HIGH-INTENSITY, DYNAMIC-STABILITY GAIT TRAINING IN PEOPLE WITH MULTIPLE SCLEROSIS - PROJECT SUMMARY THE GOAL OF THIS PHASE I/II MULTICENTER CLINICAL TRIAL IS TO IMPROVE WALKING FUNCTION, DYNAMIC STABILITY, AND COMMUNITY MOBILITY IN PEOPLE WITH MULTIPLE SCLEROSIS (MS) USING A NOVEL COMBINATION OF HIGH-INTENSITY TRAINING AND AN ENVIRONMENT THAT CHALLENGES DYNAMIC BALANCE. THERE ARE TWO CRITICAL ELEMENTS OF THIS PROPOSAL: 1) BALANCE PERTURBATIONS APPLIED DURING TREADMILL WALKING WILL ALLOW PARTICIPANTS TO DEVELOP NEUROMUSCULAR STRATEGIES TO MAINTAIN UPRIGHT POSTURE, WHICH WILL BE REFINED AND ENHANCED WITH REPEATED PRACTICE, AND 2) PATIENTS WHO ENGAGE IN HIGH-INTENSITY TRAINING WILL IMPROVE THEIR NEUROMUSCULAR AND CARDIORESPIRATORY CAPACITY TO FACILITATE THE ABILITY TO WALK FASTER AND FURTHER. INDEED, PATIENTS WITH MILD OR MODERATE MS PRESENT WITH ALTERED LOCOMOTOR AND POSTURAL CONTROL, AND THIS 2X2 FACTORIAL DESIGN WILL DELINEATE THE INDIVIDUAL AND COMBINED EFFECTS OF REPEATED EXPOSURE TO POSTURAL PERTURBATIONS DURING WALKING AND HIGH-INTENSITY EXERCISE THAT MAY ENHANCE LOCOMOTOR AND BALANCE CONTROL IN BOTH LABORATORY CONDITIONS AND IN THE REAL-WORLD, WHERE THE LOCOMOTOR AND POSTURAL DEMANDS ARE SUBSTANTIAL. THIS STUDY IS PREDICATED ON THE PREVIOUS EXPERIENCE OF THE INVESTIGATORS WITH HIGH-INTENSITY TRAINING IN PATIENTS WITH OTHER NEUROLOGICAL DISORDERS AND PROMISING RESULTS OF A PILOT STUDY DEMONSTRATING FEASIBILITY OF DYNAMIC BALANCE TREADMILL TRAINING IN PEOPLE WITH MS. THE PRESENT PROJECT WILL ATTEMPT TO DELINEATE CHANGES IN CLINICAL AND LABORATORY MEASURES OF LOCOMOTOR CAPACITY FOLLOWING WALKING- SPECIFIC EXERCISE TRAINING AT HIGH VS LOW INTENSITY AND WITH OR WITHOUT POSTURAL PERTURBATIONS DURING TREADMILL STEPPING (AIM 1). AIM 2 WILL DEMONSTRATE THE RELATIVE EFFICACY OF THESE STRATEGIES ON DYNAMIC POSTURAL CONTROL (I.E., BALANCE) AND RELATE IMPROVEMENTS IN THE RESPONSE TO PERTURBATIONS TO SECONDARY MEASURES OF COMMUNITY FALLS. FINALLY, AIM 3 WILL DELINEATE CHANGES IN COMMUNITY MOBILITY AND PARTICIPATION FOLLOWING THESE VARIOUS EXERCISE PARADIGMS. WE SUGGEST THAT HIGH-INTENSITY LOCOMOTOR TRAINING WITH POSTURAL PERTURBATIONS WILL ELICIT THE LARGEST CHANGES IN BOTH LOCOMOTOR AND BALANCE OUTCOMES, AND THESE COMBINED GAINS WILL BOOST PHYSICAL ACTIVITY IN THE COMMUNITY SETTING LEADING TO IMPROVED QUALITY OF LIFE.

ENHANCEMENTS FOR A CAREER-CHANGE PATHWAY FOR NON-COMPUTING MAJORS -THIS PROJECT WILL CONTRIBUTE TO THE NATIONAL NEED FOR WELL-EDUCATED SCIENTISTS, MATHEMATICIANS, ENGINEERS, AND TECHNICIANS BY SUPPORTING THE RETENTION AND GRADUATION OF HIGH-ACHIEVING, LOW-INCOME STUDENTS WITH DEMONSTRATED FINANCIAL NEED AT MARQUETTE UNIVERSITY. OVER ITS SIX-YEAR DURATION, THIS TRACK 2 PROJECT WILL FUND SCHOLARSHIPS TO 35 UNIQUE FULL-TIME STUDENTS WHO ARE PURSUING GRADUATE DEGREES IN COMPUTER AND INFORMATION SCIENCE/DATA SCIENCE. POST-BACCALAUREATE STUDENTS WILL BE GIVEN THE OPPORTUNITY TO FORGE A CAREER PATHWAY INTO A PROFESSIONAL MASTER OF SCIENCE (MS) IN COMPUTER AND INFORMATION SCIENCE/DATA SCIENCE PROGRAM AND WILL RECEIVE SCHOLARSHIPS, ON AVERAGE, IN FIVE ACADEMIC TERMS (OUT OF A TOTAL OF SIX TERMS). SPECIFIC PROJECT ACTIVITIES INCLUDE (I) DEVELOPMENT, DELIVERY, AND ASSESSMENT OF IMPACTFUL DISTANCE LEARNING ADAPTATIONS TO THE FOUNDATIONS OF COMPUTING BRIDGE COURSE AS WELL AS CO-CURRICULAR SUPPORTS FOR STUDENTS SEEKING A CAREER CHANGE; (II) INVESTIGATION OF FACTORS INFLUENCING THE DEVELOPMENT OF COMPUTING PROFESSIONAL IDENTITY IN POST-BACCALAUREATE CAREER-CHANGING INDIVIDUALS; (III) DISSEMINATION OF FINDINGS RE-CURRICULAR AND CO-CURRICULAR ACTIVITIES THAT PROMOTE SUCCESSFUL CONVERSION INTO COMPUTING FOR STUDENTS WHO HAVE LITTLE PRIOR ACADEMIC TRAINING IN COMPUTING. A UNIQUE FEATURE OF MARQUETTE UNIVERSITY'S MASTER OF SCIENCE IN COMPUTER AND INFORMATION SCIENCE PROGRAM IS THE PROVISION OF A MODEL FOR STUDENTS TO CROSS OVER FROM NON-STEM UNDERGRADUATE DEGREES TO HIGH DEMAND, WELL-PAID JOBS IN COMPUTING. AN IMPORTANT BROADER IMPACT OF THE PROJECT IS THE DIVERSIFICATION OF THE POOL OF QUALIFIED COMPUTING PROFESSIONALS THROUGH INCLUSION OF ACADEMICALLY TALENTED LOW-INCOME STUDENTS. THE INTELLECTUAL MERIT OF THE PROJECT IS THE GENERATION OF NEW KNOWLEDGE ON OPTIMAL BRIDGE COURSE CONTENT AND DELIVERY AS WELL AS PROGRAM ACADEMIC SUPPORTS FOR STUDENTS WHO HAVE LITTLE PRIOR ACADEMIC TRAINING IN COMPUTING. THE OVERALL GOAL OF THIS PROJECT IS TO SIGNIFICANTLY INCREASE STEM DEGREE COMPLETION OF LOW-INCOME, HIGH-ACHIEVING UNDERGRADUATES WITH DEMONSTRATED FINANCIAL NEED. SPECIFICALLY, THE PROJECT WILL UPDATE AN ONLINE COMPUTER SCIENCE BRIDGE COURSE TO EXPEDITE A POST-BACCALAUREATE CAREER PATHWAY INTO A PROFESSIONAL MASTER OF SCIENCE IN COMPUTER AND INFORMATION SCIENCE PROGRAM. THE UNDERLYING RATIONALE FOR THE PROJECT IS THE NEED FOR A MECHANISM FOR STUDENTS WITH NON-COMPUTING AND NON-STEM UNDERGRADUATE DEGREES TO ACQUIRE COMPETENCIES IN COMPUTING THAT CAN LEAD TO A MASTER OF SCIENCE DEGREE IN COMPUTER AND INFORMATION SCIENCE. THERE IS EVIDENCE OF GROWING DEMAND FOR THIS TYPE OF BRIDGE COURSE AMONG A BROAD ADULT POPULATION, FURTHERMORE, THE CAREER CHANGE OPPORTUNITY WILL HAVE A POSITIVE EFFECT ON THE LOCAL AND REGIONAL WORKFORCE THROUGH PROVISION OF TRAINING NEEDED TO FULFILL CURRENT EDUCATION AND WORKFORCE DEVELOPMENT NEEDS. THE PROJECT WILL ADVANCE DEVELOPMENT OF THE BRIDGE COURSEWORK THAT PROVIDES ACCELERATED COVERAGE OF BACKGROUND CONCEPTS FOR THE SPECIFIC NEEDS OF CAREER CHANGING INDIVIDUALS. KNOWLEDGE GENERATION IS EXPECTED REGARDING THE DEVELOPMENT OF COMPUTING PROFESSIONAL IDENTITY IN POST-BACCALAUREATE, CAREER-CHANGING INDIVIDUALS. FOR FORMATIVE AND SUMMATIVE EVALUATION OF THE IMPACT OF THE BRIDGE COURSE, QUANTITATIVE DATA WILL BE COLLECTED BY THE EVALUATION TEAM THROUGH SURVEYS, WHILE QUALITATIVE DATA WILL BE COLLECTED THROUGH ONE-ON-ONE OR GROUP INTERVIEWS. DISSEMINATION OF FINDINGS WILL OCCUR THROUGH JOURNAL ARTICLES, CONFERENCE PAPERS AND POSTERS AT COMPUTER SCIENCE EDUCATION CONFERENCES, REGIONAL PROFESSIONAL ORGANIZATIONS AS WELL AS COMMUNITY STAKEHOLDERS, BUSINESSES AND ORGANIZATIONS ACTIVELY INVOLVED WITH STUDENT PRACTICUMS. THIS PROJECT IS FUNDED BY NSF'S SCHOLARSHIPS IN SCIENCE, TECHNOLOGY, ENGINEERING, AND MATHEMATICS PROGRAM, WHICH SEEKS TO INCREASE THE NUMBER OF LOW-INCOME ACADEMICALLY TALENTED STUDENTS WITH DEMONSTRATED FINANCIAL NEED WHO EARN DEGREES IN STEM FIELDS. IT ALSO AIMS TO IMPROVE THE EDUCATION OF FUTURE STEM WORKERS, AND TO GENERATE KNOWLEDGE ABOUT ACADEMIC SUCCESS, RETENTION, TRANSFER, GRADUATION, AND ACADEMIC/CAREER PATHWAYS OF LOW-INCOME STUDENTS. THIS AWARD REFLECTS NSF'S STATUTORY MISSION AND HAS BEEN DEEMED WORTHY OF SUPPORT THROUGH EVALUATION USING THE FOUNDATION'S INTELLECTUAL MERIT AND BROADER IMPACTS REVIEW CRITERIA.- SUBAWARDS ARE NOT PLANNED FOR THIS AWARD.

LIFE AFTER SPORT: PRIOR INJURY AND SEDENTARY BEHAVIOR AS MECHANISMS OF LATER POOR HEALTH - PROJECT SUMMARY/ABSTRACT APPROXIMATELY 8.5 MILLION HIGH SCHOOL AND COLLEGIATE ATHLETES ENGAGE ANNUALLY IN COMPETITIVE SPORT. WHILE COMPETITIVE SPORT OFFERS MANY BENEFITS, IT ALSO POSES UNDERAPPRECIATED RISKS TO LONG-TERM HEALTH. COMPETITIVE SPORT PROFOUNDLY INCREASES RISK FOR INJURY (E.G., KNEE INJURY) AND MAY POSITION ATHLETES FOR EARLY ONSET OF CHRONIC DISEASES (E.G., OSTEOARTHRITIS), CHRONIC PAIN, POOR HEALTH-RELATED QUALITY OF LIFE, AND DISABILITY. PRELIMINARY EVIDENCE INDICATES FORMER ATHLETES IN MID-LIFE (MEAN AGE 53) PERFORM WORSE ON FUNCTIONAL TESTS THAN NON-ATHLETE PEERS, BUT DID NOT ACCOUNT FOR PRIOR INJURY STATUS. QUANTIFYING FUNCTION IN FORMER ATHLETES WITH AND WITHOUT A PRIOR INJURY AND NON-ATHLETE CONTROLS IS THEREFORE CRITICAL TO INFORMING TARGETED INTERVENTIONS AND ULTIMATELY INCREASING HEALTHSPANS AND QUALITY OF LIFE. ONE MODIFIABLE FACTOR THAT MAY EXPLAIN THE ASSOCIATION BETWEEN SPORTS PARTICIPATION AND POOR LONG-TERM HEALTH IS PHYSICAL ACTIVITY (PA) PATTERNS INCLUDING PROLONGED SEDENTARY BEHAVIOR (SB). EVEN AMONG ADULTS WHO MEET RECOMMENDED PA GUIDELINES, SB IS STRONGLY ASSOCIATED WITH A HOST OF POOR OUTCOMES INCLUDING CHRONIC DISEASE AND ALL-CAUSE MORTALITY. CURRENT MALE PROFESSIONAL ATHLETES ARE SUBSTANTIALLY MORE SEDENTARY OUTSIDE THEIR PRACTICE TIMES THAN NON-ATHLETES, BUT WHETHER THIS HARMFUL PATTERN OF SB ALSO EXISTS IN AMATEUR (E.G., HIGH SCHOOL, COLLEGIATE) OR FEMALE ATHLETES OR PERSISTS IN FORMER ATHLETES IS UNKNOWN. THE LONG-TERM RESEARCH GOAL IS TO IMPROVE MUSCULOSKELETAL, CARDIOMETABOLIC, AND GENERAL HEALTH AND FUNCTION ACROSS THE LIFESPAN OF MALE AND FEMALE FORMER ATHLETES, EMPHASIZING EARLY BEHAVIORAL INTERVENTIONS THAT PREVENT THIS UNDERREPRESENTED AND AT-RISK POPULATION FROM DEVELOPING CHRONIC DISEASES BY MID-LIFE. THE OVERARCHING HYPOTHESIS IS THAT FORMER ATHLETES, ESPECIALLY THOSE WITH A PRIOR INJURY, WILL HAVE POORER FUNCTION AND HEALTH IN MID-LIFE AND THAT CURRENT AND FORMER ATHLETES WILL ENGAGE IN GREATER OVERALL AND LONGER BOUTS OF SB COMPARED TO MATCHED NON-ATHLETES. USING A CROSS-SECTIONAL DESIGN, AIM 1 WILL COMPARE FUNCTION, STRENGTH, AND CARDIOMETABOLIC INDICATORS AMONG FORMER ATHLETES WITH AND WITHOUT PRIOR KNEE INJURY AND NON- ATHLETE MATCHED CONTROLS IN MID-LIFE (AGE 45-64). HYPOTHESIS 1 IS THAT FORMER ATHLETES WITH A PRIOR INJURY WILL HAVE THE POOREST (1A) FUNCTION (30-SECOND CHAIR STAND, 6-MINUTE WALK, STAIR CLIMB), (1B) QUADRICEPS AND HAMSTRING STRENGTH, AND (1C) CARDIOMETABOLIC INDICATORS (BODY COMPOSITION, CARDIOVASCULAR DISEASE RISK). AIM 2 WILL COMPARE SB AND PA PATTERNS IN CURRENT (AGE 18-25) AND MID-LIFE FORMER ATHLETES TO MATCHED NON-ATHLETE CONTROLS. HYPOTHESIS 2.1 IS THAT CURRENT ATHLETES WILL HAVE GREATER SB, LONGER SB BOUTS, AND HIGHER LEVELS OF MODERATE-TO-VIGOROUS PA. HYPOTHESIS 2.2 IS THAT FORMER ATHLETES IN MID-LIFE WILL HAVE GREATER SB, LONGER SB BOUTS, AND LOWER PA LEVELS. IDENTIFYING AND ULTIMATELY INTERVENING ON RISK FACTORS FOR EARLY CHRONIC DISEASE WILL NOT ONLY BENEFIT MANY MILLIONS OF MALE AND FEMALE FORMER ATHLETES—A GROUP IN WHICH MINORITIES ARE INCREASINGLY MORE REPRESENTED THAN IN THE OVERALL UNITED STATES POPULATION—BUT ALSO PROVIDE AN EFFICIENT MEANS TO PREVENT OR DELAY CHRONIC DISEASE DEVELOPMENT IN THE POPULATION AT LARGE, A RECENT NIH EMPHASIS (E.G., PA-18-849).

MECHANISMS OF FATIGABILITY AND THE PROTECTIVE EFFECTS OF EXERCISE IN PEOPLE WITH DIABETES - ABSTRACT PRE-DIABETES (PRE-D) IS CHARACTERIZED BY ELEVATED GLYCATED HEMOGLOBIN AND PLASMA GLUCOSE AND IS A CLINICAL PRECURSOR TO TYPE 2 DIABETES MELLITUS (T2D). PRE-D CURRENTLY AFFECTS ~90 MILLION AMERICANS. BOTH PRE-D AND T2D ARE HIGHLY ASSOCIATED WITH CARDIOVASCULAR DISEASE AND AMONG THE TOP FIVE CAUSES OF MORTALITY WORLDWIDE. EXERCISE IS THE CORNERSTONE OF MANAGEMENT AND IS MOST EFFICACIOUS DURING THE PRE-D STAGE WHEN GLYCEMIA IS BELOW THE DIABETIC THRESHOLD. HOWEVER, EXCESSIVE FATIGABILITY DURING EXERCISE (I.E., EXERCISE INDUCED REDUCTIONS IN FORCE OR POWER OF THE LIMB MUSCLES) LIMITS EXERCISE PERFORMANCE IN PEOPLE WITH PRE-D. OUR LABORATORY DEMONSTRATED THAT (1) ACROSS THE DIABETIC SPECTRUM, PEOPLE WITH PRE-D AND T2D HAVE GREATER FATIGABILITY OF LIMB MUSCLES THAN CONTROLS DUE TO MECHANISMS WITHIN THE MUSCLE, AND (2) FATIGABILITY IN PEOPLE WITH T2D WAS ASSOCIATED WITH A REDUCED BLOOD FLOW TO THE EXERCISING MUSCLE. IT IS UNKNOWN, HOWEVER, IF PEOPLE WITH PRE-D HAVE IMPAIRED VASCULAR FUNCTION AND OXYGEN DELIVERY THAT LEADS TO AN INCREASED FATIGABILITY. OUR CENTRAL HYPOTHESIS IS THAT IMPAIRED VASCULAR FUNCTION IMPEDES BLOOD FLOW AND BLUNTS SUBSEQUENT OXYGEN DELIVERY TO SKELETAL MUSCLE DURING EXERCISE, RESULTING IN EXCESSIVE FATIGABILITY OF LIMB MUSCLES IN PEOPLE WITH PRE-D. A UNIQUE AND TRANSLATIONAL ASPECT OF THIS PROPOSAL IS THE QUANTIFICATION OF THE VASCULAR RESPONSES (MACRO- AND MICRO-VASCULATURE) TO FATIGUING EXERCISE AND EXERCISING TRAINING AT DIFFERENT SITES ALONG THE VASCULAR TREE, INCLUDING, IN FEED ARTERIES (DOPPLER ULTRASONOGRAPHY), ISOLATED SKELETAL MUSCLE ARTERIOLES (EXTRACTED FROM MUSCLE BIOPSY), CAPILLARY PERFUSION (NEAR INFRARED SPECTROSCOPY, NIRS) AND CAPILLARY DENSITY (FROM MUSCLE BIOPSIES). AIM 1 WILL DETERMINE IF VASCULAR DYSFUNCTION IS A MECHANISM FOR EXCESSIVE FATIGABILITY IN PEOPLE WITH PRE-D. AIM 1.1 WILL COMPARE LEG BLOOD FLOW AND SKELETAL MUSCLE OXYGENATION IN RESPONSE TO DYNAMIC FATIGUING EXERCISE BETWEEN PEOPLE WITH PRE-D, HEALTHY CONTROLS AND T2D. GROUPS WILL BE MATCHED FOR AGE, SEX, BODY MASS INDEX AND PHYSICAL ACTIVITY LEVELS TO DETERMINE DISEASE- RELATED VASCULAR FUNCTION AND FATIGABILITY RATHER THAN INACTIVITY-RELATED CHANGES ACROSS THE DIABETIC SPECTRUM. SKELETAL MUSCLE BLOOD FLOW THROUGH THE FEMORAL ARTERY WILL BE QUANTIFIED WITH ULTRASONOGRAPHY AND SKELETAL MUSCLE OXYGENATION WITH NIRS DURING A DYNAMIC FATIGUING KNEE EXTENSION EXERCISE. AIM 1.2 WILL DETERMINE ENDOTHELIAL VASCULAR FUNCTION AT MACRO- AND MICRO-VASCULAR LEVELS IN PEOPLE WITH PRE-D AND T2D. FLOW-MEDIATED DILATION WILL BE ASSESSED IN VIVO IN THE FEMORAL ARTERY USING ULTRASONOGRAPHY AND IN ISOLATED ARTERIOLES EXTRACTED FROM THE VASTUS LATERALIS MUSCLE BIOPSIES. AIM 2 IS A CLINICAL TRIAL THAT WILL DETERMINE THE EFFECTIVENESS OF RESISTANCE EXERCISE TRAINING COUPLED WITH BLOOD FLOW RESTRICTION TO IMPROVE FATIGABILITY AND VASCULAR FUNCTION IN PEOPLE WITH DIABETES. PEOPLE WITH PRE-D AND T2D WILL PERFORM 8 WEEKS OF UNILATERAL RESISTANCE TRAINING IN WHICH ONE LEG IS EXERCISED WITH FREELY PERFUSED CONDITIONS AND THE OTHER LEG WITH BLOOD FLOW RESTRICTION. THUS, BLOOD FLOW RESTRICTION AND RESISTANCE TRAINING WILL BE USED AS A PROBE TO FURTHER UNDERSTAND THE MECHANISMS OF FATIGABILITY ALONG THE VASCULAR TREE IN PEOPLE WITH PRE-D AND T2D, AND TEST TRAINING STRATEGIES TO IMPROVE FATIGABILITY IN THIS POPULATION.

SEXUAL DIMORPHIC CIRCUITS IN PHOTOPERIODIC ENCODING AND PHOTIC PROCESSING - PROJECT SUMMARY/ABSTRACT LIGHT REGULATES NEUROLOGICAL FUNCTION THROUGH CIRCUITS THAT REMAIN POORLY DEFINED, AND THERE REMAINS A SIGNIFICANT UNMET NEED IN UNDERSTANDING PHOTIC PLASTICITY IN THE BRAIN OF BOTH SEXES. FOR EXAMPLE, SEASONAL CHANGES IN DAY LENGTH (I.E., PHOTOPERIOD) MODULATE SLEEP, ATTENTION, APPETITE, METABOLISM, MOOD, AND IMMUNE FUNCTION OCCUR IN A LARGE NUMBER OF PEOPLE, WITH UP TO 80% BEING WOMEN. THE SUPRACHIASMATIC NUCLEUS (SCN) DRIVES CIRCADIAN AND PHOTOPERIODIC RESPONSES TO LIGHT, BUT THERE IS A LARGE GAP IN OUR UNDERSTANDING OF SEX DIFFERENCES IN SCN PHOTIC AND PHOTOPERIODIC PROCESSING. OUR PRELIMINARY DATA INDICATE THAT CIRCADIAN AND PHOTOPERIODIC RESPONSES DIFFER BY SEX, WITH FEMALES DISPLAY LARGER BEHAVIORAL RESPONSES TO LIGHT MEDIATED BY DIVERGENT SCN CIRCUITS. BASED ON THESE DATA, WE PROPOSE THE CENTRAL HYPOTHESIS THAT PHOTIC PROCESSING DIFFERS BETWEEN MALES AND FEMALES BECAUSE THE RETINA-SCN CIRCUIT IS SEXUAL DIMORPHIC. WE WILL TEST THAT BIOLOGICAL SEX INFLUENCES CELLULAR RESPONSES TO LIGHT (AIM1), PHOTOPERIODIC RESPONSES TO DAY LENGTH (AIM2), AND NEURAL ADAPTATIONS IN SCN CIRCUITS (AIM3). THESE STUDIES WILL BENEFIT FROM THE COLLECTIVE EXPERTISE OF THIS COLLABORATIVE RESEARCH TEAM, WHICH SPECIALIZES IN RETINAL PHYSIOLOGY, SCN NETWORK PROPERTIES, CELLULAR IMAGING, AND COMPUTATIONAL ANALYSES. INDEPENDENT OF SEX DIFFERENCES, SUCCESSFUL COMPLETION OF THESE STUDIES WILL PROVIDE NOVEL INSIGHTS INTO NEURAL MECHANISMS OF PHOTIC PROCESSING AND PHOTOPERIODIC ENCODING TO ADVANCE UNDERSTANDING IN BOTH SEXES. GLOBAL ANALYSIS OF THE SUPRASPINAL CONNECTOME AFTER SPINAL CORD INJURY - PROJECT SUMMARY THE SUPRASPINAL CONNECTOME REFERS TO THE FULL SET OF NEURONS THAT RESIDE IN THE BRAIN AND PROJECT AXONS TO THE SPINAL CORD. THE SHEER COMPLEXITY OF THIS CONNECTOME, BOTH ANATOMICAL AND FUNCTIONAL, HAS PRESENTED STUBBORN CHALLENGES TO UNDERSTANDING AND TREATING THE HIGHLY VARIABLE DISRUPTION THAT ACCOMPANIES SPINAL INJURY. ANATOMICALLY, THE WIDE DISTRIBUTION OF THE CELL BODIES HAS MADE IT VERY DIFFICULT TO TRACE AND QUANTIFY ALL POPULATIONS SIMULTANEOUSLY, LEADING MOST STUDIES TO FOCUS ON JUST ONE OR SEVERAL MAIN SUPRASPINAL CELL TYPES. FUNCTIONALLY, DESCENDING CONTROL IS DISTRIBUTED ACROSS MULTIPLE POPULATIONS WITH OUTPUTS THAT CAN BE COMPLEMENTARY, ANTAGONISTIC, OR POTENTIALLY COMPENSATORY AFTER INJURY. THESE FUNCTIONAL RELATIONSHIPS STRONGLY AFFECT OUTCOMES AFTER SPINAL INJURY BUT ARE DIFFICULT TO CAPTURE IN SINGLE- OR SEVERAL-TRACT APPROACHES. THIS PROJECT AIMS TO ADVANCE THE FIELD THOUGH AN APPROACH THAT YIELDS A COMPREHENSIVE UNDERSTANDING OF SPINAL INJURY AT A WHOLE-CONNECTOME LEVEL AND BY PROVIDING NEW VIRAL TOOLS TO SELECTIVELY MANIPULATE SUBTYPES OF BRAIN-SPINAL NEURONS. FIRST, WE WILL USE 3D WHOLE-BRAIN AND -SPINAL IMAGING TO VISUALIZE AND PRECISELY QUANTIFY THE NUMBER AND ACTIVITY LEVEL OF SPARED NEURONS IN ALL SUPRASPINAL CELL TYPES ACROSS COHORTS OF ANIMALS THAT RECEIVE GRADED CONTUSION INJURIES, WHILE SIMULTANEOUSLY MEASURING BOTH SPONTANEOUS AND REHABILITATION-INDUCED RECOVERY OF FUNCTION. WITH WHOLE-BRAIN DATA AND ADVANCED STATISTICAL TECHNIQUES WE CAN TAKE ADVANTAGE OF THE NATURAL VARIABILITY BETWEEN INDIVIDUAL ANIMALS TO IDENTIFY THE ELEMENTS OF THE CONNECTOME THAT ARE MOST PREDICTIVE OF RECOVERY. NEXT, WE WILL SYSTEMATICALLY TEST THESE PREDICTED FUNCTIONAL RELATIONSHIPS BY TARGETED, REVERSIBLE CHEMOGENETIC SILENCING OF SELECTED SUPRASPINAL POPULATIONS BEFORE INJURY, AFTER INJURY, AND AFTER REHABILITATION. IMPORTANTLY, THESE SILENCING EXPERIMENTS WILL EMPLOY BOTH DIRECT TARGETING OF BRAIN REGIONS AND NEWLY DEVELOPED VIRAL TOOLS THAT ENABLE SELECTIVE MANIPULATION OF DEFINED SUBTYPES OF SPINALLY PROJECTING NEURONS. ADDITIONAL SURGERY IS ASSOCIATED WITH SIGNIFICANT EMOTIONAL, COSMETIC AND FINANCIAL BURDENS FOR PATIENTS AND THEIR CAREGIVERS. ALTHOUGH RADIOGRAPHY, FROZEN SECTION, TOUCH PREP AND MARGINPROBE ARE AVAILABLE FOR INTRAOPERATIVE MARGIN ASSESSMENT, THEIR ACCURACY IS VARIABLE AND MOST, EXCEPT RADIOGRAPHIC EXAMINATION, ARE TIME- AND LABOR- INTENSIVE AND NOT ROUTINELY USED. SINCE PUBLICATION OF THE 2014 SSO-ASTRO GUIDELINES FOR INVASIVE CANCER RECOMMENDING RE-EXCISION FOR POSITIVE MARGINS ONLY, THE RE-EXCISION RATES HAVE DECREASED BUT REMAIN SUBSTANTIAL (14-18%) WITH SIGNIFICANT VARIATION AMONG SURGEONS. BECAUSE THE SIZE OF BCS SPECIMENS VARIES SIGNIFICANTLY (A FEW TO >40 CM2 PER MARGIN) AND POSITIVE MARGINS OFTEN INCLUDE ONE OR MULTIPLE SITES/FOCI, A DEVICE WITH BOTH VARIABLE MARGIN COVERAGE AND MICROSCOPIC RESOLUTION THAT CAN ACCURATELY EVALUATE AN ENTIRE SURGICAL SPECIMEN WITHIN A FEW MINUTES IS HIGHLY DESIRABLE. WHILE NEW TECHNOLOGIES HAVE BEEN PROPOSED, THEY ARE EITHER POINT OR HIGH RESOLUTION DEVICES WITH A VERY SMALL FIELD-OF-VIEW THAT REQUIRES EXCESSIVE TIME TO SCAN A SPECIMEN, OR WIDE- FIELD DEVICES WITH LOW RESOLUTION AND POOR SENSITIVITY. NONE HAS DEMONSTRATED THE CAPABILITY OF ANALYZING AN ENTIRE LUMPECTOMY SPECIMEN WITH BOTH ADEQUATE RESOLUTION AND TIME EFFICIENCY IN A CLINICAL SETTING. OUR GOAL IS TO DEVELOP A DEEP LEARNING (DL) ENABLED, DEEP ULTRAVIOLET (DUV) SCANNING MICROSCOPE (DDSM) FOR SUBCELLULAR RESOLUTION AND RAPID (<5 MIN) EXAMINATION OF FRESHLY EXCISED TUMOR SPECIMENS DURING BCS. WE HYPOTHESIZE THAT THERE ARE SIGNIFICANT SUBCELLULAR OPTICAL CONTRASTS THAT CAN BE IDENTIFIED BY THE DDSM TO DIFFERENTIATE BREAST CANCER CELLS FROM NORMAL TISSUE. OUR PRELIMINARY DUV IMAGES DEMONSTRATE EXCELLENT CONTRASTS AND ACCURACY FOR IDENTIFICATION OF BREAST CANCER CELLS. WE PROPOSE THAT LARGE AND VARIABLE MARGIN COVERAGE, MICROSCOPIC RESOLUTION AND HIGH SPEED ARE ACHIEVED BY USING: 1) DUV LIGHT FOR SURFACE EXCITATION OF FRESH SPECIMENS; 2) PARALLEL IMAGING OF TWO MARGINS; 3) A LOW OPTICAL MANIFICATION FOR FAST SPEED; AND 4) DL AND SPARSE-SAMPLING (SS) TO RAPIDLY SEARCH FOR PATHOLOGICAL FEATURES OF CANCER CELLS. IN AIM 1, A NOVEL DDSM INSTRUMENT WILL BE DEVELOPED AND USED TO IMAGE 120 FRESH BREAST TISSUES. DL CLASSIFICATION ALGORITHMS WILL BE DEVELOPED AND VALIDATED USING THE 120 TISSUE SAMPLES IN AIM 2. AIM 3 WILL INTEGRATE DL AND SS ALGORITHMS INTO THE DDSM AND DEMONSTRATE FOR FAST DETECTION OF VARIABLE AMOUNT OF CANCER CELLS ON THE SURFACES OF BREAST TUMOR SPECIMENS. DDSM IS HIGHLY INNOVATIVE, COMBINING DUV MICROSCOPY, PARALLEL IMAGING, DL CLASSIFICATION, AND SS IN A FAST, COMPACT, AUTOMATED DESIGN. DURING INITIAL BCS, IF THE DDSM ACCURATELY AND EFFICIENTLY IDENTIFIES POSITIVE MARGINS, ADDITIONAL BREAST TISSUE WOULD BE REMOVED FROM THE SURGICAL CAVITY UNTIL NEGATIVE MARGINS ARE ACHIEVED AND UNNECESSARY REMOVAL OF ADDITIONAL TISSUE WOULD BE AVOIDED, THUS DECREASE THE NEED FOR ADDITIONAL SURGERY. OPIOID-INDUCED DYSREGULATION OF CORTICO-STRIATAL CIRCUITS - PROJECT SUMMARY/ABSTRACT OPIOID-BASED DRUGS ARE MAINSTAYS FOR PAIN MANAGEMENT DESPITE THE RISK OF DEPENDENCE, ADDICTION, AND COGNITIVE IMPAIRMENTS, EVEN WHEN TAKEN AS DIRECTED. YET, THE NEUROLOGICAL UNDERPINNINGS OF OPIOID ADDICTION REMAIN RELATIVELY UNDER-INVESTIGATED, THEREBY LIMITING OUR UNDERSTANDING OF THE NEURAL AND ASSOCIATED PSYCHOLOGICAL BASIS OF ADDICTION TO OPIOIDS. IMPAIRED DECISION-MAKING, STRONG PERSEVERANCE AND AN EXTRAORDINARY MOTIVATION TO OBTAIN DRUG ARE WIDELY SEEN AS CORE ASPECTS OF ADDICTION. THESE BEHAVIORS ARE EXPRESSED ONLY BY A FRACTION OF THOSE EXPOSED TO THE DRUG – HIGHLIGHTING A SIGNIFICANT DEGREE OF INDIVIDUAL VARIABILITY AND A NEED TO IDENTIFY LARGELY UNKNOWN FACTORS THAT CONVEY RISK OR RESILIENCE. HYPOFUNCTION OF PREFRONTAL CIRCUITS HAS EMERGED AS A PRIMARY MECHANISM BY WHICH THIS TRANSITION OCCURS, HOWEVER EVIDENCE REGARDING THE NATURE, SUB-CIRCUIT(S) LOCUS, AND DEVELOPMENT OF THIS HYPOFUNCTION IS LACKING. OUR PUBLISHED DATA FIND THAT SELF-ADMINISTRATION (SA) OF THE POTENT OPIOID, REMIFENTANIL (REM), PROMOTES A PROGRESSIVE HYPOACTIVE STATE IN PRELIMBIC CORTEX (PRL) PYRAMIDAL NEURONS THAT UNDERLIES IMPAIRMENTS IN COGNITIVE FLEXIBILITY, AND THAT THIS PHENOMENON OCCURS ON A FASTER TIMELINE IN FEMALES. PILOT DATA INDICATE INFLEXIBILITY ALIGNS WITH INCREASED PERSEVERATIVE DRUG SEEKING AND MOTIVATION FOR DRUG THAT IS MORE PROMINENT IN FEMALES. WE PREDICT THESE CHANGES EMERGE AS A RESULT OF CHANGES IN PL-CORE CIRCUITS, AS PILOT DATA INDICATE THAT HYPOACTIVITY IS MORE PROMINENT IN PRL-CORE PN AND THAT THIS PLASTICITY ALIGNS WITH SEX- AND CELL-TYPE SPECIFIC ADAPTATIONS IN SYNAPTIC STRENGTH AT DOWNSTREAM CORE MEDIUM SPINY NEURONS (MSNS) BASED ON EXPRESSION OF DOPAMINE TYPE I (D1) VERSUS TYPE II (D2) RECEPTORS. THIS PROPOSAL WILL COMBINE CIRCUIT-SPECIFIC EX VIVO MEASURES OF PLASTICITY, IN VIVO CA2+ IMAGING, AND CHEMOGENETICS WITH A NOVEL SA MODEL THAT PERMITS LONGITUDINAL ASSESSMENT OF INDIVIDUAL VARIABILITY IN THE DEVELOPMENT OF MOTIVATED AND PERSEVERATIVE DRUG-SEEKING AND A CLINICALLY RELEVANT OPERANT MODEL TO ASSESS COGNITIVE FLEXIBILITY (STRATEGY SHIFTING) TO IDENTIFY CHANGES FROM SYNAPSE TO FUNCTION TO BEHAVIOR. AIM1 OF THIS PROPOSAL WILL FURTHER INVESTIGATE EXPOSURE DEPENDENT CHANGES WITHIN PRL-CORE CIRCUITS USING PATHWAY-SPECIFIC EX VIVO ELECTROPHYSIOLOGY. INTERSECTIONAL CHEMOGENETIC MANIPULATIONS WILL ASSESS THE CONTRIBUTION OF THIS CIRCUIT FLEXIBILITY AND DRUG SEEKING AND WHETHER DYSFUNCTION UNDERLIES DEFICITS IN FLEXIBILITY AND CHANGES IN PERSEVERANCE/MOTIVATION. AIM2 WILL USE CA2+ IMAGING TO EXAMINE THE CORRESPONDING DISRUPTIVE EFFECTS OF OPIOID PLASTICITY ON PRL-CORE INFORMATION PROCESSING ASSOCIATED WITH DECISION MAKING AND MEASURE LONGITUDINAL CHANGES IN BASAL STATES AND ACTIVITY ENCODING OPIOID TAKING WITH PROGRESSIVE SA. AIM 3 WILL IDENTIFY SYNAPTIC MODIFICATIONS AT PRL-CORE D1- AND D2- MSN SYNAPSES. CONTRALATERAL CHEMOGENETIC TARGETING WILL DETERMINE CONTRIBUTIONS OF PRL-TO-D1-MSN AND PRL- TO-D2-MSN CIRCUITS IN FLEXIBILITY AND LONGITUDINALLY DETERMINE IF CONTRIBUTIONS TO OPIOID TAKING AND BEHAVIOR SHIFTS OVER TIME. PROPOSED STUDIES WILL PROVIDE NOVEL INSIGHT INTO PROGRESSIVELY DEVELOPING CHANGES IN FUNCTION AND BEHAVIOR, WITH THE POTENTIAL TO INFORM MORE TAILORED TREATMENTS BASED ON SEX AND STAGE OF ADDICTION.

ADVANCING FUNCTIONAL ASSESSMENT OF PEDIATRIC FLATFOOT USING WEIGHTBEARING CT - PROJECT SUMMARY THE FOOT IS A COMPLEX STRUCTURE AND SEVERE DEFORMITIES SUCH AS FLATFEET ARE COMMON AMONG CHILDREN, PARTICULARLY THOSE WITH NEUROLOGIC DISORDERS. TREATMENT OF PEDIATRIC FOOT DEFORMITIES IS CURRENTLY LIMITED BY THE INADEQUATE VISUALIZATION PROVIDED BY STANDARD OF CARE 2D RADIOGRAPHS. THESE RADIOGRAPHIC IMAGES ARE LIMITED TO A SINGLE POINT OF VIEW AND FLATTEN IRREGULARLY SHAPED, 3D FOOT BONES INTO 2D OBJECTS. WEIGHTBEARING COMPUTED TOMOGRAPHY (WBCT) IS AN EMERGING TECHNOLOGY THAT ALLOWS FOR FULL VOLUMETRIC ASSESSMENT OF THE FOOT IN WEIGHTBEARING WITH EQUIVALENT OR LESS RADIATION EXPOSURE THAN A CURRENT STANDARD OF CARE FOOT & ANKLE RADIOGRAPH SERIES. INTERNATIONAL EXPERTS HAVE CALLED WBCT A “GAME-CHANGER” DUE TO ITS ABILITY TO OBSERVE PATHOLOGIES IN THE ADULT FOOT PREVIOUSLY NOT ABLE TO BE SEEN INCLUDING SUBTALAR JOINT ALIGNMENT, IMPINGEMENT, AND SUBLUXATION WHICH HAVE IMPORTANT CLINICAL IMPLICATIONS. WBCT HAS ADVANCED KNOWLEDGE OF FOOT AND ANKLE MORPHOLOGY WITH A CONSENSUS FOR ITS USE IN ADULT SYMPTOMATIC PES PLANUS/FLATFEET. DESPITE THE SUCCESSES AND CHANGES IN CLINICAL PARADIGMS RESULTING FROM THE TECHNOLOGICAL ADVANCES OF WBCT, ITS APPLICATIONS HAVE BEEN LIMITED TO ADULT FOOT DEFORMITY AND HAVE NOT BEEN TRANSLATED TO PEDIATRIC POPULATIONS, CREATING A LARGE KNOWLEDGE GAP IN OUR UNDERSTANDING OF FOOT PATHOLOGIES THROUGHOUT GROWTH AND DEVELOPMENT. PLANUS DEFORMITY, WITH REPORTED INCIDENCE AS HIGH AS 44% OF CHILDREN, IS OF PARTICULAR INTEREST DUE TO LACK OF UNDERSTANDING IN STRUCTURAL CHANGES THAT MAY CAUSE PAIN AND DYSFUNCTION LATER IN YOUNG ADULTHOOD. OUR OVERALL OBJECTIVE FOR THE CURRENT MULTISITE PROJECT IS TO COMPLETE A COMPREHENSIVE STUDY OF THE 3D STRUCTURE AND FUNCTION OF PEDIATRIC PLANUS FEET. TO ACCOMPLISH THIS GOAL, WE PROPOSE TWO SPECIFIC AIMS. SPECIFIC AIM 1 TO COMPLETE A CROSS SECTIONAL EVALUATION OF CHILDREN WITH PLANUS FOOT DEFORMITY USING CURRENT STANDARD-OF-CARE 2D RADIOGRAPHIC ANALYSIS AND EQUIVALENT METRICS OBTAINED FROM 3D WBCT. WE WILL ALSO EXPLORE RELATIONSHIPS BETWEEN JOINT FORM AND FUNCTION. SPECIFIC AIM 2 IS TO COMPLETE A LONGITUDINAL EVALUATION OF CHILDREN WITH PLANUS FOOT DEFORMITY TO DETERMINE RADIOGRAPHIC AND FUNCTIONAL METRICS THAT ARE PREDICTIVE OF WORSENING SYMPTOMS AND ARCH COLLAPSE. THE LONG-TERM FOCUS OF THIS PROJECT IS TO PROVIDE A MORE COMPREHENSIVE ASSESSMENT OF THE 3D STRUCTURE AND FUNCTION OF PLANUS FOOT STRUCTURE IN CHILDREN. THIS WILL ALLOW FOR ACCURATE CLINICAL ASSESSMENT AND TARGETED TREATMENT PLANNING THROUGH IMPROVED RADIOGRAPHIC MEASUREMENTS AND THE ABILITY TO DIAGNOSE THESE COMPLEX FOOT DISORDERS IN DEVELOPING CHILDREN.

THIS CACREP-ACCREDITED PROGRAM HAS PROVIDED EVIDENCE-BASED AND EMERGING BEST PRACTICES IN CLINICAL MENTAL HEALTH SERVICES FOR CHILDREN, ADOLESCENTS, AND THEIR FAMILIES SINCE 2013. THE PROPOSED FOUR-YEAR PROJECT ADDRESSES THE BHWET PROGRAM OBJECTIVES BY: (A) INCREASING THE NUMBER OF NEW OR EXPANDED COMMUNITY PARTNERSHIPS WITH EXPERIENTIAL TRAINING SITES IN HIGH-NEED AND HIGH-DEMAND AREAS; (B) PROMOTING COLLABORATIVE TRAINING BY USING TEAM-BASED MODELS OF CARE TO INTEGRATE BEHAVIORAL HEALTH CARE INTO INTERPROFESSIONAL PRIMARY CARE SETTINGS; (C) RECRUITING A DIVERSE WORKFORCE INTERESTED IN WORKING WITH CHILDREN, ADOLESCENTS, AND YOUNG ADULTS; AND (D) RECRUITING, DEVELOPING, AND EXPANDING THE CAPACITY TO TRAIN CLINICAL SUPERVISORS TO SUPPORT AND MENTOR BEHAVIORAL HEALTH TRAINEES. THE UNIQUE CONTRIBUTION OF THIS GRADUATE PROGRAM WILL BE TO COMBINE A FIELD-EMBEDDED INTRODUCTION TO AMBITIOUS STEM INSTRUCTION WITH EXTENSIVE PEDAGOGICAL CONTENT KNOWLEDGE IN COMPUTER SCIENCE EDUCATION, PRODUCING HIGHLY QUALIFIED MATHEMATICS AND SCIENCE TEACHERS WHO CAN ALSO TEACH COMPUTER SCIENCE COURSES IN HIGH-NEED SCHOOLS. GRADUATES OF THE PROGRAM WILL COMPLETE REQUIREMENTS FOR MIDDLE AND SECONDARY LEVEL TEACHER CERTIFICATION IN MATHEMATICS OR SCIENCE AND REQUIREMENTS FOR COMPUTER SCIENCE EDUCATION CERTIFICATION. GRADUATES WILL THEN SERVE AS TEACHERS IN HIGH-NEED SCHOOLS, EQUIPPED WITH PEDAGOGICAL SKILLS AND QUALIFICATIONS IN COMPUTER SCIENCE AND MATHEMATICS OR SCIENCE, AND PREPARED WITH A HIGHLY DESIRABLE COMBINATION OF SKILLS TO SERVE IN HIGH-NEED ENVIRONMENTS. THIS PROJECT AT MARQUETTE UNIVERSITY INCLUDES PARTNERSHIPS WITH MILWAUKEE PUBLIC SCHOOLS, MILWAUKEE AREA CHARTER SCHOOLS, THE SHOREWOOD SCHOOL DISTRICT, AND SCHOOLS IN THE ARCHDIOCESE OF MILWAUKEE. PROJECT GOALS INCLUDE RECRUITING 24 HIGHLY QUALIFIED STEM PROFESSIONALS AND PREPARING THEM TO ENTER HIGH-NEED SCHOOL ENVIRONMENTS WITH A MASTER OF EDUCATION AND TEACHER CERTIFICATIONS. THE PROJECT WILL DEVELOP A MULTIMODAL (IN PERSON AND ONLINE) PATHWAY THAT WILL LEAD TO COMPUTER SCIENCE TEACHER CERTIFICATION ALONGSIDE MATHEMATICS OR SCIENCE TEACHER CERTIFICATION. THE PROJECT IS THEORETICALLY GROUNDED IN PRACTICE-BASED TEACHER PREPARATION, AMBITIOUS INSTRUCTION, AND PEDAGOGICAL CONTENT KNOWLEDGE FOR COMPUTER SCIENCE INSTRUCTION. IN KEEPING WITH CURRENT REFORMS IN K-12 STEM EDUCATION AND TEACHER EDUCATION, AMBITIOUS INSTRUCTION SUPPORTS ALL STUDENTS, REGARDLESS OF RACE, SOCIOECONOMIC STATUS, ETHNICITY, GENDER, AND IDENTITY, IN RIGOROUS AND CONCEPTUAL LEARNING OF STEM CONTENT. ADDITIONALLY, THE PROJECT WILL SUPPORT GRADUATES IN THEIR EARLY PROFESSIONAL PRACTICE. RESEARCH QUESTIONS WILL ADDRESS EFFECTIVE PEDAGOGIES FOR DEVELOPING AMBITIOUS TEACHING PRACTICES IN MATHEMATICS, SCIENCE, AND COMPUTER SCIENCE, PARTICULARLY FOCUSED ON PEDAGOGIES OF ENACTMENT AND SCHOLAR DEVELOPMENT OF AMBITIOUS TEACHING ACROSS STEM FIELDS THROUGH QUALITATIVE AND OBSERVATIONAL RESEARCH METHODS. THE PROJECT WILL CONTRIBUTE TO THE EMPIRICAL KNOWLEDGE BASE BY ADVANCING INTERSECTIONAL RESEARCH ON COMPUTER SCIENCE EDUCATION, AMBITIOUS STEM INSTRUCTION, AND PRACTICE-BASED TEACHER PREPARATION. THIS TRACK 1: SCHOLARSHIPS AND STIPENDS PROJECT IS SUPPORTED THROUGH THE ROBERT NOYCE TEACHER SCHOLARSHIP PROGRAM (NOYCE). THE NOYCE PROGRAM SUPPORTS TALENTED STEM UNDERGRADUATE MAJORS AND PROFESSIONALS TO BECOME EFFECTIVE K-12 STEM TEACHERS AND EXPERIENCED, EXEMPLARY K-12 TEACHERS TO BECOME STEM MASTER TEACHERS IN HIGH-NEED SCHOOL DISTRICTS. MU POSTAWARD OPTIMIZATION FOR STRATEGIC TRANSFORMATION (MU POST) -TO PROVIDE FACULTY MORE TIME TO DEVOTE TO RESEARCH ACTIVITIES, THE MARQUETTE UNIVERSITY POST-AWARD OPTIMIZATION FOR STRATEGIC TRANSFORMATION (MU-POST) PROJECT FOCUSES ON PROCESS IMPROVEMENTS THAT REDUCE THE ADMINISTRATIVE BURDEN ASSOCIATED WITH SPONSORED AWARD MANAGEMENT. SMALL AND MID-SIZED UNIVERSITIES OFTEN LACK ROBUST RESEARCH SUPPORT AND SERVICE INFRASTRUCTURE. WITH DEFINED RESOURCES AND EFFECTIVE PROCESSES SUPPORTING THEM, FACULTY CAN DEDICATE MORE TIME TO THEIR RESEARCH, COMPLETE MILESTONES ON SCHEDULE, COLLABORATE WITH COLLEAGUES, MENTOR STUDENTS, AND INCREASE THEIR PUBLICATION RATE. REDUCING ADMINISTRATIVE BURDEN THROUGH PROCESS IMPROVEMENTS LESSENS THE ORGANIZATIONAL COMPLEXITIES THAT COME WITH HAVING A GRANT AND ULTIMATELY FOSTERS GROWTH IN THE NATION?S RESEARCH CAPACITY. MU-POST DEVELOPS STRATEGIES, PROCESSES, AND MODELS THAT CAN BE UTILIZED BY SMALL TO MID-SIZE INSTITUTIONS TO GROW CAPACITY, ACCESS, AND VISIBILITY WITHIN THE POST-AWARD SECTOR. THE PROJECT HAS TWO MAJOR OBJECTIVES: 1) TO TEST THE IMPLEMENTATION OF POST-AWARD ADVOCATES, ROLES THAT BRIDGE THE GAP BETWEEN A CENTRAL SPONSORED PROGRAMS OFFICE, FACULTY MEMBERS, AND THEIR DEPARTMENTS AND OTHER UNITS, AND 2) TO DEVELOP AND IMPLEMENT BROAD CHANGES IN POST-AWARD PROCESSES AND SYSTEMS TO ALLOW FOR SCALABILITY, EFFICIENCY, AND TRANSPARENCY. THE PROJECT?S OUTCOMES ARE THE DOCUMENTATION OF MODEL PROCESSES AND USE CASES AND THE CREATION OF TOOLS AND OTHER RESOURCES THAT CAN BE USED BY EMERGING INSTITUTIONS AS A GUIDE TO STRENGTHEN THE SERVICES THEY OFFER TO HELP RESEARCHERS MORE EFFICIENTLY MANAGE THEIR AWARDS, ALLOWING THEM TIME TO FOCUS ON, AND PARTICIPATE IN, RESEARCH ACTIVITIES. THIS AWARD REFLECTS NSF'S STATUTORY MISSION AND HAS BEEN DEEMED WORTHY OF SUPPORT THROUGH EVALUATION USING THE FOUNDATION'S INTELLECTUAL MERIT AND BROADER IMPACTS REVIEW CRITERIA.- SUBAWARDS ARE NOT PLANNED FOR THIS AWARD.

SEX CHROMOSOME EVOLUTION IN LIZARDS AND SNAKES -SEX CHROMOSOMES PLAY AN IMPORTANT ROLE IN THE EMBRYONIC DEVELOPMENT OF MANY ANIMAL SPECIES BY CONTROLLING AN INDIVIDUAL'S SEX. HOWEVER, THE SEX CHROMOSOMES OF MOST ANIMALS REMAIN UNIDENTIFIED LIMITING OUR UNDERSTANDING OF BROAD PATTERNS OF SEX CHROMOSOME EVOLUTION. THIS PROJECT WILL ADVANCE THE STUDY OF SEX CHROMOSOME EVOLUTION BY INVESTIGATING LIZARDS AND SNAKES. THESE REPTILES HAVE A VARIETY OF SEX-DETERMINING SYSTEMS WITH NUMEROUS EVOLUTIONARY TRANSITIONS AMONG THEM. EACH OF THESE INDEPENDENTLY DERIVED SYSTEMS CAN ACT AS A REPLICATE EVOLUTIONARY ?EXPERIMENT? TO TEST THE GENERAL PRINCIPLES AND PROCESSES THAT GOVERN SEX CHROMOSOME EVOLUTION. THIS RESEARCH WILL FILL GAPS IN OUR KNOWLEDGE BY IDENTIFYING SEX CHROMOSOMES IN THESE UNDER-STUDIED REPTILES AND THEN USE THESE DATA TO CHARACTERIZE SOME GENERAL PROPERTIES OF SEX CHROMOSOME EVOLUTION. THE RESEARCH WILL TRAIN A POSTDOCTORAL RESEARCHER AND STUDENTS, INCLUDING PHD STUDENTS AND UNDERGRADUATES, IN GENOMICS AND BIOINFORMATICS, SKILLS THAT ARE IN HIGH DEMAND FOR CAREERS IN AGRICULTURE, BIOTECHNOLOGY, AND HEALTH CARE. RESEARCHERS WILL DEVELOP GENETIC SEX TESTS FOR SEVERAL REPTILE SPECIES. SUCH TESTS ARE IMPORTANT FOR THE MANAGEMENT AND BREEDING OF ENDANGERED SPECIES AND SEXING EMBRYONIC MATERIAL IN DEVELOPMENTAL RESEARCH. RESEARCHERS WILL ENGAGE IN OUTREACH TO PET REPTILE HOBBYISTS AND THE GENERAL PUBLIC ABOUT GENETICS, EVOLUTION, AND SEX CHROMOSOMES VIA A MUSEUM EXHIBIT AND A BOOTH AT PET REPTILE TRADE SHOWS. COMPARATIVE STUDIES OF SEX CHROMOSOMES IN ANIMAL GROUPS WITH REPEATED TRANSITIONS AMONG SEX-DETERMINING MECHANISMS, LIKE THE LIZARDS AND SNAKES, CAN HELP ANSWER LONGSTANDING QUESTIONS ABOUT SEX CHROMOSOME EVOLUTION. THIS RESEARCH WILL INTEGRATE COMPARATIVE GENOMICS, TRANSCRIPTOMICS, AND PHYLOGENETICS TO ENHANCE THE UTILITY OF LIZARDS AND SNAKES AS A MODEL CLADE TO INVESTIGATE SEX CHROMOSOME EVOLUTION AND ADDRESS THE FOLLOWING QUESTIONS: 1) ARE SEX CHROMOSOMES AN EVOLUTIONARY TRAP? 2) ARE SOME CHROMOSOMES PARTICULARLY GOOD AT BEING SEX CHROMOSOMES? 3) IS SEX CHROMOSOME DOSAGE COMPENSATION MORE LIKELY TO EVOLVE IN XX/XY VS. ZZ/ZW TAXA? 4) WHAT IS THE RELATIVE ROLE OF REARRANGEMENTS VS. SEX-BIASED RECOMBINATION IN SUPPRESSING RECOMBINATION BETWEEN THE SEX CHROMOSOMES? UPON COMPLETION, THIS PROJECT WILL ENABLE SUBSEQUENT INVESTIGATIONS INTO THE MECHANISMS THAT DRIVE SEX CHROMOSOME EVOLUTION IN LIZARDS AND SNAKES AND ESTABLISH A FRAMEWORK FOR COMPARATIVE ANALYSES OF SEX CHROMOSOME EVOLUTION ACROSS THE TREE OF LIFE. DATA COLLECTED FOR THIS PROJECT WILL HAVE BROAD UTILITY TO OTHER SCIENTISTS, INCLUDING NEW, HIGH QUALITY GENOME ASSEMBLIES FOR MULTIPLE LIZARD AND SNAKE SPECIES. THIS AWARD REFLECTS NSF'S STATUTORY MISSION AND HAS BEEN DEEMED WORTHY OF SUPPORT THROUGH EVALUATION USING THE FOUNDATION'S INTELLECTUAL MERIT AND BROADER IMPACTS REVIEW CRITERIA.

COLLABORATIVE RESEARCH: EDGE FGT: DEVELOPMENT OF FUNCTIONAL GENETIC TOOLS IN GECKOS - FUNCTIONAL GENOMIC TOOLS, SUCH AS CRISPR/CAS GENOME EDITING, ARE CRITICAL FOR INVESTIGATING THE BIOLOGY OF GENES AND HAVE TRANSFORMED OUR UNDERSTANDING OF GENOTYPE-TO-PHENOTYPE RELATIONSHIPS. HOWEVER, THE LIMITED NUMBER OF SPECIES WITH GENOME MODIFICATION METHODS PRESENTS A MAJOR BOTTLENECK IN IMPLEMENTING COMPREHENSIVE, COMPARATIVE STUDIES OF GENE FUNCTION. FOR INSTANCE, SQUAMATES (LIZARD AND SNAKES) INCLUDE MORE THAN 11,000 DESCRIBED SPECIES AND COMPRISE 40% OF ALL AMNIOTE SPECIES. YET BASIC GENE EDITING METHODS ARE ONLY AVAILABLE IN A SINGLE SPECIES, THE BROWN ANOLE LIZARD (ANOLIS SAGREI). HERE, WE PROPOSE TO: 1) ESTABLISH CRISPR/CAS GENE EDITING IN A SECOND LIZARD GROUP, THE GECKOS; 2) DEVELOP ADVANCED GENE EDITING METHODS FOR THE INTRODUCTION OF PRECISE SEQUENCE SUBSTITUTIONS, INSERTIONS, AND DELETIONS IN LIZARDS (IN BOTH ANOLIS AND GECKOS); AND 3) DISSEMINATE LIZARD GENE EDITING TECHNIQUES THROUGH TRAINING MATERIALS AND WORKSHOPS AND THE PRODUCTION OF GENOME-EDITED LIZARDS FOR RESEARCHERS. THE DEVELOPMENT OF GENE EDITING TOOLS IN GECKOS WILL INCREASE THE NUMBER OF SQUAMATE MODELS AVAILABLE FOR FUNCTIONAL GENOMIC WORK AND FACILITATE THE GENERALIZATION OF GENE FUNCTION STUDIES ACROSS SQUAMATES AND AMNIOTES. FURTHERMORE, GROWING THE FUNCTIONAL GENOMIC TOOLKIT IN LIZARDS TO INCLUDE MORE PRECISE GENE EDITING METHODS WILL EXPAND THE SCOPE OF QUESTIONS THAT CAN BE ADDRESSED IN THIS PHENOTYPICALLY DIVERSE GROUP.

BUILT IN 2002, MUSOD’S MAIN CAMPUS CLINIC, LOCATED IN MILWAUKEE, ANNUALLY SERVES NEARLY 17,000 PATIENTS FROM 53 OF WISCONSIN’S 72 COUNTIES. MUSOD PROVIDES ORAL HEALTH SERVICES THROUGH GENERAL DENTISTRY AND VARIOUS SPECIALTY CLINICS INCLUDING ADVANCED CLINICAL CARE (ACC), ENDODONTICS, PEDIATRIC , PERIODONTICS, PROSTHODONTICS, ORAL SURGERY, ORTHODONTICS, AND RADIOLOGY. NEARLY TWO-THIRDS OF THE PATIENTS ARE FROM MILWAUKEE COUNTY, MANY FROM HIGH-POVERTY NEIGHBORHOODS. MUSOD IS AMONG WISCONSIN’S LARGEST DENTAL MEDICAID (MA) PROVIDERS, ANNUALLY SERVING NEARLY 7,000 MA PATIENTS. MUSOD’S SPECIALTY CLINICS ARE NOTABLE FOR DELIVERING CARE CRITICAL TO ORAL HEALTH AND OVERALL HEALTH TO MANY MA AND UNDERSERVED PATIENTS. THIS REQUEST SUPPORTS DENTAL OPERATORY EQUIPMENT AS PART OF AN OVERALL MUSOD REVITALIZATION OF THE MAIN CLINIC’S FOUR ORIGINAL GENERAL PRACTICE DENTAL CLINICS, ALL SPECIALTY CLINICS, AND CENTRAL STERILIZATION. – MUSOD’S PROPOSED HRSA CONSTRUCTION GRANT REQUEST WILL UTILIZE ALL FUNDING TO PURCHASE 47 DENTAL OPERATORIES AND EQUIPMENT TO EDUCATE AND TRAIN PROSPECTIVE DENTISTS AND RESIDENTS TO PROVIDE CARE TO UNDERSERVED PATIENTS IN OUR ADVANCED CARE, PEDIATRIC, PROSTHODONTICS CLINICS AND PART OF THE GENERAL PRACTICE CLINICS. TYPICALLY, ONLY 10% OF DENTISTS CARE FOR PATIENTS WITH COGNITIVE/MEDICAL/PHYSICAL DISABILITIES AND THE SERVICES NEEDED ARE OFTEN NOT COVERED BY PRIVATE OR STATE INSURANCE PLANS. ACCORDING TO THE 2010 U.S. CENSUS, , 22% OF WISCONSIN’S CHILDREN HAVE A DISABILITY, MANY OF WHOM LACK REGULAR ORAL HEALTH CARE OPTIONS. THESE HRSA FUNDS TO SUPPORT UPGRADING DENTAL OPERATORIES WILL ALLOW MUSOD TO SIGNIFICANTLY IMPACT CARE FOR THIS POPULATION, WHICH HAS HIGHER RISK OF UNTREATED DENTAL CARIES AND PERIODONTAL DISEASE DUE TO PRESCRIBED MEDICATIONS, FEEDING REGIMENS, AND POOR DENTAL HYGIENE PRACTICES. ADDITIONALLY, THESE OPERATORIES ARE HEAVILY USED TO PROVIDE PATIENT CARE TO UNDERSERVED COMMUNITIES ACROSS THE WHOLE STATE OF WISCONSIN. MUSOD’S CONSTRUCTION GRANT REQUEST ALIGNS WITH HRSA’S 2019-2022 STRATEGIC PLAN GOALS 1-3: 1) IMPROVE ACCESS TO QUALITY HEALTH SERVICES; 2) FOSTER A HEALTH CARE WORKFORCE ABLE TO ADDRESS CURRENT AND EMERGING NEEDS; 3) ACHIEVE HEALTH EQUITY AND ENHANCE POPULATION HEALTH. MUSOD WILL DO THE FOLLOWING BECAUSE OF ACQUIRING THIS EQUIPMENT: - PROVIDE GREATER DIRECT ACCESS TO CARE FOR ADULTS WITH SPECIAL NEEDS TO IMPROVE THEIR ORAL HEALTH AND POSSIBLY PREVENT CERTAIN MEDICAL ISSUES AND TO REDUCE THE ORAL HEALTH DISPARITIES IN SPECIAL NEEDS PATIENTS. - INCREASE THE NUMBER OF PROSPECTIVE DENTISTS WHO ARE EDUCATED AND TRAINED TO PROVIDE CARE TO SPECIAL NEEDS DENTAL PATIENTS AND THE UNDERSERVED PATIENT POPULATIONS. - PROVIDE ONGOING PREVENTION AND INCREASE THE AMOUNT OF DENTAL CARE AVAILABLE FOR SPECIAL NEEDS PATIENTS TO MINIMIZE THE NEED FOR SEDATION AND GENERAL ANESTHESIA, THEREBY FURTHER REDUCING THE COSTS AND RISKS OF CARE. - ESTABLISH MUSOD AND EVENTUALLY OTHER DENTAL PROVIDERS AS A “DENTAL HOME” FOR SPECIAL NEEDS PATIENTS. - PROVIDE DIRECT ACCESS TO CARE FOR UNDERSERVED CHILDREN AND ADULTS IN MUSOD’S PEDIATRIC CLINIC AND GENERAL DENTISTRY CLINICS.

MECHANISMS OF FATIGABILITY AND THE PROTECTIVE EFFECTS OF EXERCISE IN PEOPLE WITH DIABETES - ABSTRACT PRE-DIABETES (PRE-D) IS CHARACTERIZED BY ELEVATED GLYCATED HEMOGLOBIN AND PLASMA GLUCOSE AND IS A CLINICAL PRECURSOR TO TYPE 2 DIABETES MELLITUS (T2D). PRE-D CURRENTLY AFFECTS ~90 MILLION AMERICANS. BOTH PRE-D AND T2D ARE HIGHLY ASSOCIATED WITH CARDIOVASCULAR DISEASE AND AMONG THE TOP FIVE CAUSES OF MORTALITY WORLDWIDE. EXERCISE IS THE CORNERSTONE OF MANAGEMENT AND IS MOST EFFICACIOUS DURING THE PRE-D STAGE WHEN GLYCEMIA IS BELOW THE DIABETIC THRESHOLD. HOWEVER, EXCESSIVE FATIGABILITY DURING EXERCISE (I.E., EXERCISE INDUCED REDUCTIONS IN FORCE OR POWER OF THE LIMB MUSCLES) LIMITS EXERCISE PERFORMANCE IN PEOPLE WITH PRE-D. OUR LABORATORY DEMONSTRATED THAT (1) ACROSS THE DIABETIC SPECTRUM, PEOPLE WITH PRE-D AND T2D HAVE GREATER FATIGABILITY OF LIMB MUSCLES THAN CONTROLS DUE TO MECHANISMS WITHIN THE MUSCLE, AND (2) FATIGABILITY IN PEOPLE WITH T2D WAS ASSOCIATED WITH A REDUCED BLOOD FLOW TO THE EXERCISING MUSCLE. IT IS UNKNOWN, HOWEVER, IF PEOPLE WITH PRE-D HAVE IMPAIRED VASCULAR FUNCTION AND OXYGEN DELIVERY THAT LEADS TO AN INCREASED FATIGABILITY. OUR CENTRAL HYPOTHESIS IS THAT IMPAIRED VASCULAR FUNCTION IMPEDES BLOOD FLOW AND BLUNTS SUBSEQUENT OXYGEN DELIVERY TO SKELETAL MUSCLE DURING EXERCISE, RESULTING IN EXCESSIVE FATIGABILITY OF LIMB MUSCLES IN PEOPLE WITH PRE-D. A UNIQUE AND TRANSLATIONAL ASPECT OF THIS PROPOSAL IS THE QUANTIFICATION OF THE VASCULAR RESPONSES (MACRO- AND MICRO-VASCULATURE) TO FATIGUING EXERCISE AND EXERCISING TRAINING AT DIFFERENT SITES ALONG THE VASCULAR TREE, INCLUDING, IN FEED ARTERIES (DOPPLER ULTRASONOGRAPHY), ISOLATED SKELETAL MUSCLE ARTERIOLES (EXTRACTED FROM MUSCLE BIOPSY), CAPILLARY PERFUSION (NEAR INFRARED SPECTROSCOPY, NIRS) AND CAPILLARY DENSITY (FROM MUSCLE BIOPSIES). AIM 1 WILL DETERMINE IF VASCULAR DYSFUNCTION IS A MECHANISM FOR EXCESSIVE FATIGABILITY IN PEOPLE WITH PRE-D. AIM 1.1 WILL COMPARE LEG BLOOD FLOW AND SKELETAL MUSCLE OXYGENATION IN RESPONSE TO DYNAMIC FATIGUING EXERCISE BETWEEN PEOPLE WITH PRE-D, HEALTHY CONTROLS AND T2D. GROUPS WILL BE MATCHED FOR AGE, SEX, BODY MASS INDEX AND PHYSICAL ACTIVITY LEVELS TO DETERMINE DISEASE- RELATED VASCULAR FUNCTION AND FATIGABILITY RATHER THAN INACTIVITY-RELATED CHANGES ACROSS THE DIABETIC SPECTRUM. SKELETAL MUSCLE BLOOD FLOW THROUGH THE FEMORAL ARTERY WILL BE QUANTIFIED WITH ULTRASONOGRAPHY AND SKELETAL MUSCLE OXYGENATION WITH NIRS DURING A DYNAMIC FATIGUING KNEE EXTENSION EXERCISE. AIM 1.2 WILL DETERMINE ENDOTHELIAL VASCULAR FUNCTION AT MACRO- AND MICRO-VASCULAR LEVELS IN PEOPLE WITH PRE-D AND T2D. FLOW-MEDIATED DILATION WILL BE ASSESSED IN VIVO IN THE FEMORAL ARTERY USING ULTRASONOGRAPHY AND IN ISOLATED ARTERIOLES EXTRACTED FROM THE VASTUS LATERALIS MUSCLE BIOPSIES. AIM 2 (A CLINICAL TRIAL) WILL ASSESS THE VASCULAR MECHANISMS FOR IMPROVED BLOOD FLOW AND FATIGABILITY IN RESPONSE TO DYNAMIC EXERCISE TRAINING WITH BLOOD FLOW RESTRICTION IN PEOPLE WITH DIABETES. PEOPLE WITH PRE-D AND T2D WILL PERFORM 8 WEEKS OF UNILATERAL RESISTANCE TRAINING IN WHICH ONE LEG IS EXERCISED WITH FREELY PERFUSED CONDITIONS AND THE OTHER LEG WITH BLOOD FLOW RESTRICTION. THUS, BLOOD FLOW RESTRICTION AND RESISTANCE TRAINING WILL BE USED AS A PROBE TO FURTHER UNDERSTAND THE MECHANISMS OF FATIGABILITY ALONG THE VASCULAR TREE IN PEOPLE WITH PRE-D AND T2D, AND TEST TRAINING STRATEGIES TO IMPROVE FATIGABILITY IN THIS POPULATION.

MARQUETTE RESEARCH FACULTY ARE HIGHLY COLLABORATIVE, INTERDISCIPLINARY, PRE- CLINICAL AND CLINICAL INVESTIGATORS, AND MAINTAIN ROBUST RESEARCH PRODUCTIVITY. GOALS OF THIS PROPOSAL INCLUDE: 1) FOCUSED RECRUITMENT OF UNDER-REPRESENTED STUDENT POPULATIONS, FIRST-GENERATION COLLEGE STUDENTS, AND WOMEN; 2) THE CREATION OF A DIDACTIC-BASED RESEARCH UNDERGRADUATE COURSE AND CONTINUOUS SEMINAR SERIES; 3) PLACING STUDENTS INTO ACTIVE RESEARCH LABS, 4) INCREASING A SENSE OF BELONGING AND PERSISTENCE, AND 5) FACILITATING GRADUATE SCHOOL APPLICATION SUBMISSION. COMPLEMENTING THE TWO YEARS OF RIGOROUS AND ORIGINAL RESEARCH EXPERIENCE FOR U-RISE PARTICIPANTS, U-RISE AT MARQUETTE HAS CREATED A THREE-CREDIT PATH TO RESEARCH COURSE TO INTRODUCE POTENTIAL STUDENT RESEARCHERS TO SKILLS AND PRACTICES OF CAREER SCIENTISTS AS WELL AS A CONTINUOUS U- RISE SEMINAR SERIES (FOUR SEMESTERS) TO ENSURE ALL PARTICIPANTS ARE ENGAGED WITH AND SUPPORTED BY THE PROGRAM. SEVERAL ALGORITHMS EXIST FOR THE CALCULATIONS OF MOLECULAR ELECTRONIC STRUCTURE (CALLED QUANTUM CHEMISTRY) USING EITHER GATE-BASED QUANTUM COMPUTERS WITH ONLY A FEW TENS OF QUBITS, SUCH AS SYCAMORE, IBM Q-MACHINES, OR USING QUANTUM ANNEALERS WITH A LARGER NUMBER OF QUBITS, SUCH AS THE D-WAVE SYSTEMS. ANOTHER ESSENTIAL COMPONENT OF CHEMICAL MODELING IS THE PROPAGATION OF THE EQUATIONS OF MOTION FOR CONSTITUENT ATOMS IN TIME AND SPACE (E.G., ALONG THE REACTION PATH), CALLED MOLECULAR DYNAMICS. MOLECULAR DYNAMICS SIMULATIONS PLAY A PIVOTAL ROLE NOT ONLY IN GUIDING AND ANALYZING EXPERIMENTS, BUT ALSO AS AN INTERDISCIPLINARY COMPUTATIONAL TOOL CAPABLE OF REACHING FAR BEYOND EXPERIMENTAL CONDITIONS IN A VARIETY OF RESEARCH TOPICS. THE PROJECT TEAM IS AMONG THE FIRST TO RUN MOLECULAR DYNAMICS SIMULATIONS ON ACTUAL QUANTUM HARDWARE -- THE DWAVE QUANTUM ANNEALER, TRYING TO HARNESS THE POWER OF QUANTUM COMPUTING FOR THE BENEFIT OF MOLECULAR DYNAMICS SIMULATIONS. THE RESEARCHERS AT MARQUETTE UNIVERSITY AND AT LOS ALAMOS NATIONAL LABORATORY WILL JOINTLY SUPERVISE UNDERGRADUATE AND GRADUATE STUDENTS AND ONE TEACHING POSTDOC TO CONDUCT RESEARCH ON QUANTUM DYNAMICS. DURING THE SUMMER, THESE STUDENTS ARE ROUTED TO LOS ALAMOS FOR INTERNSHIPS, WHERE THEY ATTEND THE QUANTUM COMPUTING SUMMER SCHOOL, INTERACT WITH OTHER SCIENTISTS AT THE INFORMATION SCIENCE AND TECHNOLOGY INSTITUTE, AND ARE IMMERSED IN THE RESEARCH ENVIRONMENT OF THE NATIONAL LAB. FOR SOCIAL ANIMALS, INDIVIDUALS OFTEN WORK TOGETHER IN A GROUP TO MANAGE THESE FLUCTUATIONS MUCH MORE EFFECTIVELY, SUCH AS THROUGH HUDDLING, YET LITTLE IS KNOWN ABOUT EXACTLY HOW ANIMALS ACCOMPLISH THIS. HONEYBEES, FOR EXAMPLE, ARE AN EXCELLENT MODEL SYSTEM TO STUDY SOCIAL TEMPERATURE REGULATION BECAUSE THEY ENGAGE IN A COOLING BEHAVIOR CALLED FANNING, IN WHICH A GROUP OF BEES FAN THEIR WINGS TO CIRCULATE AIR IN THE COLONY. IF THEY DO NOT COOL EFFECTIVELY, THE COLONY CAN DIE. THIS PROJECT WILL IDENTIFY THE MECHANISMS THAT HONEYBEES USE TO SENSE ENVIRONMENTAL FLUCTUATIONS, HOW THEIR BRAINS CHANGE IN RESPONSE TO THIS INFORMATION, HOW THEY COMMUNICATE THIS INFORMATION TO EACH OTHER, AND HOW ENTIRE COLONIES REGULATE TEMPERATURE WHEN INFORMATION CHANGES. PHYSIOLOGICAL AND GENETIC TESTING TECHNIQUES WILL BE USED TO UNDERSTAND BOTH INDIVIDUAL AND COLLECTIVE BEHAVIOR, ESPECIALLY IN CHANGING ENVIRONMENTS. THE PROJECT WILL ENGAGE THE PUBLIC 1) BY DEVELOPING A LOCAL PRISON BEEKEEPING PROGRAM TO PROVIDE TANGIBLE WORKFORCE SKILLS FOR INMATES, 2) BY WORKING WITH THE ROSE PROJECT, WHICH FOCUSES ON INTRODUCING UNDERREPRESENTED MIDDLE SCHOOL STUDENTS AND PARENTS TO STEM RESEARCH, 3) BY PRESENTING AT LOCAL BEEKEEPING AND ECOLOGY MEETINGS, AND 4) BY TRAINING AND SUPPORTING UNDERGRADUATE, GRADUATE, AND POSTDOCTORAL SCIENTISTS IN THESE CUTTING-EDGE RESEARCH AND OUTREACH TECHNIQUES. ORGANISMS MUST BE ABLE TO BEHAVIORALLY AND PHYSIOLOGICALLY MANAGE DAILY AND SEASONAL FLUCTUATIONS IN CLIMATE. SENSORY RECEPTION AND INTEGRATION OF INFORMATION IS CRITICAL TO MAINTAIN HOMEOSTASIS DURING TYPICAL THERMAL SHIFTS ACROSS DAYS OR SEASONS. COLLECTIVE BEHAVIORAL RESPONSES TO CHANGING TEMPERATURES REQUIRE EFFECTIVE RECEPTION AND INTEGRATION OF BOTH SOCIAL AND ECOLOGICAL INFORMATION. ALTHOUGH WE KNOW WHY ANIMALS THERMOREGULATE, IT IS LESS CLEAR HOW ANIMALS INTEGRATE SOCIAL INFORMATION TO BE ABLE TO THERMOREGULATE EFFECTIVELY. HONEYBEES MUST MAINTAIN 34?C IN THEIR COLONY, OR RISK DEATH OF THEIR DEVELOPING BROOD. IN THE SUMMER, HONEYBEES ENGAGE IN A COOLING BEHAVIOR CALLED FANNING, IN WHICH THEY FAN THEIR WINGS TO CIRCULATE AIR TO COOL THE COLONY. FANNER HONEYBEES UTILIZE BOTH INFORMATION FROM THEIR NESTMATES AND INFORMATION FROM THEIR ECOLOGICAL ENVIRONMENT TO COLLECTIVELY THERMOREGULATE. THE FANNING RESPONSE SHIFTS WHEN THE SOCIAL AND THERMAL INFORMATION CHANGES; HOWEVER, THE WAYS IN WHICH INDIVIDUALS OR GROUPS PERCEIVE, INTEGRATE, OR COMMUNICATE ABOUT THESE CHANGES TO BEHAVE COLLECTIVELY IS UNKNOWN. THIS PROPOSAL WILL IDENTIFY THESE MECHANISMS BY ADDRESSING FOUR AIMS: 1) DETERMINE HOW THE SOCIAL INTERACTIONS SHAPE THE FANNING RESPONSE; 2) UNCOVER THE NEURAL GENE NETWORKS ASSOCIATED WITH THE FANNING RESPONSE; 3) DEFINE TRP CHANNEL INVOLVEMENT IN THE FANNING RESPONSE; AND 4) EVALUATE COLONY-LEVEL FANNING RESPONSE IN A CHANGING ENVIRONMENT. THIS PROJECT WILL ADVANCE THE UNDERSTANDING OF HOW LIGHT AFFECTS FUNDAMENTAL BIOLOGY BY DEFINING THE NEUROBIOLOGICAL PATHWAYS USED BY LIGHT TO AFFECT BREATHING. INTEGRATED INTO THE RESEARCH PROGRAM, THIS PROJECT EMPHASIZES THE EDUCATION AND TRAINING OF UNDERGRADUATE STUDENTS IN CREATIVE PROBLEM-SOLVING SKILLS. THESE SKILLS ARE EXPECTED TO FOSTER THE NEXT GENERATION OF STEM INNOVATORS TO PROMOTE SCIENTIFIC DISCOVERY IN AN EVER-CHANGING WORLD. DEFINING THE NEUROBIOLOGICAL CIRCUITS THAT MEDIATE LIGHT?S EFFECTS ON BREATHING CAN BETTER EQUIP SOCIETY TO PREDICT AND RESPOND TO THE PHYSIOLOGICAL EFFECTS OF LIGHT POLLUTION. THE GOAL OF THIS PROJECT IS TO IDENTIFY THE RETINAL AND BRAIN PATHWAYS USED BY LIGHT TO MODULATE BREATHING AND BLOOD GAS HOMEOSTASIS. THIS PROJECT USES WHOLE-BODY PLETHYSMOGRAPHY AND A COMBINATION OF CHEMOGENIC AND VIRAL APPROACHES IN TRANSGENIC MICE TO: 1) DETERMINE THE NEURONAL POPULATIONS BY WHICH INTRINSICALLY-PHOTOSENSITIVE RETINAL GANGLION CELLS (IPRGCS) MODULATE BREATHING RATE AND MINUTE VENTILATION, AND 2) DETERMINE THE EXTENT TO WHICH IPRGC PROJECTIONS TO THE HYPOTHALAMIC BIOLOGICAL CLOCK ORGANIZE THE RESPIRATORY CHEMOREFLEX TO THE LIGHT:DARK CYCLE. THIS CONTRIBUTION IS SIGNIFICANT BECAUSE IT CRITICALLY EXPANDS THE CURRENT UNDERSTANDING OF BREATHING BEHAVIOR TO INCLUDE LIGHT AS A FUNDAMENTALLY IMPORTANT, UBIQUITOUS, AND PREVIOUSLY UNDOCUMENTED MODULATOR OF BREATHING. OVERALL, THIS PROJECT IS EXPECTED TO POSITIVELY IMPACT SEVERAL RESEARCH FIELDS, INCLUDING BASIC AND CLINICAL RESPIRATORY PHYSIOLOGY, CIRCADIAN BIOLOGY, NEUROSCIENCE, AND ECOLOGY. RICE YIELD WOULD BE ENHANCED IF GROWERS COULD PLANT TWO WEEKS EARLIER IN THE SEASON TO BETTER UTILIZE THE SPRING RAIN AND AVOID HIGH NIGHTTIME TEMPERATURES OF SUMMER, WHICH DECREASE GRAIN QUALITY AND YIELD. OUR OBJECTIVE IS TO USE THE WELL-CHARACTERIZED RICE DIVERSITY PANEL 1 (RDP1) COLLECTION OF 424 ACCESSIONS AND FOUR SUBPOPULATION-SPECIFIC RECOMBINANT INBRED LINE AND NEAR ISOGENIC LINE POPULATIONS SPECIFICALLY GENERATED FOR THIS PROJECT TO PROVIDE A BETTER UNDERSTANDING OF THE BASIC GENETIC MECHANISMS RESPONSIBLE FOR TOLERANCE TO COLD TEMPERATURES AT THE EARLY GROWTH STAGES IN RICE. TO ACCOMPLISH THIS, WE WILL (I) USE DIFFERENT RECOMBINANT INBRED LINES TO DETERMINE GENETIC MECHANISMS OF RICE COLD TOLERANCE RESPONSES BY IDENTIFYING COLD-TOLERANT-PARENT-SPECIFIC GENE CO-EXPRESSION MODULES AND THEIR CENTRAL HUB (CONTROL) GENES; (II) ASSOCIATE MODULES AND HUB GENES TO PREVIOUSLY MAPPED QUANTITATIVE TRAIT LOCI (QTL) FOR COLD TOLERANCE; (III) FINE-MAP QTL FOR COLD TOLERANCE WE PREVIOUSLY IDENTIFIED USING THE RDP1 AND FOUR RICE INBRED POPULATIONS TO NARROW DOWN COLD TOLERANCE CANDIDATE GENES; (IV) IDENTIFY CANDIDATE GENES WITHIN THE MAPPED QTL FOR COLD TOLERANCE; AND (V) VALIDATE SELECTED CANDIDATE GENES IN TRANSGENIC PLANTS WITH THE CRISPR GENE KNOCKOUT AND STRONG PROMOTER OVEREXPRESSION SYSTEMS. MECHANISMS AND THERAPEUTIC POTENTIAL OF PHRENIC PROPRIOCEPTORS AFTER SPINAL CORD INJURY - PROJECT SUMMARY/ABSTRACT BREATHING IMPAIRMENTS ARE THE PRIMARY CAUSE OF MORBIDITY AND MORTALITY FOLLOWING CERVICAL SPINAL CORD INJURY (CSCI). LIMITED TREATMENT OPTIONS, BEYOND MECHANICAL VENTILATION, ARE AVAILABLE TO MANAGE RESPIRATORY DYSFUNCTION. ELECTRICAL STIMULATION OF THE DIAPHRAGM (I.E., DIAPHRAGM PACING), IS AN EMERGING INTERVENTION USED TO MANAGE BREATHING IMPAIRMENTS IN SOME INDIVIDUALS WHO FAIL TO WEAN FROM THE MECHANICAL VENTILATOR AFTER CSCI. CLINICAL REPORTS SUGGEST THAT DIAPHRAGM PACING CAN PROMOTE VENTILATOR WEANING, BUT ALSO SHOWS PROMISE IN RESTORING INDEPENDENT BREATHING. COLLECTIVELY, OUR PRELIMINARY DATA AND LITERATURE REPORTS HAVE LED TO THE FUNDAMENTAL HYPOTHESIS THAT DIAPHRAGM SENSORY NEURONS (I.E., PHRENIC AFFERENTS) ARE A NOVEL THERAPEUTIC TARGET TO RESTORE RESPIRATORY NEURAL FUNCTION AND IMPROVE BREATHING FOLLOWING CSCI. SPECIFICALLY, OUR PRELIMINARY DATA IN ANESTHETIZED RATS SUGGEST THAT ACTIVATION OF PHRENIC PROPRIOCEPTORS INDUCES A NOVEL FORM OF PHRENIC MOTOR PLASTICITY. OUR DATA ALSO SHOW THAT DAILY DIAPHRAGM PACING IN FREELY BEHAVING RODENTS LEADS TO A LONG-LASTING INCREASE IN TIDAL VOLUME AFTER CSCI. MOREOVER, WE SHOW THAT ACTIVATION OF PHRENIC AFFERENTS IS NECESSARY FOR PACING-INDUCED RESPIRATORY RECOVERY. FINALLY, PRELIMINARY DATA DEMONSTRATE THAT PHRENIC AFFERENT DISCHARGE IS PROFOUNDLY ALTERED AFTER CSCI AND THAT DIAPHRAGM PACING RESTORES DISCHARGE PATTERNS NECESSARY FOR THE INDUCTION OF PLASTICITY. THESE RESULTS HAVE LED TO THE OVERARCHING HYPOTHESIS THAT PHRENIC PROPRIOCEPTORS INDUCE MECHANISMS OF SPINAL PLASTICITY WHICH PROMOTES THE RECOVERY OF BREATHING AFTER CSCI. AIM 1 WILL TEST THE HYPOTHESIS THAT PHRENIC PROPRIOCEPTOR ACTIVATION INDUCES A NOVEL FORM OF PHRENIC MOTOR PLASTICITY WHICH IS MECHANICALLY DISTINCT FROM OTHER FORMS OF RESPIRATORY PLASTICITY. THIS HYPOTHESIS WILL BE TESTED USING IN VIVO NEUROPHYSIOLOGY TO ENABLE STUDIES EXAMINING SPINAL MECHANISMS OF PLASTICITY. AIM 2 WILL TEST THE HYPOTHESIS THAT RESPIRATORY PROPRIOCEPTORS ARE NECESSARY FOR DIAPHRAGM-PACING INDUCED RESPIRATORY RECOVERY. THIS HYPOTHESIS WILL BE TESTED USING A RODENT MODEL OF DAILY, DIAPHRAGM PACING IN FREELY BEHAVING RODENTS AND ADENO-ASSOCIATED VIRUS (AAV) MEDIATED CHEMOGENETIC APPROACHES TO SILENCE RESPIRATORY PROPRIOCEPTORS. AIM 3 WILL TEST THE HYPOTHESIS THAT PHRENIC PROPRIOCEPTOR ACTIVITY IS ALTERED FOLLOWING CSCI AND DIAPHRAGM PACING PROMOTES DISCHARGE IN PHASE WITH BREATHING. THIS HYPOTHESIS WILL BE TESTED USING A NEUROPHYSIOLOGICAL APPROACH TO RECORD PHRENIC AFFERENT DISCHARGE AND DELIVER DIAPHRAGM PACING. OUR RESEARCH WILL IDENTIFY A NOVEL TARGET TO IMPROVE BREATHING FOLLOWING CSCI AND HAS THE POTENTIAL TO LEAD TO A PARADIGM SHIFT IN THE MANAGEMENT OF BREATHING INSUFFICIENCY AFTER CSCI – A PROBLEM WITH LIMITED TREATMENT OPTIONS AND NO CURE.

MECHANISMS OF MISFOLDED PROTEIN SEQUESTRATION AND DEGRADATION - PROJECT SUMMARY/ABSTRACT MISFOLDED PROTEINS ACQUIRE TOXIC CONFORMATIONS THAT DISRUPT ESSENTIAL CELLULAR PROCESSES, LEADING TO CELL DEATH IN DISEASES LIKE CANCER, ALZHEIMER’S, PARKINSON’S, AND HUNTINGTON’S DISEASES. THEREFORE, CELLS HAVE DEVELOPED DIFFERENT WAYS TO MANAGE MISFOLDED PROTEINS. THIS RESEARCH WILL ELUCIDATE HOW CELLS SORT MISFOLDED PROTEINS INTO DISTINCT COMPARTMENTS THAT SEQUESTER AND CONCENTRATE TOXIC, MISFOLDED PROTEINS FOR MORE EFFICIENT REFOLDING OR CLEARANCE. THE QUALITY CONTROL MECHANISMS THAT MANAGE MISFOLDED PROTEINS WEAR DOWN OVER TIME. THIS RESULTS IN THE BUILDUP OF PROTEIN AGGREGATES THAT ARE A HALLMARK OF PROTEINOPATHIES. IDENTIFYING NOVEL MECHANISMS FOR HOW CELLS SORT MISFOLDED PROTEINS INTO QUALITY CONTROL COMPARTMENTS WILL CREATE NUMEROUS NEW THERAPEUTIC STRATEGIES TO TREAT THESE DEVASTATING DISORDERS. PREVIOUS WORK HAS SHOWN THAT MISFOLDED PROTEINS ARE SPATIALLY SEQUESTERED INTO MEMBRANE-LESS COMPARTMENTS IN THE CELL. HOWEVER, WE DON’T KNOW THE MECHANISMS THAT CONTROL THE FORMATION, LOCALIZATION, AND DEGRADATION OF THESE COMPARTMENTS. THIS KNOWLEDGE GAP IS EVEN LARGER FOR AGGREGATION-PRONE PROTEINS SUCH AS THOSE THAT CAUSE DISEASE. WE WILL ADDRESS THE FUNDAMENTAL QUESTIONS OF WHY AND HOW CELLS FORM INCLUSIONS, THE ROLE OF INCLUSIONS IN DISEASE, AND CRITICALLY, HOW TO PROMOTE DEGRADATION OF THESE INCLUSIONS TO PREVENT CELL DEATH IN DISEASE. THESE STUDIES CAN ALSO SHED LIGHT ON THE APPARENT PARADOX OF PROTEIN AGGREGATES BEING BOTH TOXIC AND PROTECTIVE TO CELLS. OUR RECENT WORK ELUCIDATED A POTENTIAL MECHANISM OF DEGRADATION AS COMPARTMENTS ARE RECRUITED TO THE NUCLEUS- VACUOLE JUNCTION (NVJ) TO FACILITATE VACUOLAR CLEARANCE. ESCRT PROTEINS, NORMALLY ASSOCIATED WITH PROTEIN TRAFFICKING, PLAY AN IMPORTANT ROLE IN THIS NOVEL CLEARANCE MECHANISM. WE ARE USING A PANEL OF MISFOLDED PROTEINS TO PROBE THE INTERACTIONS REQUIRED FOR SORTING MISFOLDED PROTEINS INTO PROTEIN QUALITY CONTROL COMPARTMENTS, TRANSPORTING COMPARTMENTS TO SPECIFIC CELLULAR SITES, AND DETERMINING CLEARANCE MECHANISMS FOR THE COMPARTMENTS. WE WILL ACCOMPLISH THIS BY USING A COMBINATION OF GENETIC DELETIONS, LIVE-CELL TIME-LAPSE FLUORESCENCE MICROSCOPY, AND A SUITE OF BIOCHEMICAL TECHNIQUES FOR DETERMINING CHANGES IN THE AGGREGATION STATE OF PROTEINS PREVIOUSLY DEVELOPED BY THE PI. THE OVERALL GOAL OF OUR RESEARCH IS TO UNCOVER THE BASIC CELLULAR MECHANISMS THAT GO AWRY IN PROTEINOPATHIES SUCH AS CANCER AND NEURODEGENERATION.

COMPUTER-GUIDED ACTION PLANNING TO SUPPORT PHYSICAL ACTIVITY (CAPPA) FOR EMPLOYEES WITH CHRONIC KNEE SYMPTOMS - PROJECT SUMMARY SOCIETY NEEDS EFFECTIVE NON-OPERATIVE THERAPIES TO MANAGE CHRONIC KNEE SYMPTOMS (CKS) RESULTING FROM KNEE OSTEOARTHRITIS (KOA). CKS IS ASSOCIATED WITH PAIN, LOSS OF INDEPENDENCE, AND INCREASED MORTALITY. RATES OF CKS ARE INCREASING, FUELED BY AN AGING, OBESE, AND SEDENTARY SOCIETY. BECAUSE CKS FIRST PRESENTS IN WORKING AGE ADULTS, IT IS A MAJOR CAUSE OF WORK DISABILITY AND WORK PRODUCTIVITY LOSS. OVER 13 MILLION US ADULTS 45 YEARS AND OLDER HAVE SYMPTOMATIC KOA AND CKS,1 AND ONE-THIRD OF WORKING ADULTS (45-64 YEARS) HAVE ARTHRITIS.2-4 OF THOSE WITH KOA, OVER HALF HAVE SEVERE DISABILITY AND WILL HAVE A TOTAL KNEE REPLACEMENT (TKR). SADLY, PERSONS WITH CKS ENGAGE IN TREATMENTS WITH LOW EFFECTIVENESS WHILE WAITING FOR TKR, IN PART DUE TO THE BEHAVIORAL CHALLENGES OF ENGAGING IN REGULAR PHYSICAL ACTIVITY(PA). PHYSICAL ACTIVITY HAS BEEN SHOWN TO RELIEVE PAIN, IMPROVE FUNCTION, AND PREVENT LOSSES IN INDEPENDENCE IN INDIVIDUALS ACROSS THE KOA DISEASE SPECTRUM. DESPITE ITS VALUE, PA LEVELS REMAIN LOW AMONG ADULTS WITH CKS AND FOR ADULTS IN THE WORKPLACE. LOW PA IS LINKED TO HIGH HEALTHCARE SPENDING AND LOSSES IN WORKER PRODUCTIVITY CREATING AN INCENTIVE FOR EMPLOYER-SUPPORT PA INITIATIVES. MANY WORKPLACES HAVE EMBRACED WELLNESS PROGRAMS TO SUPPORT EMPLOYEES IN IMPROVING THEIR HEALTH, HOWEVER EMPLOYEES WITH CKS STRUGGLE WITH LOWER RATES OF PA THAN THEIR PEERS WITHOUT CKS. PREVIOUS RESEARCH HAS FOUND THAT FITBIT TRACKING CAN IMPROVE BEHAVIOR FOR SOME WITH CKS BUT OTHERS BENEFIT FROM MORE INTENSIVE FORMS OF COACHING. THE EXACT AMOUNT OF COACHING NEEDED REMAINS UNKNOWN. EMPLOYEE STAKEHOLDERS HAVE ASKED FOR LOW RESOURCE FORMS OF COACHING AVAILABLE THROUGH THE WORKPLACE. THE POTENTIAL EXISTS TO SUPPORT EMPLOYEES WITH CKS USING AN ACCEPTABLE, LOW-RESOURCE COACHING STRATEGY. THE GOAL OF THIS PROGRAM IS TO TEST A BRIEF, SCALABLE PA COACHING INTERVENTION THAT CAN ASSIST EMPLOYEES WITH CKS ATTAIN AND MAINTAIN HEALTHY PA BEHAVIOR IN TWO WORKSITES IN THE MIDWEST. CAPPA IS A 12-WEEK PILOT RANDOMIZED CONTROLLED TRIAL THAT WILL 1) USE COMPUTER-GUIDED ACTION-PLANNING BEHAVIORAL INTERVENTION TO SUPPORT EMPLOYEES IN MAKING PA ACTION PLANS FOR THEIR HEALTH, 2) USE DATA TRANSMITTED FROM A PERSONAL FITNESS TRACKER (FITBIT) TO SUPPORT COACH AND EMPLOYEE KNOWLEDGE ABOUT PA PERFORMANCE, AND 3) INFORM ON OPTIMAL STEP UP TIMES FOR PARTICIPANTS WHO DO NOT SUBSTANTIALLY INCREASE PA. CAPPA WILL PUT PA FEEDBACK IN A MOVEMENT DASHBOARD TO SUPPORT STUDY PARTICIPANTS. THE CAPPA INTERVENTION WILL USE EACH PARTICIPANT’S PREFERRED COMMUNICATION MODE (TEXT, VOICE CALL, OR VIDEO CHAT) TO ATTAIN AND SUSTAIN HEALTHY PA BEHAVIOR AT BLUE CROSS BLUE SHIELD OF ILLINOIS AND ADVOCATE AURORA HEALTH. FOLLOW-UP MEASURES OF PA, PAIN AND PHYSICAL FUNCTION ARE PLANNED AT TREATMENT COMPLETION AND THREE MONTHS FOLLOWING STUDY COMPLETION. SPECIFIC AIMS ARE TO: 1) ESTIMATE THE EFFICACY OF THE INTERVENTION TO INCREASE OBJECTIVELY- MEASURED PHYSICAL ACTIVITY, 2) EXAMINE THE FEASIBILITY, ACCEPTABILITY AND SUSTAINABILITY OF THE CAPPA INTERVENTION FOR THE RESPECTIVE WORKPLACES, AND 3) INFORM THE INITIAL STEP UP TIME FOR FUTURE STEPPED INTERVENTIONS AMONG PARTICIPANTS NOT SUBSTANTIALLY INCREASING THEIR PHYSICAL ACTIVITY. THIS STUDY LEVERAGES THE COMBINED CLINICAL AND TECHNOLOGIC EXPERTISE OF THE MEMBERS OF THE PHYSICAL ACTIVITY IN RHEUMATOLOGY RESEARCH GROUP AT NORTHWESTERN UNIVERSITY, NORTHWESTERN’S ARTHRITIS CENTER ACCELEROMETER UNIT, MARQUETTE UNIVERSITY’S BEHAVIOR, ENGAGEMENT AND HEALTH TECHNOLOGY ASSESSMENT LAB AND UBIQUITOUS COMPUTING LAB, THE BLUE CROSS BLUE SHIELD OF ILLINOIS CORPORATION, AND ADVOCATE AURORA HEALTH. IF CAPPA IS FEASIBLE AND SHOWS PRELIMINARY EFFECTIVENESS, A MULTI-SITE RANDOMIZED CONTROLLED TRIAL/U01 PROPOSAL WILL FOLLOW TO DEMONSTRATE THE EFFECTIVENESS OF THE CAPPA INTERVENTION IN LARGER WORKSITE POPULATIONS. THIS RESEARCH COULD HAVE A TREMENDOUS IMPACT ON IMPROVING SYMPTOMS AND QUALITY OF LIFE FOR THOSE WITH CKS AND EARLY KOA. IF TREATMENTS ARE SUCCESSFUL AT BLUE CROSS BLUE SHIELD OF ILLINOIS AND ADVOCATE AURORA HEALTH THEY MAY BE EMPLOYED IN THE SERVICES THEY PROVIDE TO OTHER ORGANIZATIONS.

CAREER: A SCALABLE MULTISCALE MODELING FRAMEWORK TO EXPLORE SOOT FORMATION IN REACTING FLOWS -THIS AWARD IS FUNDED IN WHOLE OR IN PART UNDER THE AMERICAN RESCUE PLAN ACT OF 2021 (PUBLIC LAW 117-2). SOOT, A CARBONACEOUS PARTICULATE MATTER FORMED DURING INCOMPLETE COMBUSTION, HAS SIGNIFICANT ADVERSE EFFECTS ON PUBLIC HEALTH AND WELFARE AND IS AN IMPORTANT FORCING AGENT IN CLIMATE CHANGE. TO ACCURATELY UNDERSTAND AND MITIGATE THE EFFECTS OF SOOT, WE NEED TO UNDERSTAND ALL THE PROCESSES RELATED TO SOOT FORMATION AND GROWTH - FROM THE INCEPTION OF SOOT AT AN ATOMIC LEVEL (AKA ATOMIC SCALE) TO ITS MATURATION IN REAL-WORLD COMBUSTION SYSTEMS AT THE DEVICE LEVEL (AKA DEVICE SCALE). UNFORTUNATELY, SUCH DETAILED MULTISCALE MODELING REMAINS A DAUNTING TASK. THIS LEADS TO A SIGNIFICANT GAP OF KNOWLEDGE AND SIGNIFICANT UNCERTAINTY IN THE PREDICTION AND CONTROL OF THE EMISSION OF SOOT AND ITS EFFECTS ON THE CLIMATE AND PUBLIC HEALTH. THIS PROJECT WILL CREATE A FRAMEWORK OF MODELS TO COMBINE SMALL-SCALE ATOMISTIC MODELING WITH LARGER-SCALE ENGINEERING MODELING OF COMBUSTION SYSTEMS. THE PROJECT WILL ENABLE A BETTER PREDICTIVE CAPABILITY FOR MODELING SOOT EMISSION FROM COMBUSTION WHICH WILL LEAD TO CLEANER COMBUSTION SYSTEMS. THE PROJECT WILL ALSO PROVIDE A DETAILED INSIGHT INTO THE PROPERTIES OF SOOT AT AN ATOMIC LEVEL ENABLING A BETTER UNDERSTANDING OF THE EFFECTS OF SOOT ON THE PLANET. IN SO DOING, THE PROJECT WILL SERVE NSF'S MISSION TO PROMOTE THE PROGRESS OF SCIENCE AND TO ADVANCE THE NATIONAL HEALTH, PROSPERITY, AND WELFARE. THE DIRECT IMPACTS OF THE TECHNICAL WORK DONE IN THIS PROJECT ARE TWO-FOLD. FIRST, IT WILL LEAD TO A MORE COMPLETE UNDERSTANDING OF THE PHYSICS OF SOOT INCEPTION AND A DETAILED INSIGHT INTO THE EVOLUTION OF SOOT IN THE REAL WORLD. SECOND, THE DEVELOPED MULTIPHYSICS AND MULTISCALE MODELING FRAMEWORK WILL OPEN UP A NEW HORIZON IN THE THEORETICAL EXPLORATION OF SOOT IN COMBUSTION. THE MULTISCALE BRIDGING STRATEGIES DEVELOPED IN THIS PROJECT CAN BE ADAPTED TO OTHER PROBLEMS THAT REQUIRE MULTISCALE AND MULTIPHYSICS EXPLORATIONS. ALONG WITH THE TECHNICAL DEVELOPMENT, THE PROJECT WILL ALSO CONDUCT OUTREACH ACTIVITIES IN COLLABORATION WITH AN ART MUSEUM TO ENCOURAGE THE COMMUNITY IN FACT- AND DATA-BASED DISCOURSE ON ISSUES SUCH AS COMPLEXITIES OF SOOT PROCESSES, THE EFFECT OF SOOT ON THE SOCIETY, ENVIRONMENTAL POLICIES, AND ENVIRONMENTAL JUSTICE, ETC. ADDITIONALLY, THERE WILL BE ACTIVITIES INVOLVING HIGH SCHOOL STUDENTS THAT WILL HELP PROMOTE SCIENTIFIC COMPUTING AND ENCOURAGE STUDENTS TO PURSUE STEM RESEARCH. THIS PROJECT WILL BRIDGE DIFFERENT DOMAINS OF PHYSICS ACROSS DIFFERENT SCALES BY UTILIZING NOVEL COMPUTATIONAL APPROACHES. AT THE ATOMIC SCALE, THIS PROJECT WILL USE TECHNIQUES SUCH AS MOLECULAR DYNAMICS TO UNRAVEL THE PHYSICS AND CHEMISTRY OF THE SOOT INCEPTION. THE RESULTS FROM THESE MODELS ALONG WITH HIGH-RESOLUTION ELECTRON MICROSCOPIC IMAGES OF ACTUAL SOOT PARTICLES WILL BE ANALYZED USING MACHINE LEARNING TECHNIQUES TO CREATE A NOVEL STOCHASTIC SOOT MODELING FRAMEWORK. THIS SOOT MODELING FRAMEWORK WILL RETAIN THE DETAILED KNOWLEDGE GAINED FROM ATOMIC-SCALE MODELS WHILE EFFICIENTLY OPERATING AT CONTINUUM-SCALE SIMULATIONS SUCH AS IN REACTING COMPUTATIONAL FLUID DYNAMICS (CFD) SIMULATIONS OF COMBUSTION DEVICES. THE STOCHASTIC SOOT MODEL WILL BE COMBINED WITH DETAILED AND ACCURATE TURBULENT CHEMISTRY AND RADIATION MODELS USING A NOVEL HYBRID EULERIAN-LAGRANGIAN APPROACH. THIS HYBRID EULERIAN-LAGRANGIAN APPROACH WILL PROVIDE A UNIQUE HYBRID DATA-TASK PARALLELISM AND AUTOMATIC LOAD BALANCING LEADING TO AN EFFICIENT AND SCALABLE FRAMEWORK FOR MULTISCALE, MULTIPHYSICS REACTING FLOW SOLVER FOR DETAILED EXPLORATION OF SOOT PROCESSES. THE APPROACH RELIES ON THE USE OF EMERGING CONCEPTS IN TRANSITION-METAL MEDIATED CATALYSIS THAT ALLOW FOR MOLECULAR FRAMEWORKS TO BE BROKEN AND REFORMED AT BONDS THAT HAVE TRADITIONALLY RESISTED MANIPULATION. THE STUDIES WILL LIKELY LEAD TO FUNDAMENTAL ADVANCES IN THE FIELD OF CATALYTIC BOND ACTIVATION AND THE KNOWLEDGE GAINED IS ANTICIPATED TO LEAD TO ECONOMIC AND ENVIRONMENTAL IMPROVEMENTS IN THE PREPARATION OF COMPLEX ORGANIC MOLECULES NEEDED FOR THE MANUFACTURE OF PHARMACEUTICALS AND OTHER VALUABLE PRODUCTS. THE BROADER IMPACTS OF THE FUNDED PROJECT SHALL EXTEND TO SUPPORTING SUMMER INTERNSHIPS THAT WILL ALLOW FOR UNDERGRADUATE STUDENTS FROM LOCAL COLLEGE STEM (SCIENCE, TECHNOLOGY, ENGINEERING AND MATHEMATICS) PROGRAMS TO RECEIVE RESEARCH EXPERIENCES IN THE YI LABORATORY. THEREFORE, ANY DYSFUNCTION OF PHOTORECEPTORS THROUGH INJURY OR DISEASE IS DEVASTATING TO AN INDIVIDUAL’S VISION. AS A RESULT, PHOTORECEPTORS ARE THE TARGET OF NUMEROUS CLINICAL AND PRECLINICAL THERAPIES, ALL AIMED AT RESTORING THEIR FUNCTION. UNSURPRISINGLY, PRECLINICAL AND CLINICAL TRIALS PLACE ENORMOUS VALUE ON OUTCOME MEASURES THAT CAN QUICKLY REVEAL POSITIVE (OR NEGATIVE) FUNCTIONAL EFFECTS AS QUICKLY AS POSSIBLE. DESPITE A VERITABLE EXPLOSION OF PRECLINICAL TREATMENTS, THERE IS A RELATIVE DEARTH OF CELLULAR-SCALE NON-INVASIVE, IN VIVO FUNCTIONAL ASSAYS FOR PHOTORECEPTORS IN THE PRECLINICAL DOMAIN. HERE, WE PROPOSE TO DEVELOP NON-INVASIVE IN VIVO FUNCTIONAL TOOLS AND TECHNIQUES TO OBTAIN AND VALIDATE CONTRAST-FREE FUNCTIONAL SIGNALS FROM PHOTORECEPTORS. THIS STUDY AIMS TO: 1) CHARACTERIZE THE INTENSITY-BASED OPTORETINOGRAM FROM PHOTORECEPTORS IN VIVO USING A TREE SHREW ANIMAL MODEL, 2) QUANTIFY AND ENHANCE THE REPEATABILITY, REPRODUCIBILITY, AND THROUGHPUT OF FUNCTIONAL IMAGING IN THE TREE SHREW, AND 3) VALIDATE THE OPTORETINOGRAM AGAINST “GOLD STANDARD” MEASUREMENTS OF PHOTORECEPTOR FUNCTION IN THE TREE SHREW RETINA USING MECHANICALLY AND PHARMACOLOGICALLY INDUCED RETINAL DEGENERATIONS. WE WILL ADDRESS THESE GOALS BY USE OF AN ANIMAL-COMPATIBLE ADAPTIVE OPTICS SCANNING LASER OPHTHALMOSCOPE, AND THE NORTHERN TREE SHREW ANIMAL MODEL (TUPAIA BELANGERI). WE WILL FIRST REVERSE-TRANSLATE THE PREVIOUSLY DEVELOPED INTENSITY-BASED OPTORETINOGRAM FROM HUMANS TO THE TREE SHREW. NEXT, WE WILL ESTABLISH THE MINIMUM MEANINGFUL CHANGE THAT CAN BE OBSERVED USING AN IORG IN THE TREE SHREW, ASSESS INTER-SESSION REPRODUCIBILITY, AND DEVELOP TOOLS TO ENHANCE EFFICIENT IMAGING IN SMALL ANIMAL MODELS. THE RESEARCH TEAM IS MOTIVATED BY RECENT CLINICAL DATA TO EXPLORE WHETHER LOCAL CHANGES IN THE ATHEROSCLEROTIC INFLAMMATORY MICROENVIRONMENT CAN EXERT A CONSIDERABLE SHIFT IN THE BIOCORROSION OF MG ALLOYS. ADDITIONALLY, THE TEAM IS INTERESTED IN UNDERSTANDING HOW CORROSION PRODUCTS FROM THE METALS INFLUENCE NEOINTIMAL PROGRESSION. THEREFORE, THE OBJECTIVES OF THIS PROJECT ARE TO: I) CLARIFY HOW PHYSIOLOGICALLY RELEVANT ATHEROSCLEROTIC INFLAMMATORY MICROENVIRONMENTS AFFECT THE CORROSION PROGRESSION OF MG BASED MATERIALS, AND II) DETERMINE WHETHER MG CORROSION CAN EXERT MEASURABLE CHANGES IN THE PROGRESSION OF THE NEOINTIMA, USING ADVANCED ELEMENTAL IMAGING AND LARGE-SCALE DATA COLLECTION IN AN APOE-/- KO MOUSE MODEL. FIRST, AN IN VITRO CO-CULTURE MODEL WILL BE RUN, USING KEY ATHEROSCLEROTIC INFLAMMATORY CELLS (FOAM CELLS) THAT ARE IMPLICATED IN PLAQUE PROGRESSION AND WILL CHARACTERIZE THEIR CELLULAR BEHAVIOR WHEN EXPOSED TO CLINICALLY USED MG ALLOYS (WE43) AND OTHER CLINICALLY RELEVANT MG ALLOYS (AZ31, WE22, ZA41). THE TEAM WILL THEN DETERMINE IF THEIR MODULATION OF THE MICROFLUIDIC DEGRADATION ENVIRONMENT VIA SECRETED REACTIVE SPECIES CAN EXERT CONSIDERABLE SHIFTS IN THE CORROSION PROGRESSION OF MG ALLOYS IN AIM I. FOR AIMS II AND III, THE TEAM WILL IMPLANT THE MG ALLOYS IN ATHEROGENIC APOE-/- TRANSGENIC MICE. AIM II WILL FOCUS ON USING AN ELEMENTAL IMAGING SYSTEM, WHICH WILL ALLOW FOR IN SITU ELEMENT DETECTION AT HIGH RESOLUTION AND SENSITIVITY. THE TEAM WILL DESCRIBE THE RELATIONSHIP BETWEEN THE IN-SITU PRESENCE OF IMPLANT DERIVED TRACE METALS AND INFLAMMATION. AIM III WILL EXPLORE THE GLOBAL RELATIONSHIPS BETWEEN MG ALLOYS, NEOINTIMAL CHARACTERISTICS, AND BIOLOGICAL VARIABLES. HERE, THE CORROSION RATE OF MG ALLOYS IN DISEASED ANIMALS WILL BE DESCRIBED AND COMPARED TO HEALTHY ANIMAL CONTROLS. OVERALL, THESE AIMS WILL ALLOW THE TEAM TO DETERMINE WHETHER 1) DISEASED NEOINTIMAL MICROENVIRONMENTS INFLUENCE THE CORROSION RATE OF MG ALLOYS, AND 2) IF NEOINTIMAL PROGRESSION IS RELATED TO THE BIOCORROSION OF MG ALLOYS. HIGHLY ORDERED AGGREGATES KNOWN AS AMYLOIDS UNDERLIE THESE DISEASE PATHOLOGIES. PROTEINS CAN ALSO UNDERGO LIQUID-LIQUID PHASE SEPARATION TO FORM AMORPHOUS AGGREGATES THAT ASSEMBLE INTO BIOMOLECULAR CONDENSATES. PROTEINS WITHIN CONDENSATES ARE DYNAMIC AND REVERSIBLE. HOWEVER, WITH AGE AND STRESS, PERSISTENCE OF PROTEINS WITHIN THESE CONDENSATES CAN LEAD TO SOLIDIFICATION AND FORMATION OF IRREVERSIBLE AGGREGATES ASSOCIATED WITH DISEASE. OUR LONG-TERM GOAL IS TO UNDERSTAND HOW CELLS MANAGE PROTEIN AGGREGATES. IT IS WELL ESTABLISHED THAT MOLECULAR CHAPERONES PLAY A CRITICAL ROLE IN THE DISASSEMBLY OF BOTH CONDENSATES AND AMYLOIDS. HOWEVER, THE FIELD IS BEGINNING TO UNLOCK THE MECHANISMS IN WHICH CHAPERONES MEDIATE THIS DISASSEMBLY. TWO DIFFERENT CHAPERONE SYSTEMS HAVE BEEN IDENTIFIED TO HAVE “DISAGGREGASE” ACTIVITY. HSP110 IS A HIGHLY CONSERVED NUCLEOTIDE EXCHANGE FACTOR THAT BINDS WITH MEMBERS OF THE HSP70 FAMILY FOR DISAGGREGATION. THE HSP104 SYSTEM ALSO BINDS WITH THE HSP70 FAMILY CHAPERONES TO DISASSEMBLE AND FRAGMENT PROTEIN AGGREGATES. IT HAS BEEN SUGGESTED THAT THESE TWO SYSTEMS MAY WORK TOGETHER TO INCREASE DISAGGREGATION EFFICIENCY. UNDERSTANDING THE MECHANISMS THAT UNDERLIE THIS SYNERGY WILL PROVIDE SIGNIFICANT LEVERAGE INTO DEVELOPING THERAPEUTIC DIRECTIONS FOR TREATING NEURODEGENERATIVE AND SYSTEMIC PROTEIN AGGREGATION DISEASES. THE OBJECTIVE OF THIS PROJECT IS TO DEFINE HOW HSP110 AND HSP104 SYSTEMS WORK TOGETHER, WHETHER IN PARALLEL OR IN CONCERT, TO MANAGE DIFFERENT TYPES OF PROTEIN AGGREGATES. DUE TO STRONG CONSERVATION OF CHAPERONE SYSTEMS ACROSS EUKARYOTIC SYSTEMS, A YEAST-BASED MODEL PROVIDES A TRACTABLE SYSTEM FOR UNDERGRADUATE RESEARCHERS TO STUDY MOLECULAR MECHANISMS THAT UNDERLIE PROTEIN DISAGGREGATION. THE GOAL OF THIS PROPOSAL IS TO UNDERSTAND HOW HSP104 AND HSP110 CHAPERONE SYSTEMS MANAGE PROTEIN DISAGGREGATION USING THREE DIFFERENT PROTEIN AGGREGATION SYSTEMS: A) THE STUDY OF BIOMOLECULAR CONDENSATES: STRESS GRANULES, B) THE STUDY OF HUMAN AMYLOID: TRANSTHYRETIN PROTEIN, AND C) THE STUDY OF A YEAST-SPECIFIC AMYLOID: [PSI+]. THIS MULTI-PRONGED APPROACH PROVIDES THE ABILITY FOR CONTROLLED EXPLORATION OF HOW THESE TWO CHAPERONE SYSTEMS COORDINATE THEIR EFFORTS TO MANAGE DIFFERENT PROTEIN AGGREGATE SUBSTRATES. WHAT THEN COULD BE THE GERMLINE SPECIFIC STRUCTURE/PATHWAY THAT EXPERIENCES A CONSERVED TEMPERATURE SENSITIVE FAILURE, BUT THAT THE SPECIFIC TEMPERATURE WHEN IT FAILS CAN BE MODIFIED? TWO POSSIBLE SUCH STRUCTURES THAT ARE GERMLINE SPECIFIC ARE THE SYNAPTONEMAL COMPLEX (SC) AND GERM GRANULES. BOTH STRUCTURES ARE NECESSARY FOR FERTILITY AND EXHIBIT LIQUID-LIKE PROPERTIES THAT MAKE THEM INHERENTLY STRUCTURALLY TEMPERATURE SENSITIVE. NOTABLY, PROTEINS WITHIN THESE STRUCTURES CONTAIN INTRINSICALLY DISORDERED DOMAINS THAT CONTRIBUTE TO THE LIQUID-LIKE PROPERTIES OF THE STRUCTURES AND ARE ALSO RAPIDLY EVOLVING. IN THE MODEL NEMATODE C. ELEGANS, BOTH STRUCTURES HAVE BEEN SHOWN TO HAVE STRUCTURAL FAILURE AROUND OR JUST ABOVE THE TEMPERATURE WHERE C. ELEGANS GOES STERILE. USING A COMPARATIVE MODEL OF CAENORHABDITIS NEMATODES THAT HAVE DIFFERENT THERMAL LIMITS OF FERTILITY, WE WILL TEST IF THE SC AND P GRANULES (NEMATODE SPECIFIC GERM GRANULES) ARE THE CRITICAL STRUCTURES THAT FAIL DURING HIGH TEMPERATURE STRESS. IN THIS GRANT WE WILL TEST THE HYPOTHESIS THAT THE LIQUID-LIKE PHYSICAL PROPERTIES OF THE SYNAPTONEMAL COMPLEX AND P GRANULES PREDISPOSES THEM TO FAILURE DURING TEMPERATURE STRESS LEADING TO LOSS OF FERTILITY AT ELEVATED TEMPERATURES USING TWO AIMS. IN AIM 1 WE WILL DETERMINE HOW SYNAPTONEMAL COMPLEX STABILITY PLAYS A ROLE IN THE THERMAL LIMIT OF FERTILITY. THESE EXPERIMENTS WILL SPECIFICALLY ADD TO OUR UNDERSTANDING HOW CONSERVED IS THE TEMPERATURE SENSITIVITY OF MEIOTIC PROCESSES. IN AIM 2 WE WILL DETERMINE HOW P GRANULE STABILITY PLAYS A ROLE IN THE SET POINT OF THE THERMAL LIMIT OF FERTILITY. THE DEVELOPMENT OF NEW TREATMENTS HAS BEEN HINDERED BY THE FIELD’S NEAR EXCLUSIVE FOCUS ON AN INDIVIDUAL’S PHYSICAL BODY MECHANICS OR METABOLIC RATE AS THE PRIMARY CAUSE OF OBESITY-RELATED RESPIRATORY PATHOPHYSIOLOGY. OUR LONG-TERM GOAL IS TO IDENTIFY NEW STRATEGIES THAT CAN EFFECTIVELY TREAT OBESITY-RELATED RESPIRATORY DISEASES. THE OVERALL OBJECTIVE OF THIS PROPOSAL IS TO DEFINE THE CIRCUITRY THAT LINKS METABOLIC NEUROBIOLOGY TO A REDUCTION IN CHEMOSENSITIVITY. OUR CENTRAL HYPOTHESIS IS THAT HYPOTHALAMIC MODULATION OF THE MIDBRAIN PERIAQUEDUCTAL GRAY (PAG) RESULTS IN OBESITY- RELATED REDUCTIONS IN CHEMOSENSITIVITY. THE RATIONALE IS THAT BY DEFINING THE CIRCUITRY THAT LINKS OBESITY-INDUCED HYPOTHALAMIC CHANGES TO REDUCED CHEMOSENSITIVITY, THE OUTCOMES OF THIS PROPOSAL ARE LIKELY TO FOSTER THE DEVELOPMENT OF NEW PHARMACEUTICAL TREATMENTS DESIGNED TO COMBAT OHS. THE CENTRAL HYPOTHESIS WILL BE TESTED IN THE FOLLOWING SPECIFIC AIMS: AIM 1. IDENTIFY THE HYPOTHALAMIC POPULATIONS THAT MODULATE THE PAG.
