THE ACADEMIC HOME OF THE CENTER IS THE GRADUATE SCHOOL AT HOWARD UNIVERSITY. THE PURPOSE IS TO ENHANCE DEVELOPING STUDENTS THROUGH EDUCATION, AND WITH INTERNSHIPS AT NOAA, TO BE A DIVERSE CADRE OF NEW PROFESSIONALS, REPRESENTATIVE OF THE NATIONS DEMOGRAPHY, AND ELIGIBLE CANDIDATES FOR THE FUTURE NOAA MISSION ENTERPRISE WORKFORCE. EXPANDQISE: TRACK 2: A QUANTUM SCIENCE EDUCATION AND RESEARCH PROGRAM FOR HBCUS: EXOTIC PHYSICS AND APPLICATIONS OF SOLID-STATE QUBITS -NON-TECHNICAL DESCRIPTION: QUANTUM INFORMATION SCIENCE AND ENGINEERING (QISE) IS WIDELY RECOGNIZED AS A KEY DRIVER FOR REALIZING GAME-CHANGING SCIENTIFIC BREAKTHROUGHS AND TRANSFORMATIVE NEW QUANTUM COMPUTING, SENSING, AND COMMUNICATION TECHNOLOGIES FOR ENSURING NATIONAL SECURITY AND MAINTAINING THE ECONOMIC COMPETITIVENESS OF THE U.S. IT IS NECESSARY TO BUILD PARTNERSHIPS BETWEEN ACADEMIA, NATIONAL LABORATORIES, AND INDUSTRY IN ORDER TO EFFECTIVELY ADVANCE THE QISE FOREFRONT BY ENGAGING A DIVERSE WORKFORCE. HISTORICALLY BLACK COLLEGES AND UNIVERSITIES (HBCUS), HOWEVER, LACK THE NEEDED FACULTY STRENGTH AND LABORATORY INFRASTRUCTURE FOR QISE TRAINING OF STUDENTS. THIS FIVE-YEAR PROJECT AIMS TO FILL THIS CRITICAL GAP BY ESTABLISHING AN EXPANSIVE QISE RESEARCH AND TRAINING PROGRAM AT HOWARD UNIVERSITY (HU) IN CLOSE PARTNERSHIP WITH NORTHEASTERN UNIVERSITY, WHERE QUANTUM RESEARCH AND EDUCATION PROGRAMS ARE WELL-ESTABLISHED. COLLABORATING NATIONAL LABORATORIES AND INDUSTRIES ARE: NATIONAL INSTITUTE OF STANDARDS AND TECHNOLOGY (NIST), NATIONAL RENEWABLE ENERGY LABORATORY (NREL), IBM, QUANTUM DESIGN, AND QNAMI. THE SCIENTIFIC FOCUS OF THE PROJECT IS ON MODELING, DISCOVERY, UNDERSTANDING, AND APPLICATIONS OF QUANTUM BITS OR QUBITS, WHICH ARE THE KEY COMPONENTS OF THE QISE ECOSYSTEM, MUCH AS THE CLASSICAL BITS UNDERLIE THE MODERN ELECTRONICS REVOLUTION. THIS EFFORT WILL ALSO PROVIDE A ROADMAP FOR OTHER HBCUS AND MINORITY SERVING INSTITUTIONS (MSIS) THROUGHOUT THE U.S. TO PROPEL THEMSELVES TOWARD LEADERSHIP ROLES IN QISE. TECHNICAL DESCRIPTION: THIS PROJECT IS PURSUING A VIBRANT QISE RESEARCH AND TRAINING PROGRAM AT HU IN CLOSE PARTNERSHIP WITH NORTHEASTERN UNIVERSITY AND COLLABORATIONS INVOLVING NATIONAL LABORATORIES (NIST AND NREL), AND INDUSTRY (IBM, QUANTUM DESIGN, AND QNAMI). THE RESEARCH AND TRAINING ACTIVITIES ARE CENTERED AROUND THREE HIGHLY INTERLINKED THRUSTS. (1) INVESTIGATION OF HYBRID STRUCTURES OF QUBITS INTERACTING WITH ELECTROMAGNETIC FIELDS, SPINS, IONS, QUANTUM DOTS, AND OTHER SYSTEMS TO GAIN A DEEPER UNDERSTANDING AND ADVANCEMENT OF QUANTUM SENSING AND TRANSDUCTION TECHNOLOGIES. (2) RATIONAL DESIGN AND IN-DEPTH EVALUATION OF DEFECT QUBITS IN TWO-DIMENSIONAL HOST MATERIALS, INCLUDING EFFECTS OF SPIN-ORBIT COUPLINGS, PHONONS, NUCLEAR SPINS, AND TEMPERATURE. AND (3), LOW-TEMPERATURE NANOSCALE MAPPING AND EXPLORATION OF QUANTUM SYSTEMS USING A COMMERCIAL CRYOGENIC SCANNING DIAMOND-NITROGEN-VACANCY MAGNETOMETER (SNVM) - THE FIRST OF ITS KIND AT AN MSI IN THE US?WITH UNPRECEDENTED RESOLUTION AND SENSITIVITY FOR UNRAVELING THE FUNDAMENTALS OF EXOTIC CHARGE AND SPIN PHENOMENA AND ACHIEVING IMPROVED QUBIT DESIGNS. UNDERGRADUATE, GRADUATE, AND POSTDOCTORAL STUDENTS AND FACULTY AT HU ARE PARTICIPATING VIGOROUSLY IN EXPERIMENTAL AND THEORETICAL RESEARCH AND TRAINING ACTIVITIES, CUTTING ACROSS ALL THREE THRUSTS IN THE PROJECT. QISE WORKFORCE DEVELOPMENT EFFORTS INCLUDE: WORKSHOPS INVOLVING HU STUDENTS AND FACULTY TO IGNITE AWARENESS ABOUT VARIOUS QISE TRAINING PROGRAMS AT THE NATIONAL LABS AND R1 UNIVERSITIES; OPPORTUNITIES TO COLLABORATE WITH NATIONAL AND INDUSTRY PARTNERS, INCLUDING ACCESS TO IBM CLOUD FOR QUANTUM COMPUTING AND NORTHEASTERN UNIVERSITY?S INTEGRATED EQUAL PLATFORM FOR HANDS-ON TRAINING IN GROWING, FABRICATING, AND CHARACTERIZING QUBIT MATERIALS AND DEVICES; AND, DEVELOPMENT OF NEW QISE COURSES TO SUPPORT A PROFESSIONAL QISE MASTER?S DEGREE PROGRAM AT HU. OUTREACH ACTIVITIES INCLUDE THE CREATION OF A WEB PORTAL TO HIGHLIGHT RECENT QISE DISCOVERIES AND VIDEO RECORDINGS OF MONTHLY PROJECT SEMINARS AND E-SUMMER SCHOOLS TO EDUCATE AND ENGAGE THE PUBLIC IN QISE TOPICS. THIS PROJECT IS JOINTLY FUNDED BY THE OFFICE OF MULTIDISCIPLINARY ACTIVITIES (MPS/OMA) AND TECHNOLOGY FRONTIERS PROGRAM (TIP/TF). ELUCIDATING OLFACTORY-BASED EPIGENETIC MEDIATION OF SOCIAL CONTEXTS ON STRESS RESPONSE ACROSS LIFE SPAN IN LOW SES INNER-CITY MINORITY POPULATIONS - ABSTRACT THERE IS SUBSTANTIAL ETHNIC AND RACIAL INEQUITY IN THE BURDEN OF SOCIAL ADVERSITIES ACROSS LIFE SPAN, AND DISPARITIES IN SEVERAL ADULT CHRONIC DISEASES CAN BE TRACED TO SOCIAL INEQUALITIES EXPERIENCED IN CHILDHOOD. SOCIAL ADVERSITIES SUCH AS POVERTY, HARSH PARENTING, NEIGHBORHOOD DISORGANIZATION, FAMILY INSTABILITY, AND PARENTAL INCARCERATION ARE PARTICULARLY PERVASIVE IN INNER-CITY, AFRICAN AMERICAN (AA) POPULATIONS; AND CAN HAVE SUBSTANTIAL IMPACT ON BIOLOGICAL PROCESSES THAT PUT THEM AT RISK FOR CHRONIC STRESS DISORDERS (E.G. POSTTRAUMATIC STRESS DISORDER, PTSD) AND METABOLIC DISEASES. PREVIOUSLY, WE OBSERVED THAT OLFACTORY BULB (OB) VOLUMES WERE SUBSTANTIALLY REDUCED IN AA ADULTS WHO DEVELOPED PTSD FOLLOWING SEVERE CHILDHOOD ADVERSITIES, COMPARED TO THOSE WITH SIMILAR EXPOSURES WHO DID NOT DEVELOP PTSD, WHICH IS CONGRUENT WITH ANIMAL STUDIES SHOWING THAT MATERNAL DEPRIVATION REDUCED OB SIZE. YET HOW THESE SOCIAL EXPOSURES BECOME TRANSLATED INTO CHRONIC HEALTH DISORDERS, IS UNCLEAR. EPIGENETIC FACTORS (I.E. MODIFICATIONS TO THE GENOME THAT ARE NOT CHANGES IN NUCLEOTIDE SEQUENCE) HAVE BEEN POSITED TO PLAY CRITICAL ROLES IN MEDIATING THE IMPACT OF ENVIRONMENTAL EXPOSURES ON HEALTH, DUE TO THEIR INFLUENCE ON DEVELOPMENTAL PLASTICITY AND LONG-TERM FUNCTIONAL BIOLOGY. OUR PROPOSED STUDY BUILDS UPON OUR R21 STUDY IN INNER-CITY AA POPULATIONS WHICH REVEALED THAT GROWTH ARREST SPECIFIC 5 (GAS5), A NON-CODING RNA (LNCRNA) LIKELY PLAYS AN EPIGENETIC ROLE AS A DECOY, DIVERTING GLUCOCORTICOIDS FROM BINDING TO GLUCOCORTICOID RESPONSE ELEMENTS (GRE) IN THE PROMOTER REGIONS OF GENES THAT RESPOND TO GLUCOCORTICOIDS AND PREVENTING DOWNSTREAM MOLECULAR AND PHYSIOLOGICAL EFFECTS. AFRICAN AMERICANS WITH ELEVATED GAS5 LEVELS, HAD LARGER OB VOLUMES, LOWER AFTERNOON CORTISOL LEVELS AND LOWER SYMPATHETIC AROUSAL INDEPENDENT OF BURDEN OF NEIGHBORHOOD DISORDER AND PERCEIVED SOCIAL STRESS AND RACIAL DISCRIMINATION. HOWEVER, OUR STUDIES ALSO REVEALED THAT SOCIAL CONNECTEDNESS AND DAILY SPIRITUAL EXPERIENCE SCALE ALSO MODERATED A BROAD SPECTRUM OF STRESS RESPONSIVE BEHAVIORS (E.G. PERCEIVED STRESS, AFFECT, SLEEP DISRUPTIONS, RISK TAKING, AND RESILIENCE), THEREBY PROVIDING A STRONG JUSTIFICATION TO INVESTIGATE GENOME-WIDE EPIGENOMIC MECHANISMS OF RESPONSE TO SOCIAL ADVERSITIES. AS A RESULT, WE PROPOSE A 5-YEAR PROSPECTIVE STUDY INVOLVING GENOME-WIDE NONCODING RNA PROFILING OF 300 AA WITH DIMENSIONAL DIFFERENCES IN CHILDHOOD SOCIAL ADVERSITIES. OUR SPECIFIC AIMS ARE: (1) CONDUCT BASELINE MICRORNA (MIRNA), LNCRNA AND MRNA PROFILING IN OLFACTORY NEURONS (ON) OF AA COHORTS TO EXAMINE ASSOCIATIONS BETWEEN NONCODING RNA (NCRNA) AND CHILDHOOD SOCIAL ADVERSITIES; (2) QUANTIFY BASELINE ASSOCIATIONS BETWEEN NCRNA LEVELS, PERCEIVED SOCIAL STRESS AND RACIAL DISCRIMINATION, SOCIAL CONNECTEDNESS, SPIRITUAL EXPERIENCE SCALE, AND BOTH BEHAVIORAL AND NEUROPHYSIOLOGIC MEASURES OF STRESS; AND (3) INVESTIGATE MEDIATIONAL ROLES OF NCRNA ON THE PREDICTIVE INFLUENCES OF CUMULATIVE EXPOSURES TO NEIGHBORHOOD STRESS, POVERTY, SOCIAL STRESS, PERCEIVED DISCRIMINATION AND OTHER SOCIAL DISADVANTAGES ON 12-MONTH TRAJECTORY IN STRESS RESPONSE BEHAVIORS. OUR OVERARCHING HYPOTHESIS IS THAT INTERACTIONS BETWEEN MIRNA AND LNCRNA WILL PARTIALLY MEDIATE EFFECTS OF THESE ADVERSE SOCIAL CONTEXTS (E.G. POVERTY, NEIGHBORHOOD DISORGANIZATION, FAMILY INSTABILITY, AND PARENTAL INCARCERATION) ON BIOLOGICAL PROCESSES RELATED TO STRESS RESPONSE ( E.G. GR SIGNALING, IMMUNE SIGNALING, CIRCADIAN MOLECULAR ALTERATIONS, AND ELEVATED SYMPATHETIC TONE) AND STRESS RESPONSIVE BEHAVIORS (PERCEIVED STRESS, PSYCHIATRIC, SLEEP DISRUPTIONS, RISK TAKING, AND RESILIENCE). SITUATED IN THE VIBRANT DC, MARYLAND, AND VIRGINIA (DMV) AREA, THE TWO INSTITUTIONS COLLECTIVELY HAVE A DIRECT IMPACT ON A POPULATION OF 4 MILLION HISTORICALLY UNDERSERVED AFRICAN AMERICANS IN THE AREA, WITH OVER 2 MILLION BLACK RESIDENTS IN THE METROPOLITAN OF WASHINGTON DC AND BALTIMORE. FOCUSED ON DMV REGION, THE INITIATIVE STRIVES TO ADDRESS HEALTHCARE DISPARITIES IN THIS DIVERSE AND SIGNIFICANT DEMOGRAPHIC. HOWARD UNIVERSITY AND JOHNS HOPKINS UNIVERSITY POSSESS DISTINCTIVE STRENGTHS THAT COMPLEMENT EACH OTHER IN ACHIEVING THE PROPOSED GOAL THROUGH INTEGRATED RESEARCH, RESEARCH EDUCATION, AND OUTREACH. THE PRIMARY OBJECTIVES ARE TO ESTABLISH AND SUSTAIN COLLABORATIVE CANCER RESEARCH PROGRAMS WITH AN EMPHASIS ON HEALTH EQUITY, TO BOOST CANCER RESEARCH EDUCATION PROGRAMS AT HU, AND TO ADVANCE CANCER CARE FOR TRADITIONALLY UNDERSERVED POPULATIONS THROUGH OUTREACH ACTIVITIES. THE H2CAREE INITIATIVE COMPRISES OF FOUR CORES (ADMINISTRATIVE, OUTREACH, RESEARCH EDUCATION, AND PLANNING & EVALUATION); TWO FULL RESEARCH PROJECTS; ONE PILOT RESEARCH PROJECT; AND ONE SHARED RESOURCE CORE (DATA SCIENCE). WITH THESE ASSETS AND ACTIVITIES, THE PARTNERSHIP WILL: 1) THROUGH RESEARCH PROJECTS, THE SHARED RESOURCE CORE, AND THE RESEARCH EDUCATION CORE, INCREASE CANCER RESEARCH AND RESEARCH EDUCATION CAPACITY AT HU; 2) THROUGH THE RESEARCH PROJECTS, IMPROVE RESEARCH PROGRAMS FOCUSING ON CANCER HEALTH DISPARITY AT SKCCC; 3) THROUGH THE RESEARCH PROJECT AND THE RESEARCH EDUCATION CORE, INCREASE THE NUMBER OF INVESTIGATORS AND STUDENTS CONDUCTING CANCER HEALTH DISPARITIES RESEARCH; 4) THROUGH THE RESEARCH EDUCATION CORE AND OUTREACH CORE, PROMOTE THE DIVERSITY OF THE CANCER RESEARCH WORKFORCE BY INCREASING THE NUMBER OF AFRICAN AMERICAN STUDENTS PURSUING CANCER RESEARCH; AND 5) THROUGH THE OUTREACH CORE, DEVELOP AND IMPLEMENT CANCER OUTREACH AND EDUCATION INITIATIVES THAT BENEFIT BOTH LOCALLY, I.E. THE DMV REGION, AND NATIONALLY. ADDITIONALLY, THE PLANNING AND EVALUATION CORE WILL CONTINUALLY ASSESS/EVALUATE OUTCOMES OF THE CORES, RESEARCH PROJECTS, AND SHARED RESOURCES. IN VIVO MRI MEASURES OF BRAIN METABOLISM IN TRAUMATIC BRAIN INJURY - PROJECT SUMMARY TRAUMATIC BRAIN INJURY (TBI) REPRESENTS A MAJOR PUBLIC HEALTH CONCERN IN THE UNITED STATES. TBI CAN RESULT IN LONG-TERM NEUROLOGICAL COMPLICATIONS, INCLUDING NEURODEGENERATION, BEHAVIORAL DYSFUNCTION, DEPRESSION AND EPILEPSY THAT SERIOUSLY AFFECT PATIENT QUALITY-OF-LIFE. BRAIN GLUCOSE UTILIZATION HAS BEEN FOUND DECREASED IN 60% OF PATIENTS WITH CHRONIC TBI AND IS ONE OF THE PROGNOSTIC INDICATORS FOR THE LONG-TERM OUTCOME OF TBI. BOTH ANIMAL AND HUMAN STUDIES HAVE IDENTIFIED THAT THE NEUROVASCULAR UNCOUPLING BETWEEN CEREBRAL BLOOD FLOW (CBF) AND BRAIN TISSUE ENERGY DEMANDS IS A KEY FACTOR FOR CEREBRAL METABOLIC CRISIS IN THE TBI BRAIN. THE NEUROVASCULAR UNCOUPLING RESULTS IN ENERGY MISMATCH THAT DISRUPTS NORMAL NEUROGLIAL FUNCTION AND ARRESTS REPAIRING PROCESS. DESPITE NUMEROUS CLINICAL TRIALS ON POTENTIAL THERAPIES, THERE IS NO U.S. FOOD AND DRUG ADMINISTRATION APPROVED DRUG THERAPY FOR THE TREATMENT OF TBI. ONE OF THE REASON CLINICAL TRIALS FAILED IS DUE TO THE INABILITY THAT CAN ACCURATELY EVALUATE THE REGION-SPECIFIC PERTURBATIONS OF BRAIN GLUCOSE METABOLISM AND DETERMINE IF A TREATMENT CAN RESTORE NEUROVASCULAR COUPLING IN THE TBI BRAIN. EXISTENT GLUCOSE MEASURING TECHNIQUES, INCLUDING MICRODIALYSIS, CONTINUING BLOOD GLUCOSE MONITORING, SPECTROSCOPY, OR POSITRON EMISSION TOMOGRAPHY (PET) WHICH USES TRACER RADIOISOTOPES, PROVIDE INSUFFICIENT RESOLUTION TO DETERMINE REGION-SPECIFIC GLUCOSE UTILIZATION IN THE BRAIN. IN THIS PROPOSAL, WE DETERMINE TO ASSESS THE UTILITY OF A NOVEL MRI-BASED CHEMICAL EXCHANGE SATURATION TRANSFER (CEST) IMAGING, TO PROVIDE THE NEEDED HIGH-RESOLUTION FOR MEASURING REGION-SPECIFIC METABOLISM FOR TBI. THE GLUCOSE DETECTING CEST MRI (GLUCOCEST) MEASURES BRAIN GLUCOSE BY DETECTING THE EXCHANGEABLE PROTON SIGNALS OF GLUCOSE WITHOUT USING RADIOISOTOPES AND GENERATES GLUCOSE MAPPING IN A RESOLUTION >100 TIMES HIGHER THAN PET SCANS. OUR PRELIMINARY AND PUBLISHED DATA HAVE DEMONSTRATED THAT GLUCOCEST MAY BE FEASIBLE TO DETECT THE DELAYED HYPOMETABOLISM OF DIFFUSE TBI IN RATS. WE PROPOSE TO COMBINE HIGH-RESOLUTION GLUCOCEST AND THE DYNAMIC CONTRAST ENHANCED (DCE) PERFUSION MRI FOR CONCOMITANT CBF MEASUREMENTS TO IDENTIFY THE NEUROVASCULAR COUPLING STATE FOLLOWING TBI OVER TIME. OUR AIMS ARE DESIGNED TO (1) IDENTIFY THE CONTRAST MECHANISM OF IN VIVO GLUCOCEST IN THE TBI BRAIN TO ENHANCE THE SENSITIVITY AND SPECIFICITY OF GLUCOCEST WITH ADVANCED ACQUISITION SCHEME AND ANALYTICAL MODELS, (2) CHARACTERIZE PERTURBATIONS IN BRAIN GLUCOSE METABOLISM AND PERFUSION DEFICITS IN THE TBI BRAIN, AND (3) DEMONSTRATE THE POTENTIAL APPLICATION OF THE DUAL-MODALITY CEST-DCE MRI TO NONINVASIVELY MONITOR THE TREATMENT EFFECTS OF A CLINICALLY-RECOMMENDED INTERVENTION FOR TBI. OVERALL, THESE STUDIES WILL PROVIDE A STRONG TECHNICAL AND SCIENTIFIC FOUNDATION TO MOVE THE FIELD FORWARD IN UTILIZATION OF ADVANCED MRI FOR PERSONALIZED CARE IN THE CLINICAL ARENA AND DETERMINE THE BEST TREATMENT STRATEGY FOR THE BRAIN INJURED PATIENTS.

HOWEVER, NONOBESE HYPERANDROGENIC (HA) FEMALE MICE DISPLAYED ANDROGEN-SPECIFIC HEPATIC INSULIN RESISTANCE INDICATING A LIPID-INDEPENDENT PATHOGENIC MECHANISM [3]. ADDITIONALLY, HIGH FRUCTOSE DIETS (HFRD) COMPARED TO HIGH FAT DIETS (HFD) DISPLAY DIFFERING MECHANISMS OF INSULIN RESISTANCE, WHERE HIGH FRUCTOSE IMPAIRS GLUCOKINASE AND GLYCOGEN SYNTHASE BUT HIGH FAT LOWERS P-AKT [6]. KETOHEXOKINASE (KHK, ALSO KNOWN AS LIVER FRUCTOKINASE) IS REQUIRED FOR HFRD-INDUCED METABOLIC DYSFUNCTION [7]. THE OVERALL AIM IS TO ESTABLISH THAT DIFFERING CAUSES OF INSULIN RESISTANCE DISPLAY CROSSTALK BETWEEN CELLULAR, MOLECULAR, AND GENETIC MECHANISMS. I WILL DEVELOP 3 MOUSE MODELS OF HEPATIC INSULIN RESISTANCE: HIGH ANDROGEN (HA)-INDUCED, HFD-INDUCED, AND HFRD-INDUCED. USING VARIOUS HEPATIC SPECIFIC KNOCKOUT (KO) MICE TO ELIMINATE THE FUNCTION OF CERTAIN PATHWAYS (ANDROGEN RECEPTOR (AR-KO), KETOHEXOKINASE (KHK-KO), AND PROTEIN KINASE C (PKC-KO)), I WILL EXAMINE THE INTERSECTING PATHOGENIC MECHANISMS UNIQUE TO EACH OF THE THREE INSULIN RESISTANT MODELS. EXPECTED OUTCOME: I HYPOTHESIZE THAT EACH MODEL OF INSULIN RESISTANCE (HA, HFD, AND HFRD) WILL CONTAIN ITS OWN UNIQUE MECHANISTIC ASPECT WITH VARYING ASPECTS OF CROSSTALK. THUS, SUGGESTING THE MOVEMENT TOWARDS TARGETED THERAPEUTIC INTERVENTIONS BASED ON THE TYPE OF INSULIN RESISTANCE.

PURPOSE: EXTREME WEATHER CAUSES HUNDREDS OF DEATHS AND HUNDREDS OF BILLIONS OF DOLLARS IN DAMAGE ANNUALLY (PRIORITIES FOR WEATHER RESEARCH (PWR) REPORT). THE US IS EXPERIENCING ABOUT SIX TIMES AS MANY BILLION-DOLLAR WEATHER AND CLIMATE DISASTERS PER YEAR THAN IN THE 1980S. THE RISKS ASSOCIATED WITH THE EXTREME WEATHER FALL DISPROPORTIONATELY ON HISTORICALLY UNDERSERVED AND SOCIALLY VULNERABLE COMMUNITIES. DATA ASSIMILATION (DA) PLAYS A CRITICAL ROLE IN NUMERICAL WEATHER PREDICTION. THE NEXT-GENERATION NOAA UNIFIED FORECAST SYSTEM (UFS) DA FACES SIGNIFICANT CHALLENGES ASSOCIATED WITH HIGH-RESOLUTION, MULTISCALE, COUPLED EARTH SYSTEM MODELING AND A LARGE AMOUNT OF DIVERSE EXISTING AND NEW OBSERVATIONS. HOWEVER, SERIOUS GAPS IN DA WORKFORCE AND THE LACK OF SUSTAINED INNOVATIVE DA RESEARCH SUPPORT INHIBIT ADDRESSING THESE CHALLENGES. WE PROPOSE TO CREATE A MULTI-UNIVERSITY CONSORTIUM FOR ADVANCED DATA ASSIMILATION RESEARCH AND EDUCATION (CADRE). CADRE WILL PARTNER CLOSELY WITH NOAA TO SIGNIFICANTLY ADVANCE DA EDUCATION AND RESEARCH. THE PROPOSING TEAM CONSISTS OF 9 UNIVERSITIES THAT COLLECTIVELY HAVE ALL EXPERTISE NEEDED TO ADDRESS ALL THE PRIORITIES OUTLINED IN THE SOLICITATION AND DIRECTLY ADDRESS ALL THREE PRIORITY AREAS IN THE OBSERVATION AND DA PILLAR OF THE PWR REPORT. SPECIFICALLY, CADRE SEEKS TO ACHIEVE THE FOLLOWING GOALS: (1) INCREASING THE NUMBER OF GRADUATE STUDENTS AND POSTDOCS FORMALLY TRAINED IN DA WITH NOAA UFS AND JEDI DA SYSTEM WORK EXPERIENCE; (2) ENHANCING THE NATIONAL AND INTERNATIONAL DA WORKFORCE PIPELINE DEVELOPMENT THROUGH TRAINING AND OUTREACH FOCUSING ON DIVERSITY AND INCLUSION; (3) SEEKING SOLUTIONS THROUGH JEDI TO CHALLENGING DA ISSUES IN THE UFS SHORT RANGE (RRFS, HAFS), MEDIUM RANGE (GFS) AND SUB-SEASONAL TO SEASONAL (S2S) PREDICTIONS; AND (4) PROMOTING INTELLECTUAL AND TECHNICAL EXCHANGES IN R2O AND O2R. IN CLOSE COLLABORATION WITH NOAA, UK MOAP/TDSA AND OTHER WEATHER ENTITIES, CADRE WILL ACHIEVE THESE GOALS THROUGH (1) ADVISING GRADUATE STUDENTS AND POSTDOCS TO PERFORM A BROAD SCOPE OF DA RESEARCH USING JEDI AND UFS MODELS, (2) IMPLEMENTING NEW DA CAPABILITIES TO UFS JEDI TO FACILITATE O2R2O, (3) DEVELOPING AND EXPANDING DA MATERIALS FOR CLASSROOM TEACHING AND PUBLIC TRAINING, (4) DISSEMINATING RESULTS THROUGH PUBLICATIONS, DA WORKSHOPS, TECHNICAL DOCUMENTATIONS, AND CODE RELEASES. WE ANTICIPATE SUPPORTING 20 GRADUATE STUDENTS VIA GRADUATE PROGRAMS AND 6 POSTDOCS (SECTION 2.2.1) TO PERFORM UFS DA RESEARCH AND TO PARTICIPATE IN NOAA VISITOR PROGRAMS. WE PROPOSE TO SPONSOR 180 PARTICIPANTS TO ATTEND DA TRAINING COURSES AND 200 TO PARTICIPATE IN DA SCIENCE WORKSHOPS, AND AN ADDITIONAL 150 STUDENTS WILL TAKE UNIVERSITY DA CLASSES ENHANCED BY CADRE. WE WILL SUPPORT 12 DA RESEARCH THRUSTS AND THEIR IMPLEMENTATION TO THE UFS THROUGH JEDI FOR POTENTIAL R2O. THE HOWARD UNIVERSITY EXPANDING RESEARCH IN ALZHEIMER'S DISEASE AND RELATED DEMENTIAS POSTBACCALAUREATE RESEARCH EDUCATION PROGRAM (HU-ERA-PREP) - PROGRAM SUMMARY THE HU-ERA PROGRAM IS DESIGNED TO SELECT AND TRAIN A DIVERSE GROUP OF RECENT BACCALAUREATE GRADUATES FOR RESEARCH CAREERS IN ALZHEIMER'S DISEASE AND RELATED DEMENTIAS (ADRD), LEVERAGING THE WELL-ESTABLISHED HU- ADAR PROGRAM AND OTHER RELATED INITIATIVES AT HOWARD UNIVERSITY. TRAINEES WILL BE PROVIDED WITH INTENSIVE, MENTORED RESEARCH EXPERIENCES AND PROFESSIONAL DEVELOPMENT ACTIVITIES TO EXPAND EXPERTISE SKILLS RELEVANT TO ADRD-FOCUSED BIOMEDICAL RESEARCH, INCLUDING CRITICAL THINKING, ETHICAL DETERMINATIONS, QUANTITATIVE AND ANALYTICAL ASSESSMENTS, AND SCIENCE COMMUNICATION SKILLS; THEREBY PROMOTING TRAINEES’ SELF-EFFICACY AND SELF-IDENTITY AS PROFICIENT, BIOMEDICAL RESEARCHERS. TRAINING ACTIVITIES FOR EACH COHORT OF HU-ERA TRAINEES WILL SPAN A 2-YEAR CYCLE. THE MAIN TRAINING COMPONENT IS FULL-TIME, INTENSIVE BIOMEDICAL RESEARCH WORK IN AN ADRD-FOCUSED LABORATORY, GUIDED BY A DEDICATED MENTOR. ADDITIONAL CRITICAL TRAINING COMPONENTS INCLUDE: AN AGING BRAIN COURSE; AN AD/ADAR SEMINAR SERIES; A COURSE ON RESPONSIBLE CONDUCT OF RESEARCH AND ETHICS; WORKSHOPS ON VARIOUS RESEARCH-RELATED TOPICS INCLUDING DATA INTERPRETATION, RIGOR, AND REPRODUCIBILITY; AND OTHER PROFESSIONAL SKILL- BUILDING ACTIVITIES. HOST RESEARCH MENTORS AND OTHER PARTICIPATING FACULTY OF THE HU-ERA PROGRAM HAVE RIGOROUS NEUROSCIENCE RESEARCH PROGRAMS AND RECORDS OF TRAINING AND MENTORING THOSE FROM DIVERSE CULTURAL, RACIAL BACKGROUNDS, AND SELF-IDENTIFIED ORIENTATIONS. MENTORS ARE MOTIVATED TO PROMOTE DIVERSITY, EQUITY, INCLUSION, AND ACCESSIBILITY IN THEIR WORK AND RESEARCH ENVIRONMENTS AND ARE ALIGNED WITH THE HU-ERA PROGRAM MISSION TO INCREASE UNDERREPRESENTED MINORITY REPRESENTATION IN THE ADRD BIOMEDICAL RESEARCH WORKFORCE AND LEADERSHIP POSITIONS. PREVIOUS RESEARCH EXPERIENCE AND INTERESTS OF EACH TRAINEE ARE DECIDING FACTORS FOR MENTEE/MENTOR PAIRINGS. BY THE COMPLETION OF THE 2-YEAR RESEARCH TRAINING PROGRAM, HU-ERA SCHOLARS WILL BE UNIQUELY AND COMPETITIVELY PREPARED TO TRANSITION INTO RESEARCH-FOCUSED ADVANCED DEGREE PROGRAMS OR COMPETITIVE PRIVATE- SECTOR RESEARCH CAREERS IN ADRD-RELATED DISCIPLINES. A SET OF WELL-DEFINED PROFESSIONAL DEVELOPMENT ACTIVITIES WILL BE IMPLEMENTED TO FACILITATE THE TRANSITION OF HU-ERA SCHOLARS INTO HIGH-LEVEL BIOMEDICAL SCIENCE RESEARCH CAREERS INCLUDING WORKSHOPS ON RESUME/CV BUILDING, SCIENTIFIC PRESENTATION SKILLS, GRADUATE SCHOOL FUNDING, AND INTERVIEW SKILLS. HU-ERA POSTBACCALAUREATES WILL ALSO HAVE ACCESS TO HU-SPONSORED GRE & MCAT REVIEW COURSES. ALL ELEMENTS OF THE HU-ERA PROGRAM WILL BE ASSESSED BY INTERNAL AND EXTERNAL EVALUATORS AT MULTIPLE, SPECIFIC TIME POINTS WITH THE GUIDANCE OF A PROFESSIONAL EVALUATOR. OVERALL PROGRAM ACHIEVEMENTS WILL BE DETERMINED BY MEASURES OF MATRICULATION OF SCHOLARS INTO PHD AND/OR MD PROGRAMS WITHIN INSTITUTIONS HAVING STRONG AD/ADRD RESEARCH PROGRAMS, OR ATTAINMENT OF ADRD-RESEARCH POSITIONS WITHIN THE PRIVATE SECTOR. LINKING DEFECTS IN DNA POLYMERASE BETA TO MITOCHONDRIAL DYSFUNCTION AND CYTOSOLIC NUCLEIC ACID SENSOR IN HELICOBACTER PYLORI ASSOCIATED INFLAMMATION - PROJECT SUMMARY TARGETING MITOCHONDRIA HAS EMERGED AS A KEY STRATEGY FOR BACTERIA TO HIJACK HOST CELL PHYSIOLOGY AND PROMOTE INFECTION. MITOCHONDRIAL DYSFUNCTION SHAPES INNATE IMMUNE RESPONSE AND PROMOTES INFLAMMATORY RESPONSES. SEVERAL STUDIES HAVE SHOWN THAT DEFECTIVE OXIDATIVE DNA DAMAGE REPAIR PATHWAYS ARE ASSOCIATED WITH MITOCHONDRIAL DYSFUNCTION AND CHRONIC INFLAMMATORY DISEASES. BASE EXCISION REPAIR (BER) IS THE PREDOMINANT PATHWAY THAT CORRECTS SMALL BASE LESIONS CAUSED BY REACTIVE OXYGEN AND NITROGEN SPECIES (RONS). BER IS THE MAJOR PATHWAY FOR THE REPAIR OF OXIDATIVE DNA LESIONS AND IS PRESENT IN BOTH NUCLEUS AND MITOCHONDRIA BY SIMILAR MECHANISMS THAT SHARE MANY OF THE CORE BER ENZYMES. DNA POLYMERASE BETA (POLB) AND GAMMA (POLG) ARE INVOLVED IN MAINTENANCE OF MITOCHONDRIAL DNA (MTDNA) INTEGRITY. THE 5’- DEOXYRIBOPHOSPHODIESTERASE (5’-DRP LYASE) FUNCTIONS OF POLB IS MORE ACTIVE THAN POLG 5’-DRP LYASE FUNCTION TO REMOVE THE 5’-DEXYRIBOSE PHOSPHATE GROUP (5’-DRP). IN ADDITION, WE CHARACTERIZED 5’-DRP LYASE DEFICIENT POLB (L22P POINT MUTATION)] THAT RESULTS IN LOSS OF 5’-DRP LYASE FUNCTION AS A MODEL TO EXAMINE WHETHER ABSENCE OF 5’-DRP LYASE IMPAIRS MITOCHONDRIAL FUNCTION AND CONTRIBUTES TO ABERRANT INFLAMMATORY RESPONSE. OUR PRELIMINARY DATA SHOW THAT BER DEFICIENT CELLS SIGNIFICANTLY ACCUMULATE MITOCHONDRIAL DNA (MTDNA) DAMAGE. MOREOVER, WE FOUND THAT HELICOBACTER PYLORI (H. PYLORI) INFECTION EXACERBATES DNA DAMAGE AND INFLAMMATORY RESPONSE IN 5’-DRP LYASE DEFICIENT MICE. HOWEVER, THERE ARE NO DATA TO EXPLAIN HOW THE BER DEFECT ( 5’-DRP LYASE DEFICIENT POLB) INFLUENCES MITOCHONDRIAL DNA MEDIATED INNATE IMMUNE RESPONSE AND ITS POTENTIAL IMPACT ON INDUCTION OF INFLAMMATION ASSOCIATED HUMAN DISEASES. IN THIS APPLICATION, WE WILL EXAMINE HOW DEFECTIVE BER ASSOCIATED MITOCHONDRIAL DYSFUNCTION CONTRIBUTES TO CYTOSOLIC NUCLEIC ACID SENSOR MEDIATED INNATE IMMUNE INFLAMMATORY RESPONSE. BASED ON OUR HIGHLY SIGNIFICANT AND ENCOURAGING PRELIMINARY FINDINGS, WE WILL TEST THE HYPOTHESIS THAT DEFECTIVE BER PROMOTES MITOCHONDRIAL DYSFUNCTION AND PROVOKES CYTOSOLIC NUCLEIC ACID MEDIATED SIGNALING IN H. PYLORI ASSOCIATED INFLAMMATION. IN THIS STUDY, WE PROPOSE THREE SPECIFIC AIMS AS FOLLOWS: I) DETERMINE WHETHER BER DEFICIENT CELLS RELEASE MTDNA IN THE CYTOPLASM; II) DETERMINE WHETHER ABERRANT REPAIR OF MTDNA MODULATES INNATE IMMUNE SIGNALING IN VITRO AND IN VIVO; AND III) DEFINE HOW THE EFFECTS OF ABERRANT BER (5’-DRP LYASE DEFICIENT POLB) WITH H. PYLORI INFECTION CONTRIBUTE TO CYTOSOLIC NUCLEIC ACID SENSOR MEDIATED INNATE IMMUNE SIGNALING AND INFLAMMATORY RESPONSE. COMPLETION OF THE PROPOSED STUDIES WILL PROVIDE NOVEL MECHANISTIC INSIGHT INTO HOW IMPAIRED MTBER AND EXTRACELLULAR BACTERIAL INFECTION CONTRIBUTE TO MITOCHONDRIAL DYSFUNCTION RELATED INFLAMMATION. FURTHER, UNLOCKING CYTOSOLIC SURVEILLANCE AND SIGNALING UPON EXTRACELLULAR BACTERIAL INFECTION MAY REVEAL NEW OPPORTUNITIES FOR FUTURE IMMUNE BASED THERAPEUTIC STRATEGY.

EPITRANSCRIPTOMIC AND POST-TRANSCRIPTIONAL REGULATION OF THE RNOME IN GRAM NEGATIVE AND FIRMICUTE BACTERIA - PROJECT SUMMARY / ABSTRACT POST-TRANSCRIPTIONAL REGULATION OF GENE EXPRESSION HAS SIGNIFICANT EFFECTS ON THE PHYSIOLOGY OF THE BACTERIAL CELLS. WHILE MUCH HAS BEEN UNCOVERED ABOUT POST-TRANSCRIPTIONAL REGULATION OF GENE EXPRESSION IN BACTERIA, THERE ARE STILL SOME KEY UNKNOWNS THAT ARE HINDERING OUR ABILITY TO UNDERSTAND HOW THESE PROCESSES WORK AND DIFFER THROUGHOUT THE PROKARYOTIC DOMAIN OF LIFE. IN THIS PROJECT, OUR OVERALL GOAL IS TO EXPAND OUR UNDERSTANDING OF THE DIVERSITY OF THESE PROCESSES IN GRAM-NEGATIVE MODEL ORGANISMS, FIRMICUTES, AND MYCOBACTERIA. OUR PROPSOSED RESEARCH PROGRAM HAS TWO SUB-PROJECTS. PROJECT #1 IS ENTITLED: EPITRANSCRIPTOMIC REGULATION OF GENE EXPRESSION THROUGHOUT THE PROKARYOTIC DOMAIN. IN SUB-PROJECT 1, WE WILL EXPAND OUR UNDERSTANDING OF RNA MODIFICATIONS WITHIN A WIDE RANGE OF BACTERIAL SPECIES. RNA MODIFICATIONS INCLUDE AN ISOMERIZATION OR CHEMICAL ADDITIONS TO CANONICAL NUCLEOTIDES WITHIN RNA. GENERALLY FOUND IN TRNA MOLECULES, RNA MODIFICATIONS ARE CRITICAL IN PROMOTING TRANSLATIONAL FIDELITY BY PROMOTING RIBOSOME MRNA DECODING ERRORS. THERE ARE SEVERAL KEY KNOWLEDGE GAPS DRIVING OUR EXPERIMENTAL APPROACH. THE REGULATORY ROLE OF RNA MODIFICATIONS, CIS-ACTING SIGNALS THAT MAY PROMOTING REGULATORY TARGETING OF RNA MODIFICATIONS, AND THE EFFECT OF GC CONTENT ON THE ABILITY OF CODON USAGE TO DRIVE TARGETING OF GENES BY MODIFIED TRNAS. WE WILL USE A MULTI-OMICS APPROACH TO ADDRESS THESE KEY KNOWLEDGE GAPS USING SEVERAL BACTERIAL MODEL ORGANISMS: ESCHERICHIA COLI, STAPHYLOCOCCUS AUREUS, CLOSTRIDIOIDES DIFFICILE, AND MYCOBACTERIUM SMEGMATIS. PROJECT #2 IS ENTITLED: POST-TRANSCRIPTIONAL GENE REGULATION VIA NOVEL PROTEINS AND SRNAS IN FIRMICUTES. IN SUB-PROJECT 2, WE WILL EXPAND OUR UNDERSTANDING OF THE REGULATORY ROLE OF NOVEL PROTEINS AND SRNAS IN THE REGULATION OF THE RNOME IN FIRMICUTES. MUCH OUR UNDERSTANDING OF SRNA IN BACTERIA HAS COME FROM EXTENSIVE STUDIES IN MODEL ORGANISMS. THIS HAS ASSISTED IN OUR UNDERSTANDING SRNA REGULATION OF BACTERIAL PHYSIOLOGY IN GENERAL. HOWEVER, RECENT STUDIES HAVE UNCOVERED HUNDREDS OF SPECIES- SPECIFIC SRNA IN S. AUREUS. THE VAST MAJORITY OF THESE ARE UNCHARACTERIZED. SINCE WE LACK A FULL PICTURE OF SRNA REGULONS IN S. AUREUS, OUR UNDERSTANDING OF BACTERIAL SRNA BIOLOGY IS INCOMPLETE. IN ADDITION, MANY MODEL ORGANISMS UTILIZE THE RNA CHAPERONE HFQ AS A SRNA CO-FACTOR IN SOME CASES TO PROMOTE SRNA-MRNA-RNASE INTERACTIONS. HOWEVER, HFQ IS DISPENSABLE FOR GROWTH AND SRNA FUNCTION. IT IS UNKNOWN IF THERE ARE OTHER PROTEIN FACTORS THAT MAY ACT AS SPECIFIC-SPECIFIC CHAPERONES IN S. AUREUS OR OTHER FIRMICUTES. WE WILL HAVE PRELIMINARY DATA TO SUGGEST THAT NOVEL REGULATORY PROTEINS MAY EXIST, IN S. AUREUS AND C. DIFFICILE, IN THE MRNA REGULATION AND MAY ALSO MODULATE LEVELS OF SRNAS. WE WILL EXECUTE RNA-IMMUNOPRECIPITATION SEQUENCING OF THIS CANDIDATE PROTEIN IN S. AUREUS AND C. DIFFICILE TO IDENTIFY ITS RNA-INTERACTOME. WE HAVE ALSO IDENTIFIED PREDICTED SRNA INTERACTIONS WITH SEVERAL IMPORTANT PROTEINS IN S. AUREUS AND EXECUTE EXPERIMENTS TO TEST THIS INTERACTION.

MECHANISMS OF CELLULAR MORPHOGENESIS THAT COORDINATE SIGNALING DURING TISSUE PATTERNING - PROJECT SUMMARY THE LONG-TERM GOAL OF OUR RESEARCH PROGRAM IS TO UNDERSTAND AND IDENTIFY THE MECHANISMS OF CELL SHAPE DYNAMICS THAT FACILITATE THE EMERGENCE OF TISSUE-WIDE PATTERNS FROM LOCAL CELL-CELL COMMUNICATION EVENTS. THE INCORRECT SPATIAL AND TEMPORAL INDUCTION OF GENES AND CELL BEHAVIORS BY PERTURBED SIGNALING CAN LEAD TO CATASTROPHIC ERRORS IN DEVELOPMENT. THEREFORE, IT IS CRITICAL TO UNDERSTAND HOW SIGNALING MECHANISMS THAT TAKE PLACE AT THE LEVEL OF A CELL OR ADJACENT CELLS ARE COORDINATED ACROSS LARGER TISSUE LENGTH SCALES TO ACHIEVE ROBUST PATTERNS AND SUPPORT TISSUE HOMEOSTASIS. CYTONEMES, OR SIGNALING FILOPODIA, ARE LONG ACTIN-RICH CELLULAR PROTRUSIONS THAT ALLOW CELLS TO PHYSICALLY INTERACT WITH, AND SIGNAL TO, OTHER CELLS AT A DISTANCE. CELLULAR PROTRUSIONS LIKE CYTONEMES HAVE BEEN SHOWN TO PLAY A ROLE IN MANY DEVELOPMENTAL PROCESSES, INCLUDING SPOT AND STRIPE PATTERNING, LIMB PATTERNING, STEM CELL NICHE MAINTENANCE, AND NEURAL PLATE PATTERNING. DESPITE THIS, RELATIVELY LITTLE IS KNOWN ABOUT THE GENERAL MECHANISMS THAT REGULATE CYTONEME-SPECIFIC FORMATION AND BEHAVIOR. WE ARE INTERESTED IN UNDERSTANDING HOW THESE PROTRUSIONS ARE REGULATED BOTH AT THE LEVEL OF CYTOSKELETAL MECHANISMS, AND AS AN INTEGRATED PART OF THE CELL-WIDE AND TISSUE-WIDE SIGNALING STATES. TO ADDRESS THIS PROBLEM, WE USE THE SPOT PATTERN OF BRISTLES ON THE DORSAL THORAX OF THE FRUIT FLY DROSOPHILA MELANOGASTER AS A MODEL SYSTEM. DURING BRISTLE PATTERNING, CYTONEMES EXTEND FROM THE BASAL SURFACE OF THORACIC EPITHELIAL CELLS TO FACILITATE LONG-RANGE, NOTCH-MEDIATED, LATERAL INHIBITION. LEVERAGING OUR STRENGTHS IN DROSOPHILA MODEL SYSTEMS GENETICS AND QUANTITATIVE LIVE MICROSCOPY, OVER THE NEXT FIVE YEARS OUR GOALS INCLUDE UNDERSTANDING HOW THE FORMATION OF CYTONEMES ARE REGULATED, ESPECIALLY DOWNSTREAM OF NOTCH-MEDIATED LATERAL INHIBITION SIGNALING. IN PARALLEL, WE AIM TO UNDERSTAND HOW CYTONEME ACTIVITY CONTRIBUTES TO CELL AND TISSUE HETEROGENEITIES THAT FACILITATE THE TIMELY FORMATION OF WELL-ORGANIZED PATTERNS. THESE STUDIES WILL NOT ONLY YIELD VALUABLE CELL BIOLOGICAL KNOWLEDGE ABOUT THE SPECIFIC REGULATION OF CYTONEMES, BUT WILL ALSO ELUCIDATE FUNDAMENTAL MECHANISMS THAT COORDINATE PATTERNING ACROSS LENGTH SCALES.

LOUIS STOKES RENEWAL STEM PATHWAYS AND RESEARCH ALLIANCE: WASHINGTON BALTIMORE HAMPTON ROADS -LOUIS STOKES ALLIANCE (WBHR-LSAMP ) -THE LOUIS STOKES ALLIANCES FOR MINORITY PARTICIPATION (LSAMP) PROGRAM ASSISTS UNIVERSITIES AND COLLEGES IN THEIR EFFORTS TO SIGNIFICANTLY INCREASE THE NUMBERS OF STUDENTS MATRICULATING INTO AND SUCCESSFULLY COMPLETING HIGH QUALITY DEGREE PROGRAMS IN SCIENCE, TECHNOLOGY, ENGINEERING AND MATHEMATICS (STEM) DISCIPLINES TO DIVERSIFY THE STEM WORKFORCE. PARTICULAR EMPHASIS IS PLACED ON TRANSFORMING UNDERGRADUATE STEM EDUCATION THROUGH INNOVATIVE, EVIDENCE-BASED RECRUITMENT AND RETENTION STRATEGIES, AND RELEVANT EDUCATIONAL EXPERIENCES IN SUPPORT OF POPULATIONS UNDERREPRESENTED IN STEM DISCIPLINES: AFRICAN AMERICANS, HISPANIC AMERICANS, AMERICAN INDIANS, ALASKA NATIVES, NATIVE HAWAIIANS, AND NATIVE PACIFIC ISLANDERS. THESE STRATEGIES FACILITATE THE PRODUCTION OF WELL-PREPARED STUDENTS HIGHLY QUALIFIED AND MOTIVATED TO PURSUE GRADUATE EDUCATION OR CAREERS IN STEM. FOR THE UNITED STATES TO REMAIN GLOBALLY COMPETITIVE, IT IS VITAL THAT IT TAPS INTO THE TALENT OF ALL ITS CITIZENS AND PROVIDES EXCEPTIONAL EDUCATIONAL PREPAREDNESS IN STEM AREAS THAT UNDERPIN THE KNOWLEDGE-BASED ECONOMY. THE WASHINGTON BALTIMORE HAMPTON ROADS ? LOUIS STOKES ALLIANCE FOR MINORITY PARTICIPATION (WBHR-LSAMP) HAS FORMED AN ALLIANCE IN RESPONSE TO THE NEED FOR A MORE DIVERSE AND SKILLED TECHNICAL WORKFORCE. THAT NEED STILL EXISTS AND IS PARTICULARLY ACUTE IN THE DISTRICT OF COLUMBIA, MARYLAND, AND VIRGINIA (DMV). THE INSTITUTIONS THAT MAKE UP THE ALLIANCE ARE DIVERSE AND INCLUDE HOWARD UNIVERSITY, LEAD INSTITUTION, BOWIE STATE UNIVERSITY, HAMPTON UNIVERSITY, MORGAN STATE UNIVERSITY, NORFOLK STATE UNIVERSITY, UNIVERSITY OF THE DISTRICT OF COLUMBIA, VIRGINIA STATE UNIVERSITY, EL CAMINO COLLEGE, HOWARD COMMUNITY COLLEGE, AND PRINCE GEORGE?S COMMUNITY COLLEGE. THROUGH THIS PROJECT, THE ALLIANCE WILL IMPLEMENT ACTIVITIES TO SUPPORT STUDENT SUCCESS BY IMPROVING RETENTION, PARTICIPATION IN UNDERGRADUATE RESEARCH, AND THE OVERALL NUMBER OF HISTORICALLY UNDERREPRESENTED POPULATIONS GRADUATING WITH STEM DEGREES FROM THE TEN ALLIANCE INSTITUTIONS. THROUGH THIS EFFORT, RESEARCHERS INVOLVED IN THIS PROJECT BELIEVE THAT THE IMPROVED RESULTS WILL STRENGTHEN THE STEM WORKFORCE IN THE DMV AND ULTIMATELY CONTRIBUTE TO AMERICA'S NATIONAL SECURITY. HOWARD UNIVERSITY CLINICAL RESEARCH NETWORK FOR HEALTH EQUITY - RESEARCH ABSTRACT AFRICAN AMERICANS EXPERIENCE SIGNIFICANT DISPARITIES IN CHRONIC DISEASE MORBIDITY AND MORTALITY. OF NOTE, ELIMINATING HEALTH DISPARITIES REQUIRES SIGNIFICANT IMPROVEMENT IN CHRONIC DISEASE MANAGEMENT, WHICH IS DEPENDENT ON THE PROVISION AND UTILIZATION OF EVIDENCE-BASED PREVENTIVE CARE. THE PROVISION OF HIGH-QUALITY PREVENTIVE SERVICES TO THE AFRICAN AMERICANS IS HINDERED BY SEVERAL THINGS INCLUDING INSUFFICIENT EVIDENCE WHICH IS A CONSEQUENCE OF UNDERREPRESENTATION OF MINORITY POPULATIONS IN CLINICAL RESEARCH TRIALS. IN FACT, UNITED STATES PREVENTIVE SERVICES TASK FORCE (USPSTF) RECOMMENDATION STATEMENTS OFTEN CITE UNDERREPRESENTATION OF MINORITIES IN CLINICAL TRIALS AS A RESEARCH GAP AND LIMITATION. THIS AFFECTS THE DEVELOPMENT OF CLINICAL PRACTICE GUIDELINES THAT MEET THE NEEDS OF MINORITY POPULATIONS WHEN THE EPIDEMIOLOGY OF A PARTICULAR DISEASE IS DIFFERENT IN MINORITY POPULATIONS. NOT ONLY IS THERE AN UNDERREPRESENTATION OF MINORITIES AS STUDY PARTICIPANTS BUT THERE IS EQUALLY SEVERE UNDERREPRESENTATION OF AFRICAN AMERICANS AS CLINICAL INVESTIGATORS, WHICH IS DUE IN PART TO UNDERREPRESENTATION OF AFRICAN AMERICANS IN THE CLINICAL RESEARCH WORKFORCE. ADDRESSING THESE DISPARITIES REQUIRES COMPLEX SYSTEMS ANALYSIS, DEVELOPMENT OF INNOVATIVE APPROACHES, GENERATION OF SCIENTIFIC EVIDENCE, AND EFFECTIVE KNOWLEDGE DISSEMINATION. FOR OVER 150 YEARS, HOWARD UNIVERSITY HAS CONTRIBUTED SIGNIFICANTLY TO THE AFRICAN AMERICAN PHYSICIAN WORKFORCE THROUGH UNDERGRADUATE AND GRADUATE MEDICAL EDUCATION. IN ADDITION, HOWARD UNIVERSITY HAS ADVANCED MINORITY HEALTH THROUGH ROBUST RESEARCH ACTIVITIES AND COMPREHENSIVE PATIENT CARE TO THE RESIDENTS OF THE DISTRICT OF COLUMBIA AND BEYOND. THE ADOPTION OF ELECTRONIC MEDICAL RECORD SYSTEMS BY HOWARD UNIVERSITY HOSPITAL AND HOWARD UNIVERSITY FACULTY PRACTICE PLAN HAS LED TO THE GENERATION OF VAST AMOUNTS OF CLINICAL DATA ABOUT THIS INNER-CITY URBAN LOW-INCOME AFRICAN AMERICAN POPULATION. TO ADDRESS THE CHALLENGES, WE PROPOSE THE FORMATION OF THE HOWARD UNIVERSITY CLINICAL RESEARCH NETWORK (HUCRN) AS AN ADDITION TO THE CONSORTIUM OF THE RCMI CLINICAL RESEARCH NETWORK FOR HEALTH EQUITY (RCMI-CRNHE). THIS EFFORT WILL LEAD TO AN INNOVATIVE, PRODUCTIVE, AND IMPACTFUL RESEARCH NETWORK FOR INVESTIGATION OF STUDIES OF HIGH IMPORTANCE IN MINORITY HEALTH AND HEALTH DISPARITIES. THE SIGNIFICANCE OF THIS PROJECT IS DERIVED FROM THE NECESSITY TO STUDY A HEALTH DISPARITY AFFECTED POPULATION, BURDEN OF CHRONIC DISEASE, PREVALENT HEALTH DISPARITIES, ENGAGEMENT OF AN UNDERREPRESENTED CLINICAL RESEARCH WORKFORCE. FURTHER WE INTEND TO INTRODUCE A NEW APPLICATION OF INNOVATIVE DATA SCIENCE CALLED MHEALTH. WE WILL DEVELOP A MHEALTH INFORMATION TECHNOLOGY INFRASTRUCTURE, CLINICAL RESEARCH INFORMATICS INFRASTRUCTURE INCLUDING A CLINICAL DATA REPOSITORY, AND WE WILL CONDUCT 2 PILOT STUDIES TO ASSESS THE EFFECTIVENESS OF THE INFRASTRUCTURE, POLICIES, AND PROCESSES THAT HAVE BEEN IMPLEMENTED.

FIVE B2B STUDENTS PER YEAR WILL BE SELECTED VIA A RIGOROUS REVIEW PROCESS AND PROVIDED EDUCATIONAL READINESS BY P2P WHILE STILL INCARCERATED, TRANSITION TO A 2-YEAR ASSOCIATES GRANTING SCHOOL (BCCC), BE PROVIDED SKILLS AND METHODS DEVELOPMENT WHILE AT BCCC, PARTICIPATE IN A 10-WEEK STEM RESEARCH INTERNSHIP AT HOWARD IN THE SUMMER IN BETWEEN YEARS 1 AND 2, THEN TRANSITION TO A 4-YEAR SCHOOL, UPON COMPLETING THEIR ASSOCIATES DEGREE AT BCCC. WE PLAN TO START A 5-PARTICIPANT COHORT EACH YEAR OF THE 5-YEAR FUNDING PERIOD. THE PROGRAM WILL ADDRESS THE FOLLOWING, WHICH IS EXPLAINED IN DETAIL WITHIN THE PROPOSAL: 1. SKILLS DEVELOPMENT WILL BE GAINED VIA THE LEADERSHIP, COLLEGE & PROFESSIONAL READINESS, AND SAT PORTIONS OF THE P2P PROGRAM AND INTRO TO RESEARCH WORKSHOPS WILL BE IMPLEMENTED DURING THE COMMUNITY COLLEGE TIME SPAN. 2. RESEARCH EXPERIENCE WILL BE OBTAINED VIA A 10-WEEK INTERNSHIP WITH ONE OF NEARLY 20 COLLABORATING INVESTIGATORS AT HOWARD UNIVERSITY AND SCHOLARS WILL ATTEND A NATIONAL MEETING TO PRESENT THEIR WORK. 3. CURRICULUM AND METHODS TO PROVIDE SUPPLEMENTAL INSTRUCTION FOR GATEWAY COURSES, DEVELOPMENT WILL INCLUDE A MOLECULAR TECHNIQUES COURSE INCORPORATED INTO GENERAL BIOLOGY (BCCC BTC 105 - TECHNIQUES AND INSTRUMENTATION FOR BIOTECHNOLOGY) 4. A. RECRUITMENT PLAN: WE WILL DO A MULTITUDE OF ACTIVITIES DETAILED IN THE PROPOSAL BUT BRIEFLY, P2P ALREADY WORKS WITH OVER 100 INDIVIDUALS PER YEAR IN THEIR ACTIVELY RUNNING PROGRAM. PRIORITY WILL BE GIVEN TO SELECT THE TOP FIVE PEOPLE OF THE P2P PROGRAM. OTHER RECRUITMENT WILL BE DONE AS WELL, SUCH AS PLACING ADS ON LOCAL HIP-HOP AND TOP 40 RADIO STATIONS AND UTILIZING CONNECTIONS ALREADY IN PLACE WITH COMMUNITY ORGANIZATIONS AND GOVERNMENT (MD-DOC) PARTNERS. 4. B. RETENTION PLAN: WE WILL PROVIDE INCENTIVES: 60% TUITION SCHOLARSHIPS, YEARLY STIPENDS, PAID INTERNSHIPS, AND MENTORING OPPORTUNITIES TO GIVE BACK. THE GOAL OF THE P2P-B2B PROGRAM IS TO CREATE SYNERGIES BETWEEN PRISONS AND 2- AND 4- YEAR COLLEGES IN BALTIMORE (CREATING A PRISON-TO-COLLEGE PIPELINE) AND TO INCREASE THE TRANSITION TO BS/BA DEGREES FROM AA DEGREES. HOWARD UNIVERSITY IS ACQUIRING A 500 MHZ SOLUTION AND SOLID-STATE NUCLEAR MAGNETIC RESONANCE (NMR) SPECTROMETER EQUIPPED WITH A BROADBAND PROBE AND A TRIPLE-RESONANCE SOLIDS PROBE TO SUPPORT THE RESEARCH OF PROFESSOR STEVEN CUMMINGS ALONG WITH COLLEAGUES AMOL KULKAMI, DHARMARAJ RAGHAVAN, SUNG KIM, AND TIMOTHY RAMADHAR. THIS INSTRUMENT FACILITATES RESEARCH IN THE AREAS OF CATALYST DESIGN, BIOCHEMISTRY, POLYMER CHEMISTRY, NANOMATERIALS, AND MEDICINAL CHEMISTRY. IN GENERAL, NMR SPECTROSCOPY IS ONE OF THE MOST POWERFUL TOOLS AVAILABLE TO CHEMISTS FOR THE ELUCIDATION OF THE STRUCTURE OF MOLECULES. IT IS USED TO IDENTIFY UNKNOWN SUBSTANCES, TO CHARACTERIZE SPECIFIC ARRANGEMENTS OF ATOMS WITHIN MOLECULES, AND TO STUDY THE DYNAMICS OF INTERACTIONS BETWEEN MOLECULES IN SOLUTION OR IN THE SOLID STATE. ACCESS TO STATE-OF-THE-ART NMR SPECTROMETERS IS ESSENTIAL TO CHEMISTS WHO ARE CARRYING OUT FRONTIER RESEARCH. THIS INSTRUMENT ENHANCES THE EDUCATIONAL, RESEARCH, AND RESEARCH TRAINING AT ALL LEVELS IN MULTIPLE DEPARTMENTS AT HOWARD UNIVERSITY, A HISTORICALLY BLACK UNIVERSITY (HBCU). THIS INSTRUMENT FURTHER PROPAGATES RESEARCH AND EDUCATIONAL OPPORTUNITIES BETWEEN THE MINORITY-SERVING INSTITUTIONS IN THE REGION INCLUDING MORGAN STATE UNIVERSITY, BOWIE STATE UNIVERSITY, AND GALLAUDET UNIVERSITY. THE AWARD OF THE 500 MHZ NMR SPECTROMETER IS AIMED AT ENHANCING RESEARCH AND EDUCATION AT ALL LEVELS, ESPECIALLY IN AREAS SUCH AS CHARACTERIZATIONS OF METAL-ORGANIC FRAMEWORKS, BIOLOGICAL SYSTEMS AND MATERIALS, AND NANOPARTICLES. SPECIFICALLY, THE INSTRUMENT IMPACTS RESEARCH FOCUSED ON FUSING CARBORANE CHEMISTRY WITH CARBENE AND PYRIDINATE LIGANDS. THE INSTRUMENT IS ALSO USED FOR STRUCTURAL INVESTIGATIONS OF COMPLEX BIOACTIVE COMPOUNDS BY STRUCTURAL AND MECHANISTIC ANALYSIS OF THE CRYSTALLINE SPONGE METHOD. ADDITIONALLY, THIS INSTRUMENT ENABLES THE COMPOSITION ANALYSIS OF STAPHYLOCOCCUS AUREUS BIOFILM. OTHER INVESTIGATIONS INCLUDE THE METABOLOME AND COMPOSITION ANALYSIS OF EXOSOME IN SEARCH OF MARKERS FOR TYPE 1 DIABETES, AND THE DETERMINATION OF THE BINDING MODE OF SMALL MOLECULES TO THE PROTEIN NLRP3 INFLAMMASOME. IN RESPONSE TO GLOBAL HELIUM SUPPLY CHALLENGES AND THE 2022 CHIPS AND SCIENCE ACT, THE ACQUISITION OF THIS INSTRUMENT INCLUDES A SUPERCONDUCTING MAGNET WITH A LOW LOSS CRYOSTAT REDUCING THE INSTITUTION?S CONSUMPTION OF HELIUM. YOUNG ADULTS WITH VIOLENT BEHAVIOR DURING EARLY PSYCHOSIS: A MIXED-METHODS ASSESSMENT AND INTERVENTION TRIAL - PROJECT SUMMARY: ALTHOUGH THE ABSOLUTE RISK OF VIOLENCE AMONG PERSONS WITH SERIOUS MENTAL ILLNESSES (SMI) IS SMALL, YOUNG PEOPLE EXPERIENCING EARLY PSYCHOSIS ARE A SUBGROUP AT HIGHEST VIOLENCE RISK. VIOLENCE IS A PUBLIC HEALTH ISSUE THAT CONTRIBUTES TO HEALTH DISPARITIES EXPERIENCED BY ADULTS WITH SCHIZOPHRENIA, INCLUDING CAREGIVER BURDEN, SOCIAL ISOLATION, AND INCARCERATION AND SUBSEQUENT EARLY MORTALITY. COGNITIVE BEHAVIORAL THERAPY (CBT) FOR PSYCHOSIS AND SIMILAR BEHAVIORAL INTERVENTIONS HAVE HAD EFFICACY IN REDUCING IMPAIRMENT AND IMPROVING FUNCTIONING AMONG YOUNG ADULTS WITH EARLY PSYCHOSIS. HOWEVER, NO IDENTIFIED STUDIES HAVE EXPLORED USING A BEHAVIORAL INTERVENTION TO REDUCE VIOLENCE AMONG YOUNG ADULTS WITH EARLY PSYCHOSIS. IN THIS APPLICATION, DR. STEPHANIE ROLIN PROPOSES A COMPREHENSIVE PATH TOWARDS BECOMING AN INDEPENDENT PHYSICIAN INVESTIGATOR CREATING AND ADAPTING EVIDENCE-BASED APPROACHES TO ASSESS AND REDUCE VIOLENCE RISK. SPECIFICALLY, THIS PROPOSAL FOLLOWS THE CDC’S MAP OF ADAPTATION, A MODEL FOR ADAPTING EVIDENCE-BASED INTERVENTIONS (EBI), TO ADAPT AND EVALUATE THE FEASIBILITY OF AN EARLY INTERVENTION SERVICES (EIS)-SPECIFIC CBT- BASED INTERVENTION TO REDUCE VIOLENCE WITH PROPOSED TARGETS CONSISTENT WITH THE TRANSDIAGNOSTIC NATURE OF RESEARCH DOMAIN CRITERIA (RDOC). THIS RESEARCH WILL OCCUR AT ONTRACKNY, THE LARGEST EIS PROGRAM IN THE UNITED STATES; ONTRACKNY HAS PROVIDED EVIDENCE-BASED COORDINATED SPECIALTY CARE TO OVER 1,800 INDIVIDUALS AT MORE THAN 20 CLINICS IN NEW YORK STATE. FIRST, DR. ROLIN WILL CONDUCT INDIVIDUAL SEMI-STRUCTURED INTERVIEWS WITH EIS CLIENTS WHO HAVE ENGAGED IN RECENT VIOLENT BEHAVIOR (N=20) AND CLINICIANS WHO HAVE WORKED WITH EIS CLIENTS WHO HAVE ENGAGED IN VIOLENT BEHAVIOR (N=10). THESE INTERVIEWS WILL USE THE THEORY OF PLANNED BEHAVIOR TO IDENTIFY KEY AREAS OF AN EXISTING CBT MODEL CALLED PICASSO (WHICH HAS SHOWN EFFICACY IN REDUCING VIOLENCE) THAT REQUIRE ADAPTATION FOR THE EIS SETTING. NEXT, DR. ROLIN WILL ADAPT THE PICASSO MODEL FOR THE EIS SETTING IN AN OPEN PILOT TRIAL, THROUGH AN ITERATIVE PROCESS THAT UTILIZES MIXED METHODS ASSESSMENT. FINALLY, DR. ROLIN WILL EVALUATE THE INTERVENTION’S FEASIBILITY THROUGH A PILOT RCT. TO FURTHER HER LONG-TERM CAREER GOAL OF BECOMING AN INDEPENDENT PHYSICIAN-INVESTIGATOR FOCUSED ON CREATING AND ADAPTING EBI TO REDUCE VIOLENCE RISK, DR ROLIN WILL PURSUE TRAINING IN THE FOLLOWING FOUR AREAS: (1) QUALITATIVE RESEARCH INCLUDING MIXED METHODS; (2) PSYCHOSOCIAL INTERVENTION ADAPTATION; (3) PSYCHOTHERAPY CLINICAL TRIALS; AND (4) GRANT WRITING. THE PRODUCT OF THIS STUDY WILL BE AN R01 OF A LARGE RCT TO TEST THE FEASIBILITY OF THE FIRST EBI OF CBT TO REDUCE VIOLENCE IN THE EIS SETTING. OVERALL, THIS AWARD WILL ENSURE DR. ROLIN’S SUCCESSFUL TRANSITION TO AN INDEPENDENT PHYSICIAN INVESTIGATOR STUDYING THE ASSESSMENT AND PREVENTION OF VIOLENCE AMONG ADULTS WITH SMI.
