLEM IS THE EXTENSIVE ATTACK SURFACE THAT ARISES FROM THE MIGRATION OF THE MOBILE NETWORK CONTROL PLANE INTO A CLOUD-BASED OPERATING ENVIRONMENT. WHILE THE INTEGRATION OF THE RAN INTELLIGENT CONTROLLER (RIC) INTO A KUBERNETES FRAMEWORK DRAMATICALLY INCREASES THE SCALABILITY AND EXTENSIBILITY OF CONTROL LOGIC, IT ALSO EXPOSES THE CONTROL PLANE TO THE BREADTH OF ADVERSARIAL TACTICS AND OPEN-SOURCE SUPPLY CHAIN VULNERABILITIES THAT PLAGUE EXISTING CLOUD ECOSYSTEMS. THE SECOND PROBLEM IS THE EXISTING LACK OF VISIBILITY INTO CORE 5G NETWORK OPERATIONS: ONE CANNOT SECURE THE MOBILE NETWORK IF ONE CANNOT OBSERVE ITS OPERATIONS WITH SUFFICIENT GRANULARITY. PROJECT SE-RAN REPRESENTS THE FIRST SECURITY-FOCUSED, BASE-STATION-INTERNAL TELEMETRY STREAM THAT WILL FACILITATE RUNTIME SECURITY MONITORING WITHIN THE O-RAN COMPLIANT 5G OPEN-SOURCE SOFTWARE (OSS) ECOSYSTEM. SE-RAN IS BASED ON FOUR GROUNDBREAKING INNOVATIONS. FIRST, SE-RAN WILL DELIVER A MODULAR BASE STATION EXTENSION (I.E., AN O-RAN SERVICE MODEL) THAT DELIVERS ADVANCED 5G-PROTOCOL LAYER-3 SECURITY AUDITING DESIGNED TO TRANSFORM THE ABILITY OF 5G OPERATORS TO TRACK THE SECURITY-RELEVANT STATE OF EVERY USER EQUIPMENT (UE) DEVICE AND BASE STATION IN THE NETWORK. SECOND, SE-RAN WILL DELIVER THE FIRST RUNTIME 5G-IDS (INTRUSION DETECTION SYSTEM) CONTROL PLANE APPLICATION FOR MALICIOUS RADIO FREQUENCY (RF)-BASED EXPLOIT AND ANOMALY DETECTION. THIRD, SE-RAN WILL INTRODUCE 5G-KUBEARMOR, THE FIRST NEAR REAL-TIME RAN INTELLIGENT CONTROLLER (NRT-RIC) SECURITY POLICY GENERATION AND ENFORCEMENT ENGINE, ENABLING 5G ADMINISTRATORS TO SECURE THE 5G CONTROL PLANE USING APPLICATION-LAYER LEAST-PERMISSIVE SECURITY POLICIES. FINALLY, IT WILL INTRODUCE THE FIRST 5GNAPP MANAGEMENT SERVICE THAT INTEGRATES ALL THREE TECHNOLOGIES UNDER A UNIFIED SECURITY INCIDENT AND EVENT MANAGEMENT (SIEM) SYSTEM. THE OVERALL BENEFIT OF THIS PROJECT WILL BE A TRANSFORMATIVE SECURITY FRAMEWORK THAT PROVIDES 5G OPERATORS WITH UNPRECEDENTED THREAT IDENTIFICATION, POLICY ENFORCEMENT, AND COMPLIANCE MONITORING THAT SPANS THE ENTIRE 5G NETWORK INFRASTRUCTURE. INSAR ADDRESSES THREE OF THE SEVEN MAJOR THEMES SUMMARIZED BY THE NATIONAL RESEARCH COUNCIL EARTH SCIENCES DECADAL SURVEY: 1) SOLID-EARTH HAZARDS AND DYNAMICS; 2) HUMAN HEALTH AND SECURITY; AND 3) LAND-USE CHANGE ECOSYSTEM DYNAMICS AND BIODIVERSITY. ALL REQUIRE GLOBAL FREQUENT ASSESSMENTS OF SOLID SURFACE DEFORMATION VIA INSAR MEASUREMENTS.FOR MAXIMUM IMPACT INSAR MEASUREMENTS MUST BE PRECISE (SUB-CM LEVEL) AND TIMELY. FREQUENT ACQUISITIONS (SUB-WEEKLY) ARENEEDED TO PROVIDE ENHANCED DEFORMATION PRECISION THROUGH TIME SERIES AVERAGING AND ENSURE THAT AN EVENT IS PROPERLY CAPTURED AND CHARACTERIZED. ORBITAL MECHANICS PREVENT SINGLE-PLATFORM SENSORS FROM SIMULTANEOUSLY ACHIEVING RAPID REVISIT TIMES AND WIDE-AREA COVERAGE. MULTIPLE PLATFORMS ARE NEEDED TO AVOID COMPROMISING COVERAGE BUT TRADITIONAL INSAR SENSORS ARE TOO EXPENSIVE (>$300M) TO REPLICATE.UNDER THE NASA ESTO IIP PROGRAM SRI INTERNATIONAL (SRI) AND OUR TEAM OF COLLABORATORS PROPOSE TO DEVELOP A COMPLETE SAR/INSAR INSTRUMENT FOR CUBESATS. THE INSTRUMENT IS CALLED CIRES-IDD (CUBESAT IMAGING RADAR FOR EARTH SCIENCE INSTRUMENT DEVELOPMENT AND DEMONSTRATION). WE WILL LEVERAGE SRIS TRL-5 CIRES RADAR HARDWARE DEVELOPED UNDER NASA ACT FUNDING SRIS TRL 4 IMAGE FORMATION SOFTWARE AND A TRL-3 HIGH-GAIN DEPLOYABLE MEMBRANE ANTENNA TO FORM THE COMPLETE INSTRUMENT.CIRES-IDD IS A THREE-YEAR PROGRAM WITH GROUND AND AIRBORNE UNMANNED AERIAL SYSTEMS (UAS) DEMONSTRATIONS TO RETIRE TECHNOLOGY RISKS AND RAISE THE CIRES-IDD SYSTEM TRL FROM 3 TO 6 IN PREPARATION FOR A FUTURE ORBITAL DEMONSTRATION CAMPAIGN. THE FOLLOWING POINTS SUMMARIZE THE MATURATION PLAN FOR EACH TECHNOLOGY PIECE.WE WILL START BY MATURING THE SAR/INSAR IMAGE-PROCESSING SOLUTION. SRI WILL APPLY THEIR EXTENSIVE SET OF SAR ALGORITHMS AND COHERENT REGISTRATION TECHNIQUES TO CIRES-IDD. WE WILL TEST AND DEBUG THE DEVELOPED SOFTWARE ON THE UAS PLATFORM FOLLOWED BY A RIGOROUS CALIBRATION CAMPAIGN. SRI EXPERIENCE WITH ADAPTING THE IMAGE FORMATION TOOLS TO DIFFERENT SYSTEMS (E.G. OPERATIONAL TRL-9 AIRBORNE RADAR SYSTEMS) ENABLES A RELATIVELY STRAIGHTFORWARD ALGORITHM DEVELOPMENT PATH BASED ON PREVIOUSLY PROVEN TECHNIQUES.OUR ANTENNA PARTNER PHYSICAL SCIENCES INC. (PSI) WILL DEVELOP THE HIGH-GAIN DEPLOYABLE MEMBRANE ANTENNA (>36 DBI). PSI WILL ITERATIVELY DEVELOP AND TEST INCREASING SCALES OF ANTENNA TEST ARTICLES FOCUSING ON DEPLOYMENT TENSIONING PACKAGING EFFICIENCY AND RF PERFORMANCE. ANTENNA RISK REDUCTION TESTS WILL INCLUDE MICROGRAVITY DEPLOYMENT AND FOLDING TESTS AMONG OTHERS. WORKING WITH NASA AMES RESEARCH CENTER AND THE NASA AIRBORNE SCIENCE PROGRAM WE WILL EXECUTE FOUR UAS FLIGHT CAMPAIGNS TOVALIDATE CIRES-IDD UTILITY AT ALTITUDE. THESE FLIGHTS WILL DEMONSTRATE THE FULL SAR/INSAR PERFORMANCE CAPABILITIES (25-M IMAGING RESOLUTION SUB-CM-LEVEL INSAR ACCURACY SNR>13 DB) AND DEMONSTRATE SCIENTIFIC RELEVANCE. WE HAVE PARTNERED WITH JET PROPULSION LABORATORY STANFORD UNIVERSITY AND THE U.S. GEOLOGICAL SURVEY TO IDENTIFY LOCAL AREAS OF INTEREST AND ASSIST WITH ANALYZING AND ASSESSING THE UTILITY OF THE DATA FOR SCIENTIFIC RESEARCH.THE LESSONS LEARNED FROM UAS TESTING WILL BE USED TO OPTIMIZE THE CIRES-IDD DESIGN FEATURES SUCH AS THERMAL RESILIENCE POWER MANAGEMENT MASS VOLUME AND SENSOR CAPABILITIES THEREBY IMPROVING SYSTEM PERFORMANCE AND REDUCING RISKS FOR FUTURE ON-ORBIT OPERATIONS. ALTHOUGH THE PROPOSED PROJECTS ULTIMATE GOAL IS TO PREPARE AND PROVE CIRES-IDD FOR FUTURE ON-ORBIT CUBESAT OPERATIONS THE UAS SAR SYSTEM RESULTING FROM THIS EFFORT WILL BECOME A USEFUL SCIENTIFIC RESEARCH PLATFORM IN ITS OWN RIGHT A CAPABILITY ALSO VALUED IN THE DECADAL SURVEY.

TRACKING HIV INFECTION & ALCOHOL ABUSE CNS COMORBIDITY WITH NEUROIMAGING - PROJECT SUMMARY ABSTRACT AS THE SUCCESSFULLY TREATED POPULATION OF HUMAN IMMUNODEFICIENCY VIRUS (HIV)-INFECTED INDIVIDUALS AGE, COGNITIVE AND HEALTH CHALLENGES OF NORMAL AGING ENSUE, BURDENED BY HIV, TREATMENT SIDE EFFECTS, AND HIGH PREVALENCE COMORBIDITIES, NOTABLY, ALCOHOL USE DISORDER (ALC) AND HEPATITIS C VIRUS (HCV). THE AGING BRAIN IS INCREASINGLY VULNERABLE TO ENDOGENOUS AND EXOGENOUS INSULT WHICH, COUPLED WITH HIV INFECTION, CAN LEAD TO HIV-ASSOCIATED NEUROCOGNITIVE DISORDER (HAND) AND SENSORIMOTOR DISTURBANCES. OUR GOAL IS TO EXPLICATE THE ROLE OF AGING IN MAGNETIC RESONANCE (MR)-DETECTABLE HIV PATHOLOGY IN THE CONTEXT OF COMMON COMORBIDITIES (ALC, HCV), WHILE CONSIDERING HIV-RELEVANT VARIABLES (NUTRITION, MEDICATION ADHERENCE AND TOXICITY, LIVER INTEGRITY). AS COMMON HIV-ASSOCIATED COMORBIDITIES OF ALC AND HCV MAY INTENSIFY INFLAMMATORY CASCADES AND POTENTIATE HIV PATHOLOGY, A COROLLARY AIM IS TO CONSIDER WHETHER CIRCULATING MARKERS OF INFLAMMATION (MICROBIAL DISLOCATION, MONOCYTE-RELATED IMMUNE ACTIVATION, PERIPHERAL CYTOKINE ELEVATIONS) CORROBORATE PROPOSED IMAGING MARKERS OF NEUROINFLAMMATION. WE WILL FOLLOW 140 PARTICIPANTS FROM OUR EXISTING COHORT AND RECRUIT 100 NEW SUBJECTS, WITH A FOCUS ON OLDER HIV-INFECTED INDIVIDUALS, USING MR METRICS CONSISTENT WITH OUR LONGITUDINAL DATABASE. IN KEEPING WITH THE REQUIREMENTS OF THE RFA, WE PROPOSE FOUR SPECIFIC AIMS: 1. INVESTIGATE THE CROSS-SECTIONAL AND LONGITUDINAL PATTERN OF BRAIN AND COGNITIVE CHANGES IN TERMS OF DISEASE AND AGE TRAJECTORIES USING QUANTITATIVE MR IMAGING AND NEUROCOGNITIVE MEASURES. 2. IDENTIFY FACTORS THAT MODIFY HIV DISEASE TRAJECTORY, INCLUDING AGE, SEX, ALCOHOL CONSUMPTION, HCV AND ITS TREATMENT, NUTRITION, MEDICATION ADHERENCE AND TOXICITY, AND HEPATIC INTEGRITY. EXPLORATORY FACTORS INCLUDE MEASURES OF DEPRESSIVE SYMPTOMS, FATIGUE, SLEEP QUALITY, AND IMPULSIVITY. 3. ESTABLISH EVIDENCE FOR NEUROINFLAMMATORY MARKERS USING FREE WATER DTI AND BRAIN METABOLITES QUANTIFIED BY MAGNETIC RESONANCE SPECTROSCOPY. 4. QUANTIFY THE EXTENT OF POSTURAL INSTABILITY AND TRUNCAL TREMOR IN RELATION TO COMORBID FACTORS AND NORMAL OR ACCELERATED AGING. ACCOMPLISHMENT OF THESE AIMS WILL ENHANCE UNDERSTANDING OF FACTORS CONTRIBUTING TO AGE AND DISEASE- RELATED COGNITIVE DECLINE, SENSORIMOTOR PROBLEMS, AND NEURODEGENERATION. CONVERGENT EVIDENCE FROM BLOOD BIOMARKERS (ELEVATIONS IN MICROBIAL DISLOCATION, MONOCYTE ACTIVATION, AND PROINFLAMMATORY CYTOKINES), MRS METABOLITES [MYO-INOSITOL (MI), CHOLINE-CONTAINING COMPOUNDS (CHO), MACROMOLECULES (MM09+LIP09] AND GLUTATHIONE (GSH)], AND DTI METRICS (FREE WATER) MAY PROVIDE INITIAL SUPPORT FOR A CONSTELLATION OF IN VIVO MARKERS OF NEUROINFLAMMATION.

NEUROFUNCTIONAL MECHANISMS OF CHANGES IN COGNITION AND MOTOR FUNCTION IN AGING WITH HIV AND PARKINSON'S DISEASE - PROJECT SUMMARY/ABSTRACT SINCE THE INTRODUCTION OF SUCCESSFUL TREATMENT FOR HUMAN IMMUNODEFICIENCY VIRUS (HIV) VIA ANTIRETROVIRAL THERAPY (ART), INDIVIDUALS INFECTED WITH HIV ARE NOW LIVING LONGER WITH ESTIMATES THAT IN THE UNITED STATES OVER HALF OF ALL HIV INFECTED INDIVIDUALS AND IN CALIFORNIA 67 PERCENT OF PEOPLE LIVING WITH HIV (PLWH) ARE OVER THE AGE OF 50. DUE TO THIS MAJOR DEMOGRAPHIC SHIFT IN THE HIV-INFECTED POPULATION IN THE US, AND AS OUTLINED IN PA-20-149, THERE IS AN URGENT NEED TO BETTER DEFINE THE UNDERLYING PATHOPHYSIOLOGY OF NEUROLOGICAL COMPLICATIONS AND NEUROCOGNITIVE DECLINE IN THE AGING HIV-INFECTED POPULATIONS. DESPITE ART TREATMENT, HIV-ASSOCIATED NEUROCOGNITIVE DISORDERS (HAND) OCCUR WITH HIGH HETEROGENEITY IN THE PATTERN AND SEVERITY OF DEFICITS, PARTICULARLY IN PEOPLE AGING ON MODERN ART REGIMENS. OUR RESEARCH POINTS TO NEUROIMAGING BIOMARKERS THAT COULD BE OF ADVANTAGE FOR IDENTIFYING DIAGNOSTIC SIGNATURES AND UNDERSTANDING THE HETEROGENEITY OF CNS DISEASE IN RELATION TO BEHAVIORAL MANIFESTATIONS, THAT WOULD NEED TO BE FOLLOWED UP WITH LONGITUDINAL RESEARCH. BRADYPHRENIA WAS INDICATIVE OF WORSE PERFORMANCE IN OTHER COGNITIVE DOMAINS, MEDIATED BY BASAL GANGLIA-LIMBIC BRAIN PATHWAY COMPROMISE, AND RELATED TO BRADYKINESIA, A MAIN SYMPTOM IN AGE-RELATED NEURODEGENERATION IN PARKINSON'S DISEASE (PD). BRADYKINESIA OCCURRED IN PLWH WITH MORE SEVERE COGNITIVE IMPAIRMENT AND COULD BE A SIGN OF DISEASE PROGRESSION INVOLVING MORE WIDESPREAD BRAIN NETWORKS. OUR NOVEL APPLICATION OF FMRI-DERIVED AMPLITUDES OF LOW FREQUENCY FLUCTUATIONS (ALFF) INDICATED DISRUPTED NEURODYNAMICS IN SUBCORTICO-CORTICAL CIRCUITS IN OLDER PLWH, WITH SOME OVERLAP WITH THE PATHOPHYSIOLOGY IN PD8. OUR RESEARCH POINTS TO PREDICTORS OF THE INTERINDIVIDUAL VARIATION IN IMPAIRMENT PROFILES RELATED TO PAST HIV SEVERITY (NADIR CD4, AIDS), CURRENT CD4 COUNT, OLDER AGE, ALCOHOL AND SUBSTANCE USE, AND SLEEP QUALITY AS A MEASURE OF RESILIENCE. HERE, WE EXTEND THIS WORK IN DIRECT RESPONSE TO PA-20-149, AIMING TO LONGITUDINALLY INVESTIGATE THE NEUROCOGNITIVE PROFILE USING THE NIMH'S RESEARCH DOMAIN CRITERIA (RDOC) FRAMEWORK TO EVALUATE PREVIOUSLY IDENTIFIED NEUROIMAGING BIOMARKERS OF COGNITIVE AND MOTOR DEFICITS IN AVIREMIC OLDER PLWH ON ART. SCIENTISTS FROM MULTIPLE DISCIPLINES MAKE WIDE USE OF ECOCYC; IT HAS BEEN CITED 4,939 TIMES, AND FROM 2019– 2021, AN AVERAGE OF 94,800 USERS PER YEAR VISITED THE ECOCYC WEBSITE. IT SERVES AS A GENERAL REFERENCE SOURCE ON E. COLI FOR EXPERIMENTAL BIOLOGISTS AND IS PARTICULARLY USEFUL FOR THE ANALYSIS OF FUNCTIONAL-GENOMICS EXPERIMENTS. THE DB SERVES COMPUTATIONAL BIOLOGISTS WHO ARE UNDERTAKING GLOBAL STUDIES OF E. COLI; METABOLIC ENGINEERS WHO ARE DEVELOPING NEW METHODS FOR CHEMICALS PRODUCTION, INCLUDING BIOFUELS; AND RESEARCHERS AND BIOINFORMATICISTS WHO ARE USING ECOCYC AS THE GOLD-STANDARD DATASET TO DEVELOP NEW COMPUTATIONAL METHODS, INCLUDING THE PREDICTION OF OPERONS, PROMOTERS, AND PROTEIN FUNCTIONAL LINKAGES. EDUCATORS ALSO USE THE DB. WE WILL UPDATE ECOCYC IN AN ONGOING FASHION TO REECT NEW INFORMATION ABOUT THE GENES, METABOLIC PATHWAYS, AND REGULATORY INTERACTIONS OF THESE IMPORTANT MODEL ORGANISMS. INFORMATION WILL BE INTEGRATED FROM THE BIOMED- ICAL LITERATURE AND FROM LARGE-SCALE EXPERIMENTS, SUCH AS DATA ON GENE ESSENTIALITY, ON NUTRIENTS SUPPORTING GROWTH, AND ON PROTEIN INTERACTIONS. WE WILL CONTINUE A COMPREHENSIVE AND ONGOING EFFORT TO RENE STEADY-STATE METABOLIC NETWORK MODELS OF THESE ORGANISMS BY VALIDATING MODEL PREDICTIONS AGAINST MANY CONDITIONS OF GROWTH AND NON- GROWTH FOR WILDTYPE AND KNOCK-OUT STRAINS. THE RESULTING MODELS WILL HAVE APPLICATIONS IN ANTI-MICROBIAL DRUG DISCOVERY AND METABOLIC ENGINEERING, AND THE MODEL DEVELOPMENT PROCESS WILL LEAD TO MANY IMPROVEMENTS IN THE ECOCYC DB. WE WILL LAUNCH A NEW EFFORT TO CURATE THE GENES AND PROTEINS OF E. COLI STRAINS OTHER THAN THE STRAIN SERVED BY ECOCYC. THOUSANDS OF E. COLI STRAINS HAVE BEEN SEQUENCED; YET IN MANY CASES, THEIR GENOME ANNOTATIONS ARE OF LOW QUALITY. WE WILL PROJECT CURATED GENE ANNOTATIONS FROM ECOCYC AND FROM OTHER E. COLI STRAINS TO ORTHOLOGS OF THOSE GENES IN THE BIOCYC DBS FOR HUNDREDS OF OTHER E. COLI STRAINS, THUS SIGNICANTLY IMPROVING THE ANNOTATION QUALITY OF OTHER E. COLI STRAINS IN A COST-EFFECTIVE FASHION. AGONISTS AT THE NOCICEPTIN/ORPHANIN FQ (N/OFQ) RECEPTOR (NOPR) HAVE SHOWN PROMISE AS MODULATORS OF THE ANTINOCICEPTIVE AND REWARDING EFFECTS OF OPIOIDS. WE HAVE FOUND THAT TWO DIFFERENT NOPR AGONISTS POTENTLY INDUCED SLEEP AND INCREASED EEG DELTA POWER IN RATS, MICE, AND NON-HUMAN PRIMATES, SUGGESTING THAT THE N/OFQ-NOPR SYSTEM MAY HAVE A PREVIOUSLY UNRECOGNIZED ROLE IN SLEEP/WAKE REGULATION. SINCE OUR STUDIES USED SYNTHETIC COMPOUNDS, THE HYPOTHESIS TO BE TESTED IN THIS PROPOSAL IS THAT THE N/OFQ-NOPR SYSTEM IS A COMPONENT OF THE ENDOGENOUS SLEEP/WAKE REGULATORY SYSTEM. FIRST, WE WILL EVALUATE WHETHER DISRUPTION OF ENDOGENOUS NOPR TONE AFFECTS THE REGULATION OF ACTIVITY, SLEEP/WAKE AND/OR BODY TEMPERATURE (TB) USING A NOVEL, INDUCIBLE NOPR (INOPR) KO MOUSE STRAIN IN WHICH NOPR CAN BE DELETED THROUGHOUT THE BRAIN BY SYSTEMIC ADMINISTRATION OF TAMOXIFEN. WE WILL EXPAND ON OUR INITIAL RESULTS WHICH INDICATE THAT, NOT ONLY DO INOPR KO MICE FAIL TO RESPOND TO NOPR AGONISM BY INCREASING NREM SLEEP, THEY EXHIBIT A PARTIAL INSOMNIA UNDER BASELINE CONDITIONS WITH LONGER WAKE BOUTS DURING THE MAJOR SLEEP PERIOD. WE WILL THEN BEGIN TO ESTABLISH THE NEUROANATOMICAL FOUNDATION FOR THE PHENOTYPES IDENTIFIED IN THE GLOBAL DELETION STUDY AND FOR SUBSEQUENT CIRCUIT-BASED STUDIES BY DETERMINING THE CHEMOTYPES OF NEURONS ACTIVATED BY NOPR AGONISM. FIRST, WE WILL DETERMINE NOPR EXPRESSION IN NEURONAL POPULATIONS KNOWN TO BE INVOLVED IN SLEEP/WAKE CONTROL. GLOBAL ASSESSMENT OF NOP AGONIST-INDUCED C-FOS EXPRESSION SHOWED THAT TWO OF 316 BRAIN AREAS STOOD OUT AS HAVING INCREASED FOS EXPRESSION: THE LATERAL HABENULA (LHB) AND THE ANTERODORSAL PREOPTIC AND ADJACENT PARASTRIAL (ADP/PS) NUCLEI. WE WILL FOLLOW UP BY OBTAINING A GLOBAL VIEW ON NOP AGONIST-INDUCED FOS EXPRESSION IN OPRL1 KI-EGFP AND PNOC-IRES-CRE (PREPRONOCICEPTIN-IRES-CRE) MICE AND USE OTHER GENETIC MARKERS TO FURTHER CHARACTERIZE AND QUANTIFY THE FOS+ NEURONS. NEXT, WE WILL DETERMINE WHETHER LOCAL NOPR DELETION AFFECTS ACTIVITY, SLEEP/WAKE AND/OR TB BY INJECTING A CRE-DEPENDENT VIRUS INTO CANDIDATE BRAIN REGIONS IN NOPRLOX/LOX MICE, PARTICULARLY FOCUSING ON THE LHB AND ADP/PS NUCLEI. WE WILL THEN CHEMOGENETICALLY ACTIVATE GLUTAMATERGIC AND NOPR+ NEURONS IN THE LHB TO DETERMINE WHETHER THESE CELLS AFFECT ACTIVITY, SLEEP/WAKE AND/OR TB AND USE THE FOS-TRAP METHOD TO CHEMOGENETICALLY REACTIVATE LHB AND ADP/PS NEURONS LABELLED AFTER NOPR AGONIST TREATMENT. THE INFORMATION OBTAINED WILL FURTHER OUR UNDERSTANDING OF THE ROLE OF THE N/OFQ – NOP SYSTEM IN SLEEP/WAKE CONTROL AND ENABLE US TO DEVELOP A CIRCUIT-BASED MODEL TO UNDERSTAND THE EFFECTS ON NOPR AGONISM ON SLEEP/WAKE PHYSIOLOGY.

DEVELOPING SCIENCE ASSESSMENTS FOR LANGUAGE DIVERSITY IN EARLY ELEMENTARY CLASSROOMS -THERE IS AN URGENT NEED FOR VETTED CLASSROOM-BASED, FORMATIVE ASSESSMENT TASKS FOR THE EARLY GRADES THAT ALIGN WITH THE NEXT GENERATION SCIENCE STANDARDS (NGSS); SPECIFICALLY, ONES THAT CONSIDER THE LANGUAGE AND LITERACY DIVERSITY OF YOUNG STUDENTS, AND EXPLICITLY INTEGRATE EQUITY AND INCLUSION FOR YOUNG SCIENCE LEARNERS IN THE DESIGN. THIS PROJECT WILL DESIGN INSTRUCTIONAL ASSESSMENT MATERIALS BY USING AN INNOVATIVE AND UNIQUE DESIGN APPROACH THAT BRINGS TOGETHER THE COHERENT AND SYSTEMATIC DESIGN ELEMENTS OF EVIDENCE-CENTERED DESIGN, AN EQUITY AND INCLUSION FRAMEWORK FOR THE DESIGN OF SCIENCE MATERIALS, AND INCLUSIVE DESIGN PRINCIPLES FOR LANGUAGE-DIVERSE LEARNERS. USING THIS THREE-PRONGED APPROACH, THIS PROJECT WILL DEVELOP A SUITE OF NGSS ALIGNED FORMATIVE ASSESSMENT TASKS FOR FIRST-GRADE SCIENCE AND A SET OF INSTRUCTIONAL MATERIALS TO SUPPORT TEACHERS AS THEY ADMINISTER THE FORMATIVE ASSESSMENTS TO STUDENTS WITH DIVERSE LANGUAGE SKILLS AND CAPACITIES. THESE RESOURCES WILL BE OF HIGH UTILITY FOR CLASSROOM TEACHERS IN THE EARLY ELEMENTARY GRADES AND OFFER AN IMPORTANT STEP TOWARDS MEETING THE NEED FOR USEABLE NGSS ALIGNED ASSESSMENTS FOR THE EARLY GRADES. THE DESIGN APPROACH WILL PROVIDE ASSESSMENT DEVELOPERS WITH A RIGOROUSLY DESIGNED AND TESTED MODEL FOR DEVELOPING THEIR OWN NGSS-ALIGNED ASSESSMENTS WHICH WILL SUPPORT THE STUDY OF SCIENCE IN EARLY ELEMENTARY CLASSROOMS AND PROMOTE FURTHER DEVELOPMENT OF NEW CURRICULUM MATERIALS, AND SUPPORT THE EVALUATION OF EXISTING CURRICULUM MATERIALS. THIS PROJECT WILL DEVELOP SMALL GROUP CHECKS AND INDIVIDUAL CHECKS AS FORMATIVE ASSESSMENTS AND STUDY THE DESIGN FEATURES, IMPLEMENTATION PRACTICES, AND AFFORDANCES OF EACH ASSESSMENT. TO ACCOMPLISH THE GOALS OF THIS PROJECT, THE PROJECT WILL BRIDGE THE NEXT GENERATION SCIENCE ASSESSMENT FOR YOUNG SCIENTISTS (NGSA-YS) AND THE EQUITY AND INCLUSION CURRICULUM DESIGN (EI-CD) APPROACHES TO GENERATE A NOVEL AND INNOVATIVE ASSESSMENT DESIGN AND DEVELOPMENT APPROACH. THE COMBINED APPROACH WILL GENERATE NEW ASSESSMENTS THAT RELIABLY MEASURE YOUNG LEARNERS? NGSS THREE-DIMENSIONAL SCIENCE PROFICIENCY AND PURPOSEFULLY POSITION EQUITY AND INCLUSIVITY AS CENTRAL TO THE DESIGN OF THE ASSESSMENTS. IN ADDITION, THE PROJECT WILL DEVELOP RESOURCES THAT GUIDE TEACHERS? IMPLEMENTATION AND SCORING OF THE ASSESSMENTS AND THEIR APPLICATION OF RESULTS TO INFORM INSTRUCTION IN LANGUAGE-DIVERSE CLASSROOMS. TO SUPPORT CLAIMS ABOUT VALIDITY, THE PROJECT WILL COLLECT DATA FROM MULTIPLE SOURCES INCLUDING EXPERT REVIEWS, COGNITIVE INTERVIEWS OF STUDENT PERFORMANCE, A SMALL CLASSROOM PILOT, AND A LARGER FIELD STUDY. THE PROJECT WILL USE ESTABLISHED PSYCHOMETRIC AND QUALITATIVE APPROACHES TO INVESTIGATE THE STRUCTURE OF SCORES FROM THE ASSESSMENT TASKS, EVALUATE WHETHER ASSESSMENT TASKS PROVIDE STUDENTS WITH SUFFICIENT AND INCLUSIVE OPPORTUNITIES TO DEMONSTRATE THEIR PROFICIENCIES IN SCIENCE AND ENSURE THAT FORMATIVE USE OF THE ASSESSMENT TASKS PROVIDES APPROPRIATE GRADE AND LANGUAGE AND LITERACY INFORMATION NEEDED TO MAKE INSTRUCTIONAL DECISIONS. CLASSROOM IMPLEMENTATION DATA WILL ALSO BE COLLECTED TO LEARN ABOUT CHALLENGES IN TEACHER ADMINISTRATION AND SCORING, AND THE USABILITY AND VALUE OF THE ASSESSMENTS TO TEACHERS. THIS PROJECT WILL CONTRIBUTE TO THE FIELD OF SCIENCE EDUCATION BY PROVIDING TEACHERS WITH FORMATIVE ASSESSMENT TASKS, RUBRICS, AND SUPPORT MATERIALS FOR USE IN THEIR CLASSROOMS AND BY PROVIDING ASSESSMENT DEVELOPERS WITH A MODEL FOR CREATING NGSS ASSESSMENTS FOR GRADE 1 THAT INCLUDES PRINCIPLES AND DESIGN DOCUMENTS FOR DEVELOPING ADDITIONAL TASKS. THE DISCOVERY RESEARCH PREK-12 PROGRAM (DRK-12) SEEKS TO SIGNIFICANTLY ENHANCE THE LEARNING AND TEACHING OF SCIENCE, TECHNOLOGY, ENGINEERING AND MATHEMATICS (STEM) BY PREK-12 STUDENTS AND TEACHERS, THROUGH RESEARCH AND DEVELOPMENT OF INNOVATIVE RESOURCES, MODELS AND TOOLS. PROJECTS IN THE DRK-12 PROGRAM BUILD ON FUNDAMENTAL RESEARCH IN STEM EDUCATION AND PRIOR RESEARCH AND DEVELOPMENT EFFORTS THAT PROVIDE THEORETICAL AND EMPIRICAL JUSTIFICATION FOR PROPOSED PROJECTS. ASSESSING THE EFFICACY AND IMPLEMENTATION OF A TECHNOLOGY-BASED MATHEMATICS INTERVENTION FOR MIDDLE SCHOOL STUDENTS -PROJECT ABSTRACT MIDDLE SCHOOL IS A CRITICAL JUNCTURE FOR MATHEMATICS LEARNING AND A POINT WHERE MANY U.S. STUDENTS GET LEFT BEHIND AND TURN AWAY FROM MATHEMATICS. THIS HAS A LASTING EFFECT, AS THESE STUDENTS ENTER HIGH SCHOOL UNPREPARED FOR THE COLLEGE PREPARATORY COURSES THAT WOULD SET THEM UP FOR MORE ADVANCED ACADEMIC AND CAREER OPPORTUNITIES. THIS PROJECT WILL INVESTIGATE THE POTENTIAL OF A NOVEL APPROACH TO MATHEMATICS INTERVENTION THAT LEVERAGES THE AFFORDANCES OF TECHNOLOGY AND EVIDENCE-BASED PEDAGOGY TO IMPROVE MATHEMATICS LEARNING FOR MIDDLE SCHOOL STUDENTS. THE MATHEMATICS INTERVENTION ENTITLED EMPIRES IS A COLLABORATIVE ACTIVITY SET IN ANCIENT MESOPOTAMIA THAT OFFERS A MULTIFACETED APPROACH IN WHICH (1) RICH, NARRATIVE MATH PROBLEMS INCREASE CONCEPTUAL COMPREHENSION; (2) ANIMATED REPRESENTATIONS OF MATHEMATICS CONCEPTS SUPPORT MATHEMATICAL UNDERSTANDING; (3) MULTIPLAYER COLLABORATION LEADS TO PEER INSTRUCTION AND MODELING; (4) SIMULATIONS OFFER EXCITING CHALLENGES THAT INCREASE MATHEMATICS RESILIENCY; AND (5) A BRIDGE CURRICULUM AIDS TRANSFER OF LEARNING TO MULTIPLE CONTEXTS, INCLUDING TRADITIONAL STANDARDIZED TESTS. THIS PROJECT WILL CONTRIBUTE TO THE RESEARCH BASE ON MIDDLE SCHOOL MATH INTERVENTIONS; THE APPLICATION OF TECHNOLOGY TO MATHEMATICS EDUCATION MORE BROADLY; AND THE POTENTIAL OF EDUCATIONAL TECHNOLOGIES TO SUPPORT MATHEMATICS MOTIVATION, RESILIENCY, AND ACHIEVEMENT FOR STUDENTS, ESPECIALLY ENGLISH LEARNERS. EMPIRES AUGMENTS TEACHERS? ABILITY TO FACILITATE LEARNING FOR A RANGE OF LEVELS AND PROVIDES TIMELY FEEDBACK, ALLOWING MATHEMATICS INTERVENTION TEACHERS FROM A RANGE OF BACKGROUNDS TO PROVIDE HIGHER-QUALITY LEARNING EXPERIENCES. THIS MODEL COULD HELP ENGLISH LEARNERS AND OTHER STUDENT GROUPS UNDERREPRESENTED IN STEM TO STAY MOTIVATED, PERSIST, AND PASS THE GATEWAY OF ALGEBRA INTO STEM CAREER PATHWAYS. THE CORE RESEARCH DESIGN WILL EMPLOY A ONE-YEAR CLUSTER RANDOMIZED CONTROLLED TRIAL. SCHOOLS WILL BE RANDOMLY ASSIGNED TO IMPLEMENT EMPIRES IN GRADE 7 MATHEMATICS INTERVENTION CLASSES DURING THE 2024/25 SCHOOL YEAR OR CONTINUE WITH BUSINESS AS USUAL. THE PRIMARY PURPOSE OF THE STUDY IS TO DETERMINE WHETHER EMPIRES IMPROVES MATHEMATICS ACHIEVEMENT ON AVERAGE, WHETHER STUDENT-LEVEL CHARACTERISTICS (E.G. ENGLISH LEARNER STATUS) MODERATE THE EFFECT OF EMPIRES, AND WHETHER STUDENT MOTIVATION, MATHEMATICS RESILIENCY, AND PEER TEACHING MEDIATE THE EFFECTS OF EMPIRES. STUDENT SCORES FROM THE STATE MATHEMATICS ASSESSMENT SERVE AS THE PRIMARY OUTCOME MEASURE. OTHER KEY MEASURES INCLUDE (1) ELECTRONIC USAGE RECORDS AUTOMATICALLY COLLECTED BY THE EMPIRES PLATFORM; (2) VALIDATED STUDENT SURVEY SCALES MEASURING MOTIVATION, MATHEMATICS RESILIENCY, AND PEER TEACHING MEDIATORS; AND (3) TEACHER IMPLEMENTATION INTERVIEWS AND SURVEYS. RESEARCHERS WILL ESTIMATE THE MAIN EFFECT OF EMPIRES USING HIERARCHICAL LINEAR MODELING, CONTROLLING FOR PRIOR ACHIEVEMENT AND OTHER COVARIATES. MODERATOR ANALYSES WILL ALSO EMPLOY HIERARCHICAL LINEAR MODELING TO EXAMINE WHETHER THE IMPACT OF EMPIRES VARIES FOR STUDENTS ACROSS POLICY-RELEVANT SUBGROUPS, INCLUDING ENGLISH LEARNERS. STRUCTURAL EQUATION MODELING WILL BE USED TO ASSESS WHETHER STUDENT MOTIVATION, MATHEMATICS RESILIENCY, AND PEER TEACHING MEDIATE THE EFFECTS OF EMPIRES ON STUDENT LEARNING. RESEARCHERS WILL ALSO EXAMINE THE RELATIONSHIP BETWEEN TEACHER AND STUDENT IMPLEMENTATION FIDELITY AND STUDENT ACHIEVEMENT. FINDINGS FROM THIS STUDY WILL HELP RESEARCHERS AND EDUCATORS BETTER UNDERSTAND DESIGN CHARACTERISTICS OF EFFECTIVE GAMES AND BEST PRACTICES FOR IMPLEMENTING THEM IN EDUCATIONAL SETTINGS, PARTICULARLY IN SETTINGS WITH HIGHER PERCENTAGES OF ENGLISH LEARNERS. DETECTION SYSTEM COMPRISING INEXPENSIVE PRINTED SENSOR ARRAYS FOR HYDROGEN GAS EMISSION MONITORING AND REPORTING (DECIPHER) THE PRIMARY GOAL OF THIS PROJECT IS TO DEVELOP A GAS DETECTION SYSTEM COMPRISING A DISTRIBUTED NETWORK OF LOW-COST SENSOR ARRAYS PRINTED WITH FUNCTIONALIZED CARBON NANOTUBES (CNTS) AND A ROBUST ANALYTICAL PLATFORM FOR MONITORING HYDROGEN GAS (H2) EMISSION IN OUTDOOR APPLICATIONS. THE DETECTION SYSTEM IS AIMED TO HAVE 5 ± 0.5 PPB SENSITIVITY WITH A FAST RESPONSE (<30 S) AND RECOVERY (<60 S) TIME, LONG LIFE (>10 YEARS) AND WILL QUANTIFY H2 CONCENTRATION WITH HIGH ACCURA-CY. THE PROJECT WILL BE EXECUTED IN THREE PHASES, EACH HAVING A 12-MONTH BUDGET PERIOD. BUDGET PERIOD 1 (BP1) WILL DELIVER A SENSOR ARRAY WITH <15 PPB SENSITIVITY AND A LOW-POWER SENSOR DE-VICE. BUDGET PERIOD 2 (BP2) WILL FOCUS ON DEVELOPMENT OF ANALYTICAL MODELS AND IMPROVEMENT OF SENSOR PERFORMANCE, FOLLOWED BY FIELD TEST 1. IN BUDGET PERIOD 3 (BP3), ANALYTICAL MODELS WILL BE FURTHER TRAINED WITH FIELD TEST DATA, AND FINALLY DEMONSTRATE THE CAPABILITIES OF THE DETECTION SYSTEM IN FIELD TEST 2. BP3 WILL CLOSE WITH A TECHNOECONOMIC ANALYSIS OF DECIPHER PROVIDING A PATHWAY FOR ADVANCING THE TECHNOLOGY FOR COMMERCIAL DEPLOYMENT.

IT WILL FUND AN HWC 2.1 MNPS SITE/DATA COORDINATOR DEDICATED TO BUILDING COLLABORATION ACROSS COMMUNITY PARTNERS (FIRST RESPONDERS, LAW ENFORCEMENT OFFICERS, AND YOUTH- AND FAMILY-SERVING ORGANIZATIONS), DISTRICT ADMINISTRATION, AND SCHOOL TEAMS TO SHARE DATA AND SOLUTIONS TO COMBAT THE EFFECTS OF SCHOOL VIOLENCE AND TRAUMA. IT WILL PROVIDE TRAINING SCHOOL STAFF AND COMMUNITY PARTNERS ON HOW TO REPORT HWC 2.1 INCIDENTS, RESPOND WITH EVIDENCE-BASED TRAUMA-SENSITIVE PRACTICES, AND MAKE REFERRALS TO MENTAL HEALTH SUPPORTS (AS WARRANTED). IT WILL INSTRUCT COACHING SCHOOL STAFF AND COMMUNITY PARTNERS TO USE DATA AND SUSTAIN CONTINUOUS IMPROVEMENT PROCESSES. THE R&D TEAM FROM SRI EDUCATION AND EVERGREEN VALLEY COLLEGE INTENDS TO INVESTIGATE AND DEVELOP AN INSTITUTIONAL LEVEL BADGING AND MICRO-CREDENTIALS PROGRAM AND SYSTEM. MANY BADGING AND MICRO-CREDENTIAL PROGRAMS DOCUMENT TECHNICAL COMPETENCIES, BUT THE FIELD LACKS UNDERSTANDING OF HOW SUCH PROGRAMS CAN INCREASE INSTITUTIONAL CAPACITY TO DEVELOP STEM TECHNICIAN STUDENTS' PROFESSIONAL SKILLS COMPETENCIES, SUCH AS COMMUNICATION, TEAMWORK, CAPACITY FOR LIFE-LONG LEARNING, AND ADAPTING TO THE WORKFORCE. THE OVERARCHING GOAL OF THE PROJECT IS TO INVESTIGATE AND DEVELOP MICRO-CREDENTIAL PROGRAMS FOR BOTH STUDENTS AND FACULTY RELATED TO TEACHING AND LEARNING PROFESSIONAL SKILLS. STEM TECHNICAL STUDENTS WILL USE BADGING TO DOCUMENT THEIR DEVELOPMENT OF PROFESSIONAL SKILLS FOR FUTURE EMPLOYMENT, SHARING THE BADGES DIGITALLY THROUGH LINKEDIN AND ON THEIR RESUMES. STEM TECHNICAL FACULTY WILL USE BADGING TO DOCUMENT THEIR LEARNING OF NEW INSTRUCTIONAL STRATEGIES TO TEACH PROFESSIONAL SKILLS IN THEIR CLASSES. PROJECT OBJECTIVES ARE TO (A) DESIGN, TEST, AND IMPLEMENT A BADGING SEQUENCE FOR PROFESSIONAL SKILLS MICRO-CREDENTIALS FOR STUDENTS TO DOCUMENT SATISFACTORY COMPLETION OF INSTRUCTIONAL ACTIVITIES; (B) DESIGN, TEST, AND IMPLEMENT A BADGING SEQUENCE THAT SUPPORTS AND DOCUMENTS INSTRUCTORS? INTEGRATION OF ACTIVITIES AND REFLECTIONS INTO COURSES; (C) DEVELOP A STRATEGY FOR ENGAGING EMPLOYERS AND LINKING TO BADGING ECOSYSTEMS TO BUILD ACCEPTANCE AND RECOGNITION OF PROFESSIONAL SKILLS MICRO-CREDENTIALS; AND (D) EXPLORE WAYS FOR COLLEGES TO TRACK PARTICIPATION, ACHIEVEMENT, AND WORKPLACE TESTIMONIALS. OBJECTIVES AND SIGNIFICANCE. THE EXTRACTION OF PLASMA FROM THE ATMOSPHERE IS ONE OF THE MOST IMPORTANT EFFECTS OF SPACE WEATHER ON EARTH AND ANY OTHER PLANET THAT HAS AN ATMOSPHERE. THE PROCESS OF EXTRACTION LIKELY RESULTS FROM A SEQUENCE OF MECHANISMS THAT START ACCELERATING UPWARD THE IONS IN THE F-REGION AND CONTINUE TO ACCELERATE THEM WELL ABOVE IT. THE CONTRIBUTION THAT EACH MECHANISM HAS ON THE ION ACCELERATION IS NOT WELL DETERMINED. FURTHERMORE THE WAVE MODES' IDENTIFICATION AS WELL AS THE HEIGHT DEPENDENCE OF THEIR AMPLITUDE IS NOT WELL ESTABLISHED. THIS PROJECT WILL TAKE FULL ADVANTAGE OF GROUND-BASED ASSETS (INCOHERENT SCATTER RADARS ISRS) SPACECRAFT ASSETS (FAST AURORAL SNAPSHOT FAST AND AKEBONO) AND MODELS (IONOSPHERE/POLAR WIND MODEL IPWM AND DYNAMIC FLUID-KINETIC DYFK) TO TRACE FOR THE FIRST TIME THE IONOSPHERIC POPULATIONS FROM THEIR SOURCE -THE F-REGION OF THE IONOSPHERE-INTO HIGH ALTITUDE WITH THE OBJECTIVE OF QUANTIFYING THE CONTRIBUTION OF THE DIFFERENT LIKELY ACCELERATION MECHANISMS AND THE ABILITY OF OUTFLOW MODELS TO PREDICT THE OBSERVED FLUXES. THE TEAM ASSEMBLED FOR THIS PROJECT HAS THE EXPERTISE AND EXPERIENCE IN INCOHERENT SCATTER RADARS (ISRS) FLUID AND HYBRID MODELS FAST AND AKEBONO WAVE DATA ANALYSIS TO SUCCESSFULLY CARRY OUT ALL TASKS OF THE PROJECT. METHODOLOGY. WE WILL CONSTRUCT AN EMPIRICAL STATISTICAL MODEL OF THE VARIATION OF WAVE AMPLITUDE WITH ALTITUDE USING FAST AND AKEBONO WAVE DATA. WE WILL USE THIS MODEL TO ESTABLISH MEAN AND VARIANCE OF WAVES' AMPLITUDES WITH ALTITUDE THAT CAN BE USED TO CONSTRAIN THE ALLOWABLE CHANGES IN THE ION EXTRACTION MODELS. WE WILL EMPLOY TWO DIFFERENT MODELS OF ION OUTFLOW: THE IPWM AND THE DYFK MODEL. WE WILL IDENTIFY ALL EVENTS IN THE ISR DATABASES (SONDRESTROM AND EISCAT) THAT CONTAIN SIGNS OF UP-FLOW IN THE FREGION WHILE EITHER FAST OR AKEBONO IS NEAR TO THE ISR LOCATION. WE WILL USE THE ISR MEASUREMENTS IN THE F-REGION TO INITIALIZE THE OUT-FLOW MODELS FROM BELOW AND SPACECRAFT MEASUREMENTS TO CONSTRAIN THEM FROM ABOVE AND MODEL THE ION FLUID (OR PARTICLES) MOTION ALONG THE FLUX TUBE WHILE CONVECTING THE FLUX TUBES HORIZONTALLY. WE WILL COMPARE THE FLUX AND PARTICLE DISTRIBUTION PREDICTED BY THE OUTFLOW MODELS (WITH THE WAVE PROPERTIES FROM THE FAST-AKEBONO EMPIRICAL MODEL AS INPUT) WITH THE FLUX AND PARTICLE DISTRIBUTION OBSERVED BY THE SPACECRAFT AND ITERATIVELY ADJUST THE WAVE-PARTICLE INTERACTION (WPI) INPUTS TO THE MODELS UNTIL SATISFACTORY AGREEMENT IS ACHIEVED WITHIN THE ERROR LIMITS IMPOSED BY THE FAST-AKEBONO EMPIRICAL MODEL. WE WILL COMPARE THE FINAL SOLUTION FOR THE WPI INPUTS TO THE WAVE OBSERVATIONS FROM THE SPACECRAFT AND WILL REPEAT THE COMPARISON METHODOLOGY FOR EVENTS AT DIFFERENT LOCATIONS OF THE POLAR IONOSPHERE AND LEVELS OF GEOMAGNETIC ACTIVITY. IMPACT OF THE WORK. THE EMPIRICAL MODEL OF WAVE POWER VERSUS ALTITUDE WILL HELP REDUCE THE LARGE UNCERTAINTY IN THE CURRENTLY ASSUMED VARIATION OF WAVE POWER WITH ALTITUDE REGION AND GEOMAGNETIC ACTIVITY. THIS INVESTIGATION WILL ALSO PROVIDE A DIRECT ESTIMATION OF THE PREDICTION ERROR THAT CAN BE ASSIGNED TO EACH OF THE MODELS AND THE QUANTIFICATION OF THE ADEQUACY OF THE ASSUMPTION OF ION CYCLOTRON RESONANCE HEATING BY EXTREMELY LOW FREQUENCY (ELF) WAVES. THE IMPROVEMENT IN THE SPECIFICATION OF ION ACCELERATION IN THE IPWM MODEL WILL BENEFIT ALL OTHER WORK USING THAT MODEL INCLUDING GLOBAL MODELING EFFORTS USING IPWM COUPLED TO THE MULTIFLUID LYON- FEDDER-MOBARRY (MFLFM) GLOBAL MAGNETOSPHERIC MODEL. PAST EXPERIENCE WITH THE GLOBAL COUPLED MODEL HAS SHOWN THAT THE COUPLED SYSTEM DYNAMICS AND GLOBAL MAGNETIC FIELD FLUCTUATIONS ARE HIGHLY SENSITIVE TO THE LOCAL DETAILS OF HOW ION ACCELERATION IS REGULATED. THEREFORE AN IMPROVED UNDERSTANDING OF LOCAL ION ACCELERATION IS EXPECTED TO HAVE A SIGNIFICANT IMPACT ON OUR ABILITY TO PREDICT GLOBAL DYNAMICS
