CHICAGO CHRONIC CONDITION EQUITY NETWORK (C3EN) - PROJECT SUMMARY HEALTH DISPARITIES IN THE BURDEN OF MULTIPLE CHRONIC CONDITIONS FOR LOW-INCOME AND RACIAL/ETHNIC MINORITY GROUPS ARE PERVASIVE AND HAVE THEIR ROOTS IN SOCIAL DETERMINANTS OF HEALTH AND UNEQUAL ACCESS TO EVIDENCE-BASED HEALTH CARE SERVICES DURING CRITICAL PERIODS OF LIFE. A LONG HISTORY OF RACIAL SEGREGATION IN THE CHICAGO REGION HAS RESULTED IN NEIGHBORHOODS THAT ARE DISPROPORTIONATELY BURDENED BY POVERTY AND RELATED SOCIAL DETERMINANTS OF HEALTH AS WELL AS POOR ACCESS TO HIGH QUALITY HEALTH RESOURCES. UNEQUAL DISTRIBUTION OF RESOURCES ACROSS THE CHICAGO REGION RESULTS IN GEOGRAPHIC “HOTSPOTS” WHERE HIGH NUMBERS OF RESIDENTS SUFFER FROM MULTIPLE CARDIOMETABOLIC CONDITIONS. INNOVATIVE LOCAL SOLUTIONS ARE NECESSARY TO BRIDGE GAPS IN THE HEALTHCARE SYSTEM, GAPS IN THE TRADITIONAL APPROACH TO DELIVERING PREVENTIVE AND MANAGEMENT SERVICES, AND GAPS GENERATED BY THE TRADITIONAL SILOED APPROACH TO SCIENCE. THE CHICAGO CHRONIC CONDITION EQUITY NETWORK (C3EN) WILL ADDRESS THESE GAPS BY STRENGTHENING COLLABORATIONS ACROSS COMMUNITY-BASED ORGANIZATIONS, PRACTICE NETWORKS, AND ACADEMIC RESEARCHERS, BY PROMOTING A COMPREHENSIVE APPROACH TO THE PREVENTION AND MANAGEMENT OF MULTIPLE CHRONIC CONDITIONS THAT ACCOUNTS FOR MENTAL HEALTH, FUNCTIONAL HEALTH AND SOCIAL LIFE, AND BY SUPPORTING INTERVENTIONS THAT ACTIVELY SEEK TO CROSS BOUNDARIES OF DISEASE-SPECIFIC MANAGEMENT, PROFESSIONAL TRAINING, COMMUNITY AND PRACTICE. THIS NETWORK WILL BE THE COMMUNICATION CHANNEL FOR MULTI-DIRECTIONAL EXCHANGE OF IDEAS REGARDING HOW TO ADDRESS DISPARITIES IN COMMUNITIES, THE COMMUNITY-BASED RESEARCH NETWORK WHERE BOTH PILOT STUDIES AND LARGER REGIONAL STUDIES OF SCALABLE INTERVENTIONS WILL TAKE PLACE, AND THE TRAINING GROUND FOR NEW INVESTIGATORS. OUR RESEARCH PROJECTS HAVE BEEN SELECTED TO DEMONSTRATE OUR SCIENTIFIC THEMATIC INTEREST IN PROMOTING A COMPREHENSIVE VIEW OF HEALTH THAT ACKNOWLEDGES THE IMPORTANCE OF PHYSICAL, PSYCHOLOGICAL, AND SOCIAL FUNCTIONING AS OUTCOMES AS WELL AS POTENTIAL AVENUES FOR HEALTH IMPROVEMENT. THE C3EN WILL BENEFIT FROM A FOUNDATION OF MULTIPLE INSTITUTES AND CENTERS AT THE UNIVERSITY OF CHICAGO AND RUSH THAT SET THE STAGE FOR CONDUCTING INTERDISCIPLINARY RESEARCH ALIGNED WITH PUBLIC HEALTH GOALS OF THE CHICAGO REGION, INTER-INSTITUTIONAL COLLABORATION, AND MODELS FOR TRAINING PROGRAMS. OUR CENTER’S SPECIFIC AIMS ARE: 1) TO BUILD A CHICAGO REGIONAL COMMUNITY-BASED RESEARCH NETWORK BASED ON COLLABORATIONS WITH KEY STAKEHOLDER GROUPS INCLUDING COMMUNITY-BASED ORGANIZATIONS, NETWORKS OF AMBULATORY PRACTICES, AND ACADEMIC INSTITUTIONS, TO IDENTIFY INNOVATIVE, EFFECTIVE, AND SCALABLE INTERVENTIONS THAT REDUCE DISPARITIES FOR MULTIPLE CHRONIC CONDITIONS; 2) TO PROVIDE CUTTING EDGE RESEARCH SUPPORT AND EXPERTISE (INTERSECTORAL HEALTH, INFORMATICS, IMPLEMENTATION SCIENCE, COMMUNITY-RESOURCE REFERRAL SOFTWARE SYSTEMS, REMOTE SENSOR TECHNOLOGY) TO FACILITATE CHRONIC CONDITION HEALTH DISPARITIES RESEARCH CONDUCTED ACROSS PRACTICE NETWORKS, IN COMMUNITY SETTINGS, AND IN THE HOME; AND 3) TO ATTRACT AND SUPPORT DIVERSE NEW INVESTIGATORS BY PROVIDING EDUCATION, TRAINING, MENTORING, SUPPORT FOR COMMUNITY AND STAKEHOLDER ENGAGEMENT, PILOT GRANT FUNDING, AND ACCESS TO RESEARCH SUPPORT.

SEES: COMMUNITY DRIVEN MANAGEMENT OF SYNCHROTRON FACILITIES FOR EARTH AND ENVIRONMENTAL SCIENCE -SYNCHROTRON FOR EARTH AND ENVIRONMENTAL SCIENCE (SEES) WILL DEVELOP, MANAGE, OPERATE, AND SUPPORT USER ACCESS TO U.S. SYNCHROTRON-BASED EXPERIMENTAL AND ANALYTICAL CAPABILITIES NECESSARY TO ADVANCE EARTH AND ENVIRONMENTAL SCIENCES. SEES WILL ENABLE CUTTING-EDGE RESEARCH AND TRAIN THE NEXT GENERATION OF SCIENTISTS WITH A FOCUS ON GENERATING NEW UNDERSTANDING OF EARTH AND ENVIRONMENTAL MATERIAL PROPERTIES, PROCESSES, AND BEHAVIOR UNDER THE FULL RANGE OF CONDITIONS FOUND ON AND WITHIN THE EARTH, FROM THE ATMOSPHERE TO THE DEEPEST INTERIOR. SEES WILL PROVIDE EXPANDED ACCESS AND SUPPORT FOR THE EARTH AND ENVIRONMENTAL SCIENCE COMMUNITY AT BEAMLINES SITUATED AT THE ADVANCED PHOTON SOURCE (APS) AT ARGONNE NATIONAL LABORATORY, ADVANCED LIGHT SOURCE (ALS) AT LAWRENCE BERKELEY NATIONAL LABORATORY, NATIONAL SYNCHROTRON LIGHT SOURCE-II (NSLS-II) AT BROOKHAVEN NATIONAL LABORATORY, AND THE STANFORD SYNCHROTRON RADIATION LAB (SSRL). THE EXPANSION IN THE NUMBER OF BEAMLINES AVAILABLE TO THE COMMUNITY UNDER THIS AWARD, COUPLED WITH NEW ANALYTICAL AND EXPERIMENTAL CAPABILITIES, WILL PROVIDE SIGNIFICANT NEW BENEFIT FOR ENVIRONMENTAL SCIENCE RESEARCH ACTIVITIES. SEES WILL LEVERAGE THE PLANNED DOE UPGRADES AT APS AND ALS TO FURTHER INCREASE RESEARCH CAPABILITIES FOR ALL EARTH AND ENVIRONMENTAL SCIENCES RESEARCH ACTIVITIES. IN THIS WAY, SEES WILL ENABLE FUNDAMENTAL RESEARCH THAT HAS DIRECT SOCIETAL BENEFITS ADDRESSING, FOR EXAMPLE, ENVIRONMENTAL STEWARDSHIP, HUMAN HEALTH, NATURAL HAZARDS, AND THE RESOURCES THAT WILL DRIVE THE TECHNOLOGY OF THE NEXT DECADE. IN TERMS OF BROADER IMPACTS, SEES WILL PLACE MAJOR EMPHASIS ON DIVERSITY AND INCLUSION OF THE PARTICIPATING SCIENTIFIC COMMUNITY, SEEKING MECHANISMS FOR BROADENING PARTICIPATION FROM MEMBERS OF COMMUNITIES UNDERREPRESENTED IN STEM. IT WILL ALSO CONDUCT A SIGNIFICANT PORTFOLIO OF EDUCATION AND OUTREACH ACTIVITIES, WORKING COLLABORATIVELY WITH EDUCATION AND OUTREACH PROGRAMS AT THE DOE-MANAGED SYNCHROTRON LIGHT SOURCES. SYNCHROTRON RADIATION BASED ANALYTICAL METHODS HAVE REVOLUTIONIZED THE STUDY OF PHYSICAL AND CHEMICAL PROPERTIES OF EARTH MATERIALS AND ASSOCIATED NATURAL PROCESSES. THESE ADVANCED CAPABILITIES HAVE BEEN PROVEN TO BE CRITICAL IN OBTAINING OUR CURRENT UNDERSTANDING OF THESE SYSTEMS, AND WILL ALLOW FUNDAMENTAL QUESTIONS ABOUT THE EARTH TO BE ADDRESSED, INCLUDING: HOW DID PLATE TECTONICS BEGIN? HOW DID DEEP MANTLE STRUCTURES FORM? WHAT IS THE CONNECTION BETWEEN INITIAL DIFFERENTIATION AND THE EARLY MECHANISMS OF CRUST FORMATION AND RECYCLING? HOW DOES OXIDATION STATE IMPACT THE DEPTH OF MELT INITIATION IN THE MANTLE? WHAT FLUID-ASSISTED PHYSICAL PROCESSES CONTROL THE RATES AT WHICH FAULTS PROPAGATE? HOW DO MINERALS, AQUEOUS COLLOIDS, AND PARTICULATE AEROSOLS AFFECT THE TRANSPORT OF CRITICAL ELEMENTS AT THE EARTH?S SURFACE? HOW LONG CAN SOILS AND OCEANS SEQUESTER CARBON AND WHAT IS THE IMPACT ON CLIMATE? HOW DO BIOGEOCHEMICAL CYCLING AND FLUID-MINERAL INTERACTIONS AFFECT BIOAVAILABILITY AND TOXICITY OF ENVIRONMENTAL CONTAMINANTS? THE AVAILABILITY OF UNIQUE EXPERIMENTAL AND ANALYTICAL CAPABILITIES IN THE BEAMLINES AT DOE-OPERATED NATIONAL SYNCHROTRONS ENABLE THOUSANDS OF CUTTING-EDGE EXPERIMENTS TO BE CONDUCTED ANNUALLY BY SCIENTISTS AND STUDENTS FROM AROUND THE WORLD. FURTHER ADVANCES WILL OCCUR WITH THE PLANNED SYNCHROTRON SOURCE UPGRADES AT APS AND ALS, ALLOWING FUNDAMENTALLY NEW TYPES OF STUDIES TO BE PURSUED. THIS PROJECT WILL ENABLE THE EARTH AND ENVIRONMENTAL SCIENCE COMMUNITY TO TAKE FULL ADVANTAGE OF THESE NEW SOURCE CAPABILITIES TO REMAIN AT THE FOREFRONT OF SCIENTIFIC DISCOVERY. THIS AWARD REFLECTS NSF'S STATUTORY MISSION AND HAS BEEN DEEMED WORTHY OF SUPPORT THROUGH EVALUATION USING THE FOUNDATION'S INTELLECTUAL MERIT AND BROADER IMPACTS REVIEW CRITERIA.- SUBAWARDS ARE PLANNED FOR THIS AWARD.

STUDY TO UNCOVER PATHWAYS TO EXCEPTIONAL COGNITIVE RESILIENCE IN AGING (SUPERAGING) - PROJECT SUMMARY (OVERALL): THE PRIMARY GOAL IS TO ESTABLISH A MULTICENTER SUPERAGING CONSORTIUM TO IDENTIFY BEHAVIORAL, HEALTH, BIOLOGIC, GENETIC, ENVIRONMENTAL, SOCIOECONOMIC, PSYCHOSOCIAL, ANATOMIC AND NEUROPATHOLOGIC FACTORS ASSOCIATED WITH SUPERAGING. THESE GOALS WILL BE ACHIEVED THROUGH AN ORGANIZATIONAL STRUCTURE WITH 3 CORES (ADMINISTRATIVE/BIOSTATISTICS, CLINICAL/IMAGING, AND BIOSPECIMEN/NEUROPATHOLOGY) AND 2 RESEARCH PROJECTS. THE CONSORTIUM WILL ENROLL 500 PARTICIPANTS ACROSS 4 US SITES LOCATED IN ILLINOIS, WISCONSIN, MICHIGAN AND GEORGIA, AND THE CANADIAN SITE IN SOUTHWEST, ONTARIO, WITH A FOCUS ON THE ENROLLMENT OF BLACK SUPERAGERS AND COGNITIVELY AVERAGE ELDERLY CONTROLS WITH SIMILAR DEMOGRAPHICS (CONTROLS). THE ADMINISTRATIVE/BIOSTATISTICS CORE WILL PROVIDE GOVERNANCE AND FISCAL OVERSIGHT, MAINTAIN SCIENTIFIC INTEGRITY, AND CREATE A CENTRALIZED BIOSTATISTICS AND DATABASE INFRASTRUCTURE TO HARMONIZE THE GOALS AND ACTIVITIES OF THE CORES, SITES, AND PROJECTS, WITH EACH OTHER, WITH THE NIA AND WITH EXTRAMURAL COLLABORATORS. THE CLINICAL/IMAGING CORE WILL STANDARDIZE CRITERIA FOR THE UNIFORM CROSS-SITE AND MULTIDISCIPLINARY CHARACTERIZATION OF SUPERAGERS, STREAMLINE RECRUITMENT INCLUDING THAT OF BLACK PARTICIPANTS, ENTER RELEVANT INFORMATION IN THE COMPREHENSIVE DATABASE, SUPPORT CO-ENROLLMENT INTO PROJECT 1, AND ENCOURAGE COLLABORATIVE VENTURES AIMING TO UNDERSTAND THE FACTORS THAT PROMOTE SUPERAGING. THE BIOSPECIMEN/NEUROPATHOLOGY CORE WILL COLLECT AND BANK BRAIN TISSUE AND BLOOD PRODUCTS FROM SUPERAGING AND CONTROL CASES, ACCORDING TO OPTIMIZED PROCEDURES. IT WILL RENDER PATHOLOGICAL DIAGNOSES, QUANTITATE SELECTED MARKERS OF NEURODEGENERATION AND NEURONAL STRUCTURE, COORDINATE THE ANALYSES OF PLASMA BIOMARKERS FOR ALZHEIMER'S DISEASE, AND MAKE SPECIMENS AVAILABLE FOR COLLABORATIVE INVESTIGATIONS. PROJECT 1 WILL USE STATE-OF- THE-ART WEARABLE TECHNOLOGY TO OBTAIN REAL-TIME MEASUREMENTS IN THE COURSE OF EVERYDAY LIFE TO CHARACTERIZE QUANTITATIVE PARAMETERS RELATED TO SLEEP, PHYSICAL ACTIVITY, AUTONOMIC RESPONSIVITY, AND SOCIAL ENGAGEMENT TO DETERMINE WHETHER SUPERAGERS HAVE RELATIVELY PRESERVED AND QUANTITATIVELY DETERMINED PHYSIOLOGIC AND BEHAVIORAL `COMPLEXITY' COMPARED TO CONTROLS. PROJECT 2 WILL USE TRANSCRIPTOMIC, GENETIC, AND PROTEIN PROFILING APPROACHES TO TEST THE HYPOTHESIS THAT SUPERAGERS WILL DEMONSTRATE SIGNIFICANT MOLECULAR DIFFERENCES IN THEIR CENTRAL AND PERIPHERAL IMMUNE AND INFLAMMATORY SYSTEM PARAMETERS COMPARED TO MATCHED CONTROL AND ALZHEIMER'S DISEASE PARTICIPANTS. BY IDENTIFYING NEUROBIOLOGIC FEATURES THAT CONTRIBUTE TO SUPERIOR MEMORY PERFORMANCE IN OLD AGE, OUTCOMES FROM THIS CONSORTIUM WILL HELP ISOLATE FACTORS THAT PROMOTE SUCCESSFUL COGNITIVE AGING AND PERHAPS ALSO PREVENT AGE-RELATED BRAIN DISEASES SUCH AS ALZHEIMER'S DISEASE.

PHYSICS FRONTIER CENTER FOR LIVING SYSTEMS -LIVING SYSTEMS CONTINUALLY EXPLORE NEW OUT-OF-EQUILIBRIUM CONFIGURATIONS AND ADAPT TO CHANGING ENVIRONMENTS FROM THEIR SPECIAL PERCH IN PHASE SPACE. THIS OPEN-ENDED ADAPTIVE ABILITY HAS ENABLED LIVING MATTER TO EMERGE FROM A PRIMORDIAL STEW OF CHEMICAL SPECIES AND DEVELOP PATHWAYS FOR SELF-REPLICATION AND DIVERSIFICATION OF PHYSIOLOGICAL FUNCTION IN RESPONSE TO EVER-CHANGING ENVIRONMENTAL CONDITIONS. A SIGNIFICANT GAP IN OUR UNDERSTANDING OF LIVING MATTER LIES IN OUR LIMITED KNOWLEDGE OF HOW LIVING SYSTEMS ADAPT, LEARN, AND EVOLVE NEW FUNCTIONS. THIS PHYSICS FRONTIERS CENTER (PFC) AWARD AIMS TO ADDRESS THIS GAP BY ESTABLISHING A CENTER FOR LIVING SYSTEMS (CLS) AT THE UNIVERSITY OF CHICAGO. THE PRIMARY OBJECTIVE OF THE CLS IS TO DEVELOP GENERAL FRAMEWORKS FOR ADAPTATION IN COMPLEX SYSTEMS. THROUGH THE PFC AWARD, THE CLS WILL FOCUS ON CREATING FRAMEWORKS TO UNDERSTAND ADAPTATION IN LIVING SYSTEMS THAT SPAN LENGTH AND TIME SCALES. THIS ENDEAVOR NECESSITATES PROGRESS IN THE NEW FIELD OF SCIENCE DEDICATED TO UNDERSTANDING INFORMATION PROCESSING AND MEMORY STORAGE IN FAR-FROM-EQUILIBRIUM SYSTEMS. TO ACHIEVE ITS GOALS, THE CLS WILL FACILITATE COLLABORATION AND INTERDISCIPLINARY RESEARCH AMONG DIVERSE SCIENTIFIC COMMUNITIES THROUGH VARIOUS CENTER-LED ACTIVITIES, INCLUDING WORKSHOPS, SEED FUNDING, EXTERNAL COLLABORATIONS, AND THE ESTABLISHMENT OF SHARED FACILITIES. THE CLS WILL CONTRIBUTE TO THE ADVANCEMENT OF PHYSICS EDUCATION BY IMPLEMENTING A MULTI-FACETED APPROACH AIMED AT ACCELERATING CULTURE CHANGE WITHIN PHYSICS EDUCATION TO TRAIN THE FUTURE PHYSICS WORKFORCE AND PROMOTE DIVERSITY WITHIN THE FIELD. THE CLS WILL DEVELOP RECRUITMENT AND SCIENCE OUTREACH PROGRAMS TO INVOLVE UNDER-REPRESENTED GROUPS AND FOSTER EFFECTIVE TRAINING CULTURE IN PHYSICS COMMUNITIES. THE CLS RESEARCH ACTIVITIES ARE ORGANIZED INTO THREE AREAS. THE FIRST FOCUSES ON UNDERSTANDING HOW EVOLUTIONARY PROCESSES THAT PLAY OUT ON THE BILLIONS-OF-YEARS TIMESCALE BUILD COMPONENTS THAT CAN ADAPT ON MUCH SHORTER TIME SCALES. THE SECOND RESEARCH AREA WILL EXPLORE THE NATURE OF THE MECHANOCHEMICAL DYNAMICAL SYSTEMS THAT CONTROL THE SHAPE AND MOTION OF CELLS AND MULTICELLULAR TISSUE. THE THIRD RESEARCH AREA SEEKS TO UNITE THE MECHANISTIC AND PHYSICAL INSIGHTS INTO ADAPTATION GLEANED FROM THE FIRST TWO RESEARCH AREAS TO REVEAL HOW COMPUTATION IS DONE IN COMPLEX SYSTEMS. THIS DISTILLS OUT THE PRIMARY VISION OF THE CENTER: TO LEARN HOW STRONGLY INTERACTING, HETEROGENEOUS BIOLOGICAL SYSTEMS PERFORM COMPUTATIONS THAT MAKE THEM ADAPTABLE TO COMPLEX INPUTS OVER A WIDE RANGE OF TIME AND SPATIAL SCALES. A SEED FUNDING MECHANISM WILL BE USED TO EXPLORE HOW IDEAS GLEANED FROM LIVING SYSTEMS CAN ADVANCE RESEARCH IN NON-LIVING SYSTEMS, INCLUDING NON-EQUILIBRIUM MATTER, OPEN QUANTUM SYSTEMS AND PHOTONICS. THIS PHYSICS FRONTIERS CENTERS AWARD IS CO-FUNDED BY THE DIVISION OF PHYSICS WITHIN THE MATHEMATICAL AND PHYSICAL SCIENCES DIRECTORATE, AND THE DIVISION OF MOLECULAR AND CELLULAR BIOSCIENCES AND THE DIVISION OF INTEGRATIVE ORGANISMAL SYSTEMS WITHIN THE DIRECTORATE FOR BIOLOGICAL SCIENCES. NSF?S CHEMMATCARS: A SYNCHROTRON X-RAY NATIONAL USER FACILITY FOR MOLECULAR SCIENCE AT THE ADVANCED PHOTON SOURCE -THE DIVISION OF CHEMISTRY AND THE DIVISION OF MATERIALS RESEARCH IN THE MATHEMATICAL AND PHYSICAL SCIENCES DIRECTORATE, ALONG WITH THE DIVISION OF CHEMICAL, BIOENGINEERING, ENVIRONMENTAL, AND TRANSPORT SYSTEMS IN THE ENGINEERING DIRECTORATE PROVIDE CONTINUING SUPPORT TO NSF'S CHEMMATCARS. THIS IS A NATIONAL USER FACILITY FOR FRONTIER RESEARCH IN CHEMISTRY, MATERIALS SCIENCE, BIOLOGY, AND ENGINEERING, EMPLOYING SYNCHROTRON X-RAYS AT THE ADVANCED PHOTON SOURCE, ARGONNE NATIONAL LABORATORY. EXPERIMENTAL FACILITIES AT NSF'S CHEMMATCARS SERVE A BROAD NATIONAL AND INTERNATIONAL COMMUNITY OF SCIENTISTS PROVIDING STATE-OF-THE-ART CAPABILITIES INCLUDING SOME NOT FOUND ANYWHERE ELSE IN THE WORLD. RESEARCH ACTIVITIES ADDRESS VITAL SOCIETAL ISSUES, INCLUDING THE DEVELOPMENT OF NEW ENERGY SOURCES, BIOMOLECULAR MATERIALS INSPIRED BY BIOLOGICAL PROCESSES, STUDIES OF BIOMEMBRANE INTERACTIONS TO UNDERSTAND THE RULES OF LIFE, ENVIRONMENTAL REMEDIATION AND CHEMICAL SEPARATIONS PROCESSES, AND NEW MATERIALS AND CATALYSTS IMPORTANT FOR A WIDE RANGE OF INDUSTRIES. THE FACILITY SERVES AS A TRAINING GROUND FOR RESEARCHERS AT ALL LEVELS AND CARRIES OUT NUMEROUS ACTIVITIES TO DEVELOP AND DIVERSIFY THE FUTURE STEM WORKFORCE. THESE INCLUDE SPECIALIZED EDUCATION, TRAINING, AND CAREER DEVELOPMENT OF STUDENTS, POSTDOCTORAL SCHOLARS, AND FACULTY IN FOREFRONT SYNCHROTRON X-RAY STUDIES IN MOLECULAR SCIENCE, AS WELL AS PROGRAMS DESIGNED TO ADDRESS THE GOAL OF BROADENING PARTICIPATION IN STEM ACTIVITIES. THIS USER FACILITY PROVIDES A UNIQUE HIGH BRILLIANCE X-RAY RESOURCE FOR THE STUDY OF ADVANCED SMALL-MOLECULE CRYSTALLOGRAPHY, LIQUID SURFACE AND INTERFACE SCATTERING, AND ANOMALOUS SMALL AND WIDE-ANGLE X-RAY SCATTERING. ADVANCED INSTRUMENTATION AT NSF'S CHEMMATCARS ENABLES FOREFRONT RESEARCH OF ORDERED AND DISORDERED SOLIDS, COMPLEX FLUIDS, AND SOFT INTERFACES ON THE ATOMIC, MOLECULAR, AND MESOSCOPIC LENGTH SCALES OVER A RANGE OF TIME SCALES FROM NANOSECONDS TO MINUTES. USERS OF NSF'S CHEMMATCARS TAKE ADVANTAGE OF ITS UNIQUE CAPABILITIES TO ADDRESS A WIDE VARIETY OF SCIENTIFIC PROBLEMS. THE MANY CAPABILITIES IN ADVANCED CRYSTALLOGRAPHY INCLUDE THE STRUCTURAL RESOLUTION OF EXCITED STATE AND HIGHLY REACTIVE SPECIES WITH PHOTO-CRYSTALLOGRAPHY. SITE-SPECIFIC MEASUREMENTS CAN DISTINGUISH THE SAME-SITE OCCUPANCY OF SIMILAR ATOMIC NUMBER ELEMENTS, AS WELL AS RELATIVE OXIDATION LEVELS OF THE SAME ELEMENT AT CRYSTALLOGRAPHICALLY DISTINCT SITES. INITIATIVES IN STRUCTURAL DYNAMICS, INCLUDING A FRONTIER EFFORT IN SMALL MOLECULE SERIAL CRYSTALLOGRAPHY, WILL INVESTIGATE SYSTEMS UNDERGOING REVERSIBLE AND IRREVERSIBLE CHEMICAL TRANSFORMATIONS. MOLECULAR-SCALE STRUCTURAL STUDIES OF LIQUID INTERFACES USING X-RAYS ADVANCE THE SYNTHESIS OF NANO- AND 2D MATERIALS FOR CATALYSIS AND ENERGY APPLICATIONS, REVEAL THE INTERFACIAL SEPARATION OF CRITICAL MINERALS, AND EXPLORE THE UNDERLYING STRUCTURE OF SEA SURFACE MICROLAYERS AND ORGANIC THIN FILM COATINGS. LIPID BILAYERS ARE UTILIZED AS MODEL SYSTEMS OF BIOMEMBRANE STRUCTURE AND DYNAMICS, PROTEIN-LIPID INTERACTIONS, AND NONEQUILIBRIUM PROCESSES. A RECENTLY COMMISSIONED CAPABILITY IN X-RAY ABSORPTION SPECTROSCOPY CAN TRANSFORM OUR UNDERSTANDING OF ELEMENTAL SPECIATION AND OXIDATION AT LIQUID INTERFACES. A FRONTIER INITIATIVE WILL PROBE ELEMENTAL AND STRUCTURAL HETEROGENEITIES AT LIQUID INTERFACES ON SMALL LENGTH SCALES. ANOMALOUS SMALL AND WIDE-ANGLE X-RAY SCATTERING (ASWAXS) PROBES ELEMENT-SPECIFIC METAL ION DISTRIBUTIONS IN DISORDERED MATERIALS TO INVESTIGATE METAL INTERACTIONS IN SUPRAMOLECULAR CHEMISTRY, METAL ION INTERACTIONS IN SOLUTION, AND ELEMENTAL DISTRIBUTIONS IN NANOMATERIALS WITH SUB-NANOMETER RESOLUTION. TISSUE DESTRUCTION AND HEALING IN CELIAC DISEASE - PROJECT SUMMARY CELIAC DISEASE (CED) IS A COMPLEX T CELL-MEDIATED ENTEROPATHY INDUCED BY DIETARY GLUTEN IN HLA-DQ2+ AND/OR HLA-DQ8+ INDIVIDUALS, WHICH CURRENTLY AFFECTS 1% OF THE GLOBAL POPULATION. A GLUTEN-FREE DIET (GFD) IS, TO THIS DATE, THE TREATMENT OF CHOICE FOR CED. HOWEVER, 50% OF CED PATIENTS ARE UNABLE TO EFFECTIVELY ADHERE TO A DIET THAT SUSTAINABLY EXCLUDES GLUTEN, WITH MANY PATIENTS SUFFERING FROM INADVERTENT GLUTEN EXPOSURE. MOREOVER, OVER 30% OF CED PATIENTS HAVE PERSISTENT HIGH SYMPTOM BURDENS, RESULTING FROM CONTINUED MUCOSAL DAMAGE, DESPITE ADHERING TO A GFD. PERSISTENT MUCOSAL DAMAGE ON A GFD IS ASSOCIATED WITH SEVERAL SEVERE COMPLICATIONS, INCLUDING MALIGNANCIES, ESPECIALLY LYMPHOMAS AND BONE DISEASES. IN ADDITION, PATIENTS WITH ACTIVE CED DISPLAY A WIDE RANGE OF CLINICAL PRESENTATIONS, INCLUDING METABOLIC DEFECTS (VITAMINS, IRON, AND CHOLESTEROL) THAT ARE NOT CORRELATED TO THE DEGREE OF TISSUE DAMAGE. ALTHOUGH MUCH PROGRESS HAS BEEN MADE IN UNDERSTANDING CED, MAJOR GAPS REMAIN, NOTABLY REGARDING THE BIOLOGICAL MECHANISMS INVOLVED IN DIFFERENT CLINICAL PRESENTATIONS AND THE INCONSISTENT HEALING PROCESS. FOR INSTANCE, IT IS POORLY UNDERSTOOD WHY, INDEPENDENTLY FROM THE DEGREE OF VILLOUS ATROPHY, CERTAIN PATIENTS DISPLAY NUTRIENT AND LIPID DEFICIENCIES, WHEREAS OTHERS HAVE NORMAL LEVELS OF VITAMINS, CHOLESTEROL, AND IRON. FURTHERMORE, WHILE THERE IS EVIDENCE FOR A ROLE OF THE MICROBIOME IN CED, WE LACK INFORMATION ON SMALL-INTESTINAL MUCOSAL MICROBIOTA IN HUMAN CED (WHICH IS MORE LIKELY TO HAVE METABOLIC EFFECTS AND DIRECTLY INTERACT WITH THE IMMUNE SYSTEM). FINALLY, WE HAVE LITTLE KNOWLEDGE ABOUT INTERACTIONS BETWEEN GLUTEN, INTESTINAL EPITHELIAL CELLS (IECS), IMMUNE CELLS, AND THE MICROBIOTA, AND HOW THEY ARE LINKED TO THE DIFFERENT CED CLINICAL PHENOTYPES. OUR RC2 PROPOSAL WILL TEST THE HYPOTHESIS THAT CED IS A HETEROGENEOUS DISORDER, WHILE ATTEMPTING TO DEFINE INTERACTIONS BETWEEN IECS, MICROBIOTA, IMMUNE SYSTEM, AND GENETICS THAT UNDERLIE DIFFERENCES IN CLINICAL PRESENTATION, SEVERITY OF TISSUE DESTRUCTION, AND THE ABILITY TO HEAL. IT WILL ALSO ADDRESS CRITICAL GAPS IN OUR UNDERSTANDING OF CED PATHOGENESIS AND CLINICAL PRESENTATIONS, AND DEVELOP TOOLS FOR NON-INVASIVE MONITORING OF CED PATIENTS. WE HAVE ASSEMBLED A TEAM OF INTERNATIONALLY RECOGNIZED EXPERTS IN THE FIELD OF CED, EPITHELIAL CELL BIOLOGY, MUCOSAL IMMUNOLOGY, MICROBIOME, AND CHEMISTRY. THE RC2 PROPOSAL IS ANCHORED AROUND MULTI-OMICS STUDIES PERFORMED IN THE CONTEXT OF CROSS-SECTIONAL AND INTERVENTIONAL GLUTEN CHALLENGE AND DE-CHALLENGE STUDIES, ON 445 WELL-CHARACTERIZED ADULT AND PEDIATRIC PATIENTS. THE PROPOSED SPECIFIC AIMS ARE: 1) DEVELOPING AN APPROACH TO PRECISION MEDICINE IN CED; 2) DECIPHERING THE MECHANISMS ASSOCIATED WITH TISSUE DESTRUCTION AND HEALING IN CED, 3) DEVELOPING NON-INVASIVE TOOLS FOR MONITORING CED PATIENTS, AND 4) DEVELOPING RESEARCH RESOURCES FOR THE SCIENTIFIC AND MEDICAL COMMUNITY TO ADVANCE PATIENT CARE AS WELL AS DISCOVERY-BASED AND HYPOTHESIS-GENERATING SCIENCE. THIS APPLICATION AIMS TO GENERATE THE MUCH- NEEDED KNOWLEDGE BASE AND RESOURCES TO FURTHER OUR UNDERSTANDING OF CED PATHOGENESIS AND ITS HETEROGENEITY, IMPROVE INDIVIDUALIZED PATIENT CARE AND FOLLOW-UP, AND DEVELOP NEW THERAPEUTIC AND PREVENTIVE TARGETS.

THE PLATFORM CONSISTS OF THREE CORE OPERATING SITES, AT UNIVERSITY OF CHICAGO (UC), TEXAS ADVANCED COMPUTING CENTER (TACC), AND THE NATIONAL CENTER FOR ATMOSPHERIC RESEARCH (NCAR), AND PARTNERS WITH THE RENAISSANCE COMPUTING INSTITUTE (RENCI) AT THE UNIVERSITY OF NORTH CAROLINA ON NETWORKING CAPABILITY DEVELOPMENT. THE HARDWARE RANGES FROM AN INVESTMENT IN DIVERSE HIGH-END DATACENTER NODES INCLUDING GRAPHIC PROCESSING UNITS (GPUS), FIELD PROGRAMMABLE GATE ARRAYS (FPGAS), AND SPECIALIZED ARCHITECTURES, TO SINGLE BOARD COMPUTERS (SBCS) THAT CAN BE COST-EFFECTIVELY DEPLOYED AT THE EDGE. USERS CAN RECONFIGURE THIS HARDWARE AT BARE-METAL LEVEL, THROUGH VIRTUALIZATION, OR CONTAINER DEPLOYMENT DEPENDING ON THEIR EXPERIMENTAL NEEDS AND THE USED HARDWARE. IN PHASE-IV, CHAMELEON WILL CONTINUE ITS MISSION TO PROVIDE AN EXPERIMENTAL PLATFORM FOR COMPUTER SCIENCE RESEARCH AND EDUCATION, AND EVOLVE FURTHER TO BETTER SATISFY EMERGENT NEEDS IN CURRENT RESEARCH. APTO: COLLABORATIVE RESEARCH: GLOBAL OBSERVATORY AND VIRTUAL LABORATORY FOR SCIENCE AND TECHNOLOGY ADVANCE -OVER THE PAST HALF-CENTURY, THE GLOBAL GEOPOLITICAL BALANCE OF SCIENTIFIC, TECHNOLOGICAL, AND ECONOMIC LEADERSHIP HAS SHIFTED, WITH CHINA?S METEORIC RISE AND THE ASCENDANCE OF NEW POWERS INCLUDING KOREA AND INDIA. TECHNOLOGICAL LEADERSHIP REQUIRES DRIVING ADVANCES AND SETTING STANDARDS THAT CATALYZE THE FUTURE OF GLOBAL PRODUCTIVITY. TO UNDERSTAND PATHWAYS THAT ENHANCE U.S. COMPETITIVENESS IN CRITICAL TECHNOLOGY CAPACITY, PRODUCTION, AND USE, THIS PROJECT WILL CREATE A GLOBAL OBSERVATORY AND VIRTUAL LABORATORY FOR U.S. SCIENCE AND TECHNOLOGY IN THE CONTEXT OF GLOBAL ADVANCEMENT. IT WILL PRODUCE DATA SETS AND TECHNOLOGY OUTCOME MODELS THAT CAPTURE THE COMPLEX AND EMERGENT INTERDEPENDENCIES AMONG TECHNOLOGIES; THE FUNDERS, RESOURCES, RESEARCHERS, AND UNIVERSITIES THAT CATALYZE AND INVENT THEM; THE WORKFORCES AND ORGANIZATIONS THAT PRODUCE THEM; AND THE MARKETS THAT CONSUME THEM. DRAWING UPON THE POWER OF DEEP NEURAL NETWORK ?TRANSFORMER? ARCHITECTURES, THE PROJECT WILL THEN BUILD A DEEP-LEARNED, CHRONOLOGICALLY TRAINED, LARGE LANGUAGE MODEL (LLM) TO FUNCTION AS A DATA-DRIVEN ?DIGITAL DOUBLE? OF THE GLOBAL TECHNO-SCIENTIFIC SYSTEM. THE LLM WILL EMBED RESEARCH ARTIFACTS (E.G., ARTICLES, PATENTS, PRODUCTS, RELATED NEWS, AND THEIR RICH META-DATA) IN A HIGH-DIMENSIONAL SPACE, MAPPING THEM TO QUANTITATIVE METRICS OF TECHNOLOGY CAPABILITY, PRODUCTION, AND USE. THE PROJECT TEAM WILL FINE-TUNE OUR LLMS TO CAPTURE CHANGES IN KEY METRICS AS CORRESPONDING TRAJECTORIES WITHIN EMBEDDING SPACE, AND THUS ENABLE THEM TO FUNCTION AS 1) A GLOBAL OBSERVATORY FOR TECHNOLOGY CATALYSIS, CAPACITY, PRODUCTION, AND USE; AND 2) A VIRTUAL LABORATORY FOR SIMULATED EXPERIMENTS THAT CAN GUIDE 3) CAUSAL ESTIMATION OF RELATIONSHIPS AMONG POLICY LEVERS (FUNDING, COMPETITION, IMMIGRATION), TECHNOLOGY PERFORMANCE, AND GLOBAL LEADERSHIP. THEY WILL ALSO TUNE THE LLMS AND RELATED MODELS TO ENABLE CUSTOMIZED EXTRACTION, STRUCTURING, AND DISAMBIGUATION OF DATA ON RESEARCH, PRODUCTS, FUNDING, AND POLICY FROM NOVEL SOURCES TO ENRICH MODELED OBSERVATIONS AND PREDICTIONS, WHICH WILL ENABLE THE CONTINUOUS INCORPORATION OF ADDITIONAL DATA AND EXTRACTION OF INSIGHT. FINALLY, THEY WILL USE THE MODELS AS RESOURCES FOR SCIENTISTS AND POLICYMAKERS BY BUILDING DASHBOARDS TO PROVIDE FUNDING AGENCIES, POLICYMAKERS, AND RESEARCHERS WITH THE SITUATIONAL AWARENESS REQUIRED TO IMPROVE THE QUALITY AND DIVERSIFICATION OF THEIR TECHNOLOGY DEVELOPMENT PORTFOLIOS. CONSORTIUM PROPOSAL FOR CMB-S4 CONTINUING DESIGN -THIS AWARD WILL FUND THE NSF SIDE OF A CONTINUING COLLABORATION WITH THE DEPARTMENT OF ENERGY (DOE) OFFICE OF HIGH ENERGY PHYSICS (HEP), TO FURTHER THE DESIGN OF THE CMB-S4 EXPERIMENT, WHICH IS A COSMIC MICROWAVE BACKGROUND (CMB) STAGE IV FACILITY, AS DEFINED DURING THE 2013 SNOWMASS CONFERENCE. FUNDING WILL SUPPORT, SPECIFICALLY, COMPLETING ASSESSMENT OF ALTERNATIVE INSTRUMENT CONFIGURATIONS WHILE ADVANCING TECHNICAL, BUDGETARY, AND ADMINISTRATIVE DELIVERABLES WITH THE INTENTION OF REACHING READINESS FOR A CONSTRUCTION PROJECT. THIS WORK BUILDS ON PREVIOUS ACTIVITY SUPPORTED BY NSF AND BY DOE/HEP. CMB-S4 USES PROVEN TECHNOLOGIES, BUILDING ON DECADES OF DEVELOPMENT, AND TAKES THEM TO UNPRECEDENTED SCALES. IF SUCCESSFUL, IT WILL PROVIDE A POWERFUL AND UNIQUE MILLIMETER-WAVE SURVEY COVERING 70% OF THE SKY AT HIGH CADENCE, AND WILL CROSS CRITICAL SCIENCE THRESHOLDS IN OUR UNDERSTANDING OF THE ORIGIN AND EVOLUTION OF THE UNIVERSE. THE DISCOVERY SPACE OF THE INFORMATION ENCODED IN CMB POLARIZATION, IN THE GRAVITATIONAL LENSING OF THE CMB, IN OTHER SECONDARY EFFECTS, AND IN YET-TO-BE-DISCOVERED SIGNALS, IS MAXIMIZED BY THE DESIGN OF CMB-S4. UNIQUE DATA PRODUCTS FROM THIS SURVEY WILL BENEFIT THE ENTIRE ASTRONOMICAL COMMUNITY AND A BROADER, MULTI-DISCIPLINARY RESEARCH COMMUNITY IN AN ERA IN WHICH MULTI-WAVELENGTH AND MULTI-MESSENGER ASTRONOMY IS THE PATH TO UNDERSTANDING AND DISCOVERY. CMB-S4 PROVIDES A POWERFUL AND SYNERGISTIC COMPLEMENT TO MAJOR UPCOMING ASTRONOMICAL SURVEYS AND FACILITIES, SUCH AS THE VERA C. RUBIN OBSERVATORY LEGACY SURVEY, THE NANCY GRACE ROMAN SPACE TELESCOPE, AND THE JAMES WEBB SPACE TELESCOPE. THE PROJECT COMES WITH A PLAN TO INCREASE DIVERSITY IN CMB-S4 SCIENCE AND IN STEM FIELDS, DRIVEN BY THREE PRINCIPLES: DIVERSITY OF PARTICIPANTS, DIVERSITY OF TRAINING, AND DIVERSITY OF OUTCOMES. EARLY-CAREER TRAINING INCLUDES ANNUAL FOUR-DAY SUMMER SCHOOLS, AND DEVELOPMENT OF PARTNERSHIPS WITH HBCUS AND URM-FOCUSED INSTITUTIONS TO INCREASE DIVERSITY FURTHER. THIS IS AN IMPORTANT AND TRANSFORMATIVE PROJECT DURING A TIME OF SIGNIFICANT BUDGET UNCERTAINTY. SECOND AND THIRD YEARS OF FUNDING WILL BE RE-EVALUATED AND REPRIORITIZED TOWARDS THE END OF THE FIRST YEAR, PENDING REVIEWS, AVAILABILITY OF FUNDING, AND PHASING OF THE WORK INVOLVED IN THE PROJECT. INTEGRATED GENETIC, OMIC, AND IMMUNOLOGIC STUDIES TO IDENTIFY ENDOTYPES AND NOVEL DRUG TARGETS FOR ASTHMA AND ALLERGIC DISEASES - ABSTRACT ASTHMA AND ALLERGIC DISEASES ARE AMONG THE MOST COMMON CHRONIC DISEASES IN CHILDREN AND ADULTS, COSTING OUR HEALTH CARE SYSTEM OVER $80 BILLION PER YEAR. RATES HAVE BEEN INCREASING OVER THE PAST 40 YEARS AND THERAPEUTIC ADVANCES HAVE BEEN INCREMENTAL. OVER 150 LOCI HAVE BEEN REPORTED IN LARGE GENOME-WIDE ASSOCIATION STUDIES (GWAS) OF ASTHMA AND ALLERGIC DISEASES, BUT THEIR INDIVIDUAL EFFECTS ARE SMALL AND THESE VARIANTS ACCOUNT A SMALL FRACTION OF THE OVERALL GENETIC RISK. MOREOVER, REMARKABLY FEW OF THE GWAS FINDINGS FOR ASTHMA AND ALLERGIC DISEASES HAVE LED TO DISCOVERIES OF CAUSAL VARIANTS OR CAUSAL GENES THAT CONTRIBUTE TO ASTHMA AND ALLERGIC DISEASE PATHOGENESIS. THE LATTER HAS BEEN PARTICULARLY CHALLENGING DUE IN PART TO THE SIGNIFICANT CLINICAL HETEROGENEITY OF THESE DISEASES, AND IN PART TO THE LAG IN THE DEVELOPMENT OF POWERFUL STATISTICAL, MOLECULAR, AND IMMUNOLOGIC TOOLS FOR BRIDGING THE TRAJECTORY FROM GWAS TO GENE DISCOVERY TO BIOLOGY TO TRANSLATION. IN THIS APPLICATION, WE PROPOSE A ROBUST AND COMPREHENSIVE STRATEGY FOR IDENTIFYING CANDIDATE CAUSAL VARIANTS AND THEIR TARGET GENES AT ASTHMA AND ALLERGIC DISEASE-ASSOCIATED LOCI, AND FOR CHARACTERIZING (I) THEIR FUNCTIONAL EFFECTS IN ASTHMA AND ALLERGIC DISEASE-RELEVANT CELLS TYPES, INCLUDING BRONCHIAL EPITHELIAL CELLS, AIRWAY SMOOTH MUSCLE AND LUNG IMMUNE CELLS, AS WELL AS PERIPHERAL IMMUNE CELLS, ALL IN RESTING AND ACTIVATED STATES; (II) THEIR DOWNSTREAM PHENOTYPIC EFFECTS ON BOTH BROAD CATEGORIES OF DISEASE GROUPS AND TRAITS IN THE UK BIOBANK RESOURCE AND ON SPECIFIC ASTHMA AND ALLERGIC DISEASE ENDOTYPES IN DEEPLY PHENOTYPED ETHNICALLY-DIVERSE SUBJECTS PARTICIPATING IN ASTHMA BIRTH COHORTS; AND (III) THEIR IMMUNOLOGIC EFFECTS IN RESTING AND ACTIVATED LUNG LYMPHOCYTES AND MYELOID CELLS AND IN “HUMANIZED LOCUS” BAC-ENGINEERED MOUSE MODELS. THESE GOALS WILL BE ACCOMPLISHED THROUGH A HIGHLY COLLABORATIVE AND SYNERGISTIC PROGRAM THAT INCLUDES TWO PROJECTS, A SERVICE CORE, AND AN ADMINISTRATIVE CORE THAT TOGETHER WILL THAT BRIDGE THE TRAJECTORY FROM GWAS TO TRANSLATION THROUGH HIGHLY INTEGRATED STUDIES BY AN EXCEPTIONAL TEAM OF INVESTIGATORS WITH EXPERTISE IN GENETICS, (EPI)GENOMICS, STATISTICAL GENETICS, AND IMMUNOLOGY. ACHIEVING THESE GOALS WILL ULTIMATELY IDENTIFY NOVEL DRUG TARGETS AND THE INDIVIDUALS MOST LIKELY TO RESPOND, PROVIDING A FRAMEWORK FOR PRECISION MEDICINE AND PERSONALIZED TREATMENT OF ASTHMA AND ALLERGIC DISEASES.

FOLLOWING BIOTERRORISM EVENTS IN 2001, THE NATIONAL INSTITUTE OF ALLERGY AND INFECTIOUS DISEASES ESTABLISHED THE REGIONAL BIOCONTAINMENT LABORATORY (RBL) NETWORK, TO PROMOTE SAFE RESEARCH IN 12 BIOSAFETY LEVEL 3 (BSL3) LABORATORIES. IN TURN, INSTITUTIONS AND UNIVERSITIES HOSTING THESE RBLS SUSTAINED THE MANAGEMENT OF THESE FACILITIES, FOSTERED A STRONG BIOSAFETY CULTURE, AND FUELED RESEARCH INQUIRIES INTO THE MOLECULAR MECHANISMS WHEREBY MICROBES CAUSE HUMAN DISEASE. THE UNIVERSITY OF CHICAGO HOSTS THE REGIONAL BIOCONTAINMENT HOWARD TAYLOR RICKETTS LABORATORY (HTRL) OPERATED BY ITS DEPARTMENT OF MICROBIOLOGY SINCE 2009. WHEN THIS NATION BEGAN TO IMPLEMENT SHUTDOWNS TO PREVENT THE SPREAD OF COVID-19, ESSENTIAL PROFESSIONALS AT THE HTRL CONTINUED KEY MANAGEMENT OPERATIONS AND PIVOTED THEIR EXPERTISE TO STUDY SARS-COV2. THIS MOMENTUM WOULD NOT HAVE BEEN POSSIBLE WITHOUT THE EXISTING EXPERTISE AND DEDICATION OF ENGINEERS, BIOSAFETY OFFICERS, VETERINARY STAFF, SCIENTISTS, AS WELL AS FINANCIAL SUPPORT FROM THE UNIVERSITY. THE GOAL OF THIS PROPOSAL IS TO PRESENT A PLAN TO MAINTAIN THE SAFE OPERATION AND MANAGEMENT OF THE BSL3 SPACE OF THE REGIONAL BIOCONTAINMENT HTRL AND TO CONTINUE TRAINING PROFESSIONALS WHO WILL PROVIDE RESEARCH SUPPORT SERVICES FOR THE DEVELOPMENT OF MEASURES NECESSARY TO MITIGATE AND PROTECT THE PUBLIC'S HEALTH FROM INFECTIOUS DISEASES THAT MAY BE THE RESULT OF INTENTIONAL, ACCIDENTAL, OR NATURALLY OCCURRING HEALTH EMERGENCY. THE BACTERIAL AND VIRAL BIOINFORMATICS RESOURCE CENTER (BV-BRC) - PROJECT SUMMARY THE MISSION OF THE NIH/NIAID BIOINFORMATICS RESOURCE CENTER (BRC) PROGRAM IS TO ACCELERATE BASIC AND APPLIED INFECTIOUS DISEASE RESEARCH BY PROVIDING ACCESS TO CUTTING EDGE BIOINFORMATIC TOOLS, KNOWLEDGEBASES, AND EXPERTISE, ENSURING THAT OUR KNOWLEDGE OF PATHOGENESIS CAN BE TRANSLATED INTO DIAGNOSTICS, THERAPEUTICS AND A PUBLIC HEALTH RESPONSE THAT MITIGATES THE MORBIDITY AND MORTALITY RESULTING FROM INFECTIOUS DISEASES. THE CURRENT NIH/NIAID-FUNDED BACTERIAL AND VIRAL BIOINFORMATICS RESOURCE CENTER (BV-BRC; CONTRACT NO. 75N93019C00076) SUPPORTED THIS MISSION BY PROVIDING A BIOINFORMATICS KNOWLEDGEBASE AND ANALYSIS PLATFORM COVERING ALL BACTERIAL AND VIRAL PATHOGENS. IN RESPONSE TO THE NIAID NOTICE OF FUNDING OPPORTUNITY, RFA-AI-23- 032, OUR PROPOSAL INTENDS TO MAINTAIN, IMPROVE, AND EXPAND THE BV-BRC TO COMBAT FUTURE INFECTIOUS DISEASE THREATS, WHILE MAINTAINING OUR COMMITMENT TO ENHANCE DIVERSITY, EQUITY, INCLUSION, AND ACCESSIBILITY, FOSTERING A MORE INCLUSIVE SCIENTIFIC COMMUNITY, AND ENSURING EQUITABLE ACCESS TO BIOINFORMATICS RESOURCES. BV-BRC WILL SUPPORT BACTERIA, ARCHAEA, VIRUSES, BACTERIOPHAGES, AS WELL AS METAGENOMIC ANALYSES, WITH PARTICULAR EMPHASIS ON THE MICROBIOMES AND VIROMES RELATED TO INFECTIOUS DISEASE AND PUBLIC HEALTH. BV-BRC WILL CONTINUE TO SUPPORT THE BASIC SCIENTIFIC RESEARCH NECESSARY TO UNDERSTAND THE BIOLOGY OF THESE ORGANISMS, THEIR PATHOGENESIS, AND DISEASE PROCESSES; SUPPORT DEVELOPMENT OF DIAGNOSTICS AND THERAPEUTICS TO COMBAT PATHOGENIC ORGANISMS; AND PROVIDE A RAPID RESPONSE FRAMEWORK TO EFFECTIVELY DEAL WITH THE INEVITABLE AND UNPREDICTABLE OUTBREAKS AND PANDEMICS. TO SUPPORT THESE OVERARCHING GOALS, WE PROPOSE TO EXTEND AND ENHANCE BV-BRC THROUGH THE FOLLOWING FOUR KEY ELEMENTS: 1) MAINTAIN AND ENHANCE THE BV-BRC KNOWLEDGEBASE TO SUPPORT EXPONENTIAL GROWTH OF DATA AND USAGE AND PROVIDE INTEGRATED ACCESS TO OMICS DATA, METADATA, ANALYSIS SERVICES AND VISUALIZATION TOOLS, PRIVATE USER WORKSPACE, AND USER DOCUMENTATION TO ALLOW USERS TO ANALYZE PUBLIC AND PRIVATE DATA AND SHARE OR PUBLISH RESULTS; 2) DEVELOP INNOVATIVE TOOLS AND TECHNOLOGIES TO PROVIDE COMPREHENSIVE SERVICES FOR VIRAL AND BACTERIAL BIOINFORMATICS, METAGENOMICS, DRUG DEVELOPMENT, AND DEVELOPING AI-DRIVEN NATURAL LANGUAGE-BASED USER INTERFACE FOR INTERACTING WITH DATA AND TOOLS, WITH EMPHASIS ON IMPROVING USER EXPERIENCE; 3) OFFER CRITICAL BIOINFORMATICS EXPERTISE, OUTREACH, AND TRAINING TO THE COMMUNITY, WITH EMPHASIS ON FOSTERING OPPORTUNITIES FOR STUDENTS AND RESEARCHERS FROM MINORITY AND UNDERSERVED COMMUNITIES BY PROVIDING FREELY ACCESSIBLE TRAINING MATERIAL AND CONDUCTING TRAINING FOR INSTRUCTORS FROM UNDERREPRESENTED INSTITUTIONS, WITH PARTICULAR FOCUS ON MINORITY SERVING INSTITUTIONS (MSIS); AND 4) PROVIDE CUTTING-EDGE SUPPORT TO RAPIDLY RESPOND TO EMERGING NEEDS, OUTBREAKS, AND PANDEMIC PREPAREDNESS BY BUILDING ON THE TOOLS AND PROCEDURES DEVELOPED DURING COVID-19 AND MPOX PANDEMICS AND ENHANCING THEM TO IMPROVE READINESS AND RESPONSE TO FUTURE OUTBREAKS AND PANDEMICS.

A HETEROCHRONIC MODEL FOR BIRTH DEFECTS IN DOWN SYNDROME - PROJECT SUMMARY THE FUNDAMENTAL QUESTION FOR THE FIELD OF DOWN SYNDROME (DS) BASIC RESEARCH IS HOW AN EXTRA COPY OF HUMAN CHROMOSOME 21 (HSA21) TRANSLATES INTO THE ORGAN-SPECIFIC PATHOLOGIES THAT ARE OBSERVED IN THE DS POPULATION. STRUCTURAL PATHOLOGIES IN DS, SUCH AS BRAIN SIZE AND HEART MALFORMATION, ARE PRESENT AT BIRTH, AND ARE GENERALLY ASSOCIATED WITH SMALLER ORGANS OR REDUCED GROWTH. THEREFORE, THEY MUST BE CAUSALLY RELATED TO STRUCTURAL ORGANOGENESIS DEFECTS, REFLECTING EMBRYOLOGIC ORIGINS. A MECHANISTIC UNDERSTANDING OF DS-SPECIFIC ORGANOGENESIS DEFECTS IS LACKING IN MOST CASES, ALTHOUGH A RICH DESCRIPTIVE LITERATURE SETS THE STAGE FOR CONCERTED MECHANISTIC STUDIES. SPECIFICALLY, HOW TRISOMY 21 (T21) CAUSES HEART OR BRAIN MORPHOGENESIS DEFECTS, CAUSING CHD OR INTELLECTUAL DISABILITY, RESPECTIVELY, IS POORLY UNDERSTOOD. THE MOLECULAR NETWORKS IDENTIFIED BY THE PROPOSED WORK, IMPLICATED IN DS ORGAN-SPECIFIC CARDIAC AND NEURONAL PROGENITORS, WILL SERVE AS A TEMPLATE FOR UNDERSTANDING THE MOLECULAR ONTOGENY OF HEART AND BRAIN DEFECTS, AS WELL AS OTHER ORGAN DEFECTS, IN DS. WE HAVE DEFINED HEDGEHOG (HH) SIGNALING AS AN EXPLICIT DEVELOPMENT TIMER DURING MAMMALIAN DEVELOPMENT, REQUIRED FOR MAINTAINING ORGAN-SPECIFIC PROGENITOR CELLS AND DICTATING THEIR DIFFERENTIATION IN TIME AND SPACE, INDEPENDENT FROM DEVELOPMENTAL PATTERNING OR PROLIFERATION. THE CONTROLLED BALANCE BETWEEN ORGAN-SPECIFIC PROGENITORS AND THEIR DIFFERENTIATED COUNTERPARTS IS FUNDAMENTAL TO COMPLEX ORGANOGENESIS. THE MOSKOWITZ LAB FOUND THAT WHEN HH SIGNALING IS ABROGATED IN CARDIAC PROGENITORS, THEY UNDERWENT PRECOCIOUS DIFFERENTIATION, RESULTING IN MORPHOGENESIS FAILURE. SIMILARLY, THE BHATTACHARYYA LAB HAS RECENTLY IDENTIFIED DEFICITS IN THE HH PATHWAY IN NEURONAL PROGENITORS THAT CONTROLS THEIR DIFFERENTIATION. WE PROPOSE THE TRANSFORMATIVE HYPOTHESIS THAT A UNIFYING CAUSE OF BIRTH DEFECTS IN DS IS FAILURE OF HETEROCHRONIC TIMING CONTROL OF ORGAN-SPECIFIC PROGENITOR DIFFERENTIATION, RESULTING IN PRECOCIOUS DIFFERENTIATION, A REDUCTION OF ORGAN-SPECIFIC PROGENITORS, MORPHOGENESIS FAILURE AND BIRTH DEFECTS IN MULTIPLE ORGANS.

LOCATED TWENTY-FIVE MILES SOUTHWEST FROM THE UNIVERSITY OF CHICAGO CAMPUS, ANL IS A FEDERALLY (DEPARTMENT OF ENERGY) OWNED SITE OPERATED BY THE UNIVERSITY OF CHICAGO (UCHICAGO LLC). HTRL IS A CDC CERTIFIED FACILITY OWNED BY THE UNIVERSITY OF CHICAGO AND OPERATED BY THE DEPARTMENT OF MICROBIOLOGY. BIOSAFETY TRAINING AND SURVEILLANCE PROGRAM INVOLVING CLASSROOM AND LABORATORY ACTIVITIES ENSURES THAT STAFF, STUDENTS AND FELLOWS COMPLY WITH STATE AND FEDERAL REGULATIONS TO SAFELY OPERATE FACILITIES AND EXPERIMENTS WITH RG2, RG3 AND SELECT AGENT PATHOGENS. THE HTRL CONSTRUCTION WAS COMPLETED IN 2008. OVER THE LAST 13 YEARS, THE HTRL HAS HOSTED RESEARCH PROGRAMS STUDYING THE BACTERIAL SPECIES YERSINIA, BRUCELLA, COXIELLA, RICKETTSIA, BACILLI, MRSA AND OTHER ESKAPE ORGANISMS, AS WELL AS VIRAL PATHOGENS INCLUDING LOW AND HIGHLY PATHOGENIC INFLUENZA, DENGUE AND MORE RECENTLY SARS-COV2. THE FACILITY CURRENTLY HOSTS RESEARCH PROGRAM ON PLAGUE, ANTHRAX AND MRSA, AS WELL AS A CORE RESEARCH FACILITY FOR SARS-COV2. THE HTRL IS THE LARGEST A-BSL3 HOLDING FACILITY IN THE REGION AND REGULARLY SUPPORTS RESEARCH FROM CONSORTIUM INSTITUTIONS INCLUDING UIC, NORTHWESTERN U AND LOYOLA U. THE HTRL IS FULFILLING ITS GOAL AS A BIOMEDICAL RESEARCH FACILITY THAT CAN RAPIDLY RESPOND TO RESEARCH NEEDS AND SUPPORT BIOMEDICAL RESEARCH ON EMERGING AND RE-EMERGING PATHOGENS, DISCOVERY AND TESTING OF NEW THERAPEUTICS AND VACCINES. THE BUILDING HAS BEEN MAINTAINED IN EXCELLENT WORKING CONDITIONS AND HAS BEEN IN CONSTANT USE SINCE ITS CONSTRUCTION. HOWEVER, WITH THE SWELLING DEMANDS FOR BSL3 AND ABSL3 SPACE AND RESEARCH EXPERTISE, THE DEVELOPMENT OF NEW TECHNOLOGIES FOR THE STUDY OF INFECTIOUS AGENTS, THIS PROPOSAL IS A REQUEST (I) TO UPGRADE SYSTEM COMPONENTS NECESSARY TO MAINTAIN BIOCONTAINMENT, TEMPERATURE/HUMIDITY/PRESSURIZATION REQUIREMENTS, AND SAFE RESEARCH OPERATIONS, AND (II) TO REPLACE RESEARCH EQUIPMENT THAT HAVE EITHER OUTLIVED THE INTENDED USEFUL LIFE OR NO LONGER MEET FACILITY REQUIREMENTS.

PAST MEASUREMENTS OF THIS KIND HAVE PROVIDED PROFOUND INSIGHTS INTO THE NATURE AND ORIGIN OF COSMIC RAYS REVEALING FOR INSTANCE INFORMATION ON ACCELERATION AND CONFINEMENT TIME SCALES AND EXPOSING SOME CONSPICUOUS DISCREPANCIES BETWEEN SOLAR AND COSMICRAY ABUNDANCES. THE MOST DETAILED INFORMATION CURRENTLY AVAILABLE COMES FROM THE ACE/CRIS MISSION BUT IS RESTRICTED TO ENERGIES BELOW A FEW 100 MEV/NUC. HELIX AIMS AT EXTENDING THIS ENERGY RANGE BY OVER AN ORDER OF MAGNITUDE WHERE IN MANY CASES NO FULLY RESOLVED MEASUREMENTS EXIST AND WHERE RELATIVISTIC TIME DILATION AFFECTS THE APPARENT LIFETIME OF RADIOACTIVE CLOCK NUCLEI. THE HELIX MEASUREMENTS WILL PROVIDE ESSENTIAL INFORMATION FOR UNDERSTANDING THE PROPAGATION HISTORY OF COSMIC RAYS IN THE GALAXY. THIS IS CRUCIAL FOR PROPERLY INTERPRETING SEVERAL INTRIGUING ANOMALIES REPORTED IN RECENT COSMIC-RAY MEASUREMENTS PERTAINING TO THE ENERGY SPECTRA OF PROTONS HELIUM AND HEAVIER NUCLEI AND TO THE ANOMALOUS RISE IN THE POSITRON FRACTION AT HIGHER ENERGY. THE PRIMARY SCIENTIFIC GOALS OF THE FULL HELIX PROGRAM ARE: - A HIGH-STATISTICS MEASUREMENT OF THE 10BE/9BE 'CLOCK RATIO' TO ~10 GEV/NUC - A HIGH-STATISTICS MEASUREMENT OF THE 3HE/4HE RATIO TO ~12GEV/NUC - THE FIRST MEASUREMENTS OF 22NE/20NE ABOVE 1 GEV/NUC - THE FIRST MASS-RESOLVED MEASUREMENTS OF 7LI/6LI AND 10B/11B ABOVE 1 GEV//NUC A NUMBER OF SECONDARY GOALS WILL ALSO BE PURSUED INCLUDING THE MEASUREMENT OF SEVERAL OTHER ISOTOPIC AND ELEMENTAL ABUNDANCE RATIOS AND FLUXES THE PROPOSED WORK WILL BE CONDUCTED BY A TEAM OF US SCIENTISTS AND ENGINEERS WITH EXTENSIVE EXPERIENCE IN COSMIC-RAY OBSERVATIONS ON BALLOONS OR IN SPACE AND SPECIFICALLY IN MAGNET SPECTROSCOPY WITH THE SMILI P-BAR AND HEAT PROGRAMS. SIGNIFICANT PARTICIPATION FROM A CREW OF MORE THAN 10 GRADUATE POST-GRADUATE AND UNDERGRADUATE STUDENTS FORMS AN IMPORTANT EDUCATIONAL ELEMENT OF THE PROGRAM.

WHAT IS THE MASS SCALE OF NEUTRINOS? THE SOUTH POLE TELESCOPE (SPT), CURRENTLY EQUIPPED WITH THE SPT-3G CAMERA, PLAYS A UNIQUE ROLE IN THE PURSUIT OF THESE QUESTIONS. THE SPT IS LOCATED AT THE NSF'S AMUNDSEN-SCOTT SOUTH POLE STATION, WHICH IS THE BEST OPERATIONAL SITE ON EARTH FOR MILLIMETER- AND SUBMILLIMETER-WAVELENGTH OBSERVATIONS. FURTHERMORE, THE UNIQUE GEOGRAPHICAL LOCATION OF THE SITE ALLOWS SPT TO OBSERVE TARGETED LOW-GALACTIC-FOREGROUND REGIONS OF THE SKY AT CONSTANT ELEVATION 24 HOURS A DAY, YEAR-ROUND, RESULTING IN THE DEEPEST HIGH-RESOLUTION MAPS OF THE SKY AT THESE WAVELENGTHS. THE SPT ALSO PLAYS A CRITICAL ROLE IN THE EVENT HORIZON TELESCOPE (EHT), A GLOBAL ARRAY OF TELESCOPES TO IMAGE THE EVENT HORIZON AROUND THE BLACK HOLE AT THE CENTER OF THE MILKY WAY GALAXY AND OTHER, MORE DISTANT GALAXIES. SHARING THE SPIRIT OF SCIENTIFIC INQUIRY WILL BE EXTENDED BEYOND THE RESEARCH COMMUNITY THROUGH A WELL-ESTABLISHED EDUCATION NETWORK AT ALL LEVELS OF THE EDUCATION CONTINUUM, FROM EARLY CHILDHOOD THROUGH GRADUATE SCHOOL. THE AWARD WILL INAUGURATE AN INTERNSHIP PROGRAM IN PARTNERSHIP WITH JOLIET JUNIOR COLLEGE, WHICH WILL FUND STUDENTS FROM UNDERREPRESENTED GROUPS IN PAID INTERNSHIP POSITIONS AT SPT INSTITUTIONS. THESE PROGRAMS ARE PART OF THE LARGER SPT INITIATIVE TOWARD BUILDING A MORE DIVERSE WORKFORCE, IN OUR FIELD AND BEYOND. THIS AWARD IS TO SUPPORT MEASUREMENTS OF THE COSMIC MICROWAVE BACKGROUND (CMB) WITH SPT-3G, THE MOST POWERFUL CMB CAMERA IN OPERATION. THE SPT-3G MAPS OF THE TOTAL INTENSITY AND POLARIZATION OF THE CMB SIGNAL ALREADY HAVE AN UNPRECEDENTED COMBINATION OF DEPTH, RESOLUTION, AND SKY COVERAGE, AND THIS AWARD WOULD SUPPORT EXPANDING THE SKY COVERAGE BY OVER A FACTOR OF TWO. THE MEASUREMENTS OF THE CMB TEMPERATURE AND POLARIZATION POWER SPECTRA AND THE CMB LENSING POTENTIAL WITH SPT-3G PLAY A CENTRAL ROLE IN PROBING CURRENT COSMOLOGICAL TENSIONS AND DETERMINING IF THEIR EXPLANATION REQUIRES PHYSICS BEYOND THE ?CDM COSMOLOGICAL MODEL. RECENTLY, A KEY THRUST OF THE SPT RESEARCH PROGRAM HAS BEEN THE USE OF HIGH-RESOLUTION SPT-3G DATA TO REMOVE THE GRAVITATIONAL LENSING SIGNAL FROM THE DEGREE-ANGULAR-SCALE DATA TAKEN WITH THE BICEP ARRAY, TO ENABLE THE DEEPEST SEARCH YET FOR PRIMORDIAL GRAVITATIONAL WAVES (PGW). DELENSING WITH THE SPT-3G DATASET WILL EXTEND THE POWER OF BA TO DETECT PGW BY MORE THAN A FACTOR OF 2.5, ACHIEVING CONSTRAINTS ON THE PRESENCE OF PGW THAT WILL BE UNSURPASSED FOR MANY YEARS. THIS AWARD ALSO ADDRESSES AND ADVANCES THE SCIENCE OBJECTIVES AND GOALS OF NSF?S WINDOWS ON THE UNIVERSE: THE ERA OF MULTI-MESSENGER ASTROPHYSICS PROGRAM. THIS AWARD REFLECTS NSF'S STATUTORY MISSION AND HAS BEEN DEEMED WORTHY OF SUPPORT THROUGH EVALUATION USING THE FOUNDATION'S INTELLECTUAL MERIT AND BROADER IMPACTS REVIEW CRITERIA.- SUBAWARDS ARE PLANNED FOR THIS AWARD.

FOR ANTIGENICALLY VARIABLE PATHOGENS SUCH AS INFLUENZA, PROTECTIVE IMMUNE RESPONSES IMPOSE A MAJOR SELECTIVE PRESSURE ON VIRAL POPULATIONS AND INDIRECTLY INFLUENCE VACCINE STRAIN SELECTION AND VACCINE EFFECTIVENESS. OUR POOR UNDERSTANDING OF THE GENERATION AND MAINTENANCE OF PROTECTIVE IMMUNITY TO INFLUENZA HINDERS VACCINE DEVELOPMENT AND THE ACCURACY OF EVOLUTIONARY FORECASTS. ANTIBODY TITERS TO THE HEMAGGLUTININ (HA) SURFACE PROTEIN WERE ESTABLISHED AS A CORRELATE OF PROTECTION 50 YEARS AGO, AND MORE RECENT EVIDENCE SHOWS MANY ANTI-HA ANTIBODIES DIRECTLY AND INDIRECTLY CONTRIBUTE TO VIRAL NEUTRALIZATION. HOWEVER, HA TITERS REMAIN ONLY MODERATELY PREDICTIVE OF AN INDIVIDUAL’S RISK OF INFECTION ON EXPOSURE, AND THE CONTRIBUTIONS OF OTHER IMMUNE RESPONSES ARE LESS WELL UNDERSTOOD. UNDERSTANDING THE CAUSES IN ADDITION TO CORRELATES OF PROTECTION COULD INCREASE THE ACCURACY OF FORECASTS OF VIRAL FITNESS AND PROVIDE RELIABLE ENDPOINTS FOR VACCINE DEVELOPMENT. HERE, WE PROPOSE COMPLEMENTARY APPROACHES TO IDENTIFY THE CORRELATES AND DRIVERS UNDERLYING PROTECTION FROM INFECTION AND HETEROGENEITY IN VACCINE RESPONSES. WE WILL INTEGRATE DIVERSE VARIABLES, INCLUDING INFECTION AND VACCINATION HISTORY, BASELINE ANTIGEN-SPECIFIC AND ANTIGEN-AGNOSTIC IMMUNE STATES, INTRINSIC CHARACTERISTICS INCLUDING AGE, SEX, AND BODY MASS TO PREDICT RESPONSES TO INFLUENZA VACCINATION AND EXTRACT MECHANISTIC INSIGHT. IN ORDER TO ADDRESS OUR SPECIFIC AIMS, WE WILL LEVERAGE DATA FROM EXISTING, LONGITUDINAL STUDIES OF IMMUNE PARAMETERS FOLLOWING INFLUENZA VIRUS INFECTIONS AND VACCINATION IN HUMANS. FIRST WE WILL USE COMPUTATIONAL AND MULTIMODAL SINGLE-CELL APPROACHES TO INVESTIGATE HOW VACCINATION AND INFECTION IMPACT HOST IMMUNE STATUS. EMERGING EVIDENCE, INCLUDING OUR OWN DATA, SUGGESTS THAT VACCINATION AND INFECTION CAN ESTABLISH NEW ANTIGEN-AGNOSTIC IMMUNE SET POINTS THAT AFFECT FUTURE VACCINE RESPONSES. NEXT WE PROPOSE TO INTEGRATE COMPLEMENTARY COMPUTATIONAL APPROACHES, SPANNING MACHINE LEARNING, CAUSAL MEDIATION ANALYSIS, AND MECHANISTIC MODELING TO PREDICT AND DEVELOP CAUSAL MECHANISTIC INSIGHT INTO VACCINE RESPONSIVENESS AND PROTECTION FROM SEVERE AND MILD INFECTION. WE WILL DEVELOP AND DISTRIBUTE A SUITE OF ACCOMPANYING TOOLS TO MAKE THESE NOVEL APPROACHES ACCESSIBLE TO BENCH AND COMPUTATIONAL BIOLOGISTS. IMPROVED PREDICTION OF IMMUNE RESPONSES, ESPECIALLY PROTECTIVE IMMUNE RESPONSES, COULD LEAD TO MORE EFFECTIVE VACCINATION STRATEGIES THAT MITIGATE VACCINE FAILURE IN DIFFERENT SUBPOPULATIONS AND IMPROVE THE PUBLIC HEALTH IMPACT OF INFLUENZA VACCINATION. THE METHODS AND TOOLS THAT WE DEVELOP CAN PROVIDE FOUNDATIONAL FRAMEWORKS TO DISSECT RESPONSES TO OTHER VACCINES AND PATHOGENS.

INTRACELLULAR ELECTROPHYSIOLOGY: AN ELECTROCHEMICAL ATLAS OF ORGANELLES - PROJECT SUMMARY THE LONG-TERM OBJECTIVE OF THIS PROPOSAL IS TO BUILD AN ELECTROCHEMICAL ATLAS OF ORGANELLES TO GUIDE THE RATIONAL MANIPULATION OF INTER-ORGANELLE CONTACTS IN THE CONTEXT OF NEURODEGENERATIVE DISEASES. A NEW MODE OF INTRACELLULAR COMMUNICATION IS EMERGING AT THE LEVEL OF ORGANELLES, WHEREBY THE MEMBRANES OF TWO JUXTAPOSED ORGANELLES ARE PHYSICALLY CONNECTED, VIA PROTEIN-PROTEIN INTERACTIONS ON THEIR CYTOPLASMIC FACES, REFERRED TO AS INTER-ORGANELLE CONTACTS. IONS AND SMALL MOLECULES ARE ACTIVELY TRANSFERRED FROM ONE ORGANELLE TO THE OTHER ACROSS THESE CONTACTS, TRAVERSING TWO SEALED MEMBRANES. INTER-ORGANELLE CONTACTS ARE VITAL TO CELL FUNCTION, TISSUE HOMEOSTASIS AND PHYSIOLOGY BECAUSE THEY REGULATE PROCESSES RANGING FROM LIPID METABOLISM TO APOPTOSIS. HOWEVER, WE STILL DO NOT KNOW WHAT SIGNALS INITIATE CONTACT FORMATION OR WHAT SWITCHES ON CHEMICAL TRANSPORT ACROSS CONTACTS, NOR CAN WE DISCRIMINATE BETWEEN FUNCTIONAL AND DYSFUNCTIONAL CONTACTS. HENCE, ALTHOUGH WE KNOW OF SPECIFIC MUTATIONS IN PROTEINS THAT DISRUPT CONTACT, LEADING TO DIVERSE NEURODEGENERATIVE DISEASES, WE STILL DO NOT KNOW HOW TO RESTORE THESE CONTACTS AND TREAT THOSE DISEASES. I POSIT THAT THE ELECTROCHEMICAL STATES OF ORGANELLES, ALONE AND IN CONTACT, WILL INFORM WHICH PATHWAYS AND MOLECULES SHOULD BE TARGETED TO RECTIFY ABERRANT CONTACTS IN DISEASE STATES. MY RATIONALE IS THAT, IF WE ABSTRACT OUT THE MOLECULAR DETAILS, INTER-ORGANELLE CONTACTS RESEMBLE NEURONAL SYNAPSES. EVEN IN SYNAPSES, IONS FLOW ON CUE ACROSS TWO SEALED, ABUTTING MEMBRANES. JUST AS ION-TRANSPORT MECHANISMS ACROSS NEURONAL MEMBRANES WERE REVEALED BY HODGKIN AND HUXLEY’S ELECTROCHEMICAL MODEL, AN ANALOGOUS MODEL OF ORGANELLE MEMBRANES WILL REVEAL ION FLOW MECHANISMS ACROSS INTER-ORGANELLE CONTACTS AND WHICH SPECIFIC FLOWS ARE IMPACTED IN DISEASE. I PROPOSE TO BUILD AN ELECTROCHEMICAL ATLAS OF ORGANELLES AS A UNIVERSAL REFERENCE TO STUDY CONTACTS IN HEALTH AND DISEASE. THIS ATLAS WILL BE A COMPENDIUM OF EQUATIONS COMPRISING ELECTROCHEMICAL MODELS OF MAJOR ORGANELLES, ALONE AND IN CONTACT. BY ENABLING US TO DISCRIMINATE NORMAL AND ABERRANT CONTACTS, I ENVISAGE THE ATLAS WILL REVEAL COMMON PATHWAYS ACROSS DISEASES THAT CAN BE TARGETED TO RESTORE CONTACTS WITH IMPACTED ION FLOWS. THE INABILITY TO ASSAY IONS OR VOLTAGE IN ORGANELLES HAS PREVENTED THE DEVELOPMENT OF ELECTROCHEMICAL MODELS OF THEIR MEMBRANES. OVER THE LAST DECADE, MY LAB DEVELOPED A CHEMICAL PLATFORM TO QUANTIFY IONS AND VOLTAGE IN ORGANELLES. BY INTEGRATING ELECTROPHYSIOLOGY TO THIS PLATFORM, I PROPOSE TO NOW MAP OUT THE ELECTROCHEMICAL CHARACTERISTICS OF ORGANELLE MEMBRANES IN ISOLATION AND IN CONTACT, AND MAKE AN ELECTROCHEMICAL ATLAS OF ORGANELLES. WE WILL APPLY THE ATLAS TO ELUCIDATE HOW CA2+ FLOW ACROSS ABERRANT CONTACTS BETWEEN THE ENDOPLASMIC RETICULUM AND THE LYSOSOME CAN BE RECTIFIED TO RESTORE LYSOSOMAL CA2+ IN PARKINSONISM. DYSREGULATED LYSOSOMAL CA2+ IS A COMMON FACTOR ACROSS MANY NEURODEGENERATIVE DISEASES AND THE VALUE OF THE ELECTROCHEMICAL ATLAS IS ITS PIONEERING ABILITY TO REVEAL COMMON PATHWAYS THAT CAN BE TARGETED FOR TREATMENT IN A DISEASE CROSS-CUTTING MANNER.

PHASE I OF THE JUSTICE COMMUNITY OVERDOSE INNOVATION NETWORK (JCOIN) CREATED UNDER THE NIDA HEAL INITIATIVE FUNDED A SERIES OF INVESTIGATIONS TARGETING KEY STAGES OF THE SUBSTANCE USE TREATMENT CONTINUUM AS PEOPLE WITH SUBSTANCE USE DISORDERS INTERACT WITH THE CRIMINAL LEGAL SYSTEM. TOGETHER WITH THE NEW RESEARCH HUBS THAT WILL BE IDENTIFIED DURING JCOIN PHASE II, THESE STUDIES WILL GENERATE A LARGE AMOUNT OF RICH DATA FROM THE MOST VULNERABLE POPULATIONS IN THE U.S. OPENING OPPORTUNITIES FOR ANALYSES BEYOND THE PRIMARY STUDY AIMS TO MAXIMIZE THE IMPACT OF INVESTMENT INTO SCIENTIFIC RESEARCH – ONE OF THE PRIMARY FOCI OF THE METHODOLOGY AND ADVANCED ANALYTICS RESOURCE CENTER (MAARC). THE MAARC WILL ALSO PROVIDE OVERALL LEADERSHIP AND COORDINATION OF CENTRALIZED DATA MANAGEMENT; DATA INFRASTRUCTURE SUPPORT; PROVIDE RESOURCES FOR ADVANCED ANALYTICAL TECHNIQUES ACROSS THE NETWORK; RESEARCH DYNAMIC CHANGES IN POLICY AND PRACTICE IN CRIMINAL-LEGAL/COMMUNITY SERVICE SETTINGS; AND CONDUCT NOVEL STUDIES APPLYING CUTTING EDGE ANALYTICAL TECHNIQUES TO EXISTING DATA AND DATA COLLECTED ACROSS THE JCOIN NETWORK: AIM 1. THE ADMINISTRATIVE CORE WILL INTEGRATE EXISTING ORGANIZATIONAL AND COLLABORATIVE INFRASTRUCTURE TO PROVIDE OVERARCHING RESOURCES AND ORGANIZATIONAL STRUCTURE AND MANAGEMENT TO THE CENTER; AIM 2. THE DATA AND ANALYTICS SUPPORT CORE WILL CONTINUE TO FACILITATE THE COLLECTION, ARCHIVING, AND STORAGE OF DATA COLLECTED BY THE RESEARCH HUBS, ASSIST WITH CLOSEOUT AND CURATION OF PHASE 1 DATA AND FACILITATE INTERNAL AND EXTERNAL DATA SHARING ACROSS BOTH JCOIN PHASES; AIM 3. THE TECHNICAL ASSISTANCE CORE WILL SUPPORT ON-DEMAND TECHNICAL ASSISTANCE (CONSULTATION) AND PROVIDE APPROPRIATE RESOURCES TO JCOIN PHASE I PIS, JCOIN PHASE II RESEARCHERS, AND SCIENTISTS ACROSS NIDA AND BEYOND; AIM 4. THE SURVEY CORE WILL LEVERAGE NORC’S EXPERTISE TO SUPPORT NATIONAL SURVEYS ABOUT TOPICS BROADLY RELEVANT TO SUBSTANCE USE, STIGMA, BEHAVIORAL HEALTH, CRIMINAL-LEGAL SYSTEMS, AND PUBLIC HEALTH WITH OPPORTUNITIES FOR SPECIALIZED SURVEYS OF CRIMINAL JUSTICE SYSTEMS AS WELL AS MODIFIED REFERRAL SAMPLING OF AMERISPEAK ADJACENT COMMUNITY MEMBERS; AIM 5. THE GEOSPATIAL CORE WILL BUILD UPON POPULAR GEOSPATIAL TOOLS, DATA AND POLICY SCANS FROM PHASE 1 TO SUPPORT INNOVATIVE GEOSPATIAL ANALYSIS THAT ADDRESS IMPORTANT GAPS IN THE ENVIRONMENTAL CONTEXT OF OVERDOSES IN CRIMINAL LEGAL INVOLVED COMMUNITY MEMBERS; AIM 6. THE MODELING CORE WILL DEVELOP AN AGENT-BASED NETWORK MODELING FRAMEWORK TO CONDUCT EXPERIMENTS OF JCOIN TRIALS AND OTHER IN-SILICO TRIALS GUIDED BY JCOIN INVESTIGATORS AND OTHER PRACTITIONERS USING MULTIPLE EXISTING AND NEW DATA SOURCES; AIM 7. THE POLICY LANDSCAPE CORE WILL INVENTORY AND CHARACTERIZE MAJOR POLICY/LEGAL ENVIRONMENTAL CHANGES WITH SPECIFIC FOCUS ON MEDICAID AT THE STATE AND NATIONAL LEVEL AND PRODUCE DATASETS AND RELATED RESOURCES THAT CAN BE USED BY JCOIN RESEARCHERS AND THE LARGER NIDA RESEARCH COMMUNITY. THIS STUDY IS PART OF THE NIH’S HELPING TO END ADDICTION LONG-TERM (HEAL) INITIATIVE TO SPEED SCIENTIFIC SOLUTIONS TO THE NATIONAL OPIOID PUBLIC HEALTH CRISIS.
